Verfahren zur Herstellung von Liposomen
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Liposomen. Ferner betrifft die vorliegende Erfindung, die kosmetische, medizinische oder pharmazeutische Nutzung solcher Strukturen.
Bestimmte, strukturell an sich durchaus nicht einheitliche Biomoleküle werden in der biochemischen Fachsprache unter dem Begriff „Lipide" zusammengefaßt. Im ursprünglichen Sinne sind unter „Lipiden" Fette zu verstehen (grch.: τo λiπoς = das Fett, Öl), also Carbonsäureester des Glycerins.
Im weiteren Sinne wird in diesen Begriff eine Gruppe von in Wasser unlöslichen Molekülen verstanden, welche sich durch wenigstens einen ausgeprägt hydrophilen Molekülbereich und wenigstens einen ausgeprägt lipophilen Molekülbereich auszeichnen. Die Phosphorsäureester acylierter Glycerine, die sogenannten „Phospholipide" und andere Verbindungen gehören zu dieser insgesamt recht inhomogenen Gruppe chemischer Verbindungen.
Aufgrund der strukturellen Gegebenheiten bilden Lipide in vitro, beispielsweise im Gemenge mit Wasser, in der Regel keine echten molekularen Lösungen, vielmehr bilden sie entweder Kolloide, schließen sie sich zumeist zu sogenannten Micellen zusammen, in welchen die lipophilen Molekülbereiche der Lipidmoleküle sich im Innern der Micelle befinden und die hydrophilen Bereiche der Lipidmoleküle den Außenbereich der Micellen darstellen, wie in Fig. 1 geschildert, oder aber sie bilden flüssigkristalline Phasen aus. Die Lipidmoleküle verfügen über einen hydrophilen Bereich (h) und einen lipophilen Bereich (I). Die in Fig. 1 dargestellte Micelle (M) stellt eine Kugel aus Lipidmolekülen dar, deren Inneres von den lipophilen Resten der
naheliegend, daß die Außenschale der Micelle von den hydrophilen Gruppen der Lipidmoleküle gebildet wird.
Von größter biologischer Bedeutung ist die Fähigkeit der Lipide, sich in den bekannten Lipiddoppelschichten anzuordnen. Lipidmembranen können beispielsweise linear, gekrümmt (kubische Phasen, LrPhasen) oder in sich geschlossen (Vesikel, LrPhasen) vorliegen.
In einer wäßrigen oder allgemein polaren Umgebung lagern sich die einzelnen Lipidmoleküle dergestalt aneinander, daß zwei monomolekulare Schichten von parallel angeordneten Lipidmolekülen an ihren lipophilen Bereichen aufeinander zu liegen kommen. Dies wird in Fig. 2 dargestellt. Die Lipidmoleküle verfügen über einen hydrophilen Bereich (h) und einen lipophilen Bereich (I). Die in Fig. 2 dargestellte Lipiddoppelschicht (L) stellt eine Membran aus Lipidmolekülen dar, deren Inneres von den lipophilen Resten der Lipidmoleküle ausgefüllt wird. Da die Lipiddoppelschicht in wäßriger Umgebung (W) vorliegt, ist naheliegend, daß die Außenschale der Lipiddoppelschicht von den hydrophilen Gruppen der Lipidmoleküle gebildet wird.
In Fig. 6 wird die Nahstruktur einer lamellaren Phase vereinfacht dargestellt. Die dargestellten Lipiddoppelschichten stellen Membranen aus Lipidmolekülen dar, die über einen hydrophilen Bereich (h) und einen lipophilen Bereich (I) verfügen. Das Innere der Membranen wird von den lipophilen Resten der Lipidmoleküle ausgefüllt. Durch „Stapelung" der Membranen entsteht die lamellare Struktur, wobei die interlamellaren Zwischenräume zwischen den Einzelmembranen mit Wasser gefüllt ist.
Obwohl sich, wie zuvor angesprochen, aufgrund der strukturellen Gegebenheiten zumeist zu sogenannten Micellen zusammenschließen, ist es unter bestimmten Bedingungen auch technisch möglich, Lipide zu Vesikeln oder Liposomen zu verarbeiten.
Vesikeln oder Liposomen sind kugelförmig geschlossene mikroskopische Objekte, welche nach außen durch eine Lipiddoppelschicht begrenzt werden und in ihrem Innern eine Wasserphase beherbergen. Dies wird in Fig. 3 dargestellt. Die Lipidmoleküle verfügen über einen hydrophilen Bereich (h) und einen lipophilen Bereich (I).
Die in Fig. 3 dargestellte Lipiddoppelschicht (L) der dargestellten Vesikel, wie in der Ausschnittsvergrößerung besser ersichtlich ist, stellt eine hohlkugelförmige Membran aus Lipidmolekülen dar, deren Inneres von den lipophilen Resten der Lipidmoleküle ausgefüllt wird. Da die Lipiddoppelschicht in wäßriger Umgebung (W) vorliegt, ist naheliegend, daß die Außenschale der Lipiddoppelschicht von den hydrophilen Gruppen der Lipidmoleküle gebildet wird. Das Zentrum der Vesikel wird von einer wäßrigen Phase ausgefüllt. Der Durchmesser von Liposomen bewegt sich zumeist in der Größenordnung von einigen (typischerweise etwa 25) Nanometem bis zu etwa 1μm.
In Fig. 4 wird eine sogenannte multilamellare Vesikel (M) im Querschnitt und mit Ausschnittsvergrößerungen gezeigt. Sie besteht aus mehreren - hier zwei - Lipiddoppelmembranen als äußere Hülle, wobei die Lipiddoppelmembranen aus Lipidmolekülen mit einem hydrophilen Bereich (h) und einen lipophilen Bereich (I) bestehen. Zwischen den Lipiddoppelmembranen ist in der Regel eine dünne Schicht einer polaren, insbesondere wäßrigen Phase (w) angeordnet. Im Innern der multilamellaren Vesikel ist eine weitere polare, meist wäßrige Phase, welche auch mit Wirkstoffen (hier: Q)) beladen sein kann. Auch für multilamellare Vesikeln ist zutreffend, daß auch die Lipiddoppelmembran mit Wirkstoffen beladen sein kann, welche dann in der Regel lipophiler Natur sind.
Die Herstellung von Liposomen beispielsweise ist dem Fachmanne durchaus geläufig. Eine Methode besteht beispielsweise in einer Ultraschallbehandlung eines Lipid-Wassersystems, wobei bei der Wahl geeigneter Lipide die erhaltenden Liposomen lediglich abfiltriert zu werden brauchen. Aber auch die Vesikelextrusion entsprechend geeigneter Lipide durch einen Membranfilter einer Porengröße von typischerweise ca. 100 nm führt zur Bildung von Liposomen.
Normalerweise werden die Grundbestandteile der die Liposomen umhüllenden Lipiddoppelmembran gewählt aus der Gruppe der Phospholipide, oft Lecithin. Gelegentlich, zumal bei den sogenannten Niosomen, werden auch nichtionische Tenside als Hüllmaterial eingesetzt.
Im allgemeinen unterscheidet man zwischen „leeren" Liposomen, welche aus einer Hülle und einem wäßrigen Innern bestehen, und „beladenen" Liposomen, deren Inneres eine wäßrige Phase darstellt, welche mit wasserlöslichen Wirkstoffen versehen ist, oder deren Hülle mit lipophilen Wirkstoffen versehen ist. Mikroorganismen können liposomal gleichsam verkapselt werden, „Transfersomen" sollen, im Gegensatze zu „normalen" Liposomen intakt in die Epidermis eindringen. Im allgemeinen allerdings penetrieren Liposomen eben nicht intakt, sondern adsorbieren an oder in den obersten Schichten des Stratum co eum. Vieles spricht dafür, daß Liposomen endozytotisch in die Körperzelle aufgenommen werden. Möglich ist neben der topischen Verabreichung von liposomenhaltigen Zubereitungen auch die intravenöse Gabe.
