Abstract
疾患状態にあるヒト組織、ヒト細胞をin vivoで検討できる疾患ヒト化モデル動物を提供する。免疫不全動物の組織内にヒト多能性幹細胞が移植されて奇形腫が形成され、疾患誘起物質が腹腔内投与されることにより疾患化される。疾患誘起物質は、ストレプトゾトシン又はアロキサンである。健常人由来iPS細胞を移植し、2か月後にストレプトゾトシンを投与して糖尿病化したマウスより生育した奇形腫内の心筋及び血管平滑筋には中性脂肪の蓄積が見られた。
Description
本発明は、疾患ヒト化モデル動物及びその疾患ヒト化モデル動物の奇形腫組織に関する。
糖尿病は、世界的にみて患者数が急増している生活習慣病である。患者数急増の主原因としては、生活習慣の変化や、遺伝的要因があげられる。患者数急増により、適切な糖尿病の予防法及び治療法の開発、原因遺伝子の解明、発症機構の解明が急務となっており、このためには、適切な疾患モデル動物が必要不可欠である。
例えば糖尿病モデル動物としては、db/dbマウス(Hummel K P. et al., Science 153, 1127-1128., 1966)、KK-Ayマウス(Nishimura M., Exp. Anim., 18:147-157., 1969)、ob/obマウス(Herberg L & Coleman DL. Horm Metab Res. 7, 410-5., 1975)、NSYマウス(Ueda H. et al., Diabetologia 38, 503-508., 1995)、AKITAマウス(Yoshioka M. et al., Diabetes 46, 887-894., 1997)等が報告されている。
KK-Ayマウス及びNSYマウスは肥満を伴うモデルである。db/dbマウス及びob/obマウスは、レプチン受容体異常あるいはレプチン産生異常による肥満に伴うモデルである。AKITAマウスは、膵β細胞異常により糖尿病を発症するモデルである。
また、特許文献1には、野生マウス(Mus musculus molossinus)由来の近交系KORマウスと、市販のアトピー性皮膚炎モデルマウスであるNCマウス(NC/Jicマウス)との交配により作出された糖尿病モデルマウスが記載されている。
しかし、げっ歯類とヒトとでは脂肪代謝等が相違するため、上述のモデルマウスでは有用性は高いものとはいえない。本発明は、疾患状態にあるヒト組織、ヒト細胞をin vivoで検討しうる疾患ヒト化モデル動物を提供することを目的とする。
本発明にかかる疾患ヒト化モデル動物は、免疫不全動物の組織内にヒト多能性幹細胞が移植されて奇形腫が形成され、疾患誘起物質が腹腔内投与されることにより疾患化されたことを特徴とする。
また、本発明にかかる疾患ヒト化モデル動物は、免疫不全動物の組織内に代謝性疾患もしくは難病患者由来のヒト多能性幹細胞が移植されて奇形腫が形成されたことを特徴とする。
本発明によれば、疾患状態にあるヒト組織、ヒト細胞をin vivoで検討することができる。
以下、添付の図面を参照して本発明の実施形態について具体的に説明するが、当該実施形態は本発明の原理の理解を容易にするためのものであり、本発明の範囲は、下記の実施形態に限られるものではなく、当業者が以下の実施形態の構成を適宜置換した他の実施形態も、本発明の範囲に含まれる。
(第1実施形態)
本実施形態にかかる疾患ヒト化モデル動物は、免疫不全動物の組織内にヒト多能性幹細胞が移植されて奇形腫が形成され、疾患誘起物質が腹腔内投与されることにより疾患化されたことを特徴とする。

免疫不全動物としては、人為的又は先天的にT細胞機能免疫又はB細胞機能免疫あるいはそれらの双方の機能が著しく低下しているか、欠除している哺乳動物(ヒトを除く)であれば使用することができ、取り扱いの容易なラット又はマウスが好ましく、特に好ましくはNOD-SCIDマウスである。
