本発明の「R5」として、好適には、水素原子、塩素原子、メチル基及びメトキシ基であり、更に好適には、水素原子である。
また、本発明では、一般式(I)を有する化合物を(1)酸処理及び(2)アルカリ処理の順で処理することによって得られた一般式(II)を有する化合物を、更に(3)アルカリ処理することによって一般式(V)を有する化合物を製造することができる。
反応に使用される酸は、通常のアセタール基の脱保護に用いられる酸であれば特に限定はないが、例えば、(濃)塩酸、臭化水素酸、硫酸、過塩素酸、リン酸のような無機酸;酢酸、ギ酸、シュウ酸、メタンスルホン酸、p-トルエンスルホン酸、カンファースルホン酸、トリフルオロ酢酸、トリフルオロメタンスルホン酸のような有機酸等のブレンステッド酸;塩化亜鉛、四塩化スズ、ボロントリクロリド、ボロントリフルオリド、ボロントリブロミドのようなルイス酸;又は酸性イオン交換樹脂であり、好適には、無機酸(特に好適には、濃塩酸)である。
反応に使用される不活性溶媒としては、本反応に不活性なものであれば特に限定はないが、例えば、ジエチルエーテル、ジイソプロピルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン、ジエチレングリコールジメチルエーテルのようなエ-テル類;メタノ-ル、エタノ-ル、n-プロパノ-ル、イソプロパノ-ル、n-ブタノ-ル、イソブタノ-ル、t-ブタノ-ル、ジエチレングリコールのようなアルコ-ル類;水であり、好適には、アルコール類(最も好適には、メタノール)、水、及び水と上記有機溶媒との混合溶媒であり、メタノールと水との混合溶媒が特に好ましい。
反応温度は、通常、0~100℃(好適には、20~80℃)である。
反応時間は、通常、30分~48時間(好適には、1~10時間)である。
(2)アルカリ処理工程
反応に使用される塩基としては、通常塩基として使用されるもので反応を阻害しないものであれば特に限定はないが、例えば、炭酸リチウム、炭酸ナトリウム、炭酸カリウムのようなアルカリ金属炭酸塩類;水酸化リチウム、水酸化ナトリウム、水酸化カリウムのようなアルカリ金属水酸化物類;リチウムメトキシド、ナトリウムメトキシド、ナトリウムエトキシド、カリウム-t-ブトキシドのような金属アルコキシド類;又はアンモニア水、濃アンモニア-メタノ-ルのようなアンモニア類であり、好適には、アルカリ金属水酸化物類であり、更に好適には、水酸化ナトリウム(例えば、25%水酸化ナトリウム水溶液)である。
反応に使用される不活性溶媒としては、本反応に不活性なものであれば特に限定はないが、例えば、ジエチルエーテル、ジイソプロピルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン、ジエチレングリコールジメチルエーテルのようなエ-テル類;メタノ-ル、エタノ-ル、n-プロパノ-ル、イソプロパノ-ル、n-ブタノ-ル、イソブタノ-ル、t-ブタノ-ル、ジエチレングリコールのようなアルコ-ル類;水であり、好適には、アルコール類(最も好適には、メタノール)、水、及び水と上記有機溶媒との混合溶媒であり、メタノールと水との混合溶媒が特に好ましい。
反応温度は、原料化合物、用いられる試薬、溶媒の種類等によって異なるが、通常、0~100℃(好適には、20~80℃)である。
反応時間は、通常、5分~10時間(好適には、30分~5時間)である。
(3)アルカリ処理工程
反応に使用される塩基としては、通常塩基として使用されるもので反応を阻害しないものであれば特に限定はないが、例えば、炭酸リチウム、炭酸ナトリウム、炭酸カリウムのようなアルカリ金属炭酸塩類;水酸化リチウム、水酸化ナトリウム、水酸化カリウムのようなアルカリ金属水酸化物類;リチウムメトキシド、ナトリウムメトキシド、ナトリウムエトキシド、カリウム-t-ブトキシドのような金属アルコキシド類;又はアンモニア水、濃アンモニア-メタノ-ルのようなアンモニア類であり、好適には、アルカリ金属水酸化物類であり、更に好適には、水酸化ナトリウム(例えば、25%水酸化ナトリウム水溶液)である。
反応に使用される不活性溶媒としては、本反応に不活性なものであれば特に限定はないが、例えば、ジエチルエーテル、ジイソプロピルエーテル、テトラヒドロフラン、ジオキサン、ジメトキシエタン、ジエチレングリコールジメチルエーテルのようなエ-テル類;メタノ-ル、エタノ-ル、n-プロパノ-ル、イソプロパノ-ル、n-ブタノ-ル、イソブタノ-ル、t-ブタノ-ル、ジエチレングリコールのようなアルコ-ル類;水であり、好適には、アルコール類(特に好適には、プロパノール又はn-ブタノ-ル)、水及び水と上記有機溶媒との混合溶媒であり、好適には、n-プロパノ-ル又はn-ブタノ-ルと水との混合溶媒である。
反応温度は、通常、0~150℃(好適には、60~120℃)である。
反応時間は、通常、30分~20時間(好適には、1~10時間)である。

