Abstract

 製造コストが低く検出方法が容易で、非常に高感度かつ信頼性の高い光学式センサー。所定の被検出物を検出するための光学式センサー1であって、所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造2と、凹凸構造2の表面に形成され局在表面プラズモン共鳴を励起可能な金属層3と、金属層3の表面に形成され被検出物を吸着する吸着素子を具備する光学式センサー。この光学式センサー1に被検出物が吸着する前と後の光学特性から被検出物を定量的に検出する。

Description

光学式センサーおよびその製造方法並びに光学式センサーを用いた検出方法
 本発明は、高感度で操作が容易な光学式センサーに関する。
 従来酵素や抗体、DNAなどといった生体分子の高い認識能を利用した被検出物の検出および定量には、PCR(Polymerase chain reaction)法や電気化学的測定手法、ELISA(Enzyme-linked immunosorbent assay)法が主に用いられている。
 電気化学的測定手法は、電極表面へ被検出物を特異的に認識する生体分子を認識素子として固定化し、認識素子へ被検出物が結合した際に生じる電気化学的なシグナル変化を検出するか、または酵素などの標識物質によるシグナル増幅を検出し、定量を行うものである。
 また、ELISA法は、生体分子と被検出物との結合後にあらかじめ標識した酵素による酵素反応や蛍光分子による被検出物の濃度に依存した発色もしくは蛍光を検出する手法である。
 しかしながら、電気化学的手法あるいはELISA法では、被検出物あるいは認識素子として用いる生体分子へ酵素や蛍光物質などの標識剤を必要とする。この標識作業は、従来高い認識能を有する生体分子の認識能を阻害すると同時に、標識作業を必要とするため、感度の低下および操作の煩雑化を招いている。また、高感度に被検出物を検出・定量する際に高額かつ大型の測定装置を必要とする。
 また、簡便な測定手法として注目されているイムノクロマト法もあるが、この場合でも金コロイドやポリスチレン粒子等の修飾が必要である。
 このような背景から、生体分子へ酵素や蛍光分子などを標識することなく(非標識)、被検出物の検出および定量が行える測定手法が望まれてきた。
 そこで、非標識での被検出物の検出および定量が可能な共鳴角のシフトを検出する表面プラズモン共鳴(Surface plasmon resonance : SPR)法や振動数変化を検出する水晶振動子(Quartz crystal microbalance : QCM)法、また、共振周波数変化を検出するカンチレバーによる非標識に被検出物の検出・定量手法が開発されている。
 しかしながら、SPR法の場合、共鳴角シフトを検出するために大型の光学系を構築する必要があるとともに、一連の操作が煩雑である。そのためオンサイトでの被検出物のモニタリングに適さない。また、QCM法では、簡便に測定は可能ではあるが、ノイズが大きく、高感度測定には適さない。そしてカンチレバー法ではデバイス作製が非常に困難という点とともに、簡易測定には適さないという問題がある。
 また、上記測定法とは別に、金や銀、白金などのナノメートルオーダーでの貴金属微粒子に発現する非線形光学現象である局在表面プラズモン共鳴(LSPR)を利用した測定法(例えば、特許文献1参照)や、粒子を三次元的に配列したフォトニック結晶としての機能を有するセンサーを用いた測定方法もある(例えば、特許文献2参照)。
 しかしながら、これらの方法では、微粒子を均一に配列することが非常に難しく再現性も低いため、感度や信頼性の点で未だ問題がある。
 また、従来のセンサーは各センサー同士の再現性が低いため、同一のセンサーを用いて、試料を接触させる前後の光学特性の違いを比較することはできでも、試料に含まれる被検出物の濃度等を変化させた際の光学特性の変化を複数のセンサーを用いて データーベース化し利用するということは困難であるという問題もあった。
 また、センサーの歩留まりやスループットが低く、コストがかかるという問題もあった。

 そこで本発明では、製造コストが低く検出方法が容易で、非常に高感度かつ信頼性の高い光学式センサーを提供することを目的とする。
 本発明は、所定の被検出物を検出するための光学式センサーであって、所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造と、前記凹凸構造の表面に形成され局在表面プラズモン共鳴を励起可能な金属層と、を具備することを特徴とする。
