Abstract
げっ歯類に男性ホルモンを投与する工程を含む、げっ歯類の毛周期における成長期から退行期への移行を促進する方法を提供する。さらには、げっ歯類において、(a)毛周期の休止期において、同調した成長期を誘導する工程、(b)男性ホルモンを投与する工程、(c)薬剤を投与する工程、及び(d)男性ホルモン投与により促進されるげっ歯類の毛周期における成長期から退行期への移行が薬剤投与により抑制される程度を評価する工程、を含む薬剤の有効性を評価する方法を提供する。本発明により簡便なin vivo男性型脱毛症モデルにおいて、発毛剤、育毛剤等の有効性を効果的に評価が可能となる。
Description
本発明は動物モデルを用いた発毛剤又は育毛剤等の効果的な評価方法に関する。
発毛剤や育毛剤等の薬剤のヒトにおける有効性を推測する手段として、動物モデルを使用した発毛・育毛作用の評価が行われている。当該評価に用いる動物としては、ウサギ、ラット、マウス、ベニガオザル等が知られている(非特許文献1)。
しかしながら、動物を用いた評価方法の中でヒトの男性型脱毛症に類似した病態である「男性ホルモンの影響に起因する成長期期間の短縮(成長期から退行期への移行の促進)」が認められる動物はベニガオザルのみであった。また、ベニガオザルを用いた薬剤の評価はフォトトリコグラム法やフォリクログラム法を用いており、薬剤による成長期間の延長作用や、成長期から退行期への移行の遅延化作用等を評価することは不可能であった。更に評価期間が6ヶ月程度必要であるため、薬剤のスクリーニングには不向きであった。他の動物では、ゴールデンシリアンハムスターが男性ホルモン投与により体毛の再成長が抑制されること、B6CBAF1マウスが男性ホルモン投与によって脱毛を生じることが報告されているものの、男性ホルモンが成長期から退行期への移行を促進することについて何ら報告はない(非特許文献2、3)。従って、ヒトの男性型脱毛症に類似した病態である毛周期における成長期から退行期への移行が促進された新たな動物モデルを作成する必要があった。
しかしながら、動物を用いた評価方法の中でヒトの男性型脱毛症に類似した病態である「男性ホルモンの影響に起因する成長期期間の短縮(成長期から退行期への移行の促進)」が認められる動物はベニガオザルのみであった。また、ベニガオザルを用いた薬剤の評価はフォトトリコグラム法やフォリクログラム法を用いており、薬剤による成長期間の延長作用や、成長期から退行期への移行の遅延化作用等を評価することは不可能であった。更に評価期間が6ヶ月程度必要であるため、薬剤のスクリーニングには不向きであった。他の動物では、ゴールデンシリアンハムスターが男性ホルモン投与により体毛の再成長が抑制されること、B6CBAF1マウスが男性ホルモン投与によって脱毛を生じることが報告されているものの、男性ホルモンが成長期から退行期への移行を促進することについて何ら報告はない(非特許文献2、3)。従って、ヒトの男性型脱毛症に類似した病態である毛周期における成長期から退行期への移行が促進された新たな動物モデルを作成する必要があった。

本発明は、ヒトの男性型脱毛症に類似した病態である、毛周期における成長期から退行期への移行が促進された新たな動物モデルを提供することを目的とする。更には、当該病態モデルにおける発毛剤や育毛剤等の薬剤の評価を短期間で効率的に行う方法を提供することを目的とする。
本発明者らは、上記課題を達成すべく検討したところ、意外にもげっ歯類に男性ホルモンを投与することにより、げっ歯類の毛周期における成長期から退行期への移行が促進されることを見いだした。
すなわち、本発明は以下の通りである。
(1)げっ歯類に男性ホルモンを投与する工程を含む、げっ歯類の毛周期における成長期から退行期への移行を促進する方法。
(2)げっ歯類において、
(a)毛周期の休止期において、同調した成長期を誘導する工程、
(b)男性ホルモンを投与する工程、
(c)薬剤を投与する工程、及び
(d)男性ホルモン投与により促進されるげっ歯類の毛周期における成長期から退行期への移行が薬剤投与により抑制される程度を評価する工程、
を含む薬剤の男性型脱毛症への有効性を評価する方法。
(3)げっ歯類が有色毛マウスである、(1)又は(2)に記載の方法。
