Description
Description of the invention
The invention relates to the field of medicine, namely to the method of obtaining material containing cells of the prostate gland 2 and intended for transplantation to patients, the material itself containing cells of the prostate gland, which is obtained by a new method, and to transplantological methods of treating fibromatosis of the uterus, chronic prostatitis and disorders of male sexual function by transplantation of the obtained material.
-С.38-42).There is a known method of obtaining material from human and mammalian fetuses (pig fetuses, cattle fetuses), which contains, in particular, islet cells of the pancreas. The method consists in the fact that the pancreas of fetuses of humans, cattle or pigs is extracted under sterile conditions, washed in Hanks' solution with antibiotics, cut into fragments of 2 mm in size, which are washed again with Hanks' solution with antibiotics. Then the fabric is immersed in a 0.1-0.395 solution of kollalitin (a preparation containing collagenase) or a mixture of equal parts of 0.195 solution of kollalitin and 0.195 solution of trypsin 72 for 20-30 minutes at a temperature of 20-22. Next, the enzyme solution is removed, the material is transferred to a 0.395 trypsin solution, placed on a magnetic stirrer and processed for 15-20 minutes. Two or three pours of the trypsin solution with single cells and small cell complexes suspended in it are made into centrifuge vials, and then centrifuged and the remainder resuspended. The obtained unsettled cells and cell complexes are fractionally separated and seeded with them in culture mattresses. Incubation is carried out at a temperature of 36-37 for 8-10 days with replacement of the growth medium every 3-4 days. At the same time, a culture was obtained, the majority of which consists of B cells (see Experimental basis of clinical transplantation of islet cells / Shumakov V.I., Blyumkin V.N., Skaletskyi N.N., Shalnev B.I. in the collection "Pancreas Transplantation glands (essays)", m., "Kanon" 1995. - P.38-42).
The disadvantages of the known method include a significant loss of cells in the process of multi-stage processing of the material, in particular, during being on a magnetic stirrer and during centrifugation, as well as during fractional (3) sedimentation of cell complexes and single cells.
там же ж).A material containing beta cells is known, which was obtained by the above-mentioned method (see ibid.).
сThe disadvantages of the known material obtained by this method include its reduced biological activity, caused by the disadvantages of the method of its production, in the process of which some cells are destroyed, and the castine is injured when it is on a magnetic stirrer and during centrifugation. with

The task is solved by the fact that when obtaining material containing cells of the prostate gland, the prostate gland of an adult young or newborn rabbit is used (the prostate gland of another mammal can be used), while the extracted prostate gland is washed in Hanks' solution with the addition of antibiotics, cut into fragments and inoculate culture mattresses with the obtained material, adding culture (growth) medium and mammalian serum, then provide the necessary (optimal) mode for cultured microfragments of the prostate gland, for which incubation is carried out at a temperature of 35-40 for two days to six weeks with single or multiple lowering the temperature to 4-25 2C and holding at a lower temperature, as well as with a temporary one-time or periodic decrease in serum concentration.
The task for the material containing cells of the prostate gland is solved by using the above-mentioned method of obtaining it.
The task set for the method of treating fibromatosis of the uterus is solved during its implementation, in the process of which a transplant of material containing cells of the prostate gland is carried out, while the specified 75 material can be transplanted into various organs and tissues: subcutaneously, intramuscularly, in particular, into the direct muscle stomach ulcer, into the liver (directly into the parenchyma or through the portal vein or branches), into the pulp of the spleen, into the splenic vein, into the peritoneal cavity, into a specially created muscle pocket, into the parenchyma of the liver, or into the great omentum.
The fundamental difference of the method of obtaining material containing cells of the prostate gland, according to this 2nd invention, is its simplicity and relatively low cost due to the fact that the prostate gland of newborn, young or adult rabbits is used as the starting material, and not embryos, as in the closest analogy. For the proposed method, it is also possible to use mammalian embryos, but this is not advisable, because the glandular structures in the embryonic prostate gland are not yet formed.
When implementing the method according to the present invention, operations that injure the cells of the prostate gland, namely centrifugation, processing on a magnetic stirrer, and the operation of processing with an enzyme to dissolve collagen fibers, are excluded. According to the present invention, it is possible to release more than 9095 prostate cells contained in the original prostate gland.
ч-To obtain material containing cells of the prostate gland, the prostate gland of mammals, in particular, newborn mammals or their fetuses, can be used. Experience proves that it is possible to use the prostate gland of cattle and their fetuses, as well as horses and pigs, human fetuses, but the most accessible and cheap material is the prostate gland of newborns, young or adult rabbits. h-
To obtain material containing cells of the prostate gland, the prostate gland is removed, for example, from a young or adult rabbit, and immersed in cold Hanks' solution with the addition of antibiotics. After removing the prostate gland from the solution, its capsule and layers of connective tissue with its blood vessels are removed. The decapsulated prostate gland is crushed into microfragments up to 1 mm in size and washed again with Hanks' solution with the addition of antibiotics.
The resulting material is sown in cultural mattresses, where a cultural (growth) medium and "cattle serum" are added. Then the seeded culture mats are placed in a thermostat, where they provide an optimal regime for the cells of the prostate gland, for which incubation is carried out at a constant temperature in the range of 35-402C for two days to six weeks, while the temperature is lowered once in depending on the need (microscopic control of the culture is carried out) to 4-252C, and withstand a constant time at a reduced temperature. The duration of exposure is determined experimentally for a specific type of cell culture. To obtain high-quality material containing prostate cells, a temporary one-time or periodically repeated reduction of serum concentration 1. The exposure time at reduced serum concentration is also determined experimentally for a specific type of prostate cell culture.

The myometrium has a somewhat heterogeneous echo structure. Conclusion. Fibromatosis of the uterine body. 4 days after the patient's admission, an intramuscular transplantation of material containing prostate cells obtained from young rabbits was performed. After another 2 days, the patient was discharged from the hospital under outpatient supervision.
During the control ultrasound, which was performed two weeks after transplantation, the following data were obtained: during the examination of the uterus, no areas of fibromatosis were detected. Myometrium of a normal structure. 70 Conclusion: no pathological changes were detected.
The next 16-month follow-up did not reveal signs of recurrence of uterine fibromatosis.
Example 2. Patient B.V., 39 years old. He has been suffering from chronic prostatitis for 8 years. Repeated courses of traditional treatment with a urologist and a sexologist did not have a positive effect. The patient underwent intramuscular transplantation of material containing cells of the prostate gland obtained from adults/5 rabbits. Already 2 weeks after the transplant, the patient began to notice a significant improvement in his state of health, increased work capacity. Over the next 2 months, the patient's sexual activity increased: the frequency of full sexual contacts increased from 2.1 per month to 2.5 per week. Such a significant improvement in sexual function was noted at least 8 months after transplantation.
