Description

プローブ顕微鏡用ホルダ、プローブ顕微鏡および試料計測方法
 本発明は、生体組織等を高空間分解能で計測する顕微鏡において、その試料を配するホルダ、当該ホルダを用いたプローブ顕微鏡および当該顕微鏡を用いた試料計測方法に関する。
 培養液中における生体基板材料への細胞の接着やその後の伸展・分化などの生体反応を計測・評価・制御する場合に、生体分子・生体組織・生体基板材料などの水和現象が重要である。このとき、水和構造は、水を主成分とする培養液中における試料-培養液界面において、試料表面と水分子の相互作用、および、水分子間での水素結合を含む相互作用から形成される三次元構造を示している(非特許文献1)。人工血管の内壁と赤血球との癒着などに代表されるいわゆる生体適合性は、この水和構造と密接に関連していると考えられている。さらに、培養液中の試料表面の凹凸、電位分布、分子やたんぱく質などの組成分布や配列構造などは、培養液中における生体分子・生体組織・生体基板材料などの生体反応に関して、特に、重要な特性である。
 培養液中における生体分子・生体組織・生体基板材料などの試料-培養液界面を観察・計測する手法として、従来は、光学顕微鏡、および、ラマン分光法、第二高調波法、和周波分光法などの非線形光学顕微鏡が用いられている。特に、和周波分光法では、試料-培養液界面における水和構造に関する水分子の配列構造を計測できる。非線形光学顕微鏡としては、例えば、特許文献1では、プローブとターゲットとの間の相互作用を、界面付近の水分子、溶媒分子、または、標識物質による第二高調波光や和周波光による表面選択性の非線形光学法が開示されている。
 一方、走査プローブ顕微鏡は、原子間力顕微鏡(AFM:Atomic  Force  Microscopy)を基礎としている。走査プローブ顕微鏡の一例である、走査ケルビンプローブ顕微鏡では、導電性探針を持ったカンチレバーと試料との間に働く静電場力をカンチレバーのたわみとして検出しながら、探針を試料表面上で走査することにより静電場力分布をマッピングする手法である。探針には静電場力以外に原子間力等も加わっており、静電場力を他の相互作用から分離する必要がある。そのために、まず、カンチレバーを振動させ、探針と試料の接触時に働く原子間力により減少する振動振幅を一定に保つように探針-試料間距離を調整する。これにより試料表面の高さ方向の位置が決定され、そこから一定の距離だけ探針を試料表面から離した状態で、カンチレバーの振動の位相変化から長距離力である静電場力を検出する(例えば、特許文献2)。

 代表的な非線形光学顕微鏡である、レーザを用いた和周波顕微鏡(Sum Frequency Microscope)は,光触媒界面、表面吸着系,半導体界面,超電導体表面の、電子状態,ボンド配向,分子配向の分布および秩序を探ることに使用されている。しかし、空間分解能は1μm程度であり、微細な構造の観察はできない。
 一方、走査プローブ顕微鏡は培養液中動作が可能であり、比較的簡便な操作により10nm程度の高分解能が得られる。しかし、試料表面の位置を検出するために探針を試料表面と接触させなければならず、測定中に探針先端が破壊されたり、試料表面が付着するなどした場合、検出される信号が不安定となってしまうという問題があった。
 また、計測対象物である細胞の生存条件を維持したまま計測しようとする場合、細胞の生存条件の一つである温度約37度を維持すると、細胞を取り囲んでいる水(液体、たとえば、培養液)が蒸発してしまい、その結果、細胞そのものが乾燥により生存状況を維持できない可能性がある。その結果、細胞の生存条件を維持したまま、細胞もしくは細胞表面からの物理情報を取得することが不可能となる。
 しかしながら、上記の従来例ではこの点について考慮されておらず、計測対象物を保持するホルダについては、記載されていない。
