具体的には、本発明の新規酵素「FA-HY」は、
(a)配列番号2に示されるアミノ酸配列からなる酵素タンパク質、
(b)配列番号2に示されるアミノ酸配列において、1又は複数個のアミノ酸が欠失及び/又は置換及び/又は挿入及び/又は付加されたアミノ酸配列を含み、かつ配列番号2に示されるアミノ酸配列からなる酵素タンパク質が有する酵素活性を有するタンパク質、あるいは
(c)配列番号1に示される塩基配列の相補鎖配列からなる核酸と、ストリンジェントな条件下でハイブリダイズする塩基配列によってコードされ、かつ配列番号2に示されるアミノ酸配列からなる酵素タンパク質が有する酵素活性を有するタンパク質である。
上記のようにアミノ酸が欠失、置換又は挿入されている場合、その欠失、置換、挿入の位置は、上記酵素活性が保持される限り、特に限定されない。
上記の「cis-12不飽和脂肪酸」、「cis-9不飽和脂肪酸」、「cis-14不飽和脂肪酸」、「cis-11不飽和脂肪酸」としては、それぞれ、12位にシス型二重結合を有する炭素数18の不飽和脂肪酸、9位にシス型二重結合を有する炭素数16の不飽和脂肪酸、14位にシス型二重結合を有する炭素数20の不飽和脂肪酸、11位にシス型二重結合を有する炭素数18又は20の不飽和脂肪酸であれば特に制限されず、例えば一価不飽和脂肪酸、二価不飽和脂肪酸、三価不飽和脂肪酸、四価不飽和脂肪酸、五価不飽和脂肪酸などが挙げられる。尚、本明細書において「脂肪酸」という場合、遊離の酸のみならず、エステル体、塩基性化合物との塩等をも包含する。「DHA」および「ミリストレイン酸」についても、遊離の酸のみならず、エステル体、塩基性化合物との塩等をも包含する。
本発明の製法1の「cis-12不飽和脂肪酸」としては、例えば、シス-9,シス-12-オクタデカジエン酸(リノール酸)、シス-6,シス-9,シス-12-オクタデカトリエン酸(γ-リノレン酸)、シス-9,シス-12,シス-15-オクタデカトリエン酸(α-リノレン酸)、シス-6,シス-9,シス-12,シス-15-オクタデカテトラエン酸(ステアリドン酸)の他、WO2013/168310で製造可能となった10-ヒドロキシ-シス-12-オクタデセン酸、10-ヒドロキシ-シス-6,シス-12-オクタデカジエン酸、10-ヒドロキシ-シス-12,シス-15-オクタデカジエン酸、10-ヒドロキシ-シス-6,シス-12,シス-15-オクタデカトリエン酸、10-オキソ-シス-12-オクタデセン酸、10-オキソ-シス-6,シス-12-オクタデカジエン酸、10-オキソ-シス-12,シス-15-オクタデカジエン酸、10-オキソ-シス-6,シス-12,シス-15-オクタデカトリエン酸、シス-5,シス-9,シス-12-オクタデカトリエン酸(ピノレン酸)、トランス-5,シス-9,シス-12-オクタデカトリエン酸(コルンビン酸)等が挙げられる。もちろん、当該基質はWO2013/168310以外の方法により得られたものであってもよい。

本発明の製法3の「cis-14不飽和脂肪酸」としては、例えば、シス-11,シス-14-エイコサジエン酸、シス-11,シス-14,シス-17-エイコサトリエン酸、シス-8,シス-11,シス-14-エイコサトリエン酸(ジホモ-γ-リノレン酸)、シス-8,シス-11,シス-14,シス-17-エイコサテトラエン酸、シス-5,シス-8,シス-11,シス-14-エイコサテトラエン酸(アラキドン酸)、シス-5,シス-11,シス-14-エイコサトリエン酸(シアドン酸)、シス-5,シス-11,シス-14,シス-17-エイコサテトラエン酸(ジュニペロン酸)等が挙げられる。

さらに、水和反応に「活性化剤」を用いてよく、例えば、NADH、NADPHからなる群から選ばれる1又は2の化合物が挙げられる。その添加濃度は水和反応が効率良く進む濃度であればよい。好ましくは0.1~20mM、より好ましくは1~10mMである。
