The method of normalizing the microflora of the stomach under the conditions of the use of proton pump blockers in the experiment includes therapy with probiotic microorganisms for 28 days. At the same time, Symbiter is used as a multiprobiotic, which is administered to experimental animals orally once a day at a dose of 14 mg/kg (1.4·1010 CFU/kg), previously dissolved in 0.5 ml of water for injections.
Description
The useful model belongs to experimental medicine and physiology and can be used to improve the effectiveness of treatment of dysbacteriosis as a pathological condition and to normalize the composition of the microflora of the digestive tract disturbed by various diseases, bacterial or chemical intoxication.
Long-term use of proton pump blockers - omeprazole, lansoprazole, rabeprazole - in the treatment of acid-dependent diseases leads to suppression of the secretion of hydrochloric acid in the stomach. Under the conditions of inhibition of hydrochloric acid secretion, gastric juice loses its bactericidal properties, thanks to which the body is largely protected from the aggression of pathogenic microorganisms. As a result, the qualitative and quantitative composition of the microflora in the stomach changes: the indicators of the seeding of normal microflora decrease and the indicators of the seeding of conditionally pathogenic ones increase. Excessive bacterial growth in the stomach is the cause of the inflammatory process in the digestive tract.
There is a known method of treating dysbacteriosis, which involves the use of the prebiotic "Extravin" (grape pomace extract) by introducing it into the body (orally, reg gesisht, ip madipa) at a dose of 0.02-0.1 g/kg of body weight per day for no less than two weeks | see description of the patent of Ukraine for the utility model "Method of treatment of dysbacteriosis" Mo 42783, IPC.
Ab1KZ31/00, AbI1R 1/00, publ. 27.07.2009). This method has a number of disadvantages. The use of prebiotics is longer in the treatment of dysbacteriosis. Secondly, the use of extravin has a number of contraindications, in particular, allergic reactions to grapes and products derived from them
The useful model is based on the task of creating a method of normalizing the stomach microflora under the conditions of using proton pump blockers in the experiment, the use of which in real conditions makes it possible to reduce the concentration of harmful microorganisms and restore the population of useful microflora in the stomach of experimental animals naturally (without the use of synthetic drugs, without causing addiction and side effects) to complete recovery or long-term remission, as well as to ensure a complex positive effect on the digestive system, in particular, to eliminate or reduce the manifestations of dyskinesia, hypochlorhydria, hypergastrinemia, etc.
The task is solved by the fact that the method of normalizing the microflora of the stomach under the conditions of the use of proton pump blockers in the experiment, according to a useful model, includes therapy with probiotic microorganisms for 28 days, while as
As a multiprobiotic, Symbiter is used, which is administered orally to experimental animals once a day at a dose of 14 mg/kg (1.4 1079 CFU/kg), previously dissolved in 0.5 ml of water for injections.

The use of the proposed method makes it possible to populate the gastrointestinal tract of rats with useful bacteria in an active form, which begin to produce a number of biologically active substances (antibiotics, vitamins, amino acids, short-chain fatty acids, enzymes that neutralize free radicals, etc.), suppression of pathogenic microflora (probiotic microorganisms compete with pathogenic bacteria for nutrients or adhesion sites). Therefore, the use of the proposed method has a complex positive effect on the digestive system, in particular, it contributes to the restoration of the ecological balance of the gastric microflora, the elimination of manifestations of dysbacteriosis, the normalization of digestive processes (synthesis of group B vitamins, the breakdown of proteins, lipids, lactose), the reduction of symptoms of dyskinesia, hypochlorhydria, hypergastrinemia, etc. , and also has an immunomodulatory effect. The active effect of the multi-probiotic drug begins immediately after its use.
An example of practical application of the method. As part of an experimental study conducted in the accredited vivarium of the Educational and Scientific Center "Institute of Biology" of Taras Shevchenko National University of Kyiv, a method of normalizing the microflora of the stomach under the conditions of using proton pump blockers in the experiment was tested.
All animals were divided into three experimental groups. The first group of animals served as a control, they were administered intraperitoneally (in/o) 0.2 ml and orally (p/o) 0.5 ml of water for injections once a day for 28 days. The animals of the second group were injected with omeprazole and 0.5 ml of water for injections once a day for 28 days. The animals of the third group were simultaneously administered omeprazole and multiprobiotic "Symbiterz" acidophilic" concentrated (Symbiter) once a day for 28 days. Omeprazole (manufactured by Zidta-AIdgisn, USA) was administered intravenously at a dose of 14 mg/kg, dissolved in 0.2 ml of water for injections.
107During the study, the quantitative and qualitative composition of the stomach microflora of the second and third groups was determined compared to the control (first group). As can be seen from Table 1, a narrow spectrum of bacteria was sown from the stomach of rats in the control group. Most often, lactic acid bacteria, mushrooms of the genus Sapaida, Epiegosossib5, E5cPegisnia soii were sown. Conditionally pathogenic microorganisms were practically not detected in the stomach. Only 10 95 animals were inoculated with Siitorasiei at a concentration of 107
CFU/g. The concentration of lactobacilli cultured from the stomach was 102 CFU/g. Quantitative indicators of seeding from the stomach of Epiegosossiv, EzsePegisnia soii and mushrooms of the Sapaida genus were also insignificant.
After 28 days of suppression of NSI secretion in the stomach of rats, the qualitative and quantitative composition of the gastric microflora underwent significant changes. The frequency of seeding of lactobacilli decreased significantly, and in 40 95 experimental rats they were not seeded at all. If in the control for lactobacilli the dDsh CFU/g was equal to 2.520.03, then after 28 days of administration of omeprazole Id CFU/g was equal to 1.3520.03 (p-0.01). At the same time, the colonization of the stomach by opportunistic microflora and fungi of the Sapaida genus increased more than 1000 times (p<0.001). The concentration increased 10 times (p<0.001).

Against the background of long-term hypoacidity of the gastric juice, the stomach became populated with opportunistic bacteria, namely EzsNegisnia soyii (lactose negative), KiebziePa, Rgoiei5, EEFarnuiosossiv aigeiv, Eiarnuiosossiv erideptides (with hemolysis), the concentration of which was significant and amounted to 105-105 CFU/g.
Thus, the blockade of the proton pump eliminated the effect of the bactericidal factor - hydrochloric acid - on microorganisms and created an optimal pH for the growth and reproduction of opportunistic microflora and significantly reduced the resistance of the gastric mucosa to colonization by microorganisms. The obtained data also made it possible to conclude that Kierzieya, Etarpuiosossi5 acgeiv, Eiarnuiosossiv erideptidie (with hemolysis) enter the stomach by migration from the intestines in an ascending way.
In rats, which were administered the multiprobiotic "Symbiter" simultaneously with omeprazole for 28 days, the quantitative and qualitative indicators of the seeding of opportunistic microflora from the stomach did not differ from the control values.
Thus, long-term use of the multi-probiotic "Symbiter" significantly reduced the number of pathogenic microorganisms in the stomach of laboratory animals, while the number of lactobacilli increased in comparison not only with the second group, but even with respect to the control group. This allows you to use the claimed method to restore the normal microflora of the stomach under the conditions of using proton pump blockers in the experiment.
So, the effectiveness of the claimed method and its advantages over the analogue are confirmed by the results of the studies given in the example of a specific implementation of the method.
The claimed method is expedient to use in medicine and experimental medicine to improve the effectiveness of treatment of dysbacteriosis as a pathological condition, to normalize the composition of the microflora of the stomach, disturbed as a result of various diseases, bacterial or chemical intoxication.
