Description

質量分析装置
 本発明は、基板表面に接触された試料を、該基板表面から脱離させ、イオン化した、試料中の測定対象物質を捕捉して質量分析する質量分析装置に関するものである。
 物質の同定等に用いられる分析法において、基板の表面に付着した測定対象物質を基板表面から脱離させ、イオン化させてその物質の質量と荷電の比によって物質を同定する、質量分析方法(Mass Spectrometry)が知られている。例えば、飛行時間型質量分析法(Time of Flight Mass Spectroscopy : TOF-MS)は、イオン化した測定対象物質を高電圧電極間で所定距離飛行させて、その飛行時間により物質の質量を分析するものである。
 このような質量分析法において測定対象物質を脱離、イオン化させる方法としては、例えば、MALDI法(マトリクス支援レーザ脱離イオン化法:matrix-assisted laser desorption ionization)や、SALDI法(表面支援レーザ脱離イオン化法:surface-assisted laser desorption ionization)がある。
 MALDI法とは、測定対象物質をマトリクス(例えば、シナピン酸やグリセリン等)に混入した試料に光を照射し、照射された光のエネルギをマトリクスに吸収させ、マトリクスとともに測定対象物質を気化させ、さらにマトリクスと測定対象物質との間でプロトン移動を発生させることで測定対象物質をイオン化する方法である。
 MALDI法は、測定対象物質に対しフラグメント化や変性等の化学的な影響の少ないソフトイオン化法として、難揮発性の物質や生体分子、合成高分子等の高分子量の物質の質量分析に広く用いられている。(特許文献1等)
 しかしながら、測定対象物質が合成高分子等の場合は、ポリマー鎖の化学構造の違いによって溶媒に対する溶解性やポリマー鎖の極性等が大きく異なり、また、主鎖構造が同じであっても平均分子量や末端基の化学構造等の違いにより様々な特性が異なるため、測定対象物質の種類に応じてマトリクス剤の種類や結晶の調製方法を最適化する必要がある。
 一方、SALDI法は、測定対象物質の脱離・イオン化を支援する機能を、質量分析用デバイスそのものに備えることによりソフトイオン化を行う方法である。例えば、特許文献2及び特許文献3では、表面にナノオーダの細孔構造を有するポーラスシリコン基板を用いた質量分析用デバイスにより、シリコンナノ構造とレーザ光との相互作用を利用してソフトイオン化を行っている。
 また、特許文献4では、シリコン基板上に金属微粒子を分散させた質量分析用デバイスを用いており、レーザ光がデバイスに照射された際に金属微粒子表面に生じる表面プラズモンを利用し、ソフトイオン化を行っている。
 これらのレーザ脱離イオン化方法を用いた質量分析法においては、特許文献5に記載されているように、レーザ光を測定対象物面内の二次元範囲に掃引させることで、測定対象物の二次元質量分析データを画像データとして取得することができる。

 上記のようにして取得される二次元質量分析データの画像データの空間分解能は、測定対象物質上のレーザ光ビームスポット径に依存する。すなわち、レーザ光のビームスポット径を小さくし、測定対象物質の脱離イオン化範囲を狭めることで、空間分解能は高まる。ここで、レーザ光のビームスポットを小さくするためには、大きな開口数(NA)でレーザ光を集光する必要があり、開口数を大きくするためには、径の大きな集光レンズを用いるか、焦点距離の短いレンズを使用する方法が考えられる。
 しかしながら、従来の質量分析装置においては、レーザ光を測定対象物質の上方から照射および集光する必要があるため、開口数の大きなレンズを用いると、イオン化された物質が質量分析部に移動する行路上の障害となり、正常に質量分析を行うことができなくなってしまう恐れがあり、十分に分解能を向上させることができない。
 本発明は、上記事情に鑑みてなされたものであり、分解能を向上させることが可能な質量分析装置を提供することを目的とするものである。
 本発明の質量分析装置は、レーザ光に対し透明であり、測定対象物質が付着されるサンプルプレートと、
 サンプルプレートが載置される、一部にレーザ光を透過する光透過部を備えた支持台と、
