糖鎖修飾された抗体等有用タンパク質のトランスジエニックニヮトリ卵黄へ の生産

[0001] 本発明は、外来性融合タンパク質遺伝子が導入されたトランスジエニック鳥類または その子孫由来の卵黄中に該外来性融合タンパク質を蓄積させ、それを分離すること よりなる糖鎖付加融合タンパク質の生産方法に関する。さらに詳しくは、卵黄中に蓄 積した該外来性融合タンパク質が、ヒトの糖鎖構造と同等もしくは類似している糖鎖 構造を有することを特徴とする糖鎖付加融合タンパク質の生産方法に関する。
背景技術
[0002] 遺伝子組換え技術の発展により、数多くの生理活性タンパク質が工業生産され、医 薬品などの用途に用いられるようになった。
一般に組換え技術を用いた有用タンパク質の多量生産には、大腸菌などの原核生 物を利用する系、酵母などの真核生物を利用する系が知られており、ある種の単純 な構造を持つタンパク質については成功を収めている。さらに、哺乳動物又は昆虫 の培養細胞を利用する系が広く利用されている。し力もながら、これらには以下に述 ベるような問題点があることも知られている。
[0003] まず、大腸菌等の原核生物を用いた系では、多量体形成及び/又は糖鎖付加とい つた翻訳後修飾が正確に行われないという問題点が知られている。また、酵母を利 用した系についても翻訳後修飾が不完全であり、生じたタンパク質が必ずしも本来の 機能を有しない場合があることが指摘されている。真核生物の培養細胞を用いた系と しては昆虫細胞を用いる系、哺乳動物の培養細胞を用いる系があるが、昆虫細胞に おいては付加される糖鎖の構造が他の生物種と大きく異なるという問題点があり、哺 乳動物の培養細胞は、その維持管理に労力と多大な費用を必要とするにも関わらず 外来性タンパク質の産生量に限界がある。
[0004] これらの問題を克服するために、動物工場が注目されてきた。遺伝子導入(トランスジ エニック)動物を用いて目的タンパク質を生産する技術である。ャギゃヒッジやゥシ等
を用いたトランスジエニック哺乳類を作製し、その乳汁中に目的タンパク質を生産させ る方法が試みられてきた。 目的タンパク質にもよる力 抗体が乳汁中に 10mg/ml発 現したという報告もある(非特許文献 1参照)。しかし、この方法にも、牛海綿症脳症( BSE)の問題や利用可能な哺乳類の個体が大きく生産、飼育、管理が大変であるこ と、性成熟の期間がャギゃヒッジで 8ヶ月、ゥシでは 15ヶ月と長いこと等の問題がある
[0005] そこで、トランスジエニック鳥類を用いて卵中に目的タンパク質を発現させる方法が検 討されている。この方法は、卵の生産性の高さや、 BSE問題が存在しないこと、性成 熟の期間がニヮトリで 5ヶ月と短いこと、個体が小さいため大量の個体の飼育が可能 であること、人工授精法が確立しており大規模のトランスジエニック群を急速に育成可 能であること、一般に卵の中が自然に無菌であること等の利点が挙げられる。
[0006] 哺乳類の抗体には、 IgM IgD IgG IgA IgEの 5種類のクラスが存在することが明 らかとなつている力 ヒトの各種疾患の診断、予防及び治療には、血中半減期が長く 、各種エフェクター機能を有する等の機能特性からヒ HgGクラスの抗体が主として利 用されてレ、る。特に抗体のエフェクター機能である抗体依存性細胞障害活性 (ADC C活性)や補体依存性細胞障害活性(CDC活性)が IgGlサブクラスの抗体でもっと も高いことから(非特許文献 2参照)、リツキサンや セプチンをはじめとするその効 果発現に高いエフェクター機能を必要とする抗腫瘍ヒト化抗体のほとんどはヒ HgGl サブクラスの抗体である。
一般に糖タンパク質を修飾する糖鎖は、生物種によってその糖鎖構造が異なるため 、糖タンパク質の作用は種特異性があることが知られている。したがって、それぞれの 種に最適な有用タンパク質を安価かつ大量に生産する方法の開発が望まれている。 つまり、ヒトへの医薬品を産生するためにはその糖鎖構造がヒト型である必要がある。