(4)男性ホルモンが、テストステロン、ジヒドロテストステロン、デヒドロエピアンドロステロン、アンドロステロン、アンドロステンジオン、性腺刺激ホルモン放出ホルモン及びこれらの誘導体から選ばれる1種以上である、(1)~(3)のいずれかに記載の方法。
(5)毛包の組織学的評価、皮膚明度、皮膚彩度及びTGF-β発現量から選ばれる1種以上を用いて成長期から退行期への移行を評価する、(1)~(4)のいずれかに記載の方法。
(6)(a)~(c)を含む工程を複数回繰り返した後に、(d)男性ホルモン投与により促進されるげっ歯類の毛周期における成長期から退行期への移行が薬剤投与により抑制される程度を評価する工程を行うことを特徴とする、(2)~(5)のいずれかに記載の薬剤の男性型脱毛症への有効性を評価する方法。
すなわち、本発明は以下の通りである。
(1)げっ歯類に男性ホルモンを投与する工程を含む、げっ歯類の毛周期における成長期から退行期への移行を促進する方法。
(2)げっ歯類において、
(a)毛周期の休止期において、同調した成長期を誘導する工程、
(b)男性ホルモンを投与する工程、
(c)薬剤を投与する工程、及び
(d)男性ホルモン投与により促進されるげっ歯類の毛周期における成長期から退行期への移行が薬剤投与により抑制される程度を評価する工程、
を含む薬剤の男性型脱毛症への有効性を評価する方法。
(3)げっ歯類が有色毛マウスである、(1)又は(2)に記載の方法。
(4)男性ホルモンが、テストステロン、ジヒドロテストステロン、デヒドロエピアンドロステロン、アンドロステロン、アンドロステンジオン、性腺刺激ホルモン放出ホルモン及びこれらの誘導体から選ばれる1種以上である、(1)~(3)のいずれかに記載の方法。
(5)毛包の組織学的評価、皮膚明度、皮膚彩度及びTGF-β発現量から選ばれる1種以上を用いて成長期から退行期への移行を評価する、(1)~(4)のいずれかに記載の方法。
(6)(a)~(c)を含む工程を複数回繰り返した後に、(d)男性ホルモン投与により促進されるげっ歯類の毛周期における成長期から退行期への移行が薬剤投与により抑制される程度を評価する工程を行うことを特徴とする、(2)~(5)のいずれかに記載の薬剤の男性型脱毛症への有効性を評価する方法。
本発明において、ヒトの男性型脱毛症に類似した病態である毛周期における成長期から退行期への移行が促進された新たな動物モデルが見いだされ、また、発毛作用又は育毛作用等を有する薬剤の効率的な評価を行うことが可能となった。
本発明の1つの態様は、げっ歯類に男性ホルモンを投与する工程を含む、げっ歯類の毛周期における成長期から退行期への移行を促進する方法である。
げっ歯類とは、例えばマウス、ラット及びハムスターが挙げられ、好ましくはマウスであり、C57BL系マウス、C3H系マウス及びCBA系マウスが特に好ましい。
男性ホルモンとは、例えばテストステロン、ジヒドロテストステロン、デヒドロエピアンドロステロン、アンドロステロン、アンドロステンジオン、性腺刺激ホルモン放出ホルモン及びこれらの誘導体が挙げられ、好ましくは、テストステロン又はジヒドロテストステロンであり、特に好ましくはテストステロンである。これらの男性ホルモンは単独で使用しても良く、2種以上の男性ホルモンを組み合わせても良い。
げっ歯類とは、例えばマウス、ラット及びハムスターが挙げられ、好ましくはマウスであり、C57BL系マウス、C3H系マウス及びCBA系マウスが特に好ましい。
男性ホルモンとは、例えばテストステロン、ジヒドロテストステロン、デヒドロエピアンドロステロン、アンドロステロン、アンドロステンジオン、性腺刺激ホルモン放出ホルモン及びこれらの誘導体が挙げられ、好ましくは、テストステロン又はジヒドロテストステロンであり、特に好ましくはテストステロンである。これらの男性ホルモンは単独で使用しても良く、2種以上の男性ホルモンを組み合わせても良い。
男性ホルモンの投与方法は、経口又は非経口のいずれでも良いが、好ましくは非経口であり、毛周期を評価する部位に直接塗布するのが特に好ましい。男性ホルモンの投与開始時期(投与期間)は、毛周期を評価する部位に直接塗布する場合、好ましくは毛周期の休止期に体毛を抜去して同調した成長期を誘導した後である。男性ホルモンの投与量は、毛周期を評価する部位に直接塗布する場合、好ましくは0.01%~0.5%の溶液であり、10μL~200μLの範囲で塗布するのが好ましい。