 支持台の、サンプルプレートが載置される面と反対の面側に配置され、レーザ光を、支持台の光透過部を通過させ、サンプルプレートの裏面側から測定対象物質に照射する光照射部であって、レーザ光を出力するレーザ光源と、測定対象物質にレーザ光を集光する集光レンズと、レーザ光の集光時に生じる収差を補正する収差補正機構とを備えた光照射部と、
 レーザ光の照射により、サンプルプレートの表面から脱離されイオン化された測定対象物質を検出する検出器とを有していることを特徴とする。
 ここで「透明」とは、入射する光が透過率50%以上であることをいうものとする。なお、透過率は75%以上であることが好ましい。
 本発明の質量分析装置においては、レーザ光をサンプルプレートの面内方向に二次元的に走査させる二次元走査機構を備えていることが好ましい。
 また、支持台が、透過部が開口された導電部材からなる支持台本体と、開口にサンプルプレートが載置される面と面一となるように設けられた光透過性の導電膜とを備えてなるものであることが好ましい。
 支持台に備えられている光透過性の導電膜は、透明な酸化物導電材料からなるものとすることができる。
 また、支持台に備えられている光透過性の導電膜は、金属メッシュであってもよい。
 サンプルプレートが、レーザ光に対し透明な基板および基板上に形成された、レーザ光が照射されることにより局在プラズモンを励起し得る金属微細構造とを有してなるものであることが好ましい。
 サンプルプレートの表面にイオン化促進剤が固着されていることが好ましい。
 二次元走査機構は、支持台に備えられた面方向駆動部から構成することができる。
 また、二次元走査機構は、光照射部に備えられた、レーザ光をサンプルプレートの面内掃引するレーザ光掃引部から構成されていてもよい。
 レーザ光の走査時にレーザ光のフォーカスを自動的に調整するオートフォーカスを備えていることが好ましい。
 本発明の質量分析装置は、レーザ光を透過するサンプルプレートを用い、サンプルプレートを支持する支持台の一部をレーザ光が透過するようにして、レーザ光を測定対象物質の上方からではなく、サンプルプレートの裏面側から照射するように構成されている。そのため、測定対象物質の飛翔光路を集光レンズが遮ることはなく、レーザ光を集光するための集光レンズの大きさが制限されないので開口数の大きいレンズを用いることができる。また、収差補正機構を備えることにより、レーザ光集光スポットを回折限界近くまで絞りこむことが可能となる。開口数の大きいレンズを用いて回折限界近くまで集光スポットを絞り込むことができるので、二次元画像データ取得時における分解能を十分に向上させることができる。

 以下、図面を参照して本発明の実施形態の質量分析装置について説明する。
 図1は、本発明の質量分析装置の一実施形態の概略構成を示す図である。本実施形態の質量分析装置1は、サンプルプレートから脱離した物質を所定距離飛行させてその飛行時間により物質の質量を分析する飛行時間型質量分析装置(TOF-MS)である。
 図1に示すように、質量分析装置1は、真空チャンバ2と、真空チャンバ2内に配置され、測定対象物質Mを含む試料を付着させる(または載置する)サンプルプレートである質量分析用デバイス(以下「デバイス」と称す。)3と、デバイス3を支持する支持台4と、デバイス3に付着した試料にレーザ光からなる測定光Lを照射して、試料中の測定対象物質Mをデバイス3から脱離させる光照射部5と、脱離した測定対象物質Mを所定方向に飛翔させる飛翔方向制御部6と、脱離した測定対象物質Mを検出して測定対象物質Mの質量を分析する質量分析部7とを有する。
 真空チャンバ2は、内部を真空にすることができるチャンバであり、図示しない吸引ポンプ等が接続されている。真空チャンバ2は、密閉された状態で吸引ポンプにより内部の空気を吸引することで内部を真空にされ、その真空状態を保つものである。
 また、真空チャンバ2には、光照射部5から射出された測定光Lを真空チャンバ2の内部に入射させるための窓18が設けられている。窓18は、外部と真空チャンバ2内の気圧差に対応することができる程度に耐圧性が高く、かつ、測定光Lを高い透過率で透過する材料で形成されている。
