Abstract
Description
本出願は2018年3月20日に出願された米国仮出願第62/645,437号の優先権を主張し、その内容は、その全体が参照により本明細書に組み込まれる。
本発明は、単離抗体に関し、特に血管内皮増殖因子受容体−2および血管内皮増殖因子受容体−3の両方を標的とする単離抗体に関する。
VEGF−AとVEGF−Cは、血管新生とリンパ管形成を担う主要なVEGFリガンドと考えられている。これまでの前臨床試験において、その受容体であるVEGFR2またはVEGFR3へのエフェクター結合の遮断は、腫瘍増殖の顕著な阻害につながることが示されている。しかしながら、VEGF−Aを遮断するためのモノクローナル抗体ベバシズマブ(アバスチン)による治療は、一部のがんにおいて進行を遅延させ、生存期間を延長させたのみであった。非特許文献2および3を参照されたい。
腫瘍悪性腫瘍の臨床的失敗を引き起こす主な要因は、がん細胞がリンパ節や遠隔部位に転移する経路となるリンパ管の増殖(リンパ管新生)を遅らせることができないことである。非特許文献4を参照されたい。
したがって、リンパ管新生の選択的阻害薬は血管新生阻害薬を補完すると考えられるが、臨床使用に利用できるものはまだない。
第二のクラスの阻害剤は、PTK787、Sutent、ソラフィニブ、Zactima、およびRecentinのようなVEGFRの細胞内チロシンキナーゼ活性を阻害するための低分子に属する。非特許文献7−11を参照されたい。
ラムシルマブ(登録商標)(Cyramza)およびCDP−791を含む他の高分子治療薬は、VEGF−AとVEGFR−2との相互作用を遮断するために使用される。非特許文献12および13を参照されたい。
これらの薬剤は選択的であり、忍容性が良好であり、一般に、正常臓器におけるVEGF阻害の結果に限定した副作用はわずかである。
対照的に、Igドメイン4−6は受容体複合体の二量体化に関与している。
VEGFR2および/またはVEGFR3の受容体二量体化はそれらの特異的リガンドの生物学的活性を実行するため、それらの下流シグナル伝達の活性化に必要なシナリオである。VEGF−AおよびVEGF−CがVEGFR3によるVEGFR2のホモ二量体化またはヘテロ二量体化を引き起こす可能性がこれまでの報告で示されていることから、VEGFR2の二量体化領域を標的とすることで、VEGFR2および/またはVEGFR3受容体を介したシグナル伝達活性化が重篤に障害され、関節リウマチにおける血管新生、リンパ管新生、腫瘍転移、糖尿病関連網膜症、血管炎などのVEGF−AおよびVEGF−C誘発病因の遮断をもたらす可能性があると仮定した。
新規に開発されたモノクローナル抗体(mAbs)の結合特殊性を調べるため、R2C7−1およびR2C9−11、ヒト静脈内皮細胞(HUVEC)細胞およびVEGFR2(HEK293T/VEGFR)を表現するHEK293T細胞をモデルとして用いた。両抗体は、HUVEC細胞においてVEGFR2に対して同等の親和性を有した(図1Aおよび1C)。R2C7−1およびR2C9−11の結合特殊性をさらに確認するため、HEK293T細胞はヒトVEGFR2またはその空ベクトル(HEK293T/VC)をエンジェイした遺伝子と安定的にトランスフェクトした。VEGFR2の発現量は、まず臨床使用VEGFR2抗体であるラムシルマブ(登録商標)により測定した。フローサイトメトリ解析から得られた結果から、新たに開発された抗体のVEGFR2結合はラムシルマブ(登録商標)と同等であることが示された。R2C7−1およびR2C9−11 mAbsの結合特殊性は、HEK293T/VCトランスフェクタンズにおいても実証され、VEGFR2、ABCAM(登録商標)9530に対する市販の抗体によって確認された微量汚れを示した(データは示していない)。
