本発明は、 D N Aの断片化方法、 D N Aシャフリング方法、 およびこ れら方法に使用するための試薬キッ トに関する。 背景技術
D N Aシャフリングは、 全長 D N Aをランダムに切断し、 適当な長さ に切断された D N Aが相互にプライマーあるいは铸型となり、 ォ一パー ラップ伸長を行うことで、相同 D N A間の組換えを誘起する方法であり、 変異遺伝子の作製方法として広く使用されている。 従来、 D N Aをラン ダムに切断し、 適当な長さの D N A断片を調製する方法として、 例えば デォキシリポヌクレアーゼ I など、 基質特異性の低いヌクレア一ゼを利 用する方法があった。 しかし、 デォキシリポヌクレアーゼ I を用いる方 法では、 以下に述べるような幾多の問題があった。
すなわち、 デォキシリポヌクレア一ゼ I による方法では、 反応に関わ るさまざまな因子 (デォキシ.リポヌクレア一ゼ I の酵素量、 基質 D N A の純度や濃度、 反応温度、 反応時間) を正確に制御し、 適当な反応時間 で反応を終結させ部分分解産物を取得する必要があった。 また、 D N A の部分分解産物と未分解産物とをァガロースゲル電気泳動などの手法に より分画しなければならなかった。 さらに、 デォキシリポヌクレアーゼ I の酵素量、 基質 D N Aの純度や濃度、 反応温度、 反応時間などのわず かな条件の違いにより断片の長さが変わり、 再現性に欠けるという欠点 も有していた。 このようにデォキシリポヌクレアーゼ I を用いた D N A の断片化は多大な労力を要する上、 再現性にも乏しいという欠点を抱え ていた。 このため、 煩雑な反応最適条件の検討を必要とせず、 またァガ
ロースゲルによる分離 · 抽出 ·精製等の煩雑な操作を必要としない、 簡 便な操作で再現性の高い D N Aの断片化および分画手法の開発が望まれ ていた。 発明の開示
本発明の課題は、 上記従来技術の問題点を解消し、 DNAシャフリ ン グにおける D N A断片試料の調製において、 簡便で、 かつ再現性の高い 手法を開発することにある。
本発明者は、 鋭意研究の結果、 DNAの塩基配列中、 チミンをランダ ムにゥラシルに置換した D N Aを調製した後、 エンドヌクレアーゼ Vを 作用させることにより、 D N Aのランダムな切断を簡便かつ再現性よく 行い得るとともに、 さらに該エンドヌクレアーゼ Vによる DN A断片化 試料を用いて実際に D N Aシャフリングを行った結果、 極めて有効に D N Aシャフリングを行えることを確認し、 本発明を完成するに至った。 すなわち、 本発明は、 以下の ( 1 ) 〜 (6) に関する。
( 1 ) DN Aの塩基配列中、 チミンをランダムににゥラシルで置換した 後、 エンドヌクレアーゼ Vを作用させることにより DNAを断片化する ことを特徴とする、 DNA断片試料の調製方法。
(2 ) 上記 ( 1 ) により調製された DNA断片試料を用いて、 DNAシ ャフリングを行うことを特徴とする、 DNAシャフリング方法。
( 3 ) デォキシゥリジントリホスフエ一ト ( d UT P) およびエンドヌ クレアーゼ Vをそれぞれ試薬として含むことを特徴とする、 D NAシャ フリングにおける DN Aを断片化するために用いる試薬キッ ト
(4) さらに、 デォキシアデノシントリホスフェート (d AT P)、 デォ キシグアノシントリホスフェート (d GT P)、 デォキシシチジンントリ ホスフエ一ト (d C T P)およびデォキシチミジントリホスフェート (d TT P)、並びに DNAポリメラーゼを試薬として含むことを特徴とする、 請求項 3に記載の試薬キッ 卜。
(5) DNAシャフリングにおける、 D N A断片化に使用するものであ る、 請求項 3または 4に記載の試薬キッ ト。
( 6 ) デォキシゥリジントリホスフェート ( d UT P )、 デォキシアデノ シントリホスフェート (dATP)、 デォキシグアノシントリホスフエ一 ト (dGTP)、 デォキシシチジンントリホスフエ一ト (d CTP) およ びデォキシチミジントリホスフェート (d TT P)、 DNAポリメラ一ゼ 並びにエンドヌクレア一ゼ Vをそれぞれ試薬として含むことを特徴とす る、 DN Aシャフリング試薬キッ ト。
以下に、 本発明を詳細に説明する。