 そこで本発明では、細胞等の計測対象物を内部に収容する容器と、前記計測対象物の少なくとも一部を覆い、かつ計測用プローブ挿入用の開口部を有する第1の蓋部と、前記第1の蓋部と連結し、前記容器を覆い、かつ、計測用プローブ挿入用の開口部を有する第2の蓋部とを有する計測用ホルダとする形状とした。
 また、このホルダを用いて、細胞等をプローブ顕微鏡にて測定するものとした。
 本発明によれば、培養液等が蒸発することなく、試料の良好な状態を維持することができるので、細胞の生存条件を維持したまま、培養液中で生体分子・生体組織・生体基板材料などと水の界面において水分子の配向度を高空間分解能で計測することが可能ととなり、細胞内または細胞塊内の特定元素の凝集位置,機能を特定できる。

 本発明は、プローブ顕微鏡において、細胞や水に代表される生体試料・水試料を計測する上でのサンプルホルダの構造について開示する。この開示に先立ち、探針-試料間に働く静電気力分布を計測する走査プローブ顕微鏡(走査ケルビンプローブ顕微鏡)の構造を図2において開示する。
 本実施例(図2)では、走査プローブ顕微鏡の一形態としての、探針増強走査和周波顕微鏡を開示する。探針1は振動子2に設置され、試料3との相対位置は振動子2により制御される。探針1は、入射する光中に置かれたときに、先端近傍に近接場光強度が増幅・集中する材料が選択される。また、ラマン分光法や和周波分光法などのようにラマン散乱を用いる場合は、表面増強ラマン散乱を有効に用いることができる、金、銀、銅、アルミなどの金属やそれらの化合物が用いられる。シリコン探針に厚さ1~20nmの金薄膜を蒸着した探針は、有効な探針の候補として用いられる。また、本実施例においては、振動子2は主に試料3の垂直方向に振動し、探針1と試料3の距離は300nm以下で制御され、また、振動子2の固有振動数は200kHz~2MHzが用いられる。本実施例では、振動子2として、長手方向に伸縮する水晶振動子が用いられるが、原子間力顕微鏡などの走査プローブ顕微鏡で一般的に用いられるチューニングフォーク型の水晶振動子、ピエゾ素子による振動子、カンチレバーにピエゾ素子を配置した振動子などを用いることができる。
 振動子2により探針1は、振動子2の固有振動数近傍の周波数(固有振動数の±1%程度以内)で、試料3の表面に対して垂直方向に振動させられる。探針1と試料3との間の相互作用(力)により、振動子2に印加する電圧と振動子2の実際の振動振幅には位相差が生じるが、その位相差は、本実施例では、振動子2に印加する交流電圧と振動子2に流入する電流との位相差により、探針-試料間の相互作用(力)がわかり、探針-試料間距離がわかる。また、この位相差を一定にしながら、走査機構4により試料3と探針1との相対位置を試料に垂直方向と試料の平面方向に走査することにより、走査プローブ顕微鏡の一方式である原子間力顕微鏡(AFM)を構成でき、試料表面の凹凸が計測できる。探針1と試料3の距離は、一般的には、最近接位置で0nm(接触)から100nmの距離まで接近するが、探針1を試料3の内部までめり込ませることも可能である。また、振動子2の振動振幅が一定の量だけ減少するようにしながら、走査機構により試料3と探針1との相対位置を試料に垂直方向と試料の平面方向に走査することにより、探針1と試料3の距離を最近接位置で0nmとすることもできる(タッピングモードAFM)。
 試料ホルダ5は、培養液6の保持・交換が可能である。また,培養液6のかわりに,水、溶媒の使用も可能である。
 探針1が近接する試料3の領域近傍にパルスレーザ光、または、同期して入力する複数のパルスレーザ光を入力し、フィルター付検出器7で出力光8の強度を計測する。本実施例では、波長532nmの緑色パルスレーザ光である第一のパルスレーザ光9と、波長が2.3~10ミクロンで可変の赤外パルスレーザ光である、第二のパルスレーザ光10を同期して入力する。出力光8をフィルター付検出器7に入力して、第一のパルスレーザ光9の周波数と第二のパルスレーザ光10の周波数との和の周波数(和周波)の強度を計測する。第二のパルスレーザ光10の周波数に依存する和周波の出力光8の強度を記録することにより和周波分光ができる。本実施例では、波数が3200カイザーのピークと波数が3400カイザーのピークを比較して、ポリカーボネートと培養液14との界面における4面体配位した水分子と非対称結合した水分子の配向の割合を特定できる。