R2C7−1およびR2C9−11 mAbsの前処理がVEGFR2およびVEGFR3ヘテロ二量化を防止できるかどうかを調べるため、高レベルVEGFR3および低レベルVEGFR2を発現するHEL92.1.7、ならびにVEGFR2およびVEGFR3(HEK293T/VEGF−R2/−R3)細胞を共発現するHEK293Tを細胞モデルとして用いた。共免疫(IP)アッセイを用いて、HEL92.1.7でVEGFR2を引き下げ、またはVEGFR3を引き下げ、HEK293T/VEGF−R2/−R3トランスフェクタにおけるVEGFR2のブロットを行ったところ、特にR2C9−11において、その受容不均化に対する阻害能力がラムシルマブ(登録商標)よりも優れていることが明らかになった。図2Aを参照されたい。
リガンドが誘導する受容体二量体化の後、受容体自己リン酸化の開始とその下流のシグナル伝達活性化は必須の事象であり、結果として生じるリガンドの生物学的活性を説明する。受容体二量体化の遮断がそのダウンストリームシグナル伝達を減弱させるのに充分であるかどうかを調べるため、HUVEC細胞を飢餓状態にし、R2C7−1またはR2C9−11 mAbまたはその抗体アイソタイプ対照(IgG2a)で前処理した。ラムシルマブ(登録商標)で同時に前処理した細胞を対照とした。その後、細胞は、VEGF−A165の処置、血清刺激、または処置なしを受けた。
新規開発抗体によるVEGF−AおよびVEGF−C誘導シグナル伝達活性化の弱毒化がHUVECおよびHLEC細胞の生物学的挙動を調節できるかどうかを調べるため、2Dチューブ形成アッセイを行った。HUVECおよびHLEC細胞を、VEGFおよび他のサプリメントの非存在下で、マトリゲル(登録商標)被覆ウェル上に播種した。細胞をラムシルマブ(登録商標)、R2C7−1、R2C9−11または抗体アイソタイプ対照(IgG2a)で前処理し、VEGF−AまたはVEGF−Cでさらに10時間刺激した。写真撮影後、チューブメッシュとマスターセグメントの総数を画像Jの血管新生アナライザーで定量した結果、R2C9−11は、ラムシルマブ(登録商標)と比較して、VEGF−A−またはVEGF−C誘発性血管チューブ形成に対して優れたまたは匹敵する抑制効果を有することが示された。図3を参照されたい。しかしながら、R2C7−1 mAbは、その対応物より明らかに弱かった。
いくつかの報告は、血管新生およびリンパ管新生における周皮細胞の中心的役割を実証している。Ozerdem U,2006,Ophthalmic Res,2006.38(5):251−4を参照されたい。周皮細胞として働くWI38線維芽細胞がインビボでリンパ管形成を促進できるかどうかを調べるため、HLEC細胞をモデルとしてマトリゲル(登録商標)プラグアッセイを行った。HLEC/WI38混合物を氷−寒冷マトリゲル(登録商標)に埋め込み、NOD−SCIDの各マイナーに皮下注射した。重合したマトリゲル(登録商標)を切除し、固定し、2または4週間パラフィンに包埋した。組織マトリゲル(登録商標)中のヒトWI38およびHLEC細胞の免疫組織化学分析を、それぞれ3,3’−ジアミノベンジジン(DAB)で染色したヒトα−小筋アクチン(SMA)および3−アミノ−9−エチルカルバゾール(AEC)で染色したCD31に特異的な抗体で二重染色することにより行った。WI38細胞はヒトSMAに特異的な抗体で検出され、2週間以内に新たなヒトリンパ管の形成が見出される前に、ヒトCD31に対する抗体でプローブされた(データは示していない)、マウス組織中の新たな血管に合流したことが実証された。4週間後、HLEC細胞は右下から左上に移動し、より大きなリンパ管を形成した(データは示さず)。このデータは、リンパ管形成における周皮細胞の重要な役割を強く支持する。
上記と同じモデルを用いて、HUVECまたはHLEC細胞をWI38と予め混合し、マトリゲル(登録商標)に包埋し、NOD−SCIDマウスに皮下接種した。マウスにラムシルマブ(登録商標)、R2C7−1、R2C9−11、または対照抗体を週2回、4週間投与した。AECによって部分的または完全に汚れたチューブをNIS−ARソフトウェアを用いて定量化した。R2C7−1またはR2C9−11で処理したマウスは対照マウスと比較した場合、血管形成の劇的な減少を示し、特にリンパ管形成に関してラムシルマブ(登録商標)よりも優れた機能を示した。図5を参照されたい。
