本発明は、ヒトの濾胞ヘルパーT細胞を検出するためのマーカーに関する。また、本発明は、該マーカー用いてヒト濾胞ヘルパーT細胞を検出する方法に関する。
濾胞ヘルパーT細胞(以下、「Tfh細胞」ともいう)は、リンパ節や脾臓などの2次リンパ組織の濾胞胚中心で、B細胞を活性化して抗体産生を促進させるヘルパーT細胞として発見された細胞である。また、Tfh細胞はケモカイン受容体のCXCR5(chemokine (C-X-C motif) receptor 5)を発現し、サイトカインのIL-21(interleukin 21)を分泌する細胞として知られている(非特許文献1および2参照)。
通常、Tfh細胞は感染防御に役立つが、全身性エリテマトーデス(SLE)などの自己免疫疾患においては、Tfh細胞は自己反応性B細胞に不適切な活性化シグナルを与えて、自己抗体を産生させると考えられている(非特許文献1参照)。また、SLE、シェーグレン症候群などの自己免疫疾患において、Tfh細胞がリンパ組織から末梢血中に漏れ出る可能性があることを示唆する報告もある。すなわち、Tfh細胞を測定して得られた結果は、Tfh細胞が関与する疾患、例えばSLEやシェーグレン症候群などの自己免疫疾患の診断および検査に利用できる可能性がある。
Tfh細胞を検出する方法としては、Tfh細胞に特異的に発現しているマーカー分子を利用する方法がある。例えば、Chtanova T.ら(非特許文献3)は、ヒトの扁桃腺から単離したTfh細胞と、ヒトの臍帯血から培養したTh1およびTh2細胞と、ヒトの末梢血から単離したメモリーT細胞(Tエフェクターメモリー(TEM)およびTセントラルメモリー(TCM))とを用いて遺伝子の発現解析を行い、Tfh細胞に特異的に発現している遺伝子を見出している。また、国際公開第2010/003002号(特許文献1)には、マウス脾細胞から単離したTfh細胞と、培養したTh1、Th2およびTh17細胞とを用いて遺伝子の発現解析を行い、Tfh細胞に特異的に発現している遺伝子を見出したことが記載されている。

Chtanova T.らは、遺伝子の発現解析において、Tfh細胞との比較対象としてTh1、Th2細胞およびメモリーT細胞を用いているが、その他のTh細胞(例えばTh9、Th17およびTh22細胞)とTfh細胞との比較は行っていない(非特許文献3参照)。国際公開第2010/003002号(特許文献1)に記載のTfh細胞に発現する遺伝子は、マウスのTfh細胞から同定した遺伝子であるので、ヒトのTfh細胞とは遺伝子の発現プロファイルが異なる可能性がある。また、この文献に記載の遺伝子の発現解析では、Tfh細胞との比較対象としてTh1、Th2細胞およびTh17細胞を用いているが、その他のTh細胞(例えばTh9およびTh22細胞)とTfh細胞との比較は行っていない。
そこで、本発明者らは、最適な条件下でヒトの末梢血から単離・培養して得たTfh細胞を用い、他のTh細胞(Th1、Th2、Th9、Th17、Th22およびTreg細胞)と比べてヒトTfh細胞に特異的に発現する遺伝子を同定し、それらの遺伝子をヒトTfh細胞の検出を可能にする新規分子マーカーとして提供することを目的とした。
本発明者らはまず、健常な成人の末梢血からTfh細胞を単離および培養した。そして、本発明者らは、得られたTfh細胞に特異的に発現する遺伝子を同定して、本発明を完成した。

本明細書において、膜タンパク質とは、細胞の膜画分に含まれるタンパク質を意味する。細胞外/分泌タンパク質とは、細胞内で合成され、細胞質膜外に局在するかまたは細胞質膜外へ分泌されるタンパク質を意味する。細胞内タンパク質とは、その局在が主に細胞内であるタンパク質を意味する。
本明細書において、あるポリヌクレオチドがTfh細胞において「特異的に発現する」との表現は、Tfh細胞以外の細胞におけるそのポリヌクレオチドの発現に比べて、Tfh細胞におけるそのポリヌクレオチドの発現が有意に高いことを意味する。
具体的には、Tfh細胞におけるそのポリヌクレオチドの発現が、Tfh細胞以外の細胞でのそのポリヌクレオチドの発現の約3倍以上であることを意味する。好ましくは、Tfh細胞におけるそのポリヌクレオチドの発現が、Tfh細胞以外のヘルパーT細胞(Th1細胞、Th2細胞、Th9細胞、Th17細胞、Th22およびTreg細胞)でのそのポリヌクレオチドの発現の約3倍以上である。
具体的には、Tfh細胞におけるそのポリヌクレオチドの発現が、Tfh細胞以外の細胞でのそのポリヌクレオチドの発現の約3倍以上であることを意味する。好ましくは、Tfh細胞におけるそのポリヌクレオチドの発現が、Tfh細胞以外のヘルパーT細胞(Th1細胞、Th2細胞、Th9細胞、Th17細胞、Th22およびTreg細胞)でのそのポリヌクレオチドの発現の約3倍以上である。
本発明のポリヌクレオチドマーカーの塩基配列自体は、既に公知である。これらは、例えばUniGene(米国国立医学図書館の国立生物情報センター(National Center for Biotechnology Information:NCBI)により提供されるデータベース)などから知ることができる。本発明のポリヌクレオチドマーカーの塩基配列のEntrez gene ID、UniGene ID、Protein IDおよびTranscript IDを表16に示す。
なお、「Probe set ID」とは、Affymetrix社のGeneChip (登録商標)で用いられる遺伝子認識のためのプローブの組(Probe set)を特定するためのID番号である。なお、本明細書の表16に示されるProbe set IDは「Human Genome U133 Plus 2.0」アレイに搭載されるProbe setのID番号である。各Probe setの標的配列を、配列番号1~182に示す。
なお、「Probe set ID」とは、Affymetrix社のGeneChip (登録商標)で用いられる遺伝子認識のためのプローブの組(Probe set)を特定するためのID番号である。なお、本明細書の表16に示されるProbe set IDは「Human Genome U133 Plus 2.0」アレイに搭載されるProbe setのID番号である。各Probe setの標的配列を、配列番号1~182に示す。
本明細書において、ポリヌクレオチドの「変異型」とは、上記の遺伝子がコードするタンパク質の性質を変化させないような変異が導入されたポリヌクレオチドを意味する。このような変異は、上記の遺伝子の塩基配列からの1もしくは複数のヌクレオチドの欠失または置換、あるいは1もしくは複数のヌクレオチドの付加を含む。
上記の変異型は、上記の各遺伝子の塩基配列と、通常は少なくとも80%、好ましくは少なくとも85%、より好ましくは少なくとも約90%、さらにより好ましくは少なくとも95%の配列同一性を有する。

本発明のポリヌクレオチドマーカーとしての遺伝子によりコードされるタンパク質を少なくとも1つ検出することにより、ヒトTfh細胞を検出することもできる。よって、少なくとも1つの上記の遺伝子によりコードされるタンパク質も本発明の1つである。すなわち、本発明のヒトTfh細胞検出用タンパク質マーカー(以下、「本発明のタンパク質マーカー」ともいう)は、上記の遺伝子から選択される少なくとも1つの遺伝子によりコードされるタンパク質、その機能的に同等な変異型およびそれらの断片である。
このようなタンパク質マーカーのアミノ酸配列は、上記のUniGeneなどから得られたポリヌクレオチドマーカーの塩基配列に基づいて得ることができる。また、上記のようなNCBIにより提供されるデータベースから入手することもできる。なお、本発明のタンパク質マーカーのアミノ酸配列についてのNCBIのProtein ID番号を、表16に示す。
タンパク質の機能的に同等な変異型とは、上記のタンパク質の機能を変化させないような変異が導入されたタンパク質を意味する。このような変異は、上記の公知のタンパク質のアミノ酸配列からの1若しくは複数のアミノ酸の欠失または置換、あるいは1若しくは複数のアミノ酸の付加を含む。

