バソプレッシン受容体(AVPR)として、V1a、V1bおよびV2の三つの受容体が知られており、V1a受容体はバソプレッシンの不安および記憶増強作用等の中枢神経系効果、平滑筋の収縮および肝臓グリコーゲン分解効果を媒介し、V1b受容体はバソプレッシンの下垂体前葉効果を媒介することが知られている。また、V2受容体は、腎臓のみに見出され、細胞内アデニル酸シクラーゼの刺激を解するバソプレッシンの抗利尿作用に影響していると考えられている(特許文献2)
上記の如き、バソプレッシン受容体を介したバソプレッシンの多用な作用から、バソプレッシン受容体アンタゴニスト(拮抗薬)は、不安障害、不安神経症、睡眠障害、うつ病、健忘症、高血圧症などの予防または治療薬として期待されている。
オキシトシン受容体並びにバソプレッシンのV1a、V1bおよびV2受容体は、いずれも、G-タンパク質結合受容体として知られる膜横断受容体のスーパーファミリーを構成している。これら受容体は、上述の如く、密接に関係する同様の薬理効果に関係しているので、オキシトシン受容体アゴニスト(作動薬)および/またはバソプレッシン受容体アンタゴニスト(拮抗薬)は、不安障害、不安神経症、睡眠障害、うつ病、健忘症、肥満症、糖尿病(特に、II型糖尿病)、アテローム性動脈硬化症、薬物依存症などの予防または治療薬として期待できる。
ヒトラクトフェリンのトリプシン消化物から、摘出モルモット回腸に対する収縮活性を指標として単離されたペプチドに、Cys-Phe-Gln-Trp-Gln-Arg(lactomedin 2)がある。該ペプチドは、ヒトラクトフェリンの第20~25残基に由来するペプチドであり、オキシトシンおよびバソプレッシンと構造上のホモロジーを共有し、モルモット回腸収縮活性が知られている。

本発明は、オキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤、オキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤を含んでなる抗不安剤、または、不安障害、不安神経症、睡眠障害、うつ病、健忘症、肥満症、糖尿病(特に、II型糖尿病)、アテローム性動脈硬化症もしくは薬物依存症の予防または治療剤、オキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤を含んでなる飲食品用組成物、または、該飲食品用組成物を含んでなる飲食品を提供しようとするものである。
本発明者らは、上記課題を解決するため鋭意研究した結果、lactomedin 2およびその誘導体に、オキシトシン受容体アゴニストおよび/またはバソプレッシン受容体アンタゴニストとしての作用があることを見出し、さらに検討を重ねて、本発明を完成するに至った。