 なお、上記は、パルスレーザ光を用いる例について説明したが、試料表面のみの計測の場合には、パルスレーザと検出器は必須ではない。
 計測を行う際には、試料を加熱する必要があり、その際に、水・培養液などの蒸発を抑え、かつ細胞が生存したままでの計測を実現する必要がある。この実現に必要な試料ホルダの構造を図1に示す。探針1をホルダへ挿入しやすい構造を実現するために、ホルダ内部に円筒状の穴を設けていることを特徴とする。11はカバーであり、その中心に円筒状の穴12を具備している。また、13は培養液の取り入れ口であり、計測中にホルダの温度維持(37℃程度が望ましいとされているが、本温度に限定されるものではない)のために、蒸発した培養液や水を補給するために設けられている。また、液体(培養液)が劣化した際には、これを排出するためにも利用できる。
 14はホルダ本体(容器)であり、円筒状の穴12とスペーサー15により固定されている。この図1に示した構造を補足するために、図3を用いて説明を行う。この図3は、図1の側面図である。ホルダカバー11、円筒状の穴12、培養液の取り入れ口13、が同心円状に具備されており、円筒状の穴の下部に15に示したスペーサーが具備されている。このように、試料18の一部を覆う第1の蓋部と、ホルダ本体14を覆う第2の蓋部(ホルダカバー)11と、第1の蓋部と第2の蓋部を連結する連結部を設けた形状となっている。第1の蓋部には、探針1を通す穴26が設けられ、第2の蓋部(ホルダカバー)11にも、探針1を通す穴12が設けられている。連結部は、空洞となっている。
 これら第1の蓋部、連結部、第2の蓋部は、ホルダ本体14へ接続されている。
 ここで、スペーサー15は、試料18の高さの分だけ下駄として設けたが、試料が平坦等の場合には、穴26があるため、スペーサーは必ずしも必須ではない。また、図1、3では、スペーサー15のような形状を図示したが、下駄あげであるため、任意の形状で構わない。
 なお、ここではホルダカバー11、円筒状の穴12、培養液の取り入れ口13を同心円状に記載したが、12の穴は探針が通れば良いので、その他の形状でも構わない。勿論、培養液の取り入れ口13の形状は、円形である必要はなく、どのような形状でも構わない。また、ホルダ本体14が円柱状であるため、ホルダカバーも円柱状の形状を示したが、ホルダ本体は、試料18を保持すれば良いので、円柱状に限らず、任意の形状で構わない。それに伴い、ホルダカバー11も、任意の形状でホルダ14と接続されることとなる。
 また、試料18の生存条件を長期間維持するために、試料18を加温すると好ましい。なお、短時間で計測が終了する場合には、加温するためのヒータは必須でない。加温する場合には、図3に示した通り、ホルダ本体14へは、ヒータ16が接続されており、サンプルホルダの温度維持を行う。また、サンプルホルダの温度の計測を目的として、ペルチェ素子などから構成される温度センサー17が接続されている。このホルダ本体上には、細胞や水に代表される試料18を配することができる。ここでは、ホルダ本体14とヒータ構造物16を接続するようにして構成したことにより、ホルダ本体を消耗品として廃棄しても、ヒータ構造物16は繰り返し使用できるため、コスト面で有利となる効果がある。
 図4は、実際のホルダの装着図である。ホルダカバー11とホルダ本体14は密着されており、スペーサー15が試料18上に軽く接触した状態で保持される。これらホルダやスペーサーを貫通する形で、円筒状の穴12を経由した探針1が試料上へアプローチすることができる。