(1-1)4-(4-メチルフェニル)ブタン酸クロリドの調製
(1-2)Compound(2)を原料として用いて、Compound(3)を製造する工程

13C NMR (CDCl3, 100MHz):20.9,22.2, 22.3, 26.5, 27.0, 32.9, 34.9, 36.4, 38.4, 38.4, 52.0, 63.7, 77.5, 97.2,106.6, 118.9, 128.3, 128.9, 130.7, 135.2, 138.8, 141.9, 156.4, 190.7
MS (FAB):m/z=469 [M+H]+
(1-3)Compound(3)を原料として用いて、Compound(1)を製造する工程
Compound(3)(30g)にメタノール(360mL)を加え、45℃に加温して溶解させた後、濃塩酸(2.6mL)を加えた。同温度にて3時間攪拌した後、25%水酸化ナトリウム水溶液(46.7mL)を加えて反応を停止した。室温まで冷却した後、5倍量(150mL)になるまで減圧濃縮し、n-プロパノール(300mL)を加えて再度5倍量(150mL)になるまで減圧濃縮し、Compound(4)とCompound(5)との混合物のn-プロパノール溶液を得た。Compound(4)とCompound(5)との混合物のn-プロパノール溶液は精製することなく次工程に使用した。
(1-3-2)脱カルボニル化工程
前工程で得たCompound(4)とCompound(5)との混合物のn-プロパノール溶液に、7倍量(210mL)になるまでn-プロパノールを加え、さらに水道水(30mL)および25%水酸化ナトリウム水溶液(51.2mL)を加え、還流するまで加熱した。還流下で7時間攪拌した後、室温まで冷却し、トルエン(300mL)と水道水(300mL)を加えて分液した。得られた有機層を5倍量(150mL)になるまで減圧濃縮し、トルエン(300mL)を加えて再度5倍量(150mL)になるまで減圧濃縮した。濃縮液に10倍量(300mL)になるまでトルエンを加え、さらにMCH(300mL)を加えて室温で30分間攪拌した後、5℃以下に冷却した。同温度で30分間攪拌した後、析出した結晶をろ過し、冷トルエンとMCHの混合溶媒(混合比:1/2)で洗浄した。結晶を40℃にて10時間以上真空乾燥し、Compound(1)の粗結晶(20.4g,収率93.2%)を得た。
(1-3-3)精製工程
前工程で得られた粗結晶(19.0g)にメタノール(285mL)を加え溶解させた後、5倍量(150mL)になるまで減圧濃縮した。濃縮液に10倍量(190mL)になるまでメタノールを加え、さらに水道水(190mL)を25℃~40℃を保つように加えた。外温を20℃に設定し、結晶が析出するまでゆっくり冷却させた後、晶出した温度にて30分間攪拌した。水道水(190mL)を追加し、さらに5℃以下に冷却して30分間攪拌した後、析出した結晶をろ過し、冷33%メタノール水で洗浄した。結晶を40℃にて10時間以上真空乾燥し、Compound(1)の結晶(18.4g、収率97.0%)を得た。
(実施例2)Compound(1)を製造する方法
(2-1)4-(4-メチルフェニル)ブタン酸クロリドの調製
4-(4-メチルフェニル)ブタン酸(8.66g)をECH(70mL)に溶解し、室温、窒素雰囲気下でDMF(10μL)、オキサリルクロリド(4.5mL)を加えた。その溶液を同温度で3時間攪拌した後、減圧度6.5kPa、外温75℃に設定し、ECH(10mL)を留去した(内温約54℃)。この調製した溶液(4-(4-メチルフェニル)ブタン酸クロリドのECH溶液)は窒素雰囲気下で一夜静置した後、特に精製することなくアシル化反応(2-2)に使用した。

- 一般式(III)を有する化合物と、一般式(IV)を有する化合物とを、常圧又は減圧下、塩基を使用せずに非極性溶媒中で還流することによって、一般式(I)を有する化合物を製造する方法。
