Abstract

Beschrieben wird ein Verfahren zur gleichzeitigen, parallelen, selektiven Anreicherung von verschiedenen DNA-Abschnitten, die aus verschiedenen Geweben durch komplexe Amplifikationen erhalten werden und gewünschte individuelle Sequenzeigenschaften haben. Diese Eigenschaften der DNA-Fragmente, vor allem das Vorhandensein von 5-Methylcytosinen, sind durch Hybridisierung auf DNA Microarrays identifizierbar. Die Anreicherung der gewünschten DNA-Fragmente erfolgt durch mehrmalige Wiederholung der Arbeitsschritte (Hybridisierung, Dehybridisierung und Reamplifikation). Das Verfahren kombiniert das SELEX-Verfahren mit komplexen DNA-Arrays, die für die Anreicherung der DNA-Fragmente ververwendet werden. Die Amplifikate werden abschließend hinsichtlich ihrer Sequenz analysiert.
