改変されたウィルス力プシドタンパク質及びその使用
技術分野
本発明は、 改変された SV40の力プシド夕ンパク質及びその使用に 関する。 本発明によれば、 遺伝子治療、 ドラッグデリバリ一などの 明
応用が期待されるウィルスカブシドタンパク質の細胞指向性を生物 工学的な手法により改変することが出来る。 こう して改変されたゥ ィルスカプシドタンパク質はウィルス書様粒子を形成することが出来 、 このウィルス様粒子に特定の遺伝子核酸や医薬活性成分を封入し て、 それらを特定の細胞組織臓器に導入する事ができる。
背景技術
従来のウィルスベクタ一及びウィルス様粒子による遺伝子導入ま たはドラッグデリバリーは、 ウィルス粒子が本来持つ標的細胞への 効率的な指向性に依存していた。 従ってこれらの方法で各生理活性 物質の導入できる細胞種はウィルス粒子自身の細胞指向性又は組織 指向性により限定されるものであった。 このため、 一般に安定で、 長期保存に耐える、 主に蛋白質と核酸のみで構成されるノンェンべ ロープウィルスにっき、 このウィルス自身のもつ細胞指向性を変え ようという技術開発は以前から行われている。 この方法の一つとし て、 細胞指向性の高い外来性べプチドをウィルスカプシドタンパク 質に挿入することが考えられる。
なお、 マウスポリオ一マウィルス (Stubenrauch K, et al. Bioc hem J. 356:867-873; Shin YC, et al. J Virol. , 77 : 11491-11498 ) 、 及びヒ トパピローマウィルス (Slupetzky K, et al. J Gen Vi
ro l . 82 : 2799-804; S ad eyen JR, e t a l . V i ro l ogy. 309 : 32-40) において、 類似した方法を用いておのおののウィルスカプシドタン パク質にペプチド鎖を挿入し、 一部の挿入体では粒子を形成するも のがあった。 しかしながら挿入されたペプチド鎖の提示、 改変カブ シドタンパク質の粒子形成能、 細胞指向性などについて同時に確認 したものはなく、 また SV40ではこの研究が初めてである。 発明の開示
しかしながら、 ウィルス力プシド夕ンパク質の細胞指向性を改変 するために、 ウィルスカプシドタンパク質のアミノ酸配列への外来 ペプチドの導入によるアミノ酸配列の改変を行なえば、 改変された ウィルスカプシドタンパク質の殆どが粒子形成能を喪失する、 導入 した外来性べプチドがウィルス粒子の表面に提示されないために外 来べプチドの期待される性質が表現されない、 などのおそれがあつ た。 そこで、 本発明は、 霊長類に感染するパポバウィルス、 特にヒ 卜への高い安全性及び高い構造安定性が期待できる SV40ウィルス力 プシドタンパク質中の、 外来性べプチドの適切な挿入部位を見出し 、 その部位に外来性ペプチドを走注することにより、 その改変カブ シドタンパク質から形成される粒子の細胞指向性を制御して、 細胞 への遺伝子または薬剤などの導入の特異性と効率の制御を行おうと するものである。
本発明者らは、 上記の課題を解決すべく、 カプシドタンパク質の 様々な領域 (図 1及び図 2を参照のこと) に外来性ペプチドを挿入 してカプシドタンパク質の改変体を作製し、 そのペプチド鎖を提示 しつつ粒子を形成しうる挿入部位を探索した。 その結果、 約 4 0の 改変体 (その一部を図 2に示す) を解析したところ、 そのうちの 2 種類の改変体、 即ち、 SV40のカプシドタンパク質の M—ループ又は
H I—ループに、 1〜数個のグリシン残基から成るスぺーサ一により 挟まれた外来性べプチドを揷入した改変体が、 粒子を正常に形成し 、 かつペプチド鎖が提示されている挿入部位を見出し (図 2及び図 3 ) 、 この具体的な改変を適用することができるウィルスやスぺー サーアミノ酸の範囲を考慮しながら、 本件発明を完成した。