 この場合、前記凹凸構造から反射される光のピーク波長と前記金属層で吸収される光のピーク波長との差が10nm以下である方が好ましい。また、前記凹凸構造は、50~500nmの範囲内の一定の直径を有する柱又は孔を50~1000nmの範囲内の一定のピッチで二次元的に周期配列する方が好ましい。また、前記金属層は、膜厚が5~200nmである方が好ましい。また、前記金属層は、表面に前記被検出物を吸着する吸着素子を具備する方が好ましい。また、前記凹凸構造は、環状オレフィン系樹脂、アクリル樹脂、ポリカーボネート、ビニルエーテル樹脂、フッ素樹脂、ポリエステル樹脂のいずれかを好適に用いることができる。
 また、本発明の別の光学式センサーは、所定の被検出物を検出するための光学式センサーであって、所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造を具備し、当該凹凸構造が環状オレフィン系樹脂、アクリル樹脂、ポリカーボネート、ビニルエーテル樹脂、フッ素樹脂、ポリエステル樹脂のいずれかから選択された樹脂によって形成されたものであっても良い。
 また、上述した本発明の光学式センサーの前記凹凸構造は、インプリント技術によって形成されたものである方が好ましい。また、前記凹凸構造や前記金属層は、前記被検出物によって溶解又は分解する材料によって形成されるものであっても良い。また、前記凹凸構造は、表面に前記被検出物を吸着する吸着素子を具備する方が好ましい。
 また、本発明は、所定の被検出物を検出するための光学式センサーの製造方法であって、所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造を形成する凹凸構造形成工程と、前記凹凸構造の表面に局在表面プラズモン共鳴を励起可能な金属層を形成する金属層形成工程と、を有することを特徴とする。
 この場合、前記凹凸構造形成工程は、前記凹凸構造をインプリント技術によって形成する方が好ましい。また、前記金属層形成工程は、前記金属層を物理気相成長法(PVD)、化学気相成長法(CVD)、メッキ法のいずれかによって形成する方が好ましい。
 また、本発明は、試料中に含まれる被検出物を検出するための検出方法であって、フォトニック結晶として機能する凹凸構造と、前記凹凸構造の表面に形成され局在表面プラズモン共鳴を励起可能な金属層とを有し、前記凹凸構造から反射される光のピーク波長と前記金属層で吸収される光のピーク波長との差が10nm以下である光学式センサーを用意し、前記光学式センサーから反射される光の特性と、前記凹凸構造に前記試料を接触させた後の前記光学式センサーから反射される光の特性と、を比較することにより、前記溶液中に含まれる被検出物を検出することを特徴とする。
 また、本発明の別の検出方法は、試料中に含まれる被検出物を検出するための検出方法であって、フォトニック結晶として機能する凹凸構造と、前記凹凸構造の表面に形成され局在表面プラズモン共鳴を励起可能な金属層とを有し、前記凹凸構造から反射される光のピーク波長と前記金属層で吸収される光のピーク波長との差が10nm以下である光学式センサーを用意し、種々の濃度の被検出物を含有する試料を前記光学式センサーに接触させて、当該光学式センサーから反射される光の特性を検出して作成したデーターベースを用意し、前記凹凸構造に前記試料を接触させた後の前記光学式センサーから反射される光の特性と、前記データーベースに蓄積された情報とを比較することにより、前記溶液中に含まれる被検出物を検出することを特徴とする。
 フォトニック結晶としての機能と局在表面プラズモン共鳴による機能とを複合することにより、非常に高感度かつ信頼性の高い光学式センサーを提供することができる。
 また、ナノインプリント技術を用いて規則性のある凹凸構造が精緻に構築された光学式センサーを製造するので、非常に高感度かつ信頼性の高い光学式センサーを低コストで大量生産することができる。また、これにより異なる光学式センサー間でも光学特性の比較が可能となり、データーベース化によって更に簡易な検出方法を提供することができる。

 本発明の光学式センサー1は、図1に示すように、所定の被検出物を検出するためのもので、所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造2と、凹凸構造2の表面に形成され局在表面プラズモン共鳴を励起可能な金属層3と、で主に構成される。
 凹凸構造2は、フォトニック結晶として機能するものであればどのようなものでも良いが、例えば、所定の直径を有する孔(図1参照)や円柱(図10参照)を所定の間隔で二次元的に周期配列した構造を用いることができる。また、凹凸構造2の間隔は、反射される光のピーク波長が可視領域となるように形成するのが好ましい。具体的には、50~500nmの直径を有する孔又は柱を50~1000nmのピッチで二次元的に周期配列したものを用いることができる。
