Abstract
Es werden die neuen Steroidester und -amide der Formel (I) beschrieben, worin R1 einen α- oder β-ständigen C¿1?-C3-Alkylrest, Sk -C C-(CH2)r-R?2¿, (E) oder (Z) -CH=CH-(CH¿2)r-R?2, -(CH¿2?)2-(CH2)r-R?2¿, wobei R2 eine veresterte oder als Amid derivatisierte Hydroxygruppe ist, und St eines der steroidalen ABC-Ringsysteme der Teilformel (A), (B) oder (C) darstellt, und R?6, R7 und R8¿ die in der Beschreibung angegebene Bedeutung haben. Die Derivatisierung erfolgt u.a. mit einer Aminosäure oder einem Peptid. Die neuen Verbindungen zeichnen sich gegenüber den zugrundeliegenden Hydroxyverbindungen ('Stammverbindungen') durch eine erheblich verbesserte Löslichkeit und zum Teil auch durch eine erhöhte biologische Wirkung und Selektivität aus. Die neuen Verbindungen sind zur Herstellung von Arzneimitteln geeignet.

Die Erfindung betrifft Steroidester und -amide, Verfahren zu ihrer Herstellung sowie ihre Verwendung als Hilfsstoffe für pharmakoiogische Untersuchungen und als Arzneistoffe.

Z Wasserstoff, Halogen, -OH, -NO2, -N3, -CN, -NR9aR9b, -NHSO2R9, -CO2R9, C1-C10-Alkyl, C1-C10-Alkoxy, C1-C10-Acyloxy, C1-C10-Acyl bedeutet,
R9a,b gleich oder verschieden sind und eine der unter R9 genannten Bedeutungen haben, R9 Wasserstoff oder C1-C10-Alkyl, und, falls R9 Wasserstoff ist, außerdem deren
physiologisch verträgliche Salze mit Basen (Z = -CO2H) bzw. Säuren
(Z = -NR9aR9b) bedeutet.
Die vom Kohlenstoffatom 17 ausgehenden Schlangenlinien zu den beiden Substituenten OH und Sk (Seitenkette) bedeuten, daß sich der jeweilige Substituent OH bzw. Sk sowohl in der α- als auch in der β-Position befinden kann.
Als Alkylgruppen R5, R5a, R6, R7, R8 und R9 sind gerad- oder verzweigtkettige Alkylgruppen mit 1-10 Kohlenstoffatomen zu betrachten, wie beispielsweise Methyl, Ethyl, Propyl, Isopropyl, Butyl, Isobutyl, tert.-Butyl, Pentyl, Isopentyl, Neopentyl, Heptyl, Hexyl,
Decyl sowie die entsprechenden ungesättigten Reste (bis zu 3 Doppelbindungen), wie beispielsweise Vinyl, Propenyl, Isopropenyl, Butenyl, Isobutenyl, Butadienyl, Hexenyl usw..
Die Alkylgruppe R1 ist eine gerad- oder verzweigtkettige Alkylgruppe mit 1-3 Kohlenstoffatomen, nämlich eine Methyl-, Ethyl-, Propyl oder Isopropylgruppe.
Die Alkylgruppen R5, R5a, R6, R7, R8 und R9 können substituiert sein durch 1-3 Halogenatome, Hydroxygruppen, C1-C10Alkoxygruppen, C6-C12-Arylgruppen, die durch 1-3 Halogenatome substituiert sein können, Heteroarylreste wie Furyl, Benzofuryl, Thienyl, Pyridyl, Pyrazolyl, Pyrimidinyl, Oxazolyl, Pyridazinyl, Pyrazinyl, außerdem Di-(C1-C4)- Alkylamine und Tri-(C1-C4)-Alkylammonium. Mögliche Alkylgruppen sind die oben genannten; bevorzugt sind solche Alkylgruppen, die nicht oder einfach substituiert sind.