本発明の製法5~8の「cis-12不飽和脂肪酸」、「cis-9不飽和脂肪酸」、「cis-14不飽和脂肪酸」、「cis-11不飽和脂肪酸」としては、上記の製法1~4に記載の基質と同様である。

具体的には、無水クロム酸2.67 gに硫酸 2.3 ml、水7.7 mlを加えた物を、アセトンを90 ml加えてクロム酸溶液を作成する。三角フラスコに、2 gの水酸化脂肪酸と、40 mlのアセトンを加え、氷上にてスターラーで撹拌しながら上記のクロム酸溶液を1滴ずつ加える。溶液が青色から抹茶色になったらクロム酸溶液の滴下をやめ、イソプロピルアルコールを用いて反応を停止する。析出した沈殿をろ紙を用いて濾過し、分液ロートに入れさらにジエチルエーテルを150 ml及びミリQ水300 mlを加えてよく振り、ジエチルエーテル層を数回ミリQ水で洗浄する。洗浄後のジエチルエーテル層に硫酸ナトリウム(無水)を適量加えて撹拌し残存水分を除去する。添加した無水硫酸ナトリウムをろ紙を用いてろ過し、得られたジエチルエーテル層をロータリーエバポレーターにて濃縮し、反応産物(オキソ脂肪酸)及び未反応の基質を抽出する。

水酸化脂肪酸、オキソ脂肪酸を含有する医薬の剤型としては、散剤、顆粒剤、丸剤、ソフトカプセル、ハードカプセル、錠剤、チュアブル錠、速崩錠、シロップ、液剤、懸濁剤、座剤、軟膏、クリーム剤、ゲル剤、粘付剤、吸入剤、注射剤等が挙げられる。これらの製剤は常法に従って調製されるが、オキソ脂肪酸等は水に難溶性であるため、植物性油、動物性油等の非親水性有機溶媒に溶解するか又は、乳化剤、分散剤もしくは界面活性剤等とともに、ホモジナイザー(高圧ホモジナイザー)を用いて水溶液中に分散、乳化させて用いる。
4℃にて保管している2%寒天を含むMRS高層培地よりラクトバチルス・アシドフィルスを15 ml のMRS液体培地(Difco製; pH 6.5)に植菌し、37℃で20時間培養を行った。培養後、3,000 rpm、4℃にて10分間遠心分離することにより集菌し、ラクトバチルス・アシドフィルスの菌体を得た。
尚、前記ラクトバチルス・アシドフィルスは、独立行政法人 製品評価技術基盤機構 特許微生物寄託センター(NPMD)に2014年1月17日に受託番号NITE BP-01788として寄託されている。
(1) ゲノムDNAの取得
前記ラクトバチルス・アシドフィルスを10 mlのMRS液体培地(Difco製)に植菌し、37℃で一晩静置培養後、遠心分離により集菌した。得られた菌体を1mlの滅菌水を用い2度洗浄した後、0.1 mlの滅菌水、0.125mlの溶菌バッファー(80 mM EDTA含有200 mM Tris-HCl buffer (pH8.0))、0.25 mlのTE飽和フェノールを加え、1分間vortexした後、遠心により上清を回収した。得られた上清0.2 mlに対し0.2 mlのPCI溶液(ナカライテスク製)を加え、ハンドシェイクした後15,000 rpm、4℃、5分の遠心により上清を0.125 ml回収し、0.0125 mlの5 M NaCl、0.31 mlのエタノールを加え、室温で10分間静置した後、15,000 rpm、4℃、5分で遠心した。上清を捨て、ペレットを0.2 mlの70%エタノールで洗浄した後ペレットを0.1 mlの滅菌水を加え、65℃10分インキュベートしてゲノムDNAを取得した。
(2) PCRによるFA-HY遺伝子の取得