[0007] シアル酸は N—ァセチルノイラミン酸が最も多ぐついで N—グリコリルノィラミン酸が 占めている。非特許文献 3では、精製したヒト IgG及びニヮトリ IgGの修飾糖鎖末端の シアル酸が他動物の IgGとは違い、 N—ァセチルノイラミン酸のみで構成されている ことが示されている。また、ニヮトリ IgGにフコースが付加されていないため高い ADC C活性を持つことが示唆されて!/、る。
[0008] 一方で、ニヮトリ卵白中に産生された外来性モノクローナル抗体の糖鎖構造にはシァ ル酸が含まれず、血中安定性が低!/、ことが示されて!/、る(非特許文献 4参照)。 また、無細胞系を用いて糖鎖を人工的に付加させる方法が開示されている力 大量 に生産可能なトランスジエニック動物法とは全く異なるものである(特許文献 1参照)。 さらに細胞を遺伝子変換して、ヒト糖鎖と同等な糖鎖を付加させる方法もある(非特許 文献 5参照) 1 大量に生産可能なトランスジエニック動物法とは全く異なるものであ 特許文献 1 :特開 2002— 238595号公報

発明の開示
発明が解決しょうとする課題
[0009] 外来性タンパク質を生産するあらゆる方法がこれまで開示されてきた力 タンパク質 によっては、修飾される糖鎖構造の違レ、から本来の効果が得られな!/、ことがあった。 これを解消するために、ヒトの糖鎖構造と同等もしくは類似している糖鎖構造を有す る外来性タンパク質を生産する方法が必要とされている。
課題を解決するための手段
[0010] 生産する外来性タンパク質が高!/、生理活性を持ったヒト用医薬品タンパク質となるた めには、糖鎖構造がヒトの糖鎖構造と同等もしくは類似している糖鎖を持たなければ ならない。
本発明者らは鋭意検討を重ねた結果、トランスジエニック鳥類卵黄中に蓄積した外来 性タンパク質が、ヒトの糖鎖構造と同等もしくは類似している糖鎖を有していることを 見出した。
よって、本発明が提供するのは、以下のとおりである。
(1)外来性融合タンパク質遺伝子が導入されたトランスジェニック鳥類またはその子
孫由来の卵黄中に該外来性融合タンパク質を蓄積させ、それを分離することを特徴 とする糖鎖付加融合タンパク質の生産方法。
(2)外来性融合タンパク質が、ヒト免疫グロブリンの定常領域との融合タンパク質であ ることを特徴とする(1)記載の糖鎖付加融合タンパク質の生産方法。
(3)外来性融合タンパク質力 TNFレセプターとヒト免疫グロブリンの定常領域との 融合タンパク質であることを特徴とする(1)記載の糖鎖付加融合タンパク質の生産方 法。
(4)卵黄中に蓄積した外来性融合タンパク質を、プロテインカラムを用いて分離精製 することを特徴とする(1)記載の糖鎖付加融合タンパク質の生産方法。
(5)ひ)〜(4)の!/、ずれ力、 1項記載の方法によって得られる糖鎖付加融合タンパク質
(6)糖鎖全体の 5パーセント以上がモノシアル酸を有する糖鎖であることを特徴とす る(5)記載の糖鎖付加融合タンパク質。
[0011] 以下に本発明を詳細に述べる。
本発明の糖鎖付加融合タンパク質の生産方法は、外来性融合タンパク質遺伝子が 導入されたトランスジエニック鳥類またはその子孫由来の卵黄中に産生される該外来 性融合タンパク質が、ヒトの糖鎖構造と同等もしくは類似している糖鎖を有することを 特徴とする。
本発明で用いられるトランスジエニック鳥類の作製法は、本発明者らによってすでに 確立されているが(国際公開第 04/016081号パンフレット)、これに限定せず、あり とあらゆる公知の方法を用いることができる。
[0012] 生殖細胞に外来性融合タンパク質遺伝子を保有する GOトランスジエニックキメラ鳥類 を、野生型鳥類、 GOトランスジエニックキメラ鳥類又はその子孫と交配させ、孵化した 雛を選別することによって G1トランスジエニック鳥類を得ることができる。 G1トランスジ エニック鳥類は血液、体細胞、精子もしくは卵由来の DNAや RNAを PCR法等によ つて調べることによって体細胞や生殖細胞への遺伝子導入の確認をすることができ る。また、遺伝子導入した外来性融合タンパク質の発現からも判断できる。該タンパク 質の発現は ELISA法や電気泳動法、活性測定等によって調べることができる。
