- the PCR protocol was optimized by evaluating different temperatures of the RT step (50 0 C; 55°C; 60 0 C) and the annealing step (52°C; 54°C; 56°C), by testing different MgCl 2 concentrations (l,6mM; 2,OmM; 2,4mM), different primer concentrations (0,4 ⁇ M-0,6 ⁇ M-0,8 ⁇ M) and different annealing times (30s and 60s), by testing different additives (DMSO and glycerine) at several concentrations (5%; 7,5%; 10%) and addition of wheat germ RNA.
- Optimal conditions in terms of sensitivity/specificity were as follows: an RT step of 30 minutes at 55°C, annealing temperature of 54°C and 1,6 mM MgCl 2 . Both DMSO and Glycerine at a concentration of 7.5% can intensify the amplification signal without altering the specificity. Addition of wheat germ RNA increases the aspecificity of the PCR reaction.

Abstract
La présente invention concerne un procédé normalisé d'amplification d'un fragment d'acide nucléique NS5B du VHC de l'un quelconque des génotypes 1 à 6, en tant qu'outil pour l'analyse des variations de séquence qui peuvent être mises en corrélation avec la résistance aux médicaments pour lutter contre le VHC.