Als Substituenten seien beispielsweise genannt Halogene wie Fluor-, Chlor- oder Bromatome, Phenyl, Dialkylamine wie Dimethylamino, Diethylamino, C1-C4-Alkoxy wie Methoxy, Ethoxy. Die C6-C12-Arylgruppen können ebenfalls durch die Halogenatome Fluor, Chlor oder Brom substituiert sein.
Als bevorzugte Alkylgruppe R1 ist eine Methylgruppe und als bevorzugte Alkylgruppen R5, R5a, R6, R7, R8 und R9 sind solche mit 1-5 Kohlenstoffatomen, wie z. B. Methyl, Ethyl, Propyl, Isobutyl, Butyl, tert.-Butyl zu nennen.
Die Cycloalkylgruppen R4 können im Ring 3-10, vorzugsweise 3-6 Kohlenstoffatome enthalten. Der Ring R4a kann durch geradkettige oder verzweigte Alkylgruppen mit 1-4 Kohlenstoffatomen substituiert sein. Beispielsweise seien Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl, Methylcyclopentyl, Methylcyclohexyl genannt.
Von den die charakteristische funktioneile Gruppe R2 tragenden Seitenketten Sk führen alle zu guten Ergebnissen, insbesondere ist jedoch eine veresterte ω-Hydroxyprop-1-enyl- und - but-1-enylkette sowie eine veresterte 3-Hydroxypropylkette hervorzuheben.
Für den Rest R2 ist eine der Gruppen -O-(Xi 1-Xi 2-Xi 3-..-Xi p), -C(=O)-(Yi 1-Yi 2-Yi 3-...-Yi p) oder -OC(=O)R3 bevorzugt.
p besitzt vorzugsweise den Wert 1, d.h. die Seitenkette Sk ist dann endständig vorzugsweise mit einer Aminosäure verestert oder stellt das Carbonsäureamid einer Aminosäure dar.
Die in R5a, R8 bzw. Z in der allgemeinen Formel I enthaltenen Alkoxygruppen sollen jeweils 1 bis 10 Kohlenstoffatome enthalten, wobei Methoxy-, Ethoxy- Propoxy- Isopropoxy- und t- Butyloxygruppen bevorzugt sind.
Bei dem C1-C10-Acyl- oder C1-C10-Acyloxyrest unter der Definition Z handelt es sich um einen C1-C10 -Alkanoyl(oxy)- oder Benzoyl(oxy)rest, vorzugsweise um eine Formyl-, Acetyl-, Propionyl- oder Isopropionylgruppe.
Vorzugsweise steht in den Verbindungen gemäß vorliegender Erfindung einer der 17- Substituenten für eine freie Hydroxygruppe, wie dies aus der allgemeinen Formel I hervorgeht. Es ist aber im der vorliegenden Erfindung auch möglich, daß diese freie Hydroxygruppe mit einem
r der Definition der Alkylgruppen R5, R5a, R6, R7, R8 und R9 genannten Reste verethert oder mit einem wie unter Z genannten C1-C10-Acylrest verestert ist.
Halogen in den Definitionen für R6, R7 und Z bedeutet Fluor, Chlor, Brom oder Iod.
Ist R8 eine Gruppe Z, so ist dies vorzugsweise eine Dialkylaminogruppe -NR9aR9b, insbesondere die Dimethylaminogruppe, eine C1-C10-Alkoxy-, insbesondere die Methoxygruppe oder ein C1-C10-Acyl-, vornehmlich der Formyl- oder Acetylrest.
Als ein gegebenenfalls mit einer Gruppe Z substituierter Arylrest R8 kommen carbocyclische oder heterocyclische Arylreste wie z.B. Phenyl, Naphtyl, Furyl, Benzofuryl, Thienyl, Pyridyl, Pyrazolyl, Pyrimidinyl, Oxazolyl, Pyridazinyl, Pyrazinyl, Chinolyl infrage.
Bevorzugte Arylreste sind Phenyl, Naphtyl, Furyl, Benzofuryl, Thienyl, Pyridyl; als substituierter Arylrest R8 sind in erster Linie der 4-Cyanophenyl- und ein 4-Halogenphenyl-, insbesondere der 4-Fluorphenylrest, zu nennen.
