用語「ハロゲノアルキル」、「ハロゲノアルコキシ」および「ハロゲノシクロアルキル」とは、それぞれ、1~7個のハロゲン原子で置換された上記アルキル、アルコキシおよびシクロアルキルを意味する。
用語「フルオロアルキル」、「フルオロアルコキシ」および「フルオロシクロアルキル」とは、それぞれ、1~7個のフッ素原子で置換された上記アルキル、アルコキシおよびシクロアルキルを意味する。
用語「単環式の非芳香族複素環基」とは、炭素原子以外に、酸素原子、硫黄原子および窒素原子から選ばれるヘテロ原子を1~4個含む4~7員の単環式の非芳香族複素環基を意味し、例えば、ピロリジニル、ピペリジニル、テトラヒドロフリル、テトラヒドロチエニル、およびモルホリルが挙げられる。

本発明の化合物は、好ましくは慢性疼痛または泌尿器疾患の予防または治療に有用であり、とりわけ慢性疼痛に有用である。
本発明の化合物のいずれの製造プロセスの間においても、関与する分子における感受性基または反応性基を保護することが、要求および/または所望される。これは、慣用の保護基を用いて達成し得る。保護基およびそれらの使用の一般的な記述については、T.W.Greeneら、"Protecting Groups in Organic Synthesis", John Wiley & Sons, New York, 2006を参照されたい。保護基は、当業者に慣用の方法を用いて、その後の工程で除去し得る。

式(1-b)で表される化合物を加水分解することで、式(2)で表される化合物を得る。この化合物またはその塩を酸塩化物化し、化合物(3)を得る。これをテトラゾリノニル化することにより、目的の式(I-b)で表される化合物を製造することができる。
PGがアルキルである化合物(1-b)の加水分解反応は、溶媒中、化合物(1-b)を塩基および水で処理することで行うことができる。
塩基としては、例えば、水酸化リチウム、水酸化ナトリウム、水酸化カリウム等の水酸化アルカリ金属;ナトリウムメトキシド、ナトリウムエトキシド等のナトリウムアルコキシドが挙げられる。溶媒としては、反応に影響を与えないものであればよく、例えば、テトラヒドロフラン、1,4-ジオキサン等のエーテル;メタノール、エタノール等のアルキルアルコール;水、またはこれらの混合溶媒が挙げられる。反応は、0℃~100℃で好適に進行する。
化合物(2)またはその塩の酸塩化物化反応は、溶媒中、触媒を伴ってまたは伴わずに、酸塩化物化剤の存在下で行うことができる。
酸塩化物化剤としては、例えば、塩化オキサリル、塩化チオニルが挙げられる。触媒としては、例えば、N,N-ジメチルホルムアミドが挙げられる。溶媒としては、反応に影響を与えないものであればよく、例えば、テトラヒドロフラン、1,4-ジオキサン、1,2-ジメトキシエタン等のエーテル;ジクロロメタン、クロロホルムおよび1,2-ジクロロエタン等のハロゲノ炭化水素;もしくは上述の塩化チオニル;またはこれらの混合溶媒が挙げられる。反応は、20℃~80℃で好適に進行する。
化合物(3)のテトラゾリノニル化反応は、溶媒中または無溶媒で、アジ化物の存在下で行うことができる。
アジ化物としては、例えば、トリメチルシリルアジドが挙げられる。溶媒としては、反応に影響を与えないものであればよい。好ましくは、無溶媒である。反応は、60℃~120℃で好適に進行する。

