Abstract
Description
沈降時の充填剤の体積(b)が前記ベッド体積(a)の1.02~1.30倍となる量の充填剤含有スラリー全量を前記小径カラム内に圧縮しながら充填した後、
通液方向を複数回切り替えながら前記充填カラムに移動相を通液することを特徴とする。
本明細書中で、用語「圧縮係数」は、下式で定義される。
大径カラムにおける圧縮係数は、充填剤の種類によっても異なるが、一般的には1.02~1.30の範囲となる。圧縮係数が高すぎると、通液時の圧力が高くなり処理能力が低下すると共に、クロマトグラフ分離の「リーディング」を招き分離性能が低下する。他方で、圧縮係数が低すぎると、充填状態が不安定となり使用中にカラムの分離性能が変化すると共に、クロマトグラフ分離の「テーリング」を招き分離性能が低下する。本発明で提供する方法は、大径カラムと同等の1.02~1.30の圧縮係数を有する小径カラムの作製に適用可能であり、その中でも特に1.04~1.20の圧縮係数を有する小径カラムの作製に好適である。
本明細書中で、「沈降体積(b)」は、以下の方法にて測定した体積を意味する。
1)0.5Mの塩化ナトリウム水溶液に懸濁した充填剤を内径2cm以上の目盛り付容器に入れて充填剤を沈降させる。
2)容器に振動を与えた後、充填剤を沈降させる。
3)2)の操作を繰返し行い、充填剤層の上端が動かなくなるまで完全に沈降させたときの充填剤層の体積を「沈降体積」とする。
なお、上記1)、2)の沈降操作は、容器を静置して重力下(1G=9.8m/s2)で行う方法と、容器を遠心機に供して遠心力下(630G以下)で行う方法があり、いずれの方法を用いてもよい。
本明細書中で、用語「スラリー」は、充填剤および液体の分散物を意味し、このスラリーはカラムに充填剤を投入又は充填する時に使用可能である。液体は沈殿物や浮遊物を含まない均一な液体であれば特に限定されるものではないが、極性溶媒からなる液体であることが好ましく、極性溶媒に可溶な添加物が含まれていてもよい。極性溶媒としては例えば、水、水溶性溶媒(メタノール、エタノールなどのアルコール類、アセトニトリルなど)、およびそれらの混合溶媒が挙げられ、アルコール類の添加は通液時の圧力を高くできることが期待できるため、より充填しづらい小径カラムの充填に有効な場合がある。添加物としては例えば、酸、塩基、塩類(特に塩化ナトリウムなど)、ポリエチレングリコールが挙げられる。水と水溶性溶媒の質量比は、例えば、100:0~50:50程度、好ましくは95:5~60:40程度、さらに好ましくは90:10~70:30程度の範囲から設定できる。添加剤の濃度は、例えば、0~10M(mol/L)程度、好ましくは0.1~5M程度、さらに好ましくは0.2~3M程度の範囲から設定できる。
本明細書中で、用語「スラリー濃度」は、下式で定義される。
スラリー濃度(%)={(スラリーに含まれる充填剤の沈降体積(b))/(スラリー体積(c))}×100
スラリー濃度は30~70%が好ましく、35~65%がより好ましく、40~60%が最も好ましい。この範囲を超えるとスラリー粘度が高くなり、均一な充填が難しくなる。また逆に、この範囲を下回ると充填に時間がかかる。
本発明において充填剤の充填工程は、空のクロマトグラフィー用カラムに必要量の充填剤を投入する工程(工程1)と、投入された充填剤を沈降・圧縮する工程(工程2)、よりなる。
投入するスラリー量(体積c)={(ベッド体積(a))×(圧縮係数)}/{(スラリー濃度)/100}
すなわち沈降時の充填剤の体積(b)が前記ベッド体積(a)の1.02~1.30倍となる量の充填剤含有スラリーを前記小径カラム内に充填する。
(工程2)における充填剤の沈降・圧縮方法としては、所定の圧縮係数が達成可能な一般的な沈降・圧縮方法であれば特に限定されないが、分離性能の高いカラムを再現性良く作製するためには以下の(圧縮法a)または(圧縮法b)、あるいは(圧縮法a)と(圧縮法b)とを組み合わせた方法が好ましい。
