Abstract
Cette invention concerne une enzyme thyroïdienne, composant unique ou majeur de la NADPH oxydase de thyroïde, l'acide nucléique codant pour cette enzyme et leurs applications diagnostiques et thérapeutiques, notamment dans les hyperthyroïdies, les hypothyroïdies, les cancers de la thyroïde et d'autres tumeurs malignes.

Enzyme thyroïdienne NADPH oxydase. acide nucléique codant pour cette enzyme et leurs applications.
La présente invention concerne une enzyme thyroïdienne, composant de la NADPH oxydase de thyroïde, l'acide nucléique codant pour cette enzyme et leurs applications diagnostiques et thérapeutiques.
La thyroperoxydase (TPO) est l'enzyme qui catalyse l'iodation de la thyroglobuline et le couplage des iodotyrosines en hormones. Mais la TPO en tant que telle ne présente aucune activité catalytique. L'activité peroxydasique n'est en effet catalysée que par le "composé I", ainsi que par les composés enzymatiques qui en dérivent. C'est la réaction d'oxydation de Thème de la TPO par une molécule de peroxyde d'hydrogène (H2O2) qui conduit à la formation du composé I. Sans H2O2, il ne peut donc y avoir de biosynthèse des hormones. Le thyrocyte contient un système enzymatique capable de produire des quantités importantes de H2O2. Il est évident que l'activité d'un tel système doit être rigoureusement contrôlée pour autoriser la formation d'une quantité de H2O2 suffisante pour la synthèse des hormones et le stockage de l'iodure, tout en évitant cependant la surproduction de cet oxydant puissant. Le rôle essentiel du peroxyde d'hydrogène dans la synthèse hormonale et le stockage de l'iode ainsi que ses propriétés oxydantes font aussi de l'enzyme qui le génère, une source potentielle de troubles thyroïdiens, rendant d'autant plus intéressante son identification moléculaire.
Bien qu'une production de H2O2 par des coupes de thyroïde ait été mise en évidence en 1970, la nature biochimique du système générateur est restée longtemps hypothétique.
En 1981 , Bjôrkman et al ont localisé cytochimiquement un système générateur de H2O2 au pôle apical de follicules ouverts de thyroïde de rat. Cette localisation coïncidait parfaitement avec celle du site majeur d'iodation, mise en évidence à la fin des années 70 à , l'interface de la membrane apicale et de la lumière folliculaire (Ekholm et al, 1981 ), et suggérait fortement qu'il s'agissait d'une enzyme associée à la membrane plasmique. Le seul système générateur de H2O2 connu associé à la membrane plasmique
étant la NADPH oxydase de phagocytes, les mêmes auteurs ont proposé que cette enzyme ou une enzyme similaire soit impliquée dans la biosynthèse des hormones thyroïdiennes.
Conformément à l'hypothèse de Bjόrkman et al, Dème et al en 1985 ont rapporté une formation de H2O2 en système acellulaire en incubant des particules de thyroïde de porc avec du NADPH, puis ont montré que cette production de H2O2 était stimulée réversiblement par des concentrations submicromolaires de Ca2+. En 1987, Nakamura et al confirmaient l'existence d'une NADPH oxydase dépendante du Ca2+ dans une fraction enrichie en membranes plasmiques et en cytosquelette d'actine.
La solubilisation de la NADPH oxydase des membranes plasmiques de thyroïde de porc s'est avérée délicate (Gorin et al, 1996). Une augmentation importante de la force ionique en présence de cytochalasine B et de détergent a été nécessaire pour obtenir son extraction de la membrane. La NADPH oxydase ainsi solubilisée est toutefois fortement affectée par ce traitement, puisqu'elle devient incapable de produire de l'H2O2. Une dialyse de plusieurs heures de l'extrait restaure la production NADPH-dépendante de H2O2l ainsi que sa dépendance vis à vis du Ca2+, montrant que si l'enzyme est formée de plusieurs facteurs, tous ses composants ont bien été solubilisés. Le pourcentage de reconstitution de l'activité varie entre 50 et 90 % selon les extraits.
A la suite de ia solubilisation, Gorin et al ne sont cependant parvenus qu'à purifier seulement partiellement l'enzyme.
Les auteurs de la présente invention ont maintenant réussi à cloner l'acide nucléique codant pour un polypeptide qui est le composant unique ou majeur de la NADPH oxydase de thyroïde, obtenant par suite la séquence d'acides aminés de ce polypeptide.
La présente invention a donc pour objet un polypeptide isolé, composant unique ou majeur de la NADPH oxydase de thyroïde comprenant une séquence d'acides aminés choisie parmi les séquences SEQ ID n° 2, ou SEQ ID n° 4. Ce composant a été désigné par p138tox-
La séquence SEQ ID n° 2 représente la séquence d'acides aminés de la p138 o de la NADPH oxydase de thyroïde humaine.
La séquence SEQ ID n° 4 représente la séquence d'acides aminés de la p138tox de la NADPH oxydase de thyroïde de porc. Sont également compris dans l'invention les polypeptides homologues des polypeptides de séquence SEQ ID n°2 ou n°4.
