Abstract
發明係關於一種單離抗體,包含重鏈互補決定區CDR1、CDR2、CDR3與輕鏈互補決定區CDR1、CDR2、CDR3,其重鏈互補決定區係包含於具有SEQ ID NO: 1或3之重鏈可變區序列,輕鏈互補決定區係包含於具有SEQ ID NO: 2或4之輕鏈可變區序列,該抗體可與血管內皮生長因子受體-2 (vascular endothelial growth factor receptor-2, VEGFR2)及VEGFR3特異性結合。
Description
本發明係關於一種可特異性標靶VEGFR2與VEGFR3的雙功能抗體,該抗體可抑制VEGFR2、VEGFR3各自的同質二聚化或彼此間的異質二聚化,但不影響與其配體的結合,因此可調節VEGFR2與VEGFR3的訊息傳遞路徑,並同時抑制血管新生與淋巴管新生。
在腫瘤的發展過程中,血管新生與淋巴管新生是必要的,並且與其是否為惡性腫瘤具高度的相關性。其生物活性都取決於血管內皮生長因子(vascular endothelial growth factor, VEGF)與其相對應之受體(vascular endothelial growth factor receptor, VEGFR)結合,可進而使受體二聚體化並產生自身磷酸化反應而活化下游訊號,以增進細胞增生、細胞遷移、血管通透性及新生血管存活。Olsson等人的研究文獻(Olsson et al., Nat Rev Mol Cell Biol, 2006. 7(5): p. 359-71.)指出,VEGF-A、VEGF-C為負責血管新生與淋巴管新生的主要VEGF配體,於先前的臨床前研究亦揭示,若阻斷效應子(effector)與VEGFR2或VEGFR3結合,可有效抑制腫瘤生長。然而,參見相關文獻(Mukherji, S.K., AJNR Am J Neuroradiol, 2010. 31(2): p. 235-6; Bagri, A., et al., Trends Mol Med, 2010. 16(3): p. 122-32),使用單株抗體bevacizumab(Avastin) 阻斷VEGF-A來進行腫瘤治療時,僅於一些癌症中可達到延遲進展和延長存活的療效。導致此惡性腫瘤臨床實驗失敗的主要原因為,無法減緩可做為癌細胞擴散到淋巴結和遠處部位途徑的淋巴管之生長,因此,淋巴管新生的選擇性抑制劑可與血管新生抑制劑互補,進而達到抑制癌症的效果,然而目前都還沒有相關的臨床應用。
目前,已經開發了三種阻斷VEGFR訊號活化的策略,除了上述的bevacizumab因具抑制細胞生長機制,可做為抗血管新生配體之外,還有aflibercept,係為一種可結合VEGF的嵌合誘餌受體,目前正在進行進階性的臨床試驗(Ellis, L.M., Semin Oncol, 2006. 33(5 Suppl 10): p. S1-7; 及 Holash, J., et al., Proc Natl Acad Sci U S A, 2002. 99(17): p. 11393-8)。第二類抑制劑屬於小分子,例如PTK787、Sutent、sorafinib、Zactima、Recentin等,可抑制VEGFR的細胞內酪胺酸激酶(tyrosine kinase)活性(Wedge, S.R., et al., Cancer Res, 2002. 62(16): p. 4645-55; Wedge, S.R., et al., Cancer Res, 2005. 65(10): p. 4389-400; Wood, J.M., et al., Cancer Res, 2000. 60(8): p. 2178-89; Mendel, D.B., et al., Clin Cancer Res, 2003. 9(1): p. 327-37; 及Wilhelm, S.M., et al., Cancer Res, 2004. 64(19): p. 7099-109) 。其他為使用大分子的療法,像是Ramucirumab (Cryamza)和CDP-791,也可用於阻斷VEGF-A與VEGFR-2之間的交互作用(Krupitskaya, Y. and H.A. Wakelee, Curr Opin Investig Drugs, 2009. 10(6): p. 597-605; and Lu, D., et al., J Biol Chem, 2003. 278(44): p. 43496-507) ,這些藥劑皆具有選擇性且耐受性良好,對於抑制正常器官中VEGF的表現,通常也不具有太大的副作用。
VEGF受體(VEGF receptor, VEGFR)是一更廣泛的受體酪胺酸激酶家族,主要由VEGFR1、2、3組成,也分別被稱為Flt-1、KDR、Flt-4。此家族成員的結構和功能性結構域(domain)具有高度相似性,皆含有7個於細胞外可與配體結合的類免疫球蛋白(immunoglobin-like)結構域,1個可供細胞表面嵌合(cell-surface docking) 的穿膜結構域,與1個可使受體自體磷酸化(trans-autophosphorylation)的雙重酪胺酸激酶結構域。例如VEGF-A、胎盤生長因子(placental growth factor)等配體,係透過與類免疫球蛋白結構域2、3與VEGFR結合,結構域2為主要的結合部分,結構域3則決定產生結合的特異性(Christinger, H.W., et al., J Biol Chem, 2004. 279(11): p. 10382-8; Fuh, G., et al., J Biol Chem, 1998. 273(18): p. 11197-204) 。相反的,類免疫球蛋白結構域4-6則參與受體複合物的二聚化。
理論上,在發生受體二聚化與轉磷酸化(transphosphorylation)時,才能持續活化下游的訊息傳遞路徑,因此可以透過抑制配體與受體的結合或阻斷二聚化發生,來抑制由受體參與而產生的促進血管新生或淋巴管新生活性。
值得注意的是,透過阻斷二聚化的發生以抑制VEGFR-2訊號傳遞,不僅能防止VEGFR2與VEGFR3的同質二聚化(homodimerization),還能干擾VEGFR2/VEGFR3的異質二聚化(heterodimerization),因此,透過抑制VEGR2與VEGFR3所參與的訊號傳遞路徑,以達到抑制腫瘤生長與腫瘤擴散的這種策略是為一種具潛力的新機制。
於一方面,本發明揭示了一種單離的抗體,該單離抗體係包含重鏈互補決定區CDR1、CDR2、CDR3與輕鏈互補決定區CDR1、CDR2、CDR3,其重鏈互補決定區係來自具有序列SEQ ID NO: 1或3的重鏈可變區序列,輕鏈互補決定區來自具有序列SEQ ID NO: 2或4的輕鏈可變區序列,該抗體可與血管內皮生長因子受體-2(vascular endothelial growth factor receptor-2, VEGFR2)與VEGFR3特異性結合。
於部分實施例中,重鏈CDR1、CDR2、CDR3係來自SEQ ID NO: 1,輕鏈CDR1、CDR2、CDR3係來自SEQ ID NO: 2,且該抗體可包括一與SEQ ID NO: 1之序列具有至少80% (例如80%、85%、90%、95%、96%、97%、98%、99%或100%)同一性之重鏈可變區相同;及一與SEQ ID NO: 2之序列具有至少80% (例如80%、85%、90%、95%、96%、97%、98%、99%或100%)同一性之輕鏈可變區。
於一些實施例中,重鏈CDR1具有SEQ ID NO: 5或8之序列,重鏈CDR2具有SEQ ID NO: 6或9之序列,重鏈CDR3具有SEQ ID NO: 7之序列,且其中輕鏈CDR1、CDR2、CDR3分別具有SEQ ID NO: 10、11、12之序列。
於本發明部分實施例中,重鏈CDR1、CDR2、CDR3係來自SEQ ID NO: 3,而且輕鏈CDR1、CDR2、CDR3係來自SEQ ID NO: 4。該抗體可包括一與SEQ ID NO: 3之序列具有至少80% (例如80%、85%、90%、95%、96%、97%、98%、99%或100%)同一性之重鏈可變區之序列,至少有80%與相同(例如80%、85%、90%、95%、96%、97%、98%、99%或100%);及一與SEQ ID NO: 4之序列具有至少80% (例如80%、85%、90%、95%、96%、97%、98%、99%或100%)同一性之輕鏈可變區。
