Abstract

Beschrieben werden genetisch veränderte Fibroblastenzellen mit den folgenden Eigenschaften: (a) ein eine Untereinheit des Transkriptionsfaktors AP-1 codierendes Gen ihres Genoms ist inaktiviert und/oder eine auf den Transkriptionsfaktor AP-1 einwirkende Komponente des Signaltransduktionsweges ist modifiziert mit der Folge einer veränderten AP-1 Aktivität (b) sie enthält in einem Expressionsvektor mit einem Promotor funktionell verknüpft das Gen von (a) in einer solchen Form, dass erst nach Induktion des Promotors die aktive Untereinheit exprimiert wird oder die von dem Gen codierte Untereinheit in inaktiver und aktivierbarer Form vorliegt. Ferner wird ein Verfahren unter Verwendung der erfindungsgemässen Fibroblastenzelle zur Identifizierung einer Verbindung beschrieben, die die AP-1 abhängige Differenzierung und/oder Proliferation von Keratinozyten fördern, hemmen oder modifizieren kann oder eines Gens, das an der AP-1, abhängigen Differenzierung und/oder Proliferation von Keratinozyten beteiligt ist, sowie die Verwendung der nach dem erfindungsgemässen Verfahren identifizierten Testverbindung bzw. des Gens zur Behandlung oder Diagnose von Erkankungen, die mit einer gestörten Proliferation und/oder Differenzierung von Keratinozyten assoziiert sind.