本発明の好ましい実施形態においては、本発明のタンパク質マーカーは、CD9のアミノ酸配列を有するタンパク質、ならびにその機能的に同等な変異型およびそれらの断片(以下、「CD9についてのタンパク質マーカー」という)である。
なお、本発明には、本発明のポリヌクレオチドマーカーとしての遺伝子によりコードされるタンパク質、その機能的に同等な変異型およびそれらの断片を、ヒトTfh細胞を検出するためのマーカーとして使用することも含まれる。
本発明のポリヌクレオチドマーカーに特異的にハイブリダイズできる分子は、該マーカーを検出するために用いることができるので、ヒトTfh細胞検出用プローブとして有用である。該プローブは、本発明のポリヌクレオチドマーカーに特異的にハイブリダイズできるDNA、RNAなどの核酸プローブ、ペプチドプローブのいずれであってもよい。ヒトTfh細胞検出用プローブとしては、特に、ポリヌクレオチドマーカーを検出するための核酸プローブ、特にDNAプローブが好ましい。
本明細書において、「特異的にハイブリダイズできる」とは、ストリンジェントな条件下で標的核酸分子(上記のポリヌクレオチドマーカー)にハイブリダイズできることを意味する。
本明細書において、ストリンジェントな条件とは、ヒトTfh細胞検出用プローブが標的ポリヌクレオチドマーカーに、該標的ポリヌクレオチドマーカー以外のポリヌクレオチドよりも検出可能に大きい程度(例えば、バックグラウンドの少なくとも2倍を超える)でハイブリダイズできる条件である。
なお、ストリンジェントな条件は、通常、配列依存性であり、そして種々の環境において異なる。一般的に、ストリンジェントな条件は、規定のイオン強度およびpHでの特定の配列の熱的融点(thermal melting point:Tm)よりも、約5℃低くなるように選択される。このTmは、(規定されたイオン強度、pHおよび核酸組成の下で)上記の標的核酸分子の塩基配列に相補的なプローブの50%が平衡してハイブリダイズする温度である。
本明細書において、ストリンジェントな条件とは、ヒトTfh細胞検出用プローブが標的ポリヌクレオチドマーカーに、該標的ポリヌクレオチドマーカー以外のポリヌクレオチドよりも検出可能に大きい程度(例えば、バックグラウンドの少なくとも2倍を超える)でハイブリダイズできる条件である。
なお、ストリンジェントな条件は、通常、配列依存性であり、そして種々の環境において異なる。一般的に、ストリンジェントな条件は、規定のイオン強度およびpHでの特定の配列の熱的融点(thermal melting point:Tm)よりも、約5℃低くなるように選択される。このTmは、(規定されたイオン強度、pHおよび核酸組成の下で)上記の標的核酸分子の塩基配列に相補的なプローブの50%が平衡してハイブリダイズする温度である。
このような条件は、当該技術において公知のポリヌクレオチド同士のハイブリダイゼーション法、例えばPCR法、マイクロアレイ法、サザンブロット法などにおいて、ポリヌクレオチド同士のハイブリダイゼーションに用いられる条件であってよい。
具体的には、pH 7.0~9.0で塩濃度が約1.5 M Naイオンより低い、より具体的には、約0.01~1.0 M Naイオン濃度(または他の塩)であり、少なくとも約30℃の条件が挙げられる。例えば、マイクロアレイ法におけるストリンジェントな条件は、37℃で50%ホルムアミド、1M NaCl、1% SDS中でのハイブリダイゼーション、および60~65℃での0.1×SSC中での洗浄を含む。
また、PCR法におけるストリンジェントな条件は、pH 7.0~9.0、0.01~0.1 MのTris HCl、0.05~0.15 M Kイオン濃度(または他の塩)、少なくとも約55℃の条件が挙げられる。
具体的には、pH 7.0~9.0で塩濃度が約1.5 M Naイオンより低い、より具体的には、約0.01~1.0 M Naイオン濃度(または他の塩)であり、少なくとも約30℃の条件が挙げられる。例えば、マイクロアレイ法におけるストリンジェントな条件は、37℃で50%ホルムアミド、1M NaCl、1% SDS中でのハイブリダイゼーション、および60~65℃での0.1×SSC中での洗浄を含む。
また、PCR法におけるストリンジェントな条件は、pH 7.0~9.0、0.01~0.1 MのTris HCl、0.05~0.15 M Kイオン濃度(または他の塩)、少なくとも約55℃の条件が挙げられる。
上記のヒトTfh細胞検出用核酸プローブの配列は、当該技術常識および本発明のポリヌクレオチドマーカーの塩基配列に基づいて、該マーカーに特異的にハイブリダイズできるように、当業者が適宜決定できる。
そのような配列は、例えば、一般に利用可能なプライマー設計ソフトウェア(例えばPrimer3(http://frodo.wi.mit.edu/cgi-bin/primer3/primer3.cgiから利用可能)やDNASISPro(日立ソフトウェアエンジニアリング株式会社))を用いて決定できる。
そのような配列は、例えば、一般に利用可能なプライマー設計ソフトウェア(例えばPrimer3(http://frodo.wi.mit.edu/cgi-bin/primer3/primer3.cgiから利用可能)やDNASISPro(日立ソフトウェアエンジニアリング株式会社))を用いて決定できる。
上記のヒトTfh細胞検出用核酸プローブは、当該技術において公知のポリヌクレオチドの合成方法により作製できる。
上記のヒトTfh細胞検出用核酸プローブは、当該技術において通常用いられる標識物質により標識されていてもよい。標識された該プローブを用いることにより、本発明のポリヌクレオチドマーカーの検出、すなわちヒトTfh細胞の検出を簡便に行うことができる。
そのような標識物質は、32Pなどの放射性同位体、フルオレセインなどの蛍光物質、アルカリホスファターゼ、セイヨウワサビペルオキシダーゼなどの酵素、ビオチンなどの当該技術において通常用いられる標識物質であり得る。

上記のヒトTfh細胞検出用核酸プローブは、1種のみを用いるか、または複数種を組み合わせて用いることにより、ヒトTfh細胞を特異的に検出できる。例えば、当該技術において公知の方法を用いて該プローブ1種以上を基板上に固定化することにより、ヒトTfh細胞検出用ポリヌクレオチドマーカーを検出するためのDNAチップまたはマイクロアレイを作製できる。
上記のヒトTfh細胞検出用核酸プローブは、例えば、PCR法により上記のポリヌクレオチドマーカーを増幅するための2種以上のプライマーのセットであり得る。
上記のヒトTfh細胞検出用核酸プローブは、例えば、PCR法により上記のポリヌクレオチドマーカーを増幅するための2種以上のプライマーのセットであり得る。
本発明のタンパク質マーカーに特異的に結合できる分子は、該マーカーを検出するために用いることができるので、ヒトTfh細胞の検出に有用である。このような分子は、タンパク質マーカーに特異的に結合できるDNA、RNAなどの核酸アプタマー、抗体などのいずれであってもよいが、より好ましくは抗体である。
また、本発明のタンパク質マーカーが酵素である場合、該酵素とそれに対応する基質とを反応させて発色、発光、蛍光などを生じさせることにより、該マーカーを検出できる。
また、本発明のタンパク質マーカーが酵素である場合、該酵素とそれに対応する基質とを反応させて発色、発光、蛍光などを生じさせることにより、該マーカーを検出できる。
上記のヒトTfh細胞検出用抗体は、例えば、次のような当該技術において公知の手順により作製できる。本発明のポリヌクレオチドマーカーである遺伝子の塩基配列または本発明のタンパク質マーカーのアミノ酸配列に基づいて、タンパク質マーカーのアミノ酸配列を有するタンパク質をコードするDNA分子を適切な発現ベクターに組み込む。得られた発現ベクターを適切な宿主細胞に導入し、得られた形質転換細胞を培養して、目的のタンパク質を得る。得られたタンパク質を精製して免疫原とし、該免疫原と所望によりアジュバントとを用いて、適切な哺乳動物、例えばラット、マウスなどを免疫する。免疫された動物の脾臓細胞などから、目的の免疫原に対する抗体を産生する抗体産生細胞をスクリーニングにより選択する。得られた抗体産生細胞を、ミエローマ細胞と融合させてハイブリドーマを得て、これをスクリーニングすることにより、上記の遺伝子によりコードされるタンパク質に特異的結合性を有する抗体を産生する抗体産生ハイブリドーマを得ることができる。得られた抗体産生ハイブリドーマを培養することにより、目的の抗体を得ることができる。
ヒトTfh細胞を検出するために用い得る核酸アプタマーは、例えば、次のような当該技術において公知の手順により作製できる。ランダムな核酸の塩基配列を有する核酸ライブラリーを公知の手法により作製し、これを試験管内進化法(SELEX法)などに付すことより標的のタンパク質(本発明のタンパク質マーカー)に特異的に結合できる核酸アプタマーを選択できる。
ヒトTfh細胞検出用タンパク質マーカーに特異的に結合できる分子は、当該技術において通常用いられる標識物質により標識されていてもよい。例えば、標識されたヒトTfh細胞検出用抗体を用いることにより、ヒトTfh細胞検出用タンパク質マーカーの検出、すなわちヒトTfh細胞の検出を簡便に行うことができる。
そのような標識物質は、32Pなどの放射性同位体、フルオレセインなどの蛍光物質、アルカリホスファターゼ、セイヨウワサビペルオキシダーゼなどの酵素、ビオチンなどの当該技術において通常用いられる標識物質であり得る。

