Abstract

本発明のAGE−2アプタマーは、グリセルアルデヒド由来終末糖化産物(AGE−2)に結合するがヒト血清アルブミンには結合せず、少なくとも35塩基からなり、そして該塩基中のシトシン含有率が少なくとも35%であるか、または該塩基中のグアニン含有率が少なくとも32%である。本発明のAGE−2アプタマーは、AGE−2を検出するために使用できるので、AGE−2が関与する疾患、例えば、糖尿病性網膜症、糖尿病性腎症、糖尿病性神経障害などの糖尿病合併症、アルツハイマー病などの神経変性疾患、悪性腫瘍の増殖、転移、浸潤などの検出薬・診断薬および予防・治療剤として利用され得る。

本発明は、 ダリセルアルデヒド由来終末糖化産物 (AGE— 2) ァプタマ 一に関する。 背景技術
AGE s (終末糖化産物) は、 グルコースなどの還元糖とタンパク質との 間の非酵素的糖化反応により生成する構造体の総称である。 AGE sは、 老 化や糖尿病に伴って中枢神経などに蓄積し、 あるいは神経 ·感覚器障害ゃ腎 症などの糖尿病による合併症の原因と考えられている。 最近では、 アルッノ、 イマ一病などの神経変性疾患、 悪性腫瘍の増殖、 転移、 浸潤などにも関与す ることが明らかになつている。
AGE sは、 グルコースだけではなく、 グルコースの自動酸化おょぴ分解 産物などの種々の糖から生成される。 このうち、 グリセルアルデヒ ド由来の AGEである AGE— 2 (図 1を参照のこと) は、 AGEレセプター (RA GE) との結合能力が高く、 この RAGEを介して、 糖尿病性網膜症や糖尿 病性腎症などの糖尿病血管合併症の発症や進展に強く関与していることが知 られてレゝる (Yaraagishi S.ら, Biochem. Biophys. Res. Commun. , 2002年, 290卷, 973 - 978頁および Okamoto Τ.ら, FASEB J., 2002年, 16卷, 1928 - 193 0頁) 。 しかし、 その診断法や阻害剤は未だほとんど開発されていない。
AGE sの測定方法については、 これまでに種々検討されている。 最も簡 単には、 AGE sが、 黄褐色であり蛍光を発するので、 その蛍光を測定する 方法がある。 しかし、 AGE sに対する特異性や感度が低いため、 特に生体 試料に対しては適切ではない。 特定の構造の AGE sについては、 HPLC 法、 GCZMS法、 LC/MS法などを用いて定量することができるが、 時 間がかかり、 例えば、 診断におけるような、 多数の試料の分析には適してい ない。
現在のところ、 AGE sのうちで構造が明らかであるカルボキシメチルリ ジン (CML) を認識する抗体 (抗 CML抗体) を用いる免疫学的測定方法 が主として行われている。 し力 し、 感度が低く、 しかも抗体自体が高価であ る。 さらに、 CM Lが生体内では糖化反応ではなく脂質の過酸化によって生 じ ¾こと、 および CMLが酸化ス トレスのマーカーと考えられるようになつ てきたことから、 抗 CML抗体を抗 AGE s抗体として用いることには問題 がある。 このように、 AGE s全体を認識する抗体は、 現在のところ得られ ていない。
近年、 一本鎖 DN Aや一本鎖 RN Aが種々の立体構造をとり、 低分子から タンパク質までの種々の化合物を認識して結合して、 抗体のような機能を有 し得ることが明らかになつている (Ellington A.D.および Szostak J.W., Na ture, 1990年, 346卷, 818 - 822頁および Tuerk C.および Gold L, Science, 1990年, 249卷, 505 - 510頁) 。 このような分子をァプタマ一と称する。 ァプ タマ一は、 ランダム配列の中から S E LEXという選別方法によって得られ 得る (Tuerk C.ら、 前出) 。