[0013] G2以降の世代のトランスジエニック鳥類は、 G1トランスジエニック鳥類を交配させるこ とによって作製される。交配型は G1トランスジエニックォスと野生型メス、 G1トランスジ エニックメスと野生型ォス、 G1トランスジエニックのォスとメス等が考えられ、さらに子 孫とその親による戻し交配も可能である。なかでも G1ォスと野生型メスの交配は、 1 羽の G1ォスに対し、複数の野生型メスを交配させることができるため、効率の点から 好ましい。
[0014] 本発明で使用する鳥類としては、特に限定されないが、家畜として利用可能な家禽 鳥類であることが好ましい。家禽鳥類としては、ニヮトリ、七面鳥、カモ、ダチョウ、ゥズ ラ、ァヒル等を挙げること力 Sできる。なかでもニヮトリは入手が容易で、産卵種としても 多産であり、卵も大きく大量飼育法が確立しており、ワクチン等の生産でその安全性 が証明されてレ、るため特に好ましレ、。
[0015] 本発明の外来性融合タンパク質は、特に限定されず、例えばヒトモノクローナル抗体 など遺伝子産業上有用な抗体、酵素、成長因子、サイト力イン等を融合させた融合タ ンパク質等が挙げられる。特に糖タンパク質でその活性に糖鎖が重要視されるタンパ ク質が好ましい。なかでも、ヒト免疫グロブリンの定常領域との融合タンパク質である のが好ましぐ TNFレセプターとヒト免疫グロブリンの定常領域との融合タンパク質で ある TNFR—Fcであるのが特に好ましい。外来性融合タンパク質として TNFR—Fc を用いた場合、卵白に生産される 50倍以上の目的タンパク質を卵黄中に蓄積させる こと力 Sでさる。
[0016] また、外来性融合タンパク質としては、近年の遺伝子工学の発展により生産が可能と なった遺伝子組換え抗体でも特にヒト化抗体、キメラ抗体が好ましいが、これに限定 しない。ここで言うヒト化抗体とは、抗体の抗原認識に機能するアミノ酸配列領域以外 をヒトが通常産生する抗体配列とした抗体を指す。ここで言うキメラ抗体とは、ある動 物内で生産される抗体の一部を、異種の動物が産生する抗体の相当領域に置き換 えた抗体を指す。置き換える領域としては特に限定しないが、血中安定性の観点か ら、ヒトが通常産生する抗体の定常領域であることが好ましい。本発明では、卵中の 蓄積がよいことから、ヒト IgGクラスの定常領域を持つ抗体の遺伝子で、特にヒト IgGl のサブクラスの定常領域を持つ抗体の遺伝子を用いた力 これに限定しない。ヒト Ig
Glのサブクラスの定常領域を持つ抗体としては、例えば抗 CD2ヒ HgG抗体等が挙 げられる。
[0017] 医療用組換え抗体には、低分子抗体と称される一群がある。免疫グロブリン IgGには 直接抗原と結合する可変領域(Fv : Fragment of variable region)と呼ばれる V H、 VLのへテロ二量体からなるドメインがあり、この Fvドメインは IgGの約 5分の 1の分 子量でありながら、単独で十分な抗原結合能を持つ。 VH、 VLドメイン間を人工的に ペプチドリンカ一で結合したものが 1本鎖抗体(scFv : single chain Fv)と呼ばれ る低分子抗体で、 VH、 VL単独よりも安定性が向上することが知られていた。
[0018] Powersら(Powers, D. Bら(2000)J Immunol Method. 251 , 123)は、この sc Fvにヒト IgGlに由来する Fc部を融合させることで、血中での安定性が増すことを見 出した。この scFv— Fc抗体は医療用として有用と考えられる力 安価な大量生産シ ステムである大腸菌では生産されな!/、。
上記 scFv—Fc抗体も、本発明の生産方法で得られる外来性融合タンパク質として 好ましいものである。
[0019] GOトランスジヱニックキメラ鳥類作製時に、上記外来性融合タンパク質遺伝子を導入 すれば、生理活性が糖鎖修飾の有無、糖鎖修飾の種類により決まるタンパク質の生 理活性をより高められ、生産が困難だった抗体医薬品やタンパク質を安価に大量に 生産できる。