従って、 本発明は、 霊長類に感染するパポバウィルスの力プシド タンパク質のカプシド粒子表面を形成するアミノ酸残基に、 1〜数 個のスぺ一サーアミノ酸残基により挟まれた外来遺伝子が挿入され ている、 改変された力プシドタンパク質を提供する。 前記霊長類に 感染するパポバウィルスとしては、 例えば SV40、 J CB、 BKVなどが挙 げられる。 また。 前記スぺ一サーアミノ酸残基としては、 例えばグ リシン、 セリン、 ァラニンなどが挙げられる。
より具体的には、 本発明は、 SV40のカプシドタンパク質の]) Eル一 プ及び H Iループの少なく とも一方に、 1〜数個ずつのグリシン残基 により挟まれた外来性ペプチドが挿入されている、 改変された SV40 の力プシドタンパク質を提供する。
前記 SV40カプシドタンパク質は、 好ましくは SV40 VP 1カプシド夕 ンパク質 (配列番号 : 13 ) である。 前記 DEループにおける外来ぺプ チド挿入部位は、 好ましくは、 SV40 VP 1カプシドタンパク質の N -末 端から数えて 127位のァミノ酸と 146位のアミノ酸との間であり、 そ して前記 HIループにおける外来ペプチド挿入部位が、 SV40 VP 1カブ シドタンパク質の N-末端から数えて 268位のアミノ酸と 277位のアミ ノ酸との間である。
更に、 本発明で使用でとる他のカプシドタンパク質として、 J CV (そのアミノ酸配列を配列番号 : 13に示す) 、 BKV (そのアミノ酸 配列を配列番号 : 14に示す) などが挙げられる。 ; T CVの予想される 外来ペプチド挿入可能部位は、 N —末端から数えて 1 19位— 138位の
アミノ酸間、 又は 260位— 269位のアミノ酸間である。 また、 BKVの 予想される外来ペプチド挿入可能部位は、 N—末端から数えて 127 位— 146位のアミノ酸間、 又は 268位— 277位のアミノ酸間である。
前記グリシン残基の数は、 好ましくは、 前記外来性ペプチドの一 方の端につき 1〜 6個であり、 例えば 3〜 6個である。

本発明はまた、 上に記載の改変された SV40のカプシドタンパク質 から形成されるウィルス様粒子を提供する。
本発明は更に、 生理活性物質を封入している、 上記のウィルス様 粒子を提供する。 この生理活性物質は、 例えば、 医薬活性成分、 又 は遺伝子治療用核酸である。

図 1 1は、 Wt粒子、 あるいは Flag (Flag- DE2) または TAT-PTD (T AT-PTD-DE2) を導入された粒子の、 HeLa細胞への導入による細胞指 向性の変化を DEループへの Flag- Tagの導入により VP 立子の細胞内 への導入が阻害されている。 TAT-PTDを DEループに導入された粒子 では導入能力が獲得されている。

ウィルスを用いて医薬活性成分や遺伝子治療用核酸を導入する場 合に問題となるのはその安全性である。 導入べクタ一として用いら れているウィルス自身が病原性を持つものが多く、 またその構造夕 ンパク質に細胞障害性を持つものもある。 一方、 SV40などのパポバ ウィルスの多くは宿主に感染しても殆ど病理的変化が見られること はない。 特に SV40に関してはポリオワクチンの混入ウィルスとして
発見された経緯から、 大規模にヒ トに接種された例があるが、 SV40 の接種が原因での病理疾患発生に関しては否定的な報告がなされて いる。
現在ではこれらのウィルスはヒ 卜には無害あるいは非常に病原性 が低いと考えられており、 そのウィルスのカプシドタンパク質の細 胞指向性を制御することが出来れば、 ヒ トへの遺伝子導入、 ドラッ グデリバリ一等において、 高い安全性が期待できる。 従って、 本発 明のおいては、 SV40の力プシドタンパク質を用い、 好ましくは、 SV 40 VP 1力プシドタンパク質を用いる。