本発明においては、 遺伝子 DN A塩基配列中のチミンをランダムにゥ ラシルに置換した D N Aを調製した後、 エンドヌクレアーゼ Vを作用さ せるが、 このエンドヌクレア一ゼ Vはゥラシルを含む D N Aを基質とし、 ゥラシル位置の 2 -3塩基下流のホスホジエステル結合を切断する活性 を有する酵素である (Huang, J. , Lu, J. , Barany, F. , and Cao, W. 2001. C em. 272, 30774-30779) 0 このため、 D N A塩基のチミンをランダムに ゥラシルに置換した DNAを調製しェンドヌクレアーゼ Vを作用させる ことで、 D N Aのランダムな断片化が達成される。 このようなエンドヌ クレア一ゼ Vによる切断反応は、 取り込まれたゥラシルの量に依存して 進行し、 基質を消費した段階で反応が終結する。 このように、 完全消化 により断片化がおこなわれるため、 デォキシリポヌクレア一ゼ I を使用 した反応時におけるような部分消化ではなく、 反応の進行状況のモニタ 一の必要がない。 また、 再現性よく反応が進行する。
本発明による DNAシャフリングは、 ( 1) ランダムな DNAの切断に
よる DNA断片試料の調製工程と、 ( 2 ) 得られた DNA断片試料を用い た D N Aシャフリング工程からなる。
( 1 ) DNA断片試料の調製工程
本発明においては、 D N Aシャフリングの対象となる遺伝子 D N Aを 錶型とし、 上流プライマーおよび下流プライマ一、 並びにデォキシアデ ノシントリホスフェート (以下、 d AT Pという。)、 デォキシグアノシ ントリホスフェート (以下、 d GT Pという。)、 デォキシシチジンント リホスフェート (以下、 d C T Pという。) およびデォキシチミジントリ ホスフェート (以下、 d T T Pという。) と、 さらにデォキシゥリジント リホスフェート (以下、 d UT Pという。) を含む反応溶液に D N Aポリ メラ一ゼを加え P C R増幅を行う。 得られる P C R産物は、 上記遺伝子 D N Aの塩基配列中チミンがゥラシルにランダムに置き換えられた二本 鎖の D N A分子である。
この場合において、 得られる D NA分子中のゥラシルの置換量は、 使 用する d T T Pと d UT Pの量比に対応して変化し、 この量比を変化さ せることにより、 ゥラシルの置換量を制御できる。 すなわち、 ゥラシル の割合を多くすれば、 ゥラシルの置換量は増大する。
次いで、 ゥラシル置換 DNA分子を、 エンドヌクレア一ゼ Vにより切 断することにより、 該 D N A分子は断片化される。 得られる D N A断片 試料は、 分子長が異なる切断 DNA分子の集合体であり、 各切断 D NA 分子の分子長は、 ランダムに置換されたゥラシルの間に挟まれた領域に 対応する。 したがって、 ゥラシルの置換量が多ければ、 より短い分子長 の切断 DNA分子が多くなり、 反対に少なければ、 より長い分子長の切 断 DN A分子が多くなる。
一方、 上記したようにゥラシルの置換量は、 P C R増幅において、 使 用する d TT Pと d UT Pの量比を変化させることにより制御可能であ るから、 単に、 P C R増幅において、 d T T Pと d UT Pの量比を制御 するという極めて簡便な手段のみで、 所定の分子長分布の DN A断片を
再現性よく得ることが可能となる。 また、 この方法によれば、 従来のよ うな DNA切断後のァガロースによる分離、 抽出、 精製等の煩雑な操作 は、 実質上必ずしも必要としないので、 エンドヌクレアーゼ Vによる切 断後の DN A断片試料を、そのまま用いて DNA シャフリングを行うこ とができる。
上記においては、 遺伝子 DN Aのゥラシル置換の手法として P C Rを 使用する方法について説明したが、 本発明においては、 ゥラシル置換の 方法はこれにとどまらない。 例えば、 D NA複製の際、 チミンの代わり に誤って取り込まれたゥラシルを除去修復する機能の欠損した宿主菌内 で DNAを複製させることによつても得られる。 このような宿主菌とし ては、例えば、ェシエリシァ コリー C J 2 3 6あるいはェシエリシァ コ リ一 BW 3 1 3等が挙げられる。 この場合、 該宿主菌を定法に従って目 的遺伝子を含むプラスミ ドなどで形質転換し、 宿主菌内でプラスミ ドを 増幅し、 定法に従ってプラスミ ドを抽出 · 精製すればよい。

