最近 、 後天性免疫不全症候群 ( エイ ズ ) の発症、 蔓延 が世界的 に玆慮さ れて お り 、 そ の予防 、 治療対策が緊急 課题 と な っ て い る 。
数年前 よ り 、 エ イ ズや 成人 T細 胞 白 血病 な ど の 原 因 ウ ィ ルス と 考え ら れる種々 の レ ト ロ ウ イ ルス が単離、 周 定され、 その逍伝子の塩基配列が決定さ れた 。 そ の結果 こ れ ら は周類ま た は周一 で あ る と報告さ れる よ う に な つ た 。 特に エイ ズの病原ウ ィ ルス と さ れた H T L V— 1Π、 L A V、 A R Vは同 じ ウ ィ ルスであ り 、 H I V ( human immunodeficiency virus) と統一 し て 命名さ れた 。

本発明 に おいて 「 e n Vタ ンノ ク 質 」 と は 、 レ 卜 ロ ウ ィ ルス の感染、 病原性を司 る envタ ンパ ク 質及びそ の変異 物や改変物 をいい 、 感染へ の影饔力 、 病原性を失 っ た envタ ンパク 質の改変物等を意味 し な い 。 本発明 に おい て 「 env遣伝子を含 ま な い 」 と は 、 envタ ン ノ ク 質 を コ ー ド し て い る遺伝子を発現 し得る形で含 ま な い こ と を 意味 し 、 レ 卜 ロ ウ ィ ル ス の感染、 病原性に 関係の な い env遺伝子由 来の遗伝子 ( env遺伝子の一部や env遺伝
子の改変物等 ) を含んでい て も構わ ない 。 W 08 8 Z 0 9 6 7 0で開示さ れた gp1 6 0及び 9ΡΊ 2 0を含 ま ない レ 卜 ロ ウ ィ ルス粒子は 9P1 2 0がないた め 内包 す る遺伝 子が細胞内 に取 り 込 ま れる可能性が少ない が 、 万が一、 内 包 す る遺伝子が細胞 内 に 取 り 込 ま れ た 場 合 、 レ ト ロ ウ ィ ルス の全遺伝子を有 し ている た め; レ 卜 ロ ウ ィ ルス を構築 し て し ま う 危険性が あ るが、 本発明 の粒子 は env 遺伝子を含んでいない ので 、 そ の よ う な危険性 は な い 。 し か し 、 本発明 に おい て も 、 env遣伝子の改変物 を含ん でいる場合、 変異な ど に よ り 病原性を回復する可 能性が あ る こ と か ら 、 env遛伝子の改変物 も含ん で い ない こ と が好 ま し い 。
本発 明 に 用 い る レ ト ロ ウ イ ル ス は 、 ウ ィ ル ス ♦ べ ク タ ー を感染さ せ る動物培養細胞 に おい て 、 gagタ ンパク 質 、 及び 又 は polタ ンパク質が発現で き 、 env遗伝子 を含 ま な いもので あれば特に 限定 さ れない 。 例 え ば、 才 ン コ ウ ィ ルス亜科ま た は 、 レ ンチウ ィ ルス亜科 に 属す る もの ウ ィ ルス、 特に 、 H I V— I 、 H I V— I、
H T L V— I 、 H T L V— Iが挙げ ら れる 。
本発明 に おい て 、 ウ ィ ルス ♦ ベ ク タ ー に 組み込 ま れる レ ト ロ ウ イ ルス の c D N Aは 、 ウ ィ ルス ♦ ベ ク タ ー を感 染さ せ る動物培養細胞 に おいて 、 gagタ ンパ ク 質、 及び ま た は polタ ンパ ク 質が発現でき 、 env遺伝子を含 ま な い も の で あ れ ば 、 特 に 限定 さ れ な い 。 例 え ば 、 ト1 1 ー 1 の gag遺伝子及び pol遺伝子を コ ー ド し 、

