本発明において一般式 〔 Π — a 〕 で表わされる出発化合物 の一つである 6 , 7 — ジ ヒ ド ロ キ シ一 3 , 4 — ジ ヒ ド ロ イ ソ キノ リ ンの製造法については、 例えば Acta Chemica Scandi- navica 20巻 , 2755頁 ( 1966) に記載されている。
また、 イ ソキノ リ ン骨格の 6 , 7 —位にメ トキシ基を導入
した 6 , Ί 一 ジ メ トキシ一 3 , 4 —ジヒ ドロイ ソキノ リ ンは 鎮痙薬である阿片アル力ロイ ドのパパべリ ン、 パパベリ ノ一 ル及びパパべラルジン等の合成岀発原料として知られる 6 , 7 —ジメ トキシイ ソキノ リ ンの中間原料であり、 その製造法 こつ ヽて ま、 例え t Journa l' o f the Amer i can Chem i ca l Soc i e ty 79巻、 3773頁 (1957) に記載されている。
しかしながら、 一般式 〔 I 一 a 〕 及び 〔 I 一 b〕 並びに 〔 BI〕 で表わされる本発明化合物は、 これまで知られておら ず新規化合物であり、 また化合物 〔 I 一 a 〕 及び 〔 I — b〕 から前記ィ ソキノ リ ン誘導体を製造することは従来知られて いない。 発'明の開示
本発明の目的ば、 新規な 3 , 4 ージヒ ドロイ ソキノ リ ン誘 導体を提供するこ とにある。 更に詳細には、 イ ソキノ リ ン骨 格の 6位及びノ又は 7位側鎖にケィ素原子を導入した 3 , 4 ージヒ ドロイ ソキノ リ ン誘導体並びにィ ソキノ リ ン誘導体を 提供することにある。
本発明の他の目的は、 医薬及び農薬の中間原料、 特に広い 抗菌スぺク トルを有し極めて高い抗菌活性を有するセファ ロ スポリ ン誘導体の中間原料として有用な新規な 3 , 4 —ジヒ ドロイ ソキノ リ ン誘導体及び新規なィ ソキノ リ ン誘導体を提 供 "Tることにある。
- 本 明の更に他の目的は、 イ ソキノ リ ン誘導体の工業的に 有利な製造法を提供するこ とにある。

本発明における前記式 〔 1 ー 3 〕 又は 〔 1 — 1>〕 で表わさ- れる 3 , 4ージヒ ドロイ ソキノ リ ン誘導体は、 前記式 〔 E― a〕 又は 〔 Π— b〕 で表わされる化合物と前述した 1官能性 シリル化剤又は 2官能性シリ ル化剤と反応させることにより 得ることができるが、 その際反応溶媒を用いなくてもよいが. 一般には溶媒を用いることができる。 殊にシリ ル化荊を多量 に使用し、 溶媒として機能をもたせる場合には、 溶媒を使用 しな く てもよい。 溶媒を使用する場合、 溶媒としては、 反応 に悪影響を与えないものであればよいが、 望ま しく は活性水
素を有しない化合物が使用される。 特に非プロ ト ン系有機溶 媒が有利に使用される。 溶媒の具体例を示すと、 例えば、 ァ セ ト ニ ト リ ノレ、 N , N _ジメ チノレホルムア ミ ド、 N , N —ジ メ チルァセ トア ミ ド、 ジェチルェ一テル、 ジォキサン、 ジメ トキシェタ ン、 テ ト ラ ヒ ドロ フラ ン、 n ^キサン、 トノレエ ン、 キシレン、 ジク ロロメ ン、 ク ロ 口ホルムなどが示され る。
また本発明は、 塩基性化合物 1 、 非プ口 ト ン系ァ ミ ド化合物