本発明のペプチド(1)は、1つ以上の不斉炭素原子を有するため、ラセミ体、ラセミ体混合物、個々のエナンチオマー(鏡像異性体)またはジアステレオマーとして存在し得る。本発明のペプチド(1)は、このような異性体型の全ておよびそれらの混合物をも含むものであるが、特に、L型のアミノ酸がペプチド結合したものが好ましい。
ジアステレオマーの分離は、分別晶出、クロマトグラフィーまたはHPLC等により、通常の方法で達成し得る。また、単独の立体異性体は、必要に応じて、光学的に純粋な対応する中間体から製造するか、または適当なキラル支持体を用い、対応するラセミ体を分割する(例えば、HPLC)か、あるいは対応するラセミ体と適当な光学的に活性な酸または塩基との反応によって形成されるジアステレオマー性の塩を分別晶出によって、製造できる。あるいは、エナンチオマーの混合物は、適当なキラル化合物と化学反応させて新規な共有結合種を形成することによって分離することができる。
本明細書中、「薬理的に許容しうる誘導体」とは、本発明に係るペプチド(1)の薬理的に許容しうる塩、溶媒和物またはプロドラッグ等(例えばエステル)を意味し、これらは患者に投与されたとき、本発明に係るペプチド(1)を与えることができるものをいう。そのような誘導体は、当業者であれば過度の実験を行なうことなく得ることができる(例えば、バーガー、医薬品化学および薬物発見(Burger's Medicinal Chemistry and Drug Discovery)、第5版、第1巻「原則と実務(Principles and Practice)」参照)。好ましい薬理的に許容しうる誘導体は、塩、溶媒和物、またはエステルである。
薬理的に許容しうる塩は、通常、必要に応じて所望の酸または塩基を用いることにより簡単に製造することができる。塩は溶液から析出させて濾取するか、溶媒を留去することにより回収することができる。適当な付加塩は非毒性の塩を形成する酸を用いて形成される。そのような塩として、塩酸塩、臭化水素酸塩、ヨウ化水素酸塩、硫酸塩、重硫酸塩、硝酸塩、リン酸塩、リン酸水素塩、酢酸塩、マレイン酸塩、リンゴ酸塩、フマル酸塩、乳酸塩、酒石酸塩、クエン酸塩、ギ酸塩、グルコン酸塩、コハク酸塩、ピルビン酸塩、蓚酸塩、オキサロ酢酸塩、トリフルオロ酢酸塩、サッカラート塩、安息香酸塩、メタンスルホン酸塩、エタンスルホン酸塩、ベンゼンスルホン酸塩およびp-トルエンスルホン酸塩が例示される。薬理的に許容しうる塩基の塩として、アンモニウム塩、ナトリウムおよびカリウム等のアルカリ金属塩、カルシウムおよびマグネシウム等のアルカリ土類金属塩ならびに第1、第2および第3アミン(例えば、イソプロピルアミン、ジエチルアミン、エタノールアミン、トリメチルアミン、ジシクロへキシルアミンおよびN-メチル-D-グルカミン)の塩を含む有機塩基との塩が例示される。
多数の有機化合物が、反応系の溶媒と複合体を形成して析出または晶出し得ることは周知である。これらの複合体は「溶媒和物」として知られている。例えば、水との複合体は「水和物」である。本発明に係るペプチド(1)の溶媒和物も本発明の範囲内である。
本発明において、「プロドラッグ」とは、患者に投与した際に、体内で、ペプチド(1)を放出するキャリアである。通常、プロドラッグは常套手段で、またはin vivoで修飾部分が切断されて親化合物が生じるよう、官能基を修飾することにより製造される(例えば、文献[T. ヒグチおよびV. ステラ、新規デリバリーシステムとしてのプロドラッグ(Prodrugs as Novel Delivery Systems)、“薬物設計における生体可逆的担体(Bioreversible Carriers in Drug Design)”、エドワード B. ロッシュ編、米国薬学会およびペルガモンプレス(American Pharmaceutical Association and Pergamon Press), A.C.S. 本発明においては、特に、プロドラッグの例として、ペプチド(1)のヒドロキシまたはアミンなどの官能基が修飾されたものを挙げることができ、該プロドラッグは患者に投与されると修飾部分が切断されてヒドロキシまたはアミンが生ずる。
(製法)
本発明において、ペプチド(1-a)を含むペプチド(1)は、ペプチド合成の常法、例えば、固相ペプチド合成(Boc(tert-ブトキシカルボニル基)、Fmoc(フルオレニルメトキシカルボニル基)などを用いる)または溶液相合成により合成することができる。一般的なBocおよびFmoc方法は、広く用いられており、以下の参考文献にも記載されている:Merrifield,J.A.Chem.Soc.,88:2149(1963);Bodanszky and Bodanszky,The Practice of Peptide Synthesis,Springer-Verlag,Berlin,7-161(1994);Stewart,Solid Phese Peptide Synthesis,Pierce Chemical,Rockford,(1984)。
本発明において、ペプチド(1-a)を含むペプチド(1)は、ペプチド合成の常法、例えば、固相ペプチド合成(Boc(tert-ブトキシカルボニル基)、Fmoc(フルオレニルメトキシカルボニル基)などを用いる)または溶液相合成により合成することができる。一般的なBocおよびFmoc方法は、広く用いられており、以下の参考文献にも記載されている:Merrifield,J.A.Chem.Soc.,88:2149(1963);Bodanszky and Bodanszky,The Practice of Peptide Synthesis,Springer-Verlag,Berlin,7-161(1994);Stewart,Solid Phese Peptide Synthesis,Pierce Chemical,Rockford,(1984)。
例えば、Fmoc固相合成方法は、固体樹脂に結合したC末端アミノ酸へのアミノ酸の段階的付加によるものである。Fmoc適合樹脂としては、Wang樹脂、HPMA-PEGA樹脂、Rink酸樹脂またはヒドロキシエチル-フォトリンカー樹脂などを好適に使用できる。ペプチド鎖のC末端は高分子樹脂に共有結合しており、保護基のついたαアミノ酸がカップリング試薬により段階的に付加される。好ましいαアミノ保護基はFmoc基であり、カップリング条件に対して安定であり、穏和な条件(通常、DMF/20%ピペリジン条件)下で容易に除去できる。また、αアミノ基以外の官能基も適宜保護される。例えば、アルギニンのグアニジノ基は、Mtr(4-メトキシ-2,3,6-トリメチルベンゼンスルホニル基)で保護することができる。反応溶媒は、ジメチルホルムアミド(DMF)、N-メチル-2-ピロリドン(NMP)、ジクロロメタン(DCM)、MeOHおよびEtOHが好ましいが、これらに限定されない。カップリング剤の例としては、ジシクロヘキシルカルボジイミド(DCC)、ジイソプロピルカルボジイミド(DIC)、HATU、HBTUがある。N末端保護基の切断は、DMF中10~100%ピペリジン中で、0~40℃、好ましくは室温で実施される。合成の最後で、最終的なFmoc保護基を上記のN末端切断処置により除去する。樹脂上に残ったペプチドは、樹脂を酸性条件下で処理することにより切断される。このとき、酸に敏感な側鎖保護基も、同時にペプチドから切断される。
(医薬品製剤)
かくして得られるペプチド(1)は、優れたオキシトシン受容体作動活性および/またはバソプレッシン受容体拮抗活性を有するので、これを活性成分として配合することにより、オキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤として、不安障害、不安神経症、睡眠障害、うつ病、健忘症、肥満症、糖尿病(特に、II型糖尿病)、アテローム性動脈硬化症もしくは薬物依存症の予防または治療に用いることができる。
かくして得られるペプチド(1)は、優れたオキシトシン受容体作動活性および/またはバソプレッシン受容体拮抗活性を有するので、これを活性成分として配合することにより、オキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤として、不安障害、不安神経症、睡眠障害、うつ病、健忘症、肥満症、糖尿病(特に、II型糖尿病)、アテローム性動脈硬化症もしくは薬物依存症の予防または治療に用いることができる。
本発明のオキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤の投与経路は、経口投与または非経口投与のいずれであってもよい。その剤形は、投与経路に応じて適宜選択することができ、例えば、注射液剤、輸液剤、散剤、顆粒剤、錠剤、カプセル剤、丸剤、腸溶剤、トローチ剤、内用液剤、懸濁剤、乳剤、シロップ剤、外用液剤、湿布剤、点鼻剤、点耳剤、点眼剤、吸入剤、軟膏剤、ローション剤、坐剤、経腸栄養剤などを挙げることができる。これは、症状に応じてそれぞれ単独でまたは組み合わせて用いることができる。これらの製剤には、必要に応じて、賦形剤、結合剤、防腐剤、酸化安定剤、崩壊剤、滑沢剤、矯味剤などの、この分野において通常用いられる補助剤が使用される。
本発明のオキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤の投与量は、投与の目的、投与対象の状況(性別、年齢、体重など)、疾患の程度などに応じて異なるが、通常、成人に対して、1日あたり、経口投与の場合、0.1mg/kg~100mg/kg、好ましくは0.3mg/kg~30mg/kg、さらに好ましくは1mg/kg~10mg/kgであり、一方、非経口投与(例えば、静脈内投与)の場合、0.003mg/kg~3mg/kg、好ましくは0.01mg/kg~1mg/kg、さらに好ましくは0.03mg/kg~0.3mg/kgである。
(医薬部外品、化粧品)
本発明のオキシトシン受容体作動および/またはバソプレッシン受容体拮抗剤は、上記のような医薬品としてだけでなく、医薬部外品、化粧品などとしても用いることができる。医薬部外品または化粧品として用いる場合、必要に応じて、医薬部外品または化粧品などの技術分野で通常用いられている種々の補助剤とともに用いられ得る。また、溶液状、懸濁液状、シロップ状、顆粒状、クリーム状、ペースト状、ゼリー状などの所望の形状で、あるいは必要に応じて成形して使用してもよい。これらに含まれる本発明の活性成分の量は、特に限定されず、使用目的、使用形態に応じて適宜選択することができる。