本発明 に 用 い る ウ イ ルス · ベ ク タ 一 ち動物細胞に 感染 し 、 gag遗伝子 、 及び po I遺伝子 を発現で き る ち のであ れば、 特 に 限定 さ れない 。 例 え ぱ、 ワ ク チニ ァ ♦ ウ ィ ル ス 、 ア ビボ ッ ク ス ♦ ゥ イ ノレス 、 バキ ュ 口 , ゥ ィ ルス な ど が あ げ ら れる 。
本発明 に用 いる gag遗伝子、 及び ま た は pol遺伝子 を組み込ん だゥ ィ ルス ♦ ベ ク タ 一 の構築方 法も 、 特に 限 定さ れない 。 例 えば 、 ウ ィ ルス ♦ ベ ク タ ー と し て ワ ク チ 二 ァ · ウ ィ ルス を用 い る場合、 組み込む遺伝子を ョ 本脳 炎ウ ィ ルス の表面抗原蛋 白質を コ ー ド する 遺伝子か ら レ 卜 ロ ウ ィ ルス の gag遺伝子、 及び zま た は pol遺伝子 に 代え る こ と 、 用 いる ワ ク チニ ァ ♦ ウ ィ ルス が必ず し も好 ま し い条件を 篛た す必要の無い こ と 以外 は特開平 1 一 7

次いで 、 構築さ れた gag遺伝子及び pol遺伝子を コ ー ドす る c D N A断片が組み込 ま れた ウ ィ ルス · ベ ク タ ー を 、 ま た は gag遺伝子を コ ー ド する c D N A 断片が組 み 込 ま れ た ウ ィ ルス ' ベ ク タ ー と po I遺伝子を コ ー ド する c D N A 断片が耝み込 ま れた ゥ ィ ルス ♦ ベク タ ー を 、 動 物培養細胞 に 感染さ せ 、 そ の培養上清中 に 開裂型の gag タ ンパ ク 質 、 及び polタ ンノ ク 質を発現させる 。
本発明で用 い る動物培養細胞 は ウ ィ ルス · ベ ク タ ー が 増殖可能なも ので あれば よ く 、 そ の具体例 と し て 、 ウ イ ルス · ベ ク タ ー が ワ ク チニ ァ ♦ ウ ィ ルスで あ る場合は 、 例 え ば T K - 4 3 ( ヒ 卜 骨肉腫由 来 ) 、 F L ( ヒ 卜 羊 膜由 来 ) 、 Hela ( ヒ 卜 子宮頸部癌由 来 ) 、 K B ' ( ヒ 卜 ^ 咽喉癌由 来 ) 、 R K 1 3 ( ゥサ ギ腎由 来 ) 、 C V — 1 ( サル腎由 来 ) 、 B S C — 1 ( サル腎由 来 ) 、 L 9 2 9 ( マ ゥ ス 結合組織由 来 ) 、 C : ( ニ ヮ 卜 リ 胚 ) 細 胞 、 D E F ( 二 ヮ 卜 リ 胎児繊維芽細胞 ) 、 S W 4 8 0 ( ヒ 卜 結腸ガ ン由来 ) な どが例示さ れる 。 ウ ィ ルス , ベ ク タ ー が ア ビボ ッ ク ス · ウ ィ ルス で あ る場合 は、 本発明で用 い る勤物培養細胞 と し て は鶏胚用 繊雜芽細胞な ど の鶏由 来 細胞な ど が例示さ れる が 、 発育鶏卵漿屎膜な ど も含む。 ウ ィ ルス · ベ ク タ 一 がバキ ュ 口 • ウ ィ ルス の場合 は 、 例 え ばス ポ ドプテ ラ ♦ フ ルギぺルダ ( Spodoptera f u rug i erda ) 由 来の S f 9 株な ど が例示さ れる 。
し か し 、 一般的な培養条件 に お い て は 、 用 い る細胞 に よ っ て は 開裂型 gagタ ンパ ク 質の発現量が低く 、 例 えば C V - 1 に H I V— Iの遣伝子を組み込ん だ ワ ク チニ ァ ♦ ウ ィ ルス を感染さ せ た 場合 に は 、 開裂型 gagタ ンパ ク 質 は ほ と ん ど発現が認め ら れな い 。 開裂型 gagタ ンパ ク 質の発現量が多い ウ ィ ルス ♦ ベ ク タ ー と動物培養細胞の 組み合せが好 ま し く 、 その よ う な例 と し て は H I V - I の遺伝子を組み込ん だ ワ ク チニ ァ ウ ィ ルス を S W 4 8 0 に感染 さ せ も の が あ る 。