Par « polypeptide homologue », on entend tout polypeptide ayant une séquence d'acides aminés qui diffère des séquences SEQ ID n°2 ou n°4 par substitution, délétion et/ou insertion d'un ou plusieurs acides aminés, à des positions telles que ces modifications ne portent pas significativement atteinte à l'activité de la p138tox.
Lesdites substitutions sont de préférence des substitutions conservatives, c'est-à-dire des substitutions d'acides aminés de même classe, tels que des substitutions d'acides aminés aux chaînes latérales non chargées (tels que l'asparagine, la glutamine, la serine, la thréonine, et la tyrosine), d'acides aminés aux chaînes latérales basiques (tels que la lysine, l'arginine, et l'histidine), d'acides aminés aux chaînes latérales acides (tels que l'acide aspartique et l'acide glutamique) ; d'acides aminés aux chaînes latérales apolaires (tels que la glycine, l'alanine, la valine, la leucine, l'isoleucine, la praline, la phénylalanine, la méthionine, le tryptophane, et la cystéine).
De préférence, une telle séquence d'acides aminés homologue est identique à au moins 65 %, de préférence au moins 75 %, de préférence au moins 85 %, de préférence encore au moins 95 % des séquences SEQ ID n° 2 ou n°4. Ces séquences homologues peuvent être notamment des séquences d'autres espèces de mammifères que l'homme ou le porc. Ces séquences homologues comprennent également tout fragment des séquences SEQID n° 2 ou n° 4 possédant l'activité biologique de la p138tox de la NADPH oxydase de thyroïde. Par "activité biologique de la p138tox", on se réfère notamment à l'activité connue et mesurable de la NADPH oxydase de thyroïde : il s'agit de la production d'H2O2 et/ou d'activités de réduction d'accepteurs d'électrons exogènes tels que par exemple le nitroblue tetrazolium (NBT) et le ferricyanure
de potassium. Cette enzyme transmembranaire transfère les électrons du NADPH cytosolique vers l'oxygène moléculaire dans la lumière du follicule thyroïdien. Elle fournit ainsi le peroxyde d'hydrogène indispensable à l'activité de la thyroperoxydase, qui peut alors oxyder l'iodure et ioder la thyroglobuline. L'homologie est généralement déterminée en utilisant un logiciel d'analyse de séquence (par exemple, Séquence Analysis Software Package of the Genetics Computer Group, University of Wisconsin Biotechnology Center, 1710 University Avenue, Madison, Wl 53705). Des séquences d'acides aminés similaires sont alignées pour obtenir le maximum de degré d'homologie (i.e. identité). A cette fin, il peut être nécessaire d'introduire de manière artificielle des espaces (« gaps ») dans la séquence. Une fois l'alignement optimal réalisé, le degré d'homologie (i.e. identité) est établi par enregistrement de toutes les positions pour lesquelles les acides aminés des deux séquences comparées sont identiques, par rapport au nombre total de positions.
La présente invention a également pour objet un acide nucléique isolé codant pour un polypeptide ayant une activité de p138tox, tel que défini ci- dessus.
La présente invention concerne plus particulièrement les acides nucléiques comprenant une séquence nucléotidique choisie parmi la séquence SEQ ID n° 1 , SEQ ID n° 3, toute séquence homologue biologiquement actif desdites séquences.
La séquence SEQ ID n° 1 représente la séquence d'ADNc codant pour la p138tox de la NADPH oxydase de thyroïde humaine. La séquence SEQ ID n° 3 représente la séquence d'ADNc codant pour la p138tox de la NADPH oxydase de thyroïde de porc.
Par "séquence nucléotidique homologue", on entend toute séquence nucléotidique qui diffère de la séquence SEQ ID n°1 ou SEQ ID n° 3 par substitution, délétion, et ou insertion d'un ou plusieurs nucléotides, à des positions telles que ces séquences nucléotidiques homologues codent pour des polypeptides tels que définis précédemment.
De préférence, une telle séquence nucléotidique homologue est identique à au moins 60 % des séquences SEQ ID n° 1 ou n° 3, de préférence au moins 70 %, de préférence encore au moins 95 %.
De manière préférentielle, une telle séquence nucléotidique homologue hybride spécifiquement aux séquences complémentaires des séquences SEQ ID n° 1 ou n° 3, dans des conditions stringentes. Les paramètres définissant les conditions de stringence dépendent de la température à laquelle 50% des brins appariés se séparent (Tm).
Pour les séquences comprenant plus de 30 bases, Tm est définie par la relation : Tm=81 , 5+0,41 (%G+C)+16, 6Log(concentration en cations) -
0,63(%formamide) -(600/nombre de bases) (Sambrook et al, Molecuiar
Cloning, A laboratory manual, Cold Spring Harbor laboratory Press, 1989, pages 9.54-9.62).
Pour les séquences de longueur inférieure à 30 bases, Tm est définie par la relation : Tm= 4(G+C) + 2 (A+T).
Dans des conditions de stringence appropriées, auxquelles les séquences aspécifiques n'hybrident pas, la température d'hybridation est approximativement de 5 à 30°C, de préférence de 5 à 10°C en dessous de Tm, et les tampons d'hybridation utilisés sont de préférence des solutions de force ionique élevée telle qu'une solution 6xSSC par exemple.