In erfindungsgemäß bevorzugten Verbindungen stehen die Substituenten R6 sowie R7 jeweils für ein Wasserstoffatom.
Zwischen den Kohlenstoffatomen 6 und 7 befindet sich vorzugsweise eine C-C-
Einfachbindung.
Die Aminosäuren HO-X-H bzw. H-Y-OH können eine natürliche oder unnatürliche, d.h. bezüglich ihrer Konfiguration oder Substitution modifizierte α-, β- oder γ- Aminosäure darstellen.
Bevorzugt werden natürliche Aminosäuren wie zum Beispiel Glycin, Alanin, Valin, Leucin,
Isoleucin, Prolin.
Eine freie Aminogruppe in HO-X-H bzw. H-Y-OH kann mit den, dem Fachmann geläufigen
Schutzgruppen, wie zum Beispiel tert.-Butyloxycarbonyl- (t-BOC), Benzyloxycarbonyl-,
Biphenylisopropyloxycarbonyl- oder 9-Fluorenylmethoxycarbonyl- versehen sein.

Die Erfindung betrifft ferner Verfahren zur Herstellung von Steroidestern und -amiden der allgemeinen Formel I, bei welchem eine Verbindung der allgemeinen Formel II,
die in der Seitenkette Sk' eine endständige, freie Hydroxylgruppe trägt (R2' = OH) und in der alle anderen Substituenten die in der allgemeinen Formel I angegebene Bedeutung haben und in St vorhandene funktionelle Gruppen gegebenenfalls nach in dem Fachmann bekannter Weise geschützt sein können, mit einer an der Aminogruppe gegebenenfalls geschützten (R5 ≠ H) Aminosäure HO-X-R5 oder einem an den Aminogruppen gegebenenfalls geschützten Peptid H-O-(Xi 1-Xi 2-Xi 3-...-Xi p)-R5 (p>1) oder einem Carbonsäurehalogenid oder -anhydrid R3-C(=O)Hal (Hai = Cl, Br) bzw. [R3-C(=O)-]2O verestert oder, wenn R2 in der Verbindung der allgemeinen Formel I ein Rest -C(=O)-(Yi 1-Yi 2-Yi 3-...-Yi p)-OR5 sein soll, die endständige Hydroxylgruppe R2' zunächst vollständig zur Carboxylgruppe oxidiert, anschließend die erhaltene Carbonsäure, gegebenenfalls nach Aktivierung, mit einer an der Aminogruppe gegebenenfalls geschützten (R5≠ H) Aminosäure R5-Y-OR5' oder einem an der Aminogruppe gegebenenfalls geschützten Peptid R5-(Yi 1-Yi 2-Yi 3-...-Yi p)-OR5 (p>1) oder einem gegebenenfalls geschützten Amin HNR5C(=O)R3 zum entsprechenden Amid umgesetzt wird sowie gegebenfalls in St, der Aminosäure, dem Peptid oder dem Amin vorhandene Schutzgruppen abgespalten werden.
Sämtliche für diesen Syntheseweg erforderlichen Reaktionsschritte (Schutz der funktionellen Gruppen in St, Schutz der Aminogruppe, Veresterungsreaktion, Oxidation der Hydroxylgruppe, Säureaktivierung durch Bildung eines "Reaktivesters", Amidbildung,
Abspaltung der Schutzgruppen) werden nach gängigen und dem Fachmann geläufigen Methoden durchgeführt.
Soll p in der gewünschten Verbindung der allgemeinen Formel I einen Wert >1 besitzen, kann, anstelle wie oben beschrieben, das Peptid oder Amid auch durch aufeinanderfolgende Reaktionsschritte an der Seitenkette sukzessive aufgebaut werden, beispielsweise
Aminoschutzgruppe darstellen sowie X1, X2,..... die vorstehend unter Xi 1...p angegebene
Bedeutung haben.