アブタマ一は、 試験管内で大量に合成できること、 抗体よりも結合力が強 いものが得られる可能性があること、 安定化できることなどの利点を有する。 そのため、 抗体と同様に、 研究、 検出、 医療などに応用され得る。 医療など への応用が検討されている例としては、 H I V— 1逆転写酵素に対する RN Aァプタマ一 (Kensch 0.ら, J. Biol. Chem. , 2000年, 275卷, 18271-1827 8頁) 、 補体 C 5に対する RNAァプタマ一 (Biesecker G.ら, I瞧 unophar m., 1999年, 42卷, 219- 230頁) 、 CMV感染を防止する RN Aァプタマ一 (Wang J.ら,. 匪, 2000年, 6卷, 571- 583頁) 、 老人性黄斑変性症の治療薬 として開発中の血管内皮細胞増殖因子に对する R N Aァプタマ一 (Ruckman J.ら, J. Biol. Chem. , 1988年, 273卷, 20556-20567頁) 、 糸球体間質増殖 性糸球体腎炎モデルラッ卜に静脈内注射すると症状が緩和される血小板由来 増殖因子に对するアブタマ一 (Floege J.ら, Am. J. Path. , 1999年, 154卷 169- 179頁) 、 ショウジヨウバエ Β 5 2タンパク質の過剰発現による異常を 正常に戻す R N Αァプタマ一 (Shi H.ら, Proc. Natl. Acad. Sci. USA, 19 99年, 96卷, 10033-10038頁). などが報告されている。 発明の開示
本発明は、 A G E— 2に特異的に結合し得るアブタマ一を提供することを 目的とする。
本発明は、 ダリセルアルデヒ ド由来終末糖化産物 (A G E— 2 ) に結合す るがヒ ト血清アルブミンには結合しないアブタマ一を提供し、 該ァプタマ一 1 少なくとも 3 5塩基からなり、 そして該塩基中のシトシン含有率が少な くとも 3 5 %であるか、 または該塩基中のグァニン含有率が少なくとも 3 2 %である。
1つの実施態様では、 上記アブタマ一は、 一本鎖 D N Aである。
他の実施態様では、 上記アブタマ一は、 少なくとも 5 0塩基かつ多くとも 1 2 0塩基からなる。

本発明によれば、 AGE— 2に特異的に結合する AGE— 2アブタマ一が 提供される。 この AGE— 2ァプタマ一を用いれば、 AGE— 2の定性 ·定 量が可能となり、 糖尿病合併症、 神経変性疾患、 悪性腫瘍などの臨床検査薬 として利用され得る。 また、 AGE— 2.アブタマ一は、 AGE— 2阻害活性 を有するので、 抗 AGE— 2剤としても利用され得る。
さらに、 本発明の AGE— 2アブタマ一は、 化学的に安価に合成すること ができる。 ざらに、 このァプタマ一を修飾することによって安定化させるこ と、 あるいはアブタマ一に蛍光または発光ドメインを付加して検出効率を上 げることも可能である。 図面の簡単な説明

アブタマ一とは、 上述のように、 特定の化合物に特異的に結合し得る一本 鎖 DNAまたは一本鎖 RNAである。 本発明においては、 この特定の化合物 が AGE— 2である。 すなわち、 本発明の AGE— 2ァプタマ一は、 AGE _ 2に結合し、 ヒ ト血清アルブミンには結合しなレ、。 一本鎖 DNAまたは一 本鎖 RN Aのいずれであってもよい。
本発明の AGE— 2ァプタマ一は、 少なくとも 35塩基からなり、 好まし くは少なくとも 50塩基かつ多くとも 1 20塩基からなる。 34塩基以下の 場合は、 AGE— 2と結合しない。
