L'invention concerne une composition ayant des propriétés vaccinantes contre le paludisme. Elle comprend trois constituants, notamment l'antigène "i72", le polypeptide R23 et le polypeptide EB200.
Description
MELANGE DE CONSTITUANTS PEPTIDIQUES AYANT DES PROPRIETES VACCINANTES CONTRE LE PALUDISME, NOTAMMENT DU A PLASMODIUM FALCIPARUM
L'invention concerne un mélange de 3 constituants peptidigues ayant des propriétés vaccinantes contre le paludisme, notamment du A Plasmodium Falciparum. plus particulièrement la capacité d'induire in vivo une réponse immunitaire protectrice, y compris la production d'anticorps immunoprotecteurs aptes à détruire les parasites du stade sanguin, responsables de la maladie chez l'homme, dans un modèle de primate représenté par le singe écureuil SAIMIRI SCIUREUS.
Ces trois constituants sont définis dans ce qui suit comme "premier constituant", "deuxième constituant" et "troisième constituant".
Le "premier constituant" est constitué par un polypeptide dérivé d'un antigène majeur de P falciparum, ayant un poids moléculaire de l'ordre de 72 kDA, d'où l'appellation "Antigène 72 DA" utilisée dans ce gui suit pour le désigner, particulièrement d'un polypeptide issu de cet antigène 72 kDA ayant la capacité, comme celui-ci :
- d'être reconnu par des anticorps protecteurs contenus dans des sérums ou des fractions immunoglobulinigues provenant d'animaux, notamment de singes SAIMIRI SCIUREUS, devenus résistants aux parasites après les infections répétées contrôlées par chimiothérapie, les dits sérums ou fractions étant susceptibles de rendre résistants des singes initialement sensibles ;
- d'être lui-même inducteur, notamment chez le singe, et plus particulièrement chez le singe SAIMIRI SCIUREUS, d'anticorps actifs contre des parasites du paludisme, plus particulièrement de P falciparu ou des parasites qui présentent les mêmes caractéristiques biologiques essentielles.
Ce premier constituant est lui-même souvent reconnu par des sérums de patients humains habitant des régions où le paludisme sévit de façon endémigue.
La séguence nucléotidigué codant pour la séguence peptidigue antigène 72 kDA a été identifiée par L.S. MATTEI et al Européen Journal of Immunology (1989) 19 : 1823 - 1828. Le premier constituant est avantageusement constitué par un polypeptide ci-après désigné "i72" comprenant les 153 acides aminés de la région C - terminale de l'antigène 72 kDA, dont la séguence en acides aminés, de même qu'une séquence d'acide nucléique codant pour le polypeptide i72 sont présentées ci-après :

constitue l 'un des "troisièmes constituants" qui peuvent être utilisés de façon avantageuse dans le mélange vaccinant selon l ' invention. Un peptide recombinant de fusion contenant cette séquence ("constituant 3" dans l ' exemple qui suit) a, comme les "constituant 1" et "constituant 2" , été produit en mettant en oeuvre la technique décrite par D. B. Smith et K.S. Johnson. Une culture de E. Coli (Escherichia coli DH 5 ) contenant le plasmide d ' expression correspondant a été déposée le 24 Juillet 1991 à la CNCM sous le n' I - 1128.
La capacité qu ' a le singe écureuil SAIMIRI SCIUREUS de produire des anticorps actifs contre des P falciparu conduit à la solide présumption que tous les antigènes dont il est question dans la présente de brevet sont également inducteurs d' anticorps immunoprotecteurs chez l 'homme, dès lors qu ' ils lui seraient administrés , par analogie avec des travaux

antérieurs faisant état de la similitude entre les anticorps contre les antigènes du parasite P falciparum présents dans les sérums de populations vivant dans des régions où le paludisme sévit de façon endémique d'une part, et les anticorps induits par les sèmes antigènes chez le singe écureuil Saimini Sciureus.