スキームIII:
式(1-b)で表される化合物を加水分解することで、式(2)で表される化合物を得る。この化合物またはその塩を酸塩化物化し、化合物(3)を得る。これをアミド化することで、式(4)で表される化合物を得る。これを化合物(5)およびヒドラジンと反応させることにより、目的の式(I-c)で表される化合物を製造することができる。
PGがアルキルである化合物(1-b)の加水分解反応は、前記スキームIIの化合物(1-b)の加水分解反応と同様にして実施することができる。
化合物(2)またはその塩の酸塩化物化反応は、前記スキームIIの化合物(2)またはその塩の酸塩化物化反応と同様にして実施することができる。
化合物(3)のアミド化反応は、溶媒中、化合物(3)をアンモニアで処理することで行うことができる。
溶媒としては、反応に影響を与えないものであればよく、例えば、テトラヒドロフラン、1,4-ジオキサン、1,2-ジメトキシエタン等のエーテル;ジクロロメタン、クロロホルムおよび1,2-ジクロロエタン等のハロゲノ炭化水素;またはこれらの混合溶媒が挙げられる。反応は、0℃~50℃で好適に進行する。
化合物(I-c)を得る反応は、化合物(4)と化合物(5)を、溶媒中または無溶媒で縮合反応し、次いで、溶媒中、ヒドラジン一水和物で処理することで行うことができる。
化合物(4)と化合物(5)の縮合反応の溶媒としては、反応に影響を与えないものであればよい。好ましくは、無溶媒である。反応は、80℃~150℃で好適に進行する。
続くヒドラジン一水和物の処理の溶媒としては、反応に影響を与えないものであればよく、例えば、酢酸が挙げられる。反応は、60℃~120℃で好適に進行する。
式(1-a)で表される化合物を加水分解することで、式(4)で表される化合物を得る。これを化合物(5)およびヒドラジンと反応させることにより、目的の式(I-c)で表される化合物を製造することができる。
化合物(1-a)の加水分解反応は、溶媒中、化合物(1-a)を塩基で処理することで行うことができる。
塩基としては、例えば、水酸化ナトリウム、水酸化カリウム等の水酸化アルカリ金属;ナトリウムt-ブトキシド、カリウムt-ブトキシド等のアルカリ金属t-ブトキシドが挙げられる。溶媒としては、反応に影響を与えないものであればよく、例えば、テトラヒドロフラン、1,2-ジメトキシエタン、1,4-ジオキサン等のエーテル;t-ブタノール等のアルコール;またはこれらの混合溶媒が挙げられる。反応は、20℃~100℃で好適に進行する。
あるいは、化合物(1-a)の加水分解反応は、無溶媒で、化合物(1-a)を酸で処理することで行うことができる。
酸としては、例えば、硫酸が挙げられる。反応は、0℃~100℃で好適に進行する。
化合物(I-c)を得る反応は、前記スキームIIIの化合物(I-c)を得る反応と同様にして実施することができる。

スキームV:
式(1-a)で表される化合物をアミドオキシム化することで、式(6)で表される化合物を得る。これをオキサジアゾロニル化することにより、目的の式(I-d)で表される化合物を製造することができる。
化合物(1-a)のアミドオキシム化反応は、溶媒中、ヒドロキシアミン塩酸塩および塩基の存在下で実施することができる。
塩基としては、例えば、トリエチルアミン、ジイソプロピルエチルアミン、ピリジン、N-メチルモルホリン等のアミンが挙げられる。溶媒としては、反応に影響を与えないものであればよく、例えば、メタノール、エタノール等のアルコールが挙げられる。反応は、20℃~80℃で好適に進行する。
化合物(6)から化合物(I-d)を得る反応は、溶媒中、塩基の存在下で、化合物(6)およびクロロギ酸エステルを縮合反応し、次いで、溶媒中、得られた化合物を環化反応することで行うことができる。
縮合反応のクロロギ酸エステルとしては、例えば、クロロギ酸2-エチルヘキシルが挙げられる。塩基としては、例えば、トリエチルアミン、ジイソプロピルエチルアミン、ピリジン、N-メチルモルホリン等のアミンが挙げられる。溶媒としては、反応に影響を与えないものであればよく、例えば、N,N-ジメチルホルムアミド、N,N-ジメチルアセトアミド、1,3-ジメチル-2-イミダゾリジノンおよびN-メチルピロリドン等のアミドが挙げられる。反応は、80℃~150℃で好適に進行する。
環化反応の溶媒としては、反応に影響を与えないものであればよく、例えば、N,N-ジメチルホルムアミド、N,N-ジメチルアセトアミド、1,3-ジメチル-2-イミダゾリジノンおよびN-メチルピロリドン等のアミドが挙げられる。反応は、100℃~180℃で好適に進行する。
カルボニル源としては、例えば、カルボニルジイミダゾール、トリホスゲンが挙げられる。塩基としては、例えば、トリエチルアミン、ジイソプロピルエチルアミン、ピリジン、N-メチルモルホリン、1,8-ジアザビシクロ[5.4.0]ウンデカ-7-エン等のアミン;酢酸ナトリウム、酢酸カリウム等のC2-C7脂肪酸アルカリ金属塩が挙げられ、好ましくは、トリエチルアミン、1,8-ジアザビシクロ[5.4.0]ウンデカ-7-エン等のアミンである。溶媒としては、反応に影響を与えないものであればよく、例えば、テトラヒドロフラン、1,4-ジオキサン、1,2-ジメトキシエタン等のエーテルが挙げられる。反応は、0℃~60℃で好適に進行する。

