Abstract

 本発明は、血流障害による虚血および虚血後の再灌流によって生じる神経細胞の遅発性細胞死を防御または救済する方法、ならびに当該遅発性細胞死を防御または救済することによって当該細胞死に起因する脳血管疾患を治療するための薬剤に関するものである。さらに本発明は、上記細胞死を評価する方法、および遅発性神経細胞死を防御または救済する因子をスクリーニングする方法に関するものである。

[0003] 脳の虚血 ·再灌流の後に迅速には生じな!/、が数日後に生じる細胞死、 、わゆる遅 発性神経細胞死の存在が知られて!/ヽる (非特許文献 5および非特許文献 6を参照の こと)。本発明者らは、核酸およびプロタミンを主成分とする魚類白子抽出物が、虚血 により引き起こされる遅発性の神経細胞死に対して抑制効果があることを見出してい る(特許文献 1を参照のこと)。この遅発性神経細胞死は、その発生が遅いことから救 済可能な細胞死であると考えられて 、る。この遅発性細胞死はアポトーシスの現象を 含むことが知られている(非特許文献 7を参照のこと)。培養細胞においてミトコンドリ ァ仲介性刺激物質(Staurosporin: STS)、デスレセプター仲介性刺激物質 (Tumo r Necrosis Factor— a :TNF α )、または Fasリガンドによって誘導される、非虚 血刺激によるアポトーシス性細胞容積減少(Apoptotic Volume Decrease :AV D)には、 Cl_チャネルおよび K+チャネルの働きによる KC1流出が関与することが明 らかになつている(非特許文献 8を参照のこと)。本発明者らは、培養細胞において本 発明に用いたカルボン酸もしくはスチルベン誘導体の容積感受性の C1—チャネルの 機能をブロックする薬剤を培養上清に直接投与することによって上記したアポトーシ ス誘導剤によるアポトーシスを防ぐ方法を発明し、脳虚血性疾患、心虚血性疾患、神 経変性疾患およびうつ血性心不全に起因する細胞死に対する可能性を言及してい る(例えば、特許文献 2を参照のこと)。しかし、特許文献 2記載の発明によって個体 における虚血性刺激による遅発性神経細胞死が防御可能であるかどうかは誰にも実 証することができず、単なる予測に過ぎない。また、ラット心臓の異所性移植時に生じ る神経細胞アポトーシス性細胞死においても、 C1—チャネルブロッカー(心摘出前お よび移植直後に投与)が抑制的に作用することが報告されている (非特許文献 9を参 照のこと)。しかし、容積感受性 C1—チャネルの抑制を目的とした方法を用いて個体に おける脳虚血 '再灌流性の遅発性神経細胞死を防御または救済する方法の開発は 、未だ全く行われていない。
容積感受性 C1—チャネルの機能にタンパク質チロシンキナーゼ(protein tyrosin e kinase : PTK)が関与することはよく知られている(例えば、非特許文献 10を参照 のこと)。また、 ΡΤΚを阻害することによって脳循環を改善し、その結果として神経細 胞死が抑えられることは知られており(非特許文献 11を参照のこと)、脳虚血時に ΡΤ Κが活性ィ匕されることも知られている(非特許文献 12を参照のこと)。しかし、海馬の 神経細胞が容積感受性 C1—チャネルを有すること、これら海馬の容積感受性 C1—チヤ ネルに対し ΡΤΚが抑制的に働くことは、全く知られていない。すなわち、 ΡΤΚが海馬 の容積感受性 C1—チャネルを制御することに基づく神経細胞死に対する防御作用は 、全く知られていない。すなわち、容積感受性 C1—チャネルの機能制御に着目した上 での、 PTK阻害剤を標的とした、虚血'再灌流性の遅発性神経細胞死を防御または 救済する方法の開発は、未だ行われていない。

上述したように、遅発性神経細胞死は、虚血 ·再灌流の数日後に生じる細胞死であ る。このような遅発性神経細胞死の防御または救済に効果的な薬剤は、全く巿場ィ匕 されて!/、な!/、。遅発性神経細胞死が関与するアポトーシス様神経細胞死に対する力 スパーゼ経路の阻害に基づくアポトーシスの阻害を目的とした薬剤の開発は行われ ている可能性があるが、細胞死の経路が複雑かつ多様であるために、未だ臨床応用 には至って!/、な 、。一方、悪性腫瘍 (すなわち癌)の転移阻害を目的とした PTK阻害 剤の開発は行われているが、脳虚血性神経細胞死に対する薬物の開発は全く行わ れていない。