本発明の卵黄中に蓄積する糖鎖付加融合タンパク質は、それが有する糖鎖構造に 特徴を持つ。糖鎖で修飾された糖タンパク質をヒトへの医薬品とする場合、その糖鎖 構造がヒトの糖鎖構造と同等もしくは類似していない場合、効果の減少及び/又は 消失が起こり、さらにはアレルギー等の投薬副作用の原因にもなりかねない。
すなわち、当該タンパク質がヒトの糖鎖構造と同等もしくは類似している糖鎖構造を 持つことにより、医薬品で問題となる血中安定性を高めることや、シグナル的作用を 強調して標的細胞あるいは標的臓器への取り込みを促進させること、また、本来付与 された生理活性を増大させること、さらには新たな生理活性の付与なども期待するこ と力 Sできる。
[0020] 糖タンパク質糖鎖の体内における挙動に最も関与しているのは、糖鎖非還元末端(
糖タンパク質に結合していない末端)のシアル酸であり、このシアル酸が除去された ァシァ口糖タンパク質は肝臓に認識されて取り込まれ、血中から速やかに消失するこ と力 SAshwell, Morellら(Adv. Enzymd. , 41 , 99, 1974)によってすでに幸告さ れている。医薬品の体内における薬効は一般に有効血中濃度を維持することが重要 であるため、従来のこの知見からの応用は、糖タンパク質のァシァ口化を防いで肝臓 での取り込みを制御し、体内循環における半減期を延長させようとする試みであった
[0021] 本発明者らは、本発明のトランスジエニック鳥類またはその子孫由来の卵中に産生さ れる外来性融合タンパク質で、特に卵黄中に産生される外来性融合タンパク質がシ アル酸を含む糖鎖で修飾されていることを見出した。また、ニヮトリ IgGに付加される 糖鎖のシアル酸は、ヒトの糖鎖と同様に N—ァセチルノイラミン酸のみで構成されて いることカすでに幸告されている(Glycobiology vol. 10 no. 5 pp477— 486, 2000)ことと、卵黄中に蓄積する外来性融合タンパク質は、トランスジェニック鳥類の 全組織で発現したタンパク質力 血流にのって卵黄へ移行したものであることから、 全組織のいずれかにはヒトの糖鎖修飾と同等の修飾をする糖修飾酵素が存在し、そ のため糖鎖構造力 Sヒトと同等もしくは類似していると推測できる。
[0022] 本発明者らは、さらにタンパク質の種類によっては、卵黄中に積極的に蓄積するもの があることを見出した。例えば、融合タンパク質 TNFR— Fcの場合、卵白中の抗体含 有量が 1 a g/ml以下もしくは 0. 1 a g/ml以下なのに対し、卵黄中の抗体含有量 は 0· 1 a g/ml以上、若しくは 1 a g/ml以上である。本発明では TNFR— Fcに関 して測定を行った力、他タンパク質でも積極的に卵黄中に蓄積するものがあることは 容易に推測できる。
[0023] G0トランスジエニックキメラ鳥類は遺伝子導入後、 21日で孵化する。該メス雛は約 5 ヶ月で性成熟をし、一日に約 1個産卵する。また、 G1以降のトランスジエニック鳥類に 関しても同様で、孵化後約 5ヶ月後に一日一個の卵を産卵するようになる。
[0024] これより、本発明で見出された卵黄中に蓄積する外来性融合タンパク質を精製、調 製することで、血中安定性が高ぐ高い生理活性を持った医薬品タンパク質を安定に 大量生産することが可能となる。
卵黄中に蓄積した外来性融合タンパク質は、プロテインカラムを用いて分離精製する ことが好ましい。本発明により卵黄中に蓄積された融合タンパク質は、例えば Fcの特 性を利用したプロテイン Aやプロテイン Gによるァフィ二ティー精製が可能である。他 、 Fcの特性を利用したあらゆる公知の手法により卵黄から精製回収することが可能で ある。また融合タンパク質をプロテアーゼ処理することにより、 目的タンパク質および F cに分離または分離回収することも可能である。
[0025] 上記の糖鎖付加融合タンパク質の生産方法によって得られる糖鎖付加融合タンパク 質もまた、本発明のひとつである。
本発明の糖鎖付加融合タンパク質は、糖鎖全体の 5パーセント以上がモノシアル酸 を有する糖鎖であることが好ましレ、。より好まし!/、モノシアル酸の割合は 5〜20パー セント、更に好ましくは 5〜; 10パーセントである。
