Abstract
본 발명은 (a) 기판 표면에 점성물질의 용액을 도포하는 단계; (b) 상기 점성물질의 용액과 프레임을 접촉시키는 단계; (c) 상기 접촉된 프레임과 기판이 이격되도록 기판, 프레임 또는 기판과 프레임을 리프팅 하여 솔리드 마이크로니들를 제조하는 단계; (d) 상기 단계 (c) 과정 중 또는 상기 단계 (c) 과정이 종료된 후 상기 솔리드 마이크로니들에 만곡형(curved shape)을 부여하는 단계; (e) 상기 만곡형(curved shape)이 부여된 솔리드 마이크로니들에 금속 증착하는 단계; (f) 상기 금속 증착된 솔리드 마이크로니들를 금속 도금하는 단계; (g) 상기 금속 도금된 솔리드 마이크로니들의 선단부를 베벨 절삭하는 단계; 및 (h) 상기 솔리드 마이크로니들를 제거하여 만곡형이 부여된 중공형 마이크로니들를 수득하는 단계를 포함하는 망막하 주사 또는 추출용 중공형 마이크로니들의 제조방법을 제공한다. 본 발명의 망막하 주사 또는 추출용 중공형 마이크로니들은 금속으로 이루어져 있어 공막 및 망막을 통과할 수 있는 강도 또는 힘이 있다. 본 발명의 망막하 주사 또는 추출용 중공형 마이크로니들은 길이 및 만곡앵글이 망막하의 적합한 위치, 예를 들어 시신경이 밀집한 망막하 중심부까지 도달 할 수 있어 약물을 전달하는 데 효과적이며 약물의 효과를 증진할 수 있다.