SCIDマウスはC.B-17系統マウスで発見された自然発症の突然変異体であるが、NOD-SCIDマウスはこのSCIDマウスを基礎にして、異種移植をさらに向上する目的で改良が行われたマウスであり、NOD近交系マウスにSCID遺伝子を導入したマウスである。NOD-SCIDマウスで異種細胞の生着性が高いのは、NOD系統に由来する補体活性、マクロファージ機能やナチラルキラー(NK)細胞活性等の減退、がその原因として考えられている。
ヒト多能性幹細胞が移植される組織は特に限定されるものではないが好ましくは精巣である。後述の実施例で示されるように、組織として精巣を選択することにより移植する細胞数を大幅に減少させることができる。精巣内に投与されるヒト多能性幹細胞は、NOD-SCIDマウスの精巣内で奇形腫を形成可能な細胞であれば、特に限定されるものでないが、例えばヒトiPS細胞又はヒトES細胞である。iPS細胞としては、Oct3/4、Klf4、c-Myc、Tert、Nanog、Oct3/4、Sox2、Lin28等の転写因子を導入した細胞があげられる。ES細胞としては、RF8細胞、JI細胞、CGR8細胞、mg1.19細胞等があげられる。
疾患ヒト化モデル動物へのヒト多能性幹細胞の移植は、多能性幹細胞を培養し、得られた培養細胞を細胞乖離液で細胞毎に乖離させ、ついで細胞を分取したのち、細胞を疾患ヒト化モデル動物に移植することにより実施できる。
細胞の乖離は、組織又は細胞培養において使用される細胞乖離液を使用することができ、例えば0.25%程度のトリプシンと1mM濃度のEDTAの混合液等を用いることができる。乖離させた細胞の分取は例えば、遠心分離等により実施することができる。
ヒト多能性幹細胞の投与は、多能性幹細胞を生理的食塩水又はPBSに懸濁させ、注射器等を用いて疾患ヒト化モデル動物の組織に移植することによって実施することができる。移植に際しては、多能性幹細胞の細胞数が、1×105~1×107個程度を用いるのが好ましい。
移植した疾患ヒト化モデル動物の体内での奇形腫形成は、当該疾患ヒト化モデル動物を無菌下に通常の飼育を行うことによって実施することができる。奇形腫形成は、用いる細胞の種類や移植した細胞数によっても異なるが、概ね2~6週間程度であるのが好ましい。
本発明において、奇形腫は疾患ヒト化モデル動物から取り出すことができ、奇形腫の取り出しは、適切な外科的方法によって奇形腫を採取すればよく、特に制限はない。取り出された奇形腫を培養液中にて細胞乖離液で処理し、奇形腫から細胞を分取することができる。細胞乖離液としては、前述の多能性幹細胞を細胞乖離液で乖離させる際に使用したものを好適に使用することができる。細胞乖離液の反応は奇形腫の大きさにもよるが、概ね3時間程度でよく、反応後、例えばダルベッコ変法イーグル培地に最終濃度10%以上になるようにウシ胎児血清を添加したものを加え、酵素反応を停止させる。
腹腔内投与される疾患誘起物質は特に限定されるものではないが、惹起させる病態が糖尿病である場合は、例えばストレプトゾトシン、アロキサン等があげられ、好ましくはストレプトゾトシンである。ストレプトゾトシンは膵ランゲルハンス島を選択的に破壊する薬物であり、1回の投与により大部分のB細胞が脱落し、1型糖尿病を引き起こす。
また惹起させる病態が高血圧症である場合は、疾患誘起物質は例えばデオキシコルチコステロン・アセテート又は高食塩食である。また惹起させる病態が肥満、高脂血症、動脈硬化症である場合は、疾患誘起物質は例えば高脂肪食、高中性脂肪食、又は高加糖食を含む高カロリー食である。また惹起させる病態が心不全又は肺高血圧症である場合は、疾患誘起物質は例えばモノクロタリンである。また惹起させる病態が繊維筋痛症である場合は、疾患誘起物質は例えばレセルピンである。また惹起させる病態が大腸炎である場合は、疾患誘起物質は例えばデキストラン硫酸、アゾキシメタン、又はハプテンである。また惹起させる病態が肺線維症である場合は、疾患誘起物質は例えばブレヲマイシンである。また惹起させる病態が癌である場合は、疾患誘起物質は例えばアゾキシメタン、7,12-Dimethylbenz(α)anthracene、4-Nitroquinoline 1-Oxideである。また惹起させる病態が腎不全である場合は、両側腎臓を部分的に切除する。