於其他實施例中,重鏈CDR1具有SEQ ID NO: 13或16之序列,重鏈CDR2具有SEQ ID NO: 14或17之序列,重鏈CDR3具有SEQ ID NO: 15之序列,且其中輕鏈CDR1、CDR2、CDR3分別具有SEQ ID NO: 18、19、20之序列。
於部分實施例中,該抗體包含一Fc區、一Fab片段、一Fab’片段、一F(ab)’2片段,一單鏈抗體、一scFV多聚體、一單價抗體、一多特異性抗體或一嵌合抗體。於部分實施例中,該抗體為一人源化抗體。
另一方面,本發明提供一種醫藥組合物,其包含本發明所揭示的抗體和醫藥學上可接受的載體。
在另一方面,本發明亦揭示一種抗體複合體(conjugate),其包含本發明所揭示的抗體與其他分子(例如一小分子藥物、一細胞標靶部分、一細胞毒殺劑、一多胜肽、一核酸分子、一可檢測標記或聚合物)。

本發明揭示可特異性標靶血管內皮生長因子受體-2(vascular endothelial growth factor receptor-2, VEGFR2)與血管內皮生長因子受體-3(VEGFR3)的雙功能抗體,該抗體可抑制VEGFR2、VEGFR3各自的同質二聚化或彼此間的異質二聚化,但不影響與其配體的結合,因此可調節VEGFR2與VEGFR3的訊息傳遞路徑,並抑制血管新生與淋巴管新生。
於部分實施例中,抗體的重鏈互補決定區(complementary determining region, CDR)CDR1、CDR2、CDR3係來自具有SEQ ID NO: 1或3的重鏈可變區序列,且其輕鏈互補決定區CDR1、CDR2、CDR3係來自具有序列SEQ ID NO: 2或4的重鏈可變區序列。
於一些實施例中,其重鏈CDR1可具有SEQ ID NO: 5、8、13或16之序列;重鏈CDR2可具有SEQ ID NO: 6、9、14或17之序列;重鏈CDR3可具有SEQ ID NO: 7或15之序列;輕鏈CDR1可具有SEQ ID NO:10或18之序列;輕鏈CDR2可具有SEQ ID NO: 11或19之序列;輕鏈CDR3可具有SEQ ID NO:12或20之序列。
於部分實施例中,該抗體與位在VEGFR2之IgD6、IgD7結構域內的表位,及VEGFR3之IgD6結構域內的表位結合,該表位可以是具結構性的。
本文所述的名詞“抗體”包括具有抗原結合能力的各種抗體結構,包含單株抗體、多株抗體、全長抗體或其片段、含有Fc區之抗體、Fab片段、Fab’片段、F(ab’)2片段、單鏈抗體、scFV多聚體、單價抗體、多價抗體、人源化抗體、嵌合抗體,但不限於上述種類。
抗體亦可與另一種分子接合,例如:小分子藥物、另一種蛋白質或胜肽、可檢測的標記、聚合物或碳水化合物。
因本發明揭示了該抗體序列與其CDRs,本領域具通常知識者可藉由使用本發明之方法或本領域已知技術,例如重組方法,以多種形式製備抗-VEGFR2/抗-VEGFR3抗體。
另外,本發明亦揭示編碼該抗體或其組成份的分離核酸分子(例如:表達載體),舉例來說,該分離核酸分子可為一個可編碼SEQ ID NO:1-20中任一序列的核酸分子,本發明還揭示含有該核酸分子的宿主細胞,該核酸分子與宿主細胞可用於產生抗體。
本發明所揭示的抗體可用於阻斷VEGFR2和/或VEGFR3的同質二聚化或VEGFR2、VEGFR3的異質二聚化、阻斷VEGFR2和/或VEGFR3的訊息傳遞或是抑制血管新生和/或淋巴管新生。該抗體亦可用於免疫試驗中(如ELISA),以檢測樣品中是否含有VEGFR2和/或VEGFR3或其片段,該抗體可結合並偵測含有IgD6-IgD7結構域的VEGFR2或VEGFR3片段。
此外,該抗體可用於治療實質固態瘤,像是膀胱癌、乳癌、子宮頸癌、大腸癌、直腸癌、腎癌、肝癌、皮膚癌、間皮瘤、肺癌、口腔癌、卵巢癌、胰臟癌、前列腺癌、肉瘤、甲狀腺癌。
本文之“樣品”可以是任何生物性樣品,像是體液樣品、血液樣品、細胞樣品、尿液樣品、唾液樣品或組織樣品。
本發明所揭示之抗體可製備成適用於各種給藥途徑的醫藥組成物,像是透過靜脈內、關節內、結膜、顱內、腹腔內、胸腔內、肌肉內、脊髓鞘內或皮下給藥等方式,該醫藥組成物可以是水溶液或是凍乾製劑,且可包含醫藥上可接受之載體,像是緩衝液、賦形劑、穩定劑或防腐劑;該醫藥組成物可包含其他可與該抗體共同作用之活性成份,像是另一種治療劑或佐劑。
以下的具體實施例僅用以說明本發明,且非以任何方式限制本發明所公開的其餘部分,且基於本案所揭示之內容,本領域具通常知識者無需進一步詳細說明即可充分利用本發明,本文所引述之公開文件皆完整的以參考文獻的方式併入本發明。
假設,可標定VEGFR-2的類免疫球蛋白結構域4-7結構域的治療用抗體可於不受配體影響的情況下達到治療性效果,且可視為被標定的VEGFR-2訊息傳遞活化的辨別模式。以下將敘述可標靶VEGFR-2的治療性抗體R2C7-1、R2C9-11之製備,以及其於體外和體內模擬血管新生和淋巴管新生模型系統中的抑制效果。
實施例1、製備可阻斷VEGF-R2/-R3二聚化的雙功能抗體
VEGFR2和/或VEGFR3的受體二聚化,是活化其下游訊息傳遞路徑,以進行其特定配體的生物活性功能的必要條件,已有文獻指出,VEGF-A和VEGF-C可導致VEGFR2的同質二聚化,或其與VEGFR3的異質二聚化。因此,假設若以VEGFR2上的二聚化區域為標靶,可能可以有效阻斷以VEGFR2和/或VEGFR3受體媒介的訊息傳遞活化,進而抑制VEGFR-A和VEGFR-C所誘導的疾病機制,像是血管新生、淋巴管新生、腫瘤轉移、糖尿病相關視網膜病變和類風濕性關節炎的血管炎等。
為了檢驗該假說,因而建構一個帶有VEGFR2二聚化區域的序列,也就是其類免疫球蛋白結構域4至7(IgD4-7)序列的質體,並送至大腸桿菌表達系統中表達,之後使用His-tag親和性管柱純化得到重組蛋白,且將此重組蛋白免疫接種至小鼠身上,再將小鼠的脾臟細胞與Sp2/0-Ag14骨髓瘤細胞融合,以產生融合瘤庫(hybridoma library)。最初,選擇兩株融合瘤細胞株(R2C7與R2C9)進行後續試驗,因為其對HUVEC細胞具有高親和性且有明顯抑制管形成的效果,為了確保兩株融合瘤各自為單一群體,兩株融合瘤都經限數稀釋方法挑選,並將最終得到的融合瘤細胞株命名為R2C7-1與R2C9-11,後續亦針對此兩株抗體的功能進行檢驗,包括其物理性質也詳細表徵。
實施例2、雙標靶VEGFR2與VEGFR3
為了檢驗新開發的單株抗體R2C7-1、R2C9-11的結合特異性,使用HUVEC細胞與表現VEGFR2的HEK293T細胞(HEK293T/VEGFR)做為實驗模型,結果顯示兩株抗體皆對HUVEC細胞表現的VEGFR2具有相當的親和力(圖1A、1C)。為進一步確定R2C7-1、R2C9-11的結合親和力,HEK293T細胞被穩定轉染編碼人類VEGFR2或其空載體(HEK293T/VC)之質體,並同時使用一種臨床上使用的VEGFR2抗體—Ramucirumab檢測其VEGFR2的表現量。流式細胞儀結果顯示,本發明新開發的單株抗體和Ramucirumab兩者結合VEGFR2的能力相當,亦檢驗R2C7-1、R2C9-11抗體對HEK293T/VC轉染子的結合特異性,結果顯示只有痕跡染色(trace staining),該結果也使用一種商業上可獲得的針對VEGFR2之抗體Abcam9530進一步確認(數據未顯示)。
R2C7-1與R2C9-11單株抗體也具有與人類淋巴管內皮細胞(HLEC)與表現VEGFR3的HEK293T細胞結合的親和力,流式細胞儀分析結果亦顯示該新開發的單株抗體針對HLECs具有獨特的親和力(如圖1B)。由於先前文獻已揭示HLEC細胞上表現了一定量的VEGFR2 (參見 Dellinger et al., 2013, PLoS ONE 8(9): e74686.),因此設計可表現VEGFR3的HEK293T細胞(HEK293T/VEGFR3)用以確認R2C7-1、R2C9-11與VEGFR3的結合特異性,並使用市售的VEGFR3抗體R&D54703來檢測HEK293T/VEGFR3上的VEGFR3表現程度(數據未顯示),透過流式細胞儀分析結果,清楚的顯示兩種抗體與Ramucirumab不同,此兩種抗體都能夠與VEGFR3結合(如圖1D),且兩者相比之下,單株抗體R2C9-11較R2C7-1可更佳的與VEGFR3結合。數據顯示該新開發的單株抗體具有與VEGFR2和VEGFR3結合的雙結合能力,亦表示其可能具有抑制血管新生與淋巴管新生的雙功能。
