Abstract
Method for production of lactoferrine concentrate and cow milk protein containing copper, at which the mild is separated into fats and milk plasma by centrifugation at 5000 g during 20-25 minutes at the temperature of 3-5°C. Casein is removed from milk plasma by acidification method 1 M HCl up to the isoelectric point of protein pH 4.6 followed by incubation during 18-24 minutes at the temperature of 23-26 °C and centrifugation during 1 hour at 19000 g and at the temperature of 2-4°C. From lactoserum (supernatant) after adding of alkali 1 M NaOH up to pH 7.2 by solution of ammonium sulphate in the range of saturation 30-80 per cent a fraction is precipitated in which is lactoferrine and protein containing copper. Obtained deposit is dissolved in distilled water and pasteurized at the temperature of 55-65 °C during 30 minutes in order to make microbiological purification of preparation. Water dialysis is made followed by vacuum lyophilization. Amount of lactoferrine and protein containing copper in volume unit of lyophilizate is determined by electrophoresis method in polyacrylamide gel with sodium dodecylsulphate. Obtained concentrate of lactoferrine and protein containing copper is subject to microbiological analysis for pathogenic flora.