公表されているラクトバチルス・アシドフィルス株の全ゲノム遺伝子配列においてWO/2013/168310のCLA-HYの遺伝子配列と相同性のある(約32%)open-reading-frame (ORF)をターゲットにした。ORFの開始コドンの5'側の配列をもとに、センスプライマー(配列番号3)を、また、終止コドンの3'側の配列を基にアンチセンスプライマー(配列番号4)を設計した。これらのプライマーを用い、ラクトバチルス・アシドフィルスのゲノムDNAを鋳型とし、PCRを行った。PCRの結果増幅された遺伝子断片約1.8 kbpの塩基配列の解読を行った結果、本遺伝子断片には開始コドンATGから始まり終止コドンTAGで終わる1,773 bpの一つのopen-reading-frame (ORF)(配列番号1)が含まれていることが判明し、本遺伝子をFA-HY遺伝子とした。本FA-HY遺伝子は配列番号2に示す590残基のアミノ酸からなるタンパク質をコードしていた。
大腸菌発現用ベクターpET21b (Novagen)とロゼッタ2(DE3)株からなる宿主ベクター系を用いた。ラクトバチルス・アシドフィルスのゲノムDNAを鋳型にPCRにて増幅したFA-HY遺伝子断片をpET21bに挿入し、発現ベクター(pFA-HY)を構築した。pFA-HYをロゼッタ2(DE3)株に形質転換し、形質転換株ロゼッタ/pFA-HY株を得た。得られたロゼッタ/pFA-HY株を0.5 mgアンピシリン、0.3 mgクロラムフェニコールを含む10 ml LB培地(1%バクトトリプトン(Difco)、0.5%酵母エキス、1%塩化ナトリウムを含む培地(pH 7.0))にて37℃、300 rpmで好気的に培養し、OD600 nmが0.5になった時点で1 M IPTGを1 μl添加し、さらに16℃にて18時間培養した。培養後3,000 rpmにて10分間遠心分離し、ロゼッタ/pFA-HY株の湿菌体を得た。
脂肪酸水和酵素誘導形質転換大腸菌を用い各種不飽和脂肪酸から水酸化脂肪酸の生成試験を行った。反応液は、脂肪酸水和酵素誘導形質転換大腸菌(湿菌体重量 0.3 g/ml)、NADH (5 mM)、FAD (0.1 mM)、不飽和脂肪酸 (100 mg)、BSA(10 mg)を含む100 mM リン酸カリウムバッファー(pH 6.5)で全量を1 mlとした。反応は、37℃にて、酸素吸着剤アネロパック(三菱化学)存在下で嫌気的に16~60時間、120 rpmで振とうしながら行った。反応後、反応液(1 ml)に対して、2 mlのクロロホルム、2 mlのメタノール、1 mlの0.5%KClを加え撹拌し、クロロホルム層を回収した。回収したクロロホルム層を遠心エバポレーターにて濃縮し、反応産物及び未反応の基質を抽出した。抽出物の一部を用いてメチルエステル化した後にガスクロマトグラフィーにて反応産物を評価した。
実施例4より得られた抽出物(基質と生成物(水酸化脂肪酸)を含む混合物)を中圧クロマトグラフィーに供し、カラムから出てきた溶液をフラクションに分けて回収した。回収した各フラクションをLC/MS及びガスクロマトグラフィーにより分析し、生成物(水酸化脂肪酸)のみが含まれたフラクションを集めてロータリーエバポレーターにて濃縮した。得られた最終生成物(水酸化脂肪酸)の一部を用いてメチルエステル化した後に、ガスクロマトグラフィーにて生成物の純度を評価した。その結果、各基質からそれぞれ純度98%以上の生成物を得た。生成物は、NMRや二次元NMR、GC-MS分析などを用いて化学構造を決定した。その結果、シス-9-ヘキサデセン酸(パルミトレイン酸)からは純度98%以上で10-ヒドロキシ-ヘキサデカン酸を得ることができた。シス-11-オクタデセン酸(シス-バクセン酸)からは純度98%以上で12-ヒドロキシ-オクタデカン酸を得ることができた。シス-9,シス-12-オクタデカジエン酸(リノール酸)からは純度98%以上で13-ヒドロキシ-シス-9-オクタデセン酸を得ることができた。シス-6,シス-9,シス-12-オクタデカトリエン酸(γ-リノレン酸)からは純度98%以上で13-ヒドロキシ-シス-6,シス-9-オクタデカジエン酸を得ることができた。