[기술분야】
<1> 본 명세서에 개시된 기술은 일반적으로 자성 구조물을 자기적으로 제어하는 방법에 관한 것이다.
【배경기술】
<2> 코드화된 입자 기반의 복합 분석 방법은 그 방식이 가지는 확장성과 빠른 반 응성 덕분에 신약발굴 및 임상진단과 같은 고효율 바이오분자 검사 분야에서 많은 주목을 받고 있다. 복합 분석 방법은 타겟 분석물을 함유하는 바이알 내에 많은 코 드화된 람침 입자들을 함께 흔합함으로써 달성될 수 있다.
<3> 다양한 샘플들에 있어서, 높은 분석 효율올 위해 많은 수의 구분되는 코드들 을 사용하는 것이 필요하다. 충분히 많은 수의 코드를 확보하기 위해 양자점 또는 형광체를 이용한 스펙트 ¾ 코드화 방식과 미세입자 표면 상에 광학적으로 측정가능 한 요소를 패터닝하는 것에 기초한 그래피 ¾ 코드화 방식이 제안되어 왔다.
<4> 하지만, 스펙트럴 코드화 방식은 코드를 구현할 여러 물질을 다루는 것과 관 련하여 스펙트럼의 겹침, 한정적인 물질 수, 고비용 및 미세한 양의 지시 물질 로 딩을 정교하게 제어함에 있어 한계가 있다. 또한 2진 바코드와 같은 그래피컬 코드 화 방식의 경우 해상도의 한계가 있어 층분한 수의 코드를 구현하기 위해 보다 넓 은 면적을 필요로 하여 결국 입자의 크기에 의해 코드 수가 한정된다. 따라서, 2진 이상의 멀티레벨 코딩방식이 제안되었으나 그 공정이 매우 복잡하고 스펙트럴 코드 화 방식의 경우와 같이 여러 종류의 지시 물질의 정확한 로딩이 요구된다.
<5> 또한, 코드화된 입자 기반 복합 분석방법에서 코드를 읽어내기 위해 미세 입 자들을 적절하게 핸들링하는 방법이 필요하다. 예를 들어 미세 입자들을 흔합용액 으로부터 선택적으로 분리해 내는 것은 그리 간단하지 않다.

<37> 자성 나노입자 (120)는 초상자성 물질 (superparamagnetic material)을 포함할 수 있다. 초상자성 물질은 자기장이 제거되어도 자성이 유지되는 강자성 물질 (ferromagnetic material)과 달리 외부 자기장이 존재할 때에만 자성을 갖게 된다. 대체로 강자성 물질의 입자 크기가 수 내지 수백 nm가 되면 초상자성 물질로 상전 이될 수 있다. 예를 들면 산화철의 경우 10nm 정도의 크기에서 초상자성을 가질 수 있다.