発明を実施するための最良の形態
遅発性の神経細胞死を防御または救済するために効果的な薬剤は、開発されてい ない。本発明者らは、上記課題を解決すべく鋭意検討を行なった。具体的には、本 発明者らは、マウス海馬初代培養神経細胞を用いて、
(1)マウス海馬初代培養神経細胞に容積感受性 C1—チャネルが存在するか;
(2)もし存在するならばマウス海馬初代培養神経細胞の容積感受性 C1—チャネルは 上述の C1—チャネルブロッカーや PTK阻害剤により抑制されるか
について検討を行なった。さらに、一過性のマウス脳虚血モデルを用いて、
(3)マウス脳虚血 ·再灌流性遅発性神経細胞死が C1—チャネルブロッカーを投与する ことによって抑制することができるかどうか;
(4)マウス脳虚血 ·再灌流性遅発性神経細胞死 PTK阻害剤を投与することによって も抑制することができるかどうか、
(5)マウス脳虚血 ·再灌流性遅発性神経細胞死の誘導の際に生じるアポトーシスを 抑制できるかどうか、
(6)脳虚血 ·再灌流中の脳循環血流量を改善できるかどうか
について検討を行なった。
[0035] その結果、本発明者らは、マウスの海馬領域の神経細胞に容積感受性 C1—チヤネ ルが存在することを初めて明らかにした。さらに本発明者らは、 C厂チャネルブロッ力 一を投与するか、または PTK阻害剤を用いることによって、当該容積感受性 C1—チヤ ネルを阻害することができることを明らかにした。
[0036] さらに本発明者らは、これらの知見に基づいて、マウスの両総頸動脈を一過性に遮 断した後にその再開通を行なうと 、う脳虚血 ·再灌流モデルを用い、遅発性神経細 胞死を、生存細胞および Zまたは変性神経細胞の形態組織学的観察、細胞質にお けるチトクローム cの増力!]、あるいは局所脳血流量の測定によって評価した。
[0037] その結果、本発明者らは、脳梗塞もしくは心筋梗塞の際、または心移植術もしくは 脳血管吻合術の脳虚血時もしくは脳虚血後の再灌流時に生じる遅発性神経細胞死 に対する治療薬剤および Zまたは防御薬剤を見出すとともに、当該薬剤の効果を判 定する方法を確立した。
[0038] 本発明を用いれば、脳虚血 ·再灌流によってもたらされる遅発性神経細胞死を、静 脈内に C1—チャネルブロッカーまたは PTK阻害剤を繰り返し投与したりすることによつ て抑制することができる。この救済メカニズムは、アポトーシス性細胞容積減少 (AVD )をもたらす容積感受性 Cl_チャネルの活性ィ匕を抑制することによって達成されるもの と考えられる。このように、本発明は、遅発性神経細胞死の初期反応である容積感受 性 C1—チャネル活性ィ匕およびその活性ィ匕シグナルを標的とした、虚血 ·再灌流性の 神経細胞傷害に対する新規防御方法を提供する。

本発明は、神経細胞死を抑制するための薬剤、および当該薬剤を用いて神経細胞 死を抑制する方法を提供する。神経細胞死は脳内の各所で起こり、種々の疾患の原 因となり、特に、遅発性神経細胞死は、その発生が遅いことから救済可能な細胞死で あると考えられているため、このような細胞死を抑制する手段を開発することができれ ば、当該細胞死が原因で生じる種々の疾患に対する有効な治療薬剤または予防薬 剤を得ることができ、非常に有益であるといえる。