シス-9,シス-12,シス-15-オクタデカトリエン酸(α-リノレン酸)からは純度98%以上で13-ヒドロキシ-シス-9,シス-15-オクタデカジエン酸を得ることができた。シス-6,シス-9,シス-12,シス-15-オクタデカテトラエン酸(ステアリドン酸)からは純度98%以上で13-ヒドロキシ-シス-6,シス-9,シス-15-オクタデカトリエン酸を得ることができた。シス-11,シス-14-エイコサジエン酸からは純度98%以上で12-ヒドロキシ-シス-14-エイコセン酸と15-ヒドロキシ-シス-11-エイコセン酸を得ることができた。シス-11,シス-14,シス-17-エイコサトリエン酸からは純度98%以上で12-ヒドロキシ-シス-14,シス-17-エイコサジエン酸と15-ヒドロキシ-シス-11,シス-17-エイコサジエン酸を得ることができた。シス-8,シス-11,シス-14-エイコサトリエン酸(ジホモ-γ-リノレン酸)からは純度98%以上で15-ヒドロキシ-シス-8,シス-11-エイコサジエン酸と12-ヒドロキシ-シス-8,シス-14-エイコサジエン酸を得ることができた。シス-5,シス-8,シス-11-エイコサトリエン酸(ミード酸)からは純度98%以上で12-ヒドロキシ-シス-5,シス-8-エイコサジエン酸を得ることができた。シス-8,シス-11,シス-14,シス-17-エイコサテトラエン酸からは純度98%以上で12-ヒドロキシ-シス-8,シス-14,シス-17-エイコサトリエン酸と15-ヒドロキシ-シス-8,シス-11,シス-17-エイコサトリエン酸を得ることができた。シス-5,シス-8,シス-11,シス-14-エイコサテトラエン酸(アラキドン酸)からは純度98%以上で15-ヒドロキシ-シス-5,シス-8,シス-11-エイコサトリエン酸と12-ヒドロキシ-シス-5,シス-8,シス-14-エイコサトリエン酸を得ることができた。10-ヒドロキシ-シス-12-オクタデセン酸からは純度98%以上で10,13-ジヒドロキシ-オクタデカン酸を得ることができた。10-ヒドロキシ-シス-6,シス-12-オクタデカジエン酸からは純度98%以上で10,13-ジヒドロキシ-シス-6-オクタデセン酸を得ることができた。10-ヒドロキシ-シス-12,シス-15-オクタデカジエン酸からは純度98%以上で10,13-ジヒドロキシ-シス-15-オクタデセン酸を得ることができた。10-ヒドロキシ-シス-6,シス-12,シス-15-オクタデカトリエン酸からは純度98%以上で10,13-ジヒドロキシ-シス-6,シス-15-オクタデカジエン酸を得ることができた。10-オキソ-シス-12-オクタデセン酸からは純度98%以上で10-オキソ-13-ヒドロキシ-オクタデカン酸を得ることができた。10-オキソ-シス-6,シス-12-オクタデカジエン酸からは純度98%以上で10-オキソ-13-ヒドロキシ-シス-6-オクタデセン酸を得ることができた。10-オキソ-シス-12,シス-15-オクタデカジエン酸からは純度98%以上で10-オキソ-13-ヒドロキシ-シス-15-オクタデセン酸を得ることができた。10-オキソ-シス-6,シス-12,シス-15-オクタデカトリエン酸からは純度98%以上で10-オキソ-13-ヒドロキシ-シス-6,シス-15-オクタデカジエン酸を得ることができた。シス-5,シス-9,シス-12-オクタデカトリエン酸(ピノレン酸)からは純度98%以上で13-ヒドロキシ-シス-5,シス-9-オクタデカジエン酸を得ることができた。トランス-5,シス-9,シス-12-オクタデカトリエン酸(コルンビン酸)からは純度98%以上で13-ヒドロキシ-トランス-5,シス-9-オクタデカジエン酸を得ることができた。シス-5,シス-11,シス-14-エイコサトリエン酸(シアドン酸)からは純度98%以上で15-ヒドロキシ-シス-5,シス-11-エイコサジエン酸を得ることができた。