<40> 자성 나노입자 (120)를 경화성 물질 (110)에 혼합하여 기계적인 교반 또는 초 음파 처리를 함으로써 컬러 코드화를 위한 조성물 (100)올 제조할 '수 있다. 자성 나 노입자 (120)는 경화성 물질 (110) 내에 부피율 (volume fraction)로 예를 들어 0.01% 내지 20%가 포함될 수 있다. 자성 나노입자 (120)가 부피율 0.01% 미만이면 반사율 이 떨어질 수 있으며 부피율이 20%를 초과하면 반사율이 더 이상 증가하지 않을 수 있다.
<4i> 경화성 물질 (110)은 광결정을 이루는 자성 나노입자들 (120)을 안정적으로 분 산시키는 분산매의 역할을 한다. 또한 가교결합에 의해 자성 나노입자들 (120)간의 간격을 고정함으로써, 자기장이 제거된 이후에도 일정한 구조색을 지속적으로 발현 하도록 할 수 있다.
<42> 경화성 물질 (110)은 경화반웅을 위해 가교결합가능 부위를 함유하는 액상의 단량체 또는 올리고머를 포함할 수 있다. 경화성 물질 (110)은 하이드로젤을 형성할 수 있는 액상의 친수성 고분자를 포함할 수 있다. 친수성 고분자는 친수성 그룹을 가지고 있어서 자성 나노입자 (120)의 분산에 적합한 고분자로서, 적절한 에너지원 에 의해 가교되어 3차원 망상구조를 갖는 하이드로젤을 형성하게 되면 자성 나노입 자들 (120)을 고정시킬 수 있다.
<43> 하이드로젤을 형성할 수 있는 경화성 물질 (110)의 예는 실리콘함유 고분자, 폴리아크릴아마이드, 폴리에틸렌옥사이드, 폴리에틸렌 글리콜 디아크릴레이트, 폴 리프로필렌 글리콜 디아크릴레이트, 폴리비닐피를리돈, 폴리비닐알코올, 폴리아크 릴레이트 및 이들의 공중합체일 수 있다. 예를 들어, 경화성 물질 (110)인 폴리에틸 렌 글리콜 디아크릴레이트 (PEGDA)는 폴리에틸렌글리콜 (PEG) 양 말단에 아크릴레이 트 작용기가 있^서 자유라디칼 증합이 일어날 경우 3차원 구조의 하이드로젤로 가 교될 수 있다. 기타, 경화성 물질 (110)은 액체에서 고체로 변할 수 있는 어떠한 형 태의 매질도 포함할 수 있다.

일 실시예에 따르면, 0FML 기술을 이용한 컬러 코드화된 자성 구조물의 제조 방법이 제공된다. 상기 제조방법은 경화성 물질 및 상기 경화성 물질에 분산된 자 성 나노입자들을 포함하는 조성물로 미세유체 채널올 채우는 단계; 상기 미세유체 채널 내의 상기 조성물에 자기장을 인가하여 상기 자성 나노입자들의 일차원적 사 슬구조를 형성하는 단계; 및 상기 조성물에 패턴화된 자외선을 조사하여 상기 일차. 원적 사슬 구조가 고정된 자유부유 입자를 형성하는 단계를 포함할 수 있다. 상기 미세유체 채널 속을 흐르는 상기 조성물에 대해 마스크 패턴을 변경하며 순차적으 로 상기 패턴화된 자외선을 조사함에 따라 다증 컬러로 코드화할 수 있다. 구조색 을 발현하는 상기 자유부유 입자 각각은 복수의 컬러 도트들을 구비할 수 있다. 상 기 복수의 컬러 도트들 각각은 경화 당시 상기 자기장의 세기에 따라 컬러가 결정 될 수 있다. 상기 경화시 자기장의 세기에 따라 상기 자유부유 입자의 컬러가 결정 되며, 상기 자외선의 패턴에 의해 자유부유 입자의 형태 및 컬러 패턴이 고정될 수 있다.
