Abstract

The present invention relates to a probiotic composition which enables to maintain effectively physiological activity of functional microorganism contained therein for a long time. More par¬ ticularly, the present invention is directed to a probiotic composition comprising a colorant which maintains effectively physiological activity of a functional microorganism for a long time and increases the convenience for use by being mixed or melted easily in both liquid type and pulverulent type medium such as water, food or fodder, etc.

Description

Description PROBIOTIC COMPOSITION Technical Field
[1] The present invention relates to a probiotic composition which enables to maintain effectively physiological activity of functional microorganism contained therein for a long time. More particularly, the present invention is directed to a probiotic composition comprising a colorant which maintains effectively physiological activity of a functional microorganism for a long time and increases the convenience for use by being mixed or melted easily in both liquid type and pulverulent type medium, such as water, food or fodder, etc. Background Art
[2] The various microorganisms which have physiological activities, for example, suppression of harmful bacteria and promotion of intestinal movement by improving the balance of microorganisms in the gastrointestinal tract, have been employed as an active principle of the so-called probiotics.
[3] The "probiotics" in this specification means a live microbial food ingredient which has a useful effect on human and animal by improving balance of microorganisms in the gastrointestinal tract.
[4] The functional microorganisms employed as an active principle of probiotics, are ready to die out under the variation of environmental factors when the microorganisms are exposed to external environment. Consequently, it is necessary to minimize the influence of environmental factors in the custody process of the live microbial food ingredient.
[5] Therefore, various methods which enable to maintain effectively the physiological activity of the microorganisms by minimizing the chance of influence of environmental factors in the custody process of the live microbial food ingredient, have been studied for long time.
[6] The methods for suppressing the death of microorganisms in the custody processes have been known up to now, may be classified into several methods for dehydrating by freeze-drying, by spray-drying (Korea patent No. 0137154, 0148340 and 2002-0097237), for capsuling or coating(Korea patent No. 2002-0069863 and 2003-0009268).
[7] Those conventional methods have the advantages of maintaining the live microbial food ingredient for a long time and of making it easy to mix the live microbial food ingredient with food.
[8] However, when the probiotics are being dissolved in water, such as the case like ad- ministering the live microbial food ingredient to fishes or feathers, it is difficult for probiotics to be dissolved in water, and even though it can be dissolved in water, the microorganisms are ready to die out by the influence of environmental factors.
[9] In other words, it is difficult to administer the capsuled or coated probiotics to fishes or feather due to the low water solubility. Also, the dried probiotics are affected largely by environmental factors, for example, the ultraviolet ray, the dissolved oxygen and the concentration of chlorine ion within water, etc., after being dissolved in water.
[10] Therefore, in this technical field, it has been eagerly requested to develop a novel probiotic composition which can be mixed or dissolved easily to both liquid type and pulverulent type medium such as water, food or fodder, etc., and can be maintained in the room temperature for a long time, and can endure under the effect of environmental factors. Disclosure of Invention Technical Problem
[11] Therefore, the primary object of the present invention is to provide a probiotic composition comprising an edible colorant, which maintains effectively physiological activity of a functional microorganism contained therein for a long time and increases convenience for use by being mixed or melted easily in both liquid type and pulverulent type medium, such as water, food or fodder, etc.
[12] Another object of the present invention is to provide a probiotic composition comprising a skim milk, which maintains effectively physiological activity of a functional microorganism contained therein for a long time and increases convenience for use by being mixed or melted homogeneously in both liquid type and pulverulent type medium, such as water, food or fodder, etc. Technical Solution
[13] The primary object of the present invention is achieved by providing a probiotic composition comprising a colorant, which maintains effectively physiological activity of a functional microorganism contained therein for a long time and increases convenience for use by being mixed or melted easily in both liquid type and pulverulent type medium, such as water, food or fodder, etc.
[14] The colorant of the present invention is a material which shows a specific color by reflecting a light of specific wave length and does not have harmful effect to the physiological function of human or animal such as cattle.
[15] The colorant of the present invention includes colorants recorded in the food additive code and Korean pharmacopoeia of KFDA (the Korea Food and Drug Administration), and the rest of colorants known to the public as they are edible even though they does not recorded in the food additive code and Korean pharmacopoeia. [16] Various kinds of the colorants are added as an ultraviolet protector for minimizing death of the microorganisms when the probiotic composition was dissolved in water, food or fodder.
[17] The colorants which are added to the probiotic composition also function as a mixing or dissolving supplementary simultaneously which make it easy for probiotic composition to be mixed or be dissolved in both liquid type and pulverulent type medium such as water, food or fodder, etc.
[18] The colorants were added before the live microbial food ingredient was freeze- dried. The mixing process of the colorant does not have a negative effect to the activity of the live microbial food ingredient during freeze-drying process or custody process.
[19] The protecting effect of the colorants employed in the present invention is demonstrated in Table 1. The experiment was carried out with an edible colorant which is not harmful to human body, and showed protecting effect in order of yellow edible colorant, green edible colorant and red colorant.