化合物(1-b)を還元することで、化合物(7)を得る。これをアミノ化することで、化合物(8)を得る。これをアルカノイル化またはアルキルスルホニル化することにより、目的の式(I-e)で表される化合物を製造することができる。
化合物(1-b)の還元反応は、溶媒中、還元剤の存在下で実施することができる。
還元剤としては、例えば、水素化ホウ素リチウム、水素化ホウ素ナトリウム、水素化アルミニウムリチウム、水素化ジイソブチルアルミニウムが挙げられ、好ましくは、水素化ジイソブチルアルミニウムである。溶媒としては、反応に影響を与えないものであればよく、例えば、トルエン、キシレン等の芳香族炭化水素;ジクロロメタン、クロロホルム等のハロゲノ炭化水素;テトラヒドロフラン、ジエチルエーテル等のエーテル、メタノール、エタノール等のアルコール;またはこれらの混合溶媒が挙げられる。反応は、-80℃~120℃で好適に進行する。
化合物(7)のアミノ化反応は、化合物(7)とアジ化物を反応させ、アジドを得て、これを還元することで行うことができる。
化合物(7)とアジ化物の反応は、溶媒(例えば、テトラヒドロフラン等のエーテル)中、塩基(例えば、1,8-ジアザビシクロ[5.4.0]ウンデカ-7-エン等のアミン)の存在下で行うことができる。反応は、0℃~50℃で好適に進行する。
アジドの還元反応は、溶媒(例えば、テトラヒドロフラン等のエーテル)中、還元剤(例えば、トリフェニルホスフィン等のホスフィン)および水の存在下で行うことができる。反応は、20℃~100℃で好適に進行する。
化合物(8)と化合物(9)の反応は、溶媒(例えば、ジクロロメタン等のハロゲノ炭化水素)中、塩基(例えば、トリエチルアミン等のアミン)の存在下で行うことができる。反応は、-80℃~20℃で好適に進行する。

スキームVII:
式(1-b’)で表される化合物をヒドラジンまたはヒドロキシアミンと反応させることにより、目的の式(I-f)で表される化合物を製造することができる。
V2がNHである式(I-f)で表される化合物は、溶媒(例えば、メタノール、エタノール、2-プロパノール等のアルコール)中、PGがアルキルである化合物(1-b’)をヒドラジンと処理することにより製造することができる。反応は、50℃~120℃で好適に進行する。
V2がOである式(I-f)で表される化合物は、溶媒(例えば、テトラヒドロフラン等のエーテル、エタノール等のアルコール、水、またはこれらの混合溶媒)中、PGがアルキルである化合物(1-b’)を加水分解し、得られたカルボン酸化合物を当業者に周知の慣用の方法でヒドロキシアミンと縮合反応し、得られたヒドロキサミド化合物を、溶媒中、塩基(例えば、水素化ナトリウム等の水素化アルカリ金属、水酸化ナトリウム等の水酸化アルカリ金属)で処理することにより製造することができる。

なお、当該反応において、1当量の化合物(12)に対し2当量の化合物(13)が縮合したスルホンイミド化合物が得られた場合は、該スルホンイミド化合物をテトラブチルアンモニウムフルオリドで処理することにより、対応する化合物(10)を得ることができる。
化合物(10)と化合物(11)の反応は、溶媒中、塩基の存在下で実施することができる。