Von den Lipiden sind insbesondere die Phospholipide von höchstem biologischem Interesse, da diese die Grundsubstanz der Zellmembranen aller lebender Zellen bzw. deren Zellorganellen darstellen.
Weit über tausend Lipide von Zellmembranen wurden bislang isoliert und identifiziert. Den weitaus größten Anteil nehmen die Phospholipide mit etwa 40 bis über 90 % des Gesamtlipidanteils der Zelle ein. Dabei sind fünf Typen von Phospholipiden dominierend: Phosphatidylcholin, Phosphatidylethanolamin, Phosphatidylserin, Car- diolipin (Diphosphatidylglycerin) und Sphingomyelin. Glycolipide sind wichtige Bestandteile der Plasmamembran, des Myelins, das endoplasmatischen Reticulums und bestimmter Organelle, beispielsweise der Chloroplasten Photosynthese treibender Organismen.
Von größter Bedeutung unter den Phosphatidylcholinen beispielsweise sind die Leci- thine, welche sich durch die allgemeine Struktur
auszeichnen, wobei Ri und R
2 typischerweise unverzweigte aliphatische Reste mit 15 oder 17 Kohlenstoffatomen und bis zu 4 cis-Doppelbindungen darstellen. Lecithine sind nicht nur in der belebten Zelle von Bedeutung, sie werden auch bevorzugt als Grundbestandteil für die äußere Schicht der gängigsten handelsüblichen Liposomen verwendet.
Den Sphingolipiden liegt als Grundstruktur das Sphingosin oder auch das Phy- tosphingosin zugrunde, welche sich durch folgende Strukturformeln auszeichnen:

3 unabhängig voneinander gesättigte oder ungesättigte, verzweigte oder unverzweigte Alkylreste von 1 bis 28 Kohlenstoffatomen darstellen, R
2 gewählt wird aus der Gruppe: Wasserstoff atom, gesättigte oder ungesättigte, verzweigte oder unverzweigte Alkylreste von 1 bis 28 Kohlenstoffatomen, Zuckerreste, mit organischen Resten veresterte oder unveresterte Phosphatgruppen, mit organischen Resten veresterte oder unveresterte Sulfatgruppen und Y entweder ein Was-
serstoffatom, eine Hydroxygruppe oder einen anderen hetero-funktionellen Rest darstellt.
Zu den häufigsten Sphingolipiden gehören die natürlich vorkommenden Ceramide, Cerebroside, Ganglioside, Sphingophospholipide, von diesen insbesondere die Sphingomyeline, Sphingosulfatide und Glycosphingoside sowie durch chemische Synthese erhältliche Analoga, von welchen nachfolgend Beispiele aufgeführt werden:
Ceramide:
Ri und R3 stellen Alkylreste dar, R2 = H.
Sphingophospholipide:
Ri und R3 stellen Alkylreste dar, R4 stellt einen Organylrest dar.
Sphingomyeline sind organylphosphorylierte Sphingolipide des Typs
Wird R2 in der Strukturformel der Ceramide gewählt aus der Gruppe der Zuckerreste, wird üblicherweise unterschieden, ob Monoglycosylceramide oder Di,- Tri bzw. allgemein Oligoglycosylceramide vorliegen. Monoglycosylceramide werden gewöhnlich Cerebroside genannt:
Oligoglycosylceramide werden meistens Ganglioside genannt.
Häufig auftretende Sphingolipide sind Ceramid I, II, III und IV, Glucosylceramid, Lac- tosylceramid sowie die Ganglioside GM 1 , 2 und 3.
Klassische Herstellungsverfahren für niedrigviskose Liposomen-Dispersionen sind das:
(1) Hydratationsverfahren:
Ein Gemisch aus Phospholipiden wird in Wasser dispergiert und anschließend in einem Glaskolben eingedampft. Es bildet sich ein Lipidfilm an der Wandung, der mit einer Pufferlösung befeuchtet wird, wobei sich spontan Liposomen bilden. Es entstehen meist multilamellare Vesikel.
(2) Beschallung mit Ultraschall:
Eine in Wasser dispergierte Phospholipidmischung wird durch Ultraschall zerkleinert, wobei Liposomen entstehen.
(3) Das „French-press"-Verfahren:
Eine in Wasser dispergierte Phospholipidmischung wird durch Druckbelastung in einem Extruder in Liposomen überführt.
(4) Die Lösungsmittel-Injektionsmethode:
Die Lipide, insbesondere Phospholipide, werden in einem organischen Lösungsmittel wie Ether, Methanol gelöst und in warmes Wasser eingespritzt, in dem der einzuschließende Stoff gelöst vorliegt. Nach Abzug des Lösungsmittels im Vakuum bilden sich meist unilamellare Vesikel aus.
(5) Die Detergentien-Methode:
Eine wäßrige Mischmizellösung aus Phospholipiden, einem Detergens und der zu verkapselnden Substanz wird hergestellt und anschließend entfernt man das Detergens durch Dialyse oder Säulenchromatographie.
(6) Die „Reverse Phase Evaporation":
Im Überschuß der organischen Phase wird in einem Puffer, den Phospholipiden und den einzuschließenden Substanzen eine Wasser-in-ÖI Emulsion hergestellt und dann die flüchtige organische Phase im Vakuum abgedampft. Am Ende des Verdampfungsvorgangs entsteht eine Suspension aus großen unilamellaren Vesikeln.
Bestimmte Lipidmischungen können, ohne die vorab beschriebenen Verfahren auszunutzen, Liposomen-Dispersionen ergeben. Diese sogenannte „Spontaneous Formation of Vesicles" wird durch Auswahl der Lipide, pH-Wert-Änderungen (Proc. Natl. Acad. Sei. USA, Vol 79, S. 1683, 1982) Verdünnungen aus ethanolhaltigen Fettalkohol- und Fettsäuremischungen (Biochemistry, Vol 17,No.18, S.3758, 1978) oder aus ethanolhaltigen Phospholipidlösungen (J. Pharm. Pharmacol. 43, S. 154, 1991) induziert. Ferner können auch Abmischungen aus Phosphatidylcholin und Lyso- phosphatidylcholin (Chemistry and Physics of Lipids, 43, S. 283, 1987) und Verdünnung von Mischungen aus langkettigen Phospolipiden mit einem kurzkettigem Phos- pholipid (Deheptanoyl-Phosphatidylcholin) spontan zu Liposomendispersionen führen. In der Literatur werden weitere Mischungen beschrieben, die spontan Vesikel bilden, zum Beispiel Mischungen aus Cholesterin und Seife (Biochemistry, Vol 17,No.18, S.3758, 1978), Seifen-Fettsäure-Mischungen (Chem. Phys. Lipids, 16, S.142, 1976), Mischungen aus Alkyltrimethylammoniumhydroxiden (J. Phys. Chem. 87, S. 5020, 1983), Mischungen aus Alkyltrimethylammoniumtosylaten und anionischen Cotensiden wie Natriumdodecylbenzolsulfonat, C8-Cι2-Alkylsulfaten (J. Phys. Chem. 96, S. 6698, 1992).