Les séquences nucléotidiques homologues comprennent également tout fragment des séquences SEQ ID n° 1 ou SEQ ID n° 3, qui code pour un peptide possédant l'activité biologique de la p138to , telle que définie précédemment. Les différentes séquences nucléotidiques de l'invention peuvent être d'origine artificielle ou non. Il peut s'agir de séquences d'ADN ou d'ARN, obtenues par criblage de banques de séquences au moyen de sondes élaborées sur la base de la séquence SEQ ID n° 1 ou n° 3. De telles banques peuvent être préparées par des techniques classiques de biologie moléculaire, connues de l'homme de l'art.
Les séquences nucléotidiques selon l'invention peuvent également être préparées par synthèse chimique, ou encore par des méthodes
mixtes incluant la modification chimique ou enzymatique de séquences obtenues par criblage des banques.
Les séquences nucléotidiques de l'invention permettent la réalisation de sondes nucléotidiques, hybridant spécifiquement avec une séquence SEQ ID n° 1 ou n° 3 selon l'invention. Les conditions d'hybridation appropriées correspondent aux conditions de température et de force ionique usuellement utilisées par l'homme du métier, de préférence dans des conditions stringentes telles que définies précédemment. De telles sondes font également partie de l'invention. Elles peuvent être utilisées comme outil de diagnostic in vitro pour la détection, par des expériences d'hybridation, notamment d'hybridation "in situ", de transcrits spécifiques des polypeptides de l'invention dans des échantillons biologiques ou pour la mise en évidence de synthèses aberrantes ou d'un mauvais épissage. Les anomalies génétiques résultant d'un polymorphisme et les éventuelles mutations peuvent être mises en évidence par la technique dite de PCR (réaction en chaîne par la polymérase) en utilisant des amorces spécifiques déterminées à partir des séquences ID n°1 ou ID n°3.
Les sondes de l'invention comportent au minimum 10 nucléotides, et de préférence au moins 14 nucléotides, préférentiellement au moins 20 nucléotides, préférentiellement encore au moins 50 nucléotides, et au maximum comportent la totalité de la séquence nucléotidique SEQ ID n° 1 ou n° 3 ou de leur brins complémentaires.
Préférentiellement, les sondes de l'invention sont marquées, préalablement à leur utilisation. Pour cela, plusieurs techniques sont à la portée de l'homme du métier comme par exemple le marquage fluorescent, radioactif, chimioluminescent ou enzymatique.
Les méthodes de diagnostic in vitro dans lesquelles ces sondes nucléotidiques sont mises en oeuvre pour la détection de synthèses aberrantes ou d'anomalies génétiques, telles que la perte d'hétérozygotie, le réarrangement génique, les mutations pontuelles et les délétions au niveau des séquences nucléiques codant pour un peptide de l'invention, sont incluses dans la présente invention.
L'invention a aussi pour objet un procédé de détection de l'expression de p138tox de la NADPH oxydase de thyroïde dans un échantillon cellulaire ou tissulaire d'un mammifère, notamment d'un homme, comprenant les étapes consistant à :
- préparer l'ARN dudit échantillon ;
- mettre en contact ledit ARN obtenu avec une sonde ayant une séquence nucléotidique capable de s'hybrider spécifiquement avec un acide nucléique tel que défini précédemment ; - détecter la présence d'ARNm hybridant avec cette sonde, indicatrice de l'expression de la p138tox de la NADPH oxydase de thyroïde.
L'invention a aussi pour objet un procédé de détection de l'expression de la p138tox de la NADPH oxydase de thyroïde dans des cellules ou un tissu de mammifère, notamment d'un homme, par hybridation in situ, comprenant les étapes consistant à :
- mettre en contact lesdites cellules ou ledit tissu avec une sonde ayant une séquence nucléotidique capable de s'hybrider spécifiquement avec un acide nucléique tel que défini précédemment ;
- détecter la présence d'ARNm hybridant avec cette sonde, indicatrice de l'expression de la p138tox de la NADPH oxydase de thyroïde.
Les séquences nucléotidiques de l'invention sont également utiles pour la fabrication et l'utilisation d'amorces oiigonucléotidiques sens et/ou antisens pour des réactions de séquençage ou d'amplification spécifique selon la technique de PCR ou toute autre variante de celle-ci.
Les séquences nucléotidiques selon l'invention peuvent par ailleurs être utilisées pour la production de polypeptides recombinants possédant l'activité biologique de la p138tox de la NADPH oxydase de thyroïde tels que précédemment définis. Ces polypeptides peuvent être produits à partir des séquences nucléotidiques définies ci-dessus, selon des techniques de production de produits recombinants connues de l'homme du métier.
Selon un mode de réalisation de l'invention, la séquence nucléotidique peut être insérée dans un vecteur d'expression, dans lequel elle est liée de manière opérante à des éléments permettant la régulation de son expression, tels que notamment des promoteurs et/ou terminateurs de transcription.