本発明のポリヌクレオチドマーカーおよびタンパク質マーカーは、Tfh細胞において特異的に発現することが当該技術において公知である分子と組み合わせて、Tfh細胞の検出に用いてもよい。そのような公知の分子としては、例えばCXCR5、IL-21などが挙げられる。
したがって、本発明の一つの実施形態においては、CD9についてのポリヌクレオチドマーカーと、CXCR5をコードする遺伝子の塩基配列を有するポリヌクレオチド、ならびにその変異型および断片との組み合わせを、ヒトTfh細胞検出用ポリヌクレオチドマーカーとして用いることができる。また、本発明の別の実施形態においては、CD9についてのタンパク質マーカーと、CXCR5のアミノ酸配列を有するタンパク質、その機能的に同等な変異型およびそれらの断片との組み合わせを、ヒトTfh細胞検出用タンパク質マーカーとして用いることができる。
上記の組み合わせによれば、Tfh細胞における特異的発現が公知である分子を単独で用いて検出する場合よりも高い精度でTfh細胞を検出することを可能にする。
したがって、本発明の一つの実施形態においては、CD9についてのポリヌクレオチドマーカーと、CXCR5をコードする遺伝子の塩基配列を有するポリヌクレオチド、ならびにその変異型および断片との組み合わせを、ヒトTfh細胞検出用ポリヌクレオチドマーカーとして用いることができる。また、本発明の別の実施形態においては、CD9についてのタンパク質マーカーと、CXCR5のアミノ酸配列を有するタンパク質、その機能的に同等な変異型およびそれらの断片との組み合わせを、ヒトTfh細胞検出用タンパク質マーカーとして用いることができる。
上記の組み合わせによれば、Tfh細胞における特異的発現が公知である分子を単独で用いて検出する場合よりも高い精度でTfh細胞を検出することを可能にする。
本発明の範囲には、細胞を含む試料中で、本発明のヒトTfh細胞検出用ポリヌクレオチドマーカーまたはヒトTfh細胞検出用タンパク質マーカーを少なくとも1つ検出することにより、ヒトTfh細胞を検出する方法(以下、本発明の方法ともいう)も含まれる。
本発明の方法の一つの実施形態においては、CD9についてのポリヌクレオチドマーカーまたはタンパク質マーカーを検出することによりヒトTfh細胞を検出することが好ましい。
また、本発明の方法の別の実施形態においては、検出感度を向上させる観点から、複数のヒトTfh細胞検出用ポリヌクレオチドマーカーまたはタンパク質マーカーを検出することが好ましい。
本発明の方法においては、本発明のポリヌクレオチドマーカーまたはタンパク質マーカーと、Tfh細胞における特異的発現が公知である分子との組み合わせをヒトTfh細胞検出用マーカーとして検出することにより、試料中のヒトTfh細胞を検出してもよい。そのような組み合わせとしては、上記のCD9とCXCR5との組み合わせが好ましい。
本発明の方法の一つの実施形態においては、CD9についてのポリヌクレオチドマーカーまたはタンパク質マーカーを検出することによりヒトTfh細胞を検出することが好ましい。
また、本発明の方法の別の実施形態においては、検出感度を向上させる観点から、複数のヒトTfh細胞検出用ポリヌクレオチドマーカーまたはタンパク質マーカーを検出することが好ましい。
本発明の方法においては、本発明のポリヌクレオチドマーカーまたはタンパク質マーカーと、Tfh細胞における特異的発現が公知である分子との組み合わせをヒトTfh細胞検出用マーカーとして検出することにより、試料中のヒトTfh細胞を検出してもよい。そのような組み合わせとしては、上記のCD9とCXCR5との組み合わせが好ましい。
本発明の方法において、細胞を含む試料としては、ヒトから採取した生体試料または人工的に培養した細胞を含む試料が挙げられる。生体試料としては、血液、組織、関節液、脳脊髄液、胸水、腹水などが挙げられる。
本発明のポリヌクレオチドマーカーの存在を検出する方法の一実施形態について説明する。
まず、細胞を含む試料から、フェノール抽出およびエタノール沈殿、市販のDNA抽出キットなどを用いる当該技術において公知の方法により、核酸(DNAまたはRNA)を抽出する。
次いで、得られた核酸試料中の本発明のポリヌクレオチドマーカーの存在を検出する。この検出には、上記のヒトTfh細胞検出用核酸プローブを用いることが好ましい。
本発明のポリヌクレオチドマーカーは、PCR法、RT-PCR法、リアルタイムPCR法、LAMP(Loop-mediated isothermal amplification)法のような核酸増幅法、サザンハイブリダイゼーション、ノザンハイブリダイゼーション、FISH(蛍光in situハイブリダイゼーション)のようなハイブリダイゼーション法、DNAチップまたはマイクロアレイ法などの当該技術において公知の方法により検出できる。これらの方法を、上記のストリンジェントな条件下で行い、上記のヒトTfh細胞検出用核酸プローブがハイブリッドを形成したことを、上記の標識物質を検出することなどにより検出して、本発明のポリヌクレオチドマーカーの存在を検出できる。
まず、細胞を含む試料から、フェノール抽出およびエタノール沈殿、市販のDNA抽出キットなどを用いる当該技術において公知の方法により、核酸(DNAまたはRNA)を抽出する。
次いで、得られた核酸試料中の本発明のポリヌクレオチドマーカーの存在を検出する。この検出には、上記のヒトTfh細胞検出用核酸プローブを用いることが好ましい。
本発明のポリヌクレオチドマーカーは、PCR法、RT-PCR法、リアルタイムPCR法、LAMP(Loop-mediated isothermal amplification)法のような核酸増幅法、サザンハイブリダイゼーション、ノザンハイブリダイゼーション、FISH(蛍光in situハイブリダイゼーション)のようなハイブリダイゼーション法、DNAチップまたはマイクロアレイ法などの当該技術において公知の方法により検出できる。これらの方法を、上記のストリンジェントな条件下で行い、上記のヒトTfh細胞検出用核酸プローブがハイブリッドを形成したことを、上記の標識物質を検出することなどにより検出して、本発明のポリヌクレオチドマーカーの存在を検出できる。
次に、ヒトTfh細胞検出用タンパク質マーカーの存在を検出する方法の一実施形態について説明する。
例えば、検出対象である該タンパク質マーカーが細胞内部に存在するタンパク質である場合、当該技術において公知の方法を用いて、細胞からタンパク質を抽出する。細胞からのタンパク質の抽出は、超音波による細胞の破砕、細胞可溶化液を用いる細胞の可溶化などの公知の方法により行うことができる。そして、本発明のタンパク質マーカーに特異的に結合する分子を用いて、得られたタンパク質試料中の該タンパク質マーカーを検出できる。具体的には、タンパク質マーカーは、ELISA法、ウェスタンブロッティング法などの当該技術において公知の方法により検出できる。この検出には、本発明のタンパク質マーカーに特異的に結合する分子として、上記のヒトTfh細胞検出用抗体を用いることが好ましい。