カラムにポンプで液体を通液して充填剤を沈降・圧縮する。通液の流速を調節することで充填剤の圧縮の程度を調節する。通液する液体は沈殿物や浮遊物を含まない均一な液体であれば特に限定されるものではないが、極性溶媒からなる液体であることが好ましく、極性溶媒に可溶な添加物が含まれていてもよい。極性溶媒としては例えば、水、アルコール類、アセトニトリル、およびそれらの混合溶媒が挙げられ、添加物としては例えば、酸、塩基、塩類、ポリエチレングリコールが挙げられる。
(圧縮法b)機械的圧縮法
カラムに可動栓を取り付け、可動栓を動かすことで沈降・圧縮する。可動栓を動かす際には、カラム両端の開口部のうち少なくとも1つを液体の排出口として開口する。
上記圧縮法a、圧縮法bなどを採用することによって、充填剤含有スラリーをカラム内で圧縮することで、全ての充填剤がカラムに充填される。この充填の際や、充填の後に、移動相をカラムに通液する。充填後の移動相の通液は、充填剤の充填状態の均一化の目的で実施される。なお充填後の移動相の通液は、後述する通液方向切り替え処理の1回目の通液を兼ねる。
本発明においては通液方向を切り替えることを特徴とする。本明細書中で「通液方向を切り替える」とは、カラムの両端の2箇所の開口部のうち移動相の入口として使用していた方を出口とし、出口として使用していた方を入口とすることで通液方向を反転させることを意味し、カラムの接続された装置の配管の弁の切り替えによっても可能であるし、また一旦カラムを装置から取り外してカラムの接続の向きを入れ替えることによっても可能である。通液方向の切り替えは充填状態の均一化を促進する効果を有する。効果的な切り替え回数は充填剤の種類、充填状態、圧縮係数、カラム内径、ベッド高、移動相の組成により異なってくるが、回数としては3回以上が好ましく、より好ましくは5回以上(特に7回~30回程度)である。また、十分に均一なカラムが得られない場合にはさらに切り替え操作を追加して通液し、均一化することができる。
通液方向を切り替えて移動相を通液する場合、初めの通液方向(以下、正方向という)と、反転方向(以下、逆方向という)の通液速度は、それぞれ同一又は異なって、例えば、100~10000cm/h、好ましくは500~7000cm/h、より好ましくは1000~5000cm/hである。正方向及び逆方向いずれの場合でも、通液速度は一定である必要はない。通液速度を変更する場合は、少なくとも最も速い速度が、例えば、1000cm/h以上、好ましくは1200cm/h以上、より好ましくは1500cm/h以上であることが好ましい。正方向1回当たり及び逆方向1回当たりの通液時間は、例えば、1分以上、好ましくは1~60分、より好ましくは1~30分である。また正方向と逆方向の通液速度パターンは、異なっていてもよいが、同じであることが好ましい。正方向と逆方向とで通液速度パターンを異ならせる場合、逆方向の通液速度が遅くてもよい。また正方向と逆方向の通液速度パターンが同じになる組み合わせ(第1のセット)を複数回実施した後、これらとは異なる設定で正方向と逆方向の通液パターンが同じになる組み合わせ(第2のセット)を複数回実施し、以後、必要に応じて第3以降のセットをそれぞれ複数回実施してもよい。
通液方向を切り替えてカラムに通液する移動相は、充填剤スラリーをカラムに充填する時の移動相と同等の溶媒が使用でき、この充填時の移動相と同じであってもよく、異なっていてもよい。異なっている場合、充填剤スラリーの溶媒に比べて添加剤濃度が低減したり、水の割合が高められたりしてもよい。
通液方向を切り替えての移動相の通液が完了した後は、必要に応じて、再度、圧縮工程(後圧縮工程)を実施してもよい。この圧縮工程の条件は、通液方向を切り替えての移動相の通液を実施する前の圧縮工程(前圧縮工程)と同様の条件であってもよく、前圧縮工程よりも強い又は弱い圧縮であってもよい。
