Abstract
Description
このファミリーに属するJAK3以外のキナーゼは広範な組織で発現しているが、JAK3は免疫細胞に限局して発現している。このことは、JAK3がIL(Interleukin:インターロイキン)-2、IL-4、IL-7、IL-9、IL-15及びIL-21等の多様な受容体を介したシグナル伝達に、共通γ鎖と非共有結合的に会合することにより重要な役割を果たすことと矛盾しない(非特許文献1、2)。
また、X連鎖重症複合免疫不全症(X-linked Severe Combined Immuno Deficiency:XSCID)と呼ばれる免疫不全症の患者集団においては、JAK3タンパクレベルの低下、若しくは共通γ鎖の遺伝子欠損が見られる。このことは、免疫抑制がJAK3を介したシグナル経路を遮断することにより生じていることを示唆する(非特許文献3、4)。動物実験では、JAK3がB-およびT-リンパ球の成熟に重要な役割を果たすだけでなく、T-リンパ球の機能維持にも重要であることが示されている。従って、このメカニズムを通じた免疫応答の制御により、臓器移植時の拒絶や自己免疫疾患のようなT-リンパ球の増殖異常が関与する疾患の治療が見込まれる。
JAK1は、JAK1ノックアウトマウスやJAK1欠損細胞の解析から、IFN(Interferon:インターフェロン)α、IFNβ、IFNγ、IL-2、IL-4、IL-6、IL-7、IL-15等の多様な受容体を介したシグナル伝達に関与していることが示されている(非特許文献5)。従って、これらのシグナル伝達を介した炎症応答の制御により、自己免疫疾患や臓器移植時の急性及び慢性拒絶のようなマクロファージやリンパ球の活性化が関与する疾患の治療が見込まれる。
JAK2は、JAK2ノックアウトマウスやJAK2欠損細胞の解析から、EPO(Erythropoietin:エリスロポエチン)α、トロンボポエチン、IFNγ、IL-3、GM-CSF等の多様な受容体を介したシグナル伝達に関与していることが示されている(非特許文献6、7、8)。これらのシグナル伝達は、骨髄中の赤血球、血小板などの前駆細胞の分化に関与すると考えられる。なお、JAK2の617番塩基のバリンがフェニルアラニンに置換されている場合があり、骨髄増殖性疾患との関与が示唆されている(非特許文献6)。従って、これらのメカニズムを通じた骨髄前駆細胞の分化の制御により、骨髄増殖性疾患の治療が見込まれる。
また、JAK阻害剤であるCP-690,550が臨床試験において関節リウマチ及び乾癬において病態改善効果を示すこと(非特許文献9、10)、サル腎移植モデルにおける拒絶効果及びマウス喘息モデルにおける気道炎症抑制効果を示すことが報告されている(非特許文献11、12)。これらの知見から、JAK阻害剤により免疫活性を抑制することは、臓器移植時の拒絶、移植後の対宿主性移植片反応、自己免疫疾患及びアレルギー性疾患の予防又は治療に有益であると考えられている。CP-690,550以外のJAK阻害作用を有する化合物としては、下記報告などにおいて既知であるが(例えば特許文献1乃至14参照)、さらなる薬剤の開発が望まれている。
なお、本特許出願の優先権主張の基礎となる特許出願の出願後に公開された特許文献において、JAK阻害作用を持つ3環性ピロロピリジン化合物が報告されているが、本発明化合物の具体的な開示はない(特許文献15参照)。

複数の化合物を反応させる場合、それらの当量関係は適宜判断して決定することが出来る。すなわち同じ当量を用いることもできるし、特定の化合物を過剰量用いることもできる。
特に、化合物(1)-6、化合物(1)-7、化合物(1)-11、化合物(2)-2、化合物(2)-3の製造においては、反応を円滑に進行させる上でマイクロウェーブ照射下で反応を行うことが有効な場合がある。
本発明化合物の一般的な製造方法において用いられる酸の例としては、有機酸(例えば、酢酸、トリフルオロ酢酸、p-トルエンスルホン酸等)又は無機酸(例えば、硫酸、塩酸等)が挙げられる。
特に、化合物(1)-11の製造においては、反応を円滑に進行させる上で酸の存在下で反応を行うことが有効な場合がある。
特に、化合物(1)-7、化合物(2)-3の製造においては、反応を円滑に進行させる上でトリフルオロメタンスルホン酸イッテルビウム(III)又はトリフルオロメタンスルホン酸スカンジウム(III)等の酸触媒の存在下で反応を行うことが有効な場合がある。