発明の効果
[0026] 本発明によれば、ヒトの糖鎖構造と同等もしくは類似している糖鎖構造を有する外来 性融合タンパク質が提供される。その結果として、高い生理活性を有するヒト用医薬 品タンパク質を大量に生産することが可能となる。本発明では卵黄中に蓄積した外 来性融合タンパク質を利用できるため、本発明は利点が高い。
発明を実施するための最良の形態
[0027] 本発明の実施例を以下に示すが、本発明はこれらの実施例により限定されるもので はない。
(実施例 l) scFv—Fc抗体発現 GOトランスジエニックキメラニヮトリの作製
本発明者らはすでに scFv— Fc抗体発現ベクターコンストラクトの作製法、ウィルスべ クタ一調製法、 GOトランスジエニックキメラニヮトリ作製法を開示している(国際公開第 2004/016081号パンフレット)。本発明はこれにしたがって一連の操作を行い、 G 0トランスジェユックキメラュヮトリを作製した。
[0028] (実施例 2) pMSCV/GAATNFR— Fcレトロウイルスベクターの構築
TNFR— Fcを発現するトランスジエニックニヮトリ作製用のウィルスベクターコンストラ タトを以下のように作製した。
TNFRの遺伝子領域は、巿販ベタター、 Invitrogen Clone Collection ID : 518
1070 (Invitrogen社製)を購入し、下記のプライマーにて PCR増幅した。下線は制 限酵素 Xholおよび制限酵素 Sealである。 5' -AATCTCGAGACCATGGCGC CCGTCGC— 3' (配列番号 1) ヒト免疫グロブリン遺伝子の定常領域 hC γ 1領域の取得方法は Hottaらによる文献 に羊ポ田に §己 されており (Journal of Bioscience and Bioengineering Vol. 98 (2004) , No. 4, p. 298— 303)、これを錄型として下記プライマーにて増幅した 。下線は制限酵素 Sealおよび制限酵素 BamHIおよび制限酵素 Clalである。 配列番号 3) 号 4)
上記 PCRで増幅した TNFR遺伝子断片を制限酵素 Xholおよび Sealで消化し、同 様に PCRで増幅した hC y 1遺伝子を制限酵素 Sealおよび BamHIで消化し、 pETB lue— 2ベクター(Novagen)の BamHIと Xholサイトに 3断片のライゲーシヨンにより 糸且み込んだ。このベクターを pET/TNFR— Fcとした。
この pET/TNFR—Fcを制限酵素 Xholおよび Clalで消化し、 Hottaらの文献 (Jou rnal of Bioscience and Bioengineering Vol. 98 (2004) , No. 4, p. 298 — 303)に記載の pMSCV/GAAの制限酵素 SalTおよび制限酵素 Clalサイトにラ ィゲーシヨンすることによって、 pMSCV/GAATNFR— Fcベクターを構築した。
[0029] (実施例 3)TNFR— Fcを発現するトランスジヱニックニヮトリの作製
TNFR— Fcを発現するトランスジエニックニヮトリの作製は、実施例 2に記載のウィル スベクターを基に、国際公開第 2004/016081号パンフレット記載の方法によりパッ ケージング細胞株を樹立した。このパッケージング細胞を基に、国際公開第 2004/ 016081号パンフレットに準じて TNFR—Fcを発現するトランスジエニックニヮトリを作

[0030] (実施例 4)卵中の TNFR— Fc濃度測定用サンプルの調製
実施例 3により誕生した G0トランスジエニックキメラニヮトリを性成熟させ、産卵した卵
を回収した(個体識別番号: # 0314、 # 0513、 # 0608、 # 0709、 # 0711、 # 07 12)。 # 0314に関しては産卵した翌日より 7日目まで採卵を行い、卵中の TNFR—
Fc濃度を定量した。