本発明の飲食品用組成物は、オキシトシン受容体作動活性および/またはバソプレッシン受容体拮抗活性に基づき、上述の如き薬理効果を奏するので、該薬理効果を備えた飲食品、例えば、健康食品、健康飲料、特定保健用食品、機能性食品、栄養補助食品、その他ヒト以外の動物に対する飼料などに用いることもできる。
本発明の飲食品用組成物を飲食物に用いる場合、必要に応じて、例えば、甘味料、香辛料、調味料、防腐剤、保存料、殺菌剤、酸化防止剤などの食品に通常用いられる添加剤を配合することができる。また、固形状、粉末状、顆粒状、溶液状、懸濁液状、シロップ状、クリーム状、ペースト状、ゼリー状などの所望の形状で、あるいは必要に応じて成形して使用してもよい。これらに含まれる本発明の活性成分の量は、特に限定されず、使用目的、使用形態に応じて適宜選択することができる。
以下、実施例をあげて本発明を具体的に説明するが、本発明はこれらの実施例によって限定されるものではない。
1.ペプチドの調製
[合成例1](ペプチド1(Cys-Phe-Gln-Trp-Gln-Arg)の合成)
市販のFmoc-Arg(Mtr)-Alko樹脂(置換率: 0.5meq/g、渡辺化学工業(株)製)0.6gをPS3型ペプチド合成機(Protein Technologies社製)の反応槽に分取し、以下のようにペプチドの合成を行った。
まず、上記の樹脂を反応容器に入れて、1mmolのFmoc-Glnと、さらに活性化剤として、1mmolのHBTUを、10mLの0.4M N-メチルモルフォリンを含むジメチルフォルムアミドに溶解したものを反応槽に加え、室温にて20分撹拌反応させた。
得られた樹脂に20容量%ピペリジンを含むジメチルフォルムアミド20mLを加えて、該樹脂からFmoc基を除去し、次いで上記のFmoc-Glnをカップリングさせた方法と同様に、C末端から順次Fmocアミノ酸をカップルさせて、Cys(tBu)-Phe-Gln-Trp-Gln-Arg(Mtr)-Alko樹脂を得た。該樹脂を10mLの脱保護液(82容量%トリフルオロ酢酸、5容量%チオアニソール、3容量%エタンジチオール、2容量%エチルメチルスルフィド、3容量%フェノール、5容量%水の混合液)中で室温にて4時間撹拌し、各アミノ酸の保護基を除去するとともに、ペプチドを樹脂から遊離させた。
該混液に、40mLの冷エーテルを添加し、ペプチドを沈殿させ、さらに冷エーテルにて3回洗浄して粗ペプチドを得た。これをODSカラム(Cosmosil 5C18-ARII、20×250mm)による逆相クロマトグラフィーにより0.1重量%トリフルオロ酢酸を含むアセトニトリルの直線的濃度勾配にて展開、精製し、標記ペプチド1を得た。なお、本品をプロテインシーケンサー(アプライド バイオシステムズ社製「492型」)により分析して、上記の配列であることを確認した。