あるいは第 3級ア ミ ン.を使用するこ とにより反応を促進する ことが可能である。 殊にこれらは溶媒又はその一部として使 用することもできる。 かかる化合物の例としては、 1 ーヒ ド ロキシベンゾ ト リ アゾール、 ト リ カプリ リ ルメ チルア ンモニ ゥムク ロ ラ イ ド、 Ν , Ν —ジメ チルホルムア ミ ド、 Ν , Ν— ジメ チルァセ トア ミ ド、 Ν —メ チルー 2 — ピロ リ ドン、 ト リ ェチルァ ミ ン、 ピリ ジン、 Ν , Ν —ジメ チルァ ニ リ ン、 Ν— メ チルモルホ リ ンなどを示すこ とができ る。
反応は、 通常一 20〜120 ての範囲の温度、 好ま し く は 0 〜 100 'Cの範囲の温度で実施され、 また圧力は減圧、 常圧、 加 圧のいずれでも差支えがな く一般には常圧 (大気圧) である。 反応時間は、 目的物の種類、 溶媒の種類、 反応促進剤を使用 する場合その種類、 反応温度などの条件により左右されるが 一般には数分乃至数十時間の範囲である。
前述の如く して本発明の反応を行ない得られた反応混合物 から、 目的物の 3 , 4 —ジヒ ドロイ ソキノ リ ン誘導体を分離 する方法は何等制限されるこ とな く 、 一般的な分離法が採用
され、 例えば、 蒸発、 蒸留、 抽出、 沈澱 · 結晶化、 吸着、 昇 華など、 あるいはこれらの組合せが用いられる。
本発明におけるィ ソキノ リ ン誘導体である前記式 ( ΠΙ〕 に おいて、 前記した 及び R z と同様に、 両者は同一もし く ば異なり置換基を有していてもよい炭素数 1〜 2 0 のアルキ ル基、 ァルケニル基及びアルコキシ基並びに炭素数 6〜 2 0 のァリール基から選ばれるものである。 これら複数の 又 は R 2 においても互いに異つていてもよい。
こ こでアルキル基としてば、 例えば、 メ チル、 ェチル、 n —プロ ビル、 i s o—フ ""口 ピ)レ、 n —ブチノレ、 sec—ブチル、 ter t—ブチノレ、 ペンチノレ、 へキ シノレ、 シク ロへキ シル、 ヘプ チノレ、 ォクチル、 ノ ニル、 デシル、 ト リ デシル、 へキサデシ ル、 ォク タデシル、 エイ コ シルなどの直鑌、 分岐妆も し く は 環状の炭素数 1〜 2 0、 好まし く は炭素数 1〜 1 0、 特に好 まし く は炭素数 1〜 6 のアルキル基が挙げられる。
アルケニル基としては例えば、 ビュル、 ァリ ル、 i s o—プ ロ ぺニノレ、 フ'テニスレ、 ぺ:ンテニゾレ、 キセ ニ レ、 シク ロ へキ セニクレ、 ォク テ二ノレ、 デセニノレ、 へキサデセニスレ、 エイ コ セ チルなどの直鎖、 分岐状もし く は環状の炭素数 1〜 2 0、 ¾ ま し く は炭素数 1 〜 1 0、 特に好ましく は炭素数 1〜 6のァ ルケニル基が挙げられる。
アルコキシ基としては、 例えば、 メ トキシ、 エ トキシ、 プ ロポキ シ、 i-so—プロボキ シ、 ブ ト キ シ、 s ec—ブ ト キ シ、 ter t—ブ ト キシ、 へキ シノレオキ シ、 シク ロへキシゾレオキ シ、 デシノレォキ シ、 へキサデシルォキ シ、 エイ コ ノレォキ シな
の直鎖、 分岐状もし く は環状の炭素数 1 〜 2 0、 好ま し く は 炭素数 1 〜 1 0、 特に好ま し く は炭素数 1 〜 6 のア ルコキ シ 基が挙げられる。
また、 ァ リ ール基としては、 例えば、 フヱニル、 ビフヱ二 リ ル、 ターフェニリ ノレ、 ナチフル、 ア ン ト リ ルなどの炭素数 6 〜 2 0、 好ま し く は炭素数 6 〜 1 5 のァ リ 一ル基が挙げら れ、 フエニルが特に好ま しい。
前記アルキル基、 アルケニル基、 アルコキ シ基又はァリー ル基は、 置換基を有していてもよ く その置換基の数は、 アル キル基、 アルケニル基、 アルコキ シ基又はァ リ ール基の種類. 炭素数などにより左右されるが、 通常 1 〜 5、 好ま し く は 1 〜 3である。 かかる置換基としては、 例えば塩素、 臭素、 ョ ゥ素、 弗素などのハロゲン原子 ; メ チル、 ェチル、 プロ ピル、 ブチルなどの低級アルキル基 ; メ トキシ、 エ トキ シ、 プロボ キ シ、 ブ トキシなどの低級ア ルコキ シ基 ; シァノ基 ; ニ ト ロ 基 ; ァセ トキ シ、 ベンゾィ ルォキ シなどのァ シルォキ シ基 ; メ トキ シカルボニル、 フエノ キ シカルボ二ルなどのァルコキ シカルボニル基 ; フエニル、 メ トキ シフエ二ノレ、 ナフチルな どのァリ ール基 ; ベンゾィノレ、 トノレオイ ルなどのァコイ ル基 などが挙げられる。
前記一般式 〔 DI〕 において、 mは 0 〜 5を示すが、 好ま し く は 0 〜 1 である。
また、 本発明における前記式 〔 IV— a 〕 又は 〔 IV— b 〕 に おいて、 R 6 及び R 7 は同一もし く は異なり、 水素原子並び に置換基を有していてもよい炭素数 1 〜 1 0 のアルキル基及
びアルケニル基、 置換基を有していてもよい炭素数?〜 1 5 のァラルキル基から選ばれる基である。
こ こでアルキル基としては、 例えば、 メ チル、 ェチル、 n ーフ'口 ピゾレ、 i so—フ。口 ピゾレ、 n —ブチゾレ、 s ec—ブチノレ、 ter t—ブチル、 ペンチスレ、 へキ シル、 シク ロへキシスレ、 ヘプ チル、 ォクチル、 ノ ニル、 デシルなどの直鎖、 分岐状も し く は環状の炭素数 1 〜 1 0、 好ましく は炭素数 1 〜 4のアルキ ル基が挙げられる。