Die zur Herstellung der Verbindungen der allgemeinen Formel I dienenden Ausgangsverbindungen der allgemeinen Formel II mit einer freien Hydroxygruppe in der 17- Seitenkette Sk' (R2' = OH) -das sind die an anderer Stelle auch als "Stammverbindungen" bezeichneten Verbindungen- sowie deren Herstellung sind in einer ganzen Reihe von Patenten, Patentanmeldungen und Veröffentlichungen beschrieben:

sowie weiteren, dem auf diesem Gebiet tätigen Fachmann bekannten,
einschlägigen Literaturstellen.
Viele steroidale Verbindungen, insbesondere solche mit einem 11β-Arylrest, sind aufgrund ihres Substitutionsmusters zum Teil sehr schlecht löslich, sodaß zum Erhalt therapeutisch relevanter Plasmaspiegel oftmals überproportional hohe Einzel- oder Mehrfachdosierungen oder aufwendige Formulierungstechniken angewendet werden müssen.
Es wurde nun gefunden, daß die Löslichkeit derartiger Verbindungen überraschenderweise ganz erheblich verbessert werden kann, wenn an geeigneter Stelle in der Stammverbindung eine freie Hydroxylgruppe verestert oder in ein Amid überführt wird. Die hierdurch erhaltenen neuen Verbindungen besitzen über die verbesserte Löslichkeit hinaus zum Teil
selbst eine erhöhte biologische Wirkung und Selektivität verglichen mit der entsprechenden Stammverbindung. Die neuen Verbindungen sind wohl per se wirksam und stellen somit nicht bloße Prodrugs dar.
Steroidale kompetitive Progesteronantagonisten, in denen eine freie Hydroxygruppe in der
17-Seitenkette verestert ist, sind beispielsweise bereits aus den folgenden europäischen
Patentanmeldungen bekannt: a) 0 186 834, b) 0 283 428 und c) 0 404 283.
Im Fall a) handelt es sich um Ester des Onapristons [11β-(4-Dimethylaminophenyl)-17α- hydroxy-17β-(3-hydroxypropyl)-13α-methyl-4,9-gonadien-3-on], d.h. um Verbindungen mit einem ABC-Ringsystem der Teilformel B mit α-ständigem Alkylrest R1 und einer veresterten
17α-(3-Hydroxypropyl)-Seitenkette. Die Kettenlänge des Acylrestes ist auf maximal 10
Kohlenstoffatome beschränkt.
Im Fall b) tragen die Verbindungen mit einem ABC-Ringsystem der Teilformel C und im
Fall c) die Verbindungen mit einem ABC-Ringsystem der Teilformel A u. a. eine 17-AlkyI-,
17-Alkenyl- oder 17-Alkinylseitenkette, die endständig eine gegebenenfalls veresterte
Hydroxyfunktion aufweisen kann.
Über eine Verbesserung der Löslichkeit der Stammverbindungen mit einer endständigen, freien Hydroxygruppe in der 17-Seitenkette durch Veresterung ebendieser Hydroxygruppe ist in diesen Druckschriften nichts ausgesagt.
Auf Grund der verbesserten Löslichkeit (Tab. 1) der erfindungsgemäßen Verbindungen ist es prinzipiell möglich, die für eine pharmakologisch gewünschte Wirkung benötigte Dosis zu reduzieren, was in der Regel zu einer Reduktion unerwünschter Nebenwirkungen und somit zu einer Vergrößerung der therapeutischen Breite führt.
Sie sind daher zur Herstellung von solchen Arzneimitteln geeignet, die bei gleicher molarer Dosierung einer Verbindung der allgemeinen Formel I wie der entsprechenden Stammverbindung in einer entsprechenden Formulierung zu einer höheren Bioverfügbarkeit des Wirkstoffs führen bzw. in denen zur Erzielung einer vergleichbaren biologischen Wirkung die Dosis einer Verbindung der allgemeinen Formel I relativ zur Stammverbindung reduziert werden kann.