特に、化合物(1)-6、化合物(2)-2の製造においては、反応を円滑に進行させる上で塩基の存在下で反応を行うことが好ましい場合がある。
以下に示す本発明化合物の一般的製造法の中では、各工程おける中間体及び最終生成物の一般式が示されるが、それらの中間体及び最終生成物の一般式の概念には、保護基で保護された誘導体も含まれる。ここにおいて保護基で保護された誘導体とは、必要に応じて、加水分解、還元、酸化、アルキル化などを行うことにより目的物に誘導可能な化合物のことを意味し、例えば化合物を有機合成化学上許容な保護基で保護したものが含まれる。
保護及び脱保護は、一般的に知られている保護基を用いて、保護・脱保護反応(例えば、プロテクティブ・グループス・イン・オーガニック・シンセシス第4版(Protective Groups in Organic Synthesis, Fourth edition)、グリーン(T.W.Greene)著、ジョン・ワイリー・アンド・サンズ・インコーポレイテッド(John Wiley & Sons Inc.)(2006年)など参照)を行うことにより実施することができる。
加水分解、還元、酸化は、一般的に知られている官能基変換法(例えば、コンプリヘンシブ・オーガニック・トランスフォーメーションズ第2版(Comprehensive Organic Transformations, Second Edition)、ラロック(R.C.Larock)著、ワイリー-ブイシーエイチ(Wiley-VCH)(1999年)など参照)を行うことにより実施することができる。
式(I)で示される化合物のうち化合物(1)-8、(1)-9、(1)-11及び(1)-12は、例えば以下のスキームの製造法により製造することができる。(スキーム中、PG1は水素原子又はTs基、TIPS基若しくはSEM基等の保護基を示し、その他の記号は前記定義に同じである。)
化合物(1)-3は、化合物(1)-2と過マンガン酸カリウム又は亜塩素酸ナトリウム等の酸化剤を用い、室温から還流温度で合成することができる。
化合物(1)-4は、化合物(1)-3とチオニルクロリド又はオキシ塩化リンを用い、0℃から還流温度で酸クロリドとした後、アンモニア-メタノール又はその等価体を用いて、0℃から還流温度で合成することができる。
化合物(1)-6は、化合物(1)-4とアミン誘導体(1)-5を用い、室温から還流温度で合成することができる。また、ブッフバルト・ハートウィグ反応で用いられる反応条件(例えば、アドバンスド・シンセシス・アンド・キャタリシス、第346巻、1599―1626ページ、2004年(Advanced Synthesis & Catalysis,2000,346,pp.1599―1626)など参照)を行うことにより実施することもできる。同反応に用いる金属種とリガンドとしては、特に限定されないが、トリス(ジベンジリデンアセトン)ジパラジウム(0)、テトラキス(トリフェニルホスフィン)パラジウム(0)、酢酸パラジウム(II)等と4,5-ビス(ジフェニルホスフィノ)-9,9-ジメチルキサンテン(Xantphos)、2-ジシクロヘキシルホスフィノ-2’,6’-ジメトキシビフェニル(SPhos)、2-ジシクロヘキシルホスフィノ-2’,4’,6’-トリイソプロピルビフェニル(XPhos)等を適宜組み合わせて用いることができる。
化合物(1)-7は、化合物(1)-6とR2CO2RQ又はR2C(ORQ)3等を用い、室温から還流温度で合成することができる(RQは水素原子又はC1-6アルキル基を示す。)。
化合物(1)-8は、化合物(1)-7と水素化ホウ素ナトリウム等の還元剤を用いて、0℃から還流温度で合成することができる。
化合物(1)-11は、化合物(1)-6とカルボニル化合物(1)-10を用い、室温から還流温度で合成することができる。
化合物(1)-9及び(1)-12は、化合物(1)-8又は(1)-11とR1-RL(RLはハロゲン原子、メタンスルホニルオキシ基、p-トルエンスルホニルオキシ基等の脱離基を意味し、R1-RLは、例えば、アルキルハライド、メタンスルホン酸エステル又は酸ハライド等が挙げられる。)で表される求電子剤及び炭酸カリウム又は水酸化ナトリウム等の塩基を用い、-78℃から還流温度で合成することができる。