上記 ( 1 ) の工程によって得られた D NA断片試料 (分子長が異なる 切断 DN A分子の集合体) を用い、 P C R法により D N Aシャフリング を行う。 P C Rは、 上記 D NA断片試料、 dAT P、 d GT P、 d C T Pおよび d T T Pおよび DNAポリメラーゼを含む反応溶液を用いて行 Ό。
これによれば、上記 D N A断片試料中の二本鎖の各切断 DNA分子は、 相互に铸型あるいはプライマ一となり、 ォ一バーラップ伸長することに より、 相同 D N A間の組換えが行われ、 上記 ( 1 ) の工程中、 最初に用 いた錶型遺伝子の変異遺伝子が形成される。 このことは、 後記実施例に おける、 終止コドンを導入した緑色蛍光蛋白質 (G F P u v) 遺伝子を 用いた実験により確かめられている。
一方、 本発明の試薬キッ トは、 少なく とも、 d UT Pおよびエンドヌ クレア一ゼ Vをそれぞれ試薬として含み、 このキッ トは、 DNAシャフ
リングにおける、 上記 ( 1 ) の D N A断片試料の調製工程に使用する。 この工程においては、 この試薬キッ トのほかに、 P C Rに使用する試薬 として、 dAT P、 d GT P、 d C T Pおよび d T T Pおよび DNAポ リメラーゼ等も使用するが、 これらは既存のものを使用してもよいし、 これら試薬を、 本発明の試薬キッ トに組み込んでもよい。
さらに、 本発明の上記 ( 2 ) の D N Aシャフリング工程においては、 d AT P、 d GT P、 d C T Pおよび d T T Pおよび DNAポリメラー ゼ等の P C Rに通常使用する試薬を使用するが、 これらを 1つの試薬セ ッ トとして、 上記本発明の試薬キッ トと組み合わせて、 DNAシャフリ ング試薬キッ トとしてもよい。
本明細書は本願の優先権の基礎である 日本国特許出願 特願 2002-232236号の明細書および/または図面に記載される内容を包含する < 図面の簡単な説明
図 1は、 本発明の実施例における実験系の概略を示す図である。 図 1 中、 矢印上は、 終始コドンの導入を示し、 矢印下は MAシャフリングを 示す。
図 2は、 ゥラシル置換量が各々異なる G F P遺伝子 D N Aを、 エンド ヌクレアーゼ Vにより切断して得られた D N A断片試料の電気泳動写真 である。 発明を実施するための最良の形態
以下、 本発明を実施例により具体的に説明するが、 本発明はこれら実 施例により特に限定されるものではない。
下記実施例の実験系の概略を図 1 に示す。 本実施例においては、 G F P u V遺伝子のコ一ド領域内に終止コ ドンを有する 2種の変異体を用い た。 一つの変異体は 6 0番目のロイシンがオーカ一終止コ ドンに変わつ たものであり、 もう一つは 1 2 0番目のバリ ンがォ一カー終止コ ドンに
変わったものである。 以下これらの変異体を 6 0 *、 1 2 0 *と記す。 これらの変異体は、 構造遺伝子が分断されているため不活性であり、 全 長の G F P u Vのように緑色蛍光を発することはない。 一方、 これらの 遺伝子が両終止コ ドンの間で組換わり、 終止コ ドンを含まない配列が生 成され全長の G F P u vの構造遺伝子が復活すれば蛍光を回復する。 こ のような実験系のもと、 エンドヌクレアーゼ Vを用いたこれらの 2種類 の遺伝子の断片化、 ならびに各断片を混合し D N Aシャフリングを行う ことで、 蛍光を回復する遺伝子があらわれるか否かを検定したものであ る。
実施例 1
( 1 ) 基質 D N Aの調製
a . 断片化の基質となる G F P u V遺伝子変異体を含むプラスミ ドの 作成 ;