新しく開発した抗体はVEGFR3を特異的に標的とすることができたので、VEGFR3を発現する細胞においてそれらの細胞毒性能力を綿密に調べた。HEL92.1.7細胞を用い、VEGFR2およびVEGFR3の発現状態を最初に特徴付けた。HEL92.1.7のフローサイトメトリ分析はVEGFR3の高い発現(〜95%)を明確に明らかにしたが、VEGFR2の低い発現(〜5%)を明らかにした。図7Aを参照されたい。NOD−SCIDマウスから分離した末梢血単核球(PBMC)を用いて抗体依存性細胞傷害(ADCC)を行った実験では、ラムシルマブ(登録商標)を含むすべての検査抗体が0.125μg/mLの低濃度の抗体でも、PBMCおよび2%児血清の存在下でHEL92.1.7腫瘍細胞を殺傷する顕著な能力を示すことが示された。図7Bを参照されたい。
新たに開発された抗体とラムシルマブ(登録商標)との併用により、より良好な腫瘍抑制効果が得られるかどうかを検討するため、R2C7−1、R2C9−11又はラムシルマブ(登録商標)を単独又は併用投与されたマウス(10例/群)を用い、HEL92.1.7をモデルとして検討された。接種7日後に抗体(計10mg/kg)の静脈内投与を行い、週2回35日間投与した。興味深いことに、R2C7−1またはR2C9−11とラムシルマブ(登録商標)との併用は、R2C7−1またはR2C9−11単独治療と比較した場合、腫瘍抑制に対してより優れた治療法を与えなかった。しかしながらR2C9−11治療単独は、他の抗体または併用よりも有意に優れた治療効果を提供することができた。図9を参照されたい。したがって、これらのデータは、HEL92.1.7のようなVEGFR3発現腫瘍細胞を標的とすることはそれらの細胞生存を減弱させるだけでなく、ベビー血清中の活性化補体の存在下で試験した抗体を標的とすることによって腫瘍細胞毒性を誘導する可能性を強く示唆する。
R2C7−1およびR2C9−11抗体の認識エピトープを明らかにするため、細胞ベースの競合酵素免疫測定法(ELISA)を実施した。HEK293T/VEGFR2トランスフェクタント(5×104/ウェル)をコーティングし、96ウェルプレート上に固定した。細胞に結合するためのR2C7−1およびR2C9−11抗体の最適濃度、対数期の最高濃度を、標準的なELISA手順を用いて決定した(データは示さず)。続いて最適濃度を有する抗体を、結合エピトープを競合させるため、VEGFR2(R2−IgD)の個々の組換え細胞外ドメインの設計用量で吸収した。R2C7−1とR2C9−11の主要認識エピトープはIgD6とIgD7の間に位置したが、わずかな差異を示した。R2C7−1はR2C9−11と比較した場合、IgD6に対してより強い結合を示した。図10を参照されたい。新しく開発された抗体のエピトープをさらに定義するため、アラニンスキャニング突然変異誘発を行った。VEGFR2のIgD6とIgD7との間の任意のアミノ酸変化は両方の抗体の結合能力の劇的な損失を引き起こし(データは示さず)、このことは新しく開発された抗体の認識エピトープが構造エピトープであり得ることを示す。
抗体のCDRを定義するため、KabatおよびChothia系を適用して、R2C7−1およびR2C9−11抗体の可変領域を定義した。両方の抗体分泌ハイブリドーマが単一クローンであることを確認するため、細胞を2つの連続した連続希釈によって選択した。抗体をコードする遺伝子は、5’−RACEまたは5’−/3’−変性IgG特異的プライマーを用いたRT−PCRによりクローン化した。表1を参照されたい。新しく開発された抗体からクローニングされたCDRを表2に示す。KabatおよびChothia系によって定義された重鎖CDR1(CDR−H1)およびCDR2(CDR−H2)のアミノ酸配列はR2C7−1およびR2C9−11で大きく異なることが示されたが、重鎖CDR3(CDR−H3)では区別がなかった。軽鎖のCDR領域を定義するため、2つの系は差異を示さなかった。これらのデータを総合すると、R2C7−1とR2C9−11の間のCDRのアミノ酸配列の変動のため、これらの2つの抗体は両方ともIgD6−7ドメインに潜在的な結合を示したが、VEGFR2とVEGFR3上に明確な結合エピトープを有する可能性がある。
(1.細胞培養)