<-«> 도 4는 다중컬러 코드화된 자성 구조물의 생성과정을 나타낸 개략도이다. 자 기장 조절과 공간적으로 제어된 UV 노출의 협동 작용을 포함하는 순차적인 공정이 사용된다. 먼저 광경화성 '수지 및 상기 광경화성 수지에 분산된 초상자성 나노입자 들을 포함하는 조성물을 준비한다. 다음 상기 조성물로 폴리디메틸실록산 (PDMS) 채 널을 채운다. 이어 외부 자기장을 조절함으로써 상기 조성물의 색상이 튜닝된다. 상기 자기장의 세기에 따라 초상자성 나노입자들에 의한 1D 사슬구조의 주기가 변 하면서 상당하는 파장의 빛을 반사시키게 된다. 일단 외부 자기장에 의해 특정 컬 러 코드가 도입되면 상기 조성물의 일부 영역에 국소적으로 패턴화된 UV를 조사한 다. 상기 패턴화된 UV의 조사를 위해 물리적인 마스크가 아닌 디지털 마이크로미러 장치 (DMD)가 사용될 수 있다. DMD는 컴퓨터로 제어되는 공간적 광조절기 (computer control led spatial light modulator)의 역할을 한다. '도 4에서, UV가 DMD로부터 반사되어 나오면서 패턴화된 UV가 만들어진다. 상기 패턴화된 UV의 조사에 의해 일 부 영역은 특정 컬러로 코드화된다. 다음 단순히 자기장의 세기를 바꾸고 DMD의 패 턴을 변경시키는 방식을 순차적으로 적용하면 연속하여 다음 코드 비트들을 생성 할 수 있다. 이러한 컬러 튜닝 및 고정화 공정은 각 비트를 생성하는 데 대략 수십 분의 일초가 걸리므로 신속하게 수행될 수 있다. 또한 각 컬러의 도입마다 일반적 인 마스크를 사용한 공정에 필요한 재정렬과정이 필요없어 공정이 간단하다. 더욱 이, PDMS 채널 내부의 산소 윤활층 (oxygen lubricant layer), 즉 금지층 (inhibition layer)은 라디칼 광중합에 의해 생성된 미세입자들이 채널 벽에 들러 붙지 않고 플로우 스트림을 따라 움직일 수 있게 한다.
<57> 이러한 성질을 이용함으로써, 광유체적 무마스크 리소그래피 (0FML)를 사용하 여 패턴화된 UV와 함께 여러 가지 레벨의 자기장의 세기 하에 다양한 색상과 모양 으로 코드화된 자성 구조물들이 생성될 수 있다. 즉, 광결정을 함유한 액체 경화성 수지를 미세유체 채널에 주입시키고 여러 가지 자기장 하에 패턴화된 UV에 의해 유 도되는 인 -시튜 광중합에 의해 자성 구조물들이 제조될 수 있다. 상기 자성 구조물 들은 다각형 모양에만 국한되지 않으며, 임의의 소망하는 모양으로 디자인될 수 있 다. 작은 컬러 도트들을 구비한 이종의 코드화된 자성 구조물들이 여러 가지 자기 장 하에서 순차적인 UV 노출에 의해 제조될 수 있다. 이 경우, 그래피컬 코드의 표 현은 색상 및 모양의 유연성 때문에 제한이 없다. <5S> 일 실시예에 따르면, 고체 매질 및 상기 고체 매질 내에 자성 나노입자들이 일정 간격으로 정렬된 자성축을 포함하는 컬러 코드화된 자성 구조물이 제공된다. . 상기 컬러 코드화된 자성 구조물은 상기 정렬된 일정 간격의 크기에 따라 빛의 회 절에 의한 구조색을 발현하는 코드 영역을 구비함으로써 멀티레벨로 코드화된 컬러 코드화될 수 있다.