 デバイス3は、測定光Lが高い透過率で透過する透明な基板と、その基板上に形成された、測定光Lが照射されると局在プラズモンを励起する金属微細構造層が形成されてなる板状部材であり、真空チャンバ2内に配置されている。なお、このデバイス3の金属微細構造層上に、測定対象物質Mを含む試料が載置される。
 図2は、デバイス3の具体例の一部を示す断面図である。
 図2に示すように、デバイス3は、透明な基板20上に透明な微細凹凸構造21が形成されており、さらにその微細凹凸構造上に金属微細構造層22を有する。
 基板20は測定光Lに対し高い透過率を有する誘電体からなり、例えばガラスから形成されている。基板20上に形成されている微細凹凸構造21は、透明な誘電体から構成され、例えば、基板20上にアルミニウム(Al)を製膜した後に、その基板を水熱反応させた時に生成されるベーマイト(AlO・OH)により構成することができる。
 水熱反応により生成されたベーマイトから構成される微細凹凸構造21は、個々の凸部が数10nm程度の稜を有する微細な三角錐形状となっているため、可視~近赤外の測定光に対しては屈折率が徐々に変化している構造と等価となる。従って、微細凹凸構造21における測定光Lの透過率は高い。
 金属微細構造層22を形成する材料としては、局在プラズモンを発生させる種々の金属を使用でき、例えば、Au、Ag、Cu、Al、Pt、Ni、Ti、およびこれらの合金等が挙げられる。また、電場増強効果をより高くすることができるため、金属微細構造層22はAu、Ag等を用いて形成することがより好ましい。
 金属微細構造層22は、微細凹凸構造21上に金属を蒸着することにより形成することができる。
 金属微細構造層22の凹凸は、測定光Lの波長よりも小さいピッチおよび/または深さを有していれば、測定光Lの照射により局在プラズモンを誘起することができる。凹凸のピッチおよび深さが200nm以下、さらには、数nm~数10nm程度であることがより好ましい。
 上記のようなデバイス3に対し、測定光が照射されると、金属微細構造層の表面に局在プラズモンが誘起され、増強電場が形成される。
 なお、本実施形態においては、サンプルプレートとして、金属微細構造層を備えた質量分析用デバイスを用いているが、本発明の質量分析装置においては、裏面側からレーザ光を照射可能なものであればよく、ガラス板のみから構成されたサンプルプレートを用いてもよい。金属微細構造層を備えない場合にはレーザ光の照射による電場増強の効果がないため、測定対象物質の脱離、イオン化のためのレーザ光パワーを多くする必要がある。従って、金属微細構造層を備えたデバイスを用いることがより好ましい。
 支持台4は、試料が載置されている面とは反対側の面からデバイス3を支持し、デバイス3を所定位置に固定するものである。支持台4の中央部には、測定光Lを透過させる光透過部としての開口が設けられている。また、支持台4はデバイス面内XY方向に移動可能な機構を有している。このXY方向に移動可能な面方向駆動部(所謂XYステージ)が、測定光をデバイスの面内方向に二次元的に走査させる二次元走査機構を構成する。
 さらに支持台4はZ方向に位置調整が可能とされていてもよい。
 光照射部23は、レーザ光源9と、レーザ光源9から拡散し、射出されたレーザ光を平行光とするコリメートレンズ10と、ミラー11と、コリメータレンズ10で平行光とされ、ミラー11で反射されたレーザ光をデバイス3上の試料に集光する集光レンズ12とを有する。
 レーザ光源9は、所定波長のレーザ光を射出する光源である。ここで、レーザ光源としては、パルスレーザを用いることが好ましい。
 集光レンズ12には、レーザ光を回折限界まで集光するために、収差の小さな対物レンズや非球面レンズを使用することが好ましい。なお、二次元質量スペクトルデータ取得(イメージング)の際の分解能を十分なものとするためには、開口数(NA)の大きな集光レンズを用いることが望ましい。具体的には、単一細胞(10μm程度)のイメージングを考えた場合、1μm程度の分解能が必要であり、想定される励起光の最短波長300nmにおいて、この分解能を得ようとするとNA0.35以上の集光レンズが必要である。また可視光を想定した場合、NA0.6以上の集光レンズを用いるのが好ましい。