實施例3、抑制因配體而產生的受體二聚化
為檢測若先使用R2C7-1和R2C9-11單株抗體進行預處理是否能防止VEGFR2和VEGFR3間的異質二聚化,因此使用具有高VEGFR3表現量與低VEGFR2表現量的HEL92.1.7細胞株和同時表現VEGFR2與VEGFR3的HEK293T細胞株(HEK293T/VEGF-R2/-R3)做為實驗模型。不論是使用共免疫沉澱技術(co-immunoprecipitation, IP)於HEL92.1.7細胞中沉澱VEGFR2並對VEGFR3進行墨點分析或於HEK293T/VEGF-R2/-R3轉染子細胞中沉澱VEGFR3並對VEGFR2進行分析,結果皆清楚顯示R2C7-1和R2C9-11單株抗體較Ramuciruman具有更佳的抑制受體異質二聚化的能力,尤其是R2C9-11(如圖2A)。
為了進一步檢測新開發的單株抗體是否具有抑制因VEGF-A與VEGF-C而產生的同質和/或異質二聚化的能力,因此使用HLEC細胞株進行鄰位連接分析法(Proximity ligation assay, PLA)測試。細胞培養於無血清培養基中,並經R2C7-1、R2C9-11、Ramucirumab或同種型控制組抗體(IgG2a)預處理,接著以VEGF-A165
或VEGF-C刺激細胞,並以受體彼此間的距離判定VEGFR2與VEGFR3間是否有同質和/或異質二聚化的形成,若小於40nm即判定為二聚化。相較於未經刺激的細胞或經IgG2a預處理的對照組,結果顯示R2C7-1、R2C9-11單株抗體具有與Ramucirumab相當的抑制因VEGF-A165
和VEGF-C而產生的受體二聚化的能力(如圖2B)。綜上所述,雖然新開發的抗體具有與Ramucirumab相當的阻斷因VEGF-A和VEGF-C而產生的受體二聚化的能力,但新開發的抗體比Ramucirumab具有更佳的抑制異質二聚化的效果。
實施例4、VEGFR2和VEGFR3下游訊息傳遞的衰減
於受體產生因配體所導致的二聚化後,必然會因配體的生物活性功能而發生受體自身磷酸化和下游訊息傳遞路徑的活化。為了檢測阻斷受體二聚化是否足以減弱其下游訊息傳遞路徑的活化,係先以R2C7-1、R2C9-11或其同種型控制組(IgG2a)等單株抗體預處理處於飢餓狀態的HUVEC細胞,並以經Ramucirumab預處理之細胞做為控制組,接著細胞會分別接受VEGF-A165處理、血清刺激或無任何處理。
針對VEGFR2的自身磷酸化與其下游訊息傳遞分子(如FAK p38MAPK)進行西方墨點法分析,結果顯示不論是新開發的單株抗體或Ramucirumab皆能抑制受體二聚化,進而有效的抑制VEGFR2的活化,特別是其Y1054殘基上的酪胺酸磷酸化,雖然R2C7-1的抑制效果比Ramucirumab稍弱一點(數據未顯示)。
與Ramucirumab所誘導的p38MARK去磷酸化不同,R2C7-1單株抗體並沒對p38MARK的活化產生任何效果,反而是有效的使FAK的活化失活。另外,與Ramucirumab相比,R2C9-11則同樣具有抑制p38MAPK活性的效果(數據未顯示)。於HLEC細胞中,兩株新開發的抗體對於因VEGF-A而誘導的訊息傳遞具有類似的效果,皆可抑制VEGFR2與VEGFR3的自身磷酸化(數據未顯示),但相反的,只有R2C9-11能有效的抑制因VEGF-C而活化的訊息傳遞路徑,進而抑制VEGFR3的活性(數據未顯示)。
綜上所述,雖然R2C7-1與R2C9-11於阻斷VEGFR2的訊息傳遞路徑的機制不同,但兩者都具有雙重阻斷的特性,可阻斷VEGFR2與VEGFR3的受體媒介的訊息傳遞活化。
實施例5、阻斷2維(2D)空間與3維(3D)空間的管形成
為檢驗新開發抗體所具備之減弱VEGF-A、VEGF-C誘導的訊息傳遞功能是否能調節HUVEC、HLEC細胞的生物行為,因此進行了2D空間的管形成試驗。先將HUVEC與HLEC細胞接種於塗有基質膠且缺乏VEGFs與任何營養補充物的的孔盤上,經Ramucirumab、R2C7-1、R2C9-11或同種型控制組抗體(IgG2a)預處理後,給予VEGF-A或VEGF-C刺激10小時,再進行影像擷取,接著使用Image J的血管新生分析儀進行管網(tube meshes)、主要管段(master segments)的總數定量,其結果顯示,相比於Ramucirumab抑制因VEGF-A或VEGF-C誘導的血管生成的能力,R2C9-11具有較佳或相當的抑制效果(如圖3),然而相較之下,R2C7-1單株抗體的抑制效果較弱。
為進一步檢驗其於3D空間中抑制發芽纖維(sprouting fibrin)形成的效果,係將HUVEC、HLEC細胞接種於含有CFSE螢光染劑的超低平板(ultra-low plate)上以形成細胞球體,之後轉移制塗有基質膠的孔盤上,以含有R2C7-1、R2C9-11、Ramucirumab或同種型控制組抗體(IgG2a)的培養基培養7天。使用NIS-AR軟體進行發芽區域與螢光強度的分析,結果顯示,與控制組細胞相比,所有的抗體皆顯示顯著性的抑制發芽纖維生成的能力(P>0.001,如圖4A、4B),且值得注意的是,該新開發的抗體皆比Ramucirumab具有更佳的抑制效果,綜合以上,R2C7-1與R2C9-11單株抗體具有不同的抑制血管、淋巴管新生的能力,其差異係取決於所使用的實驗模型。
實施例6、體內由外被細胞所誘導之淋巴管新生
部分文獻已證明外被細胞(pericyte)於血管新生和淋巴管新生中扮演重要的角色(Ozerdem U, 2006, Ophthalmic Res. 2006;38(5):251-4),因此為了檢測以WI38纖維母細胞作為外被細胞是否可促進體內淋巴管新生,使用HLEC細胞作為實驗模型並進行基質膠栓試驗,將HLEC/WI38細胞混合後植入冰冷的基質膠中,接著將基質膠經皮下接種至NOD-SCID小鼠的腹側,並分別於兩週或四週後將已聚合的基質膠切除,且固定、包埋至石蠟中。後續進行免疫組織化學染色分析,使用可針對人類α-小肌肉肌動蛋白(α-small muscle actin, SMA)與CD31的特異性抗體進行雙重染色,且分別以3,3’-diaminobenzidine (DAB)、3-amino-9-ethylcarbazole(AEC)呈色,以分析組織中基質膠內的人類WI38與HLEC細胞。結果顯示,於接種兩週後,可見被人類SMA特異性抗體所標定出的WI38細胞浸潤於小鼠組織中新生成的血管,早於可被人類CD31特異性抗體所標記的新生人類淋巴血管;於接種四週後,則可見HLEC細胞從右下方移動至左上方,以形成更大的淋巴管(數據未顯示),以上的數據可證實外被細胞確實於淋巴管新生中扮演重要的角色。
實施例7、減少體內血管新生和淋巴管新生
使用與上述相同的實驗模型,將HUVEC或HLEC細胞與WI38先混合後植入基質膠中,再將基質膠經皮下接種至NOD-SCID小鼠,接著將給予小鼠每週兩次的Ramucirumab、R2C7-1、R2C9-11或控制組抗體處理,共持續四週。使用NIS-AR軟體定量經AEC部分或完全染色的管,結果顯示相較於控制組,經R2C7-1或R2C9-11處理的小鼠的新生血管數量有顯著性的減少,且效果比Ramucirumab更好,尤其是在淋巴管新生的部分(如圖5),效果更佳。
此外,也進一步測定血管尺寸的比例,結果顯示給予R2C7-1、R2C9-11或Ramucirumab處理的小鼠,其血管管徑大於50μm的比例顯著性的下降,然而相反的,其血管管徑小於50μm的比例則顯著性的上升(如圖6)。以上現象亦也同時出現於使用HLEC細胞的淋巴管新生實驗模型上,其結果也顯示相較於Ramucirumab,經給予R2C7-1和R2C9-11處理可更有效的抑制淋巴管的生成(數據未顯示),因此,以上結果顯示相較於Ramucirumab,該可雙重標靶VEGFR2與VEGFR3的新開發抗體可提供更佳的抑制血管新生與抑制淋巴管新生效果。
實施例8、體內和體外的抗體依賴性之腫瘤細胞毒性