シス-5,シス-11,シス-14,シス-17-エイコサテトラエン酸(ジュニペロン酸)からは純度98%以上で15-ヒドロキシ-シス-5,シス-11,シス-17-エイコサトリエン酸を得ることができた。シス-4,シス-7,シス-10,シス-13,シス-16,シス-19-ドコサヘキサエン酸(DHA)からは純度98%以上で14-ヒドロキシ-シス-4,シス-7,シス-10,シス-16,シス-19-ドコサペンタエン酸を得ることができた。シス-9-テトラデセン酸(ミリストレイン酸)からは純度98%以上で10-ヒドロキシ-テトラデカン酸を得ることができた。
特開2007-259712号公報に記載される方法や、WO/2013/168310に記載される方法により得られたL.プランタルムFERM BP-10549菌株由来の精製脱水素酵素(CLA-DH)を用い水酸化脂肪酸からオキソ脂肪酸の生成試験を行った。反応液は、精製脱水素酵素 (酵素量83μg)、0.5 mM NAD+、0.5mg水酸化脂肪酸を含む50 mM リン酸カリウムバッファー(pH 8.0)で全量を1 mlとした。反応は、37℃にて、酸素吸着剤アネロパック(三菱化学)存在下で嫌気的に24時間、120 rpmで振とうしながら行った。反応後、反応液よりブライダイヤー法にて脂質を抽出した。抽出物をメチルエステル化した後ガスクロマトグラフィーにてオキソ脂肪酸の生成を評価した結果、水酸化脂肪酸13-ヒドロキシ-シス-9-オクタデセン酸を基質とした場合はオキソ脂肪酸13-オキソ-シス-9-オクタデセン酸(0.01 mg)の生成を確認した。13-ヒドロキシ-シス-9,シス-15-オクタデカジエン酸を基質とした場合は13-オキソ-シス-9,シス-15-オクタデカジエン酸(0.02 mg)の生成を、13-ヒドロキシ-シス-6,シス-9-オクタデカジエン酸を基質とした場合は13-オキソ-シス-6,シス-9-オクタデカジエン酸(0.06 mg)の生成を確認した。
同様の手法を用いることにより、各水酸化脂肪酸を基質にすることにより、生成物であるオキソ脂肪酸を得ることができる。

本発明のPPARリガンド剤のPPARα,γの活性化能の測定は、Nobuyuki Takahashiら、FEBS Letters 514 (2002) p315-322、「Dual action of isoprenols from herbal medicines on both PPARgamma and PPARalpha in 3T3-L1 adipocytes and HepG2 hepatocytes.」のMaterial and methods Reporter plasmids and luciferase assaysの欄を参照して行った。詳細には、PPARα,γリガンド活性は、PPARリガンド結合領域とGAL4 DNA結合領域との融合タンパクに対する結合および標的遺伝子活性化をルシフェラーゼの発現で評価するレポーター・アッセイにより測定した。具体的には、CV-1細胞に、PPARα,γリガンド結合領域とGAL4 DNA結合領域の融合タンパク質をコードするDNAを含むプラスミドと、GAL4結合DNA配列にルシフェラーゼをつないだレポータープラスミドを導入し、この細胞に、以下に記載のリガンドを添加して、インキュベートした後にルシフェラーゼ活性の検出を行った。
サンプルはエタノールを用いて濃度調整を行った。陰性コントロールにはエタノール、陽性対照にはPPARα,γリガンド剤 GW7647(10nM)、troglitazone(5μM)をそれぞれ用いた。