<所定圧縮係数に到達するタイミング>
目的の圧縮係数まで充填剤が充填されるタイミング(すなわち目的のベッド高になるタイミング)は、全工程が完了するまでであれば特に限定されないが、例えば、前記前圧縮工程完了時(すなわち通液方向切り替え前)であってもよく、前圧縮工程完了時には目的の圧縮係数よりも小さい圧縮係数で充填剤を充填してもよい。後者の場合、その後の通液方向を切り替えての通液を実施している時の適当な段階で目的の圧縮係数に到達してもよく、この通液方向を切り替えての通液後に後圧縮工程を実施し、この後圧縮工程の段階で目的の圧縮係数に到達してもよい。
カラムの非対称係数(As)は、カラムの充填状態を評価するパラメータとして一般的に使用されているものであり、以下に示す方法で算出することができる。
(Asの算出方法)
Asの算出は、充填済みのカラムに評価用の物質を打ち込み、移動相を一定速度で供給した際に得られるクロマトグラムから算出される。この際、評価用物質の検出には、例えばHPLC(高速液体クロマトグラフィー)に使用される検出方法を用いることができ、紫外吸収(UV)検出器や、電気伝導度検出器、示差屈折率検出器等により、精度よく検出することが可能である。
本発明においてAsは、具体的には、評価対象カラムに対して少量の評価用試料を打ち込んだ際に得られるクロマトグラムを基に算出される。クロマトグラムの取得方法には、評価用試料の違いにより、アセトンを使用する方法(クロマトグラム取得方法a)、及び塩化ナトリウムを使用する方法(クロマトグラム取得方法b)がある。
(クロマトグラム取得方法a)アセトンを使用する方法
アフィニティクロマトグラフィー用充填剤、イオン交換クロマトグラフィー用充填剤、サイズ排除クロマトグラフィー用充填剤、ハイドロキシアパタイトクロマトグラフィー用充填剤、に対して適用可能である。カラムベッド体積の2倍以上に相当する量の水を通液した評価対象カラムに対して、カラムベッド体積の1%に相当する量の濃度1%(v/v)アセトン水溶液を打ち込んだ後、水を50cm/hの流速でカラムに供給し、カラム下流に設置した紫外検出器により検出波長280nmにおけるクロマトグラムを得る。
(クロマトグラム取得方法b)塩化ナトリウムを使用する方法
アフィニティクロマトグラフィー用充填剤、イオン交換クロマトグラフィー用充填剤、サイズ排除クロマトグラフィー用充填剤、疎水性相互作用クロマトグラフィー用充填剤、および疎水基とイオン交換基を併せ持つミックスモードクロマトグラフィー用充填剤に対して適用可能である。カラムベッド体積の2倍以上に相当する量の0.5M塩化ナトリウム水溶液を通液した評価対象カラムに対して、カラムベッド体積の1%に相当する量の1M塩化ナトリウム水溶液を打ち込んだ後、移動相を50cm/hの流速でカラムに供給し、カラム下流に設置した電気伝導度検出器により電気伝導度のクロマトグラムを得る。

Asは1.00が理想とされるが、実使用においては、0.80~1.80の範囲が好適範囲とされており、特に0.90~1.50が好ましく、0.95~1.30の範囲が最も好ましい。Asがこの範囲を下回る場合には、充填剤の過剰圧縮である、又は充填状態が不均一である、若しくは充填剤層にクラッキングが生じていることが推定されるし、他方上回る場合には充填剤の圧縮が不十分であることが推定される。
(実施例1)
充填剤として、KANEKA KanCapA(株式会社カネカ製:体積基準メジアン径65~85μmの高度架橋セルロースを基材としたアフィニティクロマトグラフィー用充填剤)を使用した。充填剤を0.5Mの塩化ナトリウム水溶液にてスラリー化した後、以下の手順にてベッド高及び圧縮係数の異なる6本のカラムを作製し、評価した。
<実施手順>
1)目盛り付遠沈管にスラリーを入れ、遠心機(KUBOTA製、テーブルトップ遠心機2410)にて2000rpm(630Gに相当)で20秒間遠心し、充填剤を沈降させた。続いて、遠沈管への加振と遠心(20000rpmで20秒間)とを交互に繰返して充填剤層を完全に沈降させてスラリーに含まれる充填剤の沈降体積を求めた。