本発明は、虚血後の再灌流条件下における神経細胞死の評価方法を提供する。 本発明に係る虚血後の再灌流条件下における神経細胞死の評価方法は、 C1—チヤ ネルブロッカーまたはタンパク質チロシンキナーゼ阻害剤を用いた場合の細胞死を 測定する工程;および上記測定によって得た測定値を、 C1—チャネルブロッカーまた はタンパク質チロシンキナーゼ阻害剤を用いな 、場合の細胞死を測定して得た基準 値と比較する工程を包含することを特徴としている。一実施形態において、本発明に 係る神経細胞死の評価方法は、上記神経細胞死が遅発性神経細胞死であることが 好ましい。好ましくは、本実施形態に係る神経細胞死の評価方法は、脳梗塞もしくは 心筋梗塞の際、または心移植術もしくは脳血管吻合術の脳虚血時もしくは脳虚血後 の再灌流時における神経細胞の生存率を評価する。

[0097] 海馬神経細胞を、妊娠 16日目のマウス力も採取した胎仔より単離した。得られた初 代培養海馬神経細胞を、 10%ゥシ胎仔血清、ストレプトマイシン、ペニシリンを添加し たダルベッコ必須培地にて、 5%COインキュベータ内にて 37°Cで培養した。培養 5

〜7日目の海馬神経細胞にホールセルパッチクランプ法を適応して、電流測定を行 なった。ノツチクランプ実験には、次のような組成の溶液を使用した。細胞外液には、 124mM N—メチル D グルカミン(NMDG)、 124mM HC1、 lOmM NaCl、 4mM MgCl、 0. 33mM NaH PO、 0. 5mM EGTA、 lOmM 2— [4—(2—

HEPESを用い、浸透圧を 300mOsmに調節した。 NMDGを用いて、 pH (細胞外 液および内液ともに)を 7. 4に調整した。ホールセルパッチクランプモード形成後、チ ャネル活性ィ匕の時間経過を観察するために、保持電位一 40mV力もテスト電位 ± 60 mVを与えて電流応答を記録した。また正常浸透圧時、低浸透圧刺激後、薬剤投与 後の電流の性質を検討するために、 100mV〜 + lOOmVまでのステップパルスを 随時与えて電流応答を記録した。
[0098] 虚血 ·再灌流による遅発性神経細胞死における C1—チャネルブロッカーまたは PTK 阻害剤の効果を検討するために、マウスの両総頸動脈を脳血管クリップにて閉塞し、 クリップを除去することによって再灌流する一過性脳虚血'再灌流モデルを用いた。
[0099] 図 1にそのプロトコールを示す。具体的には、 C57ZBL6Crj (9〜: L 1週齢、雄)に、 虚血 20分前より頸静脈力も DIDS (0. 48、 12、 60mg/kg) , genistein (0. 24、 6. 0、 30mgZkg)もしくは daidzein(30mgZkg)を 15分間 0. 8 LZ分 Zg体重(24 0〜300 μ LZ匹)の流速で投与した。対照群 (ビヒクル)は化合物を含まな 、溶液(1 0% DMSOおよび 10% polysorvate 80を含む PBS)を与えた。その後、 12分 間の両総頸動脈の閉塞による脳虚血 ·再灌流を行った。体温の低下を防ぐために 1 晚 35°Cの恒温槽内で飼育した後、室温(24 ± 1°C)で 4日間飼育した。予備的な試 験において、虚血前の単回投与だけでは細胞死防御効果を観察し得なかった。また 、溶媒中に DMSOおよび polysorvate 80を添加することにより、試験化合物の溶 液中の溶解性の向上が得られた。そのために、虚血前だけでなく虚血後 1、 2、 3日 目に静脈内投与と同量の化合物を腹腔内に投与した。 4日目に屠殺し組織学的検 討を行った。
[0100] これらの脳虚血モデルでは、虚血 '再灌流 1日目では 10%ほどの神経細胞が死亡 しているが、 4日目では 50%近くの神経細胞死が生じ、いわゆる遅発性神経細胞死 が観察された。
[0101] さらに、この遅発性神経細胞死がアポトーシスを含むかどうかを明らかにするために 、そして、この機構に対する C1—チャネルブロッカーまたは PTK阻害剤の効果の有無 を検討するために、同様の実験モデル系において、海馬領域における細胞質画分 のチトクローム cの増加を、生化学的手法 (ウェスタンブロッテイング)によって観察し た。また、脳循環血流量に対する PTK阻害剤の効果を、レーザードップラー局所血 流計により測定した。局所脳血流量は、 1. Ommのレーザ一径により中大脳動脈領 域の大脳皮質を左右 6〜9ケ所計測し、その平均にて示した。化合物投与前を 100 %として算出した。