本発明の AGE— 2アブタマ一は、 該ァプタマ一を構成する塩基中にシト シンまたはグァニンのレ、ずれか一方を多く含むことが好ましい。 シトシンが 多い場合は、 シトシン含有率は少なくとも 35 %、 少なくとも 40 %あるい は少なくとも 50%であってもよい。 グァニンが多い場合は、 塩基中のグァ 二ン含有率は、 少なくとも 32 %、 少なくとも 35 %あるいは少なくとも 4 0%であってもよい。 このような塩基含有率であることにより、 アブタマ一 は AGE— 2に結合しやすくなる。
具体的な例としては、 配列表の配列番号 1から 24のいずれかに記載の塩 基配列を含む一本鎖 DNAが挙げられる。 これらの一本鎖 DNAは、 54〜 58塩基からなり、 塩基中のシトシン含有率は少なくとも 35°/0である。 別 の具体的な例としては、 配列表の配列番号 25カゝら 41のいずれかに記載の 塩基配列を含む一本鎖 DN Aが挙げられる。 これらの 本鎖 DN Aは、 61 〜66塩基からなり、 塩基中のグァニン含有率は少なくとも 32%である。 本発明の AGE— 2ァプタマ一は、 ァプタマ一を得るための一般的な方法 である SE LEX (Systematic Evolution of Ligands by Exponential enr ichment) 法によって得られ得る。 一本鎖 DNAをライプラリー源とする S E LEX法の概要を図 2に基づいて説明する。 まず、 任意の 2つのプライマ 一配列で挟まれた適当な長さのランダム配列を含むテンプレート DN Aを合 成する。 本発明においては、 ランダム配列の長さは、 35塩基〜 120塩基 が適切である。 このテンプレート DNAを P CR (Polymerase Chain React ion-:ポリメラーゼ連鎖反応) で増幅して、 ランダム DNAアブタマ一ブー ルを得る。 次いで、 このランダム DNAァプタマ一プールを標的物質と会合 させ、 結合しなかった DNAを除去し、 そして結合した DNAァプタマ一を 抽出する。 得られた DNAアブタマ一を、 上記プライマー配列を用いる PC Rによって増幅する。 このとき、 5〜8mMの Mg 2+存在下で PCRを行つ て、 複製の正確性を低下させて変異を導入しやすくすることにより、 標的物 質との会合前の DN Aァプタマ一プールに存在しない新たな DN Aァプタマ 一を含む DN Aァプタマ一プールが得られる。 新たな DN Aァプタマ一は、 結合力がより強い可能性があり、 すなわち、 DNAアブタマ一が進化する可 能性が生じる。 この進化した DNAァプタマ一プールについて、 上記の一連 の操作を 5〜1 5ラウンド繰り返すことによって、 標的物質に特異的に結合 する DNAアブタマ一が得られる。 最終ラウンドの後、 得られた DNAァプ タマ一プールは、 当業者が通常行う操作によってクローニングされ、 次いで 配列決定される。 この S E LEX法における、 テンプレート DN Aの合成、 PCRなどの操作、 ならびにクローユングおよび配列決定は、 当業者が通常 用いる方法によって行われる。 本発明の AGE— 2ァプタマ一は、 このよう にして決定された配列に基づいて、 当業者が通常用いる方法によって化学合 成することができる。
本発明においては、 S E LEX法における標的物質は AGE— 2であり、 好ましくはヒ ト血清アルブミン (HSA) との結合体である。 AGE— 2は、 培養法、 化学合成法などの任意の方法によって合成され得る。 培養法では、 例えば、 ヒ ト血清アルブミン (HSA) を、 D—グルコースと数週間インキ ュペートする力 \ あるいは D—ダリセルアルデヒ ドまたは D—グリコー ァ ルデヒドと数日間インキュベートする。 化学合成法は、 例えば、 Tessierら の方法 (Biochem. J., 2003年, 369卷, 705- 719頁) に従って行われる。 具 体的には、 ァセチル一リジンとダリセルアルデヒ ドとをリン酸緩衝液 (pH 7.-4) 中で混合し、 ざらにジエチレントリアミンペンタ酢酸おょぴ 25% メタノールを加えて、 37°Cで数日間反応させることによって得られる。 得 られた AGE— 2は、 SE LEX法において用いる場合は、 適切な固相 (例 えば、 ビーズなど) に固定化されていることが好ましい。