Ferner werden in EP 0 211 647 Liposomenzubereitungen offenbart, die durch Mischung einer C8-C24-Carbonsäure (oder einem Amin) mit einer Verbindung, die zwei ionisierbare Gruppen enthält (Dicarbonsäure, Diamin, α-Amino-ω-Carbonsäure), einem zu verkapselndes Material und Wasser entstehen. Es wird offenbart, daß zur Herstellung ein Proliposomengel erzeugt wird, das anschließend mit Wasser ver-
dünnt wird. Das Konzentrat besteht aus dem liposomenbildenen Ingredientien und der Verbindung, die zwei ionisierbare Gruppen enthält. Das Gel besteht aus einem hydratisiertem Komplex, der als Flüssigkristall vorliegt. Durch Verdünnen in eine Phosphat-Pufferlösung wird im nächsten Schritt die liposomale Dispersion aus dem Gel erhalten. Das Verfahren zur Herstellung dieser Liposomendispersion benötigt also zwei Schritte. Im ersten Schritt stellt man unter Ausschluß von Wasser aus Phospolipiden und Ölsäure eine Paste her, die anschließend mit einer vorher in Wasser aufzulösenden zweiten Verbindung, wie Arginin, zum Gel gemischt wird, das dann mit Wasser spontan Liposomen bildet.
In EP 707847 wird ein Verfahren vorgestellt, das es gestattet, aus Ketoprofen und Phospholipiden oder nichtionischen Amphiphilen spontan Liposomen herzustellen. Dabei wird zunächst der pH-Wert der Mischung auf 6-8 eingestellt, und durch Absenken des pH-Werts unter pH = 6 erhält man spontan Liposomen.
In DE 69214552 wird ein Verfahren zur Herstellung von Vesikeln für Haarpflegezubereitungen beschrieben, daß mindestens ein Lipid (amphiphil, ionisch), ein Silikon und Wasser enthält. Das beschriebene Verfahren zur Herstellung ist aufwendig, da es mehrere Schritte benötigt. Ferner wird ein Hochdruck-homogensisator benutzt. Auch die in DE 69600008 beschriebenen Lipidvesikel werden durch Ultraschall oder Hochdruckhomogenisierung erzeugt .
Aus Phospholiden lassen sich mit geeigneten Cotensiden flüssigkristalline Zubereitungen herstellen (lamellare Phasen zum Beispiel). Die lamellare Phase entsteht, wenn man geeignete Lipide mit Wasser mischt. Das Wasseraufnahmevermögen für Flüssigkristalle ist begrenzt, da durch Zusatz großer Mengen an Wasser die Abstände der Wasserkanäle und damit auch die Abstände der Lipiddoppelschichten so groß werden, daß der Flüssigkristall instabil wird.
All diese Verfahren des Standes der Technik zeichnen sich jedenfalls durch teilweise erhebliche Nachteile aus, sei es, daß sie teuer sind, sei es, daß sie unvorhersehbare Resultate zeitigen. Aufgabe der vorliegenden Erfindung war es also, diesen Nachteilen abzuhelfen.
Erstaunlicherweise werden die Nachteile des Standes der Technik behoben durch ein
Verfahren zur Herstellung von Liposomen bzw. von Dispersionen von Liposomen in einer wäßrigen Phase, dadurch gekennzeichnet, daß
(I) (a) ein Lipid oder ein Lipidgemisch,
(b) gegebenenfalls eine Ölphase und
(c) eine wäßrige Phase
(d) gegebenenfalls ein oder mehrere Tenside zusammengegeben und bei einer Temperatur homogenisiert werden, welche oberhalb der Schmelztemperatur von Lipid bzw. Lipidgemisch und der Ölphase - falls eine solche verwendet wird - liegt, und wobei
(II) die Gesamtheit der homogenisierten Komponenten (a), (b), (c) und (d) in einem Temperaturintervall [θ] eine lamellare Phase bildet,
(III) die Temperatur des homogenisierten Gemisches aus (a), (b), (c) und (d) auf einen Wert innerhalb des Temperaturintervalles [θ] (Herstellungstemperatur θ^ oder auf einen Wert oberhalb des Temperaturintervalles [θ] (Herstellungstemperatur θ2) gebracht wird,
(IV) zu dem homogenisierten Gemisch aus (a), (b), (c) und (d) Wasser zugegeben und die sich bildenden Liposomen bzw. die sich bildende Liposomendispersion auf eine Temperatur θ3 gebracht wird, welche unterhalb des Temperaturintervalles [θ] befindlich ist.
Es kann dabei durchaus vorteilhaft sein, wenn die Herstellungstemperatur t oberhalb von [θ] liegt, da oftmals oberhalb der -Phase (=lamellare Phase) eine sogenannte L2-Phase vorliegt, die aus invertierten Micellen besteht. Kühlt man ab, erhält man wieder die U-Phase, und dann schließlich Liposomen Allerdings können oberhalb der Lα-Phase auch invers hexagonale Phasen und/oder invers micellare Phasen und/oder kubische Phasen vorliegen, femer emulsionsähnliche Zustände.
Insbesondere dann, wenn - an sich fakultativ vorgesehene, aber erfindungsgemäß äußerst vorteilhaft zu verwendende - Ölkomponenten anwesend sind, können bei Herstellungstemperaturen θ2 oberhalb des Temperaturintervalles [θ] emulsionsähnii-
ehe Zustände, beispielsweise auch echte W/O-Emulsionen, entstehen, die, bei Passieren des Temperaturintervalles [θ] über den Zustand der flüssigkristallinen lamella- ren Phase durch Verdünnen und Abkühlen mit Wasser Liposomen liefern. Die Liposomen sind in diesem Fall auch multilamellar.
In Fig. 5 ist ein vereinfachtes Phasendiagramm für ein Dreikomponentensystem aus Wasser, Tensid und Ölphase beispielhaft aufgeführt. Dabei bedeuten die Symbole H eine hexagonale Phase, C eine kubische Phase, L eine lamellare Phase und ,H eine invers-hexagonale Phase.
In Fig. 7 ist vereinfacht dargestellt, welcher Art Phasenübergänge bei Variation der Parameter Temperatur (θ) und Konzentration eines Bestandteiles (z.B. der Lipide) erfindungsgemäß verlaufen können.
Aus dem Zustande Ki (entsprechend einer Temperatur θi) im Phasengebiet Lα, in dem lamellare Phasen vorliegen, kann durch Absenken der Temperatur der Zustand K2 (entsprechend einer Temperatur θ3) im Bereich Lω erreicht werden, in welchem sich Liposomen bilden.
Aus dem Zustande K3 (entsprechend einer Temperatur θ2) im Phasengebiet L2, in dem (hier als Beispiel) invertierte Micellen vorliegen, kann durch Absenken der Temperatur der Zustand l^ (entsprechend einer Temperatur θ3) im Bereich Lω erreicht werden, in welchem sich Liposomen bilden.
Aus dem Zustande K5 (entsprechend einer Temperatur Θ1) im Phasengebiet Lα, in dem lamellare Phasen vorliegen, kann durch Erhöhen der Konzentration P eines Bestandteiles (hier als Beispiel Wasser) der Zustand Ke (entsprechend einer Temperatur θ3) im Bereich erreicht werden, in welchem sich Liposomen bilden.
Aus dem Zustande K7 (entsprechend einer Temperatur θ2) im Phasengebiet L2, in dem (hier als Beispiel) invertierte Micellen vorliegen, kann durch Erhöhen der Konzentration P eines Bestandteiles (hier als Beispiel Wasser) der Zustand Ks
(entsprechend einer Temperatur θ3) im Bereich U, erreicht werden, in welchem sich Liposomen bilden.
