Abstract

 本発明は、siRNAを投与した生体試料から調製された核酸抽出物を含む被検溶液において、siRNAの二本鎖を解離させ、その後、解離した一本鎖のsiRNAに、siRNAの二本鎖の一方の鎖に相補的な配列を含む蛍光標識プローブDNAをハイブリダイズさせる第一の工程と、 第一の工程後の被検溶液に対して一分子蛍光分析を行い、分析値を取得する第二の工程と、既知の種々の濃度のsiRNAを含む基準溶液を用いて作成された検量線に基いて、第二の工程で取得した分析値からsiRNAの濃度を算出する第三の工程とを含む、一分子蛍光分析によるsiRNAの濃度測定方法に関する。本発明によれば、高い精度、簡便かつ安価でsiRNAの濃度を測定することができる。