得られたァプタマ一の AGE— 2との結合親和性は、 AGE— 2が蛍光を 発することを利用して、 AG E— 2にァプタマ一を添加したときの AG E— 2の蛍光強度の減弱によって測定できる。 このようにして、 本発明のァプタ マーのうち、 親和性の強いァプタマ一を選択することができる。
本発明の AGE— 2ァプタマ一は、 AGE— 2に特異的に結合するので、
AGE— 2を検出するために用いることができる。 本発明の AGE— 2ァプ タマ一は、 よ.り安定化させる目的で、 修飾ヌクレオチド ヌクレオチドを用 いて合成されていてもよい。 あるいは、 AGE— 2アブタマ一自体の検出効 率を上げるために、 ァプタマ一に蛍光または発光ドメインが付加されていて もよい。 したがって、 本発明の AGE— 2ァプタマ一は、 AGE— 2検出試 薬として用いられ得る。
代表的には、 本発明の AGE— 2ァプタマ一は、 例えば、 EL I SA、 組 織染色などのように、 抗体が用いられ得る種々の分析において、 抗体と同様 に使用することが可能である.。 DN Aマイクロアレイの技術を利用して、 A GE— 2ァプタマ一チップを作成することもできる。 したがって、 本発明の AGE— 2ァプタマ一は、 AGE— 2検出キットとして提供され得る。
このような AGE— 2の検出に供する試料としては、 代表的には、 種々の 生体試料 (血液、 細胞、 組織など) およびその処理物が挙げられる。 このよ うな試料について、 AGE— 2を検出することにより、 AGE— 2が関与す る疾患、 例えば、 糖尿病性網膜症、 糖尿病性腎症、 糖尿病性神経障害などの 糖尿病合併症、 アルツハイマー病などの神経変性疾患、 悪性腫瘍の増殖、 転 移、 浸潤などを検出ノ診断することができる。 したがって、 本発明の AGE 一 2-アブタマ一は、 上記の疾患の臨床検査薬または診断用キットとして提供 され得る。
また、 本発明の AGE— 2ァプタマ一は、 AGE— 2阻害活性を有するの で、 抗 AGE— 2剤として、 あるいは AGE— 2が関与する疾患、 例えば、 上記の種々の疾患の予防 ·治療剤として使用され得る。
さらに、 本発明の AGE— 2ァプタマ一は、 AGE— 2が関与する疾患の 発症機構の解明などの基礎研究にも用いられ得る。 実施例
(実施例 1 : AGE— 2ァプタマ一の作成) (1— 1 本鎖ランダムオリゴ DNAの作成)
34、 56、 または 72塩基のランダム領域およびその两側にプライマー 部位 (配列番号 42および 43) を含む一本鎖ランダムオリゴ DNAの合成 を、 次のように行った。
まず、 3, 末端のヌクレオチドが 3, 水酸基を介して結合された C PG
(controlled pore glass) 担体をカラムに詰めた。 次いで、 リボースの 5, 位の保護基であるジメ トキシトリチル基をトリクロ口酢酸によって除去 して、 脱トリチル化した。 リボースの 3, 位の水酸基がリン酸シァノエチル アミダイト誘導体である 2番目のヌクレオチドを、 脱トリチルイヒされた 1番 目のヌクレオチドの 5' 水酸基に、 塩基触媒 (テトラゾール) を用いてカツ プリングさせ、 未反応の 5, 水酸基を無水酢酸によってァセチル化した。 2 つのヌクレオチド間の結合を、 ョードを用いて酸化して、 3価のリンから 5 価のリン酸エステルへ変換した。 上記の脱トリル化からリン酸エステルへの 変換までの操作を目的の鎖長になるまで繰り返した。 ランダム部分の配列は、 カップリング反応の際に、 4種のヌクレオチドアミダイ ト混合物を使用して 行った。 反応後、 カラムからアンモニア処理によってオリゴ DNAを切り出 し、 逆相カートリッジカラムによって精製した。 凍結乾燥後、 適量の水に溶 解し、 SELEXライブラリーのテンプレート DNAとした。 (1 _ 2 : AGE— 2の合成)