R6が置換されていてもよいジアルキルアミノもしくは置換されていてもよい含窒素非芳香族複素環基である化合物(22)およびその前駆体化合物は、それぞれ対応するR6がハロゲン(とりわけフッ素)である化合物(22)およびその前駆体化合物をE5E6NH[式中、E5は置換されていてもよいアルキルを意味し、E6は置換されていてもよいアルキルを意味するか、あるいは、E5およびE6が互いにそれらの末端で結合して隣接する窒素原子と置換されていてもよい含窒素非芳香族複素環基を形成する。]と反応させることにより製造することができる。
R6が置換されていてもよいアルキルアミノである化合物(22)およびその前駆体化合物は、それぞれ対応するR6がハロゲン(とりわけ臭素、ヨウ素)である化合物(22)およびその前駆体化合物をトリメチルシリルアジドと反応させ、アミノ化合物を得て、これをE7LG10[式中、E7は置換されていてもよいアルキルを意味し、LG10は脱離基を意味し、例えば、ヨウ素等のハロゲンが挙げられる。]と反応させることにより製造することができる。
アミノ化合物とE7LG10の反応は、溶媒(例えば、N,N-ジメチルホルムアミド、N,N-ジメチルアセトアミド、1,3-ジメチル-2-イミダゾリジノン、N-メチルピロリドン等のアミド)中、塩基(例えば、水素化ナトリウム、水素化カリウム等の水素化アルカリ金属)の存在下で実施することができる。反応は、0℃~60℃好適に進行する。

(1)4-({(4-ヨードイソキノリン-3-イル)[4-(トリフルオロメトキシ)ベンジル]アミノ}スルホニル)安息香酸エチルの合成
得られた固体を酢酸(55mL)およびN,N-ジメチルホルムアミド(330mL)に懸濁し、50℃に加熱した。反応混合物にN-ヨードコハク酸イミド(37.2g、0.165mol)を加え、同温で15分撹拌した。室温まで冷却し、反応混合物に酢酸エチルおよび水を加え、重曹水を加えて中和した。生成した沈殿物をろ取し、水で洗浄し、減圧下乾燥することで白色固体(43.7g)を得た。ろ液の有機層を分離し、水および飽和食塩水で洗浄し、硫酸ナトリウムで乾燥し、ろ過し、ろ液を減圧下濃縮した。残渣に酢酸エチルを加え、生成した固体をろ取し減圧下乾燥することで、淡褐色固体(25.7g)を得た。
得られた固体を合わせ、N,N-ジメチルホルムアミド(590mL)に溶解し、4-トリフルオロメトキシベンジルブロミド(44.0g、0.173mol)および炭酸カリウム(29.8g、0.216mol)を加え、50℃で3時間撹拌した。室温まで冷却後、反応混合物に酢酸エチルを加え、水および飽和食塩水で洗浄し、硫酸ナトリウムで乾燥し、ろ過し、ろ液を減圧下濃縮した。残渣にジイソプロピルエーテルを加え、生成した固体をろ取し減圧下乾燥することで、4-({(4-ヨードイソキノリン-3-イル)[4-(トリフルオロメトキシ)ベンジル]アミノ}スルホニル)安息香酸エチル(90.8g、78%)を黄色固体として得た。

3,5-ジメチル-4-ブロモアニリン(500mg,2.499mmol)の濃塩酸(2.5mL)および水(0.5mL)の混合物に、-5℃で、亜硝酸ナトリウム(193mg,2.749mmol)の水(1.3mL)溶液を滴下した。-5℃で30分撹拌した後、(1-A)で調製した溶液を、同温で滴下した。反応温度を徐々に上げ、反応混合物を室温で終夜撹拌した。反応混合物をクロロホルムにて3回抽出し、有機層を合わせ、乾燥し、減圧下濃縮し4-ブロモ-3,5-ジメチルベンゼンスルホニルクロリドの粗生成物(375mg)を得た。これをさらに精製することなく次の工程に使用した。