本発明において、 外来性ペプチドが挿入される部位は、 SV40の力 プシドタンパク質の DE—ループ内又は HI—ループ内であり、 SV40 V Iカプシドタンパク質の場合、 —ループは当該タンパク質の N-末 端から数えて 127— 146位にあり、 HI—ループは 268— 277位にある。 好ましくは、 外来性ペプチドは、 SV40 VIカプシドタンパク質の、 D E—ループ内の 137位と 138位の間、 あるいは HI—ループ内の 272位と 275位との間に挿入される。
挿入される外来性ペプチドのそれぞれの端には 1〜数個のスぺー サーアミノ酸、 例えばグリシン、 ァラニン、 セリ ンなどが存在する 必要があり、 好ましくは 1〜 9個、 更に好ましくは 1 〜 6個、 例え ば 3〜 6個のアミノ酸が存在する。 上記のスぺーサーアミノ酸が存 在しない場合、 ウィルス様粒子の形成は困難である。
外来性ペプチドとしては、 上記の部位に挿入することによりウイ ルス様粒子の細胞指向性を変更するものが好ましく、 Flag配列 (As P - Tyr-Lys- Asp- Asp- ASP-ASP- Lys) (配列番号 : 1 ) 、 あるいは抗 原ェピトープであるヒ ト免疫不全症候群 (HIV) の TAT- PTD配列 (Ty r-Gly-Arg-Lys-Lys-Arg-Arg-Gln-Arg-Arg-Arg) (配列番号 : 2 ) 又はインテグリン認識配列 (RGD) を含むアミノ酸配列 3xRGD (Arg -
Gly-Asp-Arg-Gly-Asp-Arg-Gly-Asp) (配列番号 : 3 ) が挙げられ る。
更に、 外来性ペプチドとして、 Myc配列 (Glu-Gln-Lys- Leu - lie- S er-Glu-Glu-Asp-Leu) (配列番号 : 4) 、 HA配列 (Tyr- Pro- Tyr - As P-Val-Pro-Asp-Tyr-Ala) (配列番号 : 5 ) 、 6xHis配列 (His- His - His- His- His-His) (配列番号 : 11) 、 ポリ リジン配列 (Lys- Lys- L ys-Lys-Lys-Lys-Lys-Lys) (配列番号 : 12) 、 Tat- PTD配列 (Arg - L ys-Lys-Arg-Arg-Gln-Arg-Arg-Arg) (配列番号 : 6 ) 、 ポリ リジン 配列(3) (Lys-Lys-Lys) (配列番号 : 7 ) 、 ポリ リジン配列(6) (L ys-Lys-Lys-Lys-Lys-Lys) (配列番号 : 8 ) 、 ポリ リジン(12) (Ly s-Lys -Lys -Lys-Lys-Lys -Lys -Lys -Lys -Lys -Lys -Lys) (配歹 !J番号 : 9 ) 、 NGR配列 (Gin- Gly- Arg) (配列番号 : 10) 等があげられる。 本発明はまた、 上記の改変されたカプシドタンパク質から形成さ れるウィルス様粒子に関する。 実用的には、 このウィルス様粒子に は、 生理活性物質、 典型的には医薬活性物質又は遺伝子治療のため の核酸を封入させる。 医薬活性物質としては、 低分子化合物、 特に 低分子有機化合物、 又はポリペプチド、 タンパク質、 多糖類などの 高分子化合物であっても良い。 このような物質の例として、 ァドリ アマイシン、 シクロフォスフアミ ドなどの抗がん剤、 TNF 、 Granz yme Bなどの抗がん作用のあるタンパク質などがあげられる。
また、 遺伝子治療用の遺伝子 (核酸) としては、 特に限定されず 、 例えばアデノシンデアミダーゼ遺伝子、 p53遺伝子、 等のヒ トの 正常遺伝子、 または siRNAを発現して特定の遺伝子を RNA干渉によつ て発現抑制することができる MA、 そして siRNA自身などがあげられ る。 実施例
次に、 本発明を実施例により更に具体的に説明する。