Physiol. 501: 555— 564.を参照のこと)、ラ ット頸動脈小体タイプ 1神経分泌細胞(Carpenter E, Peers C. (1997) "Swelling- and camp-activatedし 1 currents in isolated rat carotid body type I cells. J. Physiol. 50 3: 497-511 .を参照のこと)、およびマウス小脳穎粒細胞(Patel AJ, Lauritzen I, Lazd unski M, Honore E. (1998) "Disruption of mitochondrial respiration inhibits volume- regulated anion channels and provokes neuronal cell swelling, j. Neurosci. 18: 3117 -3123.を参照のこと)のみである。そこで、海馬神経細胞に容積感受性 C1—チャネル が存在するかどうかを調べた(図 2)。
[0103] 容積感受性 C1—チャネルは分子実体が明らかにされて 、な 、ので、免疫組織学的 な手法が適応できない。そこで、その発現をパッチクランプ法による電流記録によつ て確認した。 [0104] 図 2Aに示すように、ホールセルパッチクランプ下でマウス海馬初代培養神経細胞 に低浸透圧負荷をかけると、徐々に活性化される電流が観察された。この活性化さ れた電流の逆転電位(=— 36mV)は Cl_の平衡電位(=—40mV)に近似し、外向 き整流性(図 2C)で高 、プラス電位 ( > + 80mV)にお 、て不活性化を示す(図 2B) 。これらの結果は全て容積感受性 C1—電流の特性に一致するこ。よって、マウス海馬 初代培養神経細胞に容積感受性 C1—チャネルが存在することが明らかになった。

マウス海馬初代培養神経細胞の容積感受性 C1—チャネル力 その阻害剤として知 られる DIDSや genisteinによって抑制されるかどうかを確認した(図 2)。 500 M DIDSの抑制効果はプラス電位にのみ見られ、電位依存性であった(図 2D)。また 1 OO ^ M genisteinによる抑制は電位に非依存的で、逆転電位付近を除く全ての電 位においてその抑制効果を発揮した(図 2E)。このこと力 、 DIDSと genisteinが確 力に海馬神経細胞の容積感受性 C1—チャネルを抑制することを確認した。

ラットの培養神経細胞のスタウロスポリン(STS)によって誘導されたアポトーシスは 、 C1—チャネルのブロッカーである DIDSによって救済されることが報告されている(Ta nabe S, Maeno E, Okada Y (2002) Prevention of staurosporine- induced apoptotic c ell death by DIDS and SITS in rat cardiomyocytes in primary culture." Jpn. J. Physio 1. 52 (Suppl):S34を参照のこと)。そこで、マウスの脳虚血性神経細胞死が Cl_チヤネ ルブロッカーで抑制されるかどうかを検討した。
[0107] 図 3に示すように、 12分間の虚血によって神経細胞死を誘導した場合、トルイジン ブルー染色により形態学的に観察すると、海馬 CA1領域の神経細胞は核小体が不 明瞭になり錐体細胞の縮小化が認められた(図 3、 A矢印)。一定面積あたりの残存 する形態的に正常な細胞数は、脳虚血前の約 60個(結果非表示)から 37. 1個にま で減少した(図 2、第 1棒グラフ)。虚血 15分前および再灌流後に C1—チャネルブロッ カーである DIDS (0. 48、 12、 60mgZkg)を投与しておくと、 0. 48mgZkgまたは 60mgZkgでの投与では、その効果はあまり顕著ではな力つた(図 3、図 5)。しかし、 12mgZkgの投与群においてその形態変化は少なくなり(図 3、 D)、残存する形態 的に正常な細胞数は 52. 5個に上昇した(図 4、第 3棒グラフ)。
[0108] さらに、図 3に示すように、神経変性像 (Fluoro— JadeB染色により検出)を示す神 経細胞を観察したところ、ビヒクル投与のマウスの海馬 CA1領域において、明らかに 強く蛍光染色される変性した神経細胞が観察された (図 5、 A矢印)。単位面積当たり の変性神経細胞数を計測すると 28. 4個であった(図 6、第 1棒グラフ)。 DIDS12mg Zkgを投与すると、染色される細胞が減少し(図 5、 C)。変性神経細胞数は 12. 0個 に明らかに低下した(図 6、第 3棒グラフ)。これらの結果は、 C厂チャネルブロッカー である DIDSが一過性のマウス脳虚血後の遅発性神経細胞死に対して救済的に作 用することを示している。