Ferner kann man auch über Temperaturerhöhung oder Konzentrationsänderung von K2 zu K1 t K_i zu K3, K_. zu K5 in den Bereich der lamellaren Phase bzw in den Bereich oberhalb der lamellare Phase gehen und dann wieder abkühlen und erfindungsgemäße Liposomen erhalten.
Erfindungsgemäß besonders vorteilhaft werden Phospholipide, namentlich solche, wie eingangs zum Hintergrund der Erfindung geschildert, als Lipide (a) eingesetzt. Die Grundstrukturen auf der Grundlage von Lipiddoppelmembranen können vorteilhaft auf allen gangen Lipiden natürlichen, synthetischen oder teilsynthetischen Ursprungs gewählt werden. Insbesondere vorteilhaft sind die Phospholipide wie etwa Phosphatidylcholine, Phosphatidylethanolamine, Phosphatidylserine, Cardiolipine (Diphosphatidylglycerine) und Sphingomyeline, ferner Glycolipide oder Glycerolipide.
Insbesondere von Bedeutung sind die Lecithine, Sphingolipide wie etwa Sphingosin oder Phytosphingosin, Ceramide, Cerebroside, Ganglioside, Sphingophospholipide, von diesen insbesondere die Sphingomyeline, Sphingosulfatide und Glycosphingoside sowie durch chemische Synthese erhältliche Analoga.
Beispiele für erfindungsgemäß verwendbare Lipide sind beispielsweise Dimethyldi- octadecylammoniumchlorid, Dimethyldioctadecylammoniumbromid, 1-Palmitoleyl-2- oleyl-sn-glycero-3-phosphatidylcholin, Dimyristoleylphosphatidylcholin, 1-Palmitoleyl- 2-oleyl-3-phosphatidylglyerin.
Der Verfahrensschritt (IV), kann mit Wasser einer Temperatur vorgenommen werden, die innerhalb, oberhalb oder unterhalb des Temperaturintervalles [θ] liegt, besonders vorteilhaft mit Wasser einer Temperatur von höchstens 35° C.
In Abhängigkeit von der Wahl der Komponenten, insbesondere der Lipide und, ge- wünschtenfalls, der anwesenden Tenside können die lamellaren Phasen bei Raumtemperatur auftreten oder auch nur bei höherer Temperatur oder über größere Temperaturbereiche. Die Herstellung der Liposomen kann also in Abhängigkeit vom ge-
wählten Tensidsystem sowohl bei Raumtemperatur als auch bei höherer Temperatur (z.B. 85°C) erfolgen und bei Verdünnung mit Wasser zu erfindungsgemäßen Liposomen führen.
Tenside sind amphiphile Stoffe, die organische, unpolare Substanzen in Wasser lösen können. Sie sorgen, bedingt durch ihren spezifischen Molekülaufbau mit mindestens einem hydrophilen und einem hydrophoben Molekülteil, für eine Herabsetzung der Oberflächenspannung des Wassers, die Benetzung der Haut, die Erleichterung der Schmutzentfernung und -lösung, ein leichtes Abspülen und -je nach Wunsch - für Schaumregulierung.
Bei den hydrophilen Anteilen eines Tensidmoleküls handelt es sich meist um polare funktionelle Gruppen, beispielweise -COO', -OSO3 2', -SO3 ", während die hydrophoben Teile in der Regel unpolare Kohlenwasserstoffreste darstellen. Tenside werden im allgemeinen nach Art und Ladung des hydrophilen Molekülteils klassifiziert. Hierbei können vier Gruppen unterschieden werden:
• anionische Tenside,
• kationische Tenside,
• amphotere Tenside und
• nichtionische Tenside.
Anionische Tenside weisen als funktioneile Gruppen in der Regel Carboxylat-, Sulfat- oder Sulfonatgruppen auf. In wäßriger Lösung bilden sie im sauren oder neutralen Milieu negativ geladene organische Ionen. Kationische Tenside sind beinahe ausschließlich durch das Vorhandensein einer quarternären Ammoniumgruppe gekennzeichnet. In wäßriger Lösung bilden sie im sauren oder neutralen Milieu positiv geladene organische Ionen. Amphotere Tenside enthalten sowohl anionische als auch kationische Gruppen und verhalten sich demnach in wäßriger Lösung je nach pH-Wert wie anionische oder kationische Tenside. Im stark sauren Milieu besitzen sie eine positive und im alkalischen Milieu eine negative Ladung. Im neutralen pH-Bereich hingegen sind sie zwitterionisch, wie das folgende Beispiel verdeutlichen soll:

Typisch für nicht-ionische Tenside sind Polyether-Ketten. Nicht-ionische Tenside bilden in wäßrigem Medium keine Ionen.
A. Anionische Tenside

Vorteilhaft zu verwendende kationische Tenside sind

4. Quaternäre Tenside.
5. Esterquats
Quaternäre Tenside enthalten mindestens ein N-Atom, das mit 4 Alkyl- oder Aryl- gruppen kovalent verbunden ist. Dies führt, unabhängig vom pH Wert, zu einer positiven Ladung. Vorteilhaft sind, Alkylbetain, Alkylamidopropylbetain und Alkyl- amidopropylhydroxysulfain. Die erfindungsgemäß verwendeten kationischen Tenside können ferner bevorzugt gewählt werden aus der Gruppe der quaternären Ammoniumverbindungen, insbesondere Benzyltrialkylammoniumchloride oder -bromide, wie beispielsweise Benzyldimethylstearylammoniumchlorid, ferner Alkyl- trialkylammoniumsalze, beispielsweise beispielsweise Cetyltrimethylammoniumchlo- rid oder -bromid, Alkyldimethylhydroxyethylammoniumchloride oder -bromide, Dial- kyldimethylammoniumchloride oder -bromide, Alkylamidethyltrimethylammonium- ethersulfate, Alkylpyridiniumsalze, beispielsweise Lauryl- oder Cetylpyrimidiniumchlo- rid, Imidazolinderivate und Verbindungen mit kationischem Charakter wie Aminoxide,
beispielsweise Alkyldimethylaminoxide oder Alkylaminoethyldimethylaminoxide. Vorteilhaft sind insbesondere Cetyltrimethylammoniumsalze zu verwenden.
C. Amphotere Tenside
Vorteilhaft zu verwendende amphotere Tenside sind
1. Acyl-/dialkylethylendiamin, beispielsweise Natriumacylamphoacetat, Dinatrium- acylamphodipropionat, Dinatriumalkylamphodiacetat, Natriumacylamphohydro- xypropylsulfonat, Dinatriumacylamphodiacetat und Natriumacylam- phopropionat,
2. N-Alkylaminosäuren, beispielsweise Aminopropylalkylglutamid, Alkylamino- propionsäure, Natriumalkylimidodipropionat und Lauroamphocarboxyglycinat.
D. Nicht-ionische Tenside
Vorteilhaft zu verwendende nicht-ionische Tenside sind
1. Alkohole,
2. Alkanolamide, wie Cocamide MEA/ DEA/ MIPA,
3. Aminoxide, wie Cocoamidopropylaminoxid,
4. Ester, die durch Veresterung von Carbonsäuren mit Ethylenoxid, Glycerin, Sor- bitan oder anderen Alkoholen entstehen,
5. Ether, beispielsweise ethoxylierte/propoxylierte Alkohole, ethoxylierte/ propo- xylierte Ester, ethoxylierte/ propoxylierte Glycerinester, ethoxylierte/ propoxy- lierte Cholesterine, ethoxylierte/ propoxylierte Triglyceridester, ethoxyliertes propoxyliertes Lanolin, ethoxylierte/ propoxylierte Polysiloxane, propoxylierte POE-Ether und Alkylpolyglycoside wie Laurylglucosid, Decylglycosid und Cocoglycosid.