また、本発明の他の1つの態様は、

薬剤とは、発毛作用又は育毛作用等を有する薬剤であり、例えばミノキシジル、フィナステリド、デュタステリド、ラタノプロスト、ビマトプロスト、ケトコナゾール、シクロスポリン、FK506等が挙げられる。また、本発明の方法は発毛作用又は育毛作用等を有する薬剤をスクリーニングする方法としても利用可能であるため、該薬剤には発毛作用又は育毛作用等の効果を有するか否かが不明な薬剤も含まれる。
上記の方法では、毛周期の休止期に毛周期を成長期に強制同調させることが好ましい。成長期への移行時期が同調されることにより、成長期から退行期への移行時期の動物個体ごとのばらつきが小さくなり、薬剤の効果の評価が容易となる。毛周期の休止期に毛周期を成長期に強制同調させる方法は特に限定されず、例えば体毛の抜去により毛周期を成長期に強制同調させることが可能である。抜去の方法としては、手で直接評価部位の毛を引き抜く方法、鉗子や脱毛ワックスを用いて毛を引き抜く方法等が挙げられる。
本発明の薬剤の男性型脱毛症に対する有効性の評価は、男性ホルモン投与によるげっ歯類の毛周期における成長期から退行期への移行の作用に対して、薬剤投与を併用することにより男性ホルモンの移行促進作用がどの程度抑制されるかという視点から行われる。男性ホルモン投与による毛周期における成長期から退行期への移行促進は、男性型脱毛症で認められる病態に近い症状を反映していると考えられ、薬剤の男性型脱毛症に対する予防又は治療効果を評価するのに好ましい環境となる。また、男性ホルモン投与により毛周期における成長期から退行期への移行を促進することにより成長期の期間が短縮されるため、男性ホルモンを投与しない場合と比較して薬剤の成長期に対する期間延長作用(成長期から退行期への移行遅延作用)を検出することが容易になる。薬剤の投与方法は、経口又は非経口のいずれでも良く、使用する薬剤にとって適切な投与ルートを選択すればよい。
薬剤の投与時期は、男性ホルモンの投与時期と同時期でも良く、また一定期間前後しても良い。同時期に投与される場合、1日の投与時期に特に制限は無く、男性ホルモンと同時に投与しても良く、一定期間時間を前後させて投与させても良い。
発毛・育毛効果の観察時期としては、毛包の形態が成長期後期から退行期にかけて大きく変化する観点から成長期後期から退行期が望ましい。薬剤投与の有無による効果の比較という観点からは、効果の差を検出しやすい点で男性ホルモン単独投与群における退行期に相当する時期が好ましい。マウスを用いる場合、好ましい効果の観察時期は、体毛の抜去後15日目~ 22日目であり、さらに好ましくは体毛の抜去後17日目~ 20日目に設定される。
毛周期の成長期の短縮または退行期への移行促進は、毛包の組織学的評価、皮膚明度、皮膚彩度又はTGF-β発現量の変化を測定することにより評価する。
毛包の組織学的評価は、毛包の形態が毛周期に伴って変化する(Sven Muller-Rover et al. The Journal of Investigative Dermatology 117 (2001), 3-15)という事実を利用して行うことができる。特に毛周期における成長期から退行期への移行時期には毛包の退縮が顕著に認められるため、組織学的評価が容易である。また、毛包の形態の変化は皮膚明度や皮膚彩度の変化といった指標で間接的に評価することが可能である。毛包の退縮が顕著に認められると当該部位の剃毛後の皮膚の色が成長期と比較して明るいピンク色を呈する傾向がある。このような皮膚明度や皮膚彩度の変化を測定する手段としては色査計による数値化、写真を撮影し画像処理技術を用いた数値化、肉眼観察によるスコア化等が挙げられる。皮膚明度や皮膚彩度を指標として評価する場合、動物を屠殺する必要がないため、少ない動物数で経時的評価が可能であり、成長期から退行期への移行をより詳細に評価できる。当該評価方法を使用する場合、げっ歯類は有色毛を有する動物が好ましく、特に好ましくは有色毛マウスである。有色毛マウスとしては、例えば、C57BL系マウス、C3H系マウス、CBA系マウスが挙げられる。