 また、凹凸構造2の材料としては、フォトニック結晶を形成できるものであればどのようなものでも良いが、好ましくは、大面積で均一な凹凸構造を再現性良く形成できるインプリント技術(熱インプリント、光インプリント等)を適用可能な材料が良い。例えば、環状オレフィン開環重合/水素添加体(COP)や環状オレフィン共重合体(COC)等の環状オレフィン系樹脂、アクリル樹脂、ポリカーボネート、ビニルエーテル樹脂、パーフルオロアルコキシアルカン(PFA)やポリテトラフルオロエチレン(PTFE)等のフッ素樹脂、ポリスチレン、ポリイミド系樹脂、ポリエステル系樹脂等の熱可塑性樹脂を用いることができる。また、エポキシド含有化合物類、(メタ)アクリル酸エステル化合物類、ビニルエーテル化合物類、ビスアリルナジイミド化合物類のようにビニル基・アリル基等の不飽和炭化水素基含有化合物類、等の重合反応性基含有化合物類の重合反応(熱硬化、または光硬化)によって製造される樹脂を用いることもできる。熱的に重合するために重合反応性基含有化合物類を単独で使用することも可能であるし、熱硬化性を向上させるために熱反応性の開始剤を添加して使用することも可能である。更に光反応性の開始剤を添加して光照射により重合反応を進行させて凹凸構造2を形成してもよい。熱反応性のラジカル開始剤としては有機過酸化物、アゾ化合物が好適に使用でき、光反応性のラジカル開始剤としてはアセトフェノン誘導体、ベンゾフェノン誘導体、ベンゾインエーテル誘導体、キサントン誘導体等が好適に使用できる。また、反応性モノマーは無溶剤で使用しても良いし、溶媒に溶解して塗布後に脱溶媒して使用しても良い。樹脂を塗布するための基板としては、目的のセンサー感度を損なわないものであれば任意に選択でき、環状オレフィン系樹脂、アクリル系樹脂やPETのような透明プラスチック基板、石英基板、サファイア基板等、透明基板が特に好ましい。
 なお、パターンの寸法安定性の観点から、樹脂の吸水率は3%以下である方が好ましい。
 また、熱インプリント用の樹脂としては、空気中、昇温速度5℃/minでの示差走査熱量計測定において、酸化に伴う発熱ピークの発熱開始温度(酸化開始温度)が、当該樹脂のガラス転移温度+35℃以上である方が好ましい。これは、熱インプリントによる微細加工において、樹脂劣化により発生する樹脂表面のパーティクル状物を防止できるので、凹凸構造2の転写不良や離型不良等を大幅に低減できるからである。
 この場合、熱インプリント用樹脂としては環状オレフィン系樹脂からなる方が良く、更に好ましくは、式(1)で示される繰返し単位を少なくとも一つ含む熱インプリント用樹脂である方が良い。

(式(1)中、Xはハロゲン原子又は炭素数1~12の炭化水素基である。XとR21(又はR20)は互いにアルキレン基などを介して結合していても良い。p、q、rは0、1又は2である。R1~R21はそれぞれ独立に水素原子、ハロゲン原子、脂肪族炭化水素基、及び脂環族炭化水素基である。また、R11(又はR12)とR13(又はR14)とは炭素数1~5のアルキレン基を介して結合してもよく、また、何れの基も介さずに結合してもよい。但し、R11(又はR12)とR13(又はR14)とが何の基も介さずに結合した場合、結合しない残基である、R11(又はR12)はハロゲン原子又は炭素数1~12の炭化水素基であり、R13(又はR14)は水素原子、ハロゲン原子、脂肪族炭化水素基、又は脂環族炭化水素基である。)
 凹凸構造2をこのように形成することにより、凹凸構造2に光を照射すると、フォトニック結晶の機能によって凹凸構造2に応じた特定の波長の光が反射される。また、凹凸構造2に何らかの物質が吸着し、凹凸構造2に微妙な変化があると、それに応じて反射光の波長や強度が変化する。したがって、その変化によって、所定の被検出物を検出することが可能となる。
 なお、凹凸構造2は、被検出物によって溶解又は分解する材料によって形成されるものであっても良い。例えば、被検出物が極性溶媒の場合には、凹凸構造2をこの極性溶媒に溶解する極性物質により形成し、被検出物が無極性溶媒の場合には、この無極性溶媒に溶解する無極性物質により形成することが考えられる。
 このように形成された光学式センサー1は、凹凸構造2に被検出物を接触させると凹凸構造が溶解又は分解することになるが、この場合にも凹凸構造2に変化が生じることになるので、それに応じて反射光の波長や強度が変化する。したがって、その変化によって、所定の被検出物を検出することが可能となる。
 金属層3は、局在表面プラズモン共鳴を励起するためのもので、例えば、上述した凹凸構造2の表面上に局在した状態で形成される。例えば、凹凸構造2が孔状であれば、少なくとも孔の底面に金属層3を形成すれば良く(図1参照)、柱状であれば、柱の上に金属層3を形成すれば良い(図10参照)。
 金属層3に用いる金属としては、局在表面プラズモン共鳴を励起可能なものであればどのようなものでも良く、例えば、金、銀、銅、白金、アルミニウム等が挙げられる。また、これらを単独で用いても組み合わせて用いても構わない。また、凹凸構造2に用いられる材料との接合性を考慮して、クロム等の介在層を設けても良い。
 金属層3の膜厚は任意であるが、感度を考慮した場合、好ましくは5~200nmにするのが良い。