 また、光照射部23には、レーザ光がデバイス3の透明な基板20や窓18を通過する際に生じる収差を補正するための収差補正機構が集光レンズ12よりも上流側に備えられている。例えば、ミラー11として、収差補正機能を有する、反射光の波面を制御可能なメンブレンミラーを用いることによって、集光レーザ光の収差補正を行うことができる。この場合、ミラーが収差補正機構を構成するものである。その他、集光レンズの上流側に1対の収差補正レンズを収差補正機構として備えてもよい。
 従来の装置においては、真空チャンバ内に集光レンズが配置され、デバイスの表面側からレーザ光が照射されていたため、あるいは、集光レンズが真空チャンバ外に配置される構成である場合には焦点距離が比較的長かったため収差を考慮する必要性が無かった。しかしながら、本実施形態の質量分析装置では、光源を真空チャンバ外部に配置して、レーザ光を真空チャンバに設けられた透過窓18を透過させ、さらにデバイス3を透過させる構成であり、かつNAの大きな集光レンズを用いるので、収差を補正する機構を備える必要がある。
 飛翔方向制御部6は、支持台4と質量分析部7との間に配置された引き出しグリッド14と、引き出しグリッド14とデバイス3との間に電圧を印加する可変電圧源13と、グリッド14よりも質量分析部7側における測定対象物質Mの飛翔経路を囲むカバー15とを有し、デバイス3から脱離された測定対象物質Mに一定の力を作用させ、質量分析部7に向けて飛翔させる。
 引き出しグリッド14は、デバイス3と質量分析部7との間に、デバイス3の表面に対向して配置された中空の電極である。
 可変電圧源13は、支持台4と引き出しグリッド14とに接続しており、この可変電圧源13により、支持台4と引き出しグリッド14との間を所定電位差とし、所定の電界を形成する。
 カバー15は中空の筒型部材である。カバー15は、引き出しグリッド14と質量分析部7との間に、筒型の軸が測定対象物質Mの飛翔経路と平行に、かつ、測定対象物質Mの飛翔経路を囲うように配置されている。また、カバー15は、引き出しグリッド14側の端部が引き出しグリッド14に接近し、質量分析部7側の端部が、後述する質量分析部7の検出器24に接している。
 飛翔方向制御部6は、可変電圧源13により電圧を印加することでデバイス3と引き出しグリッド14との間に電界を形成し、デバイス3から脱離された測定対象物質Mに一定の力を作用させるよう制御する。電界により一定の力が加えられた測定対象物質Mは、デバイス3から引き出しグリッド14側に所定加速度で飛翔される。さらに、飛翔している測定対象物質Mは、カバー15の中空部分を通過し、質量分析部7まで飛翔する。
 上記の通り、TOF-MSにおいては、支持台4を電極として用いるため、支持台4は基本的には導電性を有する材料で構成される。支持台4の全体が導電性である必要はないが、少なくもデバイス3を載置する面は導電性材料で構成する。なお、測定光Lを透過させる透過部は、開口であってもよいし、透過性材料で構成されていてもよい。
 図3に、支持台4の一例の上面図および断面図を示す。図3に示すように、支持台4の中央部には、測定光Lが透過するための開口4aが設けられている。開口4aがあいていてもその周囲が電極として機能するため、支持台4とグリッド電極間に電圧を印加して、測定対象物質Mを飛翔させることはできるが、図3に示すように、この開口4aの上部に透明電極材料からなる膜41を備えていることが好ましい。膜41は、支持台4の開口4aに、デバイス3が載置される面と面一となるように設けられていることが好ましい。膜は開口4a内に充填された透明部材(図示していない。)上に形成すればよい。例えば、厚み1mm程度のガラスからなる透明な基体を開口4a内に配置し、その基体上にスパッタリングによりITO(酸化スズ添加酸化インジウム)を100nm程度成膜して形成することができる。この透明電極材料からなる膜41を備えることにより、測定対象物質の飛翔時における加速を十分なものとすることができる。