【발명의 명칭】
망막하 주사 또는 추출용 중공형 마이크로니들 및 망막하 주사기 【기술분야】
본 발명은 대한민국 보건복지부의 지원 하에서 과제번호 A102003에 의해 이루어진 것으로서, 상기 과제의 연구관리전문기관은 보건산업진흥원, 연구사업명은 "본건의료연구개발사업 미래 융합 의료기기 개발 /치료기기" , 연구과제명은 "초고횡비 마이크로니들 기술을 웅용한 안질환 맞춤형 약물" 주관기관은 연세대학교 산학협력단, 연구기간은 2010.05.01 ~ 2015.03.31이다.
본 특허출원은 2012년 04월 09일에 대한민국 특허청에 제출된 대한민국 특허출원 제 10-2012-0036554 호에 대하여 우선권을 주장하며, 상기 특허출원의 개시 사항은 본 명세서에 참조로서 삽입된다.
본 발명은 망막하 주사 또는 추출용 중공형 마이크로니들의 제조방법 및 망막하 주사기 또는 추출기에 관한 것이다.
【배경기술】
물체의 상이 맺히고 시세포의 대부분이 모여 있는 눈의 망막 중심부에 위치한 황반의 병변은 시력상실을 포함한 시력장애의 중요한 원인을 제공한다. 황반변성은 삼출성과 비삼출성으로 나뉘고 삼출성의 경우 적극적인 치료가 필요하다. 현재 변성이 일어난 부위의 경계를 명확히 알 수 있는 경우는 레이저광 웅고술, 광역학요법, 항체주사 및 초자체 (유리체) 절제술 등의 시술을 시행하고 있으나 아직까지 완전한 치료법은 존재하지 않는다.
최근 23 게이지 또는 25 게이지 니들 끝에 40 게이지 폴리머 류브를 연결하여 망막하 주사기 (MedOne 社의 Retinal cannula)를 이용한 삼출성 황반변성을 치료하는 방법이 소개되었다 (참조: 도 1). 그러나, 플라스틱 폴리머의 휘는 특성 때문에 주사부위를 정확히 침투할 수 없어 망막 손상을 유발 할 수 있고 니들과의 접착부분이 쉽게 떨어져 의료 사고를 일으킬 수 있는 문제점이 있다.
한편, 대한민국 특허 게 0431454호는 내부에 주사바늘의 직경보다 큰 가이드 홈이 형성된 가이드부; 이 가이드부의 후단부에 결합되고, 내측에 이동로가 형성되는 가이드 몸체; 및 가이드 몸체의 내부에서 주사바늘을 가이드 홈에서 전후진 이동시키는 구동부를 포함하는 망막하 주사기를 개시하고 있다.
미국특허 게 5,409,457호는 미리 바늘 끝을 직각으로 꺾어 놓고, 공막의 어느 지점을 눌러서 공막을 수직으로 뚫게끔 하는 구조를 개시하고 있다. 본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다. 【발명의 내용】
【해결하려는 과제】
본 발명자들은 종래의 망막하 주사기의 문제점, 특히 망막 손상의 유발, 낮은 강도에 의한 벤딩 되는 특성에 따른 정확하지 않은 침투, 니들과의 접착 부분이 쉽게 떨어지는 문제점 등을 해결하고자 예의 연구 노력하였다. 그 결과, 본 발명자들은 중공형 마이크로니들 기술을 이용하여 망막하 주사기에 최적화 된 중공형 마이크로니들을 제작하고, 이러한 망막하 주사용 중공형 마이크로니들이 상술한 종래의 문제점들을 해결할 수 있음을 확인함으로써, 본 발명을 완성하게 되었다.
따라서, 본 발명의 목적은 망막하 주사 또는 추출용 중공형 마이크로니들의 제조방법을 제공하는 데 있다.
본 발명의 다른 목적은 망막하 주사기 또는 추출기를 제공하는 데 있다. 본 발명의 다른 목적 및 이점은 하기의 발명의 상세한 청구범위 및 도면에 의해 보다 명확하게 된다.
【과제의 해결 수단】
본 발명의 일 양태에 따르면, 본 발명은 다음의 단계를 포함하는 망막하 주사 또는 추출용 중공형 마이크로니들의 제조방법을 제공한다:
(a) 기판 표면에 점성물질의 용액을 도포하는 단계;
(b) 상기 점성물질의 용액과 프레임을 접촉시키는 단계;
(C) 상기 접촉된 프레임과 기판이 이격되도톡 기판, 프레임 또는 기판과 프레임을 리프팅 하여 솔리드 마이크로니들를 제조하는 단계;
(d) 상기 단계 (c) 과정 중 또는 상기 단계 (c) 과정이 종료된 후 상기 솔리드 마이크로니들에 만곡형 (curved shape)을 부여하는 단계 ;
(e) 상기 만곡형 (curved shape)이 부여된 솔리드 마이크로니들에 금속 증착하는 단계;
(f) 상기 금속 증착된 솔리드 마이크로니들를 금속 도금하는 단계;
(g) 상기 금속 도금된 솔리드 마이크로니들의 선단부를 베벨 절삭하는 단계 ; 및