Les signaux contrôlant l'expression des séquences nucléotidiques (promoteurs, activateurs, séquences de terminaison...) sont choisis en fonction de l'hôte cellulaire utilisé. A cet effet, les séquences nucléotidiques selon l'invention peuvent être insérées dans des vecteurs à réplication autonome au sein de l'hôte choisi, ou des vecteurs intégratifs de l'hôte choisi. De tels vecteurs seront préparés selon les méthodes couramment utilisées par l'homme du métier, et les clones en résultant peuvent être introduits dans un hôte approprié par des méthodes standard, telles que par exemple l'électroporation ou la précipitation au phosphate de calcium. Tous vecteurs de clonage et/ou d'expression tels que décrits ci- dessus, contenant une des séquences nucléotidiques définies selon l'invention, font également partie de la présente invention.
L'invention vise en outre les cellules hôtes transfectées, de manière transitoire ou stable, par ces vecteurs d'expression. Ces cellules peuvent être obtenues par l'introduction dans des cellules hôtes, procaryotes ou eucaryotes, d'une séquence nucléotidique insérée dans un vecteur tel que défini ci-dessus, puis la mise en culture desdites cellules dans des conditions permettant la réplication et/ou l'expression de la séquence nucléotidique transfectée. La présente invention a ainsi pour objet un procédé de production d'un polypeptide recombinant à activité de p138 o de la NADPH oxydase de thyroïde, dans lequel on cultive des cellules transformées dans des conditions permettant l'expression d'un polypeptide comprenant une séquence d'acides aminés choisie parmi la séquence SEQ ID n°2 ou n°4, ou toute séquence homologue et on récupère ledit polypeptide.
Ce procédé comprend généralement les étapes consistant à : i) insérer une séquence nucléotidique telle que définie précédemment dans un vecteur d'expression, ladite séquence nucléotidique
étant liée de manière opérante avec des éléments permettant la régulation de son expression ; ii) transformer une cellule hôte avec le vecteur ainsi obtenu ; iii) cultiver ladite cellule hôte dans des conditions permettant l'expression de ladite séquence nucléotidique ; iv) recueillir le peptide recombinant exprimé ; v) éventuellement purifier ledit peptide.
La présente invention rend possible une régulation de NADPH oxydase dans les cellules de thyroïde, qui produisent de l'H2O2, dans les cas où ce peroxyde serait produit de manière ou en quantité anormales.
Une production incontrôlée de peroxyde d'hydrogène (H2O2) peut en effet induire des cassures dans l'ADN (Birnboim H.C et coll., 1982) et des modifications au niveau des bases thymine et guanine (Jackson J.H. et coll., 1989). Un échappement de la NADPH oxydase à son contrôle par le calcium peut donc être notamment une cause de transformation des thyrocytes normaux en cellules cancéreuses.
Les séquences nucléotidiques selon l'invention ont donc des utilisations dans le domaine thérapeutique, notamment pour la réalisation de séquences antisens, capables de s'hybrider spécifiquement avec une séquence d'acide nucléique, y compris un ARN messager, utilisables en thérapie génique. L'invention a ainsi pour objet des séquences antisens capables d'inhiber, au moins partiellement, la production de NADPH oxydase de thyroïde, telle que définie précédemment. De telles séquences sont avantageusement constituées par celles qui constituent le cadre de lecture codant pour la p138tox de la NADPH oxydase au niveau du transcrit.
La présente invention a donc pour objet une composition thérapeutique comprenant au moins un acide nucléique comprenant une séquence antisens capable de s'hybrider spécifiquement avec un acide nucléique codant pour la p138 o de la NADPH oxydase de thyroïde, en association avec un véhicule pharmaceutiquement acceptable.
Les séquences nucléotidiques selon l'invention peuvent en outre être utilisées pour transformer des cellules cibles et leur faire exprimer un
polypeptide possédant l'activité biologique de p138tox de la NADPH oxydase de thyroïde .
L'invention a donc également pour objet une composition pharmaceutique comprenant un acide nucléique selon l'invention codant pour un polypeptide possédant l'activité biologique de la p138tox de la NADPH oxydase de thyroïde, en association avec un véhicule pharmaceutiquement acceptable, ladite composition étant destinée à être utilisée en thérapie génique. L'acide nucléique d'intérêt, de préférence inséré dans un vecteur et placé sous le contrôle d'un promoteur de transcription spécifique de la glande thyroïde, peut être administré sous forme nue ou en association avec au moins un agent facilitant la transfection dudit acide nucléique.
Une telle composition pharmaceutique peut être par exemple utilisée en thérapie génique chez des malades affectés d'hypothyroïdies, notamment liées à un défaut d'organification de l'iodure (c'est-à-dire un défaut de formation de liaison covalente de l'iodure aux protéines cellulaires), résultant d'un défaut de production d'H2O2 dans la thyroïde.
L'invention permet également l'expression ectopique de la NADPH oxydase thyroïdienne, en particulier dans les cellules cancéreuses de diverses origines en vue de leur faire produire des quantités toxiques d'H2O2 susceptibles d'entraîner l'apoptose ou la nécrose des cellules tumorales. Pour cela, la séquence d'acide nucléique ou une partie de celle-ci associée à un promoteur de transcription spécifique du tissu cancéreux est insérée dans un vecteur d'expression.