 図4に支持台の別の例の上面図を示す。図4に示すデバイス支持台4’は、開口4aに透明電極膜41に代えて金属のメッシュ42を備えている。金属のメッシュ42の網目の大きさは10μm~数十μm程度とする。測定光Lは集光させておりその集光スポットは1μm程度であり、測定光Lは網目を通過させてデバイス表面に照射することができる。測定光Lは金属のメッシュの金属線にかかると反射されてデバイス表面に照射される光量が減衰してしまうため、このようなデバイス支持台4’を用いる場合には、光照射部23からの測定光Lを網目を通過してデバイス表面に照射した状態で測定を行うようにデバイス支持台のX-Y方向への移動等により測定光の照射位置を制御すればよい。例えば、測定光Lの戻り光を検出することにより、制御可能である。
 質量分析部7は、測定光Lが照射されてデバイス3の表面から脱離され、引き出しグリッド14を通過して飛翔してきた測定対象物質Mを検出する検出器24と、検出器24による検出値を増幅させるアンプ16と、アンプ16からの出力信号を処理するデータ処理部17とを有する。なお、検出器24は、真空チャンバ2の内部に配置され、アンプ16及びデータ処理部17は、真空チャンバ2の外に配置されている。
 また、検出器24としては、例えば、マルチチャンネルプレート(MCP)を用いることができる。質量分析部7は、検出器24で検出した検出結果に基づいて、データ処理部17で測定対象物質Mのマススペクトルを検出し、測定対象物質Mの質量(質量の分布)を検出する。
 質量分析装置1は、基本的に以上のように構成されている。
 以下、質量分析装置1を用いた質量分析方法について説明する。
 まず、デバイス3の表面に測定対象物質M(もしくは測定対象物質を含む試料)を載置し、デバイス3を支持台4に載置する。
 次に、可変電圧源13からデバイス3と引き出しグリッド14間に所定電圧を印加し、所定のスタート信号により光照射部23から測定光Lを射出させ、デバイス3の裏面側から測定対象物質Mに測定光Lを照射する。
 デバイス3の金属微細構造層22に測定光Lが照射されることで、デバイス3の表面では、プラズモンに起因する増強電場が形成され、その増強電場により増強された測定光Lの光エネルギにより測定領域から測定対象物質Mが脱離する。
 脱離した測定対象物質Mは、デバイス3と引き出しグリッド14との間に形成された電界により引き出しグリッド14の方向に引き出されて加速し、引き出しグリッド14の中央の孔を通ってカバー15の中空部を検出器24の方向に略直進して飛行し、検出器24に到達して検出される。
 脱離後の測定対象物質Mの飛行速度は物質の質量に依存し、質量が小さいほど速いため、質量の小さいものから順に検出器24に到達する。
 検出器24からの出力信号は、アンプ16により所定のレベルに増幅され、その後データ処理部17に入力される。
 データ処理部17には、上記スタート信号と同期して同期信号が入力されており、この同期信号とアンプ16からの出力信号とに基づいて検出した物質の飛行時間をそれぞれ算出する。
 また、データ処理部17は、飛行時間から質量を導出してマススペクトル(質量スペクトル)を算出する。さらに、データ処理部17は、算出したマススペクトルから測定対象物質の質量を検出し、また、測定対象物質を同定する。データ処理部17は、例えば質量分析にためのプログラムが組み込まれたパーソナルコンピュータ等により構成される。
 測定対象物質Mの所定位置の質量検出が終了次第、支持台4がデバイス面内方向に移動し、異なる箇所に測定光を照射し、再度上記質量検出を繰り返すことで、測定対象物質Mの二次元質量スペクトルデータを取得することができる。
 本実施形態の質量分析装置によれば、レーザ光(測定光)を透過するサンプルプレートを用い、サンプルプレートの裏面側から照射するように構成されているので、測定対象物質の飛翔光路を集光レンズが遮ることがない。集光レンズの配置の自由度が高く、レーザ光を集光するための集光レンズの大きさも制限されないので開口数の大きいレンズを用いることができ、収差補正機構を備えていることから、レーザ光集光スポットを回折限界近くまで絞りこむことが可能である。そのため、高解像度の質量分析画像データを取得することができる。
 なお、上記実施形態のように、サンプルプレートとして表面に微細凹凸構造層を有するデバイスを用いた場合、その表面に載置される試料の面も凸凹を有するため、レーザ光を二次元走査して二次元の質量スペクトルデータを取得する際には、レーザ光のスポット位置が常に試料面に集光するために、オートフォーカスを備えていることが好ましい。