実施例 1. 改変型 VP1タンパク質の調製
本実施例では、 SV40の外部に提示される ドメインの中で BC-ルー プ、 HI-ループ、 及び DE-ループの 3つのドメイン (図 1 を参照のこ と) を対象にして、 外来のェピトープを揷入した改変型 SV40-VP 1 遺伝子の作製を行った。 SV40— VP1夕ンパク質においてその一部が 外部に提示されるループ上の特定のアミノ酸残基を、 両側に 3残基 ずつグリシンを有する Flag配列に置換した改変型 SV40- VP 1遺伝子 を作製し、 pFastBaclベクタ一に組込み、 大腸菌 DH5 a株に導入した 次に、 Sf9細胞を 1X107個づっ蒔いた 10cm径の組織培養用ディ ッ シュ (IWAKI) に、 それぞれの改変型 SV40のウィルスタンパク質(VP 1)を発現する組換えバキュロウィルスを、 m. o. i. (Multiplicity o f Infection ; 重複感染度) 5〜 10で感染させた。 各々の改変型 VP1 タンパク質を発現する組み換えバキュロウィルスは Invitrogen社製 の Bac to - Bac Baculovirus Express ion Sys temsを用 て作製した 組み換えパキュロウィルスによる感染の 72時間後に、 Sf- 9細胞を 回収し、 冷却したリン酸緩衝液(PBS)により 2回洗浄した。 氷冷した ソニケ一シヨン用緩衝液(20mM Tris- HC1 (pH7.9), (w/v)デォキ シコール酸ナトリウム(D0C)、 2mM PMSF 1 ^ g/mL キモス夕チン (c hymos tat in) 、 ァプロチニン (aprot inin) 、 ロイぺプチン ( leupe pt in) 、 アンチパイン (antipain) 、 ぺプス夕チン (pepstatin) ) 200 こ回収した細胞を懸濁させ、 氷冷しながら 20秒間超音波破砕 し、 その後 20秒氷上に置いた。 この操作を三回繰り返した後に、 そ の細胞破砕液を 15, 000g、 4°Cにて 10分間の遠心分離を行い、 その上 清を回収した。 回収した V P 1タンパク質 2 1を抗 V P 1抗体を用いてゥ
エスタンブロッテイ ングを行い、 その発現を確認した (図 3 ) 。 実施例 2. 改変型 VP 1夕ンパク質の Sf- 9細胞内ウィルス様粒子形 成の解析
発現を確認した後、 SW51Ti用 Open Top Ultraclear Tube (Beckma n)を用いて、 上記の細胞破砕液 2 Lを 20mM Tr i s- HC 1 (pH7.9)で 10倍 希釈し 20 Lとし 20— 40 % (w/v) ショ糖密度勾配遠心 ( 55, 000 rpm 、 4°Cにて 1時間) で分画した。
20 - 40 % (w/v) ショ糖密度勾配遠心により分画したサンプルを 抗 VP 1抗体を用いてウエスタンプロッティ ングを行い、 改変型 VP 1の VLP形成能を解析した。 野生型 VP1夕ンパク質により形成された VP卜 VLP (以下 Wt- VLP) は 8番目から 10番目にそのピークが検出された ( 図 3 ) 。 同様のピークが DE—ル一プ及び HI—ループに Flag配列を揷 入した改変型でも確認できた。
SW51Ti用 Open Top Ultraclear Tube (Beckman)を用いて、 200 /iL とし 20— 50 % (w/v) 塩化セシウム密度勾配遠心 ( 35, 000 rpm, 4°C にて 3時間) で分画した。 超遠心後のサンプルのうち を力一ポ ン-コロジオン膜を張った銅メッシュを上に吸着させた後、 メッシ ュを純水で洗浄し 2— 6%酢酸ウラン溶液で染色し乾燥させた。 観察 は日立社製 H- 7500電子顕微鏡を用いて行った。 前述の DE—ル一プ又 は HI—ループに FLAG配列を挿入した改変型 VP1が Wt_VLPとほぼ同じ 形状と大きさのウィルス様粒子を形成することを確認した。 下の表 1 に改変型- VP1のウィルス様粒子の形成能を解析した結果を示す ( 図 4 ) 。