 金属層3をこのように形成することにより、金属層3に光を照射すると、局在表面プラズモン共鳴によって特定の波長の光が吸収される。また、金属層3に、何らかの物質が吸着し、金属層3に微妙な変化があると、それに応じて吸収光の波長や強度が変化する。したがって、その変化によって、所定の被検出物を検出することが可能となる。
 なお、凹凸構造2、金属層3は、被検出物によって溶解又は分解する材料によって形成されるものであっても良い。例えば、被検出物が極性溶媒の場合には、凹凸構造2をこの極性溶媒に溶解する極性物質により形成し、被検出物が無極性溶媒の場合には、この無極性溶媒に溶解する無極性物質により形成することが考えられる。
 このように形成された光学式センサー1は、凹凸構造2に被検出物を接触させると凹凸構造は溶解又は分解することになるが、この場合にも凹凸構造2や金属層3に変化が生じることになるので、それに応じて反射光や透過光の波長や強度が変化する。したがって、その変化によって、所定の被検出物を検出することが可能となる。
 このように、光学式センサー1にフォトニック結晶として機能する凹凸構造2と、局在表面プラズモン共鳴を励起可能な金属層3とを形成することにより、二つの光学的な特性から被検出物を二重に検出することが可能となり、被検出物の検出精度を上げることが可能となる。
 また、凹凸構造2から反射される光のピーク波長と金属層3で吸収される光のピーク波長との差をできる限り小さく形成しても良い。なぜなら、このように形成すると、凹凸構造2によるフォトニック結晶としての反射光の変化と、金属層3による局在表面プラズモン共鳴としての吸収光の変化とが重なるため、凹凸構造2上の金属層3に被検出物が付着すると、その前後で、光学式センサー1から反射する光や吸収される光の変化が大きくなり光学式センサー1としての感度が向上するからである。
 ここで、二次元フォトニック結晶の原理式は、

 式(3)において発散条件は分極率の分母が0になることであり、このときに共鳴現象が生じる。入射光の波長に比べて十分に小さな金属層に生じる局在表面プラズモン共鳴の共鳴周波数は、金属層の誘電率ε(ω)と金属層周囲の媒質εによって決まる。入射光の波長に比べて金属層の大きさが無視できない場合には、共鳴周波数は、Mieの散乱理論を用いることで導くことが可能であるが、実際には金属層の直径や形状にも依存することが知られている。
 式(3)より、局在表面プラズモン共鳴は、単一の金属層のみでも発現が可能であることから、局在表面プラズモン共鳴の励起には、金属層の大きさ、すなわちピラーの直径のみが関与することになる。
 したがって、凹凸構造2から反射される光のピーク波長と金属層3で吸収される光のピーク波長との差をできる限り小さく形成するためには、まず、金属層3の大きさを決定して局在表面プラズモン共鳴による吸収光のピーク波長を固定し、この波長がフォトニック結晶による反射光のピーク波長と同じになるように、孔同士の距離及び平均屈折率を制御すれば良い。
 なお、凹凸構造2から反射される光のピーク波長と金属層3で吸収される光のピーク波長との差は、反射光や吸収光を受光する受光手段の性能にもよるが、10nm以下が良く、好ましくは5nm以下が良い。
 上述した凹凸構造2及び金属層3のいずれか一方又は両方の表面に被検出物を特異的に吸着する吸着素子を形成するようにしても良い。吸着素子に用いる材料としては、被検出物を特異的に結合でき、かつ、凹凸構造2や金属層3に固定できるものであればどのようなものでも良い。例えば、抗原に対しては抗体、抗体に対しては抗原、ハプテンに対しては抗ハプテン抗体、抗ハプテン抗体に対してはハプテン、DNAに対してはハイブリダイズすることができるDNA又はPNA、ビオチンに対してはアビジンあるいはストレプトアビジン、アビジンあるいはストレプトアビジンに対してはビオチンあるいはビオチン化タンパク、ホルモン受容体(例えばインスリン受容体)に対してはホルモン(例えばインスリン)、ホルモン(例えばインスリン)に対してはホルモン受容体(例えばインスリン受容体)、レクチンに対しては対応する糖鎖、糖鎖に対しては対応するレクチンなどが挙げられる。また、吸着素子は、特異的結合能を有するそれらのフラグメントあるいはサブユニットなどをも含む。さらに、細胞自体を吸着素子として選択可能であり、この場合の被検出物は、該細胞のある一部(受容体等)を特異的に認識するものでよい。
 なお、被検出物とは、凹凸構造2、金属層3、吸着素子の少なくともいずれかと吸着し得るもの、または凹凸構造2、金属層3のいずれかを溶解又は分解し得るものであり、化学物質や生体物質等がある。具体的には、抗原、抗体、レセプター、リガンド、レクチン、糖鎖化合物、RNA、DNA、PNA、ハプテン等が挙げられる。被検出物の機能から分類すると、ホルモン、イムノグロブリン、凝固因子、酵素、薬剤等が含まれる。物質名では、血清アルブミン、マクログロブリン、フェリチン、α-フェトプロテイン、CEA、前立腺特異抗原(PSA)、B型肝炎ウイルス表面抗原(HBsAg)、HIV-1P24などが挙げられる。
