Abstract

L'invention concerne une méthode permettant d'isoler des peptides antibactériens et les peptides ainsi isolés. Ladite méthode consiste: à construire une séquence de fusion d'ADN d'une séquence de tête de facteur de conjugaison de levure alpha et d'un peptide aléatoire; à introduire ladite séquence dans un vecteur de Saccharomyces cerevisiae qui contient un promoteur inductible; à digérer l'ADN linéaire non lié à l'aide d'une exonucléase III et d'une nucléase de pousse de haricot mango; à électro-transformer le vecteur de levure après purification; à cultiver le transformant pendant 24H; à détecter un noyau d'inhibition bactérienne et la vitesse d'inhibition bactérienne après induction de l'expression par un milieu liquide; à sélectionner les clones positifs et à les amplifier par PCR; et à déterminer la séquence. La méthode de l'invention peut être facilement et efficacement utilisée pour isoler des oligopeptides et des polypeptides antibactériens et pour trouver de des antibiotiques manière alternative.