例えば、検出対象である本発明のタンパク質マーカーが分泌タンパク質である場合、該タンパク質マーカーに特異的に結合する分子を用いて、細胞を含む試料中に分泌されたタンパク質マーカーを検出できる。
また、細胞を含む試料から細胞(リンパ球)を採取し、該細胞を、抗CD3抗体、抗CD28抗体、コンカナバリンA、フィトヘマグルチニン(PHA)、ホルボールミリステートアセテート(PMA)、イオノマイシンなどを用いて刺激した後、タンパク質マーカーに特異的に結合する分子を用いて、該細胞から分泌されたタンパク質マーカーを検出できる。
具体的には、本発明のタンパク質マーカーは、ELISA法、ウェスタンブロッティング法などの当該技術において公知の方法により検出できる。この検出には、本発明のタンパク質マーカーに特異的に結合する分子として、上記のヒトTfh細胞検出用抗体を用いることが好ましい。
また、細胞を含む試料から細胞(リンパ球)を採取し、該細胞を、抗CD3抗体、抗CD28抗体、コンカナバリンA、フィトヘマグルチニン(PHA)、ホルボールミリステートアセテート(PMA)、イオノマイシンなどを用いて刺激した後、タンパク質マーカーに特異的に結合する分子を用いて、該細胞から分泌されたタンパク質マーカーを検出できる。
具体的には、本発明のタンパク質マーカーは、ELISA法、ウェスタンブロッティング法などの当該技術において公知の方法により検出できる。この検出には、本発明のタンパク質マーカーに特異的に結合する分子として、上記のヒトTfh細胞検出用抗体を用いることが好ましい。
例えば、検出対象である本発明のタンパク質マーカーが細胞表面(細胞膜)に存在するタンパク質である場合、該タンパク質マーカーに特異的に結合する分子を用いて、細胞を含む試料中の細胞の表面に存在するタンパク質マーカーを検出できる。
また、細胞を含む試料から細胞の膜画分を採取し、本発明のタンパク質マーカーに特異的に結合する分子を用いて、得られた膜画分中のタンパク質マーカーを検出できる。具体的には、ELISA法、ウェスタンブロッティング法、フローサイトメトリ(FCM)に基づく方法などの当該技術において公知の方法により検出できる。
この検出には、本発明のタンパク質マーカーに特異的に結合する分子として、上記のヒトTfh細胞検出用抗体を用いることが好ましい。
また、細胞を含む試料から細胞の膜画分を採取し、本発明のタンパク質マーカーに特異的に結合する分子を用いて、得られた膜画分中のタンパク質マーカーを検出できる。具体的には、ELISA法、ウェスタンブロッティング法、フローサイトメトリ(FCM)に基づく方法などの当該技術において公知の方法により検出できる。
この検出には、本発明のタンパク質マーカーに特異的に結合する分子として、上記のヒトTfh細胞検出用抗体を用いることが好ましい。
例えば、FCMにより本発明のタンパク質マーカーを検出する場合、次のような手順で検出を行うことができる。
まず、細胞を含む試料を、適切な標識物質で標識されたヒトTfh細胞検出用抗体と接触させる。存在するのであればヒトTfh細胞は、この標識された抗体と細胞表面で結合する。よって、標識物質と結合した細胞を含む試料をフローサイトメータに通すことにより、ヒトTfh細胞を検出できる。また、所望により標識物質と結合したヒトTfh細胞を、セルソーターを用いて弁別・分取することもできる。
このようなFCMに基づく方法自体は、当業者に公知であり、反応の条件は当業者により適宜決定される。
まず、細胞を含む試料を、適切な標識物質で標識されたヒトTfh細胞検出用抗体と接触させる。存在するのであればヒトTfh細胞は、この標識された抗体と細胞表面で結合する。よって、標識物質と結合した細胞を含む試料をフローサイトメータに通すことにより、ヒトTfh細胞を検出できる。また、所望により標識物質と結合したヒトTfh細胞を、セルソーターを用いて弁別・分取することもできる。
このようなFCMに基づく方法自体は、当業者に公知であり、反応の条件は当業者により適宜決定される。
上述のように、本実施形態のヒトTfh細胞検出用ポリヌクレオチドマーカーまたはタンパク質マーカーを少なくとも1つ検出することにより、ヒトTfh細胞を特異的に検出することができる。したがって、例えば、被験者から採取した組織などの細胞を含む試料において上記のマーカーを検出することにより、該試料中のTfh細胞を特異的に検出することができる。また、上記のマーカーを用いれば、種々の細胞を含む試料からTfh細胞を単離することができる。
さらに、上記のヒトTfh細胞検出用マーカーは、SLEやシェーグレン症候群などの自己免疫疾患のようなTfh細胞が関与すると考えられている疾患の患者の診断および/または検査において利用できる可能性がある。すなわち、上記のヒトTfh細胞検出用ポリヌクレオチドマーカーおよびタンパク質マーカーからなる、Tfh細胞が関与すると考えられている疾患の患者の診断および/または検査のためのマーカー、並びにその使用も本発明の範囲に含まれる。また、そのようなマーカーを用いて、被験者が、Tfh細胞が関与すると考えられている疾患であるか否かを診断および/または検査する方法も本発明の範囲に含まれる。
さらに、上記のヒトTfh細胞検出用マーカーは、SLEやシェーグレン症候群などの自己免疫疾患のようなTfh細胞が関与すると考えられている疾患の患者の診断および/または検査において利用できる可能性がある。すなわち、上記のヒトTfh細胞検出用ポリヌクレオチドマーカーおよびタンパク質マーカーからなる、Tfh細胞が関与すると考えられている疾患の患者の診断および/または検査のためのマーカー、並びにその使用も本発明の範囲に含まれる。また、そのようなマーカーを用いて、被験者が、Tfh細胞が関与すると考えられている疾患であるか否かを診断および/または検査する方法も本発明の範囲に含まれる。
本発明を、実施例により詳細に説明するが、本発明はこれらの実施例によって限定されない。
実施例1:ヒト末梢血由来培養Tfh細胞における高発現遺伝子の解析
1.ヒト末梢血からのナイーブCD4陽性T細胞の単離
健常な成人の末梢血から得たバフィーコートをFicoll-paque plus溶液(GEヘルスケアバイオサイエンス株式会社)に重層し、これを遠心分離することにより単核球フラクションを得た。抗CD4抗体を結合させた磁気ビーズ(Miltenyi Biotec社)を用いて、上記のフラクションからCD4陽性細胞を粗精製した。
このようにして得たCD4陽性細胞を、表1に示す蛍光標識抗体を用いて染色した後、セルソーター(FACS Aria:Becton Dickinson社)を用いて、CD4+CD25-CD45RA+CD45RO-ナイーブCD4陽性T細胞(以下、「ナイーブCD4陽性T細胞」ともいう)を分離した。
1.ヒト末梢血からのナイーブCD4陽性T細胞の単離
健常な成人の末梢血から得たバフィーコートをFicoll-paque plus溶液(GEヘルスケアバイオサイエンス株式会社)に重層し、これを遠心分離することにより単核球フラクションを得た。抗CD4抗体を結合させた磁気ビーズ(Miltenyi Biotec社)を用いて、上記のフラクションからCD4陽性細胞を粗精製した。
このようにして得たCD4陽性細胞を、表1に示す蛍光標識抗体を用いて染色した後、セルソーター(FACS Aria:Becton Dickinson社)を用いて、CD4+CD25-CD45RA+CD45RO-ナイーブCD4陽性T細胞(以下、「ナイーブCD4陽性T細胞」ともいう)を分離した。
2.ナイーブCD4陽性T細胞からのTfh細胞およびTh9細胞の分化培養
上記の工程1.で得られた成人末梢血由来ナイーブCD4陽性T細胞の残余を、96ウェルプレートに1.0 x 105細胞/0.3 ml/ウェルの密度で播種した。培地は、Yssel培地を用いた。
また、上記の細胞の活性化および増殖のために、各ウェルに抗体ビーズを0.5 x 105個ずつ添加した。そして、Tfh細胞およびTh9細胞のそれぞれの分化培養に適したサイトカインおよび中和抗体(表2参照)を添加して、細胞を37℃、5% CO2のインキュベータ内で培養した。なお、用いたサイトカインの濃度は全て10 ng/mlであり、中和抗体の濃度は全て2.5μg/mlである。また、用いたサイトカインおよび中和抗体は、R&D Systems社とBiolegend社から入手した。
上記の工程1.で得られた成人末梢血由来ナイーブCD4陽性T細胞の残余を、96ウェルプレートに1.0 x 105細胞/0.3 ml/ウェルの密度で播種した。培地は、Yssel培地を用いた。
また、上記の細胞の活性化および増殖のために、各ウェルに抗体ビーズを0.5 x 105個ずつ添加した。そして、Tfh細胞およびTh9細胞のそれぞれの分化培養に適したサイトカインおよび中和抗体(表2参照)を添加して、細胞を37℃、5% CO2のインキュベータ内で培養した。なお、用いたサイトカインの濃度は全て10 ng/mlであり、中和抗体の濃度は全て2.5μg/mlである。また、用いたサイトカインおよび中和抗体は、R&D Systems社とBiolegend社から入手した。
培養開始から3日後、上記のサイトカインおよび抗体を含んだ培地で細胞を3倍に希釈し、さらに4日間(合計7日間)培養して、Tfh細胞およびTh9細胞を得た(なお、この工程で得られたTfh細胞を以下「Tfh細胞(1)」という)。
得られたTfh細胞(1)およびTh9細胞をそれぞれ二等分し、一方の各細胞を、Yssel培地およびPBSで洗浄した後、遠心分離により細胞を回収し、次のRNA抽出工程まで-80℃にて凍結保存した。これらをそれぞれ「活性化刺激なし」のTfh細胞(1)およびTh9細胞とした。また、他方の各細胞には、上記の抗体ビーズを添加し、さらに3時間培養することにより再度活性化した後、遠心分離により細胞を回収し、同様に-80℃にて凍結保存した。これらをそれぞれ「活性化刺激あり」のTfh細胞(1)およびTh9細胞とした。
得られたTfh細胞(1)およびTh9細胞をそれぞれ二等分し、一方の各細胞を、Yssel培地およびPBSで洗浄した後、遠心分離により細胞を回収し、次のRNA抽出工程まで-80℃にて凍結保存した。これらをそれぞれ「活性化刺激なし」のTfh細胞(1)およびTh9細胞とした。また、他方の各細胞には、上記の抗体ビーズを添加し、さらに3時間培養することにより再度活性化した後、遠心分離により細胞を回収し、同様に-80℃にて凍結保存した。これらをそれぞれ「活性化刺激あり」のTfh細胞(1)およびTh9細胞とした。
3.ヒト末梢血からのTfh細胞(2)の単離
健常な成人の末梢血から得たバフィーコートをFicoll-paque plus溶液(GEヘルスケアバイオサイエンス株式会社)に重層し、これを遠心分離することにより、単核球フラクションを得た。次いで、抗CD4抗体を結合させた磁気ビーズ(Miltenyi Biotec社)を用いて、上記のフラクションからCD4陽性細胞を粗精製した。このようにして得たCD4陽性細胞を、表3に示す蛍光標識抗体を用いて染色した後、セルソーター(FACS Aria:Becton Dickinson社)を用いて、Tfh細胞を分離した(なお、この工程で得られたTfh細胞を以下「Tfh細胞(2)」という)。分離のゲーティングは、表4に示すとおりに行った。