Les séquences nucléotidiques codant pour la NADPH oxydase selon l'invention peuvent également être utilisées en co-expression ectopique avec le cotransporteur basolatéral Na+/V symporter (NIS) et la thyroperoxydase (TPO) en vue d'un traitement radioisotopique (131l) de diverses tumeurs. Pour cela, une séquence nucléique de p138tox de NADPH oxydase de thyroïde associée à un promoteur de transcription spécifique du tissu cancéreux est insérée dans un vecteur d'expression et co-transfectée avec le NIS et la TPO dont l'expression est sous le contrôle du même promoteur. Le traitement permet la capture de l'131l par la tumeur, son organification (liaison covalente à des protéines cellulaires) et la destruction de la tumeur.
Les séquences nucléotidiques selon l'invention peuvent être utilisées dans la production d'animaux transgéniques dont le gène de la
NADPH oxydase thyroïdienne est "éteint", délété ou muté. Ces animaux peuvent être utilisés dans la mise au point de méthodes de diagnostic, de médicaments ou de traitements à visées thérapeutiques.
La présente invention vise également des compositions pharmaceutiques comprenant une quantité efficace d'au moins un polypeptide tel que défini précédemment, en association avec un véhicule pharmaceutiquement acceptable. De telles compositions pharmaceutiques sont particulièrement utiles dans les cas d'hypothyroïdies telles que mentionnées précédemment.
La présente invention vise également des compositions pharmaceutiques comprenant une quantité efficace d'au moins un polypeptide modifié par rapport au polypeptide p138tox de telle sorte que ledit peptide mime la zone d'interaction avec la thyroperoxydase (TPO) avec pour conséquence l'inhibition de la NADPH oxydase et/ou l'inhibition de la TPO, en association avec un véhicule pharmaceutiquement acceptable.
De telles compositions pharmaceutiques sont particulièrement utiles dans le traitement d'affections impliquant une activité anormale de la NADPH oxydase de thyroïde, que l'on souhaite bloquer ou diminuer.
Une composition pharmaceutique selon l'invention peut être notamment administrée par voie orale, parentérale, intraveineuse, intramusculaire, sous-cutanée, percutanée ou par administration intra-nasale. La préparation des compositions pharmaceutiques qui contiennent des principes actifs dissous ou dispersés dans ces dernières, est bien connue de l'homme du métier. Généralement, ces compositions sont préparées sous la forme de solutions ou de suspensions injectables. Cependant, elles peuvent aussi être sous des formes solides appropriées pour préparer des solutions, ou des suspensions extemporanement. Les préparations peuvent aussi être émulsifiées.
Les modes d'administration, les posologies et les formes galéniques des compositions pharmaceutiques selon l'invention peuvent être
déterminés de manière usuelle par l'homme du métier, notamment selon les critères généralement pris en compte pour l'établissement d'un traitement thérapeutique adapté à un patient, comme par exemple l'âge ou le poids corporel du patient, la gravité de son état général, la tolérance au traitement, et les effets secondaires constatés, etc.
Le polypeptide de l'invention est également particulièrement utile pour le criblage d'inhibiteurs de la NADPH oxydase de la thyroïde, destinés à la fabrication d'un médicament pour la prévention et/ou le traitement d'affections impliquant une activité anormale de la NADPH oxydase de thyroïde.
Parmi ces affections, on peut citer notamment les hyperthyroïdies, comme la maladie de Grave's Basedow, ou les cancers de la thyroïde, en particulier les cancers de la thyroïde donnant des métastases (à savoir notamment les cancers folliculaires et papillaires moyennement différenciés et donnant des métastases), ou autres tumeurs malignes, telles que les cancers du colon, du duodénum, de l'intestin ou encore des ovaires.
La présente invention a également pour objet un procédé de détection d'anticorps anti-NADPH oxydase dans un échantillon biologique, dans lequel on met en contact ledit échantillon biologique avec un polypeptide selon l'invention, marqué de manière détectable et on détecte les complexes p138tox-anticorps.
Le polypeptide de l'invention peut ainsi permettre le criblage de serums de malades souffrant de maladies autoimmunes de la thyroïde telle que la maladie d'Hashimoto, par la recherche d'anticorps circulants anti- NADPH oxydase.
L'invention a également pour objet un anticorps monoclonal ou polyclonal dirigé spécifiquement contre la p138tox de la NADPH oxydase de thyroïde, ou un fragment dudit anticorps capable de se lier spécifiquement à la p138tox de la NADPH oxydase de thyroïde.
Des anticorps polyclonaux peuvent être obtenus à partir du sérum d'un animal immunisé contre la p138tox de la NADPH oxydase de thyroïde selon l'invention selon les modes opératoires usuels.
Les anticorps monoclonaux peuvent être obtenus selon la méthode classique de culture d'hybridomes décrite par Kôhler et Milstein (Nature, (1975), vol. 256, pp. 495-497).
L'invention a également pour objet un procédé de production d'anticorps dirigés contre la p138to de la NADPH oxydase de thyroïde dans lequel on immunise des animaux par administration d'un vecteur d'expression contenant une séquence nucléotidique selon l'invention.