由於該新開發的抗體可特異性的標靶VEGFR3,因此進一步檢測其對表現VEGFR3的細胞是否具有細胞毒性。於本實施例中係使用HEL92.1.7細胞株,首先,先檢測其VEGFR2與VEGFR3的表現情形,透過流式細胞儀可確認其具高表現量(~95%)的VEGFR3與低表現量(~5%)的VEGFR2(如圖7A),接著從NOD-SCID小鼠體內分離出周邊血單核細胞(peripheral blood mononuclear cells, PBMCs),以進行抗體依賴性之細胞毒性試驗(ADCC)。其結果顯示,所有測試的抗體(包括Ramucirumab)於含有PBNCs與2%嬰兒血清的情況下,都明顯具有殺死HEL92.1.7腫瘤細胞的能力,甚至當抗體濃度低至只有0.125μg/ml的情況下,也具有此效果(如圖7B)。
以下,將進一步透過NOD-SCID小鼠的異種移植腫瘤來檢驗抗體的抑制腫瘤能力,具VEGFR3高表現量(~95%)或低表現量(~5%)的細胞被分選出來並擴增,分別接種至小鼠的左側或右側,於腫瘤接種7天後,開始給予小鼠每週兩次的R2C7-1、R2C9-11、Ramucirumab或IgG控制組抗體,共持續25天。結果顯示,該新開發的抗體對於不論是具高或低表現VEGFR3的腫瘤皆具有與Ramucirumab相當的治療效果(如圖8)。綜上所述,R2C7-1與R2C9-11除了具有抑制血管新生與淋巴管新生的雙功能,還可觸發ADCC的毒殺效果,進而抑制表現VEGFR3的腫瘤生長,並且於25天的療程當中,具有與Ramucirumab相當的治療效果。
實施例9、新開發的抗體與Ramucirumab的合併療法
為進一步檢測若合併使用新開發的抗體與Ramucirumab是否能提供更好的抑制腫瘤能力,係以帶有HEL92.1.7腫瘤細胞的小鼠做為實驗模型(每組10隻),分別單獨給予R2C7-1、R2C9-11或Ramucirumab或其組合。於小鼠接受接種七天後,每週給予兩次抗體處理,共持續35天(總劑量為10 mg/kg)。結果顯示,與單獨給予R2C7-1或R2C9-11之處理組相比,合併給予R2C7-1或R2C9-11與Ramucirumab並不會有更好的腫瘤抑制的治療效果,反而單獨給予R2C9-11處理是所有抗體處理組中,呈現最佳治療效果的組別(如圖9所示)。因此,以上數據顯示,標靶表現VEGFR3的腫瘤細胞(如HEL92.1.7),不但可降低其細胞存活,在有嬰兒血清中活化的補體的情況下,還可以因被抗體標靶後而產生細胞毒性。
實施例10、確認新開發抗體上的表位
為確認R2C7-1與R2C9-11所識別的表位,係進行細胞型競爭性ELISA(enzyme-linked immunosorbent assay)試驗。將HEK293T/VEGFR2轉染細胞貼附於96孔盤上並固定(5x104
細胞/孔)。使用標準ELISA程序,測定R2C7-1與R2C9-11抗體與細胞結合的最佳濃度,及對數期的最高濃度(數據未顯示)。接著,使用特定劑量的VEGFR2抗原的單個類免疫球蛋白結構域(R2-IgDs)吸收最佳濃度的抗體,以競爭其結合表位。結果顯示,R2C7-1與R2C9-11的主要識別的表位係介於IgD6與IgD7之間,但兩抗體間有些微的差異,R2C7-1比R2C9-11對IgD6具有更強的結合能力(如圖10)。為了進一步確定該新開發抗體的表位,因此對其進行丙胺酸掃描誘變試驗(alanine scanning mutagenesis),顯示VEGFR2的IgD6至IgD7之間任何的胺基酸改變,都會影響兩種抗體對其辨識的結合能力(數據未顯示)。以上結果表示,本發明之新開發抗體所識別的表位應為結構性表位。
使用上述相同方法來測定VEGFR3上抗體結合的表位,以HEK293T/VEGFR3轉染細胞做為結合對象,用來測定抗體對VEGFR3結合的最佳濃度(數據未顯示)。接著,使用特定劑量的VEGFR3抗原的單個類免疫球蛋白結構域(R3-IgDs)吸收最佳濃度的抗體,以競爭其結合表位。結果顯示,R2C7-1與R2C9-11的主要識別的表位係位於VEGFR3的IgD6,而非IgD7(如圖11A)。為進一步確認此結果,係針對不同的重組IgD蛋白進行原態膠體電泳的西方墨點法,由於先前文獻已指出VEGFR3受體於生物合成過程中,IgD5會經蛋白水解反應而被切除(參見Pajusola K et al., 1994, Oncogene. 9(12):3545-55),而且於本實施例中,也並未於細胞型ELISA中顯示其具有競爭性(如圖11A),因此後續只使用E.coli系統合成IgD1-7、IgD3-4、IgD6-7等片段。於非還原膠體電泳後,使用R2C7-1、R2C9-11抗體來免疫探測該純化的蛋白質,進而顯示該抗體對IgD6-7有高度的結合能力,但對IgD3-4卻無此現象(如圖11B)。綜上所述,此些數據皆顯示VEGFR2與VEGFR3上,R2C7-1與R2C9-11結合的表位都位於IgD6-7,且VEGFR3蛋白上的識別位特定的位於IgD6。
實施例11、互補決定區(Complementarity-determining regions, CDRs)和結合親和力
為確定抗體的互補決定區,使用Kabat、Chothia系統來確立R2C7-1和R2C9-11抗體的可變區(variable regions)。首先,為了確定分泌兩個抗體的融合瘤為單株細胞,係將細胞經過連續兩次的連續稀釋來篩選細胞,並使用5’-RACE或具特異性的5'-/3’-IgG退化性引子,複製出帶有抗體序列的基因片段,參見表1。從該新開發抗體中複製出的互補決定區係列示於表2。R2C7-1與R2C9-11兩者經由Kabat、Chothia系統定義的重鏈CDR1(CDR-H1)與CDR2(CDR-H2)胺基酸序列,結果有很大的不同,但於其重鏈CDR3(CDR-H3)則無分別。至於其輕鏈的CDR,兩系統所鑑定出來的結果顯示兩抗體並無差異。綜上所述,雖然R2C7-1和R2C9-11對於VEGFR2、VEGFR3的IgD6-7結構域都具有結合潛力,但有可能是因為R2C7-1和R2C9-11之間的CDR胺基酸序列有差異,因此才導致兩抗體對於VEGFR2、VEGFR3上有不同的結合表位。

為檢測R2C7-1、R2C9-11的結合親和力,係以VEGFR2(KDR)和VEGFR3(Flt-4)的重組IgD4-7與IgD6-7做為標靶抗原,並使用Biacore系統測量其結合親和力。結果如表3,顯示R2C9-11對VEGFR2和VEGFR3的IgD4-7與IgD6-7抗原的親和力高於R2C7-1,且任一種抗體都顯示與VEGFR2的結合強於與VEGFR3的結合,除此之外,兩種新開發的抗體與VEGFR2或VEGFR3的IgD4-72的結合都略高於與IgD6-7的結合,這些數據也顯示R2C7-1與R2C9-11所識別的表位取決於VEGFR2與VEGFR3的細胞外結構域的3維結構。

材料與方法
(1)細胞培養
NHK293T細胞以含有10%FBS和1X抗生素-抗真菌劑(GIBCO;Grand Island, NY)的DMEM於37℃在5% CO2
的培養箱中培養;人類臍靜脈內皮細胞(HUVEC)則以添加各種生長因子、細胞激素和其他SingleQuot試劑組(Lonza;Basel, Switzerland)中的補充劑的內皮細胞基礎培養基-2(Endothelial basal medium-2, EGM-2),於37℃在5% CO2
的培養箱中培養,於試驗中使用的初代細胞約為第3至第6代子代細胞;人類淋巴管內皮細胞(Lymphatic endothelial cells, LECs)購買自Promocell (Heidelberg Germany),並依照其製造者指示,培養於內皮細胞培養基MV (Promocell)。
(2)融合瘤的產生
於兩週內,使用血管內皮生長因子受體2(VEGFR2)的人類重組免疫球蛋白(immunoglobin, Ig)結構域4-7來腹腔免疫BALB/C小鼠共3次。使用基因特異性的引子與人類VEGFR2 cDNA做為模板,透過PCR編碼出帶有VEGFR2的細胞外結構域4-7的基因,並複製到pGEX-KG載體中,最後所產生的質體經由直接定序確認後,命名為pGEX-VEGFR2-ED Ig D4-7。該編碼的蛋白質是被表現於大腸桿菌表現系統,並經glutathione-conjugated Sepharose beads(Sigma-Aldrich;St. Louis, MO)進行純化,相關方法可見下文所述的「基因構築和表達」。採集小鼠的血清並使用間接型的ELISA檢測是否有抗VEGFR2抗體的存在,在偵測到高程度的抗體力價(>1:3000稀釋)後,會再對小鼠進行最後一次的IP注射增強,給予不含Freund’s佐劑的25μg重組抗原,於最後一次增強的五天後,犧牲小鼠及採集其脾臟細胞,並使用改良式的融合瘤技術(Apiratmateekul, N., P. Phunpae, and W. Kasinrerk, Cytotechnology, 2009. 60(1-3): p. 53.)將脾臟細胞與骨髓瘤細胞融合。簡述之,使用50%聚乙二醇4000(PEG 4000;Sigma-Aldrich, St. Louis, MO)將脾臟細胞(7x107