 この図4までに示したホルダの実際の使用方法を図5に示す。19はプローブ顕微鏡の制御装置であり、探針1の位置並びにフィルター付き検出器7へ到達した光量の処理を行うものである。20はヒータの制御装置であり、21は温度センサーの検出装置である。これら20に示したヒータの制御装置と21に示した温度センサーの検出装置は相互に接続されており、温度をフィードバック系により制御することで一定の所望の温度へ設定することが可能である。これら設定温度と検出温度の情報、並びにプローブ顕微鏡の制御装置19も相互に接続されており、これらの情報の伝送のハブになるのは電子計算機22である。
 本実施例にて開示したホルダを用いて、ラットの培養心筋(Primary Cell Co_,Ltd_ 株式会社プライマリーセル 心筋細胞培養キット)について、心拍数を画像計測した。まず、事前準備として、ヒータ16を加温した。一方、試料キット18をホルダ14に配置し、培養液を浸し、その後、スペーサ15を介してホルダカバー11をセットした。そして、ヒータ16とセンサー17により、ホルダの温度をほぼ一定に保ちつつ、振動子2、探針1、パルス照射光を用いて表面形状及び細胞の状態を、1時間弱観測した。時折、穴13より、培養液を補充した。この結果を図6に示す。設定した温度は39℃であるが、ホルダ表面での温度は37℃であった。心拍数は、培養容器の環境により完全に一定を保つことは困難ではあるが、おおむね1分あたり100心拍数を維持することができた。
 本実施例では、ホルダに関して、その変形例を示す。図1に示したホルダでは、水や培養液をホルダカバー上から入れるものであった。しかし、実際には、培養液が劣化する可能性もあり、また、プローブ顕微鏡の探針との干渉を回避する構造を具備する必要がある。これらの課題を解決するために、水・培養液の注入と回収をより容易に実現する方法を図7、図8にて開示する。
 図7では、培養液の取り入れ口13に加え、培養液の排出口23を具備することを特徴とするホルダを開示する。この排出口23については、ホルダの側面に具備することを特徴とする。これは、前述の通り、上部から接近するプローブのアプローチを阻害せずに、ホルダ内部での劣化した液体を排出することが容易に実現できるからである。
 更に、図8では、培養液の取り入れ口13もホルダの側面に具備することを特徴とする構造を開示している。これにより、培養液の注入、排出とも、探針への干渉の影響を回避する形で実現することが可能となった。尚、これらの注入・排出とも、実際には、マイクロシリンジなどを用いることで、注入量・排出量を制御することが可能である。人工的に注入・排出を行うだけではなく、これらの制御については、図5に示した電子計算機を用いて制御することも可能である。
 本実施例のように、培養液を補充、回収することにより、計測に、より長時間を要する場合でも、生存状態を維持したまま、計測することができる効果がある。
 本実施例では、ホルダに関して、温度計測の実施方法に関する変形例を示す。実施例1、実施例2に示したホルダの構造では、ヒータは試料ホルダの下部に設置され、ヒータと温度センサーは一体化した構造となっていた。しかし、本開示方法では、ホルダの熱伝導特性により、実際の試料での温度と異なる可能性がある。そこで、本実施例では、センサーの配置位置に関する発明を開示する。
 図9では、温度センサー17をホルダ本体内部へ挿入する構造を具備する。これにより、プラスチック材料などにより構成されるホルダの熱伝導特性を正しく反映した形で、試料3の温度を計測することが可能となる。
 また、図10では、光ファイバセンサ24を用いて試料3表面の温度を計測することが特徴である試料ホルダを開示している。本方法であれば、試料ホルダ5への温度センサー17の取り付けが不要となり、より簡易な試料ホルダが実現できる。