NHK293T細胞を、10%FBSと1X抗生物質−抗真菌物質(GIBCO(登録商標)、Grand Island、NY)で補ったDMEM中で、5%CO2インキュベータ中、37℃で培養した。ヒト臍静脈内皮細胞(HUVEC)細胞を、内皮基礎培地−2(EGM−2)および種々の増殖因子、サイトカインおよび他のサプリメント中、SingleQuotキット(Lonza(登録商標)、Basel、Switzerland)中、37℃で、5%CO2インキュベータ中で維持した。実験で使用した初代培養細胞は第3−第6世代であった。ヒトリンパ管内皮細胞(LEC)をPromoCell(登録商標)(Heidelberg、Germany)から購入し、製造者の指示に従って内皮細胞培地MV(PromoCell(登録商標))で培養した。
BALB/Cマウスを血管内皮増殖因子受容体2(VEGFR2)のヒト組換え免疫グロビン(Ig)ドメイン4〜7で、2週間隔で3回、腹腔内(IP)免疫した。VEGFR2の細胞外ドメイン4〜7をコードする遺伝子を、遺伝子特異的プライマーおよびHUVECの第1株cDNAライブラリーを鋳型としてPCRにより作製し、その後pGEX−KGベクターにクローニングした。得られたプラスミド、pGEX−VEGFR2−ED Ig D4−7を直接配列決定により検証した。コードされたタンパク質を大腸菌発現系で発現させ、以下の「遺伝子構築物および発現」の節に記載されるように、グルタチオン結合セファロースビーズ(Sigma−Aldrich(登録商標)、St.Louis、MO)を用いて精製した。血清を採取し、間接酵素免疫測定法(ELISA)により抗VEGFR2抗体の有無を検査した。高レベルの抗体力価(>1:3000希釈)が検出された後、マウスは、フロイントアジュバントの非存在下で、25μgの組換え抗原による最終IP追加免疫を受けた。最後の追加免疫の5日後、マウスを屠殺した。その脾臓細胞を回収し、改変ハイブリドーマ技術によって骨髄腫細胞と融合させた(Apiratmateekul,N.,PPhunpae and WKasinrerk,Cytotechnology,2009.60(1−3):p.5参照)。簡単に述べると、脾細胞(7×107細胞)を、50%ポリエチレングリコール4000(PEG 4000。Sigma−Aldrich(登録商標)、St.Louis、MO)を用いて、2×107のSp2/0−Ag14(ATCC(登録商標)CRL−1581)細胞と、スタンダードハイブリドーマ技術によって融合させた。HAT培地(Sigma−Aldrich(登録商標))選択後、ハイブリドーマ含有ウェルから得られた培養上清を、標準的な間接ELISA手順を用いて抗体−抗原反応性について分析した。450nm(OD450nm)≧0.60での光学濃度の値を有するハイブリドーマを陽性とみなし、陽性ウェルの単細胞クローニングを、限界希釈法を用いて行った。分泌マウスモノクローナル抗体(mAb)のアイソタイプを、マウスmAbアイソタイピングキット(Thermo Scientific(登録商標)Pierce、Rockford、IL)を用いて決定した。
抗VEGFR2 mAbを産生するハイブリドーマを、ハイブリドーマ無血清培地(Gibco)を用いて無血清条件下で培養した。mAbを、TOYOPEARL(登録商標)AF−rProtein A(Griesheim、Germany)を使用するアフィニティクロマトグラフィーによって培養上清から精製した。氷冷PBS(10×床容量)で洗浄した後、プロテインAビーズによって捕捉された抗体を、溶出緩衝液(R2C7−1については50mMグリシン−HCl、pH 3.0、またはR2C9−11については50mMグリシン−HCl/150mM NaCl、pH 3.0)で溶出した。溶出した画分を、100μLの中和緩衝液(1M Tris−HCl、pH 8.0/1.5M NaCl/1mM EDTA。1mL/チューブ)を含むチューブに集めた。ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動(SDS−PAGE)および間接ELISAによる純度および活性。mAbの濃度はBradfordタンパク質アッセイ(BIO−RAD(登録商標)、Hercules、CA)を用いて決定し、4℃で一晩PBSに対して透析した。その後、抗VEGFR2 Abを、さらなる使用のため80℃で保存した。