さらに、 本実施例では改変型 VP1によるウィルス様粒子の形成に 、 ループドメインに挿入した F 1 ag配列の両側に付加するグリシン残 基の数がどのような影響を与えるかを検討した。
前述したループドメインの内、 HI—ループを対象として Flag配列 の両側にグリシン残基 0〜 6残基まで付加させた改変型 VP 1遺伝子
(図 5 ) を、 実施例 1及び実施例 2に記載した方法とと同様にして 作製した。 図 6及び図 7並びに以下の表 2にその結果を示す。

また、 ショ糖密度勾配遠心により分画したサンプルを抗 VP1抗体 を用いてウエスタンプロッティ ングした結果から、 グリシンを 3残 基付加したものの方が 1残基または 2残基付加した場合よりウィル ス様粒子の形成効率が良いこともわかった。 以上のことから Flag配 列を挿入した改変型 VP1がウィルス様粒子を形成するには両側にス ぺーサ一としてグリシン残基が必要であり、 その数は少なく とも 3 残基が好ましいことが示された。

た方法と同様にして回収した改変型ウィルス様粒子 (改変型 VLP) 2 O Lにリ ン酸緩衝液(PBS)を加えて全量 100 こし、 それを 50 X の 抗ー Flag M2ァフィ二ティ一ゲル(Si gma)と共に 4°Cで 1時間穏やかに 攪拌した。 攪拌後、 250iiLのリン酸緩衝液(PBS)で二回洗浄し l^ g/ Lの Flagぺプチドで溶出した。 溶出したサンプルを抗 VP 1抗体を用 いてウエスタンプロッティ ングを行った。 この実験より DEループに Flag配列を挿入した改変型 VP1及び HI—ループに Flag配列を挿入し た改変型 VLPだけが、 抗 Flag抗体に結合することが確認された (図 8 ) 。
実施例 5. 改変型 VP1からのウィルス様粒子における Flag配列の 機能解析
改変型 VLPにおいて、 VLP形成時でも挿入した Flag配列がェピト一 プとしての機能を有するかを検討するために以下のような実験を行 つた。 実施例 1及び実施例 2 に記載した方法と同様にして改変型 VL Pを 60ngずつ調製し、 それぞれを抗- Flag抗体(S igma)および抗 Hi s抗 体(Sigma) 9 xgと共に 4°Cで 1時間穏やかに攪拌した。 攪拌後、 20— 40% (w/v) ショ糖密度勾配遠心により分画しウエスタンブロッテ イ ングを行った。 抗 Flag抗体を用いた場合は、 ピーク画分である 9 及び 10画分に VPIおよび抗 Flag抗体の両方が検出されたのに対し、 抗 His抗体を用いた場合はピーク画分に VPIのみが検出された (図 9 実施例 7. 改変型 VP 1夕ンパク質の大量精製
改変型のウィルス様粒子を大量調製するため、 Sf9細胞を 3X 107 個づっ蒔いた 15cm径の組織培養用ディ ッシュ (IWAKI) 10枚に、 そ れぞれの改変型 SV40のウィルス夕ンパク質(VP1)を発現する組換え バキュロウィルスを、 m.0. i. (Multiplicity of Infection ; 重複 感染度) 5〜10で感染させた。
組み換えバキュロウィルスによる感染 72時間後に、 Sf- 9細胞を回 収し、 冷却したリン酸緩衝液(PBS)により 2回洗浄した。 氷冷したソ 二ケ一シヨ ン用緩衝液(20mM Tris- HC1 (pH7.9), l% (w/v)デォキシ コール酸ナトリウム (DOC), 2mM PMSF, 1 g/mL キモス夕チン、 ァ プロチニン、 ロイぺプチン、 アンチパイン、 ぺプス夕チン) 10mLに 、 回収した細胞を懸濁させ、 氷冷しながら 10分間超音波破砕した。 この操作を 2回繰り返した後に、 その細胞破砕液を 15, 000g、 4°Cに て 10分間の遠心分離にかけ、 その上清を回収した。