4.Tfh細胞(2)の培養
上記の工程3.で得られた成人末梢血由来のTfh細胞(2)を、96ウェルプレートに1.0 x 105細胞/0.3 ml/ウェルの密度で播種した。培地は、Yssel培地を用いた。
また、上記の細胞の活性化および増殖のために、各ウェルに抗CD2/3/28抗体でコーティングした磁気ビーズ(Miltenyi Biotec社)(以下「抗体ビーズ」ともいう)を0.5 x 105個ずつ添加した。そして、Tfh細胞(2)の分化培養に適したサイトカインおよび中和抗体(表5)を添加して、細胞を37℃、5% CO2のインキュベータ内で培養した。
上記の工程3.で得られた成人末梢血由来のTfh細胞(2)を、96ウェルプレートに1.0 x 105細胞/0.3 ml/ウェルの密度で播種した。培地は、Yssel培地を用いた。
また、上記の細胞の活性化および増殖のために、各ウェルに抗CD2/3/28抗体でコーティングした磁気ビーズ(Miltenyi Biotec社)(以下「抗体ビーズ」ともいう)を0.5 x 105個ずつ添加した。そして、Tfh細胞(2)の分化培養に適したサイトカインおよび中和抗体(表5)を添加して、細胞を37℃、5% CO2のインキュベータ内で培養した。
上記サイトカインの濃度は全て10 ng/mlであり、中和抗体の濃度は全て2.5μg/mlである。また、用いたサイトカインおよび中和抗体は、R&D Systems社とBiolegend社から入手した。培養開始から3日後、上記のサイトカインおよび抗体を含んだ培地で細胞を3倍に希釈し、さらに4日間(合計7日間)培養してTfh細胞(2)を得た。
得られたTfh細胞(2)をそれぞれ二等分し、一方の各細胞を、Yssel培地およびPBSで洗浄した後、遠心分離により細胞を回収し、次のRNA抽出工程まで-80℃にて凍結保存した。これらをそれぞれ「活性化刺激なし」のTfh細胞(2)とした。また、他方の各細胞には、上記の抗体ビーズを添加し、さらに3時間培養することにより再度活性化した後、遠心分離により細胞を回収し、同様に-80℃にて凍結保存した。これらをそれぞれ「活性化刺激あり」のTfh細胞(2)とした。
得られたTfh細胞(2)をそれぞれ二等分し、一方の各細胞を、Yssel培地およびPBSで洗浄した後、遠心分離により細胞を回収し、次のRNA抽出工程まで-80℃にて凍結保存した。これらをそれぞれ「活性化刺激なし」のTfh細胞(2)とした。また、他方の各細胞には、上記の抗体ビーズを添加し、さらに3時間培養することにより再度活性化した後、遠心分離により細胞を回収し、同様に-80℃にて凍結保存した。これらをそれぞれ「活性化刺激あり」のTfh細胞(2)とした。
5.ヒト末梢血からのTh1、Th2、Treg、Th17およびTh22細胞の単離
(1)ヒト末梢血からのTh1、Th2、Th17およびTh22細胞の単離
健常な成人の末梢血から得たバフィーコートをFicoll-paque plus溶液(GEヘルスケアバイオサイエンス株式会社)に重層し、これを遠心分離することにより単核球フラクションを得た。抗CD4抗体を結合させた磁気ビーズ(Miltenyi Biotec社)を用いて、上記のフラクションからCD4陽性細胞を粗精製した。このようにして得たCD4陽性細胞を、表6に示す蛍光標識抗体を用いて染色した後、セルソーター(FACS Aria:Becton Dickinson社)を用いて、Th1、Th2、Th17およびTh22の各細胞を分離した。なお、分離のゲーティングは、表7に示すとおりに行った。

(2)ヒト末梢血からのTreg細胞の単離
上記の工程(1)と同様にして得たCD4陽性細胞を、表8に示す蛍光標識抗体を用いて染色した後、上記のセルソーターを用いて、CD4high CD25high CD127internal-negative細胞をTreg細胞として純化した。

6.Th1、Th2、Treg、Th17およびTh22細胞の培養
(1)Th1、Th2、Th17およびTh22細胞の培養
上記の工程5.(1)で得られた成人末梢血由来のTh1、Th2、Th17およびTh22細胞をそれぞれ96ウェルプレートに1.5 x 105細胞/0.3 ml/ウェルの密度で播種した。培地は、Yssel培地(IMDM、1%ヒトAB型血清、0.25% BSA、1.8 mg/l 2-アミノメタノール、40 mg/lトランスフェリン、5mg/lインスリン、2mg/lリノール酸、2mg/lオレイン酸、2mg/lパルミチン酸、1%ペニシリン/ストレプトマイシン)を用いた。また、上記の細胞の活性化および増殖のために、各ウェルに抗CD2/3/28抗体でコーティングした磁気ビーズ(Miltenyi Biotec社)(以下、「抗体ビーズ」ともいう)を0.75 x 105個ずつ添加した。そして、Th1、Th2、Th17およびTh22細胞のそれぞれの分化培養に適したサイトカインおよび中和抗体を添加して、細胞を37℃、5% CO2のインキュベータ内で培養した。なお、用いたサイトカインおよび中和抗体を表9に示す。
(1)Th1、Th2、Th17およびTh22細胞の培養
上記の工程5.(1)で得られた成人末梢血由来のTh1、Th2、Th17およびTh22細胞をそれぞれ96ウェルプレートに1.5 x 105細胞/0.3 ml/ウェルの密度で播種した。培地は、Yssel培地(IMDM、1%ヒトAB型血清、0.25% BSA、1.8 mg/l 2-アミノメタノール、40 mg/lトランスフェリン、5mg/lインスリン、2mg/lリノール酸、2mg/lオレイン酸、2mg/lパルミチン酸、1%ペニシリン/ストレプトマイシン)を用いた。また、上記の細胞の活性化および増殖のために、各ウェルに抗CD2/3/28抗体でコーティングした磁気ビーズ(Miltenyi Biotec社)(以下、「抗体ビーズ」ともいう)を0.75 x 105個ずつ添加した。そして、Th1、Th2、Th17およびTh22細胞のそれぞれの分化培養に適したサイトカインおよび中和抗体を添加して、細胞を37℃、5% CO2のインキュベータ内で培養した。なお、用いたサイトカインおよび中和抗体を表9に示す。
上記のサイトカインの濃度は、IL-6を50 ng/mlとし、IL-6以外を10 ng/mlとした。また、上記の抗体の濃度は、抗IFN-γ抗体を10μg/mlとし、抗IL-4抗体および抗TGF-β抗体を2.5μg/mlとした。なお、サイトカインおよび中和抗体は、それぞれR&D systems社およびeBioscience社から入手した。培養開始から3日後、上記のサイトカインおよび抗体を含んだ培地で細胞を3倍に希釈し、さらに7日間(合計10日間)培養して、Th1、Th2、Th17およびTh22細胞を得た。得られた各細胞をそれぞれ二等分し、一方の各細胞を、Yssel培地およびPBSで洗浄した後、細胞を遠心分離により回収し、次のRNA抽出工程まで-80℃にて凍結保存した。これらをそれぞれ「活性化刺激なし」のTh1、Th2、Th17およびTh22細胞とした。また、他方の各細胞に上記の抗体ビーズを添加し、さらに3時間培養することにより再度活性化した後、細胞を遠心分離により回収し、同様に-80℃にて凍結保存した。これらをそれぞれ「活性化刺激あり」のTh1、Th2、Th17およびTh22細胞とした。
(2)Treg細胞の培養
上記の工程5.(2)で得られたTreg細胞を、上記の工程6.(1)と同様にYssel培地で培養し、そして抗体ビーズを用いて活性化した。さらに、培地には、サイトカインとしてIL-2およびTGF-β1(R&D systems社)、ならびに中和抗体として抗IFN-γ抗体、抗IL-4抗体(eBioscience社)および抗IL-6抗体(BD bioscience社)を添加した。なお、これらのサイトカインおよび中和抗体は、それぞれ10 ng/mlおよび5μg/mlの濃度で用いた。培養開始から3日後、培養開始時と同量のサイトカインおよび中和抗体を添加した。さらに3日間培養した後、細胞を二等分し、一方には活性化に用いた抗体ビーズを添加せず、他方には抗体ビーズを添加して、さらに3時間培養して、それぞれ「活性化刺激なし」および「活性刺激あり」のTreg細胞を得た。その後、各Treg細胞を遠心分離により回収し、次のRNA抽出工程まで-80℃にて凍結保存した。
上記の工程5.(2)で得られたTreg細胞を、上記の工程6.(1)と同様にYssel培地で培養し、そして抗体ビーズを用いて活性化した。さらに、培地には、サイトカインとしてIL-2およびTGF-β1(R&D systems社)、ならびに中和抗体として抗IFN-γ抗体、抗IL-4抗体(eBioscience社)および抗IL-6抗体(BD bioscience社)を添加した。なお、これらのサイトカインおよび中和抗体は、それぞれ10 ng/mlおよび5μg/mlの濃度で用いた。培養開始から3日後、培養開始時と同量のサイトカインおよび中和抗体を添加した。さらに3日間培養した後、細胞を二等分し、一方には活性化に用いた抗体ビーズを添加せず、他方には抗体ビーズを添加して、さらに3時間培養して、それぞれ「活性化刺激なし」および「活性刺激あり」のTreg細胞を得た。その後、各Treg細胞を遠心分離により回収し、次のRNA抽出工程まで-80℃にて凍結保存した。
7.フローサイトメータによる分化培養細胞の純度確認
分化培養を行った各細胞(それぞれ2.5×105細胞)を含む細胞懸濁液に、ホルボールミリステートアセテート(PMA:50 ng/ml)およびイオノマイシン(1μM)を加えて細胞を刺激した。4時間後にブレフェルディンA(10μg/ml)を添加して、さらに2時間培養した。その後、各細胞をリン酸緩衝生理食塩水(PBS)で洗浄し、4%パラホルムアルデヒドで固定した。固定後、サポニン緩衝液(0.5%サポニン、0.5%ウシ血清アルブミン(BSA)、1mMアジ化ナトリウム(PBS中))で各細胞を処理して細胞膜の透過性を亢進させた。そして、細胞に蛍光標識(FITC、PE、APC、PerCP-Cy5.5、Alexa647など)した抗IFN-γ抗体、抗IL-4抗体、抗IL-17抗体、抗Foxp3抗体、抗IL-9抗体、抗IL-22抗体、または抗IL-21抗体を反応させた。反応後、各細胞をサポニン緩衝液、次いで0.5% BSA含有PBSで洗浄し、FACS Canto II(Becton Dickinson社)を用いて解析することにより各細胞の純度を確認した。純度の確認に用いた項目は、それぞれTh1細胞:IFN-γ、Th2細胞:IL-4、Th17細胞:IL-17A、Treg細胞:Foxp3(転写因子)、Th9細胞:IL-9、Th22細胞:IL-22 、Tfh細胞(1)および(2):IL-21である。
分化培養を行った各細胞(それぞれ2.5×105細胞)を含む細胞懸濁液に、ホルボールミリステートアセテート(PMA:50 ng/ml)およびイオノマイシン(1μM)を加えて細胞を刺激した。4時間後にブレフェルディンA(10μg/ml)を添加して、さらに2時間培養した。その後、各細胞をリン酸緩衝生理食塩水(PBS)で洗浄し、4%パラホルムアルデヒドで固定した。固定後、サポニン緩衝液(0.5%サポニン、0.5%ウシ血清アルブミン(BSA)、1mMアジ化ナトリウム(PBS中))で各細胞を処理して細胞膜の透過性を亢進させた。そして、細胞に蛍光標識(FITC、PE、APC、PerCP-Cy5.5、Alexa647など)した抗IFN-γ抗体、抗IL-4抗体、抗IL-17抗体、抗Foxp3抗体、抗IL-9抗体、抗IL-22抗体、または抗IL-21抗体を反応させた。反応後、各細胞をサポニン緩衝液、次いで0.5% BSA含有PBSで洗浄し、FACS Canto II(Becton Dickinson社)を用いて解析することにより各細胞の純度を確認した。純度の確認に用いた項目は、それぞれTh1細胞:IFN-γ、Th2細胞:IL-4、Th17細胞:IL-17A、Treg細胞:Foxp3(転写因子)、Th9細胞:IL-9、Th22細胞:IL-22 、Tfh細胞(1)および(2):IL-21である。
8.トータルRNAの抽出
上記の工程1.、2.、4.および6.で得た各細胞からトータルRNAを抽出するために、それぞれRNeasy Plus MiniキットおよびRNeasy microキット(QIAGEN社)を用いた。なお、具体的な操作は、各キットの添付文書の記載に従って行った。