<62> 또한 컬러 코드화된 자성 구조물을 자기적 제어를 통해 용이하게 컬러 코드 의 정보를 분석할 수 있다 . 일 실시예에 따르면 , 자성 나노입자들의 정렬에 의해 컬러 코드화된 자성 구조물을 함유한 용액을 용기에 도입하는 단계; 상기 용액에 외부 자기장을 인가하여 상기 컬러 코드화된 자성 구조물들을 용기의 벽면으로 수 집하는 단계; 및 상기 컬러 코드의 정보를 분석하는 단계를 포함하되, 상기 자성 구조물 중 컬러 코드가 위치하는 면이 상기 용기의 벽면에 대해 이차원적으로 배열 되도록 하는 자성 구조물의 자기적 제어방법이 제공된다.

<72> 종래의 자성 비드들은 내부에 무작위로 분산된 자성 물질들을 함유하므로 이 러한 움직임의 제어에 한계가 있을 수 있다. 구체적으로는 무작위로 분산된 자성 물질들을 함유한 자성 비드의 경우, 자기력선에 끌려가는 힘만을 받고 각 자성 물 질의 자화 방향을 외부 자기장의 방향으로 돌리려는 힘이 평균화되어 사라질 수 있 다. 반면, 자성축을 구비한 미세 입자들의 경우 일차원적 사슬 구조가 형성되어 각 자성 나노입자의 자화 방향이 외부 자기장과 같은 방향으로 가도록 하는 힘들이 합 쳐져서 전체 입자를 그 방향으로 움직이게 하는 힘으로 작용할 수 있다. 이러한 성 질을 이용해 입자들의 움직임을 제어할 수 있다. 예를 들어, 자성축을 구비한 미세 입자들은 수백만 개의 마이크로 스케일 회전 교반기들로서 서로 협동하여 작동될 수 있다. 도 8B에 도시한 바와 같이, 실제로 이렇게 자성 미세 입자 자체를 자기적 제어를 통한 회전을 이용해 반응을 시켰을 경우, 미세 입자의 희전으로 인해 주위 물질과의 접촉이 증가해 생체물질 반응의 반응성이 증가하는 것을 관찰했다. 자성 미세 입자의 회전 없이 미세 입자와 DNA를 10분 간 반웅시켰을 경우의 미세 입자 탐침 영역의 경계 부분의 형광 세기는 회전을 주어 10분 간 반웅시켰을 경우에 비 해 4배가 넘게 증가하였다. 이러한 반웅성의 증가는 회전 없이 30분 간 반웅시켰을 때보다 훨씬 크게 나타났다.