G F P u v遺伝子以外のベクタ一部分は、 D a t e ら (Date, T. , Tanihara, K. , and Numura, N. 1990. Construction of Escherichia coli vectors for express ion and mutagenesis: synthesis of human c-Myc protein that is initiated at a non-AUG codon in exon 1. Gene. 90, 141-144) により作成された p TD— t a cをもとに、 該プラスミ ドの有 する唯一の N c o I部位を N d e I認識部位に変換したプラスミ ド p T D - t a c N dを作成した (p TD— t a cの CCATGG配列が CATATG と 置き換わっている)。 次に、 G F P u v遺伝子の該ベクタ一へのクロ一二 ン グ は、 以下の よ う に行われた。 即ち 、 2 つ の プラ イ マ 一 ( 5'-GGCATATGAGTAAAGGAGAAGAACTTTT-3'、 配列表の配列番号 1 及び 5'-GGTAATGGTAGCGACCGGCG-3\ 同配列番号 2 ) を用いて、 クロンテック社 製の G F P u vをコードするプラスミ ド pG F P u vより G F P遺伝子 領域を定法に従い P C R法により増幅した。 次に、 増幅断片を制限酵素 N d e I及び E c o R Iで切断し、 先に同様の制限酵素により切断して おいた p TD— t a c N dを定法に従いライゲーシヨンにより連結した
こうして得られた G F P u vを発現する組み換えプラスミ ド p T G u v 1の塩基配列を配列表の配列番号 3に示す。 また、 G F P遺伝子領域の 塩基配列を同配列番号 4に示す。
b . 終止コ ドンの導入 ;
こうして得られた G F P u Vを発現する組み換えプラスミ ドに対し、 ス トラタジーン社製 Q u i k C h a n g eキッ トにより部位指定突然変 異を行い、 6 0番目及び 1 2 0番目の位置のアミノ酸置換を行った。 即 ち、 2組のプライマー ( 6 0番目に対して
5' -CTACCTGTTCCATGGCCAACATAAGTCACTACTTTCTCTTATGG-3\ 同配列番号 5 及び 5' -CCATAAGAGAAAGTAGTGACTTATGTTGGCCATGGAACAGGTAG-3\ 同配列番 号 6、 1 2 0番目に対して
5' -GAAGTCAAGTTTGAAGGTGATACCCTTTAAAATCGTATCGAGTTAAAAGGTATTG-3'、同 配列番号 7、 及び
5, -CAATACCTTTTAACTCGATACGATTTTAAAGGGTATCACCTTCAAACTTGACTTC-3'、 同 配列番号 8 ) を用い、 Q u i k C h a n g eキッ トのマニュアルに従い 実験を行った。 こうして得られた不活性型の G F P u V遺伝子を組み込 んだプラスミ ドを p T G u v 6 0 *および p T G u v 1 2 0 *と名付け た。
c . ゥラシルによる置換 ;
断片化の基質となるゥラシルを含む D NAは、 p T G u v 6 0 *及び p T G u v l 2 0 *より、 G F P u v遺伝子をはさむ上流 (p TD _ f wd、 5'- AGCGGATAACAATTTCACACAGG- 3'、 同配列番号 9 ) 及び下流プライ マー (p T D— r e v、 5' -GTAAAACGACGGCCAGTGCC-3'、 同配列番号 1 0 ) を用い、 d UT P存在下で各変異体の G F P遺伝子領域を T a q 2 0 0 0 D N Aポリメラーゼ (ス トラタジーン社製) により? C R増幅するこ とにより増幅された二本鎖 D NA断片を用いた。 その際、 d T T Pと d UT Pの合計濃度を 0. 2 mMとし、 両ヌクレオチドの比率を種々に変え て行った ( 1 : 0、 0. 7 5 : 0. 2 5、 0. 5 : 0. 5、 0. 2 5 :
0. 7 5、 0 : 1 )。 すなわち、 d T T Pと d UT Pの濃度の組み合わせ は、 0. 2 mM : 0 mM、 0. 1 5 m M : 0. 0 5 mM、 0. I mM : 0. l mM、 0. 0 5 m M : 0. 1 5 mM、 0 m M : 0. 2 mMとした。 反応溶液の組成は、 1 0 mM トリス-塩酸緩衝液 (p H 8. 3 )、 5 O mM 塩化カリウム、 1. 5 mM 塩化マグネシウム、 各 2 5 M プ ライマ一 (p TD— : f wd及び p TD— r e v;)、 各 0. 2 mMの d AT P, d C T P, d G T P及び各種濃度の d T T P · d U T P混合溶液、 1 n g 鍀型プラスミ ド、 および 0. 2 5ユニッ トの T a q 2 0 0 0 D N Aポリメラーゼである。増幅反応は、 9 5 °Cで 1分間保温後、 9 4°C 3 0秒、 5 5 °C 3 0秒、 7 2 3 0秒の温度サイクルを 3 0回繰り返し、 最後に 7 2 °Cで 5分間保温することにより行った。
