Definitions
- the present invention is Orientia tsutsugamushi ).
- Tsutsugamushi disease (scrub typhus) is caused by the human infection of Orientia tsutsugamushi (hereinafter referred to as "tsutsugamushi bacteria"), which is mediated by tick larvae. It is a type of acute febrile infection that occurs mainly in the Asia- Pacific region, including the Russian coast, Japan, China, Malaysia, Thailand, Vietnam, and India, with a potential population of one billion people.
- Tsutsugamus bacteria are classified as a single species, but there are many serotypes with different major membrane antigen structures. To date, more than 20 Tsutsugamus serotypes have been distinguished according to the difference of these antigens. Are classified as Gilliam, Karp and Kato (Eisemann ans Osterman 1985, Am J Trop Med Hyg. Nov; 34 (6): 1173-8; Akira Tamura and Akiyoshi Kawamura 1991, J Feb. 1991, Vol. 29, No. 2; WOO-HYUN CHANG et al. 1993, J. CLIN. MICROBIOL. Mar. 1993, p. 598-605).
- Tsutsugamushi disease vaccine is based on the treatment of formalin or gamma-irradiation with killed vaccine (Killed-vaccine), 56KDa (56-kDa Type-Specific Antigen Gene (TSA56) protein, and 47KDa envelope protein (Subunit vaccine).

Infect. Immun. 65, 1541-1545; Yu et al., 2005. Am. J. Trop. Med. Hyg. 72 , 4
- Sca proteins of Tsutsugamu were expressed on the outer membrane of the bacteria as self-transporting proteins. These Sca proteins are divided into five subtypes: ScaA, ScaB, ScaC, ScaD, and ScaE. The amino acid sequences of the proteins, except ScaB, found only in Boryeong strains, were found to be relatively homologous between serotypes (ScaA: 65.8). ⁇ 81.8%, ScaC: 77.4-97.5%, ScaD: 69.7-93.8%, ScaE: 62.8-85.3%) (Ha et al., 2011 Infect Immun 79: 1718-27).
- the present invention confirms that ScaA and ScaC induce antibody responses in patients with Tsutsugamus disease, and this is completed by confirming that ScaA can function as a vaccine.
- the inventors have isolated the genomic DNA of the Tsutsugashimi Boryeong serotype as confirmed in the following example, and produced a primer based on the genetic information (GeneBank acession number AM494475.1) of the Boryeong serotype. (Underlined) and the corresponding amino acid sequence (underlined)), amplified DNA fragments were inserted into the pET28a vector to transform into E.
- the present inventors further divided a portion of the passenger domain region of the ScaA and ScaC protein produced and isolated from Escherichia coli to confirm that a portion of the passenger domain region of the ScaA and ScaC proteins functions as a vaccine.
- the Tsutsugamu strain (Born serotype) was infected with PBS, and the survival rate was calculated. All the ScaC-inoculated test groups died within 12 days of vaccination. However, in the test group inoculated with ScaA, one animal died on the 13th day of inoculation, and the other four animals survived (survival rate 80%).
- TSA56 used as a positive control survived (100% survival), and PBS used as a negative control all died within 11 days of inoculation with Boryeong strain (0% survival).

Abstract
Description
(Orientia
Nov;34(6):1173-8; Akira Tamura and Akiyoshi Kawamura 1991, J.CLIN.MICROBIOL. Feb.1991, Vol.29,No.2; WOO-HYUN CHANG et al. 1993, J.CLIN.MICROBIOL. Mar.1993,p.598-605). (Chang WH et al. 1995. J Korean Med Sci 10:227-38 ;227-38 ; Lee et al. 2010, J.Trop Med Hyg 83(4):930-5 Yang et al 2011, PLoS ONE 6(8):e22731).
(Watt G, Parola P.2003. Curr Opin Infect Dis. Oct;16(5):429-36 ; Strickman D et al. 1995. Lancet. 1996 Jul 13;348(9020):86-9.).
Trop.Dis.Bull. 37, 269~270; Seong et al., 1997. Infect.Immun. 65, 1541~1545; Yu et al., 2005. Am.J.Trop.Med.Hyg. 72, 458~46).
(ScaA: 65.8~81.8%, ScaC: 77.4~97.5%, ScaD:69.7~93.8%, ScaE: 62.8~85.3%) (Ha et al., 2011 Infect Immun 79:1718-27).

한편 아래의 실시예는 ScaA 항원을 TSA56(56-kDa Type-Specific Antigen, 56kDa) 항원과 함께 사용할 경우 백신으로서의 효과가 더 뛰어남을 보여준다.
(Seong et al., 1997. Infect.Immun. 65, 1541-1545; Yu et al., 2005. Am.J.Trop.Med.Hyg. 72, 458-46).
2010 May;78(5):1915-23.).
(Infect Immun. 1999 67(11): 6194~6197).

현탁된 세포를 Polytron homogenizer(Wheaton Inc. Millville, NJ)를 이용하여 균질화한 후 5분 동안 200 x g로 원심분리 하였다.
참고로 [도 1]에서 SP는 signal peptide, TM은 transmembrane, AT는 autotransporter domain, AA는 아미노산을 가리킨다.

Sca
Sca
ELISA
Immunoassay plates(96-well plates; Nunc, Rochester, NY)에 각 ScaA, ScaC 및 TSA56 항원을 0.05M bicarbonate buffer (pH 9.5)에 1㎍/㎖의 농도로 첨가한 뒤 100㎕/well로 분주하여 4℃에서 18시간 동안 코팅하였다.

Sca