 オートフォーカスは、具体的には分割フォトディテクタおよび結像光学系を備えた焦点位置検出機構と集光レンズまたはサンプルプレートの光軸方向(z方向)位置調整機構とから構成することができる。レーザ光の試料への照射時に、微細凹凸構造層からのレーザ光の微弱な反射光を分割フォトディテクタ上に結像し、レーザ光のデフォーカスを分割フォトディテクタの信号強度比として検出し、その強度比の差(デフォーカス分)をレンズまたはサンプルプレートの光軸方向位置調整機構にフィードバックすることで、オートフォーカスを行うことができる。
 次に、本発明の質量分析装置の第2の実施形態について説明する。
 図3は、本発明の質量分析装置の第2の実施形態の概略構成を示す図である。図3に示す質量分析装置25は、光照射部23’の構成が第1の実施形態のものと異なる。光照射部23’以外の構成については質量分析装置1と同様であるので、同一の部材および構成には同一符号を付してその説明は省略する。
 本実施形態において光照射部23は、コリメートレンズにより平行光化されたレーザ光を集光レンズ12側に反射するミラーとして、カルバノミラー対11’を備えている。カルバノミラー対11’を用いて、集光レンズ12に対し角度を設けてレーザ光を照射することで、デバイス3の表面におけるレーザ集光点を、表面に沿って二次元状に掃引させることができる。すなわち、本質量分析装置25においては、ガルバノミラー対11’が二次元走査機構であるレーザ光掃引部を構成している。
 ガルバノミラー対11’を用いれば、質量分析装置1のように支持台4をX-Y方向に移動させて測定対象物質M上のレーザ光集光点を移動させるよりも、より位置精度高く、高速に集光点を操作することが可能となり、高解像度かつ高速に二次元質量スペクトルデータを取得することができる。
 また、本実施形態においては、ガルバノミラー対11’と集光レンズ12との間に1対のレンズからなる収差補正レンズ19が備えられており、光照射部23において生じる収差を補正し、精度よくデバイス表面にレーザ光を照射することができる。
 本第2の実施形態の質量分析装置においても、第1の実施形態の場合と同様に、高解像度の質量分析画像データを取得することができる。
 上記各実施形態の質量分析装置において、サンプルプレート3の表面には、イオン化促進剤が固着されていることが好ましい。イオン化促進剤としては、例えばシナピン酸、フェルラ酸、ゲンチジン酸、ジスラノール等が挙げられる。
 イオン化促進剤を備えることにより、より定パワーのレーザ光(測定光)により、試料の脱離、イオン化が可能となり、測定対象物質の変性や損傷を抑制することができる。
 さらに、サンプルプレートである質量分析用デバイス3の表面に、測定対象物質Mを捕捉可能な表面修飾Rが施されていてもよい。例えば、測定対象物質が抗原であるような場合は、その抗原と特異的に結合可能な抗体により表面を修飾しておくことにより、試料接触面上の測定対象物質Mの濃度を高められるので、感度を向上させることができる。
 図6Aは、質量分析用デバイス3の表面に表面修飾Rが施された様子を示す断面図であり、図6Bは、図6Aに示すデバイス3から測定対象物質が脱離した状態を示す断面図である。なお、図6A及び図6Bでは視認しやすくするために表面修飾Rおよび表面修飾Rの構成要素は拡大して示してある。
 図6Aに示すように、表面修飾Rは、デバイス3の表面に、デバイス3の表面と結合する第1のリンカー機能部Aと、測定対象物質Mと結合する第2のリンカー機能部Cと、第1のリンカー機能部Aと第2のリンカー機能部Cとの間に介在し、測定光の照射により生じる電場で分解する分解機能部Bとを有するものである。図示例では、測定対象物質Mは、表面修飾Rを介して、質量分析用デバイスの測定領域の近傍に配置されている。
 なお、表面修飾Rは、第1のリンカー機能部Aと、分解機能部Bと、第2のリンカー機能部Cとを全て備えた一つの物質であってもよいし、それぞれが異なる物質からなっていてもよい。また、第1のリンカー機能部Aと分解機能部B、あるいは、分解機能部Bと第2のリンカー機能部Cが一つの物質であってもよい。
 ここで、デバイス3に測定光Lが照射されると、金属微細構造層22の表面で局在プラズモンが発生し、測定領域の表面において増強電場が発生する。また、測定光の光エネルギーは、測定領域の表面に発生した増強電場により、表面付近において高められる。