SW41Ti用 Open Top Ultraclear Tube (Beckman)に密度の異なる塩 化セシウム溶液(50%, 40¾, 30%, 20% (w/v))を密度の高いほうから 順番に 1.5mlづっ重層し、 その上に、 上記の細胞破砕液 5mLを重層し 、 SW41Tiロータ一を用いて 30, 000 rpm、 4°Cにて 3時間の遠心分離を 行った。 この操作の後に、 密度勾配の中程に白く形成される SV40ゥ ィルスのウィルス様粒子の層を回収し 37% (w/v)塩化セシウム溶液で 希釈した後、 SW55Ti用 Open Top Ultralear Tubeに移し、 これを SW5 5Ti口一夕一で 50, 000 rpm、 4°Cにて 20時間遠心分離を行い、 再度形 成されたウィルス様粒子の層を回収した。
実施例 8. 改変型ウィルス様粒子の機能解析
挿入した各々の mot ifが改変型ウィルス様粒子 (Flag—ウィルス 様粒子、 3xRGD—ウィルス様粒子、 TAT- PTD—ウィルス様粒子) の細 胞に対する吸着と侵入過程にどのような影響を及ぼすのかを検討す る目的で以下のような実験を行った。
10cm径の組織培養用ディ ッシュ (IWAKI) に HeLa細胞 (ヒ ト子宮 頸癌細胞) を lxlO6個になるように蒔いた。 また、 野生型ウィルス 様粒子及び実施例 7において調製した改変型ウィルス様粒子を、 そ の濃度が 1細胞あたり 4χ104、 4χ105、 及び 4χ106個になるようにリ ン酸緩衝液(PBS)で希釈した。
上記のようにして調製した野生型ウィルス様粒子及び改変型ウイ ルス様粒子 (4χ104、 4xl05、 4xl06 VLPs/細胞) に培地を加えて l m L (MEM培地、 10%FBS) としたものを前述した HeLa細胞に接種し、 室温で 15分おきにロッキングした。 ロッキングを 4回行った後、 9 m Lの培地を加え 37°Cで 1時間インキュベートした。 次に、 サンプルを 回収し、 リン酸緩衝液(PBS)で二回洗浄した。 以下、 目的に応じて 2 種類の方法で HeLa細胞を回収した。
( 1 ) スクレーパーによる回収
HeLa細胞表面に付着したウイルス様粒子及び細胞内に取り込まれ たウィルス様粒子の両方を検出するためにスクレーパー (IWAKI) を用いて回収した。
( 2 ) トリプシンによる回収
HeLa細胞内に取り込まれたウィルス様粒子のみを検出する目的で 10cm径の組織培養用ディ ッシュ (IWAKI) 1枚につき 0.25% Trypsi n-EDTA(Gibco) を lmL加え、 37°Cで 5分間インキュベートしディ ッシ ュから剥がれた細胞を回収した。
前述の方法で回収した細胞を冷却したリ ン酸緩衝液(PBS)により 2回洗浄し、 回収した細胞を氷冷しておいたソニケーシヨ ン用緩衝 液 200 / Lに懸濁し、 60秒間超音波破砕した。 これをウエスタンプロ ッティ ング法にかけ、 抗 VP1抗体を用いて、 HeLa細胞に吸着した VP1 タンパク質及び取り込まれた VP 1タンパク質の検出を行った。 表 3 にその結果を示す。
更に、 結果を図 10に示す。 図 10において :
Aは、 TAT- PTDまたは 3xRGDアミノ酸配列を HI- Loopに挿入されたゥ ィルスカプシド夕ンパク質からなる粒子の電子顕微鏡写真を示す。 各粒子は直径約 50nmの約ほぼ球形で、 野生型の粒子 (ここでは示さ ず) と見かけ上差は認められない。 下の黒い線は lOOnmを示す。