(h) 상기 솔리드 마이크로니들를 제거하여 만곡형이 부여된 중공형 마이크로니들를 수득하는 단계 . 본 발명자들은 종래의 망막하 주사기의 문제점 특히 망막 손상의 유발, 낮은 강도에 의한 벤딩 되는 특성에 따른 정확하지 않은 침투, 니들과의 접착 부분이 쉽게 떨어지는 문제점 등을 해결하고자 예의 연구 노력하였다. 그 결과, 본 발명자들은 증공형 마이크로니들 기술을 이용하여 망막하 주사기에 최적화 된 중공형 마이크로니들을 제작하고, 이러한 망막하 주사용 중공형 마이크로니들이 상술한 종래의 문제점들을 해결할 수 있음을 확인하였다. 본 발명의 방법을 각각의 단계에 따라 상세하게 설명하면 다음과 같다:
단계 (a): 기판 표면에 점성물질의 용액을 도포하는 단계
본 발명의 방법에 따르면 우선 중공형 마이크로니들의 몰드인 솔리드 마이크로니들를 제조하기 위하여 기판 표면에 점성물질의 용액을 도포한다. 중공형 마이크로니들의 몰드인 솔리드 마이크로니들를 제조하기
위하여 이용되는 물질은 점성물질이다. 본 명세서에서 용어 "점성물질" 은 일정 온도 이상에서 점성이 낮은 유체 형태를 갖지만 온도를 낮추어 유리화 온도에 접근시켰을 때 높은 점성을 가지는 물질을 의미한다. 본 발명에서 이용되는 점성물질에는 아크릴계 중합체 아마이드계 중합체, 아세틸계 중합체, 비닐계 중합체, 에폭시계 중합체, 실리콘계 중합체, 설폰 수지, 폴리카보네이트계 중합체 또는 이들의 공중합체를 포함하나 이에 쎄한되지 않고 당업계에서 통상적으로 사용되는 모든 점성물질이 사용될 수 있다. 바람직하게는 본 발명에서 이용되는 점성물질은 유체화된 경우에 점성을 갖는다. 이러한 점성은 점성물질의 종류, 농도 및 온도, 유기용매 등에 따라 다양하게 변화시킬 수 있으며 , 본 발명의 목적에 적합하게 조절할 수 있다. 보다 바람직하게는 본 발명의 점성물질은 유체화된 경우 200000 cSt(centistoke) 이하의 점성을 나타낸다.
점성물질의 유체화는 당업계에 공지된 다양한 방법을 통해 실시할 수 있다. 예를 들어, 점성물질이 액상 폴리머인 경우는 유체화 과정이 필요 없으며, 열가소성 수지인 경우 녹는점 이상의 온도에서 가열한 후 다시 온도를 낮춰서 유리화 온도에 접근시킴으로써 점성을 갖는다. 또한 적합한 유기 용매 (예컨대, 탄소수 1-4의 무수 또는 함수 저급 알코올, 아세톤, 에틸 아세테이트, 클로로포름 1,3-부틸렌글리콜, 핵산, 디에틸에테르 및 부틸아세테이트 등)에 폴리머 물질을 용해시켜 유체화시킬 수 있다.
본 명세서에서 용어 "도포" 는 대상 표면을 어떤 특정 물질의 일정한 두께의 층을 만드는 것을 의미한다. 표면을 제공하는 기판은 폴리머, 유기화학 물질, 금속, 세라믹, 반도체 등의 물질로 제조된 것이다.
본 발명의 바람직한 구현예에 따르면, 본 발명의 점성물질 용액의 도포 두께는 10-500 urn 범위에서 조절되며, 보다 바람직하게는 50-200 범위에서 조절되고, 가장 바람직하게는 75-165 범위에서 조절된다.
본 발명의 바람직한 구현예에 따르면, 상기 (a)단계의 점성물질은 유기용매에 의해 제거되는 고분자 화합물이다.
본 명세서에서 용어 "유기용매에 의해 제거되는 고분자 화합물" 은 분자량 5,000 이상의 천연 또는 합성 화합물로서 유기용매에 대한 용해성을 갖는 화합물을 말한다.
본 발명에서 이용되는 고분자 화합물은 중공형 마이크로니들 제작을
위한 금속 증착 및 도금 후에 제거가 용이하여야 하는 바, 본 발명자들은 금속 도금된 솔리드 마이크로 구조체 성분인 고분자 화합물을 유기용매로 용해시킴으로서 이를 제거하였다.
본 발명에서 이용되는 고분자 화합물은 보다 바람직하게는 AS(acrylonitrile styrene), 폴리 아마이드 폴리에틸렌, 폴리에스테르ᅳ 폴리아크릴, 폴리아세틸 스틸론, 테프론, 폴리염화비닐, 폴리우레탄, 나일론, 설폰 수지 또는 에폭시 폴리머이다ᅳ 가장 바람직하게는 본 발명의 고분자 화합물은 에폭시 폴리머이다.
본 발명에서 이용되는 유기 용매는 바람직하게는 벤젠 를투엔, 크실렌 (자일렌) , 핵산, 에테르, 아세톤, 알코을 및 아민을 포함하나, 이에 제한되지 않고 통상적으로 각각의 고분자 화합물의 용해에 이용되는 모든 극성 또는 비극성 용매가 사용될 수 있다. 예를 들어 고분자 화합물로서 에폭시 폴리머가 사용될 경우, NMP(N-methyl pyrrolidine)를 용매로 사용할 수 있다. 단계 (b): 점성물질의 용액과 프레임을 접촉시키는 단계