L'invention vise également des animaux immunisés (tels que des souris) par injection d'un vecteur d'expression contenant une des séquences nucléotidiques définies selon l'invention en vue de faire produire des anticorps dirigés contre la NADPH oxydase.
Les anticorps ou fragments d'anticorps de l'invention sont par exemple des anticorps chimériques, des anticorps humanisés, des fragments
Fab et F(ab')2. Ils peuvent également se présenter sous forme d'immunoconjugués ou d'anticorps marqués. Par exemple, ils peuvent être associés à une toxine, telle la toxine diphtérique ou à un produit radioactif.
Les anticorps ainsi produits peuvent être notamment utilisés pour détecter et/ou doser la NADPH oxydase de thyroïde dans tout échantillon biologique susceptible d'en contenir. L'invention a donc également pour objet un procédé de détection et/ou de dosage immunologique de la NADPH oxydase de thyroïde dans un échantillon biologique d'un mammifère, notamment d'un homme, dans lequel : i) on met en contact ledit échantillon biologique avec un anticorps tel que défini précédemment, marqué de manière détectable ; ii) on observe la formation d'un complexe anticorps-NADPH oxydase de thyroïde, indicateur de la présence de NADPH oxydase de thyroïde dans ledit échantillon.
Les anticorps de l'invention peuvent donc être avantageusement mis en œuvre dans toute situation où l'expression de la NADPH oxydase de thyroïde doit être observée.
En particulier, la NADPH oxydase de thyroïde est un marqueur de la différenciation de thyrocytes.
L'identification de certains cancers thyroïdiens (par exemple papillaires, folliculaires, anaplasiques,...) ou autres tumeurs malignes citées plus haut, peut être facilitée par la possibilité de détecter, à l'aide de ces anticorps, l'expression (ou la non-expression) de ce marqueur cellulaire spécifique du thyrocyte.
Les exemples ci-dessous illustrent l'invention sans en limiter la portée.
EXEMPLES
I - Purification de l'enzyme
La purification d'une flavodeshydrogénase (FAD) appartenant à l'oxydase de thyroïde de porc a permis le microséquençage de deux peptides (de 8 et 16 acides aminés respectivement) ayant une séquence inconnue dans les banques de données :
Cette purification a pu être réalisée selon le protocole suivant : L'enzyme extraite de membranes de thyroïde de porc par 2M KCI en présence de 0,2% Triton X 100 (Serva, Heideiberg, Allemagne) est chromatographiée sur Octyl sepharose (support hydrophobe). Après un lavage de la colonne par un détergent (10 mM CHAPS, Sigma Chemical Co, St Louis, Colorado), l'enzyme est éluée par un tampon sans FAD contenant 0,2M KCI et 1 % Triton X 100. Cette étape permet une purification de 30 fois de la flavoproteine. L'enzyme est ensuite fixée en « batch » sur FAD agarose pendant une nuit à 4°C et éluée par 250 μl de tampon d'échantillon ( SDS- PAGE "sample buffer", Laemmli, 1970). La totalité du volume d'élution est déposé dans un puits unique d'un gel d'acrylamide à 8% (conditions de Laemmli). Les protéines sont révélées par coloration à l'amidoblack. La bande d'intérêt (180 kDa) est découpée du gel (3,5 μg de protéine) et séchée et incubée dans 50 mM de tampon Tris pH 6, 0,03 % SDS, 0,2 μg/ml d'endoprotéase Lys-C de Boehringer (une nuit à 35°C) pour être soumise à un microséquençage.
Le volume de surnageant a été réduit à 200 μl sous vide et injecté dans une précolonne Ax300 (2,1 x 30 mm) de Perkin-Elmer suivie d'une colonne HPLC en phase inverse 218TP52 Vydac (2,1 x 250 mm). Les peptides ont été élues avec un gradient linéaire de 2 à 35 % (vol/vol) de 0,1 % d'acide trifluoroacétique dans l'acétonitrile pendant 90 minutes à une vitesse de 200 μl/min. Les pics majeurs ont été séquences.
Les deux peptides séquences présentent les séquences suivantes : peptide A = AWPPPRLYTEALQEK et peptide B = X(V)QLIN(R/P)QDQ(T)HFVHHYEN(P), ce peptide B contenant huit acides aminés HFVHHYEN certains.
Il - Clonage de l'ADNc de porc
A partir du peptide de 8 acides aminés, une amorce de 30 nucléotides a été déterminée pour l'obtention de l'extrémités 3' de l'ADNc par la technique dite de RACE (Rapid Amplification of cDNA extremity). Cette technique permet d'amplifier des séquences d'acides nucléiques d'un ARN entre un site interne et une des extrémités 5' ou 3' inconnues. Pour cela les ARN ont été extraits à partir de thyrocytes de porc cultivés pendant 4 jours en présence de 10 μM de forskoline. Culture de thyrocytes de porc
Les thyrocytes porcins sont préparés selon un protocole décrit dans Haye B. et al, 1985. Les cellules sont isolées des glandes par trypsinisation et suspendues dans un milieu Eagle's MEM (pH 7,4) contenant 10% (vol/vol) de sérum de veau, de la pénicilline (200 U/ml) et de la streptomycine (50 μg/ml). Les cellules (2 x 106 cellules/ml) sont cultivées en suspension pendant 4 jours à 37°C avec 10 μM de forskoline. Les follicules reconstitués sont collectés par centrifugation à 200 x g pendant 7 min et congelés dans l'azote liquide avant extraction des ARNs. Préparation des ARN Les ARN totaux sont extraits des cellules par la technique du guanidium de thiocyanate (Chomczynski et coll., 1987).
