Description
The subject of the present invention is essentially a device for raising the level of a liquid, in a cell, well or the like.
It also relates to a multi-well plate for cell culture, microtiter or other use, equipped with this device.
Frequently used in biology, particularly in cell biology, cell plates or multiple wells for containing one or more substances to be analyzed.
But when one wants to measure the optical density of a substance in solution in a liquid and contained in a well, for example according to the method described by F. BURMEISTER et al., In "Preprints of the XIVth IFSCC Congress,
Barcelona, 1986, vol. I, it is necessary that the height of the liquid is large enough to allow an optical density measurement that is sufficiently accurate, and therefore reliable.
In fact, the optical density, which is measured by illuminating a transparent or translucent sample with a generally monochromatic light beam, is a function of the length of the optical path of the light beam, the concentration of the substance in the solution to be measured and the optical coefficient. absorption of the light proper to the substance, as established by the following formula Do = s C lo
in which
- Do is the optical density.
- E is the molar extinction coefficient, the value of which depends on the wavelength of the light used and the nature of the dissolved substance.
- 1, est la longueur du trajet optique dans l'échantillon de solution.- C is the concentration of the substance in the solution, and
- 1, is the length of the optical path in the solution sample.
However, very often, one is faced with samples of very small volume and whose height in the well and therefore the optical path, is very small, which contributes to a significant inaccuracy of the optical density measurement. This inaccuracy becomes even more important, therefore much more troublesome in the case where the value of the molar extinction coefficient of the sample analyzed is low. Moreover, the increase in the height of the sample in the well by adding additional liquid does not constitute a solution to the problem of improving the accuracy of the measurements, because this contribution has the effect of reducing the concentration of the sample. the substance due to the dilution carried out.
The present invention aims to solve the above problems.
For this purpose, it relates to a device for raising the level of a liquid containing a dissolved substance, in a cell, well or the like, characterized by a sleeve with a wall of variable thickness and open at both ends, which is insertable into said cell or well.
According to another feature of this device, the sleeve is secured to an element comprising a plurality of such sleeves.
This element can be a bar or a plate.
It may comprise lines of notching between the sleeves to allow their separation so as to use only part of the set of sleeves.
The invention also relates to a multi-well plate or multiple wells allowing, for example, cell culture or microtiteration of samples, characterized in that it is provided with a device meeting the above characteristics.
This plate can be covered by a plate having on one side as many sleeves as wells in the plate.
The invention also relates to the use of the above device or plate for measuring the optical density of a substance dissolved in this liquid.
It relates more particularly to the use of this device for the determination of substances contained in a cell culture medium, these substances having for example been produced by cells in culture, or having been released into the medium after solubilization of cells, those if it has been immunologically labeled.
But other features and advantages of the invention will appear better in the detailed description which follows and refers to the accompanying drawings, given solely by way of example, and in which
FIG. 1 is a perspective view of a sleeve inserted in a well
FIG. 2 is a perspective view of a bar provided with such sleeves
FIG. 3 is a perspective view similar to FIG. 2 but showing individually separable sleeves of the bar.
- Figure 4 is a sectional view of the bar of Figure 2, passing through the axis of the sleeves, and illustrated in mounted position on a row of wells; and
- Figure 5 is a partial perspective view of a multiwell plate ready to receive a plate having on one side sleeves according to the invention.
With particular reference to FIG. 1, it can be seen that a device according to the invention is, in general, constituted by a sleeve 1 that can be inserted in a cell or well 2 formed in an element 3 and that can contain a liquid in which any substance is dissolved.
The sleeve 1 is open at both ends to allow the passage of the light beam, and its wall 4 may have a variable thickness, which characterizes the caliber of the sleeve 1.
Thus, as is easily understood, the sleeve 1, once introduced into the well 2, will reduce the volume of this well as a function of the thickness of the wall 4 and therefore allow the rise of the liquid in the well 2 of the desired height according to the caliber used.
The sleeve 1, as well as the element 3 may be of any material such as for example a transparent plastic disposable or reusable from one measurement to another after cleaning.
Thus, a light beam passing through the well and the liquid that it contains, preferably in a direction substantially parallel to the well axis, which is generally substantially cylindrical, will make it possible to measure the optical density of the substance dissolved in the liquid of which the height in the well will be large enough to provide the desired sensitivity, accuracy and efficiency of the measurement.
As seen in Figures 2 and 3, the sleeve 1 may be secured to a support 5 having a plurality of such sleeves projecting from one side of said support.
As can be seen in FIG. 2, the support 5 may consist of a bar carrying a plurality of sleeves 1 whose spacing will correspond to the spacing between the wells 2 of a well line formed in the element 3.
The bar-shaped support 5 may comprise score lines 6 between the sleeves 1, which makes it easy to separate them from each other for insertion into a particular well formed in the element 3.
FIG. 4 shows a linear assembly of sleeves 1 belonging to a bar 5, as shown in FIG. 2, which line of sleeves is inserted into the corresponding spacing wells 2 formed in the element 3.
The sleeves 1 may also be integral with a support in the form of a plate or plate 7, as can be seen in FIG. 5, where said sleeves 1 project from the same face of the plate 7.
This plate 7 with associated sleeves may include as many sleeves 1 as cells or well 2, provided in a multi-well plate P for example cell culture, microtitre samples, dosing operations or even other uses.
It goes without saying that sleeves 1 can be used in only certain wells 2 of the plate P, for example by means of a bar 5 with breakable or non-breakable sleeves, as can be seen in FIGS. .
Of course, the invention is not limited to the embodiment described and illustrated which has been given by way of example.
Thus, the material and the wall thickness of the sleeves 1 may be any, as well as the geometric position of the sleeves on a plate, a bar or any other support.
This means that the invention includes all the technical equivalents of the means described and their combinations if they are carried out according to its spirit.