ヒ ト血清アルブミン (HSA) (Sigma社製) を無菌状態で、 D—グリセ ルアルデヒ ドと 37°Cにて 7日間インキュベートした。 未反応糖を、 リン酸 緩衝生理食塩水 (PBS) の透析によって除去した。 使用する前に、 En d o s p e c y ES— 20 Sシステム (生化学工業) を用いて、 エンドトキ シンがないことを確認した。 (1 -3 :. AGE— 2のビーズへの結合)
上記 1—2で得た AG E― 2を、 PIERCE社の SulfoLink (登録商標) 力ッ プリングゲル (カタログ番号 20401) を使用して製品の指示書に従って、 次のようにビーズに固定ィヒした。
まず、 カップリングゲルをカラムにとり、 カップリング緩衝液 (50mM T r i s— HC 1, 5 mM EDTA, pH8. 5) で平衡ィ匕した。 AG E— 2をカップリング緩衝液に溶解し、 カップリングゲルと混合して、 室温 で 1時間インキュベートした。 反応終了後、 カップリング緩衝液でカラムを 数回洗浄した。 L—システィンをカップリング緩衝液に溶解し、 カップリン グゲルと混合して、 室温で 30分間インキュベートした。 反応終了後、 カツ プリング緩衝液および PB Sでカラムを数回洗浄した。 なお、 反応前および 反応後の吸光度を測定することによって、 AGE— 2の固定化量を算出した。 固定化後のゲル (ビーズ) を小分けし、 使用するまで冷喑所に保存した。

HSAを、 PIERCE社の SulfoLink (登録商標) カップリングゲル (カタ口 グ番号 2040 1) および UltraLink (登録商標) EDC/DADPA固定化キット (カタログ番号 53154) を使用して、 ビーズに固定ィ匕した。

上記 1 _ 1で合成したランダムオリゴ DN Aを铸型にして、 f o r w a r dプライマー (配列番号 42) と r e V e r s eプライマー (配列番号 4 3) による PCR (1サイクル: 94。C, 15秒; 55°0, 15秒; 72°C, 15秒 X I 2サイクル) を行った。 増幅後、 f o r w a r dプライマーのみ を用いた非対称 PCRによりプラス鎖を増幅した (1サイクル: 94°C, 1 5秒; 55°C, 15 ; 72°C, 1 5秒 X 45サイクル) 。 増幅したプラス 鎖をァガロー.スゲル電気泳動によって精製し、 S ELEX用 DNAライブラ リーとした。 この SELEX用 DNAライブラリーを PB Sに溶かし、 9 5°Cで 5分加熱し、 室温に戻した。 次いで、 SELEX用 DNAライブラリ 一と上記 1 _ 3で調製した A G E _ 2を固定化させたビーズとを混合し、 室 温で 30分間インキュベートした。 インキュベート後、 PBSでビーズを数 回洗浄した。 適量の水をビーズに加えて混合し、 100°Cで 5分間加熱し、 AGE- 2ビーズに結合した DNAを解離させて回収した。 回収した DNA と上記 1 _ 4で調製した HS.Aを固定化させたビーズとを混合し、 室温で 1 0分間インキュベートした。 素通りした (HS Aに結合しなかった) DNA を回収して、 エタノール沈殿法によって濃縮した。 濃縮した DN Aを錡型に して、 上記の操作を 5〜1 5ラウンド繰り返した。 この時、 5〜8mMの M g +存在下で PC Rを行って、 変異を導入させた。