2)スラリー全量をグラスフィルターに移し、フィルター上で充填剤を移動相(0.5Mの塩化ナトリウムを含む20%エタノール水溶液)で洗った後、充填剤を移動相でスラリー化した。1)で求めた沈降体積とスラリー体積よりスラリー濃度を算出した(スラリー濃度50%)。
3)カラム内径と表1に示す目的のベッド高及び圧縮係数、及び2)で求めたスラリー濃度より充填に必要なスラリー量を算出し、必要量のスラリーを0.5cm径ガラスカラムに投入した。
4)移動相(0.5Mの塩化ナトリウムを含む20%エタノール水溶液)を153cm/hの流速でカラムに通液して充填剤を沈降させた後、流速を増加させ(ベッド高4.9cm(圧縮係数1.12)迄:6115cm/h、ベッド高5.1cm(圧縮係数1.08)迄:4586cm/h、ベッド高5.3cm(圧縮係数1.04)迄:4586cm/h、ベッド高17.4cm(圧縮係数1.12)迄:7338cm/h、ベッド高18.0cm(圧縮係数1.08)迄:6115cm/h、ベッド高18.7cm(圧縮係数1.04)迄:5197cm/h)、充填剤層を表1に示す目的のベッド高よりも0.5cm高い位置まで各々圧縮した。その後、機械的圧縮法により充填剤層を目的のベッド高まで圧縮した。
5)移動相を4586cm/hの流速でカラムに通液し、1分毎に計19回通液方向を切り替えた。続けて、移動相の通液速度を3669cm/hとし、1分毎に計19回通液方向を切り替えた。
<結果>
カラムの性能を前述のAsの算出方法に従い評価した。評価用試料としてアセトンを使用した。性能の結果を表1に示す。
充填剤として、MabSelect SuRe(GE Healthcare社製:体積基準メジアン径85μmの高度架橋アガロースを基材としたアフィニティクロマトグラフィー用充填剤)を使用した。充填剤を0.5Mの塩化ナトリウム水溶液にてスラリー化し、以下の手順にてベッド高及び圧縮係数の同じ2本のカラムを作製し、当該手法の再現性を確認した。
<実施手順>
1)メスシリンダーにスラリーを入れ、重力下による沈降と振動を繰り返して充填剤層を完全に沈降させてスラリーに含まれる充填剤の沈降体積を求め、スラリー濃度を算出した(スラリー濃度50%)。
2)カラム内径と表2に示す目的のベッド高及び圧縮係数、及び1)で求めたスラリー濃度より充填に必要なスラリー量を算出し、必要量のスラリーを0.66cm径ガラスカラムに投入した。
3)移動相(0.5M塩化ナトリウム水溶液)を877cm/hの流速でカラムに通液して充填剤層を一次圧縮した後、機械的圧縮法により充填剤層を目的のベッド高である7cmまで圧縮した。
4)移動相を3508cm/hの流速でカラムに通液し、10分毎に計11回通液方向を切り替えた。
<結果>
カラムの性能を前述のAsの算出方法に従い評価した。評価用試料として塩化ナトリウムを使用した。性能の結果を表2に示す。
充填剤として、Fractogel EMD COO-(M)(Merck社製:粒子径40~90μmの合成ポリマーを基材としたイオン交換クロマトグラフィー用充填剤)を使用した。充填剤は0.5Mの塩化ナトリウム水溶液にてスラリー化した。
<実施手順>
1)メスシリンダーにスラリーを入れ、重力下による沈降と振動を繰り返して充填剤層を完全に沈降させてスラリーに含まれる充填剤の沈降体積を求めた。
2)スラリー全量をグラスフィルターに移し、フィルター上で充填剤を2Mの塩化ナトリウム水溶液で洗った後、充填剤を2Mの塩化ナトリウム水溶液でスラリー化した。1)で求めた沈降体積とスラリー体積よりスラリー濃度を算出した(スラリー濃度50%)。
3)カラム内径と表3に示す目的のベッド高及び圧縮係数、及び2)で求めたスラリー濃度より充填に必要なスラリー量を算出し、必要量のスラリーを0.66cm径ガラスカラムに投入した。