図 3に示すように、虚血の 15分前にビヒクルを投与し 12分間の虚血性神経細胞死 を誘導した場合、一定面積あたりの残存する形態的に正常な細胞数は 37. 1であつ た(図 4、第 1棒グラフ)。虚血 15分前および再灌流後 1、 2、 3日後の計 4回、 PTK阻 害剤である genistein (l . 2、 6、 30mgZkg)を投与しておくと、海馬 CA1領域の神 経細胞の形態変化が抑制され (図 3、 E、 F、 G)、単位面積当たりの残存する形態的 に正常な細胞数は、それぞれ 48. 2、 50. 8、 55. 7に上昇した(図 4、第 5、 6、 7棒グ ラフ)。変性神経細胞を観察したところ、ビヒクル投与群の海馬 CA1領域において、 明らかに変性した神経細胞が観察された(図 5、 A)。 Genisteinを投与すると、変性 神経細胞の出現は低下した(図 5、 D、 E、 F)。単位面積あたりの変性神経細胞数は 、濃度依存的に 14. 0、 11. 7、 5. 8と明らかに減少した(図 6、第 5、 6、 7棒グラフ)。 さらに、 genisteinの PTK活性を有さない類似体である daidzein (30mgZkg)の投 与群では、形態的正常細胞の低下、変性神経細胞の増加を抑制できな力つた(図 3 、 H ;図 4、第 8棒グラフ;図 5、 H ;図 6、第 8棒グラフ)。これらの結果は、 PTK阻害剤 である genisteinが一過性のマウス脳虚血後の遅発性神経細胞死に対して救済的に 作用することを示している。

アポトーシス性細胞死の過程において、チトクローム cのミトコンドリアから細胞質へ の放出は、細胞死の出現に最も重要な現象であるということが知られている(非特許 文献 7を参照のこと)。また、 DIDSはラットの培養神経細胞のスタウロスポリン(STS) によって誘導されたアポトーシスを救済することが報告されている(Tanabe S, Maeno E, Okada Y (2002) Prevention of staurosporme- induced apoptotic cell death by DI DS and SITS in rat cardiomyocytes in primary culture." Jpn. J. Physiol. 52 (Suppl):S 34を参照のこと)。
[0111] 一過性脳虚血誘導後の海馬細胞質において、チトクローム c (cyt— c)の増加がみ られた(図 7、ビヒクル投与群(1〜4レーン))。一過性脳虚血を誘導する 15分前に C1 —チャネルブロッカー(図 7、第 5〜第 8レーン(DIDS (12mg/kg) ) )または PTK阻 害剤(図 7、第 9〜第 12レーン (genistein (30mg/kg) ) )を投与したマウスの脳海 馬領域において、チトクローム cの増加が抑制された。これらの結果は、 C1—チャネル ブロッカーである DIDSまたは PTK阻害剤である genisteinが一過性のマウス脳虚血 後のアポトーシスに対して救済的に作用することを示している。