(1 -6 : クローニング)
5〜 1 5ラウンド後に得られた DNAを、 f o r w a r dプライマー (配 列番号 42) および r e V e r s eプライマー (配列番号 43) を用いる P CR-によって増幅し、 ァガロースゲル電気泳動によって精製し、 そして AG E— 2特異的 DNAを得た。 この DNAをクローニングベクター (Invitrog en社: Zero Blunt (登録商標) T0P0 (登録商標) PCR Cloning Kit for Sequ encing (カタ口グ番号 K 2875 J 10) ) へ導入し、 次のようにして配列 を決定した。
まず、 AGE— 2特異的 DNA (PCR産物) とクローニングベクター (TOPOベクター) とを混合し、 室温で 5分間インキュベートした。 反応 終了後、 反応液の一部をコンビテントセルに加え、 氷冷下で 30分間インキ ュペートした。 42°Cにて 30秒間ヒートショックした後、 氷上で 2分間冷 却した。 冷却した反応液に、 キット中に含まれている SOC培地を加えて 3 7 °Cにて 1時間培養した。 適量を寒天プレート (50 gZmLアンピシリ ンを含む LB培地) に播種し、 37°Cにて一晩培養した。 無作為に数十個の クローンを選ぴ、 アルカリ法によってプラスミ ド DN Aを調製した。
(1- 7 :配列決定)
上記 1一 6で得られたプラスミ ド DNA中の AGE— 2特異的 DNAの配 列を、 Applied Biosystem社の ABI377を用いて BigDye Terminator Cycle seq uence法によって つ 7こ。
その結果、 34塩基の一本鎖 DNAからは、 AGE— 2に結合するものが 得られなかった。 56塩基の一本鎖 DNAからは、 配列番号 1〜24に示す 54〜58塩基の一本鎖 DNA、 すなわち AG E _ 2ァプタマ一が得られた (表 1) 。 なお、 72塩基の一本鎖 DNAについても AGE— 2に結合する ものが得られたが、 配列決定は行っていない。

表 1に示すように、 得られた AGE— 2ァプタマ一はいずれも、 該ァプタ マーを構成する塩基中のシトシン含有率が 35%以上であった。
(実施例 2 : AGE— 2ァプタマ一の作成)
64塩基のランダム領域おょぴその両側にプライマー部位 (配列番号 42 および 43) を含む一本鎖ランダムオリゴ DNAを铸型に用いたこと以外は、 実施例 1と同様にして AGE— 2ァプタマ一の作成を行った。
その結果、 配列番号 25〜41に示す 61〜66塩基の一本鎖 DNA、 す なわち AGE— 2アブタマ一が得られた (表 2) 。

表 2に示す.ように、 得られた AGE— 2アブタマ一はいずれも、 該ァプタ マーを構成する塩基中のグァニン含有率が 32%以上であった。
次いで、 得られた AGE— 2ァプタマ一の配列に基づいて、 上記 1— 1と 同様にホスホアミダイ ト法により、 各 AGE— 2アブタマ一を化学合成した。
(実施例 3 : AGE— 2ァプタマ一による AGE— 2蛍光阻害実験) まず、 AGE— 2の蛍光特4を測定するために、 蛍光分光光度計 (FP— 777 ; 日本分光) を用いて、 励起波長および消光波長を決定した。 その結 果、 励起は 380 nmおよび消光は 470 n mで最大蛍光を示した。 そこで、 25〜: L 00 μ g/mLの AGE— 2を用いて、 励起波長 380 nmでの蛍 光強度を測定して検量線を作成した (図 3A) 。