본 명세서에서 유체화 된 점성물질ᅳ 바람직하게는 고분자 화합물의 코팅 후 점성물질의 계면 위에 리프팅 프레임을 접촉시킨다. 본 발명의 바람직한 구현예에 따르면, 이 때 사용되는 리프팅 프레임의 직경은 변화가 가능하며 직경 1-1,000 urn, 가장 바람직하게는 10—500 im 범위에서 조절된다.
본 발명에서 이용되는 리프팅 프레임의 바람직한 예는 캐뉼러 형태의 스테인리스 프레임 및 통로가 있는 관형태의 프레임을 포함한다.
본 발명의 바람직한 구현예에 따르면, 본 발명에서 이용되는 프레임은 주사기바늘이다. 예를 들어, 실린지 및 주사기바늘로 이루어진 주사기에 있어세 상기 주사기바늘 상에 중공형 마이크로니들을 형성시키면 매우 효율적인 망막하 주사기가 제공될 수 있다.
보다 바람직하게는 본 발명에서 이용되는 프레임은 실린지 연결부가 장착된 주사기바늘이다.
이와 같이, 마이크로니들를 주사용바늘에 부착하여 일체형으로 제작하면, 망막하 주사기가 제공되는 이점이 있다.
상기 프레임으로서의 주사기바늘은 바람직하게는 23 게이지 이상, 보다 바람직하게는 23-34 게이지, 보다 더 바람직하게는 23-30 게이지 또는 23-27 게이지의 주사기바늘이다. 단계 (c): 접촉된 프레임과 기판이 이격되도록 기판, 프레임 또는 기판과 프레임을 리프팅 하여 솔리드 마이크로니들를 제조하는 단계
본 명세서에서 사용된 용어, "솔리드 마이크로니들" 는 중공의 형성 없이 일체형으로 제작된 마이크로니들 및 중공형 마이크로니들의 주형을 의미한다.
본 발명에서는 유체화 된 점성물질 (바람직하게는 고분자 물질)의 온도를 낮추어 유리화 온도 (Glass temperature)에 접근시키면서 점성을 증가시킨다.
본 발명의 바람직한 구현예에 따르면 망막하 주사에 적합한 증공형 마이크로니들을 제공하기 위하여, 유체화된 (예컨대, 가열된 유체)의 점성물질에 중공형 리프팅 프레임을 완전히 접촉한 후 빠른 속도 약 3000- 5000 m/s로 상향식 리프팅하여 솔리드 구조물을 제작하며, 제작되는 솔리드 구조물은 상온에서 급속히 유리화 시킨다.
단계 (c)에서의 점성은 최종적으로 제조되는 중공형 마이크로니들의 다양한 외형적 요소, 즉 유효길이, 내경, 외경 날카로움 및 종횡비 등에 영향을 미치며, 특히 솔리드 마이크로니들 및 중공형 마이크로니들의 유효길이를 변화시키는 변수로 작용한다. 단계 (C)에서 점성물질의 점성이 클수록 중공형 마이크로니들의 유효길이가 증가하게 된다.
본 발명의 바람직한 구현예에 따르면, 상기 점성물질의 온도는 유리화 온도 (glass transition temperature: Tg)보다 높고 130°C보다 낮은 범위에서 조절됨으로써 상기 점성물질의 점성이 조절된다.
본 명세서에서 용어 "유리화 온도 (glass transition temperature)" 는 점성을 된 유체 형태의 물질의 솔리드화가 이루어지는 온도를 말한다. 따라서 유리화 온도보다 낮은 온도에서는 이미 고형화 된 물질에 대한 리프팅 공정이 불가능하며, 온도가 너무 높으면 점성이 낮아져 역시 리프팅 공정이 불가능하므로 솔리드 마이크로니들를 제작할 수 없다. 본 발명의 바람직한 구현예에 따르면, 단계 (c)에서 조절되는
점성물질의 점성은 50-10,000 Poise 이고, 보다 바람직하게는 80-8,000 Poise이며, 보다 더 바람직하게는 100-6,500 Poise 이다.
본 명세서에서 용어 "이격" 이란 서로 접촉된 기질들 간의 거리를 증가시켜 사이를 벌려놓는 것을 말한다. 본 발명자들은 점성물질과 접촉한 프레임의 리프팅 (상향이동)에 의하여 솔리드 마이크로니들를 제작하였으나, 프레임을 고정시키고 기판을 하향이동 시키거나, 프레임과 기판을 동시에 상향 및 하향으로 이동시킴으로서 이격시키는 방법이 모두 가능하다 .
본 발명에 따르면, 점성을 지닌 폴리머의 리프팅 속도를 조절함으로써 최종적으로 제조되는 중공형 마이크로니들의 다양한 외형적 요소, 즉 유효길이, 내경, 외경, 날카로움 및 종횡비 등을 조절할 수 있으며, 특히 솔리드 마이크로니들의 유효길이, 증공형 마이크로니들의 유효길이를 조절 할 수 있다.
본 명세서의 용어 "리프팅 속도" 는 프레임 또는 기판의 상향 또는 하향이동 속도 뿐 아니라 프레임과 기판이 동시에 상향 및 하향으로 이동할 경우 이들 간의 멀어지는 상대속도를 포함하는 의미이다.