培養ウェルを氷冷マトリゲル(登録商標)(80μL/ウェル、BD Biosciences、San Jose、CA)でコーティングし、続いて37o℃で30分間インキュベートしてゲル化させた。LECおよびHUVECを、VEGFおよび他のサプリメントの非存在下で、EGM−2中で8時間飢餓させた。トリプシン処理後、細胞を、示されるように、2μg/mLのVEGF−A165またはVEGF−C(Cys156Ser)(R&D、McKinley Place、NE)の存在下で、ラムシルマブ(登録商標)(5μg/mL、Eli Lilly(登録商標)、Indianapolis、IN)、抗VEGFR2モノクローナル抗体(20μg/mL、R2C7−1またはR2C9−11)を含む培地(R&D、McKinley Place、NE)に再懸濁し、続いて、それぞれ、LECまたはHUVECについて37℃で、4時間または10時間、プレコートされたウェルに播種した。NIKON(登録商標)Digital Slight DS−U3 system(東京、日本)を用いて、それぞれのウェルの全体画像を撮影し、形成された分岐節、メッシュ、およびチューブの長さの総数を、画像Jの血管新生分析器によって定量した。データは、手段±S.E.として三連で表した。
細胞をリン酸緩衝生理食塩水(PBS)で洗浄し、2mM PBSベースのEDTAで解離させ、遠心分離によって収集した。非特異的結合を遮断するため、細胞を4℃で1時間、1%ウシセラミアルブミン(BSA)で潜伏させた。PBS洗浄後、VEGFR2(1×105/反応)を発現するHUVECまたはHEK293T細胞を、2μG/mL抗VEGFR2 mAbs(R2C7−1またはR2C9−11)、ラムシルマブ(登録商標)(Eli Lilly(登録商標)、Indianapolis、IN)またはそれらのイソタイプ制御免疫グロビンG(IgG)(BioLegend、San Diego、CA)で、4Cでさらに1時間処理した。VEGFR3発現のため、VEGFR3(Santa Cruz、Dallas、TX)またはウサギIgG対照(ABCAM(登録商標)、Burlingame、CA)に対して1h免疫プローブした。1%BSA含有PBSで広範囲に洗浄した後、細胞を、それらの対応する二次免疫、4μG/mLゴート抗IgG(BioLegend)、FITC(ABCAM(登録商標))と共役したドンキー抗ウサギIgG、またはマウス抗ヒトで染色した。PE(BioLegend)と共役したIgGを4℃で1h洗浄し、その後、細胞をPBS+1%パラホルムアルデヒド及び1%BSA中で再浮遊させた。VEGFR2およびVEGFR3の細胞表面発現を、BD FACS Calibur(登録商標)フローサイトメータ(Becton Dickinson、Franklin Lakes、NJ)を用いて分析した。
抗VEGFR2(R2C7−1およびR2C9−11)mAbによるVEGFR2およびVEGFR3ホモ−/ヘテロ−二量体化の遮断を、製造者の指示に従ってDuolinkインサイチュPLAキット(Sigma−Aldrich(登録商標)、St.Louis、MO)を用いて調査した。簡単に述べると、LECを8ウェルチャンバースライドに播種し、R2C7−1、R2C9−11、またはラムシルマブ(登録商標)(Eli Lilly(登録商標))の存在下で無血清培地と共に16時間培養した。その後、細胞をVEGF−C(500ng/mLで15分間)、VEGF−C(Cys156Ser)(2μg/mLで10分間)またはVEGF−A165(50ng/mLで5分間)で刺激した。細胞をPBSで洗浄し、4%パラホルムアルデヒドで15分間固定し、ブロッキング緩衝液(Sigma−Aldrich(登録商標))で30分間、RTでブロックした。2μg/mLのVEGFR2またはVEGFR3(Santa Cruz、Dallas、TX)に対する市販の抗体で染色した細胞を対照とした。特異的オリゴヌクレオチド(PLAプローブ)に連結した対応する種特異的二次抗体、抗ウサギPLUSまたは抗マウスMINUS(Sigma−Aldrich(登録商標))を添加した。