水酸化脂肪酸、オキソ脂肪酸の代表として、(1)13-ヒドロキシ-シス-9-オクタデセン酸(「13-OH LA」と示す)、(2)13-オキソ-シス-9-オクタデセン酸(「13-OXO LA」と示す)、(3)10,13-ジヒドロキシ-オクタデカン酸(「10,13-OH LA」と示す)、(4)13-ヒドロキシ-シス-9,シス-15-オクタデカジエン酸(「13-OH ALA」と示す)、(5)13-オキソ-シス-9,シス-15-オクタデカジエン酸(「13-OXO ALA」と示す)、(6)10,13-ジヒドロキシ-シス-15-オクタデセン酸(「10,13-OH ALA」と示す)(7)13-ヒドロキシ-シス-6,シス-9-オクタデカジエン酸(「13-OH GLA」と示す)、(8)13-オキソ-シス-6,シス-9-オクタデカジエン酸(「13-OXO GLA」と示す)、(9)10,13-ジヒドロキシ-シス-6-オクタデセン酸(「10,13-OH GLA」と示す)、(10)15-ヒドロキシ-シス-5,シス-8,シス-11-エイコサトリエン酸(「15-OH AA」と示す)のPPARα、γリガンド活性のデータを図1及び図2に示す。このうち、(4)、(6)、(8)及び(9)の希少脂肪酸は新規脂肪酸である。

本出願は、日本で出願された特願2014-011855(出願日:平成26年1月24日)を基礎としており、その内容はすべて本明細書に包含されるものとする。
Claims (34)
- 以下の(a)~(c)のいずれかの酵素タンパク質。
(a)配列番号2に示されるアミノ酸配列からなる酵素タンパク質
(b)配列番号2に示されるアミノ酸配列において、1又は複数個のアミノ酸が欠失及び/又は置換及び/又は挿入及び/又は付加されたアミノ酸配列を含み、かつ水和反応を触媒する酵素活性を有するタンパク質
(c)配列番号1に示される塩基配列の相補鎖配列からなる核酸と、ストリンジェントな条件下でハイブリダイズする塩基配列によってコードされ、かつ水和反応を触媒する酵素活性を有するタンパク質 - 請求項1に記載の酵素タンパク質を含む、ラクトバチルス・アシドフィルス(Lactobacillus acidophilus)菌体または菌体破砕物。
- 請求項1に記載の酵素タンパク質をコードする核酸。
- 請求項3に記載の核酸を含むベクター。
- 請求項4に記載のベクターで形質転換された宿主細胞。
- 請求項5に記載の宿主細胞を培養し、該培養物から請求項1記載の酵素タンパク質を回収することを含む、該酵素の製造方法。
- 12位にシス型二重結合を有する炭素数18の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、13位に水酸基を有する炭素数18の水酸化脂肪酸を製造する方法。
- 12位にシス型二重結合を有する炭素数18の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、13位に水酸基を有する炭素数18の水酸化脂肪酸を誘導し、該水酸化脂肪酸から、脱水素反応又は化学的酸化により、13位にカルボニル基を有する炭素数18のオキソ脂肪酸を製造する方法。
- 9位にシス型二重結合を有する炭素数16の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、10位に水酸基を有する炭素数16の水酸化脂肪酸を製造する方法。
- 9位にシス型二重結合を有する炭素数16の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、10位に水酸基を有する炭素数16の水酸化脂肪酸を誘導し、該水酸化脂肪酸から、脱水素反応又は化学的酸化により、10位にカルボニル基を有する炭素数16のオキソ脂肪酸を製造する方法。
- 14位にシス型二重結合を有する炭素数20の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、15位に水酸基を有する炭素数20の水酸化脂肪酸を製造する方法。