個細胞)與2x107
Sp2/0-Ag14細胞(ATCC CRL-1581 TM)以標準的融合瘤技術進行融合,經HAT培養基(Sigma-Aldrich)篩選後,使用標準的間接型ELISA分析從含有融合瘤的孔中獲得的培養物上清液,而於450nm (OD450nm
)下,光密度值≥0.60的融合瘤判定為陽性,並將之以限數稀釋技術進行單細胞的複製。使用鼠單株抗體同種型試劑組(Thermo Scientific Pierce, Rockford, IL)測定被分泌出的小鼠單株抗體的種型。
(3)抗體的純化
使用融合瘤無血清培養基(Gibco)於無血清的情況下培養可生產抗VEGFR2單株抗體的融合瘤,並使用TOYOPEARL AF-rProtein A (Griesheim, Germany)進行親和層析法從培養物的上清液中純化出單株抗體,經冰冷的PBS(10x bed volume)清洗過後,被Protein A磁珠純化的抗體經Elution緩衝液(R2C7-1的為50 mM glycine-HCl, pH 3.0;R2C9-11的為50 mM glycine-HCl/150 mM NaCl, pH 3.0)溶析出該抗體,每1mL溶析的流份(fraction)收集於含有100μL中和緩衝液(1 M Tris-HCl, pH 8.0/1.5 M NaCl/1 mM EDTA) 的管中,並分別經十二烷基硫酸鈉聚丙烯酰胺凝膠電泳(SDS-PAGE)和間接ELISA測定純度與活性,該單株抗體的濃度則由Bradford蛋白質測定法(Bio-Rad, Hercules, CA),並於4℃下使用PBS進行透析過夜,最後所得的抗VEGFR2抗體儲存於-80℃以備後續使用。
(4)管生成試驗
先將培養用孔盤以冰冷的基質膠(80 μL/孔;BD Biosciences, San Jose, CA)進行包被,並於37℃下培養30分鐘以形成凝膠。LECs與HUVECs先於缺乏VEGFs和其他補充劑的情況下,培養於EGM-2中飢餓8小時,接著經胰蛋白酶處理後,將細胞再懸浮於含有Ramucirumab(5 μg/mL;Eli Lilly, Indianapolis, IN)、抗VEGFR2單株抗體(20 μg/mL;R2C7-1或R2C9-11)的培養基中,並添加2μg/mL的VEGF-A165(Cys156Ser)或VEGF-C(R&D; McKinley Place, NE),接著於37℃下將LEC或HUVEC接種至預先包被好的孔盤上培養,分別培養4小時或10小時。使用NIKON Digital Slight DS-U3系統(Tokyo, Japan)拍攝孔盤的全影像,並透過Image J的Angiogenesis Analyzer進行分支節點、管網、管長的總數定量,數據皆為三重複並以平均值±S.D.值呈現。
(5)流式細胞儀分析
使用磷酸鹽緩衝生理鹽水(PBS)沖洗細胞,並溶於以PBS為基底的EDTA中,再經離心分離收集細胞,並於4℃將細胞培養在含有1%牛血清蛋白的PBS中1小時,以阻斷其產生非特異性的結合,後續經PBS沖洗後,再於4℃下,以2μg/mL的抗VEGFR2單株抗體(R2C7-1或R2C9-11)、Ramucirumab (Eli Lilly, Indianapolis, IN)或同種型控制組免疫球蛋白G (IgG)處理表現VEGFR2的HUVEC或HEK293T細胞(1x105
/反應)1小時。為了檢測VEGFR3的表現,先以VEGFR3抗體(Santa Cruz, Dallas, TX)或控制組的兔子IgG的抗體(Abcam, Burlingame, CA)為免疫探針,偵測表現VEGFR3的LEC或HEK293T細胞,後續經含1%BSA的PBS充分沖洗後,以各自相對應的二級抗體,分別為4μg/mL帶有PE的山羊抗小鼠IgG(Biolegend)、帶有FITC的驢抗兔子IgG(Abcam)或帶有PE的小鼠抗人IgG(Biolegend),於4℃下染色細胞1小時,接著再沖洗一次細胞,並重新懸浮於含1%多聚甲醛、1%BSA的PBS中,並使用BD FACSCaliburTM
流式細胞儀(Becton Dickinson, Franklin Lakes, NJ)分析細胞表面上VEGFR2和VEGFR3的表現情形。
(6) 原位鄰位連接分析法(In situ proximity ligation assay, PLA)
依照製造商的使用說明,使用Duolink in-situ PLA試劑組(Sigma-Aldrich, St. Louis, MO)檢測抗VEGFR2單株抗體(R2C7-1、R2C9-11)阻斷VEGFR2與VEGFR3的同質/異質二聚化的效果。簡言之,LECs接種於8孔的細胞培養用玻片,並培養於含有R2C7-1、R2C9-11或Ramucirumab (Eli Lilly)的無血清培養基中16小時,接著於分別使用VEGF-C (500 ng/mL共15分鐘)、VEGF-C (Cys156Ser) (2 μg/mL共10分鐘)或VEGF-A165 (50 ng/ml共5分鐘)刺激細胞後,以PBS沖洗細胞,並以4%多聚甲醛固定15分鐘,再以阻隔緩衝液(Blocking buffer, Sigma-Aldrich)於室溫下作用30分鐘,後續進行細胞染色,以2 μg/mL市售的抗VEGFR2或VEGFR3(Santa Cruz, Dallas TX)抗體染色做為控制組,並添加可與特定寡核甘酸(PLA探針)連接的各自相對應的物種特異性二級抗體,透過加入含有連接酶(Sigma-Aldrich)的溶液(最終濃度為1 單位/μL)於37℃下作用30分鐘已完成PLA探針的連接反應,並將連接完成的PLA探針於37下與含有聚合酶(Sigma-Aldrich)的溶液(濃度為10 單位/μL)作用100分鐘,以擴增訊號,之後以緩衝液B(Sigma-Aldrich)沖洗,再以DAPI染出玻片上的細胞核,然後以Duolink in-situ mounting medium(Sigma-Aldrich)將玻片封片,另外,使用同種型的IgG做為控制組,以上染色的螢光訊號(或點)皆以MetaMorph Offline軟體進行影片成像與分析。
(7) 共免疫沉澱分析(Co-immunoprecipitation)
為檢測抗VEGFR2抗體對VEGF-C所誘導的受體二聚化是否具有抑制作用,先將LEC細胞接種於直徑6公分的培養皿上,直到細胞長至70-80%滿,接著將細胞沖洗乾淨後,培養於含有20 μg/mL抗體(R2C7-1、R2C9-11或同種型控制組抗體)的無血清培養基中16小時,隨後,將細胞分為以VEGF-C(500 ng/mL)刺激15分鐘,或無使用VEGF-C處理,然後先以冰冷的PBS沖洗兩次,再使用RIPA緩衝液(20 mM Tris-HCl (pH 7.5)/ 150 mM NaCl/ 1 mM Na2EDTA/ 1 mM EGTA/ 1% NP-40/ 1% sodium deoxycholate/ 2.5 mM sodium pyrophosphate/ 1 mM β-glycerophosphate/ 1 mM Na3VO4/ 1 µg/mL leupeptin)收集細胞。於免疫沉澱試驗,裂解物(1 mg/樣品)先使用Protein-A/G-agarose磁珠(Thermo Fisher; Waltham, MA)進行預處理,以最小化其他非特異性的結合,之後再與抗VEGFR3抗體(1 μg/樣品, Santa Crus)共培養,以連續旋轉方式於4℃隔夜培養,免疫沉澱的蛋白質以天然的聚丙烯醯胺膠體電泳的方式分離,並以市售的VEGFR3抗體(Santa Crus)進行免疫墨點,再使用ECL 西方墨點系統(Thermo Scientific)進行呈色,如下所述。
(8) 原態膠體電泳