9.マイクロアレイ発現解析
Two-Cycle Target Labeling and Control Reagents(Affymetrix社)を用いて、上記の工程7.で抽出した各細胞のトータルRNA(10~100 ng)をcDNAに逆転写し、さらにビオチン化cRNAへの転写反応を行った。次いで、増幅したビオチン化cRNA(20μg)を断片化した。なお、具体的な操作は、キットに添付の説明書の記載に従って行った。
上記で得られた各細胞由来のビオチン化cRNA(15μg)を検体として、それぞれGeneChip Hunman Genome U-133 Plus 2.0 Array(Affymetrix社)に添加し、GeneChip Hybridization Oven 640(Affymetrix社)に移して、45℃、60 rpmの条件下で16時間ハイブリダイゼーションを行った。ハイブリダイゼーション終了後、上記のマイクロアレイを、GeneChip Fluidic Station 450(Affymetrix社)を用いて洗浄および蛍光標識を行い、該マイクロアレイを、GeneChip Scanner 3000 7G(Affymetrix社)を用いてスキャンし、蛍光強度データを取得した。

上記で得られた各細胞由来のビオチン化cRNA(15μg)を検体として、それぞれGeneChip Hunman Genome U-133 Plus 2.0 Array(Affymetrix社)に添加し、GeneChip Hybridization Oven 640(Affymetrix社)に移して、45℃、60 rpmの条件下で16時間ハイブリダイゼーションを行った。ハイブリダイゼーション終了後、上記のマイクロアレイを、GeneChip Fluidic Station 450(Affymetrix社)を用いて洗浄および蛍光標識を行い、該マイクロアレイを、GeneChip Scanner 3000 7G(Affymetrix社)を用いてスキャンし、蛍光強度データを取得した。
10.ヒトTh22細胞に特異的に発現する遺伝子の選択
上記の工程8.で得られた蛍光強度のデータを、発現解析ソフトウェアGeneSpring GX Ver.11(Agilent Technologies社)を用いて、MAS5アルゴリズムにより標準化して、各細胞における遺伝子についての相対蛍光強度を得た。この相対蛍光強度は、細胞における遺伝子の発現量に相当する。得られたTfh細胞(1)における各遺伝子の相対蛍光強度を、ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9およびTh22細胞における相対蛍光強度と比較した。Tfh細胞(1)における相対蛍光強度が、ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9およびTh22細胞のいずれよりも3倍以上高く、かつ有意に発現する遺伝子(ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9、Th22細胞およびTfh細胞(1)の8群間における相対蛍光強度のANOVA統計解析により、「p値<0.05」を示した遺伝子)をTfh細胞(1)に特異的に発現している遺伝子として同定した。
また同様に、Tfh細胞(2)における相対蛍光強度が、ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9およびTh22細胞のいずれよりも3倍以上高く、かつ有意に発現する遺伝子(ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9、Th22細胞およびTfh細胞(2)の8群間における相対蛍光強度のANOVA統計解析により、「p値<0.05」を示した遺伝子)をTfh細胞(2)に特異的に発現している遺伝子として同定した。なお、この選択工程において用いた各検体の数を、表10に示す。
上記の工程8.で得られた蛍光強度のデータを、発現解析ソフトウェアGeneSpring GX Ver.11(Agilent Technologies社)を用いて、MAS5アルゴリズムにより標準化して、各細胞における遺伝子についての相対蛍光強度を得た。この相対蛍光強度は、細胞における遺伝子の発現量に相当する。得られたTfh細胞(1)における各遺伝子の相対蛍光強度を、ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9およびTh22細胞における相対蛍光強度と比較した。Tfh細胞(1)における相対蛍光強度が、ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9およびTh22細胞のいずれよりも3倍以上高く、かつ有意に発現する遺伝子(ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9、Th22細胞およびTfh細胞(1)の8群間における相対蛍光強度のANOVA統計解析により、「p値<0.05」を示した遺伝子)をTfh細胞(1)に特異的に発現している遺伝子として同定した。
また同様に、Tfh細胞(2)における相対蛍光強度が、ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9およびTh22細胞のいずれよりも3倍以上高く、かつ有意に発現する遺伝子(ナイーブCD4陽性T細胞、Th1、Th2、Treg、Th17、Th9、Th22細胞およびTfh細胞(2)の8群間における相対蛍光強度のANOVA統計解析により、「p値<0.05」を示した遺伝子)をTfh細胞(2)に特異的に発現している遺伝子として同定した。なお、この選択工程において用いた各検体の数を、表10に示す。
「活性化刺激なし」および「活性化刺激あり」の各条件における、Tfh細胞(1)およびTfh細胞(2)に特異的に発現している遺伝子および該遺伝子についての細胞間の発現量の比を表11~14に示す。表11および12に記載される遺伝子は、それぞれ「活性化刺激なし」および「活性化刺激あり」のTfh細胞(1)に特異的に発現する遺伝子である。表13および14に記載される遺伝子は、それぞれ「活性化刺激なし」および「活性化刺激あり」のTfh細胞(2)に特異的に発現する遺伝子である。なお、表中の「局在」とは、各遺伝子によりコードされるタンパク質の細胞における局在を意味する。
本実施例においては、Chtanova T.ら(非特許文献3参照)によりTfh細胞での特異的発現が見出された遺伝子SGPP2(sphingosine-1-phosphate phosphotase 2)、CXCL13(chemokine (C-X-C motif) ligand 13)、IL-21(interleukin 21)およびMAF(v-maf musculoaponeurotic fibrosarcoma oncogene homolog)も同定された。これらの既知遺伝子を表15に示す。
このように、本実施例においてはTfh細胞での特異的発現が既知である遺伝子も精度よく同定できたことから、上記のTfh細胞および他のTh細胞サブセットを用いる遺伝子探索のアプローチによって、Tfh細胞に特異的に発現する遺伝子を同定できることがわかる。
本発明者らは、上記の同定した遺伝子から表15に示される4遺伝子を除いて、Tfh細胞に特異的に発現する161遺伝子を本発明のTfh細胞検出用ポリヌクレオチドマーカーとした。