(第2実施形態)
本実施形態にかかる疾患ヒト化モデル動物は、免疫不全動物の組織内に代謝性疾患もしくは難病患者由来のヒト多能性幹細胞が移植されて奇形腫が形成されたことを特徴とする。
本実施形態にかかる疾患ヒト化モデル動物は、免疫不全動物の組織内に代謝性疾患もしくは難病患者由来のヒト多能性幹細胞が移植されて奇形腫が形成されたことを特徴とする。
代謝性疾患は、特に限定されるものではないが、例えばATGL遺伝子欠損症、1型及び2型糖尿病、高血圧症、心臓血管疾患、中性脂肪蓄積心筋血管症から成る群より選択されてもよい。なおATGL遺伝子欠損症は原発性中性脂肪蓄積心筋血管症(triglyceride deposit cardiomyovasculopathy;TGCV)である。TGCVは、2008年、わが国の心臓移植待機症例より見出された新規疾患単位であり、心筋細、冠状動脈硬化巣に中性脂肪が蓄積する結果、重症心不全、不整脈をきたす難病である。また、難病疾患においても、特に限定されるものではなく、アルツハイマー病、筋ジストロフィー症、筋萎縮性側索硬化症、糖原病などの疾患群より選択されても良い。
第2実施形態にかかる発明では、中性脂肪蓄積心筋血管症等の希少疾患iPS細胞等を用いる事で、病態の解析にも有用である。
(1)-A ヒトiPS細胞の培養と奇形腫作成
既にリプログラムされたヒト正常iPS細胞を未分化状態で維持しながら継代培養した。移植2継代前より、Feeder細胞なしの培養へ変更し0.5x106個/1精巣の細胞を確保した。Rhoキナーゼ阻害薬(ROCK inhibitor)使用後、酵素処理を行い、遠心操作にて細胞を回収し、Matrigelを加え細胞懸濁液(0.5x106個/5μl+1μlのROCK inhibitor, 4℃)を作製した。麻酔下に、6週齢のNOD-SCIDマウスの腹部を切開し、精巣を出した。精巣内へ注射し、漏れがない事を確認の上、腹腔内へ精巣を戻した。腹部を縫合し、保温しながら麻酔からの回復を待って、ケージにマウスを戻した。

(1)-B 代謝疾患モデルマウスの作成
上記(1)と同様にして正常iPS細胞を6週齢のNOD-SCIDマウスの両側精巣内へ移植した。移植2か月後に腹部超音波にて奇形腫の形成を確認した。その後、代謝疾患モデルを作製する為、薬物投与を行った。糖尿病モデルを作成するため、ストレプトゾトシン50mg(クエン酸バッファーにて懸濁)を1日目、100mgを2日目に腹腔内投与した。投与前、1週後及び奇形腫回収後の血糖を測定した。血糖値が350mg/dl以上のマウスを糖尿病モデルマウスとした。移植4か月後(糖尿病化2か月後)に片方の奇形腫を回収した。
上記(1)と同様にして正常iPS細胞を6週齢のNOD-SCIDマウスの両側精巣内へ移植した。移植2か月後に腹部超音波にて奇形腫の形成を確認した。その後、代謝疾患モデルを作製する為、薬物投与を行った。糖尿病モデルを作成するため、ストレプトゾトシン50mg(クエン酸バッファーにて懸濁)を1日目、100mgを2日目に腹腔内投与した。投与前、1週後及び奇形腫回収後の血糖を測定した。血糖値が350mg/dl以上のマウスを糖尿病モデルマウスとした。移植4か月後(糖尿病化2か月後)に片方の奇形腫を回収した。
(2) 難病疾患患者iPS細胞の奇形腫及びモデルマウスの作成