4)2M塩化ナトリウム水溶液を88cm/hの流速でカラムに通液して充填剤を沈降させた後、流速を1754cm/hに増加させ、充填剤層を一次圧縮した。その後、機械的圧縮法により充填剤層を目的のベッド高である7cmまで圧縮した。
5)カラム内を移動相(0.5M塩化ナトリウム水溶液)に置換した。
6)移動相を1754cm/hの流速で10分間カラムに通液した後、通液方向を切り替えて1315cm/hの流速で10分間カラムに通液した。この操作を再度繰返し、3回の通液方向の切り替えで計40分間通液した。
<結果>
カラムの性能を前述のAsの算出方法に従い評価した。評価用試料として塩化ナトリウムを使用した。性能の結果を表3に示す。
充填剤として、KANEKA KanCapA(株式会社カネカ製:体積基準メジアン径65~85μmの高度架橋セルロースを基材としたアフィニティクロマトグラフィー用充填剤)を使用した。充填剤を0.5M塩化ナトリウム水溶液にてスラリー化し、以下の手順にてベッド高及び圧縮係数の同じ2本のカラムを作製し、当該手法の再現性を確認した。
<実施手順>
1)メスシリンダーにスラリーを入れ、重力下による沈降と振動を繰り返して充填剤層を完全に沈降させてスラリーに含まれる充填剤の沈降体積を求めた。
2)スラリー全量をグラスフィルターに移し、フィルター上で充填剤を20%エタノール水溶液で洗った後、充填剤を20%エタノール水溶液でスラリー化した。1)で求めた沈降体積とスラリー体積よりスラリー濃度を算出した(スラリー濃度50%)。
3)カラム内径と表4に示す目的のベッド高及び圧縮係数、及び2)で求めたスラリー濃度より充填に必要なスラリー量を算出し、必要量のスラリーを1.0cm径ガラスカラムに投入した。
4)20%エタノール水溶液を381cm/hの流速でカラムに通液して充填剤を沈降させた後、流速を1529cm/hに増加させ、充填剤層を一次圧縮した。その後、機械的圧縮法により充填剤層を目的のベッド高である25cmまで圧縮した。
5)カラム内を移動相(水)で置換した後、移動相を764cm/hで5分間、1528cm/hで5分間、2293cm/hで5分間通液した。通液方向を切り替えて同様の通液を行った。
6)移動相を2293cm/hの流速でカラムに通液し、2分毎に計17回通液方向を切り替えた。
<結果>
カラムの性能を前述のAsの算出方法に従い評価した。評価用試料として塩化ナトリウムを使用した。性能の結果を表4に示す。

カラムの性能を前述のAsの算出方法に従い評価した。評価用試料として塩化ナトリウムを使用した。性能の結果を表7に示す。本比較例は、充填剤、カラムサイズ、圧縮係数、充填方法のいずれも実施例4と同じであり、充填後の通液条件のみ実施例4と異なるが、通液方向を複数回切り替えながら通液した実施例4ではAsは1.01および1.20と好適範囲内であったのに対し、通液方向の切り替えが1回であった本比較例ではAsは0.49と好適範囲を大きく下回った。
充填剤として、MabSelect SuRe(GE Healthcare社製:体積基準メジアン径85μmの高度架橋アガロースを基材としたアフィニティクロマトグラフィー用充填剤)を使用した。充填剤は0.5Mの塩化ナトリウム水溶液にてスラリー化した。ベッド高及び圧縮係数の同じ2本のカラムを作製した。
<実施手順>
1)メスシリンダーにスラリーを入れ、重力下による沈降と振動を繰り返して充填剤層を完全に沈降させてスラリーに含まれる充填剤の沈降体積を求め、スラリー濃度を算出した(スラリー濃度50%)。
2)カラム内径と表8に示す目的のベッド高及び圧縮係数、及び1)で求めたスラリー濃度より充填に必要なスラリー量を算出し、必要量のスラリーを0.66cm径ガラスカラムに投入した。
3)移動相(0.5M塩化ナトリウム水溶液)を877cm/hの流速でカラムに通液して充填剤層を一次圧縮した後、機械的圧縮法により充填剤層を目的のベッド高である7cmまで圧縮した。
<結果>