こ こ で発現さ れる g a gタ ンパ ク 質の 開裂 は p 1 0な ど プ ロ テ ア ーゼ活性を有する polタ ンパ ク 質 に よ る も の と 考え ら れる 。
培養上清か ら の開裂型 gagタ ンパ ク 質の精製 は特 に 限 定 さ れない 。 例 えば、 常法 ( サイ エ ン ス ( Science ) 、 2 2 4 、 4 9 7 — 5 0 0、 ( 1 9 8 4 ) ) に従 っ て 、 上 清を シ ョ 糖 、 あ る い は酒石酸 カ リ ウ ム密度勾配遠心を行 い 、 更 に セ シ ウ ム ク ロ ラ イ ド沈降平衡遠心を行え ば よ い 開裂型 gagタ ンパ ク 質 は粒子を形成 し て 、 回収さ れる 。
本発明 の 開裂型 gagタ ンパ ク 質に よ り 構成さ れる粒子 は 、 例 え ば、 レ ト ロ ウ イ ルス が H I V— Iで 、 比重 よ り 分画 し た 場合 、 比重が H I V— I粒子 と 周 じ 約
1 . 1 5の フ ラ ク シ ョ ン に 認 め ら れる 。
精製の各段階 に おい て ど の フ ラ ク シ ョ ン 中 に 粒 " f が存 在 す る か の確認の方法 は特 に 限定 さ れな い 。 例 え ば 、 gagタ ンパ ク 質に対す る 抗体を使 う 方法や逆転写 酵素活
性を測定 す る方法がある 。 本発明 の 開裂型 gagタ ンパ ク 質に よ り 構成さ れる粒子 は逆転写酵素を 内包 し て い て も 内包 し て いな く て も か ま わないが 、 通常、 少な く と も一 部 は逆転写酵素を内包する 。 従 っ て 、 逆転写酵素活性を 測定すれば、 本発明 の粒子を含む フ ラ ク シ ョ ン は判別で ぎる。
逆転写酵素活性の測定 は常法 に従え ば よ い 。 例 え ば 、 微生物学実習提要 ( 東京大学医科学研究所学友会編 ) の 記述 に準 じ て組み換え ウ ィ ルス感染細胞上清を Po l y-Aを 鍩型、 ol igo-dTをプラ イ マ 一 と し て 、 "2 P — d T T P の 酸不溶性画分への取 り 込みを測定すれば よ い 。
本発 明 の粒子は 、 ウ ィ ルス ' ベ ク タ ー を感染さ せ た 動 物培養細胞の生体膜由 来の脂質二重膜 と考 え ら れる ェ ン ベ ロ ープ様のもの を伴 っ て いる 。 こ の粒子 は 、 脂質二重 膜を有 し た ま ま でも抗原 と し て 機能するが 、 脂質二重膜 がある こ と が好 ま し く な い場合は 、 タ ンパク 質が失活 し ない条件下でのエ ー テル等の有機溶媒処理や 、 凍結融解 処理に よ り 破碎 し 、 除去す る こ と がで きる 。
こ の粒子 は細胞への感染を司 る envタ ンパク 質及びそ の遣伝子も含 ま ないので、 病原性を示さ ない 。 そ の他の 遺伝子 は含んで い て も含んで いな く て も構わな い 。 周様 に 、 開裂型 gagタ ンパ ク 質に よ り そ の構造が形成さ れて いれば、 envタ ンパ ク 質以外の タ ンパ ク 質 を含んで い て も 含んで いな く て も構わ ない 。 ま た 、 エ ンベ ロ ー プ様の 脂質二重膜の有無を 問わ な い 。 こ の粒子 は抗原性を有 し
さ ら に そ の抗原性 は envタ ンパク 質 に 比べ て 変異が少な い と 考え ら れる gagタ ンパ ク 質 、 ま た は内包さ れて いる 場合 に は po Iタ ンパク質お よ び gagタ ンパ ク 質に 由 来す るも のであ る 。 さ ら に 、 自然界の ウ ィ ルス粒子の一部 と 周一ま た は類似の構造を有 し て い る こ と か ら 、 gagタ ン パク 質、 po Iタ ンパ ク 質 、 あ る い はそ の単な る混合物以 上の抗原性も期待で き る 。