본 명세서에서 용어 "증착 (deposit ion)" 이란 물질의 기계적 강도를 높이기 위해 코팅시키고자 하는 물질을 물리적 방법 또는 화학적 방법으로 기화 또는 승화시켜서 원자 또는 분자 단위로 기판 표면에 웅고되도록 함으로써 피막을 형성시키는 것을 말한다. 본 발명의 증착은 당업계에서 통상적으로 이용되는 모든 물리적 증착 (Physical Vapor Deposition) 및 화학적 증착 (Chemical Vapor Deposition)이 사용될 수 있다.
본 발명의 바람직한 구현예에 따르면, 본 발명의 증착용 금속은 스테인레스강, 알루미늄 (A1), 크롬 (Cr), 니켈 (Ni), 금 (Au), 은 (Ag), 구리 (Cu), 티타늄 (Ti), 코발트 (Co) 또는 이들의 합금이다. 보다 바람직하게는, 은거울반웅 (tol lens react ion)을 이용하여 화학적으로 은 (Ag)을 증착시킨다.

증공형 마이크로니들 제조 기술들 (예컨대, 대한민국 특허 제 781702호 및 특허출원 제 2010-0066940호)은, 금속 도금 이전에 마이크로니들의 선단부를 마스킹 하는 과정을 필수적으로 포함한다. 본 발명은 이러한 과정을 실시하지 않으면서 제작시간 단축과 공정의 편의성을 증대한다.
종래의 기술들은 "도포 ― 리프팅 - 금속증착 ― 팁부분 마스킹 ― 도금 - 솔리드 구조물 제거 ― 베벨 절삭" 의 과정을 따르지만, 본 발명은
"도포 - 리프팅 - 금속증착 - 도금 - 베벨 절삭 - 솔리드 구조물 제거" 의 과정에 의해 망막하 주사 또는 추출용 중공형 마이크로니들를 제공한다. 본 발명에서 이용되는 도금 두께는 바람직하게는 5-100 urn, 보다 바람직하게는 10-50 이다.
본 발명에서 이용되는 도금 재료는 예를 들어 니켈, 스테인리스강, 알루미늄, 크름, 코발트계 합금, 티타늄 및 그 이들의 합금을 포함하나, 이에 제한되지 않고 생체 적용 가능한 금속으로서 독성이나 발암성이 없으며, 인체 거부반웅이 없으며, 인장강도와 탄성률, 내마모성 등 기계적 성질이 양호하고, 인체 내 부식 환경에 견딜 수 있는 내부식성을 갖추는 금속으로서 당업계에 알려진 모든 금속이 사용될 수 있다.