カラムの性能を前述のAsの算出方法に従い評価した。評価用試料として塩化ナトリウムを使用した。性能の結果を表8に示す。本比較例は、充填剤、カラムサイズ、圧縮係数、充填方法のいずれも実施例2と同じであり、充填後のカラムへの通液の有無のみ実施例2と異なるが、通液方向を複数回切り替えながら通液した実施例2ではAsは1.00と好適範囲内であったのに対し、充填後の通液を実施しなかった本比較例ではAsは0.61および0.49と好適範囲を大きく下回った。
充填剤として、Fractogel EMD COO-(M)(Merck社製:粒子径40~90μmの合成ポリマーを基材としたイオン交換クロマトグラフィー用充填剤)を使用した。充填剤は0.5Mの塩化ナトリウム水溶液にてスラリー化した。
<実施手順>
1)メスシリンダーにスラリーを入れ、重力下による沈降と振動を繰り返して充填剤層を完全に沈降させてスラリーに含まれる充填剤の沈降体積を求めた。
2)スラリー全量をグラスフィルターに移し、フィルター上で充填剤を2Mの塩化ナトリウム水溶液で洗った後、充填剤を2Mの塩化ナトリウム水溶液でスラリー化した。1)で求めた沈降体積とスラリー体積よりスラリー濃度を算出した(スラリー濃度50%)。
3)カラム内径と表9に示す目的のベッド高及び圧縮係数、及び2)で求めたスラリー濃度より充填に必要なスラリー量を算出し、必要量のスラリーを0.66cm径ガラスカラムに投入した。
4)2M塩化ナトリウム水溶液を88cm/hの流速でカラムに通液して充填剤を沈降させた後、流速を1754cm/hに増加させ、充填剤層を一次圧縮した。その後、機械的圧縮法により充填剤層を目的のベッド高である7cmまで圧縮した。
5)カラム内を移動相(0.5M塩化ナトリウム水溶液)に置換した。
6)移動相を1754cm/hの流速で10分間カラムに通液した後、通液方向を切り替えて1315cm/hの流速で10分間カラムに通液した。
<結果>
カラムの性能を前述のAsの算出方法に従い評価した。評価用試料として塩化ナトリウムを使用した。性能の結果を表9に示す。本比較例は、充填剤、カラムサイズ、圧縮係数、充填方法のいずれも実施例3と同じであり、充填後の通液条件のみ実施例3と異なるが、通液方向を複数回切り替えながら通液した実施例3ではAsは0.98と好適範囲内であったのに対し、通液方向の切り替えが1回であった本比較例ではAsは0.77と好適範囲を下回った。

Claims (8)
- クロマトグラフィー用小径カラムの作製方法であって、充填剤を所定の圧縮係数にてカラムに充填した後、カラムへの通液方向を複数回切り替えながら移動相を通液することを特徴とする方法。
- クロマトグラフィー用小径カラムの作製方法であって、充填剤を所定の圧縮係数よりも小さい圧縮係数にてカラムに充填した後、カラムへの通液方向を複数回切り替えながら移動相を通液し、その後、所定の圧縮係数となるよう充填剤を圧縮することを特徴とする方法。
- クロマトグラフィー用小径カラムの内径が3cm以下である、請求項1または2に記載の方法。
- 所定の圧縮係数が1.02~1.30である、請求項1~3のいずれかに記載の方法。
- クロマトグラフィー用小径カラムに充填剤を充填して、所定ベッド体積(a)の充填カラムを製造する方法であって、
沈降時の充填剤の体積(b)が前記ベッド体積(a)の1.02~1.30倍となる量の充填剤含有スラリー全量を前記小径カラム内に圧縮しながら充填した後、
通液方向を複数回切り替えながら前記充填カラムに移動相を通液する請求項1~4のいずれかに記載の方法。 - 充填剤の基材がアガロース、セルロース、または合成ポリマーである、請求項1~5のいずれかに記載の方法。
- 充填剤の体積基準メジアン径が10μm~500μmである、請求項1~6のいずれかに記載の方法。
- 請求項1~7のいずれか1項記載の方法で作製された、非対称係数が0.80~1.80の範囲であるクロマトグラフィー用小径カラム。
