B-7729; Epiegosossiv Taesiit, deposited under registration number B-7789; Epiegosossiv5 yigap, deposited under registration number B-7749; Eliegosossi5 digap5, deposited under registration number B-7769. The invention also relates to a method of manufacturing an escherichia-enterococcal inactivated liquid vaccine and a method of treatment and prevention with an escherichia-enterococcal inactivated liquid vaccine against Escherichia coli and enterococcus.
The group of inventions belongs to medical microbiology, the pharmaceutical industry, in particular to vaccines, namely to vaccines against intestinal enterobacteria and enterococcus, methods of their production and methods of treatment and prevention, and can be used for the treatment and prevention of local and systemic, acute and chronic diseases of bacterial etiology in children and adults: nose, additional nasal sinuses, pharynx, conjunctiva and eyes, oral cavity, tongue, gums, teeth, trachea, bronchi, lungs, organs of the genitourinary system, gall bladder, abdominal cavity, skin, subcutaneous tissue etc., as an alternative to antibiotics or as a method of additional treatment compatible with antibiotics in life-threatening septic conditions.
Escherichia coli is a bacterium (motile, rod-shaped microorganism) that normally resides in the human intestine, does not cause harm, and is even used for the manufacture of medicinal products (e.g., Colibacterin, Mutaflor, etc.). But there are its varieties that can cause damage to the stomach and intestines. They are the cause of the development of a special type of acute intestinal infection, which is called escherichia or coli infection (from the Latin name of the Escherichia coli: EuspPegisnpia soii). Usually, an intestinal infection caused by Escherichia coli affects the stomach and small intestine. In cases when the usual non-pathogenic Escherichia coli from the intestines through contact and household contact (normal physiological infection) occupies other territories (mucous membranes of other organs and systems), it turns from a symbiotic non-pathogenic species of bacteria into a completely pathogenic strain that causes severe chronic bacterial diseases with recurrent lifelong course and in the case of the emergence of complete resistance to antibiotics (in cases of aggressive and constant use of them with the aim of destroying E. coli or other bacteria) - even with possible fatal consequences.
Enterococci (lat. Epiegosossi5) are a genus of spherical or slightly elongated gram-positive bacteria that were originally assigned to the genus Streptococcus (Zieriosossib) of group Y. In connection with the significant differences between enterococci and streptococci discovered in 1984, they were assigned to the genus Epiegosossib5 , and in 2001 - to the independent family Enterococci (Epiegosossaceae). Placement usually occurs in pairs (diplococci) or in short chains.
Станом на 2017 рік, до ентерококів зараховано майже 60 видів бактерій, найбільш патогенними з яких є ентерокок фекальний (Епієегососси5 ГТаеєсаїї5), ентерокок феціум (Епівгососсив Таесішт) та ентерокок дуранс (Епівгососси5 игапб5), які зазвичай також розглядають як симбіотичні мікроорганізми кишечнику людини та навіть використовують для виготовлення лікарських засобів (напр., препарат Лінекс).As of 2017, enterococci include almost 60 species of bacteria, the most pathogenic of which are Enterococcus faecal (Epiegosossi5 GTaesaii5), Enterococcus faecium (Epivgosossiv Taesisht) and Enterococcus durans (Epivgosossi5 igapb5), which are usually also considered as symbiotic microorganisms of the human intestine and even used for the manufacture of medicinal products (e.g., Linex drug).

113299/.EzsPegispia soii, KierziePa rpeshtopiae Epiegosossiz Taesaiii5, Rgoiei5 tigabiii5, Rgoiei5 todopii and is used for the prevention and treatment of recurrent infections of the urinary tract of bacterial origin (NERz:/Lymly. emgotapt.sot.tsa/zigomak ZTAOMAS ia113299/.
The composition of the Strovak vaccine does not take into account the current strain of bacteria that dominates the etiology of a particular disease in a particular patient (for example, Escherichia cystitis, enterococcal prostatitis), which makes the effect of the vaccine less pronounced with a less pronounced positive clinical effect.
The origin of the bacterial strains in the Strovac vaccine is unknown.
The use of the Strovac vaccine is limited to diseases of the genitourinary system.

After that, the cultures are mixed in equal volumes, packaged and sealed (KO Mo 2429012 C1, AbIK 39/116, 20111.
The disadvantage of this method is that the received vaccine contains a large number of ballast cells of EbsPegispia soya, which carry an excessive antigenic load, which negatively affects the formation of immune defense against the toxins of Escherichia coli, streptococci and enterococci, in addition, this vaccine can be reactogenic.
The method also uses formalin, which reduces its immunogenic effect and immunological and positive clinical responses after vaccination. 60 This method, like the previous analogues, is intended for vaccination of animals, not people.