- 14位にシス型二重結合を有する炭素数20の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、15位に水酸基を有する炭素数20の水酸化脂肪酸を誘導し、該水酸化脂肪酸から、脱水素反応又は化学的酸化により、15位にカルボニル基を有する炭素数20のオキソ脂肪酸を製造する方法。
- 11位にシス型二重結合を有する炭素数18又は20の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、12位に水酸基を有する炭素数18又は20の水酸化脂肪酸を製造する方法。
- 11位にシス型二重結合を有する炭素数18又は20の不飽和脂肪酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、12位に水酸基を有する炭素数18又は20の水酸化脂肪酸を誘導し、該水酸化脂肪酸から、脱水素反応又は化学的酸化により、12位にカルボニル基を有する炭素数18又は20のオキソ脂肪酸を製造する方法。
- 12位にシス型二重結合を有する炭素数18の不飽和脂肪酸が、リノール酸、γ-リノレン酸、α-リノレン酸、ステアリドン酸、10-ヒドロキシ-シス-12-オクタデセン酸、10-ヒドロキシ-シス-6,シス-12-オクタデカジエン酸、10-ヒドロキシ-シス-12,シス-15-オクタデカジエン酸、10-ヒドロキシ-シス-6,シス-12,シス-15-オクタデカトリエン酸、10-オキソ-シス-12-オクタデセン酸、10-オキソ-シス-6,シス-12-オクタデカジエン酸、10-オキソ-シス-12,シス-15-オクタデカジエン酸、10-オキソ-シス-6,シス-12,シス-15-オクタデカトリエン酸、ピノレン酸又はコルンビン酸である請求項7または8に記載の方法。
- 9位にシス型二重結合を有する炭素数16の不飽和脂肪酸が、パルミトレイン酸である請求項9または10に記載の方法。
- 14位にシス型二重結合を有する炭素数20の不飽和脂肪酸が、シス-11,シス-14-エイコサジエン酸、シス-11,シス-14,シス-17-エイコサトリエン酸、ジホモ-γ-リノレン酸、シス-8,シス-11,シス-14,シス-17-エイコサテトラエン酸、アラキドン酸、シアドン酸又はジュニペロン酸である請求項11または12に記載の方法。
- 11位にシス型二重結合を有する炭素数18又は20の不飽和脂肪酸が、シス-バクセン酸、シス-11-シス-14-エイコサジエン酸、シス-11,シス-14,シス-17-エイコサトリエン酸、ジホモ-γ-リノレン酸、ミード酸、シス-8,シス-11,シス-14,シス-17-エイコサテトラエン酸又はアラキドン酸である請求項13または14に記載の方法。
- シス-4,シス-7,シス-10,シス-13,シス-16,シス-19-ドコサヘキサエン酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、14-ヒドロキシ-シス-4,シス-7,シス-10,シス-16,シス-19-ドコサペンタエン酸を製造する方法。
- シス-4,シス-7,シス-10,シス-13,シス-16,シス-19-ドコサヘキサエン酸から、請求項1に記載の酵素タンパク質を用いた水和反応により、14-ヒドロキシ-シス-4,シス-7,シス-10,シス-16,シス-19-ドコサペンタエン酸を誘導し、該水酸化脂肪酸から、脱水素反応又は化学的酸化により、14-オキソ-シス-4,シス-7,シス-10,シス-16,シス-19-ドコサペンタエン酸を製造する方法。
- シス-9-テトラデセン酸(ミリストレイン酸)から、請求項1に記載の酵素タンパク質を用いた水和反応により、10-ヒドロキシ-テトラデカン酸を製造する方法。
- シス-9-テトラデセン酸(ミリストレイン酸)から、請求項1に記載の酵素タンパク質を用いた水和反応により、10-ヒドロキシ-テトラデカン酸を誘導し、該水酸化脂肪酸から、脱水素反応又は化学的酸化により、10-オキソ-テトラデカン酸を製造する方法。