レ 卜 □ ウ ィ ル ス は エ イ ズ 、 成 人 T 細 胞 白 血病 、 癌 、 リ ュ ー マ チ等の 多 く の難病の病原 ウ ィ ルス で あ る と 確認 ま た は推定 さ れて お り 、 レ 卜 ロ ウ ィ ルス が原因で起 こ る 病気の多 く は効 果の あ る ワ ク チン は知 ら れて い な か っ た が 、 本発明 の レ ト ロ ウ イ ルス の コ ア と 同一 ま た は類似の 構造を有 し て い る粒子 は優れ た 効 果が期待で き る 。
か く し て 本発明 に よ れば 、 レ ト ロ ウ イ ルス 由 来の gag タ ンパ ク 質を コ ー ド す る c D N A 及び po Iタ ンパ ク 質を コ ー ドす る c D N A を ウ ィ ルスの増殖 に非必須なゲノ ム 領域に 組み込ん だウ ィ ルス · ベ ク タ 一 を感染さ せ た 細胞 を培養 し 、 開裂型の gagタ ンパ ク 質 に よ り 構成さ れ 、 envタ ンパ ク 質及び env遺伝子を含 ま ない レ ト ロ ウ ィ ル スの コ ア構造物 が得 ら れる 。 こ の構造物 は 、 レ 卜 ロ ウ ィ ルス粒子の構造の一部 と 同一 の構造を持つ が感染性を持 た な い こ と か ら 、 こ れを有効成分 と す る こ と に よ り 、 有 効な レ 卜 ロ ウ ィ ルス · ワ ク チ ン が得 ら れる こ と が期待で ぎる 。
( 実施例
以下実施例を挙げて本発明 を更 に 具体的 に説明 する 。 実施例 1
(1) 組み換えベ ク タ ー P U H Kの作製 ( 第 1 図参照 ) p U C 9 ( フ ア ルマ シ ア社製 〉 を E c o R I 、 及び P s t Iで消化後 、 ポ リ メ ラ ーゼで処理 し 、 切 断端を平 滑末端 と し 、 連結 し 、 E c o R I部位-の欠損 し た
P U C 9を作製 し た 。 E c o R I部位欠損 P U C 9を H i n d IEで 切 断後、 ワ ク チニ ァ ウ ィ ルス T K遺伝子を 含むワ ク チニ ァ ウ ィ ルス W R株 の H i n d DI J 断片 に連 結 し 、 組み換え べ ク タ一 p U Η Kを得 た 。
(2) 組み換え ベ ク タ ー Ρ Ν Ζ 6 8 Κ 2の作製 ( 第 2 図参照 )
ワ ク チニ ァ ウ ィ ルス W R株の 7 . 5 Κプ ロ モ ー タ ー ( モス ら ( Hoss et. al ) 、 セル ( Cel l ) 、 1 2 5、 8 0 5 — 8 1 3、 ( Ί 9 8 1 ) ) を P U C 1 9 ( フ ア ルマ シ ァ社製 ) の H i n c 部位に挿入 し た 後 、 H i n d I と E c o R I で頫次 ¾ 断 し て 、 7 . 5 Kプ ロ モ ー タ 一 の 前後 に ポ リ リ ン カ 一部位を有す る D N A断片 ( 約 3 5 0 bp) を得た 。
こ の D N A断片の ヌ ク レ ア ーゼ処理 し た も の 、 及 び ( 1 )で得たプラ ス ミ ド P U H Kの E c 0 R I消化物 を ポ リ メ ラ 一ゼ処理 し た も の を連結 し 、 得 ら れた組み換え プラ ス ミ ド を 0 N Z 6 8 K 2 と 命名 し た 。
(3) 組 み換えプラ ス ミ ド gag の作製 ( 第 3図参 照 )