 更に、図11では、ヒータ16の代替えとして、光ファイバ25を用いたレーザもしくは光に代表される電磁波の照射により、試料ホルダ5の加熱を行うことを特徴とする試料ホルダ5の構造を開示する。実際の照射に使用するレーザの波長は、試料が生体物質であることから考えて、赤外線の帯域、もしくは、試料ホルダの材質が有する吸収波長体であることが望ましい。このように、外部から電磁波(光)照射することにより、直接ホルダをヒータで加熱するよりも、計測時に、電磁ノイズの影響を低減することができる。
 本実施例では、ホルダの着脱構造を図12に示す。このように、ホルダ本体14と、スペーサ15からホルダカバー11を着脱可能とすることで、ホルダ本体(容器)14内の細胞を入れ替えて再計測することが可能となる。図12の上と下はそれぞれ洗浄し、繰り返し利用可能である。また、計測中でも、時折大気に開放することで、細胞が呼吸可能となり、より、生存条件を高く維持したまま、長時間計測が可能となる。

Claims (13)

  1.  計測対象物を内部に収容する容器と、
     前記計測対象物の少なくとも一部を覆い、かつ計測用プローブ挿入用の開口部を有する第1の蓋部と、
     前記第1の蓋部と連結し、前記容器を覆い、かつ、計測用プローブ挿入用の開口部を有する第2の蓋部とを有することを特徴とする計測用ホルダ。
  2.  前記第1の蓋部には、前記容器側に、前記容器から所定の高さを保つためのスペーサーが形成されていることを特徴とする請求項1記載の計測用ホルダ。
  3.  前記第2の蓋部には、液体注入及び/または液体排出用の穴が形成されていることを特徴とする請求項1記載の計測用ホルダ。
  4.  前記穴の配置位置は、前記第2の蓋部の上部であることを特徴とする請求項3記載の計測用ホルダ。
  5.  前記穴の配置位置は、前記容器の側部であることを特徴とする請求項3記載の計測用ホルダ。
  6.  さらに、前記容器を加熱するための加熱用素子が設けられていることを特徴とする請求項1記載の計測用ホルダ。
  7.  さらに、前記容器の温度を計測するための温度計測用センサーを具備することを特徴とする請求項6記載の計測用ホルダ。
  8.  前記加熱用素子及び/または前記温度計測用センサーは、前記容器からの着脱式であることを特徴とする請求項7記載の計測用ホルダ。
  9.  前記加熱用素子は、前記容器に設けられていることを特徴とする請求項6記載の計測用ホルダ。
  10. 前記加熱用素子は光ファイバであることを特徴とする請求項6記載の計測用ホルダ。
  11. 前記温度計測用センサーは光ファイバであることを特徴とする請求項7記載の計測用ホルダ。
  12.  振動子と、
     前記振動子先端に設けられた探針と、
     前記探針が、計測対象物の表面を走査することによって、前記計測対象物表面を計測する手段と、
     前記計測対象物を内部に収容する容器と、前記計測対象物の少なくとも一部を覆い、かつ前記探針の挿入用の開口部を有する第1の蓋部と、前記第1の蓋部と連結し、前記容器を覆い、かつ、前記探針の挿入用の開口部を有する第2の蓋部とを有する計測用ホルダとを有することを特徴とするプローブ顕微鏡。
  13.  計測対象物を内部に収容する容器と、前記計測対象物の少なくとも一部を覆い、かつ計測用プローブ挿入用の開口部を有する第1の蓋部と、前記第1の蓋部と連結し、前記容器を覆い、かつ、計測用プローブ挿入用の開口部を有する第2の蓋部と、を有する計測用ホルダを用い、
     前記計測用ホルダの前記容器内に、前記計測対象物を液体とともに収容するステップと、
     前記計測対象物の温度を計測する温度計測用センサーにて検知された温度に従って、前記計測対象物を加熱するための加熱用素子の加熱温度を調整しながら、前記計測用プローブが、前記計測用ホルダを介して、前記計測対象物の表面を走査することによって、前記計測対象物の表面を計測するステップと、を有することを特徴とする試料計測方法。