 また、本発明の光学式センサーは、金属層3がないフォトニック結晶としての機能を有するだけのものであっても、ナノインプリント技術を適用し易い形状と材料で形成されたものであるという点で従来のセンサーより優れている。なぜなら、ナノインプリント技術は、微細な構造を精密かつ容易に加工することができるため、製造コストが低く、非常に高感度かつ信頼性の高いセンサーを提供することが可能なためである。また、従来の光学式センサーは再現性が低く、製造された光学式センサー同士の誤差が非常に大きいため、異なる光学式センサー間で光学特性の比較をするということは困難であった。しかしながら、ナノインプリント技術で形成された光学式センサーは、同一のものを再現性良く大量生産できるため、異なる光学式センサー間であっても光学特性を比較することができる。したがって、被検出物の濃度による光学特性の変化を データーベース化することにより、より簡易な検出を行うことができるという利点もある。更に、金属層3を形成した光学式センサー1を製造するための中間体としての意義も有している。
 次に、本発明の光学式センサーの製造方法について説明する。
 本発明の光学式センサーの製造方法は、所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造2を形成する凹凸構造形成工程と、凹凸構造2の表面に局在表面プラズモン共鳴を励起可能な金属層3を形成する金属層形成工程と、で主に構成される。
 凹凸構造形成工程は、所定の形状が二次元的に正確に周期配列された凹凸構造であって、フォトニック結晶として機能するものを形成できるのであればどのようなものでも良いが、好ましくはインプリント技術を用いるのが良い。
 熱インプリント技術を用いる場合には、まず、熱可塑性樹脂9からなるフィルム又は基板等を用意し、これに所望のパターンを形成する。
 この熱可塑性樹脂としては、例えば、環状オレフィン重合体や環状オレフィン共重合体(COC)等の環状オレフィン系樹脂、アクリル樹脂、ポリカーボネート、ビニルエーテル樹脂、パーフルオロアルコキシアルカン(PFA)、ポリテトラフルオロエチレン(PTFE)等のフッ素樹脂、ポリエステル系樹脂等を用いることができる。
 凹凸構造2の形成は、まず、ニッケル等の金属やセラミックス、ガラス状カーボン等の炭素素材、シリコンなどから形成された型10を、熱可塑性樹脂9のガラス転移温度以上に加熱し、これを当該熱可塑性樹脂からなるフィルム又は基板に押圧する(図2(a))。次に、当該熱可塑性樹脂をガラス転移温度以下に冷却した後、離型する。これにより、型に形成されたパターン10a(凹凸構造2の反転パターン)が転写される(図2(b))。
 なお、凹凸構造2の形成方法はこれに限られるものではなく、半導体微細加工技術等を適用して形成することも可能である。
 金属層形成工程は、局在表面プラズモン共鳴を励起可能な金属を局在した状態で凹凸構造2上に形成できるものであればどのようなものでも良いが、例えば、物理気相成長法(PVD)により金属を凹凸構造2上に堆積させて形成すれば良い(図2(c))。また、化学気相成長法(CVD)や無電界めっきなどのめっき法等によって形成しても良い。この際、金属層3の膜厚は任意であるが、感度を考慮した場合、好ましくは5~200nmにするのが良い。
 また、図示しないが、凹凸構造2又は金属層3に吸着素子を形成する場合には、疎水性相互作用による物理的吸着や、薬品等による金属層3への化学的結合などを用いて形成すれば良い(吸着素子形成工程)。
 なお、取扱を容易にするために、センサー性能を損なわない形状で筐体や枠を構築してチップ化してもよい。
 次に、本発明の光学式センサーの使用方法(検出方法)について下記に説明する。
(1)まず、被検出物等が吸着されていない状態の光学式センサー1に光21を照射し、反射光22の光学特性、特に、反射光のピーク波長や強度に関する情報を取得する(図1又は図10参照)。光源30としてはどのようなものでも良いが、例えば、タングステン・ハロゲン光源や発光ダイオード、LED、重水素ランプ、有機EL、レーザー等を用いれば良い。また、反射光を検出する光検出手段40には、例えば、マルチチャンネル分光器やCCDイメージセンサー、CMOSセンサー等を用いることができる。また、取得した情報の処理には、コンピュータ等の情報処理手段50を用いれば良い。
(2)次に、被検出物が含まれ得る試料を本発明の光学式センサー1の凹凸構造2上に接触させた後、同様に反射光22の光学特性に関する情報を取得する。
(3)最後に、光学式センサーから反射される光の光学特性と、凹凸構造2に試料を接触させた後の光学式センサーから反射される光の光学特性とを比較することにより、溶液中に含まれる被検出物の有無や量を検出する。