D'une façon générale, le mélange vaccinant de l'invention contient des proportions de ces trois constituants qui peuvent être ajustées par le spécialiste, selon leurs activités individuelles, mesurables par exemple dans un test du type de celui dont la description sera donnée dans l'exemple. A titre purement indicatif ces proportions pourront, pour chacun des trois constituants correspondre à une valeur comprise entre 5% et 80%, notamment entre 20% et 80% en moles du mélange, étant naturellement entendu que le total des trois proportions choisies ne saurait excéder 100 -t en moles. Les polypeptides responsables des activités relatives des trois constituants peuvent être utilisés soit seuls, en particulier lorsqu'ils présentent eux- mêmes une activité immunogène suffisante, le cas échéant en présence d'un adjuvant immunologique approprié, par exemple du type muramylpeptide ou analogue, soit sous formes oligomérisées, dans le cas où une telle oligo érisation renforcerait l'induction d'une réponse immunitaire, soit encore au sein de protéines de fusion (comme dans l'exemple qui suit), soit enfin à l'état conjugué à des protéines porteuses. Des techniques pour produire de tels conjugés sont décrites, par exemple, dans la demande de brevet français 89 04847/2 645 877 déposée le 12 Octobre 1989. L'invention concerne également des protéines de fusion dans lesquelles des séquences peptidiques respectivement issues des "premier", "deuxième" et "troisième" constituants seraient incluses dans une seule et même protéine, ces séquences peptidigues étant
liées les unes aux autres, soit bout à bout, soit par l'intermédiaire d'éléments peptidigues de pontage.
Chacune de ces séquences peptidiques est réduite à la taille nécessaire lui permettant de contribuer à l'induction d'anticorps reconnaissant le constituant auquel elle correspond, lorsque cette protéine de fusion est administrée à un hôte approprié.
Par exemple, la protéine de fusion comprendrait des séquences liées bout à bout respectivement issues du polypeptide "i72", du polypeptide EB 200 et du polypeptide R23.
Les séquences des deux derniers peptides mentionnés sont le cas échéant limitées à des fragments contenant chacun 3 à 4 motifs des séquences répétitives dégénérés que possèdent respectivement EB200 et R 23.
De telles protéines fusionnant les 3 constituants peuvent être produites par des techniques de génie génétique, notamment par transformation d'un hôte cellulaire compétent par un vecteur modifié par un acide nucléique contenant lui-même des séquences nucléotidiques successives codant pour les séquences polypeptidiques correspondantes dans une phase de lecture commune, sous le contrôle d'un promoteur reconnu par les polymérases de l'hôte cellulaire.
L'invention concerne naturellement toutes compositions de vaccins dans lesquelles le susdit mélange de polypeptides se trouverait associé à des véhicules pharmaceutiquement acceptables facilitant leur administration à l'homme.
Les résultats qui suivent, obtenus chez le singe, n'en sont pas moins hautement significatifs du caractère vaccinant potentiel du mélange concerné chez l'homme. Lors d'une première expérience, ce mélange contenait les "constituant 1", "constituant 2" et "constituant 3" dans les proportions molaires suivantes ( ëlcrige I) :
Les expériences d'immunisation de singes Saimiri avec ce mélange d'antigènes recombinants de Plasmodium falciparum ont été conduites comme suit : Animaux : 16 singes Saimiri sciureus d'élevage (FI nés en captivité), phénotype Guyana K14-7, de 1 À 2 années d'âge : 1 animal donneur de parasites et 15 animaux répartis en 5 groupes expérimentaux. Les méthodes expérimentales de manipulation et d'infection des singes Saimiri ont été ceux décrits par Gysin et al (J. Parasitoi., 66 : 1003-1009, 1980) et Gysin & Fandeur (Amer. J. Trop. Med. & Hyg. 32 : 461-467, 1983).