B及び Cは、 野生型粒子 (Wt) 、 Flag, 3 xRGD, TAT- PTDの各アミ ノ酸配列を挿入されたウィルスカプシドタンパク質からなる各粒子 を CV- 1細胞に感染させ、 1時間後に細胞を回収し、 細胞に侵入し た粒子量 (VP1の量) を Western blotで検出した。 Inputで示される ように感染させる粒子量を変え、 細胞内で検出される VP1量の比較 を試みた。 Cell associatedとは感染させた細胞を トリプシンなど のタンパク質分解酵素を用いずに回収したもので、 細胞表面に接着 あるいは細胞内に侵入した粒子の検出を試みた。 Trypsinizedとは 感染させた細胞を トリプシン処理で回収する事により、 細胞表面に 結合した粒子を消化し、 細胞内に残る粒子 (VP 1 ) のみの定量を目 的とした。
Wtは Bおよび Cでみられるように細胞に接着 (cell associated参 照) 、 そして侵入(trypsinized参照)できるが、 Flagを導入した粒 子は細胞への接着の段階で阻害がみられる (B) 。 また 3XRGDは Wtと 同様に細胞に接着できるが、 侵入は非常に効率が悪かった (C) 。 一方、 TAT- PTDを導入した粒子は Wtと同様に細胞内に侵入できる事 が明らかであった (C) 。
このことは、 ペプチド鎖の導入自体が元々の力プシドタンパク質 の細胞指向性を失わせる結果になること、 そして TAT- PTDのような ペプチド鎖の導入により、 新たな細胞指向性を獲得できる事を示し ている。
実施例 9. アミノ酸の導入による細胞指向性の変更
野生型粒子 (Wt) 、 あるいは Flag (Flag- DE2) 又は TAT- PTD (Ta t-PTD-DE2) の各アミノ酸を DEループに挿入されたウィルスタンパ ク質からなる各粒子を、 HeLa細胞に感染させ、 2時間後に細胞を回 収し、 細胞に侵入した粒子量 (VP1の量) を Western Blotで検出し た。 細胞の回収の際にトリプシンで細胞表面に付着する粒子を分解
し、 細胞内に侵入した粒子のみを検出した。 Inputで示されるよう にほぼ同量の粒子を細胞導入に用いたが、 Internalizedで示される ように細胞内に検出された VP1の量は Wtに比べ、 Flag- DE2は殆ど検 出できず、 また TAT- PTD- DE2は Wtよりわずかに低いが、 VP1を細胞内 に検出できた。 この結果は、 HIループへのペプチド鎖導入の場合と 同様に、 DEループにペプチド鎖を導入すること自体が元々のカプシ ド夕ンパク質の細胞指向性を失わせ、 そして TAT- PTDのようなぺプ チド鎖の導入により新たな細胞指向性が獲得できる事を示している (図 11) 。
実施例 10. 改変型ウィルス様粒子の機能解析(2)
実施例 8に記載の実験を反復した。 但し、 トリプシンを用いる方 法に代えて、 共焦点顕微鏡を用いて観察を行なった。 HIループに F1 ag配列を挿入した Flag— HI1、 HIループに 3 XRGD配列 (RGD-RGD-RGD ) を挿入した 3XRGD— ΗΠ、 DEループに FFlag配列を挿入した Flag— DE2及び DEループに 3XRGD配列 (RGD- RGD-RGD) を挿入した 3XRGD— DE2、 並びに野生型 VLP (Wt) を試験した。 抗— Vpl抗体により検出 した結果を図 12に示し、 共焦点顕微鏡を用いて観察した結果を図 13 に示す。
図 13において、 白い球状体は細胞核を示し、 その周囲に雲状に分 布する灰白色領域は細胞に取り込まれた VLPの集合を示す。
上記の結果、 HIループ又は DEループに 3XRGD配列 (RGD-RGD- RGD ) を挿入した 3XRGD— DE2は、 野生型 VLPと同等に細胞に取り込まれ た。 結果を表 3にまとめる。

実施例の結果のまとめ