毛周期の成長期から退行期への移行が進むに従い毛包周辺部位におけるTGF-βの発現量が増大することが知られているため、毛周期の成長期の短縮または退行期への移行促進は、毛包周辺組織におけるTGF-β発現量の変化を測定することでも評価可能である。TGF-β発現量を評価する方法としては、評価部位の毛包を含む皮膚をホモジネートし、RT-PCR法を用いてTGF-βのmRNA量を測定する方法、ELISA法を用いてTGF―βタンパク量を測定する等の方法が挙げられる。
薬剤の有効性の評価は、上述の(a)~(c)の工程を1回行った後、(d)の工程を行うことで実施可能であるが、1回の工程で判定出来ない場合、又は十分な判定ができない場合は、2回目以降の毛周期においても(a)~(c)の工程を行うことが可能である。2回以上の毛周期において(a)~(c)の工程を行った後に、最終的に(d)の工程を行うことにより、薬剤の効果を検出する感度を向上させることが可能となる。この場合、男性ホルモン及び薬剤の投与時期に特に限定はないが、1回目の工程における男性ホルモン及び薬剤の投与開始から、最終的な効果判定時期の直前まで継続的に投与することが好ましい。
薬剤の有効性の評価は、上述の(a)~(c)の工程を1回行った後、(d)の工程を行うことで実施可能であるが、1回の工程で判定出来ない場合、又は十分な判定ができない場合は、2回目以降の毛周期においても(a)~(c)の工程を行うことが可能である。2回以上の毛周期において(a)~(c)の工程を行った後に、最終的に(d)の工程を行うことにより、薬剤の効果を検出する感度を向上させることが可能となる。この場合、男性ホルモン及び薬剤の投与時期に特に限定はないが、1回目の工程における男性ホルモン及び薬剤の投与開始から、最終的な効果判定時期の直前まで継続的に投与することが好ましい。
以下に、試験例を挙げ、本発明を更に詳しく説明する。なお、本発明はこれら実施例に限定されるものでは無い。
試験例1:男性ホルモン負荷マウス退行期抑制評価モデルの作成
<試験方法>
C57BL/6マウス(8週齢、メス)の背部体毛を休止期に抜去し、同調した成長期を誘導した。体毛抜去10日後よりエタノールに溶解した0.05%、0.15%或いは0.5%テストステロンを1日1回、1匹当たり0.1mLを測定日の前日まで塗布した。マウスは一群5匹使用した。体毛抜去から18日及び19日後にマウスを屠殺した後、評価部位を剃毛し、背部皮膚の明度を色彩色査計(CR-400、コニカミノルタ)にて測定し、明度をLuminescence値(L値)として数値化した。更に評価部位の病理切片を作成し、ヘマトキシリン・エオジン染色を実施した後組織学的に観察し、毛周期の判定を行った。
<試験結果>
表1に示すとおり、体毛抜去18日後、19日後共にテストステロン塗布によりL値が上昇し、評価部位の毛周期がより早く退行期に移行していることが示唆された。また、病理組織学的観察の結果、図1に示すとおり、テストステロン塗布群の毛包に退行期特有の退縮が認められることから、毛周期がより早く退行期に移行していることを確認した。
<試験方法>

試験例2:男性ホルモン負荷マウス退行期抑制評価モデルにおける皮膚内TGF-β量の検討
<試験方法>
C57BL/6マウス(8週齢、メス)の背部体毛を抜去し、同調した成長期を誘導した。体毛抜去10日後よりエタノールに溶解した、0.15%或いは0.5%テストステロンを1日1回、1匹当たり0.1mLを測定日の前日まで塗布した。マウスは一群5匹使用した。体毛抜去から17日後にマウスを屠殺した後、背部皮膚を採取して蛋白を抽出し、ELISAによりTGF-β1およびTGF-β2量を測定した。ELISAはPromega社のEmax ImmunoAssay Systemを用いた。
<試験結果>
表2に示すとおり、0.15%以上のテストステロン塗布により、TGF-β1、β2いずれもエタノールの塗布に比べて有意に増加した。TGF-βは、毛包が毛周期上の成長期から退行期へ移行する際に、深く関与していることが報告されている因子であり、テストステロンの塗布により、毛包が通常よりも早く退行期に移行することが示唆された.