難病患者iPS細胞を未分化状態で維持しながら継代培養した。移植2継代前より、Feeder細胞なしの培養へ変更し0.5x106個/1精巣の細胞を確保した。Rhoキナーゼ阻害薬(ROCK inhibitor)使用後、酵素処理を行い、遠心操作にて細胞を回収し、Matrigelを加え細胞懸濁液(0.5x106個/5μl+1μlのROCK inhibitor, 4℃)を作製した。麻酔下に、6週齢のNOD-SCIDマウスの腹部を切開し、精巣を出した。精巣内へ注射し、漏れがない事を確認の上、腹腔内へ精巣を戻した。腹部を縫合し、保温しながら麻酔からの回復を待って、ケージにマウスを戻した。移植3~4か月後に開腹し、奇形腫及び、その他のマウス臓器・血液を回収した。
(3)奇形腫及びマウス臓器等の解析
摘出した奇形腫を、4%パラホルムアルデヒド固定し、シュクロースにて凍結保護を行った。凍結し、クリオスタットにて薄切し、スライドガラスへ貼り付け、HE染色にて組織構造を確認した。
難病患者iPS細胞を未分化状態で維持しながら継代培養した。移植2継代前より、Feeder細胞なしの培養へ変更し0.5x106個/1精巣の細胞を確保した。Rhoキナーゼ阻害薬(ROCK inhibitor)使用後、酵素処理を行い、遠心操作にて細胞を回収し、Matrigelを加え細胞懸濁液(0.5x106個/5μl+1μlのROCK inhibitor, 4℃)を作製した。麻酔下に、6週齢のNOD-SCIDマウスの腹部を切開し、精巣を出した。精巣内へ注射し、漏れがない事を確認の上、腹腔内へ精巣を戻した。腹部を縫合し、保温しながら麻酔からの回復を待って、ケージにマウスを戻した。移植3~4か月後に開腹し、奇形腫及び、その他のマウス臓器・血液を回収した。
(3)奇形腫及びマウス臓器等の解析
摘出した奇形腫を、4%パラホルムアルデヒド固定し、シュクロースにて凍結保護を行った。凍結し、クリオスタットにて薄切し、スライドガラスへ貼り付け、HE染色にて組織構造を確認した。
心筋トロポニンT(cTNT)抗体を用いて奇形腫内の心筋細胞を、α-smooth musle actin (SMA)抗体を用いて血管平滑筋細胞を、Adipophilin抗体は細胞内脂肪滴構成膜を、ジアミノベンジジン法もしくは蛍光染色法にて同定した。Oil-Red O染色は脂肪滴を、LipidToxは中性脂肪を同定するために、化学染色する。その他目的となる細胞腫に応じて、抗体及び化学染色にて組織評価を行った。
図1に示されるように、健常人由来iPS細胞より発生した・生育した奇形腫内の心筋及び血管平滑筋では中性脂肪の蓄積は見られなかった(Negative Control)。健常人由来iPS細胞を移植し、2か月後にストレプトゾトシンを投与して糖尿病化したマウスより発生した・生育した奇形腫内の心筋及び血管平滑筋において中性脂肪の蓄積が見られた(DM-TGCV)。中性脂肪心筋血管蓄積症(TGCV)患者由来iPS細胞より発生・生育した奇形腫内の心筋及び血管平滑筋において中性脂肪の蓄積が見られた(n=4奇形腫, Positive Control)。ヒトATGL欠損iPS細胞を用いた場合は、ストレプトゾトシンを投与しなくても細胞要因のみで心筋・血管平滑筋細胞に脂肪蓄積が見られた。
採取した血液は静置処理にて血清を回収し、外注にて評価した。糖尿病化されたマウスに発生及び生育する、奇形腫内心筋細胞の細胞内中性脂肪蓄積量を定量的に解析する為、cTNT染色(赤色蛍光)及びLipidTox染色(緑色蛍光)を行い、cTNT陽性細胞(赤色)内のLipidTox(緑色)陽性領域をKeyenceBZ-9000を用い、自動解析を行った。
図2に示されるように、ヒトATGL欠損iPS細胞を用いた場合は、ストレプトゾトシンを投与しなくても細胞要因のみで心筋細胞に脂肪蓄積が見られた。