6. Sucroseester, -Ether
7 Polyglycerinester, Diglycerinester, Monoglycerinester

Vorteilhaft ist ferner die Verwendung einer Kombination von anionischen und/oder amphoteren Tensiden mit einem oder mehreren nicht-ionischen Tensiden.
Besonders überraschend ist, daß die Liposomengröße steuerbar ist über das relative Verhältnis von Tensiden (zum Beispiel anionischem Tensid) zu Phospholipid, d.h. je mehr (z.B. anionisches) Tensid enthalten ist, desto kleiner ist das Liposom insgesamt, geringere Anteile an (z.B. anionischen) Tensiden führen automatisch bei gleichem Gesamtgehalt an Phospholipid und (z.B. anionischen) Tensiden zu größeren Lipiden. Es besteht ein linearer Zusammenhang zwischen Tensidkon- zentration und Liposomenradius, so daß einfach auszurechnen ist, wie groß die Liposomen werden sollen.
Ferner wurde überraschend gefunden, daß sich die Liposomengröße auch bei nichtionischen (z.B Glyceryl Stearat Citrat) oder kationischen Tensiden (z.B. Cetyl- trimethylammoniumbromid) in analoger Weise steuern läßt.
Es ist demnach erfindungsgemäß möglich, mit denselben Inhaltsstoffen nur durch relative Veränderungen der grenzflächenaktiven Inhaltsstoffe transparente, translu- cente oder weiße Vesikellösungen herzustellen.
Ferner wurde überrraschend gefunden, daß auch Glycerinester (z.B Glycerin- monocaprylat, -caprinat, laurat) oder Glycerinether (2-Ethylhexylglycerinether, in Gegenwart geeigneter Cotenside (z. B. aus der Gruppe der Acyl-Lactylate) zu Vesikel- Dispersionen führen.
Erfindungsgemäß kann die Größe der Vesikel gesteuert werden durch
a) die Konzentration des zugesetzten Cotensids/Coemulgators (anionisch/nicht- ionisch/amphoter) b) Gesamtkonzentration der Bilayer bildenen Lipide c) grenzflächenaktive Wirkstoffe als Cotensid bzw. Coemulgator (z. B Ascorbylpalmitat) d) Konzentration und Art der zu verkapselnden Ölko ponente e) Einsatz krümmungsinduzierender Polymere (u.a. hydrophob modifizierte Polymere)
Weiterhin können die Vesikel auch noch weiter verdünnt werden, ohne daß sie zerfallen (z.B. In Micellen).
Die Ölphase kann vorteilhaft gewählt werden aus folgender Substanzgruppe: Mineralöle, Mineralwachse
Öle, wie Triglyceride der Caprin- oder der Caprylsäure, vorzugsweise aber Rizinusöl;
Fette, Wachse und andere natürliche und synthetische Fettkörper, vorzugsweise Ester von Fettsäuren mit Alkoholen niedriger C-Zahl, z.B. mit Isopro- panol, Propylenglykol oder Glycerin, oder Ester von Fettalkoholen mit Alkansäuren niedriger C-Zahl oder mit Fettsäuren; Alkylbenzoate;
Silikonöle wie Dimethylpolysiloxane, Diethylpolysiloxane, Diphenylpoly- siloxane sowie Mischformen daraus.
Die Ölphase wird vorteilhaft gewählt aus der Gruppe der Ester aus gesättigten und/oder ungesättigten, verzweigten und/oder unverzweigten Alkancarbonsäuren einer Kettenlänge von 3 bis 30 C-Atomen und gesättigten und/oder ungesättigten, verzweigten und/oder unverzweigten Alkoholen einer Kettenlänge von 3 bis 30 C-Atomen, aus der Gruppe der Ester aus aromatischen Carbonsäuren und gesättigten und/oder ungesättigten, verzweigten und/oder unverzweigten Alkoholen einer Kettenlänge von 3 bis 30 C-Atomen. Solche Esteröle können dann vorteilhaft gewählt werden aus der Gruppe Isopropylmyristat, Isopropylpalmitat, Isopropylstearat, Isopro- pyloleat, n-Butylstearat, n-Hexyllaurat, n-Decyloleat, Isooctylstearat, Isononylstearat, Isononylisononanoat, 2-Ethylhexylpalmitat, 2-Ethylhexyllaurat, 2-Hexyldecylstearat, 2-Octyldodecylpalmitat, Oleyloleat, Oleylerucat, Erucyloleat, Erucylerucat sowie synthetische, halbsynthetische und natürliche Gemische solcher Ester, z.B. Jojobaöl.
Ferner kann die Ölphase vorteilhaft gewählt werden aus der Gruppe der verzweigten und unverzweigten Kohlenwasserstoffe und -wachse, der Silkonöle, der Dialkylether, der Gruppe der gesättigten oder ungesättigten, verzweigten oder unverzweigten Alkohole, sowie der Fettsäuretrigiyceride, namentlich der Triglycerinester gesättigter und/oder ungesättigter, verzweigter und/oder unverzweigter Alkancarbonsäuren einer Kettenlänge von 8 bis 24, insbesondere 12 - 18 C-Atomen. Die Fettsäuretriglyce-
ride können beispielsweise vorteilhaft gewählt werden aus der Gruppe der synthetischen, halbsynthetischen und natürlichen Öle, z.B. Olivenöl, Sonnenblumenöl, Sojaöl, Erdnußöl, Rapsöl, Mandelöl, Palmöl, Kokosöl, Palmkernöl und dergleichen mehr.
Auch beliebige Abmischungen solcher Öl- und Wachskomponenten sind vorteilhaft im Sinne der vorliegenden Erfindung einzusetzen. Es kann auch gegebenenfalls vorteilhaft sein, Wachse, beispielsweise Cetylpalmitat, als alleinige Lipidkomponente der Ölphase einzusetzen.
Vorteilhaft wird die Ölphase gewählt aus der Gruppe 2-Ethylhexylisostearat, Octyldo- decanol, Isotridecylisononanoat, Isoeicosan, 2-Ethylhexylcocoat, Cι2.i5-Alkylbenzoat, Capryl-Caprinsäure-triglycerid, Dicaprylylether.
Besonders vorteilhaft sind Mischungen aus C12.15-Alkylbenzoat und 2-Ethylhexyliso- stearat, Mischungen aus Cι2.15-Alkylbenzoat und Isotridecylisononanoat sowie Mischungen aus Cι2.15-Alkylbenzoat, 2-Ethylhexylisostearat und Isotridecylisononanoat.
Von den Kohlenwasserstoffen sind Paraffinöl, Squalan und Squalen, hydrogeniertes Polyisobuten vorteilhaft im Sinne der vorliegenden Erfindung zu verwenden.
Vorteilhaft kann die Ölphase ferner einen Gehalt an cyclischen oder linearen Silikonölen aufweisen oder vollständig aus solchen Ölen bestehen, wobei allerdings bevorzugt wird, außer dem Silikonöl oder den Silikonölen einen zusätzlichen Gehalt an anderen Ölphasenkomponenten zu verwenden.