C57BL/6マウス(8週齢、メス)の背部体毛を抜去し、同調した成長期を誘導した。体毛抜去10日後より溶媒、5%ミノキシジルローション或いは5%フルタミドローションを1日1回、測定前日まで1匹当たり0.1mL塗布し、その30分後にエタノール或いはエタノールに溶解した0.15%テストステロンを1匹当たり0.1mL塗布した。上記薬剤等の塗布は測定日の前日まで行った。マウスは一群5匹使用した。体毛抜去から17日後にマウスを屠殺した後、評価部位を剃毛し、背部皮膚の明度を色彩色査計(CR-400、コニカミノルタ)にて測定し、L値として数値化した。
<試験結果>

試験例4:男性ホルモン負荷マウス退行期抑制評価試験(2)
<試験方法>
C57BL/6マウス(8週齢、メス)の背部体毛を抜去し、同調した成長期を誘導した。最初の体毛抜去から30日後に再び休止期に入ったマウスの背部体毛を再度抜去し、再び同調した成長期を誘導した。最初の体毛抜去10日後より溶媒或いは5%ミノキシジルローションを1日1回、1匹当たり0.1mL塗布し、その30分後にエタノール或いはエタノールに溶解した0.01%テストステロンを1匹当たり0.1mL塗布した。上記薬剤等の塗布は測定日の前日まで行った。マウスは一群7匹使用した。1回目の体毛抜去から47日後にマウスを屠殺した後、評価部位を剃毛し、背部皮膚の明度を色彩色査計(CR-400、コニカミノルタ)にて測定し、L値として数値化した。
<試験結果>
表4に示すとおり、テストステロン塗布によりL値が上昇し、評価部位の毛周期がより早く退行期に移行していることが示唆された。更に5%ミノキシジルローションの塗布によりテストステロンによるL値の上昇が有意に抑制され、退行期への移行が抑制された。本結果から、5%ミノキシジルローションは継続投与により複数の毛周期に渡って退行期移行抑制作用が持続することが分かった。
<試験方法>
C57BL/6マウス(8週齢、メス)の背部体毛を抜去し、同調した成長期を誘導した。最初の体毛抜去から30日後に再び休止期に入ったマウスの背部体毛を再度抜去し、再び同調した成長期を誘導した。最初の体毛抜去10日後より溶媒或いは5%ミノキシジルローションを1日1回、1匹当たり0.1mL塗布し、その30分後にエタノール或いはエタノールに溶解した0.01%テストステロンを1匹当たり0.1mL塗布した。上記薬剤等の塗布は測定日の前日まで行った。マウスは一群7匹使用した。1回目の体毛抜去から47日後にマウスを屠殺した後、評価部位を剃毛し、背部皮膚の明度を色彩色査計(CR-400、コニカミノルタ)にて測定し、L値として数値化した。
<試験結果>
表4に示すとおり、テストステロン塗布によりL値が上昇し、評価部位の毛周期がより早く退行期に移行していることが示唆された。更に5%ミノキシジルローションの塗布によりテストステロンによるL値の上昇が有意に抑制され、退行期への移行が抑制された。本結果から、5%ミノキシジルローションは継続投与により複数の毛周期に渡って退行期移行抑制作用が持続することが分かった。

本発明により発毛剤、育毛剤等の毛周期における成長期期間の延長作用及び退行期への移行の遅延作用の評価する簡便なin vivo男性型脱毛症モデルを提供することが可能になった。
Claims (6)
- げっ歯類に男性ホルモンを投与する工程を含む、げっ歯類の毛周期における成長期から退行期への移行を促進する方法。
- げっ歯類において、
(a)毛周期の休止期において、同調した成長期を誘導する工程、
(b)男性ホルモンを投与する工程、
(c)薬剤を投与する工程、及び
(d)男性ホルモン投与により促進されるげっ歯類の毛周期における成長期から退行期への移行が薬剤投与により抑制される程度を評価する工程、
を含む薬剤の男性型脱毛症への有効性を評価する方法。 - げっ歯類が有色毛マウスである、請求項1又は2に記載の方法。
- 男性ホルモンが、テストステロン、ジヒドロテストステロン、デヒドロエピアンドロステロン、アンドロステロン、アンドロステンジオン、性腺刺激ホルモン放出ホルモン及びこれらの誘導体から選ばれる1種以上である、請求項1~3のいずれかに記載の方法。
- 毛包の組織学的評価、皮膚明度、皮膚彩度及びTGF-β発現量から選ばれる1種以上を用いて成長期から退行期への移行を評価する、請求項1~4のいずれかに記載の方法。
- (a)~(c)を含む工程を複数回繰り返した後に、(d)男性ホルモン投与により促進されるげっ歯類の毛周期における成長期から退行期への移行が薬剤投与により抑制される程度を評価する工程を行うことを特徴とする、請求項2~5のいずれかに記載の薬剤の男性型脱毛症への有効性を評価する方法。