リガーゼ(最終濃度1単位/μL、Sigma−Aldrich(登録商標))を含む液を37℃で30分間添加することにより、PLAプローブ反応のライゲーションを行った。シグナル増幅は連結したPLAプローブを37℃で100分間ローリングサークル増幅することにより、ポリメラーゼ含有溶液(10単位/μL、Sigma−Aldrich(登録商標))中で行った。緩衝液B(Sigma−Aldrich(登録商標))で洗浄した後、スライドをDAPIで染色して細胞核を可視化し、Duolinkインサイチュ マウント培地(Sigma−Aldrich(登録商標))でマウントした。さらに、アイソタイプ対照IgGを対照として使用した。蛍光シグナル(またはドット)を画像化し、MetaMorph Offlineソフトウェアによって分析した。
抗VEGFR2抗体によるVEGF−C誘導受容体二量体化に対する阻害効果を決定するため、LEC細胞を、それらが70〜80%コンフルエンシーに達するまで、直径6cmの皿に播種した。細胞を洗浄し、20μg/mLの抗体(R2C7−1、R2C9−11、またはアイソタイプ対照抗体)を含有する無血清培地で16時間飢餓させた。続いて、細胞を、VEGF−C(500ng/mL)を用いてまたは用いずに15分間刺激し、氷冷PBSで2回洗浄し、RIPA緩衝液(20mM Tris−HCl(pH7.5)/150mM NaCl/1mM Na2 EDTA/1mM EGTA/1% NP−40/1%デオキシコール酸ナトリウム/2.5mMピロリン酸ナトリウム/1mM β−グリセロリン酸/1mM Na3 VO4/1μg/mLロイペプチン)によって回収した。免疫沈降のため、溶解物(1mg/サンプル)をプロテイン−A/G−アガロースビーズ(Thermo Fisher(登録商標)、Waltham、MA)で2時間前洗浄し、非特異的結合を最小限に抑え、次いで抗VEGFR3抗体(1μg/サンプル、Santa Cruz)と4℃で一晩インキュベートし、免疫沈降したタンパク質を天然ポリアクリルアミドゲル電気泳動によって分離し、市販のVEGFR3抗体(Santa Cruz)で免疫ブロットし、以下に記載するように、ECLウェスタンブロットシステム(Thermo Scientific(登録商標))を用いて可視化した。
VEGFR2およびVEGFR3の受容体二量体化状態もまた、ネイティブゲル電気泳動(PAGE)によって測定した。細胞を、20μg/mLのR2C7−1またはR2C9−11を含有する無血清培地で16時間飢餓状態にした。ラムシルマブ(登録商標)およびアイソタイプ対照抗体を、それぞれ陽性および陰性対照として使用した。細胞をVEGF−C(500ng/ml)で15分間刺激し、低張溶解緩衝液(20mM Tris−HCl、pH7.4、10mM NaClおよび1mM PMSF)によって回収した。等量のサンプルたんぱく質を4×ネイティブサンプルバッファー(400mM Tris−HCl、pH7.5、40%グリセロールと0.02%ブロモフェノールブルー)で混合し、事前鋳造されたネイティブPAGEゲル(BIO−RAD(登録商標))を用いてネイティブPAGEにより解消した。VEGFRの二量体化およびリン酸化状態を、抗VEGFR2(Santa Cruz)、抗VEGFR3(Santa Cruz)または抗p−VEGFR3抗体(Cell Applications、San Diego、CA)によって検出した。
プロテイン溶解物は、ホスファターゼ阻害剤カクテル(Sigma−Aldrich(登録商標))またはRIPA緩衝液を添加した溶解緩衝液(20mMのTris−HCl pH8.0/2mM EDTA/1mM Na3 VO4/10%グリセロール/1% Triton X−100/プロテアーゼ)を用いて得られた。HUVECおよびLEC細胞を、処置の1日前に播種した。抗VEGFR2抗体、R2C7−1またはR2C9−11(示されるように、5、10、20μg/mL)を培地に添加し、37℃で10分間インキュベートした後、HUVECのため20ng/mLのVEGF−A165(R&D)を刺激した。