[21] Another object of the present invention above-mentioned is achieved by providing a probiotic composition comprising a skim milk, which maintains effectively physiological activity of a functional microorganism contained therein for long time and increases convenience for use by being mixed or melted homogeneously in both liquid type and pulverulent type medium, such as water, food or fodder, etc.
[22] The skim milk of the present invention means liquid type or powder type of milk which is dried after being defatted.
[23] Various waters such as civil water, subterranean water, surface water and sea water etc. can be employed as the water for dissolving the probiotic composition, and these waters differ from refined water in several factors. Namely, various conditions such as chlorine ion concentration of water, dissolved oxygen of water etc. may affect the activity of live microbial food ingredient.
[24] A stabilizer generally employed for minimizing the effect of the above factors is Sodium Sulfite, L-Cysteine etc.
[25] However, the stabilizer is not effective to suppress death of live microbial food ingredient when the probiotic composition is dissolved in the civil water contained a lot of chlorine ions.
[26] The skim milk is more effective than Sodium Sulfite or L-Cysteine to minimize the effect of chlorine ion in the civil water for the live microbial food ingredient as shown in Table 2.
[27] A skim milk is added before a live microbial food ingredient is freeze-dried. The probiotic composition which includes the skim milk, can maintain the activity of microorganisms even though the probiotic composition is dissolved in the water which contains chlorine ion.
[28] The skim milk which is added to a probiotic composition also functions as a mixing or dissolving supplement which promotes mixing or dissolving of the probiotic composition in both liquid type and pulverulent type medium, such as water, food or fodder, etc.

[30] Practical and preferred embodiments of the present invention will be illustrated in more detail in the following Examples. The present invention is not limited to them and can allow various modifications and alterations performed by those skilled in the art. Advantageous Effects
[31] A dried probiotic composition of the present invention which comprises edible colorant or skim milk, can be mixed and dissolved easily in both liquid type and pulverulent type medium, such as water, food or fodder, etc. and can minimize the effect of environmental factors such as ultraviolet ray or diluted water to the physiological activity of microorganism in the live microbial food ingredient.
[32] Therefore, the live microbial food ingredient of the present invention can be employed conveniently with little attention when an operator combines the probiotic composition with a fodder for cattle in the field. Also, the probiotic composition of the present invention can be employed usefully in the market in which the demands for the liquid type of the live microbial food ingredient increases since the effect of the probiotic composition of the present invention is identical with that of prior live microbial food ingredient. Best Mode for Carrying Out the Invention
[33] Example 1. The change of the activity of the lactic ferments and the protection effect of the skim milk on the activity of the live bacteria according to the kinds of diluted water
[34] The activity change of the lactic ferment, which was dissolved in 0.9% isotonic solution (produced by dissolving 0.9% sodium chloride in distilled water) or pH7.5 PBS buffer, was compared with the activity change of the lactic ferment which was dissolved in service water, sea water, or subterranean etc. (Table 3). When over 0.05 wt % of skim milk was added to civil water, the result shows that the addition of the skim milk is effective to the maintenance of the activity of lactic ferment (Table 4).

The ultraviolet protection effect by the edible colorant [38] Cultured lactic ferment was diluted as three thousand fold with 50mM sodium phosphate buffer (pH7.5) which was added 0.9 wt % of sodium chloride. After 0.001 wt %, 0.005 wt %, 0.01 wt % and 0.04 wt % of the green edible colorant was added respectively, the number of live microorganisms was measured at five-minute intervals under the strong ultraviolet. The result shows that the survival rate of the lactic ferment increases when over 0.005 wt % of edible colorant was added as shown Table 5.

[40] The experiment shows that the survival rate of lactic ferments does not affected by edible colorant, when ultraviolet was not irradiated. (Table 6).

[44] The chickens were shipped after 30 days since the chickens stared to eat standing. When the average weight and survival rate of the chickens were compared with those of the chickens to which prior probiotics composition was administrated every day for 30 days, there is no difference between them as seen in Table 7. This result shows that the lactic ferment probiotic composition for animals which contains the edible colorant and skim milk is safe and effective.

The result is demonstrated in Table 7.
[48] The survival rate between the experiment group and the control group shows the difference of approximately 3% and fodder demand rate of experiment group was higher than that of control group approximately 8%. This result shows the lactic ferment probiotic composition for animals which contains the edible colorant and skim milk of the present invention is safe and effective.

[50] Practical and preferred embodiments of the present invention was illustrated such as above Examples, however, the present invention is not limited to them and, those skilled in the art will also appreciate that such equivalent embodiments do not depart from the spirit and scope of the invention as set forth in the appended claims.

Claims
[1] A probiotic composition comprising an edible colorant.
[2] The probiotic composition according to Claim 1, wherein content of said colorant is 0.005 wt % to 5 wt %.
[3] A probiotic composition comprising a skim milk.
[4] The probiotic composition according to Claim 3, wherein content of said skim milk is 0.05 wt % to 50 wt %. [5] A probiotic composition comprising an edible colorant and a skim milk.