 この高められたエネルギーにより表面修飾Rの分解機能部Bが分解され、図6Bに示すように、測定対象物質Mに第2のリンカー機能部Cが結合されたものが、測定領域表面から脱離される。
 このように表面修飾を用いることで、測定対象物質を微細構造体の表面から脱離させることができる。また、表面修飾Rを介して測定対象物質Mはデバイス3と結合しているので、測定対象物質Mは、測定領域のデバイス3の表面から離れて存在させることができる。
 ここで、デバイス3の表面において得られる電場増強効果は、局在プラズモンにより生じる近接場光による電場増強効果であるので、表面からの距離に対して指数関数的に減少していくものである。従って、図6Aに示すように、測定対象物質Mが表面から比較的離れて存在させることにより、測定対象物質Mに照射される測定光の光エネルギーは、電場増強による影響の少ないものとすることができる。すなわち、増強された光エネルギーにより測定対象物質Mがダメージを受けることを抑制でき、高精度な質量分析が可能となる。
 なお、この表面修飾Rの分解機能部Bは前述のイオン化促進剤に相当する。
 上記実施形態においては、透明基板20上にベーマイトからなる透明な微細凹凸構造21およびその上に金属微細構造層22を備えてなる質量分析用デバイス3をサンプルプレートとして用いるものとしたが、このような測定光の照射により表面に局在プラズモンを誘起し得る金属微細構造層の構成は上記実施形態に限るものではない。以下、他の形態の質量分析用デバイス(サンプルプレート)について説明する。
 図7は、質量分析用デバイスの他の一例の概略構成を示す斜視図である。
 図7に示す質量分析用デバイス80は、透明基板82と透明基板82上に配置された多数の金属微粒子84とで構成されている。基板82は、上記実施形態の場合と同様のものを用いればよく、測定光に対して高い透過率を有し、金属微粒子を電気的に絶縁して支持可能な材料からなる。例えば、石英やサファイヤ等が挙げられる。
 多数の金属微粒子84は局在プラズモンを誘起し得る大きさの微粒子であり、基板82の一面上に分散された状態で固定されている。
 また、金属微粒子84は、上述した金属微細凹凸構造層を構成する各種金属で形成することができる。また、金属微粒子84の形状は特に限定されず、例えば、球、ディスク型、直方体型でもよい。金属微粒子84の最大長(球、ディスク型であればその直径、直方体型であればその長辺の長さ)が測定光となるレーザ光の波長よりも小さければ、レーザ光の照射により局在プラズモンが誘起される。
 このような構成の質量分析用デバイス80であっても、金属微粒子が配置された検出面に測定光が照射されることで、増強電場を発生させることができる。
 図8は、質量分析用デバイスの他の一例の概略構成を示す上面図である。図8に示す質量分析用デバイス90は、透明基板92と基板92上に配置された多数の金属ナノロッド94とで構成されている。透明基板92は、上記の例と同様である。
 金属ナノロッド94は、局在プラズモンを誘起し得る大きさであり、短軸長さと長軸長さが異なる棒状の金属ナノ粒子であり、基板92の一面に、分散された状態で固定されている。金属ナノロッド94は、その短軸長さが3nm~50nm程度、長軸長さが25nm~1000nm程度であり、長軸長さが測定光となるレーザ光の波長よりも小さければ、レーザ光の照射により局在プラズモンが誘起される。
金属ナノロッド94は、上述した金属微粒子と同様の金属で作製することができる。なお、金属ナノロッドの詳細な構成については、例えば、特開2007-139612号公報に記載されている。
 このような構成の質量分析用デバイス90を用いても金属ナノロッドが配置された検出面に測定光が照射されることで、増強電場を発生させることができる。
 さらに、図9は、質量分析用デバイスの他の一例の概略構成を示す斜視図である。
 図9に示す質量分析用デバイス95は、透明基板96と基板96上に配置された多数の金属細線98とで構成されている。透明基板96は、上述した基板と同様の構成である。
