Description

The present invention essentially relates to a device for raising the level of a liquid, in a cell, well or the like.
It also relates to a multi-well plate for cell culture, microtitration or other use, equipped with this device.
Cell or multiple well plates frequently intended to contain one or more substances to be analyzed are frequently used in biology, in particular in cell biology.
But when it is desired to measure the optical density of a substance in solution in a liquid and contained in a well, for example according to the method described by F. BURMEISTER et al., In "Preprints of the XlVth IFSCC Congress, Barcelona, 1986, vol.I, the height of the liquid must be large enough to allow a measurement of optical density which is sufficiently precise and therefore reliable.
En effet, la densité optique qui se mesure en éclairant un échantillon transparent ou translucide par un 5 faisceau lumineux généralement monochromatique, est fonction de la longueur du trajet optique du faisceau lumineux, de la concentration de la substance dans la solution à mesurer et du coefficient d'absorption de la lumière propre à la substance, comme établi par la 0 formule suivante : D0 = ε c l0 dans laquelle :Indeed, the optical density which is measured by illuminating a transparent or translucent sample with a generally monochromatic light beam, is a function of the length of the optical path of the light beam, of the concentration of the substance in the solution to be measured and of the coefficient substance-specific light absorption, as established by the following formula 0: D 0 = ε cl 0 in which:
- D0 est la densité optique.- D 0 is the optical density.
- ε est le coefficient d'extinction molaire, dont la valeur dépend de la longueur d'onde de la lumière utilisée et de la nature de la substance dissoute.- ε is the molar extinction coefficient, the value of which depends on the wavelength of the light used and the nature of the dissolved substance.
C est la concentration de la substance dans la solution, etC is the concentration of the substance in the solution, and
10 est la longueur du trajet optique dans l'échantillon de solution.1 0 is the length of the optical path in the solution sample.
However, very often, we are faced with samples of very small volume and whose height in the well and consequently the optical path, is very small, which contributes to a notable imprecision of the optical density measurement. This imprecision becomes still much greater, therefore much more troublesome in the case where the value of the molar extinction coefficient of the sample analyzed is low. Furthermore, increasing the height of the sample in the well by adding additional liquid does not constitute a solution to the problem of improving the accuracy of the measurements, since this contribution has the effect of reducing the concentration of the substance due to the dilution achieved.
The object of the present invention is to solve the above problems. To this end, it relates to a device for raising the level of a liquid containing a dissolved substance, in a cell, well or the like, characterized by a wall sleeve of variable thickness and open at its two ends, which is insertable into said cell or well.
According to another characteristic of this device, the sleeve is integral with an element comprising a plurality of such sleeves.
This element can be a bar or a plate.
It can include notching lines between the sleeves to allow their separation so as to use only part of the set of sleeves.
The invention also relates to a plate with cells or multiple wells allowing for example the culture of cells or the microtitration of samples, characterized in that it is provided with a device corresponding to the preceding characteristics.
This plate can be covered by a plate comprising 'on one side as many sleeves as wells in the plate.
The invention also relates to the use of the above device or plate for measuring the optical density of a substance dissolved in this liquid.
It relates more particularly to the use of this device for the dosing of substances contained in a cell culture medium, these substances having for example been produced by cells in culture, or else having been released into the medium after solubilization of cells, these possibly having received an immunological labeling.
However, other characteristics and advantages of the invention will appear better in the detailed description which follows and which refers to the appended drawings, given solely by way of example, and in which.
Figure 1 is a perspective view of a sleeve inserted into a well.
Figure 3 is a perspective view similar to Figure 2 but showing individually separable sleeves of the bar.
Figure 4 is a sectional view of the bar of Figure 2, passing through the axis of the sleeves, and illustrated in the mounted position on a row of wells.
Figure 5 is a partial perspective view of a multiwell plate ready to receive a plate having on one side of the sleeves 1 according to the invention.
With particular reference to FIG. 1, it can be seen that a device according to the invention is, in general, constituted by a sleeve 1 which can be inserted into a cell or well 2 formed in an element 3 and which may contain a liquid in which any substance is dissolved.
The sleeve 1 is open at its two ends to allow the passage of the light beam, and its wall 4 can have a variable thickness, which characterizes the caliber of the sleeve 1.
Thus, as is easily understood, the sleeve 1, once introduced into the well 2, will reduce the volume of this well as a function of the thickness of the wall 4 and will therefore allow the liquid to rise in the well 2 of the desired height depending on the size used.
The sleeve 1, as well as the element 3 may be of any material such as for example a transparent plastic material for single use or reusable from one measurement to another after cleaning.
Thus, a light beam passing through the well and the liquid it contains, preferably in a direction substantially parallel to the axis of the well which is generally substantially cylindrical, will allow a measurement of the optical density of the substance dissolved in the liquid whose the height in the well will be large enough to provide the desired sensitivity, accuracy and efficiency of the measurement.
As seen in Figures 2 and 3, the sleeve 1 may be integral with a support 5 having a plurality of such sleeves projecting from one side of said support.
As can be seen in FIG. 2, the support 5 can consist of a bar carrying a plurality of sleeves 1, the spacing of which corresponds to the spacing between the wells 2 of a line of wells made in the element 3. The bar-shaped support 5 can include notching lines 6 between the sleeves 1, which makes it easy to separate them from one another so as to insert them into such and such a well formed in the element 3.
On a montré sur la figure 4, un ensemble linéaire de manchons 1 appartenant à une barrette 5, comme montré sur la figure 2, laquelle ligne de manchons est insérée dans les puits 2 d'espacement correspondant, ménagés dans 1 'élément 3.FIG. 4 shows a linear set of sleeves 1 belonging to a bar 5, as shown in FIG. 2, which line of sleeves is inserted into the wells 2 with corresponding spacing, formed in the element 3.
The sleeves 1 can also be secured to a support in the form of a plate or plate 7, as can be seen in FIG. 5 where said sleeves 1 project from the same face of the plate 7.
This plate 7 with associated sleeves may comprise as many sleeves 1 as there are cells or wells 2, provided in a plate P with multiple wells allowing, for example, cell culture, microtitration of samples, assay operations or even other uses.
It goes without saying that sleeves 1 can be used in only certain wells 2 of plate P, for example using a bar 5 with breakable or non-breaking sleeves, as seen in FIGS. 2 and 3 .
Of course, the invention is in no way limited to the embodiment described and illustrated, which has been given only by way of example.
This is how the material and the wall thickness of the sleeves 1 can be any, as well as the geometrical position of the sleeves on a plate, a bar or any other support.
This means that the invention includes all the technical equivalents of the means described and their combinations if these are carried out according to the spirit.