本発明のポリヌクレオチドマーカーとしての遺伝子の一覧を表16に示す。なお、表16中、抗CD2/3/28抗体による活性化刺激をしたか又はしていないTfh(1)およびTfh(2)細胞において、特異的な発現が認められた遺伝子には「●」を付した。
表16から明らかなように、本発明の161遺伝子は、Tfh細胞検出用ポリヌクレオチドマーカーとして有用であることが分かる。
実施例2:ヒト末梢血由来培養Th1、Th2およびTfh細胞におけるTfh細胞検出用タンパク質マーカーの発現解析
1.各Th細胞の調製
ヒト末梢血中のTfh細胞、Th1細胞およびTh2細胞をセルソーター(FACS Aria:Becton Dickinson社)を用いて分離した。分離した各Th細胞を、96ウェルプレートに1.0 x 105細胞/0.3 ml/ウェルの密度で播種した。培地は、Yssel培地を用いた。各Th細胞の活性化および増殖のために、各ウェルに抗CD2/3/28抗体でコーティングした磁気ビーズ(Miltenyi Biotec社)を0.5 x 105個ずつ添加した。そして、各Th細胞の分化培養に適したサイトカインおよび中和抗体を添加して、細胞を37℃、5% CO2のインキュベータ内で培養した。なお、分離に使用した抗体およびゲーティング、並びに各Th細胞の培養条件は、実施例1と同じである。

2.CD9測定用試料の調製
調製した各Th細胞(5×106 cells/ml)に、PE標識抗ヒトCD9抗体(BioLegend社)を終濃度が0.2μg/mlとなるように添加し、4℃で20分間反応させた。反応後、0.5% BSAを含有するPBSを添加し、遠心分離により各Th細胞を回収した。洗浄後の各Th細胞を、0.5μg/ml 7-アミノ-アクチノマイシンD(7-AAD)および0.5% BSAを含有するPBSに懸濁し、CD9測定用試料(5×106 cells/ml)を調製した。抗CD9抗体に代えてPE標識マウスIgG1アイソタイプコントロール(Biolegend社)を終濃度1.0μg/mlとなるように添加し、4℃で20分間反応させネガティブコントロール用試料(5×106 cells/ml)を調製した。
調製した各Th細胞(5×106 cells/ml)に、PE標識抗ヒトCD9抗体(BioLegend社)を終濃度が0.2μg/mlとなるように添加し、4℃で20分間反応させた。反応後、0.5% BSAを含有するPBSを添加し、遠心分離により各Th細胞を回収した。洗浄後の各Th細胞を、0.5μg/ml 7-アミノ-アクチノマイシンD(7-AAD)および0.5% BSAを含有するPBSに懸濁し、CD9測定用試料(5×106 cells/ml)を調製した。抗CD9抗体に代えてPE標識マウスIgG1アイソタイプコントロール(Biolegend社)を終濃度1.0μg/mlとなるように添加し、4℃で20分間反応させネガティブコントロール用試料(5×106 cells/ml)を調製した。
3.CXCR5測定用試料の調製
上記2.のCD9測定用試料の調製における、PE標識抗CD9抗体に代えて、終濃度1.0μg/mlのPerCP/Cy5.5標識抗CXCR5抗体(BDバイオサイエンス社)を用い、同様の操作を行うことで、各Th細胞のCXCR5測定用試料(5×106 cells/ml)を調製した。
上記のPerCP/Cy5.5標識抗CXCR5抗体に代えて、PerCP/Cy5.5標識抗マウスIgG2b抗体(BioLegend社)を終濃度1.0μg/mlとなるように添加し、4℃で20分間反応させてネガティブコントロール用試料(5×106 cells/ml)を調製した。
上記2.のCD9測定用試料の調製における、PE標識抗CD9抗体に代えて、終濃度1.0μg/mlのPerCP/Cy5.5標識抗CXCR5抗体(BDバイオサイエンス社)を用い、同様の操作を行うことで、各Th細胞のCXCR5測定用試料(5×106 cells/ml)を調製した。
上記のPerCP/Cy5.5標識抗CXCR5抗体に代えて、PerCP/Cy5.5標識抗マウスIgG2b抗体(BioLegend社)を終濃度1.0μg/mlとなるように添加し、4℃で20分間反応させてネガティブコントロール用試料(5×106 cells/ml)を調製した。
4.IL-21測定用試料の調製
調製した各Th細胞試料に含まれるIL-21陽性細胞を測定するために、各試料からIL-21測定用試料を調製した。2.5×105 cells/mlとなるように調製した各試料に、ホルボールミリステートアセテート(PMA:50 ng/ml)およびイオノマイシン(1μM)を加えて細胞を刺激した。4時間後にブレフェルディンA(10μg/ml)を添加して、さらに2時間培養した。その後、細胞をリン酸緩衝生理食塩水(PBS)で洗浄し、4%パラホルムアルデヒドで固定した。固定後、サポニン緩衝液(0.5%サポニン、0.5%ウシ血清アルブミン(BSA)、1mMアジ化ナトリウム(PBS中))で細胞を処理して細胞膜の透過性を亢進させた。そして、細胞に蛍光標識(PE)した抗IL-21抗体を反応させた。反応後、細胞をサポニン緩衝液、次いで0.5% BSA含有PBSで洗浄し、洗浄後の細胞を0.5% BSAを含有するPBSに懸濁し、IL-21測定用試料を調製した。
上記のPE標識抗IL-21抗体に代えてPE標識抗マウスIgG1抗体(BioLegend社)を終濃度1.0μg/mlとなるように添加し、4℃で20分間反応させてネガティブコントロール用試料を調製した。