Vorteilhaft wird Cyclomethicon (Octamethylcyclotetrasiloxan) als erfindungsgemäß zu verwendendes Silikonöl eingesetzt. Aber auch andere Silikonöle sind vorteilhaft im Sinne der vorliegenden Erfindung zu verwenden, beispielsweise Hexamethylcy- clotrisiloxan, Polydimethylsiloxan, Poly(methylphenylsiloxan).
Besonders vorteilhaft sind ferner Mischungen aus Cyclomethicon und Isotridecylisononanoat, aus Cyclomethicon und 2-Ethylhexylisostearat.
Die wäßrige Phase der erfindungsgemäßen Zubereitungen enthält gegebenenfalls vorteilhaft
Alkohole, Diole oder Polyole niedriger C-Zahl, sowie deren Ether, vorzugsweise Ethanol, Isopropanol, Propylenglykol, Glycerin, Ethylenglykol, Ethy- ienglykolmonoethyl- oder -monobutylether, Propylenglykolmonomethyl, -mo- noethyl- oder -monobutylether, Diethylenglykolmonomethyl- oder -monoethyl- ether und analoge Produkte, ferner Alkohole niedriger C-Zahl, z.B. Ethanol, Isopropanol, 1 ,2-Propandiol, Glycerin sowie insbesondere ein oder mehrere Verdickungsmittei, welches oder welche vorteilhaft gewählt werden können aus der Gruppe Siliciumdioxid, Aluminiumsilikate, Polysaccharide bzw. deren Derivate, z.B. Hyaluronsäure, Xanthangummi, Hydroxypropylmethylcellulose, besonders vorteilhaft aus der Gruppe der Polyacrylate, bevorzugt ein Polyacrylat aus der Gruppe der sogenannten Carbopole, beispielsweise Car- bopole der Typen 980, 981 , 1382, 2984, 5984, jeweils einzeln oder in Kombination.
Erfindungsgemäß können Wirkstoffe sehr vorteilhaft gewählt werden aus der Gruppe der Antioxidantien. Es ist dabei vorteilhaft, Antioxidantien als einzige Wirkstoffklasse zu verwenden, etwa dann, wenn eine kosmetische oder dermatologische Anwendung im Vordergrunde steht wie die Bekämpfung der oxidativen Beanspruchung der Haut. Es ist aber auch günstig, die erfindungsgemäßen Zubereitungen mit einem Gehalt an einem oder mehreren Antioxidantien zu versehen, wenn die Zubereitungen einem anderen Zwecke dienen sollen, z.B. als Desodorantien oder Sonnenschutzmittel.
Besonders vorteilhaft werden die Antioxidantien gewählt aus der Gruppe bestehend aus
Aminosäuren (z.B. Histidin, Tyrosin, Tryptophan) und deren Derivate, Imidazole (z.B. Urocaninsäure) und deren Derivate, Peptide wie D,L-Carnosin, D-Carnosin, L-Carno- sin und deren Derivate (z.B. Anserin), Carotinoide, Carotine (z.B. α-Carotin, ß-Ca- rotin, Lycopin) und deren Derivate, Liponsäure und deren Derivate (z.B. Dihydroli- ponsäure), Aurothioglucose, Propylthiouracil und andere Thiole (z.B. Thioredoxin, Glutathion, Cystein, Cystin, Cystamin und deren Glycosyl-, N-Acetyl-, Methyl-, Ethyl-,
Propyl-, Amyl-, Butyl- und Lauryl-, Palmitoyl-, Oleyl-, γ-Linoleyl-, Cholesteryl - und Glycerylester) sowie deren Salze, Dilaurylthiodipropionat, Distearylthiodipropionat, Thiodipropionsäure und deren Derivate (Ester, Ether, Peptide, Lipide, Nukleotide, Nukleoside und Salze) sowie Sulfoximinverbindungen (z.B. Buthioninsulfoximine, Homocysteinsulfoximin, Buthioninsulfone, Penta-, Hexa-, Heptahioninsulfoximin) in sehr geringen verträglichen Dosierungen (z.B. pmol bis μmol/kg), ferner (Metall)- Chelatoren (z.B. α-Hydroxyfettsäuren, α-Hydroxypalmitinsäure, Phytinsäure, Lactoferrin), α-Hydroxysäuren (z.B. Zitronensäure, Milchsäure, Apfelsäure), Huminsäure, Gallensäure, Gallenextrakte, Bilirubin, Biliverdin, EDTA, EGTA und deren Derivate, ungesättigte Fettsäuren und deren Derivate (z.B. γ-Linolensäure, Linolsäure, Ölsäure), Folsäure und deren Derivate, Ubichinon und Ubichinol deren Derivate, Vitamin C und Derivate (z.B. Ascorbylpalmitate, Mg - Ascorbylphosphate, Ascorbylacetate), Tocopherole und Derivate (z.B. Vitamin E - acetat), Vitamin A und Derivate (Vitamin A - palmitat) sowie Koniferylbenzoat des Benzoeharzes, Ru- tinsäure und deren Derivate, Ferulasäure und deren Derivate, Butylhydroxytoluol, Butylhydroxyanisol, Nordihydroguajakharzsäure, Nordihydroguajaretsäure, Trihydro- xybutyrophenon, Harnsäure und deren Derivate, Zink und dessen Derivate (z.B. ZnO, ZnSO4) Selen und dessen Derivate (z.B. Selenmethionin), Stilbene und deren Derivate (z.B. Stilbenoxid, Trans-Stilbenoxid) und die erfindungsgemäß geeigneten Derivate (Salze, Ester, Ether, Zucker, Nukleotide, Nukleoside, Peptide und Lipide) dieser genannten Wirkstoffe.
Besonders vorteilhaft im Sinne der vorliegenden Erfindung können öllösliche Antioxidantien eingesetzt werden.
Die Menge der Antioxidantien (eine oder mehrere Verbindungen) in den Zubereitungen beträgt vorzugsweise 0,001 bis 30 Gew.-%, besonders bevorzugt 0,05 - 20 Gew.-%, insbesondere 1 - 10 Gew.-%, bezogen auf das Gesamtgewicht der Zubereitung.
Sofern Vitamin E und/oder dessen Derivate das oder die Antioxidantien darstellen, ist vorteilhaft, deren jeweilige Konzentrationen aus dem Bereich von 0,001 - 10 Gew.-%, bezogen auf das Gesamtgewicht der Formulierung, zu wählen.
Sofern Vitamin A, bzw. Vitamin-A-Derivate, bzw. Carotine bzw. deren Derivate das oder die Antioxidantien darstellen, ist vorteilhaft, deren jeweilige Konzentrationen aus dem Bereich von 0,001 - 10 Gew.-%, bezogen auf das Gesamtgewicht der Formulierung, zu wählen.
Als vorteilhafte Wirkstoffe können aber auch - alternativ oder zusätzlich - verwendet werden ein oder mehrere Substanzen gewählt aus der Gruppe Acetylsalicylsäure, Atropin, Azulen, Hydrocortison und dessen Derivaten, z.B. Hydrocortison-17-valerat, Vitamine, z.B. Ascorbinsäure und deren Derivate, Vitamine der B- und D-Reihe, sehr günstig das Vitamin Bi, das Vitamin B12 das Vitamin DL aber auch Salicylsäure, Natriumsalicylat, Enzyme, DNA, Bisabolol, ungesättigte Fettsäuren, namentlich die essentiellen Fettsäuren (oft auch Vitamin F genannt), insbesondere die γ- Linolensäure, Ölsäure, Eicosapentaensäure, Docosahexaensäure und deren Derivate, Chloramphenicol, Coffein, Prostaglandine, Thymol, Campher, Extrakte oder andere Produkte pflanzlicher und tierischer Herkunft, z.B. Nachtkerzenöl, Borretschöl oder Johannisbeerkemöl, Fischöle, Lebertran aber auch Ceramide und ceramidähnliche Verbindungen und so weiter.