使用原態膠體電泳(PAGE)測量VEGFR2與VEGFR3的受體二聚化狀態,首先細胞先飢餓培養於含有20 μg/mL的R2C7-1或R2C9-11的無血清培養基16小時,並分別以Ramucirumab、同種型控制組抗體為陽性對照與陰性對照組,接著以VEGF-C(500 ng/ml)刺激細胞15分鐘後,以低滲透裂解緩衝液(20 mM Tris-HCl, pH 7.4、10 mM NaCl、1 mM PMSF)收集細胞,將等量的蛋白質樣品與4x天然樣品緩衝液(400 mM Tris-HCl, pH7.5、40% glycerol、 0.02% bromophenol blue)混合,再利用預先製作好的PAGE膠(Bio-Rad)進行原態膠體電泳,並透過抗VEGFR2、抗VEGFR3或抗p-VEGFR3抗體(Santa Cruz、Santa Cruz、Cell Applications; San Diego, CA)檢測VEGFRs的二聚化和磷酸化狀態。
(9) 西方墨點分析
使用裂解緩衝液(20 mM Tris-HCl pH 8.0/ 2 mM EDTA/ 1 mM Na3VO4/ 10% glycerol/ 1% Triton X-100/protease)並加入磷酸酶抑制劑混合物(Sigma-Aldrich)或RIPA緩衝液裂解細胞,以得蛋白質裂解物。HUVEC與LEC細胞於處理前一天先接種,HUVEC細胞先添加抗VEGFR2抗體,如R2C7-1或R2C9-11,於培養基中(5, 10, 20 µg/mL),並於37℃下處理10分鐘,後續再以20 ng/mL的VEGF-A165(R&D)刺激HUVEC,而LEC細胞則先與R2C7-1或R2C9-11抗體(20 μg/mL)預培養,再以VEGF-C(500 ng/mL共15分鐘), VEGF-C (Cys156Ser) (2 μg/mL共10分鐘) or VEGF-A165 (50 ng/mL共5分鐘) (R&D)刺激,之後以RIPA緩稱液收集細胞,並分別以Ramucirumab(5 µg/mL;Eli Lilly)與同種型控制組IgG(R2C7-1與R2C9-11使用小鼠IgG(Biolegend); rabbit IgG for VEGFR3抗體使用兔子IgG(Abcam))為陽性對照組與陰性對照組,萃取得到的蛋白質濃度以Bradford蛋白質測定試驗(Bio-Rad)測量,並取同量的總蛋白質(50 µg/樣品)以12%十二烷基硫酸鈉聚丙烯酰胺凝膠(Bio-Rad)進行電泳分離,再將蛋白質轉移到硝酸纖維素膜上(HybondTM-P; Amersham Biosciences, Piscataway, NJ),經阻隔緩衝液(3% BSA/0.1% Tween-20 in TBS)於常溫進行組隔作用兩小時後,該膜再與抗VEGFR2 (Santa Cruz)、抗VEGFR3(Santa Cruz)、抗p-VEGFR3(Tyr1230/1231) (Cell applications)、抗p-FAK、抗p-p38ERK(Cell Signaling; Danvers, MA)、抗β-Actin(MilliporeSigma; Burlington, MA)或抗tubulin的初級抗體於4℃隔夜培養,後續再與各自對應的帶有辣根過氧化物酶(horseradish peroxidase,HRP)二級抗體培養,並使用SuperSignal West Pico 化學發光(chemiluminescence) (Thermo Scientific)使有產生免疫反應的蛋白質呈色,並以X放射線底片(FUJU; Tokyo, Japan)檢測信號。
(10)細胞型競爭性ELISA
使用細胞型的競爭性ELISA測定抗VEGFR2抗體R2C7-1、R2C9-11的抗體表位。先將表現VEGFR2或VEGFR3的HEK293T細胞以每孔50000細胞/100μL的濃度接種於96孔盤上,並以37℃進行隔夜培養,經充分沖洗後,使用含1%戊二醛的PBS溶液固定細胞,並以含0.1%Tween-20的PBS溶液(PBST)沖洗三次,接著以含3%BSA的PBS溶液於37℃處理細胞30分鐘,以避免產生非特異性的結合,另一方面,R2C7-1、R2C9-11抗體(8 μg/孔盤)被指定劑量(0.2, 1.0, 5.0 μg/孔盤)的VEGFR2或VEGFR3重組蛋白的單個類球蛋白結構域(IgD4、D5、D6、D7)於37℃下吸收,共處理30分鐘,之後再將表現VEGFR2或VEGFR3的細胞與該經過預吸收的R2C7-1或R2C9-11抗體於37℃共培養1小時,並以大量的PBST沖洗3次,最後使用帶有HRP的山羊抗小鼠IgG為探針,於37℃下作用1小時以偵測結合的抗體,接著加入ABTS呈色(37℃,處理30分鐘),並以SpectraMax M5 microplate reader (Molecular Device; San Jose, CA)於OD405
下測量。
(11)基因構築與表達
將VEGFR2的細胞外球蛋白(Ig)結構域構築至pGEX-4T-1質體(GE Healthcare; Chicago, IL)中,並於勝任大腸桿菌細胞系統中表現重組蛋白。該VEGFR2的細胞外球蛋白結構域與其次級結構域(subdomain)6、7的基因(NM_002253.2)是以質體EX-I0114-Lv205 (GeneCopoeia; Rockville, MD)為模板,使用基因特異性引子進行PCR製造而成,並以BamH1、Xhol(NEB; Ipswich, MA)進行限制性消化再複製至pGEX-4T-1載體(GE Healthcare; Chicago, IL),且分別命名為VEGFR2-ED Ig D1-7、D6-7。再將基因構體於42℃下以熱休克方式處理40秒,以將之轉型至JM109細胞(Promega; Madison, WI),並於LB瓊脂上以Ampicillin(50μg/mL)篩選,再以標準菌落PCR和基因定序確認具抗生素抗性細胞中的插入基因,陽性細胞培養於含有Ampicillin的LB培養基中,以1mM的IPTG(Sigma-Aldrich)於37℃處理3小時,誘導細胞表達蛋白質,後續沉澱細胞,打散懸浮於冰冷的裂解緩衝液(10 mM Tris-HCl pH8.0/0.15 M NaCl/1 mM EDTA)並添加100μg/mL的溶菌酶、10 mM的PMSF、5單位的benzonase(Sigma-Aldrich),經音波震動處理後,以10000xg離心30分鐘獲得可溶性片段,並以帶有glutathione的Sepharose磁珠(Sigma-Aldrich)純化出GST標記的重組蛋白,再以10 mM的還原態glutathione溶析出蛋白質與PBS透析過夜,最後以SDS-PAGE測定重組蛋白的純度。
為了於真核細胞中表達VEGFR3的細胞外結構域基因,係將欲插入的基因複製至pCMV6-XL6-myc載體(OriGene; Rockville, MD)以表達該蛋白質。簡言之,以pCMV6-XL6-VEGFR3 (來自J.L.Su博士)為模板,使用基因特異性引子進行PCR而擴增得VEGFR3的細胞外Ig結構域與其次級結構域的基因(NM_002020.4) 。於限制性消化後,將插入片斷轉殖至pCMV6-XL6(OriGene; Rockville, MD)的EcoR1與Xbal位點中,再轉型至DH5α細胞(Thermo Scientific)以放大基因,並以直接定序的方式確定所得之VEGFR3-ED Ig D1-7、D3-4、D6-7-myc質體。
為了表達和純化由上述質體所編碼的重組蛋白,將之以Turofect轉染試劑(Thermo Scientific)轉染至HEK293細胞,步驟皆參照產品說明書指示,並使用anti-Myc antibody-mobilized管柱純化出編碼的蛋白質,再經Native Page Novex Bis-Tris gel system(Invitrogen; Carlsbad, CA)溶於原態PAGE膠體中,然後於硝酸纖維素膜上進行免疫墨點。另外,為了檢測VEGFR2抗體的交叉反應,再使用R2C7-1或R2C9-11抗體為探針針測墨跡,並以山羊抗小鼠IgG-HRP(Santa Cruz)抗體檢測,接著以SuperSignal West Pico chemiluminescence(Thermo Scientific)使訊號呈色。
本說明書中公開的所有技術特徵可以任意組合方式組合,本說明書中公開的每個技術特徵可以由相同、等同或類似目的的替代技術特徵取代,因此,除非另有明確說明,否則所公開的每個技術特徵僅為一系列相同或類似技術特徵的實施例。