<8i> 디코더부 (950)는 상기 탐침 분자와 상기 목적 분자가 결합한 상기 자성 구조 물을 관측하고 디코드화한다. 디코더부 (950)는 전하결합소자 (CCD) 카메라를 구비하 여 촬영을 통해 얻은 픽샐의 위치정보와 RGB 정보로부터 각 코드의 위치 및 색상 정보를 동시에 얻을 수 있다. 디코더부 (950)에서의 측정을 위해 자기장 인가부 (940)는 반응이 이루어진 자성 구조물을 수집하기 위해 자기장을 인가할 수 있다.
<82> 자성축을 갖는 자성 구조물은 상기 컬러 코드화된 미세입자 외에도 다중 방 향성 자성축을 갖는 구조물일 수 있다.
<83> 일 실시예에 따르면, 고체 매트릭스; 상기 고체 매트릭스의 일 부분에 위치 하며 고정된 자성축을 구비한 제 1 영역; 및 상기 제 1 영역과 연결되면서 상기 고체 매트릭스의 다른 부분에 위치하며 고정된 자성축을 구비한 게 2 영역을 포함하는 자 성 구조물이 제공된다. 여기서 상기 제 1 영역의 자성축과 상기 제 2 영역의 자성축 의 배향이 서로 다르다. 또한 외부 자기장의 인가에 따라 상기 제 1 영역의 자성축 과 상기 제 2 영역의 자성축이 각각 가지는 이종 자기 이방성에 의하여 상기 제 1 영 역과 상기 제 2 영역은 서로 다른 움직임을 가질 수 있다. 상기 고체 매트릭스는 고 분자 재질로 이루어질 수 있다. 상기와 같이 자성 구조물이 영역별로 방향이 다른 자성축들을 구비함으로써 균일한 자기장 내에서 서로 다른 방향으로 움직이는 마이 크로 액츄에이터를 구현할 수 있다.
<84> 상기 고체 매트릭스의 적어도 한 부분이 외부 물체에 고정될 수 있다. 예를 들어 상기 고체 매트릭스의 적어도 한 부분은 미세유체 채널 내벽에 고정될 수 있 다. 이 경우 캔틸레버 형태의 마이크로 액츄에이터를 구현할 수 있다.
<85> 도 10은 다중 방향성 자성축들을 포함하는 고분자 미세 구조물을 제작하는 과정을 나타낸 도면이다. 각 과정에서 왼쪽은 정면도이고 오른쪽은 측면에서 바라 본 단면도이다. 실험 장비는 먼저 다양한 모양의 고분자 미세 구조물의 제작을 가 능하게 하는 마스크를 사용하지 않는 0FML 장비와 외부 자기장을 생성하고 조절하 기 위한 영구 자석으로 이루어진다. 이 장비로, 순차적인 과정이 진행된다. 먼저, 유리 기판이 미세 구조물의 일부분을 기판에 붙지 않도록 해주는 금지층 (inhibition layer)을 만들어 주는 고분자 물질로 얇게 i팅된다. 이렇게 고분자 물질이 코팅된 유리 기판에 미세 구조물의 일부가 붙게 만들기 위해 원하는 부분의 코팅 물질을 제거한다. 그리고 그 위에 미세유체 채널이 부착되고 그 안으로 경화 성 물질과 자성 나노 입자들이 섞인 액체가 채워진다. 균질한 자기장이 이 미세유 체 채널을 가로질러 형성이 되면, 자성 나노입자들은 생성된 자기력선 방향을 따라 나노 입자의 사슬을 형성하게 된다. 이 상태에서 마스크를 사용하지 않는 0FML 시 스템을 통해 유체의 일부 안에 이 사슬들을 고정하면서 짧은 시간 안에 원하는 모 양으로 광경화하여 원하는 방향으로 자성축이 고정된 제 1 영역을 형성한다. 경화 후에 자기장의 방향을 원하는 방향으로 바꾸고 새로 부여한 자기장의 방향으로 자 성축을 갖는 액체를 새로 광경화시켜 제 2 영역을 형성한다. 이러한 반복적인 자기 력선 방향의 재설정과 고분자 광경화를 통한 고정 과정이 미세 구조물을 완성할 때 까지 반복된다. 구조물의 제작이 완료되면 남은 액체는 미세 구조물을 제어하기 적 합한 액체 환경으로 바꾸어 준다. 이 방법은, 열경화법에 비해 매우 빠른 시간 내 에 이루어지므로 다양한 방향으로 자기 조립된 나노 입자들의 상태를 고해상도로 고분자 내에 고정시킬 수 있고, 그러므로 다증 방향성 자성 나노입자를 포함한 마 이크로 구조물의 완전한 제작까지 걸리는 시간은 매우 짧다.
<86> 도 11은 다양한 형태로 여러 방향으로 정렬된 자성 나노입자를 가진 미세 구 조물의 예들을 나타낸 사진이다. 스케일 바는 모두 200 卿이다. 도 11의 A, B, C 및 D에서 볼 수 있듯이 한 구조물 내에 다양한 자성축들을 가지는 고분자 미세 구 조물올 매우 다양한 모양으로 용이하게 제작할 수 있다. 이러한 방법으로 만든 고 분자 미세 구조물은 기존의 고분자 미세 구조물이 가지고 있던 제어의 한계를 뛰어 넘어 복잡한 움직임을 갖도록 자기적으로 제어할 수 있다.
<87> 도 12는 다양한 방향의 자성축들을 포함한 미세 구조물에 균일한 자기장을 걸어주어 다양한 움직임을 구현한 결과를 나타낸다. 도 12A의 미세 구조물은 서로 수직한 네 방향의 자기 나노입자 사술을 포함하고 있고, 외부 자기장을 걸어주었을 때 각 부분이 서로 다른 방향으로 회전력을 받기 때문에 기존의 미세 구조물에서는 관찰하기 어려웠던 새로운 움직임을 보여준다. 0FML을 이용하여 다양한 모양으로 만드는 것이 가능하고 자기 나노입자 사슬의 방향도 자유롭게 설정할 수 있기 때문 에 도 12B와 12(:에서 보이는 바와 같이 2D 혹은 3D에 상관없이 다양한 움직임을 연 출할 수 있다.

【청구항 1】
자성 나노입자들이 정렬된 자성축을 구비한 자성 구조물올 함유한 용액을 제 공하는 단계; 및
상기 용액에 대해 외부 자기장을 인가하여 상기 자성 구조물의 움직임을 제 어하는 단계를 포함하는 자성 구조물의 자기적 제어방법.