Obgleich auch die Verwendung hydrophiler Wirkstoffe erfindungsgemäß begünstigt ist, ist ein weiterer Vorteil der erfindungsgemäßen Zubereitungen, daß auch gerade öllösliche bzw. iipophile Wirkstoffe mit besonders großer Wirksamkeit biologisch verfügbar gemacht werden.
Erfindungsgemäße Verfahren können in Abwesenheit einer Ölkomponente beispielsweise folgendermaßen vorteilhaft durchgeführt werden:
a) Das oder die Lipide (wieder vorteilhaft: Phospholipide, Glycerinester, -ether) wird oder werden bei Raumtemperatur (gewünschtenfalls mit den zu verkapselnden Inhaltsstoffen) in einem Lösungsmittel, insbesondere einem polaren Lösungsmittel (z.B. einem Polyol wie Glycerin, Erythrit, Dipropylenglycol, Propylenglycol, Sorbit oder aber auch Ethanol) gelöst. Ein Tensid (in Wasser gelöst) wird zugesetzt, bis der Fiüssigkristall auftritt und die Mischung wird mit Wasser und weiteren wasserlöslichen Zusätzen verdünnt unter Bildung erfindungsgemäßer Liposomen.
b) Das oder die Lipide (wieder vorteilhaft: Phospholipide, Glycerinester, ether) wird oder werden in der Wärme (25-85°C) (gewünschtenfalls mit den zu verkapselnden Inhaltsstoffen) in einem Lösungsmittel, insbesondere einem polaren Lösungsmittel (z.B. einem Polyol wie Glycerin, Erythrit, Dipropylenglycol, Propylenglycol, Sorbit oder aber auch Ethanol) gelöst. Ein Tensid (in Wasser gelöst) wird zugesetzt, bis der Flüssigkristall auftritt und die Mischung wird mit Wasser und weiteren wasserlöslichen Zusätzen verdünnt unter Bildung erfindungsgemäßer Liposomen.
c) Das oder die Lipide (wieder vorteilhaft: Phospholipide, Glycerinester, -ether) wird oder werden in der Wärme (25-85°C) mit einem Tensid und gewünschtenfalls mit zu verkapselnden Inhaltsstoffen in einem Lösungsmittel, insbesondere einem polaren Lösungsmittel (z.B. einem Polyol wie Glycerin, Erythrit, Dipropylenglycol, Propylenglycol, Sorbit oder aber auch Ethanol) gelöst. Es wird Wasser zugesetzt, bis der Flüssigkristall auftritt und die Mischung wird mit Wasser und weiteren wasserlöslichen Zusätzen verdünnt unter Bildung erfindungsgemäßer Liposomen.
Erfindungsgemäße Verfahren können bei Anwesenheit einer Ölkomponente beispielsweise folgendermaßen vorteilhaft durchgeführt werden:
d) Das oder die Lipide (wieder vorteilhaft: Phospholipide, Glycerinester, -ether) wird oder werden bei Raumtemperatur mit einem Tensid, einer oder mehreren Ölkomponenten und gewünschtenfalls mit zu verkapselnden Inhaltsstoffen in einem Lösungsmittel, insbesondere einem polaren Lösungsmittel (z.B. einem Polyol wie Glycerin, Erythrit, Dipropylenglycol, Propylenglycol, Sorbit oder aber auch Ethanol) gelöst. Es wird Wasser zugesetzt bis der Flüssigkristall auftritt und die Mischung wird mit Wasser und weiteren wasserlöslichen Zusätzen verdünnt unter Bildung erfindungsgemäßer Liposomen.
e) Das oder die Lipide (wieder vorteilhaft: Phospholipide, Glycerinester, -ether) wird oder werden in der Wärme (25-85°C) mit einem Tensid, einer oder mehreren Ölkomponenten und gewünschtenfalls mit zu verkapselnden Inhaltsstoffen in einem Lösungsmittel, insbesondere einem polaren Lösungsmittel (z.B. einem Polyol wie Glycerin, Erythrit, Dipropylenglycol, Propylenglycol, Sorbit oder aber auch Ethanol) gelöst. Es wird Wasser zugesetzt bis der Flüssigkristall auftritt und die Mischung
wird mit Wasser und weiteren wasserlöslichen Zusätzen verdünnt unter Bildung erfindungsgemäßer Liposomen.
Im folgenden werden vorteilhafte Ausführungsbeispiele der vorliegenden Erfindung aufgeführt, ohne daß eine Beschränkung auf diese Beispiele beabsichtigt ist.
Herstellunqsbeispiel 1 für Vesikel-Dispersion
15 mg Natriumlauroyllactylat und 95 mg Phospholipon 90 werden in 500 ml Ethanol bei 80°C gelöst. Anschließend tropft man bei dieser Temperatur Wasser zu, bis eine flüssigkristalline Phase auftritt . Weiteres Verdünnen dieser Phase mit 13.5 g Wasser liefert eine Dispersion von Liposomen mit mittlerem Radius von ca. 145 nm.
Herstellunqsbeispiel 2 für Vesikel-Dispersion
16 mg Natriumlauroyllactylat und 104 mg Phospholipon 90 werden in 1027 mg Glycerin bei 80°C gelöst. Anschließend tropft man bei dieser Temperatur Wasser zu, verdünnt weiter mit 11.1 g Wasser und erhält eine Dispersion von Liposomen mit mittlerem Radius von ca. 117 nm.
Herstellungsbeispiel 3 für Vesikel-Dispersion
17 mg Natriumlauroylsarcosinat und 100 mg Phospholipon 90 werden in 500 ml Ethanol bei 80°C gelöst. Anschließend tropft man bei dieser Temperatur Wasser zu, bis eine flüssigkristalline Phase auftritt . Weiteres Verdünnen dieser Phase mit 11.2 g Wasser liefert eine Dispersion von Liposomen mit mittlerem Radius von ca. 148 nm.
Herstellungsbeispiel 4 für Vesikel-Dispersion
18 mg Natriumlauroyllactylat und 100 mg Phospholipon 90 werden in 1041 mg Propylenglycol bei 80°C gelöst. Anschließend verdünnt man bei dieser Temperatur mit 12.0 g Wasser und erhält eine Dispersion von Liposomen mit mittlerem Radius von ca. 161 nm.
Herstellungsbeispiel 5 für Vesikel-Dispersion
16 mg Cetyltrimethylammoniumbromid und 105 mg Phospholipon 90 werden in 996 mg Proylenglycol gelöst. Anschließend gibt man insgesamt 12 g Wasser zu und erhält eine liposomale Dispersion (117 nm Radius)
Herstellungsbeispiel 5a für Vesikel-Dispersion
13 mg Cetyltrimethylammoniumbromid und 114 mg Lecithin (Phospholipon 90) werden in 1025 mg Proylenglycol gelöst. Anschließend gibt man insgesamt 12 g Wasser zu und erhält eine liposomale Dispersion (133 nm Radius).
Herstellunqsbeispiel 5b für Vesikel-Dispersion
9 mg Cetyltrimethylammoniumbromid und 123 mg Lecithin (Phospholipon 90) werden in 1004 mg Proylenglycol gelöst. Anschließend gibt man insgesamt 12 g Wasser zu und erhält eine liposomale Dispersion (191 nm Radius).