化合物(1)-8、(1)-9、(1)-11及び(1)-12において、PG1が保護基の場合は、さらに脱保護反応を行うことによりPG1が水素原子である化合物を得ることができる。
化合物(2)-3は、化合物(2)-2の水酸基をPG2で保護した後、オルトギ酸メチル又はその等価体を用いて、室温から還流温度で合成することができる。
化合物(2)-4は、化合物(2)-3と水素化ホウ素ナトリウム等の還元剤を用いて、0℃から還流温度で合成することができる。
化合物(2)-5は、化合物(2)-4とR1-RL(RLはハロゲン原子、メタンスルホニルオキシ基、p-トルエンスルホニルオキシ基等の脱離基を意味し、R1-RLは、例えば、アルキルハライド、メタンスルホン酸エステル又は酸ハライド等が挙げられる。)で表される求電子剤及び炭酸カリウム又は水酸化ナトリウム等の塩基を用い、-78℃から還流温度でアルキル化した後、PG2を脱保護することで合成することができる。
化合物(2)-6は、化合物(2)-5と2-ヨードキシ安息香酸又はクロロクロム酸ピリジニウム等の酸化剤を用い、-78℃から還流温度で合成することができる。
化合物(2)-8は、化合物(2)-6と化合物(2)-7及び2-ピコリンボラン又はトリアセトキシ水素化ホウ素ナトリウム等の還元剤を用い、室温から還流温度で合成することができる。
化合物(2)-10は、Raが水素原子である化合物(2)-8とスルホニルクロリド(2)-9を用い、炭酸カリウム又はトリエチルアミン等の塩基を用い、-78℃から還流温度で合成することができる。
化合物(2)-11は、化合物(2)-5とRe-RL(RLはハロゲン原子、メタンスルホニルオキシ基、p-トルエンスルホニルオキシ基等の脱離基を意味し、Re-RLは、例えば、アルキルハライド、メタンスルホン酸エステル又は酸ハライド等が挙げられる。)、α、β不飽和ニトリル化合物又はα、β不飽和スルホニル化合物等の求電子剤及び炭酸カリウム又は水酸化ナトリウム等の塩基を用い、-78℃から還流温度で合成することができる。
化合物(2)-5、(2)-8及び(2)-10において、PG1が保護基の場合は、さらに脱保護反応を行うことによりPG1が水素原子である化合物を得ることができる。
本発明の式(I)で示される化合物は、必要に応じて医薬的に許容され得る塩に変換することも、又は生成した塩から遊離させることもできる。本発明の医薬的に許容され得る塩としては、例えば、
アルカリ金属(リチウム、ナトリウム、カリウムなど)、
アルカリ土類金属(マグネシウム、カルシウムなど)、
アンモニウム、
有機塩基、
アミノ酸、
無機酸(塩酸、臭化水素酸、リン酸、硫酸など)、又は、
有機酸(酢酸、クエン酸、マレイン酸、フマル酸、酒石酸、ベンゼンスルホン酸、メタンスルホン酸、p-トルエンスルホン酸など)との塩が挙げられる。
プロドラッグとは、化学的又は代謝的に分解できる基を有する医薬品化合物の誘導体であり、加溶媒分解により又は生理的条件下のインビボ(in vivo)において分解され薬理学的に活性な医薬品化合物へと誘導される化合物である。適当なプロドラッグ誘導体を選択する方法および製造する方法は、例えば デザイン オブ プロドラッグス(エルゼビア、アムステルダム 1985)(Design of Prodrugs (Elsevier, Amsterdam 1985))に記載されている。本発明の場合、水酸基を有する場合は、その化合物と適当なアシルハライド、適当な酸無水物又は適当なハロアルコキシカルボニル化合物とを反応させることによって製造されるアシルオキシ誘導体のようなプロドラッグが例示される。プロドラッグとして特に好ましい構造としては-O-COC2H5、-O-CO(t-Bu)、-O-COC15H31、-O-CO(m-CO2Na-Ph)、-O-COCH2CH2CO2Na、-OCOCH(NH2)CH3、-O-COCH2N(CH3)2又は-O-CH2OC(=O)CH3などがあげられる。本発明を形成する化合物が-NH-基を有する場合は、-NH-基を有する化合物と適当な酸ハロゲン化物、適当な混合酸無水物又は適当なハロアルコキシカルボニル化合物とを反応させることにより製造されるプロドラッグが例示される。プロドラッグとして特に好ましい構造としては、-N-CO(CH2)20OCH3、-N-COCH(NH2)CH3、-N-CH2O(C=O)CH3等が挙げられる。
