Abstract

L'invention concerne une méthode d'échantillonnage d'acide nucléique transdermique peu invasive, qui consiste à percer la couche supérieure de la peau (20) pour atteindre l'épiderme sous-jacent, à l'aide de plusieurs microprojections (4). Des cellules cutanées vivantes de l'épiderme sous-jacent sont désorganisées, ce qui les amène à libérer leurs contenus, notamment leurs acides nucléiques (c.-à-d. l'ADN, l'ARN, leurs fragments ou autres matières d'acide polynucléique trouvés dans les noyaux et/ou les mitochondries de cellules). L'acide nucléique est recueilli sur les surfaces des microprojections et/ou dans un réservoir de collecte d'acide nucléique séparé. L'acide nucléique recueilli est ensuite analysé par des techniques de réaction en chaîne de la polymérase (PCR). Facultativement, un dispositif d'aspiration (10) opère une dépression partielle à travers des ouvertures (8) ménagés dans l'élément à microprojections (6), jusqu'aux microcoupures pratiquées dans la peau (20), afin d'améliorer l'écoulement de liquides (c.-à-d. organiques) intracellulaires et extracellulaires contenant l'acide nucléique.