 金属細線98は、局在プラズモンを誘起し得る線幅の線状部材であり、基板92の一面に格子状に配置されている。金属細線98は、上述した金属微細構造層や金属微粒子と同様の金属で作製することができる。また、金属細線98の作製方法は、特に限定されず、蒸着、メッキ等、金属配線を作製する種々の方法で作製することができる。
 ここで、金属細線98の線幅は、例えば、50nm以下、特に30nm以下であることが好ましい。また、金属細線98の配置パターンは、特に限定されない。例えば、複数の金属細線を交差させずに、互いに平行に配置してもよい。また、金属細線の形状も直線に限定されず、曲線であってもよい。
 このような構成の質量分析用デバイス95であっても、金属細線98が配置された検出面に測定光が照射されることで、局在プラズモンに起因する増強電場を発生させることができる。
 なお、質量分析用デバイスとしては、上述した構成に限るものではなく、ぞれぞれの局在プラズモンを誘起し得る構造を組み合わせた構成としてもよい。
 以上、本発明に係る質量分析装置について詳細に説明したが、本発明は、以上の実施形態に限定されるものではなく、本発明の要旨を逸脱しない範囲において、各種の改良や変更を行ってもよい。
 例えば、本実施形態では、質量分析装置がTOF-MSである場合を例に説明したが、イオン化された試料イオンの質量分析を行う装置としては、TOF型のものに限らず、IT(Ion Trap;イオントラップ型)、FT(ICR)(Fourier-Transform Ion Cyclotron Resonance;フーリエ変換型)、また複数の質量分析手法を組み合わせた手法であるQqTOF(Quadrupole-TOF;四重極-TOF型)、TOF-TOF(TOF連結型)などの質量分析装置を用いることができる。

Claims (10)

  1.  レーザ光に対し透明であり、測定対象物質が付着されるサンプルプレートと、
     該サンプルプレートが載置される、一部に前記レーザ光を透過する光透過部を備えた支持台と、
     前記支持台の、前記サンプルプレートが載置される面と反対の面側に配置され、前記レーザ光を、前記支持台の前記光透過部を通過させ、前記サンプルプレートの裏面側から前記測定対象物質に照射する光照射部であって、前記レーザ光を出力するレーザ光源と、前記測定対象物質に前記レーザ光を集光する集光レンズと、該レーザ光の集光時に生じる収差を補正する収差補正機構とを備えた光照射部と、
     前記レーザ光の照射により、前記サンプルプレートの表面から脱離されイオン化された前記測定対象物質を検出する検出器とを有していることを特徴とする質量分析装置。
  2.  前記レーザ光を前記サンプルプレートの面内方向に二次元的に走査させる二次元走査機構を備えていることを特徴とする請求項1記載の質量分析装置。
  3.  前記支持台が、前記透過部が開口された導電部材からなる支持台本体と、該開口に前記サンプルプレートが載置される面と面一となるように設けられた光透過性の導電膜とを備えてなるものであることを特徴とする請求項1または2記載の質量分析装置。
  4.  前記光透過性の導電膜が、透明な酸化物導電材料からなるものであることを特徴とする請求項3記載の質量分析装置。
  5.  前記光透過性の導電膜が、金属メッシュであることを特徴とする請求項3記載の質量分析装置。
  6.  前記サンプルプレートが、前記レーザ光に対し透明な基板および該基板上に形成された、該レーザ光が照射されることにより局在プラズモンを励起し得る金属微細構造とを有してなるものであることを特徴とする請求項1から5いずれか1項記載の質量分析装置。
  7.  前記サンプルプレートの表面にイオン化促進剤が固着されていることを特徴とする請求項1から6のいずれかに記載の質量分析装置。
  8.  前記二次元走査機構が、前記支持台に備えられた面方向駆動部であることを特徴とする請求項2から7いずれか1項記載の質量分析装置。
  9.  前記二次元走査機構が、前記光照射部に備えられた前記レーザ光を前記サンプルプレートの面内掃引するレーザ光掃引部であることを特徴とする請求項2から7いずれか1項記載の質量分析装置。
  10.  前記レーザ光の走査時にレーザ光のフォーカスを自動的に調整するオートフォーカスを備えていることを特徴とする請求項2から9いずれか1項記載の質量分析装置。
