Abstract

La présente invention se rapporte à des réactifs, à des procédés et à des appareils permettant de prétraiter des échantillons sanguins afin d'obtenir des fractions de sang contenant sensiblement toutes les cellules cibles dans des volumes spécimens réduits qui sont adaptés à un enrichissement ultérieur. L'invention a trait des systèmes de traitement d'échantillons, destinés à enrichir des populations cellulaires dont la densité est inférieure à approximativement 1,09 g/ml, à partir d'environ 30 ml de sang total. L'on combine le sang total avec un réactif dense à compatibilité cellulaire, afin d'élever la densité à environ 1,09 g/ml. La centrifugation provoque l'accumulation des cellules cibles et de la plupart des globules blancs contenus dans la couche leuco-plaquettaire dans la partie supérieure, sous une couche de recouvrement tampon à faible densité. Les volumes d'échantillons réduits sont adaptés à des procédés ultérieurs d'enrichissement immunomagnétique ou autres, avant la détection et le dénombrement. Lesdits systèmes de traitement facilitent, conjointement avec d'autres procédures d'enrichissement, la détection de cellules cibles rares, telles que les cellules tumorales circulantes (CTC), lesquelles constituent des marqueurs utiles pour le diagnostic du cancer et le contrôle de la réponse thérapeutique.