PTKの阻害剤の 1つである PP1は、脳虚血中の血管拡張作用を示すことが報告さ れている(非特許文献 11を参照のこと)。局所脳血流を、レーザードップラー血流計 を用いて測定した。図 8に示すように、虚血中の大脳皮質の局所血流量は脳虚血前 (100%)と比較して 25%にまで低下する力 10分間の再灌流で徐々に回復し、虚 血前付近に戻る(図中、 Genistein投与群(黒丸)、ビヒクル群(白丸)およびコント口 ール(ビヒクル無投与群))。しかしながら、脳虚血前 15分間の genistein (30mgZk g)投与を行った場合は、虚血中の脳血流の 30〜35%と高ぐさらに、再灌流後の局 所脳血流量は約 2分間で回復する。特に、再灌流直後 30秒間で 50%回復し虚血前 の 80%にまで回復する。しかし、ビヒクル群では 20%程度 (虚血前の 40%に回復)し か回復しない。この結果は、 PTK阻害剤である genisteinが一過性のマウス脳虚血' 再灌流中に血管拡張などの循環改善を引き起こし、その結果、脳血流の低下を抑制 し、その後生じるであろう遅発性神経細胞死に対して救済的に作用することを示して いる。
[0113] このように、本発明は、海馬初代培養神経細胞における容積感受性 C1—チャネルの 存在を明らかにし、該容積感受性 C1—チャネル力 厂チャネルブロッカーまたは cr チャネルを制御する PTKの阻害剤により抑制されることを見出したことに基づいて完 成されたものである。さらに、神経初代培養細胞で見出した知見に基づき、本発明者 独自の「個体における脳虚血 '再灌流の評価系」を駆使し、可溶化剤を含む溶媒に 薬物を溶解することにより循環中での動態を改善し、かつ脳虚血前だけでなく数日間 にわたり薬物を繰り返し投与する新しい工夫を重ねて確立した方法がなければなし 得なかったものである。
[0114] 尚、発明を実施するための最良の形態の項においてなした具体的な実施態様また は実施例は、あくまでも、本発明の技術内容を明らかにするものであって、そのような 具体例にのみ限定して狭義に解釈されるべきものではなぐ本発明の精神と次に記 載する特許請求の範囲内で、いろいろと変更して実施することができるものである。 産業上の利用の可能性
[0115] 本発明を用いれば、個体における神経細胞死、特に、脳虚血 ·再灌流によってもた らされる遅発性神経細胞死を抑制することができる。本発明における投与経路が静 脈内投与であることによって、臨床的治療への適用が比較的容易となる。さらに,本 発明において用いられた繰り返し投与のよる方法の工夫は本発明をより効率的に応 用するための有用な技術の開発に役立つ。また、脳梗塞もしくは心筋梗塞の際、また は心移植術もしくは脳血管吻合術の脳虚血時もしくは脳虚血後の再灌流時に本発 明を適用することによって、生体の生存にとって最も重要な臓器である脳、特に神経 細胞の障害の軽減が可能となる。
[0116] 神経細胞の虚血 ·再灌流性による遅発性神経細胞死を防御または救済するために 効果的な薬剤または方法というものが、これまで開発されていな力 たので、本発明 によって、遅発性神経細胞死を防御および Zまたは救済することが可能となり、神経 細胞の遅発性細胞死に起因する様々な脳障害に対する有効な治療および Zまたは 予防に関する技術の開発に大いに役立つ。

