実施例2で構築したpUCPgapNP1TmG418を用いて大腸菌を形質転換して、得られた形質転換体を、5mlの2YT培地(1.6% tryptone bacto(Difco社製)、1.0% yeast extract bacto(Difco社製)、0.5% NaCl)で培養し、得られた菌体からQIAprep spin miniprep kit(QIAGEN社製)を用いて、pUCPgapNP1TmG418を取得した。
実施例1で構築したpUC-PmoxLPmoxFdTmを用いて大腸菌を形質転換し、得られた形質転換体を5mlの2YT培地(1.6% tryptone bacto(Difco社製)、1.0% yeast extract bacto(Difco社製)、0.5% NaCl)で培養し、得られた菌体からQIAprep spin miniprep kit(QIAGEN社製)を用いて、pUC-PmoxLPmoxFdTmを取得した。
実施例4で得られた新規ポリペプチド発現/完全ヒト型化抗TNF-α Fab抗体発現形質転換体と完全ヒト型化抗TNF-α Fab抗体発現形質転換体を2mlのBMGY培地(1% yeast extract bacto(Difco社製)、2%polypeptone(日本製薬社製)、0.34% yeast nitrogen base Without Amino Acid and Ammonium Sulfate(Difco社製)、1% Ammonium Sulfate、0.4mg/l Biotin、100mMリン酸カリウム(pH6.0)、2% Glycerol)、また、BMGMY培地(1% yeast extract bacto(Difco社製)、2% polypeptone(日本製薬社製)、0.34% yeast nitrogen base Without Amino Acid and Ammonium Sulfate(Difco社製)、1% Ammonium Sulfate、0.4mg/l Biotin、100mMリン酸カリウム(pH6.0)、1% Glycerol、2% Methanol)に接種し、これを30℃、120rpm、72時間振盪培養後、遠心分離(12,000rpm、5分、4℃)により培養上清を回収した。菌体濃度はOD600にて測定した。
新規ポリペプチド発現/完全ヒト型化抗TNF-α Fab抗体発現形質転換体の培養上清への抗TNF-α Fab抗体分泌発現量は、サンドイッチELISA(Enzyme-Linked Immunosorbent Assay)法により以下に示す方法で行った。ELISA用プレート(マキシソープ;NUNC社製)に固定化バッファー(0.1N重炭酸ナトリウム溶液,pH9.6)にて2500倍希釈したGoat Anti-Human IgG (Fab Specific) Antibody(SIGMA社製)を各ウェル50μlずつ添加し、終夜4℃でインキュベートした。
Claims (9)
- 以下の(a)、(b)または(c)のいずれかのポリペプチド;
(a)配列番号32または33に示すアミノ酸配列からなるポリペプチド;
(b)配列番号32または33に示すアミノ酸配列において、1もしくは複数個のアミノ酸が置換、欠失、挿入及び/又は付加したアミノ酸配列からなるポリペプチド;
(c)配列番号32または33に示すアミノ酸配列と85%以上の配列同一性を有するアミノ酸配列からなるポリペプチド。 - 以下の(d)、(e)、(f)または(g)のいずれかの遺伝子;
(d)配列番号9または34に示す塩基配列からなる遺伝子;
(e)配列番号9または34に示す塩基配列と相補的な塩基配列とストリンジェントな条件下でハイブリダイズする塩基配列からなる遺伝子;
(f)配列番号9または34に示す塩基配列と85%以上の配列同一性を有する塩基配列からなる遺伝子;
(g)配列番号32または33に示すアミノ酸配列からなるポリペプチドをコードする塩基配列からなる遺伝子。 - 請求項2に記載の遺伝子を含有するベクター。
- 請求項3に記載のベクターにより宿主細胞を形質転換して得られる形質転換体。
- 宿主細胞が、酵母、細菌、真菌、昆虫細胞、または動物細胞である、請求項4に記載の形質転換体。
- 酵母がメタノール資化性酵母である、請求項5に記載の形質転換体。
- メタノール資化性酵母がオガタエア属酵母又はコマガタエラ属酵母である、請求項6に記載の形質転換体。
- 請求項4~7のいずれかに記載の形質転換体を培養し、培養物から目的タンパク質を回収する工程を含む、タンパク質の製造方法。
- 培養が、グルコース、及び/又はグリセロール、及び/又はメタノールを炭素源とすることを特徴とする、請求項8に記載のタンパク質の製造方法。