5.フローサイトメータを用いたCD9タンパク質の発現解析
上記のようにして調製したCD9測定用試料、CXCR5測定用試料およびIL-21測定試料を、FACSCanto II(BDバイオサイエンス社)およびFACS DIVAソフトウェア(BDバイオサイエンス社)を用いて解析した。解析により得られた蛍光強度のヒストグラム(粒度分布)を図1に示す。なお、図1において、ヒストグラムの縦軸は細胞数を示し、横軸は蛍光強度を示す。また、ヒストグラムの右上の数値は、各測定用試料における、全細胞数に対するマーカー陽性細胞数の比率(%)を示す。ここで、陽性細胞と陰性細胞の判定は、ネガティブコントロールにおける最大の蛍光強度を基準として行った。すなわち、ネガティブコントロールにおける最大の蛍光強度よりも高い蛍光強度を示す細胞を陽性細胞と判定した。逆に、ネガティブコントロールにおける最大の蛍光強度以下の蛍光強度を示す細胞を陰性細胞と判定した。陽性細胞比率は、全細胞数に対する陽性細胞数の比として算出した。
上記のようにして調製したCD9測定用試料、CXCR5測定用試料およびIL-21測定試料を、FACSCanto II(BDバイオサイエンス社)およびFACS DIVAソフトウェア(BDバイオサイエンス社)を用いて解析した。解析により得られた蛍光強度のヒストグラム(粒度分布)を図1に示す。なお、図1において、ヒストグラムの縦軸は細胞数を示し、横軸は蛍光強度を示す。また、ヒストグラムの右上の数値は、各測定用試料における、全細胞数に対するマーカー陽性細胞数の比率(%)を示す。ここで、陽性細胞と陰性細胞の判定は、ネガティブコントロールにおける最大の蛍光強度を基準として行った。すなわち、ネガティブコントロールにおける最大の蛍光強度よりも高い蛍光強度を示す細胞を陽性細胞と判定した。逆に、ネガティブコントロールにおける最大の蛍光強度以下の蛍光強度を示す細胞を陰性細胞と判定した。陽性細胞比率は、全細胞数に対する陽性細胞数の比として算出した。
図1より、CD9は、Th1細胞およびTh2細胞よりもTfh細胞において特異的に高発現していることが分かる。また、CD9測定用試料のTfh細胞の陽性細胞比率は、Tfh細胞についての既知マーカーであるCXCR5の測定用試料の場合と比べて高いことがわかる。さらに、Tfh細胞についての既知マーカーであるIL-21の測定用試料の場合では、Tfh細胞だけでなくTh1細胞の陽性細胞率も高い値を示しているが、CD9測定用試料の場合ではTh1細胞やTh2細胞の陽性細胞率は低く、Tfh細胞のみ高い値を示していることがわかる。以上のことから、CD9タンパク質は、Tfh細胞検出用タンパク質マーカーとして好適に利用可能であることが分かった。
実施例3:Tfh細胞検出用マーカー分画に含まれるTfh細胞比率の解析
1.分画試料の調製
実施例2で調製した培養Tfh細胞を、PerCP/Cy5.5標識抗CXCR5抗体で染色してCXCR5測定用試料を調製した。また、培養Tfh細胞をPerCP/Cy5.5標識抗CXCR5抗体およびPE標識抗ヒトCD9抗体で染色してCXCR5/CD9測定用試料を調製した。セルソーター(FACS Aria:Becton Dickinson社)を用いて、CXCR5測定用試料からCXCR5陽性の細胞を分離した。また、該セルソーターを用いて、CXCR5/CD9測定用試料からCXCR5陽性且つCD9陽性の細胞を分離した。
この分離した細胞をそれぞれ96ウェルプレートに1.5 x 105細胞以下/0.3 ml/ウェルの密度で播種した。培地として、IL-2(R&D systems社)を終濃度10 ng/mlとなるように添加したYssel培地を用い、細胞を37℃、5%CO2のインキュベータ内で3日間培養して、各CXCR5分画試料およびCXCR5/CD9分画試料を調製した。
1.分画試料の調製
実施例2で調製した培養Tfh細胞を、PerCP/Cy5.5標識抗CXCR5抗体で染色してCXCR5測定用試料を調製した。また、培養Tfh細胞をPerCP/Cy5.5標識抗CXCR5抗体およびPE標識抗ヒトCD9抗体で染色してCXCR5/CD9測定用試料を調製した。セルソーター(FACS Aria:Becton Dickinson社)を用いて、CXCR5測定用試料からCXCR5陽性の細胞を分離した。また、該セルソーターを用いて、CXCR5/CD9測定用試料からCXCR5陽性且つCD9陽性の細胞を分離した。
この分離した細胞をそれぞれ96ウェルプレートに1.5 x 105細胞以下/0.3 ml/ウェルの密度で播種した。培地として、IL-2(R&D systems社)を終濃度10 ng/mlとなるように添加したYssel培地を用い、細胞を37℃、5%CO2のインキュベータ内で3日間培養して、各CXCR5分画試料およびCXCR5/CD9分画試料を調製した。
2.IL-21陽性細胞測定のための試料処理
上記1の分画前の培養Tfh細胞試料、ならびに分画後のCXCR5分画試料およびCXCR5/CD9分画試料に含まれるIL-21陽性細胞を測定するために、各試料からIL-21測定用試料を調製した。2.5×105 cells/mlとなるように調製した各試料に、ホルボールミリステートアセテート(PMA:50 ng/ml)およびイオノマイシン(1μM)を加えて細胞を刺激した。4時間後にブレフェルディンA(10μg/ml)を添加して、さらに2時間培養した。その後、細胞をリン酸緩衝生理食塩水(PBS)で洗浄し、4%パラホルムアルデヒドで固定した。固定後、サポニン緩衝液(0.5%サポニン、0.5%ウシ血清アルブミン(BSA)、1mMアジ化ナトリウム(PBS中))で細胞を処理して細胞膜の透過性を亢進させた。そして、細胞に蛍光標識(PE)した抗IL-21抗体を反応させた。反応後、細胞をサポニン緩衝液、次いで0.5% BSA含有PBSで洗浄し、洗浄後の細胞を0.5% BSAを含有するPBSに懸濁し、IL-21測定用試料を調製した。
上記のPE標識抗IL-21抗体に代えてPE標識抗マウスIgG1抗体(BioLegend社)を終濃度1.0μg/mlとなるように添加し、4℃で20分間反応させネガティブコントロール用試料を調製した。

上記のPE標識抗IL-21抗体に代えてPE標識抗マウスIgG1抗体(BioLegend社)を終濃度1.0μg/mlとなるように添加し、4℃で20分間反応させネガティブコントロール用試料を調製した。
3.IL-21陽性細胞の測定
上記2で調製した各IL-21測定用試料をFACSCanto II(BDバイオサイエンス社)およびFACS DIVAソフトウェア(BDバイオサイエンス社)を用いて解析した。各IL-21測定用試料において、全細胞数に対するIL-21陽性細胞数の比率を、Tfh細胞比率(%)として算出した。
ここで、陽性細胞と陰性細胞の判定は、ネガティブコントロールにおける最大の蛍光強度を基準として行った。すなわち、ネガティブコントロールにおける最大の蛍光強度よりも高い蛍光強度を示す細胞を陽性細胞と判定した。
反対に、ネガティブコントロールにおける最大の蛍光強度以下の蛍光強度を示す細胞を陰性細胞と判定した。陽性細胞比率は、全細胞数に対する陽性細胞数の比として算出した。
上記2で調製した各IL-21測定用試料をFACSCanto II(BDバイオサイエンス社)およびFACS DIVAソフトウェア(BDバイオサイエンス社)を用いて解析した。各IL-21測定用試料において、全細胞数に対するIL-21陽性細胞数の比率を、Tfh細胞比率(%)として算出した。
ここで、陽性細胞と陰性細胞の判定は、ネガティブコントロールにおける最大の蛍光強度を基準として行った。すなわち、ネガティブコントロールにおける最大の蛍光強度よりも高い蛍光強度を示す細胞を陽性細胞と判定した。
反対に、ネガティブコントロールにおける最大の蛍光強度以下の蛍光強度を示す細胞を陰性細胞と判定した。陽性細胞比率は、全細胞数に対する陽性細胞数の比として算出した。
4.Tfh細胞濃縮率の算出
上記3で得られた各試料のTfh細胞比率(%)に基づいて、Tfh細胞検出用マーカー分画におけるTfh細胞濃縮率を算出した。具体的には、分画前のTfh細胞比率(%)に対する、各分画のTfh細胞比率(%)の比をそれぞれ算出した。得られた比をTfh細胞濃縮率として、CXCR5分画試料およびCXCR5/CD9分画試料におけるTfh細胞濃縮率を図2に示した。図2より、CXCR5/CD9分画は、既知のTfh細胞のマーカーであるCXCR5よりも高い濃度でTfh細胞が濃縮されることが分かる。したがって、CD9タンパク質とCXCR5タンパク質との組み合わせは、CXCR5タンパク質単独に比べて、Tfh細胞検出用タンパク質マーカーとして有用であることが分かった。
上記3で得られた各試料のTfh細胞比率(%)に基づいて、Tfh細胞検出用マーカー分画におけるTfh細胞濃縮率を算出した。具体的には、分画前のTfh細胞比率(%)に対する、各分画のTfh細胞比率(%)の比をそれぞれ算出した。得られた比をTfh細胞濃縮率として、CXCR5分画試料およびCXCR5/CD9分画試料におけるTfh細胞濃縮率を図2に示した。図2より、CXCR5/CD9分画は、既知のTfh細胞のマーカーであるCXCR5よりも高い濃度でTfh細胞が濃縮されることが分かる。したがって、CD9タンパク質とCXCR5タンパク質との組み合わせは、CXCR5タンパク質単独に比べて、Tfh細胞検出用タンパク質マーカーとして有用であることが分かった。

- 請求項2に記載のポリヌクレオチドマーカーと、CXCR5をコードする遺伝子の塩基配列を有するポリヌクレオチド、ならびにその変異型および断片との組み合わせからなる、ヒト濾胞ヘルパーT細胞検出用ポリヌクレオチドマーカー。
- 請求項1に記載の遺伝子から選択される少なくとも1つの遺伝子または請求項2に記載の遺伝子によりコードされるタンパク質、ならびにその機能的に同等な変異型および断片である、ヒト濾胞ヘルパーT細胞検出用タンパク質マーカー。
- 請求項2を引用する請求項4に記載のタンパク質マーカーと、CXCR5のアミノ酸配列を有するタンパク質、その機能的に同等な変異型およびそれらの断片との組み合わせからなる、ヒト濾胞ヘルパーT細胞検出用タンパク質マーカー。
- 細胞を含む試料における、請求項1~3のいずれか1項に記載のヒト濾胞ヘルパーT細胞検出用ポリヌクレオチドマーカーまたは請求項4もしくは5に記載のヒト濾胞ヘルパーT細胞検出用タンパク質マーカーを少なくとも1つ検出することを含む、ヒト濾胞ヘルパーT細胞を検出する方法。