Souche de Plasmodium falciparum : la souche de parasites utilisés a été la souche Palo Alto (FUP-1) adaptée au singe Saimiri en 1979 (Gysin et al J. Parasitol. 66: 1003-1009, 1980). Identification des groupes d'animaux d'expérience : les animaux utilisés dans l'expérience proviennent de l'élevage de Saimiri sciureus de l'Institut Pasteur de la Guyane française, appartenant au phénotype Guyana de karyotype K14-7. Tous sont nés en captivité (génération FI) et avaient entre 1 et 2 ans d'âge en mars 1991. La distribution des animaux dans les groupes a été faite par tirage- au-sort à partir d'un groupe pool initial de 15 singes. Les numéros d'identification de chaque singe ont été ceux affectés à la naissance et correspondent aux fiches individuelles dans le fichier établi dans le programme informatisé de l'unité de primatologie de l'Institut Pasteur de la Guyane Française.
Groupe I - Immunisation avec le mélange de la série d'antigènes de Pasteur : i72pGEx, EB200, R23, dorénavant appelés molécules recombinantes Pasteur. Singes 89053, 89023, 89080.
Groupe II - Contrôles non immunisés. Singes n* 89126, 89043, 90001.
Immunisations : les animaux ont été immunisés l'état intact (non splénectomisés). Tous les antigène ont été utilisés dans la dose de 100 μg de protéin fusion par injection émulsionnée dans 1 ml d'adjuvan PAO (polyalofine) de Behringwerke, injecté dans plusieurs points par voie sous-cutanée dans la régio dorsale de l'animal.
Nombre d'injections : chaque animal a reçu 4 doses vaccinantes aux jours : 0 (zéro) , 21, 42 et 75 de 1'expérience.
Evaluation de la protection immune. Tous les animaux ont été splénectomisés le jour 85 de l'expérience, soit 10 jours après la dernière dose immunisante. Au jour 90, les animaux ont été soumis à une inoculation d'épreuve avec une dose de 100,000 globules rouges parasités par Plasmodium falciparum, souche Palo-Alto, provenant d'un anima 1 donneur (Saimiri splénoctomisé 10 jours avant l'infection) infecté avec l'échantillon d'un stabilat congelé de parasites (P. faciparu , souche Palo-Alto, FUP-1) , une semaine auparavant.
La figure l illustre les résultats obtenus concernant l'évolution des parasitémies (P sur l'axe des ordonnées) en fonction du nombre de jours (J) suivant les inoculations d'épreuve chez les singes vaccinés et chez les singes contrôle.
Les courbes (a) relatives aux animaux du groupe II témoignent d'un développement rapide de la parasitémie dans les 4 à 6 jours suivant l'inoculation des animaux avec les globules rouges parasités obligeant l'expérimentateur à introduire un traitement chi iothérapique au 7ème jour après l'infection .
Au contraire les courbes (b) relatives aux animaux du groupe I font apparaître une guérison complète de 2 des animaux vaccinés, et pour le troisième un retard à
la parasitémie qui ne se développe qu'à compter du lOème jour.
Il est intéressant de remarquer que les trois courbes (b) y compris celle relative à l'animal chez lequel la parasitémie a tout de même fini par se développer, témoignent toutes d'un accroissement puis même d'une réduction de la paratisémie entre les jours 4et 6.
Compte tenu de la nature virulente des parasites utilisés dans les innoculations d'épreuve et des résultats moins favorables obtenus à ce jour avec d'autres peptides immunogènes, la guérison totale chez deux singes vaccinés et le retard dans le développement de la parasitémie chez le troisième doivent être dans tous les cas interprétés comme le résultat d'un effet vaccinant réel de la composition selon l'invention.
Une deuxième expérience a été réalisée chez le singe Saimiri sciureus, suivant le même protocole d'immunisation que pour l'expérience précédente, sauf que les singes n'ont pas été splénectomisés. La composition molaire du mélange utilisé est la suivante :
"Constituant 1" : 46%
"Constituant 2" : 13% (fusion dans le pGEX) II "Constituant 3" : 41%
Trois singes ont été immunisés avec le mélange de protéines recombinantes (Constituant l≈PfEB-pGEX ; Constituant 2=R23-pGEX) contenant lOOμg de chaque antigène en présence d'adjuvant PAO (Singes 89084, mort entre les jours 14 et 21, 89077 et 89021) et deux singes contrôles ont reçu seulement l'adjuvant (Singes 89061 et 89095). Les singes ont reçu 4 injections aux jours 0, 21, 42 et 75,suivies d'une injection de 106 parasites, P. falciparum souche Palo Alto, au jour 89.