次いで、 100 μ g/mLの AGE— 2に対して、 最終濃度 25〜: L 00 nMとなるように各アブタマ一 (配列番号 1〜1 5) を添加し、 蛍光強度を 測定した (図 3 Bおよび Cを参照) 。 ァプタマ一を 25 nM添加したときの AGE— 2の蛍光強度の減弱に基づいて、 ァプタマ一単位モル数 (nmo 1) -あたりに結合する AGE— 2の質量 (n g) を算出した。 結果を表 3に 示す。

(実施例 4 : ゥシ周皮細胞を用いたアポトーシス実験)
ゥシ周皮細胞を、 屠殺後のゥシから取り出して、 ダルベッコ改変イーグル 培地 (Gibco BRL, Rockville, MD) に 20%ゥシ胎児血清 (ICN Biomedicals Inc., Aurora, 0H) を添加した培地中で継代培養した。 培養したゥシ周皮細 胞を、 20 μ gZmLの AGE— 2および Ι Ο Ο μ g/mLのァプタマ一 (配列番号 1〜22) とともに 37 にて 2日間インキュベートした。 コン トロールではァプタマ一の代わりに HS Aを添加して、 ならびに陽性コント ロールでは AGE— 2を添加して、 上記と同様にインキュベートした。 2日 後、 細胞をトリプシンで剥がし、 [3H〕 —チミジンの取込実験を行って、 生存細胞の数をカウントした。 生存細胞数から、 図 4に示すようにして、 ァ ポトーシス阻害率を算出した。 結果を表 4に示す。 表 4

本発明の AGE— 2ァプタマ一は、 AGE— 2を検出するために使用でき るので、 AGE— 2が関与す.る疾患、 例えば、 糖尿病性網膜症、 糖尿病性腎 症、 糖尿病性神経障害などの糖尿病合併症、 アルツハイマー病などの神経変 性疾患、 悪性腫瘍の増殖、 転移、 浸潤などの検出薬 ·診断薬として利用され 得る。 さらに、 本発明の AGE— 2ァプタマ一は、 AGE— 2阻害活性を有 するため、 AGE— 2が関与する疾患、 例えば、 上記の種々の疾患の予防 ' 治療剤として使用され得る。 特に、 糖尿病患者の増加に伴って糖尿病合併症 も増加しているため、 早期発見および治療に有用である。 また、 本発明の A GE— 2アブタマ一は、 AGE— 2が関与する疾患の発症機構の解明などの 基礎研究にも用いられ得る。

Claims

請求の範囲
1. ダリセルアルデヒド由来終末糖化産物 (AGE— 2) に結合するがヒ ト 血清アルブミンには結合しないァプタマ一であって、 該ァプタマ一が、 少な くとも 35塩基からなり、 そして該塩基中のシトシン含有率が少なくとも 3
5%であるか、 または該塩基中のグァニン含有率が少なくとも 32%である、 ァプタマ一。
2. 前記アブタマ一が、 一本鎖 DNAである、 請求項 1に記載のアブタマ一。
3. 前記アブタマ一が少なくとも 50塩基かつ多くとも 120塩基からなる、 請求項 1または 2に記載のアブタマ一。
4. 前記塩基中のシトシン含有率が少なくとも 40%である、 請求項 1から 3のいずれかの項に記載のァプタマ一。
5. -前記塩基中のシトシン含有率が少なくとも 50%である、 請求項 4に記 載のァプタマ一。
6. 前記塩基中のグァニン含有率が少なくとも 35%である、 請求項 1から
3のいずれかの項に記載のァプタマ一。
7. 前記塩基中のグァニン含有率が少なくとも 40%である、 請求項 6に記 載のアブタマ一。
8. 前記一本鎖 DNAが、 配列表の配列番号 1から 24のいずれかに記載の 塩基配列を含む、 請求項 2に記載のアブタマ一。
9. 前記一本鎖 DNAが、 配列表の配列番号 25から 41のいずれかに記載 の塩基配列を含む、 請求項 2に記載のアブタマ一。
10. 請求項 1から 9のいずれかの項に記載のァプタマ一を含む、 AGE— 2検出試薬。