対応原料化合物を参考例23と同様に処理することにより下記第16表記載の化合物を得た。
1.TRPM8阻害試験
試験化合物:
上記実施例に記載の化合物を、TRPM8阻害試験に用いた。
方法:
試験化合物の機能活性はカルシウム感受性蛍光色素を用いて細胞内カルシウム濃度変化を測定することによって評価した。蛍光シグナルの変化は、浜松フォトニクスのFunctional Drug Screening System (FDSS)による細胞イメージング技術を用いて測定した。細胞内カルシウム濃度の上昇はメントールによって活性化させることで検出可能であった。
ヒト型TRPM8を安定発現させたHEK293細胞をフラスコで培養した。アッセイ当日、フラスコから培地を除き、細胞をリン酸緩衝食塩液(phosphate-buffered saline: PBS)で洗浄後、2 mmol/L エチレンジアミン四酢酸二ナトリウム塩(EDTA・2Na)含有PBSで回収した。次いで細胞を、3 μmol/L Fura-2AMおよび0.01% Pluronic F-127を含む測定溶液で60分間インキュベートした。引き続き、ウェルあたり20,000-50,000個の懸濁細胞を各種濃度の試験化合物と各ウェル内にて20分間37℃でインキュベートした。100 μmol/L メントールによって惹起される細胞内カルシウム濃度変化は、FDSSを用いて2分間測定した。50%阻害濃度(IC50値)は4点での濃度反応試験から算出した。濃度反応曲線は各データポイントあたり4ウェルの平均値を用いて作成した。
結果:
各検体のIC50値を下記第17表に示す。
試験化合物:
上記実施例に記載の化合物を、ラットにおけるTRPM8拮抗作用試験に用いた。
方法:
試験化合物のin vivo拮抗活性をラットwet-dog shakes (WDS)モデルにおいて評価した。ラットは、TRPM8作動薬であるメントールに応答して身震い行動(shaking behavior)を示す。メントール投与の前にTRPM8遮断薬でラットを前処置すると、観測される身震い行動が抑制される。
Sprague Dawley (SD)系雄性ラットにおけるTRPM8遮断薬のメントール誘発性身震い行動に対する抑制活性を評価するために、メントール負荷(50 mg/kg, 腹腔内投与, 10% マクロゴール15ヒドロキシステアレート/生理食塩水溶液)の1、2、または4時間前に試験化合物(3 mg/kg, 経口投与, 0.5% メチルセルロース溶液; N = 3-4/群)を投与した。メントール投与後、自発的WDSの回数を5分間計測した。ビヒクル前処置と比較した自発的WDS行動の抑制を抑制率(%)として表し、以下のように計算する。
阻害率(%) = [1-(試験化合物投与群におけるWDS回数/ビヒクル投与群におけるWDS回数)] x100。

- 一般式(I):
R2は、水素原子、または置換されていてもよいシクロアルキルであり、
R3は、置換されていてもよいアルキル、または置換されていてもよいアルコキシであり、
Zは、CH、またはNであり、
環Aは、下式(i)、(ii)、または(ix):
R4は、置換されていてもよいアルキルであり、
R6は、置換されていてもよいアルキル、置換されていてもよいシクロアルキル、置換されていてもよいアルコキシ、置換されていてもよいアルキルアミノ、置換されていてもよいジアルキルアミノ、置換されていてもよい単環式の含窒素非芳香族複素環基、置換されていてもよいフェニル、またはハロゲンであり、
R7は、水素原子、置換されていてもよいアルキル、またはハロゲンであり、
X1およびX2は、それぞれ独立して、テトラゾリル、テトラゾリノニル、置換されていてもよいトリアゾリル、トリアゾリノニル、オキサジアゾロニル、置換されていてもよいアルカノイルアミノメチル、または置換されていてもよいアルキルスルホニルアミノメチルであるか、あるいは、
R4およびX2が互いにそれらの末端で結合して隣接するベンゼンとインダゾリノニル、またはベンゾイソオキサゾロニルを形成し、
Y1およびY2は、共にCHであるか、あるいは、Y1およびY2の一方がCH、他方がNである。]
で表される化合物またはその薬理的に許容し得る塩。 - R1が、C1-C6アルキル、C1-C6ハロゲノアルキル、またはC3-C7シクロアルキルであり、
R2が、水素原子、またはC3-C7シクロアルキルである請求項1記載の化合物またはその薬理的に許容し得る塩。 - R3が、C1-C6ハロゲノアルキル、またはC1-C6ハロゲノアルコキシである請求項1または2記載の化合物またはその薬理的に許容し得る塩。
- 環Aが、式(i)、または式(ii)であり、
R4が、C1-C6アルキルであり、
X1およびX2が、それぞれ独立して、テトラゾリル、テトラゾリノニル、トリアゾリル、C1-C6アルキルトリアゾリル、C1-C6ハロゲノアルキルトリアゾリル、トリアゾリノニル、オキサジアゾロニル、C2-C7アルカノイルアミノメチル、C1-C6アルキルスルホニルアミノメチル、またはC1-C6ハロゲノアルキルスルホニルアミノメチルであるか、あるいは、
R4およびX2が互いにそれらの末端で結合して隣接するベンゼンとインダゾリノニル、またはベンゾイソオキサゾロニルを形成する請求項1~3いずれか1項記載の化合物またはその薬理的に許容し得る塩。 - 環Aが、式(i)、または式(ii)であり、
R4が、C1-C6アルキルであり、