H I V - I の全 c D N Aを含 む P N L 4 3 を B g ^ I と E c o R ェ で 消化後、 低融点 ァ ガ ロ ー ス電気泳動で約 5 . Ί kbp の断片を分離 し 、 フ エ ノ ール抽 出 に よ り そ の 断片を 回収 し た
こ の 断片中 に は 、 第 3 図 に 示す よ う に 、 gag遺伝子 、 及び pol遺伝子領域が含 ま れて い る 。
次 に 、 (2-)で得た 0 N Z 6 8 K 2を B a m H I と E c 0 R ェ で 消化 し 、 先 に 調製 し た gag遺伝 子; 及び pol遺伝子を含む D N A断片 と ラ イ ゲー シ ヨ ン し て 組み 換えプラ ス ミ ド W- gag を得た 。
(4) 組み換え ワ ク チニ ァ ウ ィ ルス の作出 ( 第 3 図参 照 〉

( T K " ) "1 4 3細胞 に上記プラ ー ク 形成 ウ ィ ルス を接 種 し 、 4 5分後、 1 %寒天 · 1 2 %牛胎児血清 ♦ 2 5 ^ 2 ^ B U d R加 Engle M E M培地を積層 し 、 3 日 間 培養後、 感染細胞を 0 . 0 1 %ニ ュ ー 卜 ラル ♦ レ ッ ドで 染色 し た 。 形成さ れた プラ ー ク をパスツ ール ピぺ ッ 卜 で 単離 し 、 Eagle M E M培地 中 に希釈す る 。 単離 し た ブラ ー ク にっ き 、 再度同様の操作を緩 り 返 し 、 プラ ー ク を純 化 し た 。 純化 し た プラ ー ク よ り 得 ら れた ウ ィ ルス を増殖 させ 、 増殖 ウ ィ ルス よ り D N Aを調製 し 、 gag遺伝子 、 及び pol遺伝子を コ ー ド す る c D N Aをプ ロ ブ と す る ド ッ 卜 プ ロ ッ ト ハイ プ リ ダイ ゼ ー シ ヨ ン に よ り 、 得 ら れ た組み換え ワ ク チニ ァ ウ ィ ルスが H I V— I 由 来の gag 遺伝子 、 及び pol遺伝子を コ ー ドす る D N Aをゲノ ム内 に持つ組み換え ウ ィ ルスで あ る こ と を確認 し 、 V — 2 0 一 1 — 8 と命名 し た 。
(6) 組み換え体の細胞上清で の発現
耝耩培養用 9 cnデ ィ ッ シ ュ に培養 し た S W 4 8 0細胞 に Bi.o. i. 1 0〜 2 0の V— 2 0 — 1 — 8 を接種 し 、 3 7 で 、 1 時簡感染させた後 、 ウ ィ ルス液を抜取 り 、 細胞を Eagle M E Mで洗净 し 、 5 %牛胎児血清加 Eag I e M E M を加えた 。 1 6時間後 、 細胞上清を回収 し 、 1 0 IDH卜 リ ス ♦ 塩歷 ( PH7 . 4 ) - 0 . 1 M N a C - 1 ηιΗ
E D T A溶液で作製 し た 2 0〜 6 0 % の酒石酸 カ リ ウ ム の密度勾 配上 に 重層 し 、 ロ ー タ ー S W 4 7 ( ベ ッ クマ ン 製 ) で 3 5 , 0 0 0 rpm 、 1 6時間遠心 し た 。 遠心後 、
各 フ ラ ク シ ョ ン に 分け た 。

I M N a C ^ — I mHE D T A溶液 加 え 、 3 7 °Cで 1 時間反応さ せ た 。 反応中 に 、 二 卜 ロ セル ロ ー ス フ ィ ル タ ー に鉛筆で 2 間 隔の碁盤の 目 を書い て お き 、 反応終了後、 各 フ ラ ク シ ョ ン及び対照の反応液 1 0 ^ _£ を二 卜 ロ セル ロ ー ス フ ィ ル タ ー上の各区画 に スポ ッ 卜 し た 。 こ の フ ィ ル タ ー を l乾に よ り 完全 に 乾燥さ せ た後、 5 % 卜 リ ク ロ ロ 酢酸 一 5 %ピ ロ リ ン酸ナ ト リ ウ ム液 中で 1 5分間振盪 し な が ら 洗净 し 、 未反応の放射性前駆休を 除去 し た 。 