현재까지 개발된 마이크로니들의 길이는 최대 2 mm에 불과하다. 본 발명은 이러한 한계점을 극복하여 망막하 주사에 적합한 유효길이를 제공한다. 본 발명은 안구 외벽 중 가장 강도가 높은 공막과 망막을 투과할 수 있고, 망막하 부위에 약물을 주입하여 치료가 가능한 수준의 길이를 갖는다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다. 실시예
실시예 1: 망막하 주사 또는 추출용 마이크로니들 디바이스의 제작 I
14,000 cSt 의 점도를 갖는 SU-8 2050 포토레지스트 (Microchem 사로부터 구입)를 사용하여 솔리드 마이크로니들를 제작하였다. 1.5 X 1.5 cm 의 커버글라스 위에 1000 RPM 으로 SU-8 2050 을 도포하여 두께가 약 160 를 유지시켰다. 상기 커버글라스를 핫플레이트 위에 120°C로 약 30 분 동안 가열하여 SU-8 2050 이 유동성을 유지하도록 하였다. 이어, 끝이 평평하고 실린지연결부가 부착된 23 게이지의 주사바늘을 접촉하여 수직으로 들어 을려 직경이 50 μη\, 길이가 5-10 画 인 솔리드

14,000 cSt 의 점도를 갖는 SU-8 2050 포토레지스트 (Microchem 사로부터 구입)를 사용하여 솔리드 마이크로니들를 제작하였다. 1.5 X 1.5 cm 의 커버글라스 위에 1000 RPM 으로 SU-8 2050 을 도포하여 두께가 약 160 를 유지시켰다. 상기 커버글라스를 핫플레이트 위에 120°C로 약 1 시간 동안 가열하여 SU-8 2050 이 유동성을 유지하도록 하였다. 이어, 끝이 평평한 23 게이지의 주사바늘을 접촉하여 수직으로 들어 올려 직경이 20-60 , 길이가 5-10 隱 인 솔리드 구조체를 제작하였다. 이 경우, 상기 커버글라스 즉 기판의 온도를 90° C (폴리머의 부착력 : 1Nᅳ 점도 : 100 Poise) 와 60° C (폴리머의 부착적 : 2N, 점도 : 6ᅳ 500 Poise)까지 천천히 낮추는 동안 리프팅 프레임 (주사바늘)을 10 / /s 의 속도로 5 분간 각각 리프팅 하였다. 제작된 솔리드 구조체 위에 틀렌스 시약 (Tollen' s reagent)을 사용하여 은도금을 시행한 후, 니켈 전해도금을 실시하였다. 니켈 전해도금은 1 A/dm2 당 0.206 m/min 으로 75 분간 처리하여 도금된 금속 두께가 20 가 되도록 하였다. 파이버 레이저를 이용하여 도금된 구조물 끝부분에 150 의 베벨을 주어 절삭하고 SU-8
리무버 (Microchem 사로부터 구입) 또는 아세톤을 사용하여 솔리드 구조물을 제거하여 중공형 마이크로니들 형태의 망막하 주사 또는 추출용 마이크로니들 디바이스를 제작하였다.
제작된 망막하 주사 또는 추출용 마이크로니들은 상단부 외경 110 / , 내경 40 , 하단부 직경 350 , 길이 3-6画의 중공형 마이크로니들이다. 제작된 중공형 마이크로니들의 경도는 1-2 N 값을 나타내며, 이는 망막을 천공 할 수 있는 최소의 강도보다 큰 값이다. 실시예 3: 망막하 주사 또는 추출용 마이크로니들 디바이스의 제작 m