( 2 ) エンドヌクレア一ゼ Vによる D N Aの断片化
上記 ( 1 ) c . で得られた各 P C R産物 (各 1. 5 H g ) を精製後、 1 0 mM HE P E S - KOH (p H 7. 4)、 5 0 mM 塩化ナトリ ゥム、 0 · 5 mM 塩化マンガン、 0. 5ユニッ トのエンドヌクレア一 ゼ V (トレビジェン社製) 中、 3 7 °Cで 2 4時間反応させた。 反応液を キアゲン社製 Q I A q u i c kキッ トにより精製し、 3 0 j 1 の水に溶 解した。 6 0 *の試料に関し、 一部 ( 3 1 ) をァガロースゲル電気泳 動により分離し、 断片のサイズを見積もった (図 2 )。 なお、 図中 Mレー ンは、 分子量マ一カーついて行った泳動図である。 その結果、 d TT P = 0. 2 mM、 d UT P = 0 mM条件下で得られた D N A試料では全長 DNAのままであるが (レーン 1 )、 d T T P = 0. 1 5 mM, d UT P = 0. 0 5 mM (レーン 2 )、 d T T P = 0. l mM、 d UT P = 0. 1 mM (レーン 3 ) と d UT Pの濃度が高くなるにつれ D N A断片長が短 くなり、 d T T P = 0. 0 5 mM、 d UT P = 0. 1 5 mM条件下で得 られた D N A断片試料が 2 0 0〜4 0 0 b pの断片サイズを与え(図 2 , レーン 4 )、 この DNA断片試料を用いて以下の DNAシャフリング実験 を行った。
( 3 ) D N Aシャフリング
上記 ( 2 ) で得られたレーン 4に対応する D NA断片試料を用い、 P C R反応による D N Aシヤフリングを行った。 反応溶液の組成は、 1 0 mM トリスロ塩酸緩衝液 (p H 8 . 3 )、 5 0 mM 塩化カリウム、 1. 5 mM 塩化マグネシウム、 各 0. 2 mMの d AT P、 d C T P, d G T P、 d T T P、 各変異体の D N A断片各 0. 5 β g および 0. 2 5ュニッ トの T a Q 2 0 0 0 D NAポリメラ一ゼである。 増幅反応は、 9 5 °Cで 1分間保温後、 9 4 °C 3 0秒、 5 5 °C 3 0秒、 7 2 °C 3 0秒 の温度サイクルを 3 0回繰り返し、 最後に 7 2 °Cで 5分間保温すること により行った。
実施例 2
( 1 ) DN Aシャフリング産物から全長遺伝子の増幅
上記 ( 2 ) で得られた D N Aシャフリング産物から全長遺伝子を再増 幅するため、 G F P遺伝子をはさむプライマ一を用いて P C Rを行った。 反応溶液の組成は、 1 0 mM トリスロ塩酸緩衝液 (p H 8. 3 )、 5 0 mM 塩化カリウム、 1. 5 mM 塩化マグネシウム、各 2 5 M プ ライマ一 ( p TD— i wd及び p TD— r e v)、 各 0. 2 mMの d AT P、 d C T P、 d GT P、 d TT P、 上記 ( 3 ) で得られた反応溶液 1 l 、 および 0. 2 5ュニッ トの丁 & 2 0 0 0 D NAポリメラーゼで ある。 増幅反応は、 9 5 で 1分間保温後、 9 4°C 3 0秒、 5 5 °C 3 0秒、 7 2 °C 3 0秒の温度サイクルを 2 5回繰り返し、 最後に 7 2 で 5分間保温することにより行った。
( 2 ) ライブラリの構築とスクリーニング
得られた増幅断片を精製し、 制限酵素 N d e l及び E c o R Iで切断 した。 同様に、 p TD— t a c N dベクタ一に関しても同様の制限酵素 で消化し、 両 D NA断片をライゲ一シヨン反応により連結させた。