(試験の概要)
本試験で用いる高架十字迷路(Eleveted plus maze:EPM)は、2つのオープンアーム(24cm×5cm×0.5cm)と2つのクローズドアーム(24cm×5cm×13cm)からなり、それらのアームは床から50cm高くなった中央プラットフォームと結合している。高い位置にあるにも関わらず、クローズドアームの周りには囲いがあるために、マウスは安全に歩行することができる。一方オープンアームの周囲は開放されていて囲いがないために、オープンアームを歩行するマウスは高い位置から転落するという不安感を感じる。そのために、マウスがオープンアームにいる時間が長いほどマウスの不安感は緩和されており、抗不安活性の指標となる。

(試験方法)
本試験においては、オープンアームの一つに面している中央プラットフォーム上にマウスを置いて、試験を開始した。5分の試験時間の間、オープンアーム内で過ごした累積時間を記録した。オープンアーム内で過ごした時間のパーセンテージを不安の指標として計算した。

[試験1](脳室内投与)
高架十字路試験開始の20分前に、検体1(ペプチド1)を人工脳脊髄液(以下の組成からなる水溶液:124mM NaCl、3mM KCl、26mM NaHCO3、2mM CaCl2/2H2O、1mM MgSO4/7H2O、1.25mM KH2PO4、10mM D-Glucose;「aCSF溶液」という。)に溶解した液(4μL)を、ddYマウス(雄性、4週齢、日本エスエルシー(株);n=6)に投与した(投与量は、検体1の量に換算して0.1nmolに相当する量)。かかる投与群を検体投与群とした。
コントロール群(n=6)には、aCSF溶液を、同様にして投与した。
また、検体1の抗不安活性が、オキシトシン受容体(OTR)を介してのものであることを明らかにするため、OTR拮抗薬併用群(n=6)として、検体1(0.1nmolol)に加え、その10倍量のOTR拮抗薬(1nmol)を併せて投与し、検体1による抗不安活性が抑制されるかどうかを観察した。
さらに、OTR拮抗薬投与群(n=6)として、OTR拮抗薬(1nmol)のみを投与した。
各群について、上記試験方法に従い、高架十字迷路試験を行い、マウスがオープンアームにいる時間の割合(%)を算出し、各群毎に、平均値(mean)および標準誤差を算出した。結果を表1に示す。

さらに、OTR拮抗薬投与群(n=6)として、OTR拮抗薬(1nmol)のみを投与した。
各群について、上記試験方法に従い、高架十字迷路試験を行い、マウスがオープンアームにいる時間の割合(%)を算出し、各群毎に、平均値(mean)および標準誤差を算出した。結果を表1に示す。
