本発明の第 1の態様は、細胞を撮影して取得された細胞画像を処理し、該細胞画 像内に含まれる各細胞の特徴量を測定する特徴量測定部と、測定された特徴量の 分布を作成する特徴量分布作成部と、作成された特徴量の分布において、連続的 に分布する特徴量を有する細胞を特異的な細胞として抽出する特異細胞抽出部とを 備える細胞画像処理装置である。

的な特徴量を有する細胞を抽出することとした力、これに代えて、 2以上の特徴量を 使用して分布を作成することとしてもよい。この場合、 2以上の特徴量を使用して単一 のパラメータを演算し、該単一のパラメータについて分布を作成してもよい。
[0027] 例えば、図 6A、図 6Bに示すように、神経細胞のサイズを、該神経細胞の細胞体の 輪郭に内接する内接円の半径によって表現し、神経細胞の細径突起部の長さを輪 郭の周囲長により表現し、周囲長を内接円半径で規格化することにより新たな単一の ノ ラメータ(周囲長/内接円半径)が得られる。例えば、細胞が完全に円形で突起の 伸張が見られない場合には 1、突起が十分に長い場合には 2. 5のように表現できる。 この値に基づき、特異的な細胞を抽出することとしてもよい。
標準的なモデルとしては、活性の高い神経細胞は、サイズが大きく細径突起の長さ も長いが、活性の低い神経細胞は、サイズが小さく細径突起の長さも短い。したがつ て、パラメータにより分布を求めることにより、標準的なモデルに従わない特異的なも の、例えば、サイズが大きく細径突起の長さが短いものや、サイズが小さく細径突起 の長さが長いものを特異的な細胞として抽出することができる。
[0028] また、 2以上の特徴量を用いて分布を作成した場合でも単発的に存在する特異的 な細胞をノイズとして抽出することができる。すなわち、ノイズの特徴として、複数の特 徴量に関して特異的なものということがある。具体的には、培養容器中の小さいゴミな どはサイズが非常に小さく輝度も高い。そして、これは分布上では単発的なものとし て現れる。一方、通常の細胞も死滅過程ではサイズが小さくなり、輝度も高くなる傾向 があるが、ある程度のサイズを保っている場合が多い。したがって、サイズが非常に 小さく輝度が高いものはゴミなどのノイズとして除去することができる。
[0029] また、本実施形態においては、上述したように、作成された特徴量の分布をグルー プ化して特徴的な細胞を抽出するので、抽出された各細胞の画像を、図 5に示される ように、グループ毎に並べて、あるいはグループ毎にまとめて表示することとしてもよ い。そして、図 5に示されるように特徴的な細胞をその細胞周辺を含め、あるいはその 単一細胞だけを切り取った状態での画像として並べて表示し、観察者がその画像を 選択すると、選択された特徴的な細胞あるいはその細胞周辺の特徴量データなどが 示されるようにしてあよい。
このようにすることで、グループ化された共通の特徴を有する細胞を、観察者に比 較可能に提示することができ、最終的な観察者の目視による判断を容易にすることが できるという利点がある。
[0030] また、特異的なものは単一の原因で起こるのではなぐいくつかの複合的な要素に よって引き起こされるのが一般的であるから、特異的なものとして抽出'表示された細 胞をさらに所定の要素で分類し、分類毎に並べて表示することとしてもよい。
例えば、ある種のホルモンに対して、細胞内のレセプタが発現する反応を見る場合 において、レセプタの発現は、例えば蛍光タンパク遺伝子を導入することで観察でき
[0031] ここで、反応の大きさは発現した蛍光タンパクの量、すなわち、各細胞を蛍光顕微 鏡で観察した際の蛍光量(明るさ)で定量することができる。そこで、特異的なものとし て「蛍光量が少ないもの」、セルサイクルとの関連付けとして「DNA量」により分類す ることが効果的である。
[0032] また、観察者が「細胞が小さ!/、場合、細胞が死にかけて弱って!/、ると考えられ、この 場合、細胞の反応過程が機能しなくなつているはずである」との仮説を立てた場合に 、特異的な細胞として抽出された蛍光量の少ない細胞を、それらのサイズによって分 類したデータを表示することにより、観察者の仮説の検証作業を補助することができ また、抽出された各細胞に対して、形態のパラメータにより分類を行い、いくつかの 特徴的な細胞グループに分類し、図 5と同様の画像表示方法を用いてグループ毎に 細胞を表示することによりユーザの利便性を増すことができる。
Claims
[1] 細胞を撮影して取得された細胞画像を処理し、該細胞画像内に含まれる各細胞の 特徴量を測定する特徴量測定部と、
測定された特徴量の分布を作成する特徴量分布作成部と、
作成された特徴量の分布にお!/、て、連続的に分布する特徴量を有する細胞を特 異的な細胞として抽出する特異細胞抽出部とを備える細胞画像処理装置。
[2] 前記特異細胞抽出部が、不連続に分布する特徴量を有する細胞を除外する請求 項 1に記載の細胞画像処理装置。
[3] 前記特徴量測定部が、各細胞の複数の特徴量を測定し、
前記特徴量分布作成部が、複数の特徴量の相関において特徴量の分布を作成す る請求項 1に記載の細胞画像処理装置。
[4] 前記特異細胞抽出部の前記所定範囲の設定が変更可能である請求項 1に記載の 細胞画像処理装置。
[5] 前記特徴量分布作成部により作成された特徴量の分布において、連続的に分布 する特徴量を有する細胞をグループ化し、グループ毎に表示する表示部を備える請 求項 1に記載の細胞画像処理装置。
[6] 細胞を撮影して取得された細胞画像を処理し、該細胞画像内に含まれる各細胞の 特徴量を測定する測定ステップと、
測定された特徴量の分布を作成する分布作成ステップと、
作成された特徴量の分布にお!/、て、連続的に分布する特徴量を有する細胞を特 異的な細胞として抽出する抽出ステップとを含む細胞画像処理方法。
[7] 前記抽出ステップが、不連続に分布する特徴量を有する細胞を除外する請求項 6 に記載の細胞画像処理方法。