Herstellunqsbeispiel 6 für Vesikel-Dispersion
18 g Laurylglycosid (Plantaren 1200) und 127 mg Phospholipon 90 werden in 1048 mg Propylenglycol gelöst. Anschließend gibt man insgesamt 12 g Wasser zu und erhält eine liposomale Dispersion (283 nm Radius).
Herstellunqsbeispiel 7 für Vesikel-Dispersion
20 mg Glycerylstearatcitrat und 131 mg Phospholipon 90 werden in 1027 mg Proylenglycol gelöst. Anschließend gibt man insgesamt 12 g Wasser zu und erhält eine liposomale Dispersion (235 nm Radius).
Herstellungsbeispiel 8 für Vesikel-Dispersion mit Ölkomponente:
87 mg Natriumlauroyllactylat und 260 mg Phospholipon 90 werden in 126 mg Ethanol und 82 mg Dicaprylylether gelöst Dann wird bei 80°C langsam Wasser zugetropft. Es bildet sich ein Gel, das mit Wasser verdünnt wird. Es bilden sich multilamellare Vesikel. Insgesamt wird mit 8 g Wasser verdünnt.
Herstellunqsbeispiel 9 für Vesikel-Dispersion mit Ölkomponente:
79 mg Natriumlauroyllactylat und 305 mg Phospholipon 90 werden in 80 mg Ethanol und 85 mg Dicaprylylether gelöst. Dann wird bei 80°C langsam Wasser zugetropft.
Es bildet sich ein Gel, das mit Wasser verdünnt wird. Es bilden sich multilamellare Vesikel. Insgesamt wird mit 7 g Wasser verdünnt.
Herstellungsbeispiel 10 für Vesikel-Dispersion
49 mg Polyglycerinmonoisostearat und 188 mg Lecithin (Phospholipon 90) werden in 1670 mg Proylenglycol gelöst. Anschließend gibt man insgesamt 20 g Wasser zu und erhält eine liposomale Dispersion (156 nm Radius).
Herstellungsbeispiel 10a für Vesikel-Dispersion
20 mg Polyglycerinmonoisostearat und 146 mg Lecithin (Phospholipon 90) werden in 1270 mg Proylenglycol gelöst. Anschließend gibt man insgesamt 15 g Wasser zu und erhält eine liposomale Dispersion (192 nm Radius).
Herstellungsbeispiel 10b für Vesikel-Dispersion
10 mg Polyglycerinmonoisostearat und 101 mg Lecithin (Phospholipon 90) werden in 990 mg Proylenglycol gelöst. Anschließend gibt man insgesamt 12 g Wasser zu und erhält eine liposomale Dispersion (214 nm Radius).
Herstellungsbeispiel 11 für Vesikel-Dispersion
51 mg Natriumlauroyllactylat und 211 mg Lecithin (Phospholipon 90 H) werden in 1630 mg gelöst. Anschließend gibt man insgesamt 20 g Wasser zu und erhält eine liposomale Dispersion (80 nm Radius).
Herstellbeispiel 12 für Vesikel-Dispersion
178 mg Lecithin (Phopholipon 90) werden in 1145 mg Propandiol gelöst und es wird 238 mg Natriumcocoylglutamat (Tamina) zugegeben. Bei 80°C wird mit insgesamt 15 g Wasser verdünnt (163 nm Radius).
Herstellbeispiel 13 für Vesikel-Dispersion
145 mg Lecithin (Phopholipon 90) und 53 mg Hydroxyethylamonium Methosulfat (Dehyquat F75) werden in 1201 mg Propandiol gelöst und mit insgesamt 15 g Wasser verdünnt (142 nm Radius).
Herstellbeispiel 14 für Vesikel-Dispersion
215 mg Lecithin (Phopholipon 90) und 55 mg Polyglycerin-dipalmitostearat (Polydermanol PS-60-DS) werden in 1830 mg Propandiol gelöst. Bei 80°C werden 1800 mg Wasser zugegeben. Man läßt auf Raumtemperatur abkühlen. Es entsteht ein transparenter Flüssigkristall, der bei langsamem Zutropfen von Wasser in eine Vesikelsuspension übergeht (insgesamt 18 g Wasser; 128 nm Radius).
Herstellbeispiel 15 für Vesikel-Dispersion
188 mg Lecithin (Phopholipon 90) und 49 mg Polyglycerin-dipalmitostearat (Polydermanol GE-14-DA) werden in 1670 mg Propandiol gelöst. Bei 80°C werden 1500 mg Wasser zugegeben. Es bildet sich eine lamellare flüssigkristalline Phase, die auch bei Raumperatur erhalten bleibt. Verdünnen mit Wasser führt zu einer Vesikelsuspension (156 nm Radius).
Herstellbeispiel 16 für Vesikel-Dispersion
182 mg Lecithin (Phopholipon 90) und 273 mg Laurethersulfat, 25%ig, werden in 1206 mg Propandiol gelöst und bei 80°C werden 15 g Wasser zugegeben. Vesikelsuspension (133 nm Radius).
Herstellbeispiel 17 für Vesikel-Dispersion
126 mg Lecithin (Phopholipon 90) und 24 mg Ascorbylpalmitat werden in 980 mg Propandiol gelöst und bei 80°C werden 12 g Wasser zugegeben. Vesikelsuspension (142 nm Radius).
Herstellbeispiel 18 für Vesikel-Dispersion
206 mg Lecithin (Phopholipon 90) und 53.8 mg Natriumlauroyllactylat, 14 mg Ascorbylpalmitat werden in 1990 mg Propandiol gelöst und mit 20 g Wasser verdünnt. Vesikelsuspension (124 nm Radius).
Herstellbeispiel 19 für Vesikel-Dispersion
220 mg Lecithin (Phopholipon 90) und 57 mg Natriumluroyllactylat, 12 mg Ascorbinsäure werden in 1710 mg Propandiol gelöst und mit 20 g Wasser verdünnt. Vesikelsuspension (286 nm Radius).
Herstellbeispiel 20 für Vesikel-Dispersion
180 mg Lecithin (Phopholipon 90 H) und 48 mg Tocopheryl PEG-1000 Succinat (Eastman) werden in 1570 mg Propandiol gelöst und mit insgesamt 20 g Wasser verdünnt. Vesikelsuspension (256 nm Radius).
Herstellbeispiel 21 für Vesikel-Dispersion
344 mg Glycerylmonocaprylat und 46 mg Natriumlauroyllactylat werden bei 80°C miteinander verrührt und anschließend mit 24.6 g Wasser verdünnt. Vesikelsuspension (180 nm Radius).
Herstellungsbeispiel 21a für Vesikel-Dispersion
446 mg Glycerylmonocaprylat und 116 mg Natriumlauroyllactylat werden bei 80°C miteinander verrührt und anschließend mit 24.4 g Wasser verdünnt. Vesikelsuspension (148 nm Radius).
Herstellungsbeispiel 21b für Vesikel-Dispersion
400 mg Glycerylmonocaprylat und 165 mg Natriumlauroyllactylat werden bei 80°C miteinander verrührt und anschließend mit 24.4 g Wasser verdünnt. Vesikelsuspension (127 nm Radius).
Herstellungsbeispiel 21c für Vesikel-Dispersion
397mg Glycerylmonocaprylat und 272 mg Natriumlauroyllactylat werden bei 80°C miteinander verrührt und anschließend mit 24.3 g Wasser verdünnt. Vesikelsuspension (119 nm Radius).