また、 # 0513、 # 0608、 # 0709、 # 0711、 # 0712ίこ ϋして (ま産^]初日 ίこ採^]し 、卵中の TNFR— Fc濃度を定量した。卵黄は 50% (W/V)、卵白は 10% (V/V) となるように PBSを用いて希釈し、凍結保存して測定用サンプルとした。
[0031] (実施例 5) ELISA法による卵中 TNFR— Fcの定量
PBSで希釈した抗ヒト IgG抗体(コスモバイオ社製)を ELISAプレートに 100 μ g/w ell入れて 4°Cでー晚静置した。 PBS— 0. 05%Tween20溶液を 200 1で各 wellを 3回洗净した後、 PBS - 0. 05%Tween20溶 ί夜一 2%スキムミノレクを l SO l/well 入れた。
室温で 2時間静置後、 wellを 200 1の PBS— 0. 05%Tween20溶液で 3回洗浄し 、実施例 4で調製した卵黄、卵白サンプルを 120 1入れ、 4°Cでー晚静置した。この ELISAプレートを室温に戻した後、 PBS— Tween20溶液で 3回各 wellを洗浄し、 P BS - O. 05%Tween20溶液で希釈した Peroxide (POD)標識抗ヒト IgG抗体(コス モバイオ社製)を 100 1/well入れて室温で 1時間静置した。
[0032] PBS - 0. 05%Tween20溶液で wellを 4回洗浄し、発色液(10mgの o—フエ二レン ジァミン(片山化学工業製)を lmlのメタノールに溶解し、蒸留水で 100mlとしたもの に 10 μ 1の過酸化水素水(和光純薬工業製)を加えて調製した) 100 1を wellに加 えた。 8M硫酸を 50 1添加して反応を止め、 490nmの蛍光強度をプレートリーダー で測定して、標準検量線から濃度を計算した。調製した 1サンプルにっき 3well行い 、平均したものを結果とした。
標準検量線は精製した TNFR— Fcを用いて作製した。
実施例 2で作製した TNFR— Fc融合タンパク質ベクターコンストラクトをリポフエクショ ン法により GP293細胞に導入し、その培養上清を 4°C、 10分間、 3000rpmで遠心し て、固形物を除去した。この上清を冷却しながら攪拌し、 50%飽和となるよう細かく砕 いた硫酸アンモニゥムを徐々に加え(313g硫酸アンモニゥム/ 1000ml水)、タンパ ク質を沈殿させた。これを 4°Cでー晚静置した後、 4°Cで 10分間、 15000rpmで遠心
して沈殿を完全に沈降させ、少量の PBSで溶解した。 2Lの PBSで 3回透析して硫酸 アンモニゥムを除去した。
[0033] 精製用のプロテイン Gカラム(日本ガイシ社製)の初期洗浄を Binding Buffer (Na HPO · 2Η O 1. 56g/l、 NaHPO - 12H O 7. 16g/l) 10mL. Wash Buffer

(酢酸 20%、蒸留水 80%) 10ml, Binding Buffer 10mlの順で行った(流速 2ml/ 分)。 PBSに溶解したタンパク液を lml/分で流し、 TNFR— Fcをカラムに吸着させ た。 Binding Buffer20mlを 1. 7ml/分で流して不要なタンパクを除去し、 Elutio n Buffer (グリシン 7. 507g/l、 2N HC1にて pH2. 5—3. 0に調製)を 1. 5ml/ 分で流して TNFR - Fcを溶出した。
溶出分画を PBS (2Uで 3回透析し、精製 TNFR— Fcとし、波長 280nmの吸光度よ りタンパク濃度を定量した。
[0034] # 0314の定量結果を図 1に、 # 0513、 # 0608、 # 0709、 # 0711、 # 0712の定 量結果を図 2に示す。図 1より採卵日により若干のぶれはあるものの、卵白中に比べ 卵黄中に多く蓄積していることが分かる。また、図 2より卵黄中への蓄積は遺伝子導 入したニヮトリ個体によるものではないことが分かる。
[0035] (実施例 6)卵黄中に蓄積した scFv— Fcの糖鎖解析用のサンプルの調製