以上に示す疾患の治療および予防を行う場面が想定されるが、これらに限定されることはない。

シリカゲル薄層クロマトグラフィーでの精製は、特に記述がない場合は、メルク製PLCプレートを用いた。

本発明化合物の薬理学的アッセイを以下に示す。
本発明化合物の、JAKに対する阻害活性を測定した。
各酵素(JAK1、JAK2、JAK3、Tyk2)はカルナバイオサイエンス社から購入した。
酵素の基質(以下、基質)はLANCE Ultra ULight―JAK―1(Tyr1023) Peptide(パーキンエルマー社)を使用した。
基質のリン酸化の検出抗体は、LANCE Ultra Europium-anti-phospho tyrosine antibody (PT66) (パーキンエルマー社)を使用した。
その他の試薬はそれぞれ以下から購入した。
アデノシン三リン酸(ATP):シグマ社
4-(2-ヒドロキシエチル)-1-ピペラジンエタンスルホン酸(HEPES): ドウジンドウ社
グリコールエーテルジアミン四酢酸(EGTA):ドウジンドウ社
塩化マグネシウム(MgCl2):和光純薬社
ジチオトレイトール(DTT):和光純薬社
Tween20:シグマ社
エチレンジアミン四酢酸(EDTA):ドウジンドウ社
アッセイバッファーは以下の組成のものを用いた。
HEPES(pH7.5):50mM
EGTA:1mM
MgCl2:10mM
DTT:2mM
Tween20 :0.01%(重量/重量)
化合物は、1μM、0.1μM、0.01μM、0.001μM、0.0001μM、0.00001μMの6濃度とした。
酵素濃度、ATP濃度は各酵素(JAK1、JAK2、JAK3、Tyk2)試験で、
JAK1酵素試験では酵素濃度は0.5μg/mL、ATP濃度は70μM、
JAK2酵素試験では酵素濃度は0.013μg/mL、ATP濃度は10μM、
JAK3酵素試験では酵素濃度は0.025μg/mL、ATP濃度は3μM、
Tyk2酵素試験では酵素濃度は0.25μg/mL、ATP濃度は20μM、とした。
酵素の基質は6nMとした。
EDTAは11mMとした。
PT66は2nM~3nMとした。
その後、基質希釈液、ついでATPを添加し、室温で30分インキュベーションした。
その後、EDTA希釈液、ついでPT66希釈液を添加し、室温で1時間インキュベーションした。
ARVO―HTSにより蛍光を測定し、化合物濃度の対数値と阻害活性をプロットすることでIC50値を得た。表27及び28に、合成例化合物のJAK1、JAK2、JAK3及びTyk2酵素アッセイの結果を示す。なお表中の「*」は、IC50>1μMを表す。
本発明化合物である合成例化合物1~67の、JAKを経由するサイトカインシグナルに対する阻害活性を、ラット全血を用いたSTATリン酸化アッセイにて測定した。
Lewisラット雌は日本チャールス・リバー社より購入した。
IL-6はペプロテック社より購入した。
FITC(fluorescein isothiocyanate)蛍光標識された抗CD3抗体(FITC-CD3)はイ-バイオサイエンス社より購入した。
BD Phosflow Lyse/Fix Buffer、
BD Phosflow Perm Buffer III、
BD Pharmingen Stain Buffer、
BD Phosflow STAT-1(pY701)PE(R-Phycoerythrin)蛍光標識(以下、BD Phosflow STAT-1)
はBD(ベクトンディキンソン)社より購入した。
希釈の濃度、および後述するチューブ中への添加量は、チューブ中で、それぞれ以下の終濃度に成るように調製した。
化合物は、1μM、0.1μM、0.01μMの3濃度または10μM、1μM、0.1μMの3濃度とした。
IL-6は100ng/mLとした。
FITC-CD3は1μg/mLとした。
また、本特許出願の優先権主張の基礎となる特許出願の出願後に公開されたWO2013/024895に開示の化合物A(Compound A;実施例b53)、化合物B(Compound B;実施例b138)、化合物C(Compound C;実施例b137)、化合物D(Compound D;実施例b122)及び化合物E(Compound E;実施例b127)の、ラット全血シグナルアッセイの結果を以下に示す。
JAKシグナルを経由する細胞増殖に対する、本発明化合物の阻害活性は、ヒト赤白血病細胞株TF-1を用いて測定可能である。