圖1為流式細胞儀直方圖組,顯示分別使用單株抗體(monoclonal antibodies, mAbs)R2C7-1與R2C9-11辨識VEGFR2與VEGFR3,圖1A與1B分別為初代人類臍靜脈內皮細胞(human umbilical vein endothelial cells, HUVEC)與人類淋巴管內皮細胞(human lymphatic endothelial cells, HLEC),係使用其第3到第6子代的初代細胞進行實驗;圖1C與圖1D分別為過度表達VEGFR2與VEGFR3的HEK293T細胞(HEK293T/VEGFR2, HEK293T/VEGFR3),如圖所示,HEK293T細胞可穩定的被轉染人類VEGFR2或VEGFR3基因,並使用可分別針對VEGFR2或VEGFR3的抗體(Abcam9530, R&D54703)於BD FACSJazzTM
細胞分選儀,免疫篩選出具高度表達VEGFR2或VEGFR3的轉染子(transfectants),接著將全部的初代細胞或轉染子(1x105
細胞/反應)以R2C7-1或R2C9-11(10 μg/mL;箭頭)進行染色,同時以IgG2a同種型抗體(CTL)做為陰性控制組,而以Ramucirumab (RAM)(10 μg/mL)為VEGFR2的陽性控制組,除此之外,僅被染上二級抗體的細胞將視為背景值,而染色呈陽性的細胞則以總細胞的百分比方式呈現(圖1C、1D;最上部)。
圖2為阻斷VEGFR2與VEGFR3之受體二聚化的墨點圖與統計圖,圖2A為共免疫沉澱之結果。將具有高VEGFR3表現量與低VEGFR2表現量的紅白血病細胞HEL92.1.7(1x1010
細胞/反應),給予R2C7-1、R2C9-11或Ramucirumab(20 μg/mL;RAM)處理,或是未經任何處理,並以IgG2a同種型抗體(CTL)為陰性控制組,於37℃下處理24小時後,細胞再經胰蛋白酶處理與離心,並以1%(w/v)的多聚甲醛固定並用甘胺酸溶液中和,最後用PBS洗滌。於經離心而取得的可溶性片段中加入25μg針對VEGFR2的抗體(Abcam9530),並於室溫下培養2小時。與抗體接合的蛋白質複合物可透過Protein A/G進行免疫沉澱(immunoprecipitated, IP),並對HEL92.1.7細胞的VEGFR3進行免疫墨點(immune-blotted, WB,圖左),而全可溶性片段以VEGFR3和β-肌動蛋白特異性抗體進行免疫探測以達標準化。相同的,為了沉澱VEGFR3並對VEGFR2做墨點分析,係將HEK293T/VEGFR2細胞經帶有VEGFR3之質體進行轉染(圖右)。於轉染24小時後,將轉染細胞給予R2C7-1、R2C9-11或Ramucirumab(20 μg/mL)處理,或是未經任何處理,並繼續以前述方式進行免疫沉澱。圖2B為鄰位連接分析法(Proximity ligation assay, PLA)之結果圖,藉由PLA檢測單株抗體R2C7-1與R2C9-11(各10μg/mL)抑制VEGFR2和/或VEGFR3受體二聚化的抑制效果。人類淋巴內皮細胞(human lymphatic endothelial cell; HLEC)接種並培養於含有R2C7-1、R2C9-11或Ramucirumab(RAM)的無血清培養基中16小時,隨後以VEGF-A165
或VEGF-C刺激細胞。每個實驗皆進行三重複,根據代表圖顯示其螢光產物的存在,來證明VEGFR2和/或VEGFR3的同質或異質二聚化為緊密接近(>40 nm)。細胞核以DAPI染色,其螢光訊號係使用Image J analysis於螢光顯微鏡下以40x放大倍率進行形象化與量化,數據結果皆以平均值±S.D.值表示,係表示於未經抗體處理的細胞中(CTL)所佔之百分比,此外,經同種型對照(IgG2a)處理的細胞亦做為另一控制組,而細胞未經任何刺激的,或細胞以VEGF-A165
或VEGF-C刺激的分別用空白、雙交叉或灰色條表示。使用t
-test計算有經抗體處理之細胞與未經抗體處理之細胞間的統計學上顯著差異,*表示P >0.05。
圖3係顯示VEGFR2抗體對於人類臍靜脈內皮細胞(HUVEC)與人類淋巴管內皮細胞(HLEC)的阻斷效果。圖3A,HUVEC細胞培養於EGM-2,並且不給予任何補充物與VEGF,讓細胞飢餓8小時,之後經胰蛋白酶處理後,將細胞懸浮於含有10 μg/mL Ramucirumab、R2C7-1、R2C9-11或同種型控制組的抗體(IgG2a)的培養基中,並接種至已用基質膠包被的孔盤上,之後於37℃下使用2 μg/mL VEGF-A165
刺激細胞10小時,上述每個實驗皆為三重複;圖3B,類似上述步驟,一樣將HLEC培養於飢餓狀態下,並同樣以上述抗體進行預處理,且於37℃下給予2 μg/mL VEGF-C刺激細胞4小時。實驗結果皆以影像方式擷取,並使用Image J的Angiogenesis Analyzer定量管網(tube meshes)數目、主要接合(master junction)、管的長度、小管區域,數據相較於控制組(IgG2A) 以百分比表示(平均值±S.D.值,三重複),*、**、***分別表示P >0.05、P >0.01、P >0.001。
圖4係顯示各種VEGFR2抗體於3D血管新生模型中的抑制效果。內皮細胞(HUVEC與HLEC)以CFSE螢光來標記,並於37℃下培養於ultra-low 96孔盤中48小時,以形成細胞球體(cell spheroids)。之後,透過離心將細胞球體轉移並附著至經基質膠包被的孔盤上培養30分鐘,再將附著的細胞球體培養於含有10 μg/mL R2C7-1、R2C9-11、Ramucirumab(RAM)或同種型控制組抗體(CTL)的培養基中,並使用光學和螢光顯微鏡觀察與拍攝細胞球體核心的形狀,培養第7天後所擷取之圖像為HUVEV、HLEC的最終圖像,螢光強度皆由NIS-AR軟體進行分析,總面積與總發光強度中減去細胞球體核心可得到發芽纖維(sprouting fibrin)的面積與發光強度,發芽區域(sprouting area,圖4A)與其螢光強度(圖4B)之數據皆以平均值±S.D.值的百分比值呈現,並以t
-test測定各種VEGFR2抗體與CTL抗體之間的統計學上顯著差異,***表示>0.001。
圖5係顯示各種VEGFR2抗體於體內抑制血管新生與淋巴管新生的抑制效果。將HUVEC(1x106
)/WI38(2 x 105
)的細胞,或HLEC(1x106
) /WI38(2 x 105
)的細胞混合物懸浮於200μL的冰冷基質膠中,接著使用具備24英吋針頭的冰冷注射器,將該細胞混合物分別皮下接種於NOD-SCID小鼠的左右兩腹側。於細胞接種後隔天,開始進行每週兩次的靜脈注射,小鼠分別給予10 mg/kg劑量的R2C7-1、R2C9-11、Ramucirumab(RAM)或控制組抗體IgG2a(CTL)並持續4週。之後切下小鼠腹側已聚合的基質膠,以10%福馬林固定隔夜,並包埋於石蠟後切為5 μm厚的組織切片,接著使用雙重染色的方法進行免疫組織化學染色分析,係使用具人類α-平滑肌肌動蛋白(α-SMA)特異性的抗體以針對WI38、人類CD31抗體辨別HUVEC、HLEC,後續使用NIS-AR軟體分析與定量呈AEC染劑陽性的血管或淋巴管的數量,數據以百分比方式(平均值±S.D.值)呈現,以比較三個獨立實驗與對照組之間的差異,且各個VEGFR2抗體與CTL抗體間的統計學上顯著差異皆經過t

圖6係顯示小鼠經VEGFR2抗體處理後,大、小血管的比率差異。將基質膠/HUVEC細胞混合物(1x106
細胞/500μL基質膠栓)經皮下接種的方式接種至NOD-SCID小鼠的腹部中線處。於接種後第7、14、21、28天,透過腹腔注射的方式給予小鼠10 mg/kg劑量的IgG1 CTL、Ramucirumab(RAM)、IgG2a CTL、R2C7-1、R2C9-11,並於第35天犧牲小鼠,該組織基質膠以10%福馬林固定24小時,包埋於石蠟並切為5 μm厚的組織切片,再以抗人類CD31(hCD31)抗體進行免疫組織化學染色來辨別HUVEC細胞的血管形成,以分析抗體於體內抑制血管新生的效果。使用顯微鏡測量呈hCD31陽性的血管直徑,並分成直徑小於50μm與大於50μm兩組,再接著定量兩組中的血管數量,左側圖組係顯示經各種不同抗體處理過後,於直徑大於50μm組別中血管數目/總血管數的比例;右側圖組係顯示經各種不同抗體處理過後,於直徑小於50μm組別中血管數目/總血管數的比例,兩次獨立實驗的數據皆以平均值±S.D.值呈現,各個VEGFR2抗體與其相對應之CTL抗體間的統計學上顯著差異皆經過t