RACE
L'ADNc complet est obtenu par 3' et 5' RACE selon le protocole donné par CLONTECH dans le kit "SMART RACE cDNA Amplification". Les différents produits d'amplification sont clones dans le vecteur pCR-XL-TOPO (Invitrogen).
III - Clonage de l'ADNc humain
Le clonage de l'ADNc humain est réalisé en utilisant également la technique du 3' et 5' RACE. Les réactions de PCR sont faites à partir d'ADNc de thyroïdes humaines (Marathon-Ready cDNA) commercialisés par CLONTECH. L'amorce déterminée à partir du peptide porcin de 8 acides aminés a permis d'obtenir l'extrémité 3' de l'ADNc humain. L'extrémité 5' est obtenue par 5' RACE en utilisant une amorce spécifique choisie dans l'extrémité 3' de la séquence humaine. Les produits d'amplification sont clones dans le plasmide pCR-XL-TOPO (Invitrogen) puis séquences.
IV - Clonaαe de l'ADNc de rat :
Le clonage de l'ADNc est réalisé en utilisant la technique RACE (kit SMART RACE de Clontech), à partir d'1 μg d'ARNm total de glande thyroïdienne de rat. Les produits de PCR ont été clones dans un vecteur pCR- XL-TOPO (Invitrogen) pour le séquençage.
La séquence SEQ ID n° 5 en annexe représente l'ADNc de 5689 paires de bases, contenant un cadre de lecture ouvert codant pour un polypeptide de 1 517 acides aminés (SEQ ID n° 6).
V- Expression tissulaire de la NADPH oxydase clonée
1 ) Analyse de l'expression tissulaire de l'enzyme humaine par Northern Blot :
L'ARNm de 5 kb correspondant à l'expression de l'enzyme humaine clonée n'a pu être détecté par Northern Blot que dans le tissu thyroïdien. Cette observation va dans le sens d'une participation de l'enzyme clonée à la fonction spécifique des thyrocytes, c'est-à-dire la biosynthèse des hormones thyroïdiennnes.
Les thyrocytes porcins cultivés sans forskoline n'expriment pas l'ARNm de 5 kb alors que son expression est maintenue, voire accentuée en présence d'AMP cyclique.
Ces résultats sont en accord avec ceux de Raspé et Dumont, 1995, qui rapportent que la TSH induit l'expression du générateur de H O2 des thyrocytes de chien via l'activation de la cascade de l'adénylate cyclase.
2) Analyse de l'expression tissulaire de l'enzyme de rat par RT- PCR : L'ARNm total (2 μg) a été traité avec la transcriptase inverse
Thermoscript 15 U (GIBCO BRL) dans 20 μl de tampon de PCR selon le protocole du fabricant pendant 60 minutes à 60°C. L'ADNc de rat a été amplifié pendant 30 cycles de température (95° C, 15 sec. ; 55°C, 20 sec; 72°C, 30 sec.) dans un thermocycleur GeneAmp 2400 (Perkin Elmer) dans 25 μl de tampon PCR préchauffé contenant 2,5 unités d'AmpliTaq Gold (Perkin Elmer), 250 nM de chaque amorce oligonucléotidique sens et anti-sens et 200 μM de chaque dNTP. Les amorces sens et anti-sens de PCR (GIBCO BRL) ont été construites à partir de la séquence d'ADNc de rat et étaient respectivement : 5'- CAAATCGTCCATGGGTACCC-3' (SEQ ID n° 7) et 5'- TCAACAGTTGTCAGAAATAG-3' (SEQ ID n° 8). Un produit d'amplification de 655 paires de bases a été recherché dans divers tissus de rat adulte. De fortes concentrations d'ARNm de la NADPH oxydase chez le rat ont été trouvées dans l'intestin et le colon, de plus faibles concentrations étant trouvées dans le duodénum.
VI - Localisation chromosomigue du gène de la NADPH oxydase de thyroïde
Les auteurs de la présente invention ont recherché par la technique FISH d'hybridation in situ fluorescente la localisation du gène de la p138tox de la NADPH oxydase de thyroïde dans le génome humain. A cette fin,
50 métaphases normales ont été analysées, à l'aide d'une sonde (vecteur pCR-XL-TOPO contenant le produit de 4 kb humain de la 5'-RACE), selon la
technique de Valent et al, 1996 et 1997. On a utilisé 40 ng de sonde marquée avec 120 ng de compétiteur (ADN de placenta humain) dénaturés.
Dans 40 des métaphases observées, un signal fluorescent a été détecté sur la bande chromosomique 15q15.

Claims
1 . Polypeptide isolé ayant une activité de p138tox de la NADPH oxydase de thyroïde, comprenant une séquence d'acides aminés choisie parmi les séquences SEQ ID n°2 ou n°4, ou toute séquence d'acides aminés homologue.