 なお、予め試料に含まれる被検出物の濃度等を変化させた際の反射光の光学特性を光学式センサー1で検出してデーターベース化し、このデータと凹凸構造2に試料を接触させた後の光学式センサーから反射される光の光学特性とを比較することにより、溶液中に含まれる被検出物の有無や量を検出しても良い。この場合、(1)の操作は不要になる。
 また、光学式センサー1に光を照射し、透過光23の光学特性に関する情報を利用しても良い(図3又は図11参照)。この場合、下記のように使用すれば良い。
(1)まず、被検出物等が吸着されていない状態の光学式センサー1に光21を照射し、透過光23の光学特性、特に、透過光23のピーク波長や強度に関する情報を取得する。
(2)次に、被検出物が含まれ得る試料を本発明の光学式センサー1の凹凸構造2上に接触させた後、同様に透過光23の光学特性に関する情報を取得する。
(3)最後に、光学式センサーを透過した光の光学特性と、凹凸構造2に試料を接触させた後の光学式センサーを透過した光の光学特性とを比較することにより、溶液中に含まれる被検出物の有無や量を検出する。
 なお、この場合にも、予め試料に含まれる被検出物の濃度等を変化させた際の透過光の光学特性を光学式センサー1で検出して データーベース化し、このデータと凹凸構造2に試料を接触させた後の光学式センサーを透過する光の光学特性とを比較することにより、溶液中に含まれる被検出物の有無や量を検出しても良い。この場合、(1)の操作は不要になる。
 次に本発明の実施例について説明する。
 熱インプリント技術を用いて、ピラー状パターンが形成されたニッケル金型からガラス転移温度(Tg)が163℃の環状オレフィン系樹脂フィルム(日本ゼオン社製、商品名:ゼオノアフィルムZF-16:厚み100μm)に当該パターンを転写し、直径約250nm、深さ150nmの孔がピッチ500nmで複数配置された凹凸構造を有する光学式センサーAを作製した。
 熱インプリントは次の方法で行った。まず、前記樹脂フィルムに対し、予め205℃に加熱したニッケル金型を圧力2MPaで180秒間押圧する。次に、ニッケル金型と樹脂フィルムを100℃まで冷却した後、この樹脂フィルムからニッケル金型を離型し、孔状の凹凸構造を形成する。なお、熱インプリントには、SCIVAX社のインプリント装置(VX)を使用した。
 次に、光学式センサーAを用いて、濃度の異なる蛋白質の吸着検出を試みた。
 蛋白質としては、妊娠中に産生されるホルモンであるヒト絨毛性ゴナドトロピン(Human chorionic gonadotropin:hCG)を用い、これを超純水によって1pg/ml、10pg/ml、100pg/mlの濃度に調整した。次にこの溶液を光学式センサーAへ滴下し、5分間室温にて静置することにより、ヒト絨毛性ゴナドトロピンを凹凸構造2上へ物理的に吸着させた。静置後は、超純水によって洗浄操作を行い、余剰のhCGを除去し、乾燥させた後に光学特性の評価を行った。
 光学特性の評価は、タングステン・ハロゲン光源(Ocean Optics,LS-1)からの白色光をマルチチャンネルファイバー(Ocean Optics,r200-7UV/VIS)を介して光学式センサーAの凹凸構造2上へ垂直に入射し、その反射光をマルチチャンネル分光器(Ocean Optics,USB4000)によって検出した。なお、光学特性評価は、波長350nm~1000nmの範囲において行った。
 蛋白質の吸着前後における光学特性を図4に示す。また、蛋白質の濃度に対する反射光のピーク波長の変化を図5に、強度の変化を図6に示す。
 その結果、蛋白質の濃度が1pg/mlの場合にはピークシフトが観察され(図5参照)、濃度が10pg/ml、100pg/mlの場合には強度の増加を観察することができた(図6参照)。
 熱インプリント技術を用いて、ピラー状パターンが形成されたニッケル金型からガラス転移温度(Tg)が163℃の環状オレフィン系樹脂フィルム(日本ゼオン社製、商品名:ゼオノアフィルムZF-16:厚み100μm)に当該パターンを転写し、直径約100nm、深さ150nmの孔がピッチ200nmで複数配置された凹凸構造を有するフィルムBと、直径約250nm、深さ150nmの孔がピッチ500nmで複数配置された凹凸構造を有するフィルムCを作製した。
 熱インプリントは次の方法で行った。まず、前記樹脂フィルムに対し、予め205℃に加熱したニッケル金型を圧力2MPaで180秒間押圧する。次に、ニッケル金型と樹脂フィルムを100℃まで冷却した後、この樹脂フィルムからニッケル金型を離型し、孔状の凹凸構造を形成する。なお、熱インプリントには、SCIVAX社のインプリント装置(VX)を使用した。
 次に、フィルムBとフィルムCの凹凸構造の表面に真空蒸着法によって金(Au)からなる厚さ約30nm程度の金属層を形成した。