LEC細胞をR2C7−1またはR2C9−11抗体(20μg/mL)とプレインキュベートし、VEGF−C(500ng/mL、15分間)、VEGF−C(Cys156Ser)(2μg/mL、10分間)またはVEGF−A165(50ng/mL、5分間)(R&D)で刺激し、次いでRIPA緩衝液によって回収した。ラムシルマブ(登録商標)(5μg/mL。Eli Lilly(登録商標))および20μg/mLの適切なアイソタイプ対照IgG(BiolegendからのR2C7−1およびR2C9−11に対するマウスIgG。ABCAM(登録商標)からのVEGFR3抗体に対するウサギIgG)を、それぞれ陽性対照または陰性対照として用いた。抽出たんぱく質の濃度をBradfordたんぱく質アッセイ(BIO−RAD(登録商標))で測定した。12%ドデシル硫酸ナトリウム−ポリアクリルアミドゲル(BIO−RAD(登録商標))で、等量の総タンパク質(50μg/サンプル)を分離した。タンパク質をニトロセルロース膜(HYBOND(登録商標)−P、Amersham(登録商標)Biosciences、Piscataway、NJ)に移した。ブロッキング緩衝液(TBS中3%BSA/0.1%Tween−20)で、室温で2時間ブロッキングした後、膜を、抗VEGFR2(Santa Cruz)、−VEGFR3(Santa Cruz)、p−VEGFR3(Tyr1230/1231)(細胞適用)、p−FAK、p−p38ERK(Cell Signaling、Danvers、MA)およびβ−アクチン(MilliporeSigma、Burlington、MA)またはチューブリンに対する抗体と共に4℃で一晩インキュベートし、続いて西洋ワサビペルオキシダーゼに連結した適切な二次抗体とインキュベートした。SUPERSIGNAL(登録商標)West Pico chemiluminescence(Thermo Scientific(登録商標))を用いて免疫反応性タンパク質を可視化し、シグナルをX線フィルム(FUJI(登録商標)、Tokyo、Japan)上で検出した。
R2C7−1およびR2C9−11抗VEGFR2抗体のエピトープを、細胞ベースの競合ELISAによって決定した。VEGFR2またはVEGFR3を発現するHEK293T細胞を、50000細胞/100μL/ウェルの密度で96ウェルプレート上に播種し、37℃で一晩インキュベートした。広範な洗浄後、細胞をPBS中の1%グルタルアルデヒドで固定し、次いでPBS(PBST)中の0.1%Tween−20で3回洗浄した。非特異的結合を減少させるため、細胞を3%BSA含有PBSで、37℃で30分間ブロックし、一方、R2C7−1およびR2C9−11抗体(8μg/ウェル)を、VEGFR2またはVEGFR3組換えタンパク質の個々の免疫グロブリン様ドメイン(IgD4、D5、D6およびD7)の指示用量(0.2、1.0、または5.0μg/ウェル)で、37℃で30分間吸収させた。続いて、VEGFR2またはVEGFR3を発現する細胞を、予め吸収されたR2C7−1またはR2C9−11抗体と共に37℃で1時間インキュベートし、続いてPBSTで3回広範に洗浄した。結合した抗体をHRPと結合したヤギ抗マウスIgGを用いて37℃で1時間プローブし、ABTS(37℃で30分間)を加えることによって可視化し、OD405でSpectraMax M5マイクロプレートリーダ(Molecular Devices(登録商標)、San Jose、CA)によって測定した。
VEGFR2の細胞外免疫グロビン(Ig)ドメインをpGEX−4T−1プラスミド(GE(登録商標)Healthcare、Chicago、IL)に構築し、組換え蛋白質をコンピテントEcoli細胞系で発現させた。VEGFR2遺伝子(NM_002253.2)の細胞外Igドメインおよびそのサブドメイン6および7を、プラスミドEX−I0114−Lv205(GeneCopoeia、Rockville、MD)を鋳型として使用し、BamH1およびXhoI(NEB、Ipswich、MA)で制限的に消化し、pGEX−4T−1ベクター(GE(登録商標)Healthcare、Chicago、IL)にクローニングし、それぞれVEGFR2−ED Ig D1−7およびD6−7と命名した遺伝子特異的PCRプライマーによって作製した。