ァ ラルキル基としては、 例えば、 ベンジル、 フ エネチル、 3 —フエ二 レプロ ピノレ、 1 —メ チノレー 2 —フエ ニル'ェチル、 4 一フエニルブチル、 1 , 1 一 ジメ チルー 2 — フ エ二ルェチ ゾレ、 5 —フ エ二 レペンチル、 6 —フ エニル'へキ シル、 4 ーフ ェニ レシク ロへキシ レ、 7 フ エ二 Jレへフ。チ レ、 8 —フ エ二 ルォク チル、 9 一フ エニルノ ニルなどの炭素数 7 〜 1 5、 好 ましく は炭素数了〜 1 0 のァ ラルキル基が挙げられる。
本発明において、 前記一般式 〔 M〕 で表わされるイ ソキノ '; ン誘導体は、 前記一般式 〔 I 一 b 〕 で表わされる 3 , 4 — ジヒ ドロイ ソキノ リ ン誘導体を脱水素せしめることにより製 造されるが、 この場合、 使用する脱水素触媒としては脱水素 して芳香族核を形成しう る脱水素触媒であればいかなる触媒
でもよ く 、 例えば、 パラジウム、 白金、 ルテニウムなどの貴 金属触媒、 硫黄、 セ レン、 R a n e y ニ ッケル、 ジク ロ ロジシァ ノ キノ ンなどのキノ ン類などがあり、 好ま し く は貴金属触媒 特に好ま し く はパラジゥム触媒が挙げられる。
上記脱水素反応は、 液相、 気相のいずれにおいても実施で きるが、 通常液相で実施される。 気相で行う場合一般にはキ ャ リ ア ' ガスを用いて行う。 キャ リ ア · ガスとしては反応に 悪影響を与えないものであればよいが、 例えば窒素、 へリ ウ ムなどが挙げられる。
液相で行う場合、 必ずしも反応溶媒を用いな く てもよいが、 一般には溶媒を使用する。 溶媒を使用する場合、 溶媒として は反応に悪影響を与えないものであればよいが、 具体例を示 すと、 例えばデカ リ ン、 クメ ン、 P — シメ ン、 キシレン、 二 ト ロベンゼン、 ナフタ リ ンなどが挙げられる。
また上記反応は通常 50〜250 ての範囲の温度、 好ま し く は 100〜200 'Cの範囲の温度で実施され、 また圧力は減圧、 常 圧、 加圧のいずれでも差支えがな く 一般には常圧 (大気圧) である。 反応時間は、 出発化合物の種類、 触媒の種類、 溶媒 の種類、 反応温度などの反応条件により左右されるが一般に は数分乃至数十時間の範囲である。
反応装置の型式については、 一般に使用される型式であれ ば何等制限されることはないが、 例えば固定床、 懸濁床など 通常塔型、 槽型の反応装置が使用される。
また、 本発明における前記一般式 〔 n〕 で表わされるイ ソ キノ リ ン誘導体は、 前記一般式 〔 IV— a 〕 又は 〔 IV— b 〕 で
表わされる化合物と 2官能性シリ ル化剤とを反応せしめるこ とによっても製造されるが、 この場合 2官能性シリ ル化剤と

