Abstract

Den beim Einbringen von Fremdmaterial in höhere eukaryotische Zellen, insbesondere bei der Transfektion mit DNA, auftretenden Toxizitätsproblemen wird begegnet, indem in der Zelle Genprodukte zur Expression gebracht werden, die die durch den Transfektionsprozeß ausgelöste Apoptose blockieren und/oder indem die Zellen mit anti-inflammatorischen Substanzen behandelt werden. Bevorzugt werden als anti-Apoptose-Gene Bcl-2, E1B 19K oder ein anti-apoptotisch wirksames Gen vom Hühner-Adenovirus CELO, als anti-inflammatorisch wirkende Substanz Adenovirus VA1, das in Form von VA1-DNA in die Zelle eingebracht wird, verwendet. Mit Hilfe dieser Maßnahmen kann eine lang anhaltende Genexpression erzielt werden.