図3及び図4に示されるように、中性脂肪(LipidTox)と細胞内脂肪滴構成膜Adipophilinの2重染色において筋繊維構造のみられる組織内において、同じ部位でお互いが陽性であり、中性脂肪は細胞内に存在している事が確認できた。いずれの条件のマウス心筋細胞及び血管平滑筋において中性脂肪の蓄積は見られなかった。
図5に示されるように、糖尿病化しなかった奇形腫内及び、糖尿病化した奇形腫内に存在する心筋細胞(cTNT陽性細胞)内の中性脂肪(LipidTox)を定量的に解析し、陽性面積比がそれぞれ、0.059±0.027%及び15.06±3.51%であり、t検定においてp=0.01であり、優位に糖尿病化した奇形腫内の心筋細胞に中性脂肪の蓄積が見られた(各群n=4奇形腫、及び各奇形腫4切片ずつ)。
下記に、STZ投与正常iPS細胞移植群(DM)、PBS投与正常iPS細胞移植群(Control)、及び、中性脂肪心筋血管蓄積症(ATGL遺伝子欠損)患者由来iPS細胞移植群(ATGL(-))における移植後4か月後(STZ投与2か月後)の血液データを示す。
表1に示されるように、DM群に関しては、STZ投与の影響により、高血糖状態及びInsulin値の低下が見られた。なお、DM群に関してGOT/GPT/γGTPの上昇パターンにおけるGOT優位の上昇は、心筋等への脂肪蓄積による組織障害が考えられる(なお、マウス心筋には脂肪蓄積なし)。ATGL(-)群に関しては、奇形腫内にて脂肪滴代謝ができないため、エネルギー代謝不全の影響で高血糖化がみられた。
下記に、STZ投与正常iPS細胞移植群(DM)、PBS投与正常iPS細胞移植群(Control)、及び、中性脂肪心筋血管蓄積症(ATGL遺伝子欠損)患者由来iPS細胞移植群(ATGL(-))における脂肪代謝タンパクの免疫染色による発現を示す。なお、表2において、-:Negative stain、±:Faint stain、+: Positive stain、++:Massive stainである。
表2に示されるように、細胞要因(ATGL遺伝子欠損)によって、奇形腫内の筋・血管にORO陽性細胞内脂肪滴が観察された。マウスの糖尿病化によって、ORO陽性細胞内脂肪滴が著明に増加し、ATGL発現が筋肉・血管で上昇し、CGI-58は血管で増加していた。コントロール群ではこれらの酵素の上昇はみられなかった。
希少難病及び代謝性疾患の病態解析に利用できる。糖尿病性合併症の発症機序解明や治療法開発に利用できる。
Claims (10)
- 免疫不全動物の組織内にヒト多能性幹細胞が移植されて奇形腫が形成され、疾患誘起物質が腹腔内投与されることにより疾患化されたことを特徴とする疾患ヒト化モデル動物。
- 免疫不全動物の組織内に代謝性疾患もしくは難病患者由来のヒト多能性幹細胞が移植されて奇形腫が形成されたことを特徴とする疾患ヒト化モデル動物。
- 前記免疫不全動物はNOD-SCIDマウスであることを特徴とする請求項1又は2に記載の疾患ヒト化モデル動物。
- 前記組織は精巣であることを特徴とする請求項1乃至3の何れか1項に記載の疾患ヒト化モデル動物。
- 前記ヒト多能性幹細胞は、ヒトiPS細胞又はヒトES細胞であることを特徴とする請求項1乃至4の何れか1項に記載の疾患ヒト化モデル動物。
- 前記疾患誘起物質は、ストレプトゾトシン又はアロキサンであることを特徴とする請求項1に記載の疾患ヒト化モデル動物。
- 前記代謝性疾患は、中性脂肪蓄積心筋血管症であることを特徴とする請求項2に記載の疾患ヒト化モデル動物。
- 前記ヒト多能性幹細胞は、ヒトATGL欠損iPS細胞であることを特徴とする請求項2に記載の疾患ヒト化モデル動物。
- 前記疾患化は、I型糖尿病化であることを特徴とする請求項1に記載の疾患ヒト化モデル動物。
- 請求項1乃至9の何れか1項に記載の疾患ヒト化モデル動物に由来することを特徴とする奇形腫組織。