Die neuen Verbindungen der allgemeinen Formel I sind also wertvolle pharmazeutische Wirkstoffe. So verfügen sie über eine starke Affinität zum Gestagenrezeptor und besitzen einen überraschend großen Bereich an antigestagenen, antiglucocorticoiden, antimineralcorticoiden und antiandrogenen Eigenschaften. Diese wichtigen biologischen Wirksamkeiten können für medizinische Zwecke genutzt werden.
Wirkstoffe dieser Art mit ausgeprägter antigestagener Aktivität sind zur Auslösung von Aborten geeignet, da sie das zur Aufrechterhaltung der Schwangerschaft erforderliche Progesteron vom Rezeptor verdrängen. Sie sind deshalb wertvoll und interessant im Hinblick auf ihre Verwendung zur postcoitalen Fertilitätskontrolle. Die erfindungsgemäßen Verbindungen der allgemeinen Formel I sind auch zur Herstellung von Präparaten für die Empfängnisverhütung für die Frau geeignet (WO-A 93/23020).
Sie können außerdem gegen hormonelle Unregelmäßigkeiten, zur Menstruationsauslösung und zur Geburtseinleitung eingesetzt werden. Weitere Indikationsgebiete im Bereich der Gynäkologie sind die Behandlung von den mit einer Dysmenorrhoe einhergehenden Beschwerden sowie der Endometriose.
Schließlich können sie für die Behandlung von hormonabhängigen Carcinomen verwendet werden.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel I weisen auch eine antiglucocorticoide Aktivität auf und können somit auch als Arzneimittel zur Therapie corticoid-induzierter Störungen (Glaukom) sowie zur Bekämpfung von Nebenwirkungen, die bei langfristiger Behandlung mit Glucocorticoiden auftreten (Cushing-Syndrom) eingesetzt werden. Sie ermöglichen daher auch die auf eine Supersekretion der Glucocorticoide zurückzuführenden Störungen, vor allem die Adipositas, Arteriosklerose, Hypertension, Osteoporose, Diabetes sowie die Insomnie zu bekämpfen.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel I mit antiandrogener Aktiviät können bei der Behandlung der Hypertrophie und des Prostatakarzinoms verwendet werden. Sie ermöglichen weiterhin eine spezifische Therapie von Androgenisierungserscheinungen bei Frauen: die pathologische Behaarung bei Hirsutismus, die androgenetische Alopezie sowie die gesteigerte Talgdrüsenfunktion bei Akne und Seborrhoe sind günstig beeinflußbar.
Die Erfindung betrifft somit auch Arzneimittel auf Basis der pharmazeutisch verträglichen, d.h. in den verwendeten Dosen nicht toxischen Verbindungen der allgemeinen Formel I, zusammen mit den üblichen Hilfs- und Trägerstoffen.
Schließlich betrifft die vorliegende Erfindung auch die Verwendung der Verbindungen der allgemeinen Formel I zur Herstellung von Arzneimitteln.
Die erfindungsgemäßen Verbindungen können nach an sich bekannten Methoden der Galenik zu pharmazeutischen Präparaten für die enterale, percutane, parenterale oder lokale Applikation verarbeitet werden. Sie können in Form von Tabletten, Dragees, Gelkapseln, Granulaten, Suppositorien, Implantaten, injizierbaren sterilen wäßrigen oder öligen Lösungen, Suspensionen oder Emulsionen, Salben, Cremes und Gelen verabreicht werden.
Der oder die Wirkstoffe können dabei mit den in der Galenik üblichen Hilfsstoffen wie z.B. Gummiarabikum, Talk, Stärke, Mannit, Methylcellulose, Laktose, Tensiden wie Tweens oder Myrj, Magnesiumstearat, wäßrigen oder nicht wäßrigen Trägern, Paraffinderivaten, Netz-, Dispergier-, Emulgier-, Konservierungsmitteln und Aromastoffen zur Geschmackskorrektur (z.B. etherischen Ölen) gemischt werden.
Eine Dosiseinheit enthält etwa 0,1-100 mg Wirkstoff(e). Die Dosierung der erfindungsgemäßen Verbindungen liegt beim Menschen bei etwa 0,1-1000 mg pro Tag.
