Abstract
Description
The invention relates to the field of medicine and biology, namely to the field of cytology and immunology and can be used for comprehensive assessment of the body's resistance.
084,The well-known "Method of determination and/or quantitative detection and/or separation of apoptotic cells in a sample or part of a sample" (patent for invention UUO 9527903 JSC, dated 10.19.95, Izobreteniya stran mira (1996). - Vyipusk 084,
48) involves the contact of the studied sample of cells or tissue with a reagent - a polypeptide or protein from the class of annexins, which has an affinity for phosphatidylserine of the plasma membrane of apoptotic cells.
Detection and/or quantification of apoptotic cells is carried out directly by detection of a label that is connected to annexin. Such labels can be radioactive markers, fluorescent, enzyme, protein, individual dyes, and immunoglobulins. For example, flow cytometry, image cytometry or image analysis is used to detect the presence or absence of a label. In the indirect method of detection, the sample comes into contact with an unlabeled reagent with a high affinity for phosphatidylserine, and the amount of bound agent is determined using reagent-specific antibodies. In this case, any immunoassay technique can be used. It is proposed to qualitatively and quantitatively determine apoptotic cells after incubation of the sample with substances that can induce or inhibit apoptosis. The proposed method is used for diagnostics.
The disadvantage of using the absolute value of spontaneous and induced apoptosis for diagnosis is the assessment of only the final result of the apoptotic process without taking into account numerous factors that can lead to apoptosis (the state of oxidant and antioxidant systems, the level of tumor necrosis factor, the state of cell membranes, estimated by the amount of platelet aggregation).
The purpose of the proposed method is to increase the informativeness and reliability of the method by determining the levels of spontaneous and modulator-induced, for example, dexamethasone, apoptosis and the apoptosis induction index, namely the ratio of spontaneous and induced, for example, dexamethasone, apoptosis, simultaneously with a comprehensive assessment of the state of the oxidant-antioxidant system , cell membranes by the value of the aggregation properties of platelets and the level of spontaneous and tumor necrosis factor induced, for example, by lipopolysaccharide.
The task is achieved by the fact that the resistance of the organism, which is determined in a known way by isolating mononuclear cells, incubating them with dexamethasone, staining with a dye, followed by determining the indices of spontaneous and induced, for example, dexamethasone, apoptosis, according to the proposed method, the values of peroxidation are measured lipids, catalase, platelet aggregation and levels of spontaneous and induced, for example, lipopolysaccharide, tumor necrosis factor and determine the apoptosis induction index, namely, the ratio of apoptotic indices of spontaneous and induced, for example, dexamethasone, apoptosis and at values of the apoptosis induction index ranging from 0 .6106 to 0.6301 and values of spontaneous and induced, for example, by dexamethasone, apoptosis, lipid peroxidation, catalase, platelet aggregation and spontaneous and induced, for example, by lipopolysaccharide, tumor necrosis factor within the normal range diagnose that the body's reactivity is within the normal range, p A decrease in the level of all determined indicators, except for the apoptosis induction index, diagnoses a decrease in the body's resistance due to exhaustion, with an increase in the level of the apoptosis induction index above 0.301 and an increase in all other studied indicators, a diagnosis is made of stressing the body's resistance with sufficient compensatory capabilities.
4009.The claimed method is performed as follows. Mononuclear cells are isolated from heparinized blood by centrifugation with an acceleration of 4009 for 25-35 minutes in a density gradient of ficol-verografin (p-1077-1078), a ring of mononuclear cells is removed. The obtained cells are pipetted and centrifuged twice for 8-12 minutes with an acceleration of 2009 in a neutral buffer solution, for example, rpozrnaiye rimbeg zooshiop with pH 7.2. Double cultures are prepared from the washed cells at a concentration of 5-x106 cells/ml, which are placed in a nutrient medium containing 1095 fetal calf serum in a 1:1 ratio. A solution of dexamethasone with a final concentration of 1075-1077M is added to one of the double cultures. Cell cultures are incubated for 12-24 hours at a temperature of 37"C. After incubation, the cells are pipetted into a solution of a neutral buffer, for example, rpozrpage ribeg zoishiop with a pH of 7.2, centrifuged for 3-7 minutes at an acceleration of 4009. The percentage ratio of live and dead cells is calculated by using a standard test with trypan blue. Washed cells are incubated for 25-35 minutes at a temperature of 37"C with a solution of Noesnvzi 33342 dye at a concentration of 0.05-0.15 μg/ml in a ratio of cell suspension/dye solution - 1:1-2:1 , then centrifuged with the addition of a neutral buffer solution, for example, a buffer solution with a pH of 7.2, for 3-7 minutes with an acceleration of 4009.
Cells are fixed with a fixing mixture, placed on a glass slide, covered with a coverslip and sealed. The preparations are viewed under the illumination of a fluorescent lamp at a magnification of 7x90. The apoptotic index is defined as the percentage ratio of the number of cells with signs of apoptosis and 2000 cells detected in the field of view of the microscope. The apoptotic index is determined both in cells that were incubated only in the nutrient medium - the index of spontaneous apoptosis, and in cells that were incubated in the nutrient medium in the presence of dexamethasone - the index of induced apoptosis. The apoptosis induction index was determined as the ratio of spontaneous and induced apoptosis indices in this patient. Values of lipid peroxidation and the level of blood catalase, levels of spontaneous and induced, for example, lipopolysaccharide, tumor necrosis factor, the value of platelet aggregation were determined according to generally accepted methods. With the values of the apoptosis induction index ranging from 0.6106 to 0.6301 and the values of spontaneous and induced, for example, dexamethasone, apoptosis, lipid peroxidation, catalase, platelet aggregation and spontaneous and induced, for example, lipopolysaccharide, tumor necrosis factor within the normal range it is diagnosed that the body's reactivity is within the normal range, with a decrease in the level of all determined indicators, except for the apoptosis induction index, a decrease in the body's resistance due to exhaustion is diagnosed, with an increase in the level of the apoptosis induction index above 0.6301 and an increase in all other studied indicators, resistance stress is diagnosed body with sufficient compensatory capabilities. A decrease in antioxidant activity with an increase in the pro-oxidant system and apoptosis induction index was diagnosed as the beginning of decompensation of the body's resistance. A decrease in the level of induction of tumor necrosis factor with an increase in its spontaneous level was diagnosed as evidence of the presence of an infectious process or hypoxia in the body, with the latter a decrease in the antioxidant system was observed. A significant increase in the aggregation ability of platelets was diagnosed as the possibility of a critical condition in the patient.