この遺伝子構築物を、42℃で40秒間の熱ショックによってJM109細胞(PROMEGA(登録商標)、Madison、WI)に形質転換し、そしてLB寒天上のアンピシリン(50μg/mL)によって選択した。抗生物質耐性細胞における遺伝子挿入は、標準的なコロニーPCRおよび遺伝子配列決定により確認した。陽性細胞をアンピシリン含有LBブロス中で増殖させ、1mM IPTG(Sigma−Aldrich(登録商標))により37℃で3時間タンパク質発現を誘導した。細胞をペレット化し、氷冷溶解緩衝液(10mM Tris−HCl pH8.0/0.15 M NaCl/1mM EDTA)中で、100μg/mLリゾチーム、10mM PMSFおよび5単位のベンゾナーゼ(Sigma−Aldrich(登録商標))を添加して疑った。超音波処理後10,000×gで30分間の遠心分離によって可溶性画分を得た。GSTタグ化組換えタンパク質をグルタチオン結合セファロースビーズ(Sigma−Aldrich(登録商標))で捕捉し、10mM還元型グルタチオンで溶出し、PBSに対して一晩透析した。組換えタンパク質の純度をSDS−PAGEによって調べた。

- 配列番号1または配列番号3を有する重鎖可変領域配列由来の重鎖相補性決定領域1、重鎖相補性決定領域2、および重鎖相補性決定領域3と、
配列番号2または配列番号4を有する軽鎖可変領域配列由来の軽鎖相補性決定領域1、軽鎖相補性決定領域2、および軽鎖相補性決定領域3とを含み、
血管内皮増殖因子受容体−2および血管内皮増殖因子受容体−3の両方に特異的に結合する、単離抗体。 - 前記重鎖相補性決定領域1、前記重鎖相補性決定領域2、および前記重鎖相補性決定領域3が配列番号1由来であり、
前記軽鎖相補性決定領域1、前記軽鎖相補性決定領域2、および前記軽鎖相補性決定領域3が配列番号2由来である、請求項1に記載の単離抗体。 - 前記重鎖相補性決定領域1が配列番号5または配列番号8由来であり、
前記重鎖相補性決定領域2が配列番号6または配列番号9由来であり、
前記重鎖相補性決定領域3が配列番号7由来であり、
前記軽鎖相補性決定領域1、前記軽鎖相補性決定領域2および前記軽鎖相補性決定領域3がそれぞれ配列番号10、配列番号11および配列番号12由来である、請求項2に記載の単離抗体。 - 前記重鎖相補性決定領域1、前記重鎖相補性決定領域2、および前記重鎖相補性決定領域3が配列番号3由来であり、
前記軽鎖相補性決定領域1、前記軽鎖相補性決定領域2、および前記軽鎖相補性決定領域3が配列番号4由来である、請求項1に記載の単離抗体。 - 前記重鎖相補性決定領域1が配列番号13または配列番号16由来であり、

前記軽鎖相補性決定領域1、前記軽鎖相補性決定領域2および前記軽鎖相補性決定領域3がそれぞれ配列番号18、配列番号19および配列番号20由来である、請求項4に記載の単離抗体。 - Fc領域、Fab断片、Fab’断片、F(ab’)2断片、一本鎖抗体、scFV多量体、一価抗体、多重特異性抗体、またはキメラ抗体を含む、請求項1〜5のいずれか一項に記載の単離抗体。
- ヒト化抗体である、請求項1〜6のいずれか一項に記載の単離抗体。
- 請求項1〜7のいずれかに記載の単離抗体および薬学的に許容される担体を含む、医薬組成物。
- 請求項1〜7のいずれか一項に記載の単離抗体および別の分子を含む、単離抗体結合体。
- 請求項1〜7のいずれか一項に記載の単離抗体と細胞を接触させることを含み、
細胞における血管内皮増殖因子受容体−2および/または血管内皮増殖因子受容体−3のホモダイマー化、または、前記血管内皮増殖因子受容体−2および前記血管内皮増殖因子受容体−3のヘテロダイマー化を阻害する、ダイマー化阻害方法。 - 請求項1〜7のいずれか一項に記載の単離抗体と細胞を接触させることを含み、
細胞における血管内皮増殖因子受容体−2および/または血管内皮増殖因子受容体−3シグナル伝達を阻害する、シグナル伝達阻害方法。 - 請求項1〜7のいずれか一項に記載の単離抗体の有効量をヒトを除く対象に投与することを含み、

- 前記ヒトを除く対象に、別の治療薬を投与することをさらに含む、請求項13に記載の治療方法。