ノ ) メ チルビ二ルシ ラ ンなどが挙げられる。
この反応の際、 H 20を共存させると、 前記式 〔 IE〕 におけ

かかる化合物は、 予め前記 2官能性シリ ル化剤を H 20と反

これらシリ ル化剤は、 種々の割合で使用できる。 前記式 ( IV - a ) 又は 〔 IV— b 〕 で表わされる化合物 1 モル当り、 0. 5 モル以上、 好ま し く は 1. 0 モル以上となるような割合が 有利に使用され、 上限は一般に 1 0 モル以下が望ま しい。
また本発明において、 前記一般式 〔 IV— a 〕 又は 〔 IV— b〕 で表わされる化合物における R 6 及びノ又は R 7 並びに上記
P 6 一般式 〔 IV— b 〕 で表わされる化合物における 〉Cぐ を y " R 7 一旦金属原子と置換させその後これをシリ ル化剤と反応せし めることにより前記一般式 〔 ΠΙ〕 で表わされるイ ソキノ リ ン 誘導体を得てもよいが、 その場合置換させる金属原子と して は、 例えば、 リ チウ ム、 ナ ト リ ウ ム、 カ リ ウムなどのアル力

この場合塩基性化合物、 非プ口 ト ン系ァ ミ ド化合物あるい は第 3級ァ ミ ンを使用することにより反応を促進することが 可能である。 殊にこれらは溶媒またはその 1部として使用す ることもできる。 かかる化合物の例としては、 1—ヒ ドロキ シベンゾ ト リ アゾール、 ト リ カプリ リ ルメ チルア ンモニゥム ク ロライ ド、 Ν , Ν —ジメ チルホルムア ミ ド、 Ν , Ν —ジ ^
チルァセ トア ミ ド、 N —メ チルー 2 — ピロ リ ドン、 ト リ ェチ ルァ ミ ン、 ピリ ジン、 N , N —ジメ チルァニリ ン、 N —メ チ ルモルホ リ ンなどを示すこ とができる。

本発明における前記一般式 ( V ) で表わされるイ ソキノ リ ン誘導体は、 前記一般式 〔 I — a 〕 で表わされる 3 , 4 —ジ ヒ ドロイ ソキノ リ ン誘導体を脱水素せしめるこ とにより製造 される。 この場合、 使用する脱水素触媒としては脱水素して 芳香族核を形成しう る脱水素触媒であればいずれの触媒でも よ く 、 例えば、 ノ、。ラ ジウム、 白金、 ルテニウムなどの貴金属 触媒、 硫黄、 セ レ ン、 Ra n ey ニ ッケル、 ジク ロ ロジシァノ キ ノ ンなどのキノ ン類などがあり、 好ま し く は貴金属触媒、 特 に好ま し く はパラジウム触媒が挙げられる。
本発明における上記反応は、 液相、 気相のいずれにおいて も実施できるが、 通常液相で実施される。 気相で行う場合、
一般にはキャ リ ア ' ガスを用いて行う。 キャ リ ア · ガスとし ては反応に悪影響を与えないものであれば 'よいが、 例えば窒 素、 ヘリ ウムなどが挙げられる。
液相で行う場合、 必ずしも反応溶媒を用いなくてもよいが、 一般には溶媒を使甩する。 溶媒を使用する場合、 溶媒として は反応に悪影響を与えないものであればよいが、 具体例を彔 すと、 例えば、 デカ リ ン、 クメ ン、 p —シメ ン、 キシレン、 ニ ト ロベンゼン、 ナフタ リ ンなどの非水素溶媒が挙げられる。