Die nachfolgenden Beispiele dienen der näheren Erläuterung der vorliegenden Erfindung:
BEISPIEL 1
(Z)-6'-(4-CyanphenyI)-9,11α-dihydro-17β-hydroxy-17α-[3-(1-oxopropoxy)-1-propenyl]- 4,H-naphtho[3',2',1':10,9,11] estr-4-en-3-on
Die Lösung von 400 mg (0,77 mmol) (Z)-6'-(4-Cyanphenyl)-9,11α-dihydro-17β-hydroxy- 17α-(3-hydroxy-1-propenyl)-4'H-naphtho[3',2',1':10,9,11]estr-4-en-3-on in einem Gemisch aus 4 ml wasserfreiem Dichlormethan und 4 ml wasserfreiem Pyridin versetzt man unter einer Atmosphäre von trockenem Argon mit 0,2 ml Propionsäurechlorid und läßt 1 Stunde bei 23°C rühren. Man gießt in eine gesättigte wäßrige Natriumhydrogencarbonatlösung, extrahiert mehrfach mit Dichlormethan und trocknet die vereinigten organischen Extrakte über Natriumsulfat. Nach Filtration und Lösungsmittelabzug reinigt man den erhaltenen Rückstand durch Chromatographie an 200 ml neutralem Aluminiumoxid der Aktivitätsstufe III mit einem Laufmittelgemisch aus n-Ηexan und Ethylacetat. Isoliert werden 346 mg (0,60 mmol, 78%) der Titelverbindung als kristalliner Feststoff.
BEISPIEL 2
(Z)-6 -(4-Cyanphenyl)-9,11α-dihydro-17β-hydroxy-17α-[3-[(1-oxopentyl)oxy]-1- propenyl]-4'H-naphtho[3',2',1':10,9,11]estr-4-en.3.on
In Analogie zu Beispiel 1 werden 400 mg (0,77 mmol) (Z)-6'-(4-Cyanphenyl)-9,11α-dihydro- 17β-hydroxy-17α-(3-hydroxy-1-propenyl)-4'H-naphtho[3,,2',1':10,9,11]estr-4-en-3-on unter Verwendung von Pentansäureanhydrid umgesetzt. Man isoliert nach Aufarbeitung und Reinigung 392 mg (0,65 mmol, 84%) der Titelverbindung als kristallinen Feststoff.

(Z)-6'-(4-Cyanphenyl)-9,11α-dihydro-17β-hydroxy-17α-[3-[(1-oxoheptyl)oxy]-1- propenyl]-4'H-naphtho[3',2',1':10,9,11] estr-4-en-3-on

Man isoliert nach Aufarbeitung und Reinigung 345 mg (0,55 mmol, 71%) der Titelverbindung als farblosen Schaum.

(Z)-17α-[3-[(Aminoacetyl)oxy]-1-propenyl]-6'-(4-cyanphenyl)-9,11α-dihydro-17β- hydroxy-4'H-naphtho[3',2',1':10,9,11] estr-4-en-3-on
780 mg (1,13 mmol) der nach Beispiel 13 dargestellten Verbindung versetzt man bei 0°C unter einer trockenen Atmosphäre aus Argon mit 5 ml Trifluoressigsäure und läßt 20 Minuten rühren. Man gießt in eine gesättigte wässrige Natriumhydrogencarbonatlösung, extrahiert mehrfach mit Dichlormethan und trocknet die vereinigten organischen Extrakte über Natriumsulfat. Nach Filtration und Lösungsmittelabzug reinigt man den erhaltenen Rückstand durch Kristallisation aus Ethylacetat und isoliert 287 mg (0,49 mmol, 43%) der Titelverbindung als kristallinen Feststoff.

Die Lösung von 400 mg (0,77 mmol) (Z)-6'-(4-Cyanphenyl)-9,11α-dihydro-17β-hydroxy- 17α-(3-hydroxy-1-propenyl)-4'H-naphtho[3',2',1':10,9,11]estr-4-en-3-on in einem Gemisch aus 4 ml wasserfreiem Dichlormethan und 4 ml wasserfreiem Pyridin versetzt man unter einer Atmosphäre von trockenem Argon mit 200 mg N-(tert.-butoxycarbonyI)-glycin, 20 mg 4- Dimethylaminopyridin, 184 mg Dicyclohexylcarbodiimid und läßt 16 Stunden bei 23°C rühren. Man filtriert von ausgefallenem Harnstoff ab, wäscht mit wenig Dichlormethan nach und reinigt den nach Lösungsmittelabzug erhaltenen Rückstand durch Chromatographie an 200 ml feinem Kieselgel mit einem Gradientensystem aus n-Hexan und Ethylacetat. Isoliert werden 373 mg (0,55 mmol, 72%) der Titelverbindung als farbloser Schaum.

Beispiel 41a
(Z)-6'-(4-Fluorphenyl)-17α-(3-hydroxyprop-1-enyl)-9,11α-dihydro-17β-hydroxy-4'H- naphtho[3',2',1':10,9,11]estr-4-en-3-on
Die Lösung von 435 mg (852 μmol) der nach Beispiel 41b dargestellten Verbindung in 5 ml wasserfreiem Tetrahydrofuran versetzt man mit 43 mg Palladium/Bariumsulfat (10%ig), 0,44 ml Pyridin und hydriert bei einer Atmosphäre bis zur Aufnahme der berechneten Wasserstoffmenge. Den nach Filtration und Lösungsmittelabzug erhaltenen Rückstand reinigt man durch Chromatographie an feinem Kieselgel unter Verwendung eines Gradientensystems aus n-Ηexan, Ethylacetat und Methanol. Isoliert werden 318 mg (620 μmol, 73%) der Titelverbindung als kristalliner Feststoff.

Beispiel 41b
6'-(4-FluorphenyI)-17α-(3-hydroxyprop-1-inyl)-9,11α-dihydro-17β-hydroxy-4'H- naphtho[3',2',1': 10,9,11]estr-4-en-3-on
Die Lösung von 3,4 g (4,86 mmol) der nach Beispiel 41c dargestellten Verbindung in 170 ml Aceton versetzt man unter einer Atmosphäre aus Argon mit 10 ml einer 4N Salzsäure und erwärmt 2,5 Stunden auf 50°C. Man gießt in eine gesättigte Natriumhydrogencarbonatlösung, extrahiert mit Dichlormethan und trocknet die vereinigten organischen Extrakte über Natriumsulfat. Den nach Filtration und Lösungsmittelabzug erhaltenen Rückstand reinigt man durch Chromatographie an feinem Kieselgel unter Verwendung eines Gradientensystems aus n-Hexan und Ethylacetat. Isoliert werden 2,32 g (4,54 mmol, 93%) der Titelverbindung als farbloser Schaum.

6'-(4-FIuorphenyl)-3-(3,3-dimethyltrimethylendioxy)-17α-[3-(tetrahydropyran-2-yloxy)- prop-1-inyl]-9,11α-dihydro-4'H-naphtho[3',2',1':10,9,11]estran-5α,17β-dioI
Die Lösung von 15,6 g (17,3 mmol) 6'-(4-Nonafluorbutylsulfonyloxy)-3-(3,3- dimethyltrimethylendioxy)-17α-[3-(tetrahydropyran-2-yIoxy)-prop-1-inyl]-9,11α-dihydro- 4'H-naphtho[3',2',1':10,9,11]estr-4-en-5α,17β-diol, das man in Analogie zu dem in DE 4216003 beschriebenen Verfahren hergestellt hat, in einem Gemisch aus 140 ml Toluol und 60 ml Ethanol versetzt man unter einer Atmosphäre aus Argon mit 0,4 ml einer 2M Natriumcarbonatlösung, 1,45 g Lithiumchlorid, 2,62 g 4-Fluorbenzolboron-säure, 988 mg Tetrakistriphenylphosphinpalladium (0) und erhitzt 1 Stunde auf 95°C. Zur Vervollständigung der Reaktion wird nochmals die gleiche Menge Katalysator zugegeben und weitere 1,5 Stunden erhitzt. Die erkaltete Mischung wird mit Ethylacetat verdünnt, mit gesättigter Natriumchloridlösung gewaschen und die organische Phase über Natriumsulfat getrocknet. Den nach Filtration und Lösungsmittelabzug erhaltenen Rückstand reinigt man durch Chromatographie an feinem Kieselgel unter Verwendung eines Gradientensystems aus n-Ηexan und Ethylacetat. Isoliert werden 10,2 g (14,6 mmol, 84%) der Titelverbindung als farbloser Schaum.

6'-(4-Fluorphenyl)-3-(3,3-dimethyltrimethylendioxy)-17α-[3-(tetrahydropyran-2-yloxy)- propyl]-9,11α-dihydro-4'H-naphtho[3',2',1':10,9,11]estran-5α,17β-diol
Die Lösung von 800 mg (1,14 mmol) der nach Beispiel 41c dargestellten Verbindung in 30 ml wasserfreiem Ethanol versetzt man mit 10 mg Palladium/Kohle (10%ig) und hydriert bei einer Atmosphäre bis zur Aufnahme der berechneten Wasserstoffmenge. Den nach Filtration und Lösungsmittelabzug erhaltenen Rückstand reinigt man durch Chromatographie an feinem Kieselgel unter Verwendung von n-Hexan und Ethylacetat. Isoliert werden 640 mg (910 μmol, 80%) der Titelverbindung als farbloser Schaum.

OH aus der Gruppe der Aminosäuren Glycin, Alanin, Valin, Leucin, Isoleucin, Prolin ausgewählt ist.

13. Verfahren zur Herstellung von Steroidestern und -amiden der allgemeinen Formel I, dadurch gekennzeichnet, daß eine Verbindung der allgemeinen Formel II, die in der Seitenkette Sk' eine endständige, freie Hydroxylgruppe trägt (R2' = OH) und in der alle anderen Substituenten die in der allgemeinen Formel I angegebene Bedeutung haben und in St vorhandene funktionelle Gruppen gegebenenfalls nach in dem Fachmann bekannter Weise geschützt sein können, mit einer an der Aminogruppe gegebenenfalls geschützten (R5 ≠ H) Aminosäure HO-X-R5 oder einem an den Aminogruppen gegebenenfalls geschützten Peptid H-O-(Xi 1-Xi 2-Xi 3-...-Xi p)-R5 (p>1) oder einem Carbonsäurehalogenid oder -anhydrid R3-C(=O)Hal (Hal = Cl, Br) bzw. [R3-C(=O)-]2O verestert oder, wenn R2 in der Verbindung der allgemeinen Formel I ein Rest -C(=O)-(Yi 1-Yi 2-Yi 3-...-Yi p)-OR5 sein soll, die endständige Hydroxylgruppe R2' zunächst vollständig zur Carboxylgruppe oxidiert, anschließend die erhaltene Carbonsäure, gegebenenfalls nach Aktivierung, mit einer an der Aminogruppe gegebenenfalls geschützten (R5≠ H) Aminosäure R5-Y-OR5' oder einem an der Aminogruppe gegebenenfalls geschützten Peptid R5-(Yi 1-Yi 2-Yi 3-...-Yi p)-OR5 (p>1) oder einem gegebenenfalls geschützten Amin HNR5C(=O)R3 zum entsprechenden Amid umgesetzt wird sowie gegebenfalls in St, der Aminosäure, dem Peptid oder dem Amin vorhandene Schutzgruppen abgespalten werden.
14. Arzneimittel, enthaltend mindestens eine Verbindung der allgemeinen Formel I gemäß Anspruch 1 sowie einen pharmazeutisch verträglichen Träger.
15. Verwendung der Verbindungen der allgemeinen Formel I gemäß Anspruch 1 zur Herstellung von Arzneimitteln.