圖7係顯示各種VEGFR2抗體的抗體依賴性細胞毒性(antibody-dependent cytotoxicity, ADCC)。圖7A為VEGFR2與VEGFR3的表現程度,如箭頭所示,分別使用具VEGFR2特異性的抗體(Abcam9530)或具VEGFR3特異性的抗體(R&D54703)進行HEL92.1.7細胞染色,受體的表現程度百分比係經由與呈現同種型控制組抗體(CTL)染色的細胞相比而得;圖式7B表示其抗體依賴性細胞毒性,HEL92.1.7細胞(5x104
/每孔,25μL)接種到96孔盤,並接著加入25 µL/每孔之所設定濃度的R2C7-1、R2C9-11或Ramucirumab (RAM),並於37℃下培養15分鐘。另外,使用PBMC分離試劑套組(MyBioSource) 採集NOD-SCID小鼠的周邊血單核細胞(peripheral blood mononuclear cell, PBMC),並將之以4.0x106
細胞/mL的濃度懸浮於含有4%幼兔血清的RPMI1640培養基中,之後將50μL的PBMC效應物與孔中的標的HEL92.1.7細胞以4:1的比例混合,再加入PrestoBlueTM
,最後將細胞混合物於37℃下培養20小時,後續透過測定其螢光強度(560nm/ 590nm)來鑑定細胞活力。
圖8係顯示各種VEGFR2抗體抑制腫瘤生長的能力。使用流式細胞分選儀(Beckman Coulter CytoFLEX S),分離出具有VEGFR3高表現量的HEL92.1.7細胞(即為圖8A中的VEGFR3-H,其佔比於全族群中大於95%),及呈VEGFR3低表現量的HEL92.1.7細胞(即為圖8B中的VEGFR3-L,其佔比於全族群中小於5%),並進行細胞擴增。分別將高表現量與低表現量的細胞透過皮下注射的方式,注射至同一小鼠背側的右側與左側(1x106
/小鼠),於腫瘤接種後第7天,以5隻小鼠為一組,每組分別給予5 mg/kg劑量的特定抗體處理(RAM、R2C7-1、R2C9-11),每週給予兩次,共持續25天,同種型控制組IgG2a(CTL)也以同樣步驟進行,每週皆測量每隻小鼠的腫瘤大小兩次,於小鼠犧牲後測量其平均腫瘤大小(圖8A與圖8B中的小圖),不同抗體處理組與對照組的組間差異係由t-test分析,*、**分別表示P >0.05、P >0.01。
圖9係顯示不同VEGFR2單株抗體合併使用下的抑制腫瘤生長能力。將HEL92.3.1細胞經皮下接種方式,接種至NOD-SCID小鼠(1x106
/小鼠),如圖所示,10隻小鼠為一組,共6組,於接種7天後,透過靜脈注射分別給予R2C7-1、R2C9-11、RAM(每隻小鼠劑量皆為5 mg/kg),或是RAM(5 mg/kg)再加佐R2C7-1、R2C9-11(5 mg/kg)其中一種的組合,每週兩次,共持續35天,於只施以單種VEGFR2抗體的小鼠,另給予同樣劑量的IgG2a同種型控制組抗體(CTL),使每隻小鼠體內的抗體總量皆為10 mg/kg。使用修改過的橢圓型公式:[腫瘤體積 = 1/2(長度 × 寬度2
)] (Tomayko MM, Reynolds CP., 1989)計算每隻小鼠的腫瘤體積,數據皆以平均值±S.D.值表示腫瘤大小的平均值(如小圖),VEGFR2抗體與CTL抗體間的統計學上顯著差異係經由t

圖10係顯示R2C7-1、R2C9-11抗體於VEGFR2上的結合表位(binding epitopes)特徵。將表現VEGFR2(293T/VEGFR2)的HEK293T細胞,以每孔1x106
細胞/100μL的濃度接種於96孔盤中,並於37℃下隔夜培養。另外,使用標準ELISA程序,測定R2C7-1、R2C9-11抗體與細胞結合的最佳濃度,並將最佳濃度的R2C7-1、R2C9-11單株抗體(分別為200μg/mL、50μg/mL)與所設定劑量的VEGFR2抗原的單個類免疫球蛋白結構域(R2-IgDs)結合。將經戊二醛固定並洗滌的293T/VEGFR2細胞,與被結合的抗體於37℃下共培養30分鐘,後續使用帶有HRP的山羊-抗-小鼠IgG抗體探測被結合的抗體,接著添加ABTS呈色,並以SpectraMax微量盤式分析儀於OD405
下測量吸光值。數據皆以平均值±S.D.值表示同種型控制組(CTL)三重覆實驗的百分比,R2C7-1與R2C9-11於VEGFR2抗原上的潛在辨識表位位置(分別為上圖與下圖)由圖中橫跨IgD6與IgD7的黑線所示。
圖11係顯示於VEGFR3抗原上測定其與R2C7-1、R2C9-11抗體的結合表位。圖11A為細胞型競爭性ELISA結果圖,先將表現VEGFR3(293T/VEGFR3)的HEK293T細胞塗覆於96孔盤上。依前述,測得R2C7-1、R2C9-11單株抗體的最佳濃度(數據未示)。將最佳濃度的R2C7-1、R2C9-11單株抗體與所設定劑量之VEGFR3抗原的單個類免疫球蛋白結構域(R3-IgDs)結合。使用帶有HRP的二級抗體探測被結合的抗體,接著添加ABTS顯色,並以SpectraMax微量盤式分析儀於OD405

Claims (12)
- 一種單離抗體,係包含:(a)SEQ ID NO:5之重鏈可變區CDR1、SEQ ID NO:6之重鏈可變區CDR2、SEQ ID NO:7之重鏈可變區CDR3、SEQ ID NO:10之輕鏈可變區CDR1、SEQ ID NO:11之輕鏈可變區CDR2、SEQ ID NO:12之輕鏈可變區CDR3;或(b)SEQ ID NO:8之重鏈可變區CDR1、SEQ ID NO:9之重鏈可變區CDR2、SEQ ID NO:7之重鏈可變區CDR3、SEQ ID NO:10之輕鏈可變區CDR1、SEQ ID NO:11之輕鏈可變區CDR2、SEQ ID NO:12之輕鏈可變區CDR3;或(c)SEQ ID NO:13之重鏈可變區CDR1、SEQ ID NO:14之重鏈可變區CDR2、SEQ ID NO:15之重鏈可變區CDR3、SEQ ID NO:18之輕鏈可變區CDR1、SEQ ID NO:19之輕鏈可變區CDR2、SEQ ID NO:20之輕鏈可變區CDR3;或(d)SEQ ID NO:16之重鏈可變區CDR1、SEQ ID NO:17之重鏈可變區CDR2、SEQ ID NO:15之重鏈可變區CDR3、SEQ ID NO:18之輕鏈可變區CDR1、SEQ ID NO:19之輕鏈可變區CDR2、SEQ ID NO:20之輕鏈可變區CDR3;其中該抗體可與血管內皮生長因子受體2(VEGFR2)和血管內皮生長因子受體3(VEGFR3)產生特異性結合。
- 如請求項1所述之單離抗體,其中該抗體為一人源化抗體。
- 如請求項1所述之單離抗體,其中該抗體為一包含一Fc區、一Fab片段、一Fab'片段、一F(ab)'2片段,一單鏈抗體、一scFV多聚體、一單價抗體、一多特異性抗體或一嵌合抗體之抗體。
- 一種醫藥組合物,係包含如請求項1-3任一所述之單離抗體與一醫藥學上可接受的載體。
- 一抗體接合物,係包含請求項1-3任一所述之單離抗體與一分子。
- 一種使用請求項1-3任一所述之單離抗體製備抑制細胞中VEGFR2和/或VEGFR3的同質二聚化或VEGFR2和VEGFR3的同質二聚化的藥物上的用途,係包含使該細胞與該單離抗體接觸。
- 一種使用請求項1-3任一所述之單離抗體製備抑制細胞中VEGFR2和/或VEGFR3訊息傳遞路徑的藥物上的用途,係包含使該細胞與該單離抗體接觸。
- 一種使用請求項1-3任一所述之單離抗體製備抑制一個體之血管新生和/或淋巴管新生的藥物上的用途,係包含給予該個體一有效劑量的該單離抗體。
- 一種使用請求項1-3任一所述之單離抗體製備治療一個體之實體固態瘤的藥物上的用途,係包含給予該個體一有效劑量的該單離抗體。
- 如請求項9所述之用途,其中該實體固態瘤係選自膀胱癌、乳癌、子宮頸癌、大腸癌、直腸癌、腎癌、肝癌、皮膚癌、間皮瘤、肺癌、口腔癌、卵巢癌、胰臟癌、前列腺癌、肉瘤與甲狀腺癌。
- 如請求項9所述之用途,進一步包含給予該個體另一種治療劑。
- 一種檢測生物樣品中之VEGFR2和/或VEGFR3或其片段的方法,係包含:將該生物樣品與請求項1-3任一所述之抗體接觸;與測定該抗體與VEGFR2和/或VEGFR3或其片段間的特異性結合。