実施例 1により誕生した G0トランスジエニックキメラニヮトリを性成熟させ、産卵した卵 を回収した。 3羽の卵より卵黄を 15ml回収した。これに PBSを 60mlカロえ、 4°C、 10分 間、 15000rpmで遠心し、上清を回収した。この過程を 4回繰り返した。これに rProt einA Sepharose Fast Flow (商品名、アマシャム社製)を lml加え、 4°Cで 2時 間撹拌した。その後、 4°C、 5分間、 15000rpmで遠心し、上清を除き、 PBSで 5回洗 净した。これを FISHERbrand screening column (商品名、フィッシャー 'へノレス ケア社製)に添加し、 PBS20mlで洗浄し、 0. 1Mグリシン— HC1 (ρΗ2· 7)で溶出し 、Tris— HCl (pH8. 0)で中和し、 10mM酢酸アンモニゥムで透析し、糖鎖解析用の サンプルとした。
[0036] (実施例 7)卵黄中に蓄積した scFv— Fcの糖鎖解析
実施例 6で調製したサンプルを糖鎖解析した。
サンプル 2. Omgを 10mmol/l炭酸水素アンモニゥム 2Lに対してー晚透析した。こ
れをヒドラジン分解用試験管 6本に採取し、凍結乾燥した。その後ヒドラクラブ S— 20 4 (生化学工業社製)内にセットし、減圧下 50°Cで 12時間乾燥した。無水ヒドラジン 2 mlを減圧下で注入し、 100°Cで 2時間ヒドラジン分解を行った。 2時間後、減圧下でヒ ドラジンを留去した。室温に戻し、ヒドラジン分解用試験管を取り出した。
ヒドラジン分解後の各試験管に酢酸アンモニゥム緩衝液 200 11 1を加えて攪拌した。 これにトリメチルピリジン 10 1を加えて攪拌した。さらに無水酢酸 10 1を加えて攪拌 した。すべて室温で 20分間反応させて、その後遠心エバポレーターで乾固した。 精製用のセルロースカートリッジ (タカラバイオ社製)の初期平衡を行った。
セルロースカートリッジを精製水 10mlで 3回洗浄した。これをセルロースカートリッジ 用溶媒 2 (50vol%エタノール) 10mlで洗浄した。さらに、セルロースカートリッジ用溶 媒 1 (67vol%ブタノール /17vol%エタノール) 10mlで平衡化した。
[0037] これに上記の乾固したサンプル 6本分をまとめて 400 H 1の精製水で溶解し、 lOOOOr pm、 2分で遠心後、上清にエタノール 0. 4mlと 1ーブタノール 1. 6mlを加えて攪拌 した。これを平衡化したカートリッジに注入した。その後カートリッジをセルロースカー トリッジ用溶媒 1 (67vol%ブタノール /17vol%エタノール) 10mlで洗浄し、セル口 一スカートリッジ用溶媒 2 (50vol%エタノール) 2mlで溶出し、溶出液をリアクションチ ユーブ 5本に分注し、遠心エバポレーターで乾固した。
ひとまず、カップリング試料を調製した。
2—ァミノピリシン (PALSTATION Pyridylamination Reagent Kit 糖鎖分析 用、タカラバイオ社製)のバイャル 1本(300mg)に酢酸 100 1を加え、オーブン中 で加熱溶解した。
[0038] 次に、還元試料を調製した。
Borane― dimethylamine complex (PAL¾ ΓΑΤΙΟΝ Pyridylamination Re a gent Kit 糖鎖分析用、タカラバイオ社製) 25. 5mgを秤量し、酢酸 128 1を加え て溶解した。
先ほど、遠心エバポレーターで乾固した各チューブに調製したカップリング試薬 20 1を加えて攪拌した。これを 90°Cのオーブンに揷入し、 60分間反応させた。これに 還元試薬 20 1を加えて攪拌した。これを 80°Cのオーブンに揷入し、 60分間反応さ
せた。
上記 5本に試料をまとめてゲルろ過を実施し、ピリジンアミノ化糖鎖を精製した。以下 に条件を示す。
カラム: TSKgel トヨパール HW— 40F 1. 5cm X 50cm (東ソ一社製) 溶出溶媒: 10mmol/l炭酸水素アンモニゥム
フラクション:約 4ml/チューブ 計 50本
