Abstract

L'invention concerne un procédé servant à déterminer le niveau d'expression d'un gène intéressant, ou d'un groupe de gènes intéressants, dans un échantillon biologique. Le procédé comporte la préparation d'une courbe normalisée pour chaque gène intéressant, mise en oeuvre par le dosage d'une série de dilutions préparées à partir d'ARN complet dans un test d'amplification par PCR de 5'-exonucléase à la transcriptase inverse. Un cycle de seuil (Ct) pour chaque élément de la série transcrite inverse - amplifiée est déterminé et tracé par opposition au relevé de la quantité d'ARN complet de la série de dilutions. Le tracé est utilisé pour déterminer un ARN équivalent à partir duquel on détermine l'ARN normalisé équivalent du gène intéressant de l'échantillon. Le dosage peut être utilisé pour déterminer l'effet d'un traitement de l'échantillon sur le niveau d'expression d'un gène intéressant ou d'un groupe de gènes intéressants.