- 脱水素反応のための脱水素酵素が乳酸菌由来である請求項8、10、12、14~18、20または22に記載の方法。
- 乳酸菌がラクトバチルス・プランタルム(Lactobacillus plantarum)FERM BP-10549菌株である請求項23に記載の方法。
- 請求項7の方法によって製造される、13-ヒドロキシ-シス-9,シス-15-オクタデカジエン酸、13-ヒドロキシ-シス-6,シス-9,シス-15-オクタデカトリエン酸、10,13-ジヒドロキシ-シス-6-オクタデセン酸、10,13-ジヒドロキシ-シス-15-オクタデセン酸、10,13-ジヒドロキシ-シス-6,シス-15-オクタデカジエン酸、10-オキソ-13-ヒドロキシ-シス-6-オクタデセン酸、10-オキソ-13-ヒドロキシ-シス-15-オクタデセン酸、10-オキソ-13-ヒドロキシ-シス-6,シス-15-オクタデカジエン酸、13-ヒドロキシ-シス-5,シス-9-オクタデカジエン酸または13-ヒドロキシ-トランス-5,シス-9-オクタデカジエン酸。
- 請求項11の方法によって製造される、15-ヒドロキシ-シス-11,シス-17-エイコサジエン酸、15-ヒドロキシ-シス-8,シス-11-エイコサジエン酸、15-ヒドロキシ-シス-8,シス-11,シス-17-エイコサトリエン酸、15-ヒドロキシ-シス-5,シス-11-エイコサジエン酸または15-ヒドロキシ-シス-5,シス-11,シス-17-エイコサトリエン酸。
- 請求項13の方法によって製造される、12-ヒドロキシ-シス-14-エイコセン酸、12-ヒドロキシ-シス-14,シス-17-エイコサジエン酸または12-ヒドロキシ-シス-8,シス-14,シス-17-エイコサトリエン酸。
- 請求項8の方法によって製造される、13-オキソ-シス-6,シス-9-オクタデカジエン酸、13-オキソ-シス-6,シス-9,シス-15-オクタデカトリエン酸、10,13-ジオキソ-シス-6-オクタデセン酸、10,13-ジオキソ-シス-15-オクタデセン酸、10,13-ジオキソ-シス-6,シス-15-オクタデカジエン酸、13-オキソ-シス-5,シス-9-オクタデカジエン酸または13-オキソ-トランス-5,シス-9-オクタデカジエン酸。
- 請求項12の方法によって製造される、15-オキソ-シス-11,シス-17-エイコサジエン酸、15-オキソ-シス-8,シス-11-エイコサジエン酸、15-オキソ-シス-8,シス-11,シス-17-エイコサトリエン酸、15-オキソ-シス-5,シス-11-エイコサジエン酸または15-オキソ-シス-5,シス-11,シス-17-エイコサトリエン酸。
- 請求項14の方法によって製造される、12-オキソ-シス-14-エイコセン酸、12-オキソ-シス-14,シス-17-エイコサジエン酸、12-オキソ-シス-8,シス-14-エイコサジエン酸、12-オキソ-シス-5,シス-8-エイコサジエン酸または12-オキソ-シス-8,シス-14,シス-17-エイコサトリエン酸。
- 請求項19の方法によって製造される、14-ヒドロキシ-シス-4,シス-7,シス-10,シス-16,シス-19-ドコサペンタエン酸。
- 請求項20の方法によって製造される、14-オキソ-シス-4,シス-7,シス-10,シス-16,シス-19-ドコサペンタエン酸。
- 請求項25~32のいずれか1項に記載の水酸化脂肪酸またはオキソ脂肪酸を含む脂肪酸含有物。
- 請求項25~32のいずれか1項に記載の水酸化脂肪酸またはオキソ脂肪酸を生産するための、請求項1に記載の酵素タンパク質を含む、ラクトバチルス・アシドフィルス(Lactobacillus acidophilus)菌体または菌体破砕物の使用。