제 1 항에 있어서,
상기 움직임은 상기 자성축이 갖는 자성 토크에 의한 회전 또는 상기 외부 자기장의 밀도가 밀해지는 방향의 힘에 의한 이동인 자성 구조물의 자기적 제어방

제 1 항에 있어서,
상기 자성 구조물의 표면에 탐침 분자가 존재하며, 상기 외부 자기장의 회전 에 따라 상기 자성 구조물이 회전되어 상기 용액 내의 목적 분자와의 반응을 촉진 하는 자성 구조물의 자기적 .제어방법 .
【청구항 4】
자성 나노입자들의 정렬에 의해 컬러 코드화된 자성 구조물을 함유한 용액을 용기에 도입하는 단계 ;
상기 용액에 외부 자기장을 인가하여 상기 컬러 코드화된 자성 구조물들을 용기의 벽면으로 수집하는 단계; 및
상기 컬러 코드의 정보를 분석하는 단계를 포함하되,
상기 자성 구조물 중 컬러 코드가 위치하는 면이 상기 용기의 벽면에 대해 이차원적으로 배열되도록 하는 자성 구조물의 자기적 제어방법 .

고체 매트릭스; 및 상기 고체 매트릭스 내에 고정된 자성축을 포함하되, 상기 자성축은 일정 간격을 두고 일축 방향으로 정렬된 자성 나노입자들의 정렬구조를 가지며, 외부 자기장의 변화에 따라 상기 자성축이 갖는 자성 토크에 의해 힘을 받아 움직이는 자성 구조물.

제 5 항에 있어서,
상기 고체 매트릭스의 표면에 탐침 분자가 고정된 자성 구조물.

고체 매트릭스; 상기 고체 매트릭스의 일 부분에 위치하며 고정된 자성축을 구비한 제 1 영역; 및 상기 제 1 영역과 연결되면서 상기 고체 매트릭스의 다른 부분 에 위치하며 고정된 자성축을 구비한 제 2 영역을 포함하되 ,
상기 제 1 영역의 자성축과 상기 제 2 영역의 자성축의 배향이 서로 다르며, 외부 자기장의 인가에 따라 상기 제 1 영역의 자성축과 상기 제 2 영역의 자성 축이 각각 가지는 이종 자기 이방성에 의하여 상기 제 1 영역과 상기 제 2 영역이 서 로 다른 움직임을 갖는 자성 구조물.

제 7 항에 있어서 ,
상기 고체 매트릭스의 적어도 한 부분이 외부 물체에 고정되는 자성 구조물.

경화성 매질에 분산된 자성 나노입자 조성물을 비혼화성 용매 중에 분산시켜 에멀견을 형성하는 단계;
상기 에멀젼에 자기장을 인가하고 광조영하여 자성축이 내재된 미세구체 형 태의 자성 구조물을 제조하는 단계;
상기 자성 구조물의 표면에 탐침 분자를 고정화하는 단계;
상기 자성 구조물을 목적 분자를 함유한 용매에 흔합하는 단계; 및
외부 자기장을 인가하여 상기 탐침 분자가 고정된 상기 자성 구조물을 회전 또는 이동시킴으로써 목적 분자와의 반웅을 촉진시키는 단계를 포함하는 자성 구조 물의 자기적 제어방법.
【청구항 10]
제 9 항에 있어서,
상기 반웅 후 세기에 기을기가 있는 외부 자기장을 인가함으로써 상기 자성 구조물을 상기 용매로부터 분리해 내는 단계를 더 포함하는 자성 구조물의 자기적 제어방법.
【청구항 11]
탐침 분자를 구비하면서 자성 나노입자들의 정렬에 의해 컬러 코드화된 자성 구조물을 제공하는 제 1 원료 공급부;
목적 분자를 함유한 용액을 제공하는 거 12 원료 공급부;
상기 탐침 분자와 상기 목적 분자가 반웅할 수 있도록 상기 자성 구조물과 상기 용액이 흔합되는 원료 흔합부; 및 상기 자성 구조물을 상기 용액 중에서 회전시키거나 이동시키도록 상기 용액 에 자기장을 가하는 자기장 인가부를 포함하는 자성 구조물의 자기적 제어장치.
【청구항 12]
제 11 항에 있어서,
상기 탐침 분자와 상기 목적 분자가 결합한 상기 자성 구조물을 관측하고 디 코드화하기 위한 디코더부를 더 포함하는 자성 구조물의 자기적 제어장치.