Claims

請求の範囲
[I] Cl_チャネルブロッカーまたはタンパク質チロシンキナーゼ(PTK)阻害剤を含むこ とを特徴とする脳虚血後の神経細胞死を防御または救済するための薬剤。
[2] 上記神経細胞死が個体における遅発性神経細胞死であることを特徴とする請求の 範囲 1に記載の薬剤。
[3] 上記神経細胞死が核内 DNAの断片化を伴うことを特徴とする請求の範囲 1または 2に記載の薬剤。
[4] 上記神経細胞死は、チトクローム cのミトコンドリア力もの放出の顕著な低下を伴うこ とを特徴とする請求の範囲 1〜3の何れか 1項に記載の薬剤。
[5] 上記 C1—チヤネノレブロッカー力 4, 4,― diisothiocyanostilbene― 2, 2,— disul fonic acid (DIDS)であることを特徴とする請求の範囲 1〜4の何れ力 1項に記載の 薬剤。
[6] 上記 PTK阻害剤が、 genisteinであることを特徴とする請求の範囲 1〜4の何れ力 1 項に記載の薬剤。
[7] 上記神経細胞死が、脳梗塞もしくは心筋梗塞の際、または心移植術もしくは脳血管 吻合術の脳虚血時もしくは脳虚血後の再灌流時に生じることを特徴とする請求の範 囲 1〜6の何れか 1項に記載の薬剤。
[8] C1—チャネルブロッカーまたは PTK阻害剤を虚血後の再灌流条件下における神経 細胞に投与する工程を包含することを特徴とする神経細胞死を防御または救済する ための方法。
[9] 上記神経細胞死が遅発性神経細胞死であることを特徴とする請求の範囲 8に記載 の方法。
[10] 上記神経細胞死が核内 DNAの断片化を伴うことを特徴とする請求の範囲 8または 9に記載の方法。
[II] 上記神経細胞死力 細胞死を引き起こすチトクローム cのミトコンドリアからの放出の 活性の顕著な低下を伴うことを特徴とする請求の範囲 8〜 10のいずれか 1項に記載 の方法。
[12] 脳虚血時および虚血後の再灌流時に生じる脳血流量低下の改善を伴うことを特徴 とする請求の範囲 8〜: L Iのいずれか 1項に記載の方法。
[13] 上記 C1—チヤネノレブロッカーが、 4, 4' - diisothiocyanostilbene - 2, 2, 一 disul fonic acid (DIDS)であることを特徴とする請求の範囲 8〜12の何れ力 1項に記載 の方法。
[14] 上記 PTK阻害剤が、 genisteinであることを特徴とする請求の範囲 8〜12の何れか 1項に記載の方法。
[15] 上記神経細胞死が、脳梗塞もしくは心筋梗塞の際、または心移植術もしくは脳血管 吻合術の脳虚血時もしくは脳虚血後の再灌流時に生じることを特徴とする請求の範 囲 8〜14の何れ力 1項に記載の方法。
[16] 個体を用いた虚血後の再灌流条件下における神経細胞死の評価方法であって、
C1—チャネルブロッカーまたはタンパク質チロシンキナーゼ阻害剤を用いた場合 の細胞死を測定する工程;および
上記測定によって得た測定値を、 C1—チャネルブロッカーまたはタンパク質チロシ ンキナーゼ阻害剤を用いな 、場合の細胞死を測定して得た基準値と比較する工程 を包含することを特徴とする方法。
[17] 上記神経細胞死が脳虚血性の遅発性神経細胞死であることを特徴とする請求の範 囲 16に記載の神経細胞死の評価方法。
[18] 脳梗塞もしくは心筋梗塞の際、または心移植術もしくは脳血管吻合術の際に生じる 脳虚血時もしくは脳虚血後の再灌流時における神経細胞の生存率を評価することを 特徴とする請求の範囲 16または 17に記載の神経細胞死の評価方法。
[19] 上記細胞死を測定する工程が、ミトコンドリア脱水素酵素の活性を指標に行なうこと を特徴とする請求の範囲 16〜18のいずれか 1項に記載の神経細胞死の評価方法。
[20] 上記細胞死を測定する工程が、核内 DNAの断片化を指標に行なうことを特徴とす る請求の範囲 16〜18のいずれか 1項に記載の神経細胞死の評価方法。
[21] 脳血流量の変化を測定する工程をさらに包含することを特徴とする請求の範囲 16
〜20のいずれか 1項に記載の神経細胞死の評価方法。
[22] マウスの両総頸動脈を一過性に遮断した後にその再開通を行なう工程をさらに包 含することを特徴とする請求の範囲 16〜21のいずれ力 1項に記載の神経細胞死の 評価方法。
[23] 神経細胞の遅発性細胞死を防御または救済する因子をインビトロでスクリーニング する方法であって、
培養するための細胞を調製する工程;
グルコースを含まない培地中にて当該細胞を低酸素条件下または無酸素条件下 で培養する低酸素処理工程;
グルコースを含む培地中にて有酸素条件下で当該虚血処理に供した細胞を培養 する再酸素処理工程;および
細胞死を測定する工程
を包含し、
当該低酸素処理工程時もしくは当該再酸素処理工程時またはこれらの前後に候 補因子を培地中に添加して測定した細胞死を、当該候補因子を添加することなく測 定した細胞死と比較する工程
をさらに包含することを特徴とするスクリ一二ング方法。
[24] 上記細胞死を測定する工程力 ミトコンドリア脱水素酵素の活性、核内 DNAの断 片化、カスパーゼ活性、ミトコンドリア力も細胞質へのチトクローム cの放出、または核 の形態変化を指標とすることを特徴とする請求の範囲 23に記載のスクリーニング方 法。
