Abstract

Cette invention se rapporte à des procédés servant à obtenir des enzymes qui fixent des substrats cibles et qui catalysent des réactions souhaitées. Des protéines à barreau α/β sont classées en deux classes sur la base de la structure des couvercle catalytiques. Les couvercles peuvent être greffés sur des échafaudages, avec des modifications mineures additionnelles au niveau des résidus conservés et non conservés, pour former des enzymes de produits candidats permettant de trier les propriétés souhaitées. La conception d'une nouvelle enzyme qui fixe un substrat cible et qui catalyse une réaction choisie est facilitée par la sélection d'un échafaudage qui fixe le substrat et d'un couvercle catalytique de la bonne classe pour la réaction souhaitée. Une mutagenèse ciblée ou focalisée peut être utilisée pour affiner la fixation du substrat et la catalyse.