ヒ ト免疫不全症候群ウィルス (HIV) の TAT- PTD 配列 (Tyr- Gly_A rg-Lys-Lys-Arg-Arg-Gln-Arg-Arg-Arg) (配列番号 : 2 ) 又はイン テグリン認識配列 (RGD) を含むアミノ酸配列 3xRGD ( Arg-Gly-Asp- Arg-Gly-Asp-Arg-Gly-Asp) (配列番号 : 3 ) 、 あるいは Flag配列
(Asp- Tyr- Lys- Asp-Asp- Asp- Asp- Lys) (配列番号 : 1 ) などを SV4 0 VPlカプシドタンパク質の表面ループ、 具体的には VP1アミノ酸 13 6番目から 139番目の間、 あるいは 272番目から 275番目の間に導入し 、 そしてその改変型 VP1で粒子を形成させた。 導入したアミノ酸配 列の如何に関わらず、 改変型 VP1は粒子を形成することができた。
また挿入されたアミノ酸配列は、 改変型 VP1を変性しなくても抗 体などで認識させる事ができ、 粒子表面に提示されていた。 この粒 子を用いて CV-1あるいは HeLa細胞などに導入させたところ、 Flagを 導入した改変型粒子は細胞に接着する事が殆どできなかつた。 一方 3 xRGDを導入した粒子は細胞に接着はできたが、 野生型に比べて侵 入の効率は低かった。 ところが、 TAT- PTD配列を導入した粒子は細 胞に接着、 そして侵入できその効率は野生型 VP1で構成された粒子 より少しであるが上回っていた。 つまり野生型のウィルス力プシド タンパク質がもともと持つ細胞指向性がぺプチド鎖揷入によって不 活化されていたが、 挿入したペプチド鎖の機能によって新しい細胞 指向性が与えられた事を示している。
産業上の利用可能性
外来性ぺプチド鎖をウイルスカプシド夕ンパク質の適所に挿入す る事により、 そのカプシドタンパク質の粒子形成を阻害することな く効率的なペプチド鎖の提示を行う ことができた。 このペプチド鎖 の挿入により、 ウィルスカプシドタンパク質がもつ元々の細胞指向 性が阻害され、 そして導入したぺプチド鎖で粒子の細胞指向性を制 御することができた。
Claims
1 . 霊長類に感染するパポバウィルスのカプシ ドタンパク質の力 プシ ド粒子表面を形成するアミ ノ酸残基に、 1 〜数個のスぺーサ一 アミ ノ酸残基により挟まれた外来性ペプチドが挿入されている、 改 変されたカプシドタンパク質。

2 . 前記霊長類に感染するパポバウィルスが SV40、 J CV又は BKVで ある、 請求項 1 に記載の改変されたカプシ ドタンパク質。
3 . 前記スぺ一サーアミ ノ酸残基が、 グリ シン、 セリ ン又はァラ ニンである、 請求項 1 又は 2 に記載の改変されたカプシドタンパク 質。 囲
4 . SV40のカプシド夕ンパク質の DEループ及び HIループの少なく とも一方に、 1〜数個ずつのグリ シン残基により挟まれた外来性べ プチドが揷入されている、 改変された SV40のカプシドタンパク質。
5 . 前記 SV40力プシド夕ンパク質が SV40 VP 1力プシ ドタンパク質 である、 請求項 4に記載の改変された SV40のカプシドタンパク質。
6 . 前記 DEループにおける外来ペプチド挿入部位が、 SV40 VP 1力 プシ ドタンパク質の N -末端から数えて 127位のアミ ノ酸と 146位のァ ミノ酸との間であり、 そして前記 HIループにおける外来ペプチド揷 入部位が、 SV40 VP 1カプシドタンパク質の N-末端から数えて 268位 のアミ ノ酸と 277位のアミ ノ酸との間である、 請求項 5 に記載の改 変された SV40のカプシドタンパク質。
7 . 前記グリ シン残基の数が、 前記外来性ペプチドの一方の端に つき 1 〜 6個である、 請求項 4〜 6 のいずれか 1項に記載の改変さ れた SV40のカプシドタンパク質。
8 . 前記グリ シン残基の数が、 前記外来性ペプチドの一方の端に つき 3 〜 6個である、 請求項 7 に記載の改変された SV40のカプシド
タンパク質。