Les parasité ies ont été suivies pendant 27 jours et tous les singes ont été traités au jour 26 après le challenge. Les résultats sont présentés dans la figure 1. Le singe 89021 a présenté un taux de parasitémie maximum de 0,48% au jour 16 après le challenge. Les trois autres singes, 89077, 89061 et 89095, ont présenté des parasitémies de 2.3, 2.5 et 2.6%, respectivement, aux jours 15-16. Ces résultats témoignent donc d'une baisse importante de la paratisémie aussi chez les animaux non splénectomisés.
L'invention concerne naturellement aussi les mélanges d'anticorps spécifiques qui peuvent être obtenus à partir de constituants respectifs des mélanges susdits de polypeptides. Ces mélanges d'anticorps peuvent être administrés in vivo, notamment dans le but d'interférer avec une paratisémie due à une infection par un parasite humain, notamment du type P. falciparum.
Enfin, l'invention étend de même ses effets à des mélanges des séquences nucléiques dans lesquelles les différentes séquences nucléiques correspondantes auront été recombinées entre elles, dans des conditions autorisant leur expression sous la forme d'une protéine de fusion comprenant des séquences peptidiques correspondant aux séquences des trois constituants (entiers ou partiels) précédemment pris en considération.
Claims
1. Mélange vaccinant contre la paludisme comprenant trois constituants définis dans ce qui suit comme "premier constituant", "deuxième constituant" et "troisième constituant", respectivement caractérisés en ce que :
- le "premier constituant" est constitué par un polypeptide dérivé d'un antigène majeur de P falciparum, ayant un poids moléculaire de l'ordre de 72 kDA, d'où l'appellation "Antigène 72 kDA" utilisée dans ce qui suit pour le désigner, particulièrement d'un polypeptide issu de cet antigène 72 kDA ayant la capacité, comme celui-ci :
* d'être reconnu par des anticorps protecteurs contenus dans des sérums ou des fraction.-. immunoglobuliniques provenant d'animaux, notamment de singes SAIMIRI SCIUREUS, devenus résistants aux parasites après les infections répétées contrôlées par chimiothérapie, les dits sérums ou fractions étant susceptibles de rendre résistants des singes initialement sensibles ;
* d'être lui-même inducteur, notamment chez le singe, et plus particulièrement chez le singe SAIMIRI SCIUREUS, d'anticorps actifs contre des parasites du paludisme, plus particulièrement de P falciparum ou des parasites qui présentent les mêmes caractéristiques biologiques essentielles. ou par une protéine de fusion contenant la précédente séquence peptidique.

Mélange selon la revendication 1 ou la revendication 2, caractérisé en ce que la proportion de chacun des constituants est en une proportion comprise entre 5% et 80%, notamment entre 20 et 80% en moles de mélange, étant naturellement entendu gue le total des trois proportions choisies ne saurait excéder 100 % en moles.
4. Polypeptide de fusion incluant en une seule molécule des séquences correspondant à celles des trois constituants du mélange selon la revendication 1 ou la revendication 2.
5. Mélanges d'anticorps spécifigues gui peuvent être obtenus à partir des constituants respectifs des mélanges de polypeptides selon la revendication 1 ou la revendication 2.
6. Composition immunogène contenant les constituants des compositions des revendications 1 à 4,
FEUILLE DE REMPLACEMENT associées à un véhicule pharmaceutiguement acceptable facilitant leur administration in vivo sous forme de vaccin.
7. Fragments d'acides nucléiques recombinés ou non avec des séquences d'ADN hétérologues, caractérisés en ce gu'ils codent pour les polypeptides des constituants des mélanges selon la revendication 1 ou la revendication 2 ou pour le polypeptide de fusion de la revendication 4.