反応液の一部をェシエリシァ コリー J M 1 0 9のコンビテントセル に導入し、 1 0 O g /m l のアンピシリン及び 0. 5 mMのイソプロ
ピル一 j3— D—チォガラク トピラノシドを含む L B寒天プレート上、 3 7 °Cで 1 6時間培養しコロニーを形成させた。 形成したコロニー数を白 色光下で計測し、 また緑色蛍光を発するコロニー数を 3 6 5 Mの紫外線 を照射し計測した。 その結果、 全 1 0 0クローンのうち、 1 0個が蛍光 性であった。
( 3 ) D N Aシャフリングライブラリ一により得られた蛍光性クローン の塩基配列
( 2 ) で得た蛍光性コロニー 1 0個を培養し、 定法によりプラスミ ド を精製し、 定法により G F P遺伝子の塩基配列を決定した。 その結果、 蛍光性コロニーのいずれにおいても 6 0番目、 1 2 0番目の位置に終止 コ ドンを含まない全長の G F P u V遺伝子をコードするものであった。 すなわち、 DNAシャフリングの基質として用いた終止コ ドンを含む遺 伝子はもともと存在した終止コ ドン間で組換わっており、 D N Aシャフ リングが行われていることが判明した。 産業上の利用可能性
従来、 DNAシャフリングにおける遺伝子 D N Aの断片化工程におい ては、 基質特異性の低いデォキシリポヌクレアーゼによる方法がよく用 いられているが、 この方法では、 反応に関わるさまざまな因子 (デォキ シリポヌクレアーゼ I の酵素量、 基質 DNAの純度や濃度、 反応温度、 反応時間) を正確に制御し、 直当な反応時間で反応を集結させ部分分解 産物を取得する必要があった。 これに対して、 本発明は、 DNAシャフ リングにおける遺伝子 D N Aの断片化工程において、 遺伝子 DNAの塩 基配列中、 チミンをゥラシルに所定の量置換させた後、 エンドヌクレア ーゼ Vを作用させ、 置換されたゥラシル部位の 2— 3塩基下流において 該 DNAを切断して、 D NA断片試料を得るものであり、 基質を消費し た段階で反応が終結するため、 完全消化により断片化がおこなわれ、 反 応の最適条件の検討や反応の進行状況のモニターの必要がないものであ
り、 得られる D N A断片試料の分子長の分布状態は、 ゥラシルの置換量 を制御することにより行うものであり、 極めて簡便な操作でよく、 しか も再現性が良好なものである。 本明細書に引用されたすベての刊行物は、 その内容の全体を本明細書 に取り込むものとする。 また、 添付の請求の範囲に記載される技術思想 および発明の範囲を逸脱しない範囲内で本発明の種々の変形および変更 が可能であることは当業者には容易に理解されるであろう。 本発明はこ のような変形および変更をも包含することを意図している。
Claims
1. D NAの塩基配列中、チミンをランダムにゥラシルで置換した後、 エンドヌクレアーゼ Vを作用させることにより DN Aを断片化すること を特徴とする、 DNA断片試料の調製方法。
2. 請求項 1 により調製された D NA断片試料を用いて、 DNAシャ フリングを行うことを特徴とする、 DN Aシャフリング方法。
3. デォキシゥリジントリホスフエ一ト (d UT P ) およびエンドヌ クレアーゼ Vをそれぞれ試薬として含むことを特徴とする、 DNAを断 片化するための試薬キッ ト
4. さらに、 デォキシアデノシントリホスフェート (d AT P)、 デォ キシグアノシントリホスフエ一ト (d GT P)、 デォキシシチジンントリ ホスフェート (d C T P)およぴデォキシチミジントリホスフェート (d T T P)、並びに DNAポリメラーゼを試薬として含むことを特徴とする 請求項 3に記載の試薬キッ 卜。
5. D N Aシャフリングにおける、 DN A断片化に使用するものであ る、 請求項 3または 4に記載の試薬キッ ト。
6 · デォキシゥリジントリホスフエ一ト (d UT P)、 デォキシアデノ シントリホスフェート ( d A T P )、 デォキシグアノシントリホスフエ一 ト (d GT P)、 デォキシシチジンントリホスフェート (d C T P) およ びデォキシチミジントリホスフエ一ト (d TT P)、 DNAポリメラ一ゼ 並びにびエンドヌクレアーゼ Vをそれぞれ試薬として含むことを特徴と する、 DN Aシャフリング試薬キッ ト。