各画分にっレ、て吸光度(290nm)を測定した。
推定ピリジンアミノ化オリゴ糖画分より 200 μ 1ずつ分取し、減圧乾固後、 2 μ 1の精製 水に溶解した。これを TLCにプロットし、風乾後、 2w/v%オルシノール/硫酸を噴 霧、 100°Cで発光させ、ピリジンアミノ化糖鎖の溶出位置を確認した。その後、ピリジ ンァミノ化オリゴ糖画分を減圧乾固し、精製水 500 1に溶解した。
その後、 HPLC (イオン交換)で分析し、シアル酸の有無を確認した。分析条件を以 下に示す。なお、糖鎖標準品についても同様に分析し、溶出位置を比較した。また、 分析に先立ち、精製水 10 1を注入してグラジェント分析し、ゴーストピークのないこ とを確言忍した。標準品はタカラノ ィォ社製の PA— Sugar ChainOOl , 021、 023、 0 24、 012、 013、 014、 015および PA— Glucose Oligomer (DP = 3— 22)を用い た。
装置: LC 1 OAvpシステム(島津製作所社製)
カラム: TSK— gel DEAE- 5PW 7. 5cm X 7. 5mm I. D. (東ソ一社製) カラム温度:室温
移動相:

グラジェント:(1) / (2) = 100 : 0· ν/ν 0分→100 : 0 5分→70 : 30 30分 移動相流速: 0. 5ml/min
試料注入量: 10 1
検出波長: Ex. 310應、 Em. 380nm
HPLC (イオン交換)分析を行った結果より、ァシァ口糖鎖が主成分ではあった力 モ
ノシァ口糖鎖(6. 7%)も検出された。これより、卵黄中に産生される scFv— Fcの糖鎖 にはシアル酸が付加されていることが分かった。この結果を図 3に示す。
[0039] (実施例 8)卵黄中に蓄積した抗 CD2ヒ HgG抗体の糖鎖解析
本発明者らはすでに抗 CD2ヒト IgG抗体発現ベクターコンストラクトの作製法、ウィル スベクター調製法、 GOトランスジエニックキメラニヮトリの作製法を開示している(国際 公開第 2004/016081号パンフレット)。本発明はこれにしたがって一連の操作を 行い、 GOトランスジェユックキメラュヮトリを作製した。
卵黄中に蓄積した抗 CD2ヒ HgG抗体の糖鎖解析用のサンプルの調製は、実施例 6 に準じて行った。また卵黄中に蓄積した抗 CD2ヒ HgG抗体の糖鎖解析は、実施例 7 に準じて行った。
HPLC (イオン交換)分析を行った結果より、ァシァ口糖鎖が主成分ではあった力 モ ノシァ口糖鎖(6. 0%)が検出された。これより、卵黄中に産生される抗 CD2ヒ HgG抗 体の糖鎖にはシアル酸が付加されていることが分かった。

[図 3]実施例 7で行った卵黄中に蓄積した scFv— Fcの糖鎖解析の結果を示す。糖 鎖非還元末端のシアル酸が除去されたァシァ口糖鎖が主成分ではあった力 微量の モノシァロ糖鎖(6. 7%)が検出された。これより、卵黄中に産生される scFv— Fcの 糖鎖にはシアル酸が付加されていることが分かった。
Claims
[1] 外来性融合タンパク質遺伝子が導入されたトランスジエニック鳥類またはその子孫由 来の卵黄中に該外来性融合タンパク質を蓄積させ、それを分離することを特徴とする 糖鎖付加融合タンパク質の生産方法。
[2] 外来性融合タンパク質が、ヒト免疫グロブリンの定常領域との融合タンパク質であるこ とを特徴とする請求項 1記載の糖鎖付加融合タンパク質の生産方法。
[3] 外来性融合タンパク質が、 TNFレセプターとヒト免疫グロブリンの定常領域との融合 タンパク質であることを特徴とする請求項 1記載の糖鎖付加融合タンパク質の生産方 法。
[4] 卵黄中に蓄積した外来性融合タンパク質を、プロテインカラムを用いて分離精製する ことを特徴とする請求項 1記載の糖鎖付加融合タンパク質の生産方法。
[5] 請求項;!〜 4のいずれか 1項記載の方法によって得られる糖鎖付加融合タンパク質。
[6] 糖鎖全体の 5パーセント以上がモノシアル酸を有する糖鎖であることを特徴とする請 求項 5記載の糖鎖付加融合タンパク質。