The EkoPrimavak vaccine has no side effects. Only local (hyperemia, swelling, and pain at the injection sites) and general (increased body temperature, skin rash, as a manifestation of bacterial toxicoderma) reactions to vaccine administration are possible. Such reactions are of a natural physiological nature and are caused by the activation of local immunity (at the site of vaccine administration) and the general immune response with the release of toxins from bacteria against which the specific EcoPrimavak vaccine is directed, due to the rapid destruction of bacteria after their immune recognition by the adjusted immune system.
In the prototype, the development of allergic reactions during the use of the vaccine is possible, and the claimed vaccine has no allergic reactions, moreover, the vaccine causes a hypoallergenic effect with a significant decrease in the elevated levels of IDE before treatment.

Child K., 6 years old, diagnosis: chronic cystitis and chronic pyelonephritis, confirmed by the results of an ultrasound examination of the kidneys (ultrasound) with the debut at the age of 2 years after another acute respiratory illness and subsequent periodic relapses. A urine culture of Escherichia coli (E5cPegispia soii) was repeatedly isolated during bacteriological examination of urine. Courses of antibacterial therapy were regularly conducted, taking into account the sensitivity of selected bacterial cultures to antibiotics. However, the frequency of recurrences of cystitis increased dynamically, accompanied by a periodic increase in body temperature and pain syndrome in the area of the kidneys. The parents went to the clinic during another exacerbation (2 weeks after the course of antibiotic therapy the day before) with leukocyturia of 25-30 cells in the field of vision and moderate proteinuria (protein - 0.066 g/l). During bacteriological examination of warm urine (three days in a row), three identical cultures of E5cPegispia soyii were isolated. The escherichia-enterococcal vaccine EkoPrimavak was prepared. A treatment course of 10 subcutaneous injections every other day was carried out. The child's condition improved, urine tests came close to normal. After 1 month, the second course of immunization with bacterial polyvalent vaccine UroPrimavak was carried out, also with 10 injections. During 2 years, only 5 courses of treatment with bacterial vaccines were carried out. There was a stable clinical remission, there was no more need to prescribe antibiotics. The child continues to be under outpatient supervision with periodic monitoring studies of general urine analysis and bacteriological examination of warm urine.

Patient T., 18 years old. She came to the clinic on September 10, 2014, with a diagnosis of congestive discs of the optic nerves of both eyes with reduced vision (MOYU-0.4; MOY5-0.3). The previous course of treatment lasted 2 years and 9 months with almost zero results - the stagnation worsened. The etiology of these stagnant discs has not been verified. She passed additional examinations for viral infections in the clinic - no active forms were detected; the state of immunity and rheumatism tests are normal; there were no serological signs of autoimmune systemic diseases. A bacteriological diagnosis of urogenital dysbacteriosis of enterococcal etiology with the onset of cystitis (in March 2014) and nephrodysbacteriosis, which proceeded with manifestations of the syndrome of chronic bacterial intoxication with endotoxicosis, moderate toxemia of resorptive genesis, secondary hypochromic anemia, episodes of cephalgia and predominant asthenovegetative symptoms, was suspected and confirmed bacteriologically.
It was suspected that congestive discs of optic nerves, loss of vision, as well as changes on magnetic resonance imaging (MRI) and vasogram of cerebral vessels have a toxic etiology associated with endotoxicosis of bacterial etiology.
The first course of treatment with a bacterial escherichia-enterococcal vaccine was carried out
EcoPrimavak from 10 subcutaneous injections. She came for a follow-up examination after 2 months (December 15, 2014). It turned out that 15 days after the start of treatment, visual acuity increased almost to normal - 90-100 95 (it was only 30-40 95) and visual signs of swelling of the optic nerve discs of the left eye completely disappeared. Then, 2.5 months after the start of treatment, the signs of edema of the optic disc and the right eye completely disappeared. In addition, according to the ultrasound examination of the vessels of the brain, in 2.5 months, symmetrical blood flow through the middle cerebral arteries was completely restored and positive dynamics were noted in the disturbed venous outflow from the skull cavity. 3 months after the end of the 1st course, the 2nd course of immunization with Escherichia enterococcal vaccine EkoPrimavak of 10 injections was carried out and then the 3rd additional course of vaccination with the UroPrimavak vaccine, also of 10 injections. During the next 4 years of catamnestic there were no more observations of relapses of vision loss (100 95 vision in both eyes), cystitis and bacteriuria.