 なお、真空蒸着は、真空度1.0×10-3Pa以下において、蒸着速度1.0オングストローム以上で金(Au)を堆積させることにより行った。
 次に、フィルムB、フィルムCの金属層表面にHuman fibrinogen(被検出物)を吸着する抗体(吸着素子)を固定し、光学式センサーBおよび光学式センサーCを作製した。
 抗体の固定は次の方法で行った。まず、DDA(4,4'0Dithiodibutylic acid)にてフィルムB、フィルムCの表面へ自己組織化単分子層を形成し、カルボキシル基(-COOH)を導入する。次に、400 mM WSC(Water soluble carbodiimide)/100 mM NHS混合液にてNHS基を導入する。これに、100 mg/mlにPBS(Phosphate buffered saline, pH 7.4)にて調製した抗体溶液を添加し、抗体を固定する。最後に、1M Ethanol amine溶液を添加し、未反応NHS基をブロッキングする。
 次に、光学式センサーB、Cを用いて、濃度の異なるHuman fibrinogenの吸着検出を試みた。Human fibrinogenの吸着は、1fg/ml,1pg/ml,1ng/ml,1μg/ml,1mg/mlの濃度に調製したHuman fibrinogen溶液を光学式センサーB、Cに滴下し、10分間室温にて静置することにより、凹凸構造上の抗体(吸着素子)へ吸着させた。静置後は、超純水によって洗浄操作を行い、余剰のHuman fibrinogenを除去し、乾燥させた後に光学特性の評価を行った。
 光学特性の評価は、タングステン・ハロゲン光源(Ocean Optics,LS-1)からの白色光をマルチチャンネルファイバー(Ocean Optics,r200-7UV/VIS)を介して光学式センサーB、Cの凹凸構造2上へ垂直に入射し、その透過光をマルチチャンネル分光器(Ocean Optics,USB4000)によって検出した。
 光学式センサーBに対するHuman fibrinogenの吸着前後における光学特性を図7に、光学式センサーCに対するHuman fibrinogenの吸着前後における光学特性を図8に示す。
 その結果、光学式センサーB、CのいずれもHuman fibrinogenの濃度に応じて吸光度が変化した。また、Human fibrinogen濃度が1pg/ml以下においても吸光度の変化を観察することができた。
 熱インプリント技術を用いて、ピラー状パターンが形成されたニッケル金型からガラス転移温度(Tg)が163℃の環状オレフィン系樹脂フィルム(日本ゼオン社製、商品名:ゼオノアフィルムZF-16:厚み100μm)に当該パターンを転写し、直径約200nm、深さ150nmの孔がピッチ200nmで複数配置された凹凸構造を有するフィルムDを作製した。
 熱インプリントは次の方法で行った。まず、前記樹脂フィルムに対し、予め205℃に加熱したニッケル金型を圧力2MPaで180秒間押圧する。次に、ニッケル金型と樹脂フィルムを100℃まで冷却した後、この樹脂フィルムからニッケル金型を離型し、孔状の凹凸構造を形成する。なお、熱インプリントには、SCIVAX社のインプリント装置(VX)を使用した。
 次に、フィルムDの凹凸構造の表面にHuman fibrinogen(被検出物)を吸着する抗体(吸着素子)を固定し、光学式センサーDを作製した。
 抗体の固定は次の方法で行った。まず、Anti-human fibrinogen antibody溶液(10μg/ml)に浸漬し1時間静置させ、疎水性相互作用にて抗体を凹凸構造に固定する。これを超純水で洗浄し、乾燥する。
 次に、光学式センサーDを用いて、濃度の異なるHuman fibrinogenの吸着検出を試みた。Human fibrinogenの吸着は、まず、リン酸緩衝液(20mM,pH7.4)に1fg/ml,1pg/ml,1ng/ml,1μg/mlの濃度に調製したHuman fibrinogen溶液を用意し、これに光学式センサーDを1時間浸漬させた。次に、超純水によって洗浄操作を行い、余剰のHuman fibrinogenを除去し、乾燥させた後に光学特性の評価を行った。
 光学特性の評価は、タングステン・ハロゲン光源(Ocean Optics,LS-1)からの白色光をマルチチャンネルファイバー(Ocean Optics,r200-7UV/VIS)を介して光学式センサーB、Cの凹凸構造2上へ垂直に入射し、その透過光をマルチチャンネル分光器(Ocean Optics,USB4000)によって検出した。
 光学式センサーDに対するHuman fibrinogenの吸着前後における光学特性を図9に示す。
 その結果、光学式センサーDは、Human fibrinogenの濃度に応じて吸光度が変化した。また、Human fibrinogen濃度が1pg/ml以下においても吸光度の変化を観察することができた。