제작된 솔리드 구조체 위에 를렌스 시약 (Tollen' s reagent)을 사용하여 은도금을 시행한 후, 니켈 전해도금을 실시하였다. 니켈 전해도금은 1 A/dm2 당 0.206 mi/min 으로 75 분간 처리하여 도금된 금속 두께가 20 가 되도톡 하였다. 파이버 레이저를 이용하여 도금된 구조물 끝부분에 150 의 베벨을 주어 절삭하고 SU-8 리무버 (Microchem 사로부터 구입) 또는 아세톤을 사용하여 솔리드 구조물을 제거하여 중공형 마이크로니들 형태의 망막하 주사 또는 추출용 마이크로니들 디바이스를 제작하였다.
제작된 망막하 주사 또는 추출용 마이크로니들은 상단부 외경 120 , 내경 50 , 하단부 직경 350 urn, 길이 5-8隱의 중공형 마이크로니들이다. 제작된 중공형 마이크로니들의 경도는 1-2 N 값을 나타내며, 이는 망막을 천공 할 수 있는 최소의 강도보다 큰 값이다.
상기 실시예에서, 폴리머의 온도 및 리프팅 속도 등을 조절하여 다양한 디멘견 특성 (내경 , 직경 및 길이)을 갖는 망막하 주사 또는 추출용 마이크로니들을 제작하였다. 실시예 4: 망막하 주사 또는 추출용 마이크로니들을 이용한 망막 박리 치료 제작된 망막하 주사 또는 추출용 마이크로니들의 작동성 (workability)이 우수한 지 여부를 확인하기 위하여, 상기 제작된 망막하 주사 또는 추출용 마이크로니들 증에서 실시예 1 의 마이크로니들을 이용하여 망막박리 (Retina Detachment)를 실시하였다.
동결상태의 돼지 눈을 2 시간 동안 해동시킨 후 돼지 눈을 고정틀에 고정시켰다. 동결상태에서 해동된 돼지 눈은 망막박리가 심하게 일어나 있으므로 이를 이용하여 실험하였다. 돼지 눈 1 시 방향과 7 시 방향에 수압보조장치 (Water supply)와 광원을 연결하였다. 이어, 유리체를 제거하고 본 발명의 망막하 주사 /추출용 중공형 마이크로니들을 사용하여 망막에 천공한 후 추출작업을 시행하였다. 천공하기 전 망막상태는 막 대부분이 맥락막에서 떨어져서 유리체강 (Vitreous)을 채우고 있었다. 본 발명의 중공형 마이크로니들을 사용하여 천공하고 LEICA 기기를 사용하여 600 inmHg 의 압력으로 추출을 시도하였으며, 추출을 시작하고 수초 내에 망막박리가 사라지고 망막과 맥락막이 붙어 있음을 확인할 수 있었다. 따라서 ᅳ ,본 증공형 마이크로니들은 망막하 주사 또는 추출용으로 뿐만 아니라 망막손상을 최소화 하면서 망막과 맥락막 사이 물질을 효을적으로 제거할 수 있음을 알 수 있다. 이상으로 본 발명의 특정한 부분을 상세히 기술하였는바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구,체적인 기술은 단지 바람직한 구현예일 뿐이몌 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
Claims
【특허청구범위】
【청구항 11
다음의 단계를 포함하는 망막하 주사 또는 추출용 중공형 마이크로니들의 제조방법 :
(a) 기판 표면에 점성물질의 용액을 도포하는 단계 ;
(b) 상기 점성물질의 용액과 프레임을 접촉시키는 단계;
(c) 상기 접촉된 프레임과 기판이 이격되도록 기판, 프레임 또는 기판과 프레임을 리프팅 하여 솔리드 마이크로니들를 제조하는 단계;
(d) 상기 단계 (c) 과정 중 또는 상기 단계 (c) 과정이 종료된 후 상기 솔리드 마이크로니들에 만곡형 (curved shape)을 부여하는 단계;
(e) 상기 만곡형 (curved shape)이 부여된 솔리드 마이크로니들에 금속 증착하는 단계 ;
(0 상기 금속 증착된 솔리드 마이크로니들를 금속 도금하는 단계;
(g) 상기 금속 도금된 솔리드 마이크로니들의 선단부를 베벨 절삭하는 단계; 및
(h) 상기 솔리드 마이크로니들를 제거하여 만곡형이 부여된 중공형 마이크로니들를 수득하는 단계.

제 1 항에 있어서, 상기 프레임은 주사기바늘인 것을 특징으로 하는 제조방법.

제 2 항에 있어서, 상기 주사기바늘은 23 게이지 이상의 주사기바늘인 것을 특징으로 하는 제조방법 .
【청구항 4】
제 1 항에 있어서, 상기 점성물질의 온도는 유리화 온도 (glass temperature)보다 높고 130°C보다 낮은 범위에서 조절됨으로써 상기 점성물질의 점성이 조절되는 것을 특징으로 하는 제조방법.

제 1 항에 있어서, 상기 도금 금속은 스테인레스강, 알루미늄 (A1), 크름 (Cr), 니켈 (Ni), 금 (Au), 은 (Ag), 구리 (Cu), 티타늄 (Ti), 코발트 (Co) 또는 이들의 합금인 것을 특징으로 하는 제조방법.

제 1 항에 있어서 상기 만곡형이 부여된 상기 중공형 마이크로니들는 10ο-70ο의 만곡앵글을 가지는 것을 특징으로 하는 제조방법.

제 6 항에 있어서, 상기 만곡앵글은 40ο-60ο인 것을 특징으로 하는 제조방법.

제 1 항에 있어서, 상기 베벨 절삭은 상기 중공형 마이크로니들의 선단부의 베벨앵글을 5ο-50ο로 하는 것을 특징으로 하는 제조방법.

제 8 항에 있어서, 상기 중공형 마이크로니들의 선단부의 베벨앵글은 50-150인 것을 특징으로 하는 제조방법.