TF-1細胞(ATCC(American Type Culture Collection))は、5%ウシ胎仔血清(FBS:Fetal Bovine Serum)、1ng/mL GM―CSF(Granulocyte Macrophage Colony―Stimulating Factor)添加RPMI1640培地でCO2インキュベータ(5体積%CO2、37℃)にて増殖維持する。試験時にPBSで洗浄したTF-1細胞を、5%FBS添加RPMI1640培地に懸濁後、96ウェル培養プレートに1ウェルあたり1×104個を播種する。その培養プレートの各ウェルに化合物を添加し、37℃にて30分間インキュベーション後に、IL-4又はIL-6などのサイトカインを、それぞれ添加する。その後、培養プレートは、CO2インキュベータ(5体積%CO2、37℃)で3日間インキュベーションする。
細胞増殖度はWST-8試薬(キシダ化学社)を用い、取扱説明書に従いアッセイ可能である。WST-8試薬を培養プレートの各ウェルに添加後、CO2インキュベータ(5体積%CO2、37℃)で4時間インキュベーションする。発色したホルマザン色素は、マイクロプレートリーダーを用いて450nmの吸光度を測定することで検出可能である。化合物濃度の対数値と抑制活性をプロットすることで、IC50値を算出することができる。
JAK阻害作用が有効である疾患において、化合物が経口吸収性を有することは、疾患治療において好ましく、本発明化合物の経口吸収性は、ラットを用いて測定可能である。
化合物を0.6mg/mLの濃度になるよう、0.5%メチルセルロース液に懸濁する。その懸濁液を3mg/kg/5mLの用量で、Lewisラット雌(日本チャールス・リバー社)に、ゾンデを用いて強制経口投与する。その後、ヘパリンを抗凝血薬として用い、化合物投与後(0.5~8時間後)、経時的にラット頚静脈より採血する。得られた血液を遠心分離機により17652m/s2で10分間遠心分離し、血漿を得る。得られた血漿を、液体クロマトグラフィータンデム質量分析装置(LC/MS/MS:ウォーターズ社)を用いて分析することで、経口投与後(0.5~8時間後)における、化合物の血漿中濃度推移を算出することができる。
JAK阻害作用が有効である疾患のうち、特に関節リウマチでの治療効果を実験動物モデルで確認するには、ラットを用いたコラーゲン誘発関節炎モデル(プロスタグランジン アンド アザー リピッド メディエータース、第66巻、317-327ページ、2001年(Prostaglandin & other Lipid Mediators,2001,66,pp.317-327))を用いることができる。
ウシII型コラーゲン溶液(コンドレックス社)と不完全フロイントアジュバント(ディフコ社)を等量混合し、エマルジョン化して免疫溶液を作製する。次に、Lewisラット雌(日本チャールス・リバー社)の背部4箇所および尾の付け根部1箇所へ、ハミルトンシリンジを用いて免疫溶液を100μL/箇所で皮内投与する。免疫溶液投与7日後に、再度免疫溶液を同様に皮内投与する。
投与する化合物は、上記2.全血シグナルアッセイで得られたサイトカインシグナル阻害のIC50値と、4.経口吸収性で得られた化合物の血漿中濃度より、適宜決めた濃度になるよう、0.5%メチルセルロース液に懸濁する。この化合物懸濁液を、2回目の免疫後から連日経口投与する。
2回目の免疫後2~3週間で腫れた後肢甲部の厚さをノギスにて測定することで、化合物による関節炎抑制度合いを算出することができる。
製剤例1

式(I)で表される化合物と乳糖を60メッシュのふるいに通す。コーンスターチを120メッシュのふるいに通す。これらをV型混合機にて混合する。混合末に低粘度ヒドロキシプロピルセルロース(HPC-L)水溶液を添加し、練合、造粒(押し出し造粒 孔径0.5~1mm)した後、乾燥する。得られた乾燥顆粒を振動ふるい(12/60メッシュ)で篩過し顆粒剤を得る。
以下の成分を含有するカプセル充填用散剤を製造する。

静脈用製剤は次のように製造する。

- 請求項11に記載のJAK阻害剤を有効成分として含有する関節リウマチ治療薬。
- 請求項1乃至10の何れか一項に記載の化合物、該化合物の互変異性体若しくはその医薬的に許容され得る塩又はそれらの溶媒和物を有効成分として含有する医薬。