2. Acide nucléique isolé codant pour un polypeptide ayant une activité de p138tox de la NADPH oxydase de thyroïde selon la revendication 1.
3. Acide nucléique selon la revendication 2 comprenant une séquence nucléotidique choisie parmi la séquence SEQ ID n°1 ou n°3 ou toute séquence homologue desdites séquences.
4. Vecteur de clonage et/ou d'expression contenant une séquence nucléotidique selon l'une quelconque des revendications 2 ou 3.
5. Cellule hôte transformée par un vecteur selon la revendication

6. Anticorps mono- ou polyclonaux ou leurs fragments, anticorps chimériques ou immunoconjugués, caractérisés en ce qu'ils sont capables de reconnaître spécifiquement un polypeptide selon la revendication 1 .
7. Procédé de production d'un polypeptide recombinant à activité de p138tox de la NADPH oxydase de thyroïde, dans lequel on cultive des cellules transformées selon la revendication 5 dans des conditions permettant l'expression d'un polypeptide comprenant une séquence d'acides aminés choisie parmi la séquence SEQ ID n°2 ou n°4, ou toute séquence homologue et on récupère ledit polypeptide.

8. Sonde nucléotidique comprenant au moins 10 nucléotides, capable d'hybrider spécifiquement, dans des conditions stringentes, avec un acide nucléique selon l'une des revendications 2 ou 3.
9. Procédé de détection de l'expression de p138tox de la NADPH oxydase de thyroïde dans un échantillon cellulaire ou tissulaire, comprenant les étapes consistant à :
- préparer l'ARN dudit échantillon ;
- mettre en contact ledit ARN obtenu avec une sonde ayant une séquence nucléotidique capable de s'hybrider spécifiquement avec un acide nucléique tel que défini dans la revendication 2 ou la revendication 3 ;
- détecter la présence d'ARNm hybridant avec cette sonde, indicatrice de l'expression de p138to de la NADPH oxydase de thyroïde.
10. Procédé de détection de l'expression de p138tox de la NADPH oxydase de thyroïde dans des cellules ou un tissu par hybridation in situ, comprenant les étapes consistant à :
- mettre en contact lesdites cellules ou ledit tissu avec une sonde ayant une séquence nucléotidique capable de s'hybrider spécifiquement avec un acide nucléique tel que défini dans la revendication 2 ou la revendication 3 ; - détecter la présence d'ARNm hybridant avec cette sonde, indicatrice de l'expression de p138tox de la NADPH oxydase de thyroïde.

Procédé de détection et/ou de dosage immunologique de la NADPH oxydase de thyroïde dans un échantillon biologique dans lequel : i) on met en contact ledit échantillon biologique avec un anticorps tel que défini à la revendication 6, marqué de manière détectable ; ii) on observe la formation d'un complexe anticorps-NADPH oxydase de thyroïde, indicateur de la présence de NADPH oxydase de thyroïde dans ledit échantillon.
12. Composition thérapeutique comprenant au moins un acide nucléique comprenant une séquence antisens capable de s'hybrider
spécifiquement avec un acide nucléique selon la revendication 2 ou 3, en association avec un véhicule pharmaceutiquement acceptable.
13. Composition thérapeutique comprenant une séquence nucléotidique selon l'une quelconque des revendications 2 ou 3 éventuellement placée sous le contrôle d'un promoteur spécifique des cellules thyroïdiennes ou d'un promoteur spécifique de cellules cancéreuses, en association avec un véhicule pharmaceutiquement acceptable.
14. Composition pharmaceutique comprenant une quantité efficace d'au moins un polypeptide selon la revendication 1 , en association avec un véhicule pharmaceutiquement acceptable.
15. Composition pharmaceutique comprenant une quantité efficace d'au moins un polypeptide modifié par rapport au polypeptide selon la revendication 1 de telle sorte que ledit peptide mime la zone d'interaction avec la thyroperoxydase (TPO) avec pour conséquence l'inhibition de la NADPH oxydase et/ou l'inhibition de la TPO, en association avec un véhicule pharmaceutiquement acceptable.
16. Utilisation d'un polypeptide selon la revendication 1 pour le criblage d'inhibiteurs de la NADPH oxydase de la thyroïde, destinés à la fabrication d'un médicament pour la prévention et/ou le traitement d'affections impliquant une activité anormale de la NADPH oxydase de thyroïde.
17. Procédé de détection d'anticorps anti-NADPH oxydase dans un échantillon biologique, dans lequel on met en contact ledit échantillon biologique avec un polypeptide selon la revendication 1 , marqué de manière détectable et on détecte les complexes p138tox-anticorps.
18. Procédé de production d'anticorps dirigés contre la p138to de la NADPH oxydase de thyroïde dans lequel on immunise des animaux par
administration d'un vecteur d'expression contenant une séquence nucléotidique selon l'une quelconque des revendications 2 ou 3.
19. Animaux immunisés par administration d'un vecteur d'expression contenant une séquence nucléotidique selon l'une quelconque des revendication 2 ou 3 pour la production d'anticorps dirigés contre la NADPH oxydase.