前述の如く'して本発明の反応を行った反応混合物から、 目 的物のィ ソヰノ リ ン誘導体を分離する方法は、 前記の場合と 同様何等制限されることな く、 一般的な分離法が採用され、 例えば蒸発、 蒸留、 抽出、 沈殺 · 結晶化、 吸着、 昇華などあ るいはこれらの組合せが用いられる。 産業上の利用可能性
本発明により提供された 3 , 4 —ジヒ ドロイ ソキノ リ ン誘
導体及びイ ソキノ リ ン誘導体は、 熱に安定で蒸気圧が高く、 蒸留による精製が容易であり非極性溶剤に対する溶解性が高 く 、 抽出あるいは溶媒に対する溶解度差を利用した固体生成 による分離精製を効率的且つ容易に行う ことができる。 また 合成反応、 例えば、 セフユム核の 3位を 4級化するのに中間 原料として使用する場合、 非極性溶剤に対する溶解性が高い ので、 反応操作が容易であり反応性も高く収率もよい、 など の効果を与える有用な新規化合物である。
また本発明によれば前記一般式 〔 I 一 a 〕 又は 〔 I 一 b 〕 で表わされる 3 , 4 —ジヒ ドロイ ソキノ リ ン誘導体を脱水素 せしめることにより前記式 ( EI ) 又は ( V ) で表おされるィ ソキノ リ ン誘導体を効率よ く製造するこ とができ る。
本発明により提供された 3 , 4 —ジヒ ドロイ ソキノ リ ン誘 導体及びィ ソキノ リ ン誘導体は、 医薬及び農薬の中間原料、 例えばセファ ロスポ リ ン骨格と反応後脱シリ ル化する こ とに より ( 6 R , 7 R ) 一 Ί — 〔 ( Z ) — 2 — ( 2 —ァ ミ ノ チア ゾールー 4 —ィ ノレ) 一 2 — ( 2 —カルボキシプロポー 2 —ィ ルーォキシィ ミ ノ ) ァ セ ト ア ミ ド〕 一 3 — ( 3 , 4 — ジ ヒ ド 口 一 6 , 7 —ジヒ ドロキ シ一 2 —イ ソキノ リ ニゥム) メ チル 一 3 —セフヱム一 4 一カルボキ シレー トなどの抗菌力が強く しかも抗菌スぺク トルの巾の広いセフ ァ ロスポリ ンを与える などセファ ロスポリ ン化合物の 3位側鎖の構成材料となり得 る中間原料として有用な新規化合物である。

6 , 7 —ジヒ ド ロキ シ一 3 , 4 — ジヒ ド ロ イ ソ キノ リ ン 3.26 gをテ ト ラ ヒ ド ロ フ ラ ン 4 中に加え、 これに室温で 水素化ナ ト リ ウ ム 1.44 gを加え攪拌下 4 0 て、 常圧で 2時間 反応させた。 次いでこ の反応液に攪拌下、 あらかじめ tert— ブチノレジメ チルク ロ ロ シラ ン 7。 55 gをテ ト ラ ヒ ド ロ フ ラ ン 2 O ffl£中に加えて得られた混合物を滴下し 5 0 'C、 常圧で 1. 5時間反応させた。
冷却後濾別し、 ケークをテ ト ラヒ ドロフ ラ ンで洗浄後回収 した濾液を減圧蒸留し:
