Abstract

L'invention concerne un procédé de culture cellulaire qui comprend une étape de prolifération dans laquelle le nombre de cellules précurseurs est multiplié, et une étape de différenciation dans laquelle les cellules précurseurs multipliées sont transformées en cellules neuronales. L'étape de prolifération consiste à incuber les cellules précurseurs dans un milieu de prolifération contenant un facteur de croissance de fibroblastes basique (FGF basique). L'étape de différenciation consiste à incuber les cellules précurseurs dans des milieux de différenciation de manière à former un agrégat cellulaire n'adhérant sur aucune surface de la cuve d'incubation. Dans une forme de réalisation préférée, les cellules sont incubées dans un tambour tournant. Les milieux de différenciation peuvent également comprendre au moins un agent de différenciation. L'invention concerne en outre un procédé de traitement d'un trouble neurologique, tel que la maladie de Parkinson, un procédé permettant de transplanter un produit génique dans le cerveau d'un patient, un dosage destiné à des substances ayant une activité neurologique, et une culture cellulaire.