続い て 5 % 卜 リ ク ロ ロ 醉酸 中で 5分、 そ し て 5 0 %エ タ ノ ー ル中で 5分洗净 し た 。 乾燥後、 碁盤の 目 にそ っ て各 区画 を切 り 放 し 、 シ ン チ レ ー シ ヨ ンバイ アル に入れて 、 ト ルエ ンシ ン チ レ ー タ ー 中 で 液休シ ンチ レ一 シ ヨ ン カ ウ ン タ 一 に よ り 取 り 込みを測定 し た 。 対照 の 3 倍以上の 取 り 込みを 陽性 と し 、 逆転写 酵素活性が あ る と 判断 し た 。
次いで 、 逆転写酵素活性が認め ら れ た フ ラ ク シ ョ ン を

逆転写酵素活性の検出 さ れる比重約 1 . 1 5 の分画を さ ら に セ シ ウ ム ク ロ ラ イ ド沈降平衡遠心 に よ り 精製 し 、 逆転写酵素活性の検出 さ れる比重約 1 . 1 5 の分画 に P 2 4 が確認さ れた 。
いわ ゆ るネガテ ィ ブ染色法 に よ り 、 上記の p 2 4 が確 認さ れた 分画を電子顕微鏡 に よ り 観察す る と ワ ク チニ ァ ウ ィ ルス と は異 なる直径約 1 1 0〜 Ί 5 0 nmの球形 の粒 子が認め ら れた 。

本発 明 に よ れば、 開 裂型の gagタ ンパク質 よ り 構成さ
れる レ ト ロ ウ イ ルスの コ ア構造物 が感染性をもつ 又は感 染性を発現さ せる envタ ンパ ク 質も env遺伝子を も 含 ま ず に 製造 さ れる こ と と な る ので 、 安全性の点で 問題が よ り 少 く な い レ ト ロ ウ イ ルス ♦ ワ ク チン が提供さ れる可 能 性が髙 く 、 産業上の利用 可 能性が高い 。
Claims
1. レ ト ロ ウ イ ルス に属 す る ウ ィ ルス の env遣伝子 を含 まず レ ト ロ ウ イ ルス に属する ウ ィ ルス の少な く と も gag遺伝子及び pol遺伝子を組み込ん だ ワ ク チニ ァ * ゥ ィ ルスを細胞 に感染させ、 ま た は レ ド ロ ウ ィ ルス に属す る ウ ィ ルス の env遣伝子を含 ま ず レ ト ロ—ウ ィ ルスに 属す る ウ ィ ルスの少な く と も gag遗伝子を組み込ん だ ヮ ク チ ニ ァ · ウ ィ ルス と レ 卜 ロ ウ ィ ル ス に 属 す る ウ ィ ル ス の envl伝子 を含 まず レ ト ロ ウ イ ルス に属する ウ ィ ルスの 少な く と も po I遺伝子を組み込ん だワ ク チニ ァ ♦ ウ ィ ル スを周一の細胞 に感染させ 、 感染させた 細胞を培養する こ と に よ る 、 envタ ンパク 質を含 ま ない レ 卜 ロ ウ ィ ルス ♦ コ ァ構造物粒子の製造方法 。

3. レ 卜 ロ ウ ィ ルス に 厲する ウ ィ ルス が H I V— I H I V— E、 H T L V— ェ 、 ま た は H T L V— 1で ある 請求項 2記載の レ ト ロ ウ イ ルス * コ ア構造物粒子 の製造 方法。
4. 細胞が ヒ 卜 結腸ガ ン由来で あ る S W 4 8 0細胞 で あ る 請求項 3 記載の レ 卜 ロ ウ ィ ルス ♦ コ ア構造物粒子 の製造方法。
5 . 請求項 1 、 2 、 3 、 ま た は 4 記載の レ ト ロ ウ イ ルス ♦ コ ア構造物粒子の製造方法 に よ り 製造さ れた レ 卜 ロ ウ ィ ルス ♦ コ ア構造物粒子 。
6 . 請求項 5 記載の レ 卜 ロ ウ ィ ルス ♦ コ ア 構造物粒 子を有効成分 と する抗 レ 卜 ロ ウ ィ ルス , ワ ク チ ン 。
