Abstract

Die vorliegende Erfindung betrifft ein Verfahren zum Herstellen von heterologem Protein in einem filamentösen Pilz sowie dafür geeignete Promotoren. Ferner betrifft die vorliegende Erfindung Vektoren und Wirtszellen sowie einen Kit und deren Verwendung. Aufgabe der vorliegenden Erfindung ist es, ein Verfahren zum herstellen von heterologem Protein in einem filamentösen Pilz bereitzustellen, das eine effiziente Produktion heterologer Proteine erlaubt und bei dem eine Kontamination des erzeugten Proteins durch vegetative Sporen vermieden wird. Erfindungsgemäß wird diese Aufgabe durch ein Verfahren zum Herstellen von heterologem Protein in einem filamentösen Pilz gelöst, das (a) das Kultivieren eines homothallischen Pilzes der Familie Sordariaceae, der eine Espressionskassette enthält, die in funktioneller Verbindung die folgenden Elemente enthält: einen in dem Pilz der Familie Sordariaceae aktiven Promotor; ein heterologes Gen und einen in dem Pilz der Familie Sordariaceae aktiven Terminator, sowie (b) das Ernten des erzeugten Proteins in an sich bekannter Weise umfasst.