 以上より、本発明の光学式センサーは、従来の分析手法では分析が困難であるか、または大型・高額な分析装置を必要としていた1pg/ml以下という微量な濃度であっても、簡便かつ高感度に検出可能であることが示された。

Claims (16)

  1.  所定の被検出物を検出するための光学式センサーであって、
     所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造と、
     前記凹凸構造の表面に形成され局在表面プラズモン共鳴を励起可能な金属層と、
    を具備することを特徴とする光学式センサー。
  2.  前記凹凸構造から反射される光のピーク波長と前記金属層で吸収される光のピーク波長との差が10nm以下であることを特徴とする請求項1記載の光学式センサー。
  3.  前記凹凸構造は、50~500nmの範囲内の一定の直径を有する柱又は孔を50~1000nmの範囲内の一定の間隔で二次元的に周期配列したものであることを特徴とする請求項1又は2記載の光学式センサー。
  4.  前記金属層は、膜厚が5~200nmであることを特徴とする請求項1ないし3のいずれかに記載の光学式センサー。
  5.  前記金属層は、表面に前記被検出物を吸着する吸着素子を具備することを特徴とする請求項1ないし4のいずれかに記載の光学式センサー。
  6.  前記凹凸構造は、環状オレフィン系樹脂、アクリル樹脂、ポリカーボネート、ビニルエーテル樹脂、フッ素樹脂、ポリエステル系樹脂のいずれかによって形成されたものであることを特徴とする請求項1ないし5のいずれかに記載の光学式センサー。
  7.  所定の被検出物を検出するための光学式センサーであって、
     所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造を具備し、当該凹凸構造が環状オレフィン系樹脂、アクリル樹脂、ポリカーボネート、ビニルエーテル樹脂、フッ素樹脂、ポリエステル系樹脂のいずれかから選択された樹脂によって形成されたものであることを特徴とする光学式センサー。
  8.  前記凹凸構造は、インプリント技術によって形成されたものであることを特徴とする請求項1ないし7のいずれかに記載の光学式センサー。
  9.  前記凹凸構造は、前記被検出物によって溶解又は分解する材料によって形成されるものであることを特徴とする請求項1ないし8のいずれかに記載の光学式センサー。
  10.  前記金属層は、前記被検出物によって溶解又は分解する材料によって形成されるものであることを特徴とする請求項1ないし9のいずれかに記載の光学式センサー。

  12.  所定の被検出物を検出するための光学式センサーの製造方法であって、
     所定の形状が二次元的に周期配列されフォトニック結晶として機能する凹凸構造を形成する凹凸構造形成工程と、
     前記凹凸構造の表面に局在表面プラズモン共鳴を励起可能な金属層を形成する金属層形成工程と、
    を有することを特徴とする光学式センサーの製造方法。
  13.  前記凹凸構造形成工程は、前記凹凸構造をインプリント技術によって形成することを特徴とする請求項12記載の光学式センサーの製造方法。
  14.  前記金属層形成工程は、前記金属層を物理気相成長法(PVD)、化学気相成長法(CVD)、メッキ法のいずれかによって形成することを特徴とする請求項12又は13記載の光学式センサーの製造方法。
  15.  試料中に含まれる被検出物を検出するための検出方法であって、
     フォトニック結晶として機能する凹凸構造と、前記凹凸構造の表面に形成され局在表面プラズモン共鳴を励起可能な金属層とを有し、前記凹凸構造から反射される光のピーク波長と前記金属層で吸収される光のピーク波長との差が10nm以下である光学式センサーを用意し、
     前記光学式センサーから反射される光の特性と、前記凹凸構造に前記試料を接触させた後の前記光学式センサーから反射される光の特性と、を比較することにより、前記溶液中に含まれる被検出物を検出することを特徴とする検出方法。
  16.  試料中に含まれる被検出物を検出するための検出方法であって、
     フォトニック結晶として機能する凹凸構造と、前記凹凸構造の表面に形成され局在表面プラズモン共鳴を励起可能な金属層とを有し、前記凹凸構造から反射される光のピーク波長と前記金属層で吸収される光のピーク波長との差が10nm以下である光学式センサーを用意し、
     種々の濃度の被検出物を含有する試料を前記光学式センサーに接触させて、当該光学式センサーから反射される光の特性を検出して作成したデーターベースを用意し、
     前記凹凸構造に前記試料を接触させた後の前記光学式センサーから反射される光の特性と、前記データーベースに蓄積された情報とを比較することにより、前記溶液中に含まれる被検出物を検出することを特徴とする検出方法。
